Résumé :
Perspective on behavioral finance, we take a new look at the characteristics of investors’ risk preference, building the D-GARCH-M model, DR-GARCH-M model, and GARCHC-M model to investigate their changes with states of gain and loss and values of return together with other time-varying characteristics of investors’ risk preference. Based on a full description of risk preference characteristic, we develop a GARCHCS-M model to study its effect on the return skewness. The top ten market value stock composite indexes from Global Stock Exchange in 2012 are adopted to make the empirical analysis. The results show that investors are risk aversion when they gain and risk seeking when they lose, which effectively explains the inconsistent risk-return relationship. Moreover, the degree of risk aversion rises with the increasing gain and that of risk seeking improves with the increasing losses. Meanwhile, we find that investors’ inherent risk preference in most countries displays risk seeking, and their current risk preference is influenced by last period’s risk preference and disturbances. At last, investors’ risk preferences affect the conditional skewness; specifically, their risk aversion makes return skewness reduce, while risk seeking makes the skewness increase.

Résumé :
Ayurveda the science of medicine deals with the aim of Swastha Sharir. The Rachna Sharir is the ayurveda term which mainly deals with the anatomical and physiological compositions of body. Sushrut samhita is one of those Ayurvedic classic which deals with human anatomy and physiology. It gives precise description of Garbha vruddhi and vikas kram in detail. The knowledge of Sharir starts from Garbha. The understanding of Garbha helps to manage healthy progeny. According to Ayurveda, the union of shukra (spermatozoa), shonita (ovum) and Atma (soul) inside the uterus is known as garbha (embryo). The fertilization between Shukra and Shonita produces zygote which further develops into fetus. The Ritu, Kshetra and Ambu etc. plays vital role in the proper development of fetus.. Different components originating fromFive Elements takes part in the formation, development of the garbha. The whole process of development of the foetus from two cells to mature foetus is called Garbhavakranti. Itincludes the basis of the earliest embryology which was known to the human being. In Ayurvedic texts the process of chronological development of the foetus is explained clearly. The understanding of Garbha developmenthelps to acquire necessary conditions which required for the better management of pregnancy. Extensive study and discussion is essential to understand ancient concept of Garbha vikas kram. Review of Ayurvedic text along with modern text will help to obtain knowledge regarding this. This study is an attempt to elaborate and compare both ancient and modern concept of foetal development. Keywords: Shukra, Shonit, Garbha, Garbhavakranti, Garbha vikas kram.

Vilnius UniversityBy reconsidering the fragments from early texts on north Indian temple architecture, namely the Purāṇas and their supposed sources, this paper sets out to explore the accounts of arrangement of the plans for temple ground plans and the modes of proportional measurement. It is contended that the general system of proportional measurement, called sāmānya or sarvasādhāraṇa and elaborated on in the texts under discussion, comprises temple garbha-shrines of various scales and forms, from which measurements for the entire temple structure are derived. The importance given to the garbha-shrine is attested by the method of classification of diverse temple types that are distinguished by the geometrical form of the ground plan of the garbha-shrine. It is consequently suggested that the arrangement of the ground plan of a garbha-shrine adapted by temple architects most probably reiterated the practices used for building Vedic altars, the layouts of which, as the Śulbasūtras state, might have served as models for structuring the ground plans of garbha-shrines.

Résumé :
AbstractIt has been shown that mice with complete deficiency of all 4.1R protein isoforms (4.1-/-) exhibit moderate hemolytic anemia, with abnormal erythrocyte morphology (spherocytosis) and decreased membrane stability. Here, we characterized the Gardos channel function in vitro and in vivo in erythrocytes of 4.1-/- mice. Compared with wild-type, the Gardos channel of 4.1-/- erythrocytes showed an increase in Vmax (9.75 ± 1.06 vs 6.08 ± 0.09 mM cell × minute; P < .04) and a decrease in Km (1.01 ± 0.06 vs 1.47 ± 1.02 μM; P < .03), indicating an increased sensitivity to activation by intracellular calcium. In vivo function of the Gardos channel was assessed by the oral administration of clotrimazole, a well-characterized Gardos channel blocker. Clotrimazole treatment resulted in worsening of anemia and hemolysis, with decreased red cell survival and increased numbers of circulating hyperchromic spherocytes and microspherocytes. Clotrimazole induced similar changes in 4.2-/- and band 3+/- mice, indicating that these effects of the Gardos channel are shared in different models of murine spherocytosis. Thus, potassium and water loss through the Gardos channel may play an important protective role in compensating for the reduced surface-membrane area of hereditary spherocytosis (HS) erythrocytes and reducing hemolysis in erythrocytes with cytoskeletal impairments. Résumé :
Human red blood cells pretreated with low-ionic-strength solutions and resuspended in saline respond biphasically to extracellular Ca. At first, addition of Ca causes a large transient K efflux of as much as 600 mM . liter cell H2O-1 . h-1; this is followed by a decrease in K flux below control levels. The first phase (phase I) resembles the Gardos effect in several respects. It is inhibited by oligomycin, by external K, and by increased exposure time to Ca. Further, the K permeability of phase I is similar to that of the Gardos effect (5 X 10(-8)-9 X 10(-8) cm/s), and the cells hyperpolarize in a low-K medium when Ca2+ is added. However, phase I is not identical to the Gardos phenomenon. For example, La, which prevents the Gardos response, is ineffective on phase I. Moreover, external Ba prevents the development of phase I but not the Gardos response, whereas internal Ba prevents the Gardos response. Attempts to demonstrate a Ca leak or pump failure during phase I have failed; passive Ca movements of both treated and normal cells are similar. The results suggest that low-ionic-strength solution exposes Ca-sensitive sites to the external medium; these sites are maintained when the cells are returned to saline.

Résumé :
It has recently been shown that the Gardos channel activity of mouse erythrocytes can be modified by endothelins, suggesting a functional linkage between endothelin receptors and the Gardos channel. Using 86Rubidium (86Rb) influx, effects were estimated of proinflammatory molecules such as platelet activator factor (PAF), endothelin-1 (ET-1), interleukin-10 (IL-10), and regulated on activation normal T cells expressed and secreted (RANTES) on the Gardos channel activity in human normal and sickle red cells. It was found that PAF (EC50: 15 ± 7 nM), RANTES (EC50, 9 ± 6 ng/mL [1.2 ± 0.8 nM]), IL-10 (EC50, 11 ± 8 ng/mL [204 ± 148 nM]), and ET-1 (EC50, 123 ± 34 nM) induce a significant increase in Gardos channel activity—between 28% and 84%—over the control. In addition, these agents modify the Gardos channel affinity for internal Ca++ (K0.5) by 2- to 6-fold. Biochemical evidence is provided for the presence of ET receptor subtype B in sickle and normal red cells. Furthermore, it was found that ET-1, PAF, RANTES, and IL-10 induce a significant increase in red cell density (P < .05). These data suggest that activation of the Gardos channel is functionally coupled to receptor motifs such as C-X-C (PAF), C-C (RANTES), and ET receptor subtype B. Thus, cell volume regulation or erythrocyte hydration states might be altered by activation of the Gardos channel by cytokines in vivo. The role of these mediators in promoting sickle cell dehydration in vivo is under investigation.

Résumé :
The Ca2+-sensitive K+ channel of human red blood cells (RBCs) (Gardos channel, hIK1, hSK4) was implicated in the progressive densification of RBCs during normal senescence and in the mechanism of sickle cell dehydration. Saturating RBC Ca2+ loads were shown before to induce rapid and homogeneous dehydration, suggesting that Gardos channel capacity was uniform among the RBCs, regardless of age. Using glycated hemoglobin as a reliable RBC age marker, we investigated the age–activity relation of Gardos channels by measuring the mean age of RBC subpopulations exceeding a set high density boundary during dehydration. When K+ permeabilization was induced with valinomycin, the oldest and densest cells, which started nearest to the set density boundary, crossed it first, reflecting conservation of the normal age–density distribution pattern during dehydration. However, when Ca2+ loads were used to induce maximal K+ fluxes via Gardos channels in all RBCs (Fmax), the youngest RBCs passed the boundary first, ahead of the older RBCs, indicating that Gardos channel Fmax was highest in those young RBCs, and that the previously observed appearance of uniform dehydration concealed a substantial degree of age scrambling during the dehydration process. Further analysis of the Gardos channel age–activity relation revealed a monotonic decline in Fmax with cell age, with a broad quasi-Gaussian Fmax distribution among the RBCs.

Résumé :
Abstract Arginine (ARG) supplementation (5% arginine in chow) restores plasma ARG levels in S+S-Antilles mice to that found in C57BL mice and is associated with a reduction in Ca++ activated K+ channel (Gardos channel) activity, MCHC and high density red cells (Romero, Blood 99(4):2002). However the mechanisms for these effects are not entirely clear. Endothelin-1 (ET) is a potent vasoconstrictor that has been shown to activate Gardos channel activity and reduce MCHC. We hypothesized that reduced ET could account for the decrease in Gardos channel activity. We report here that arginine diet reduced ET levels in mouse plasma from 137 ± 10 to 89 ± 17 pmol/ml using a modified HPLC technique originally proposed by Kumarathasan (Anal Biochem299:37, 2001). We measured Gardos channel activity in ex vivo red cells as the influx of 86Rb in the presence or absence 10μM clotrimazole. Consistent with our previous report, we observed that ARG supplementation was associated with a decrease in Gardos Channel activity. In addition, incubation of red cells with 500 nmol/L of ET for 15 min at 37°C stimulated an increase in Gardos channel activity in cells from both ARG supplemented vs non-supplemented mice that was sensitive to 1 mmol/L of the specific ET receptor B antagonist, BQ788 (9.4 ± 2.5 vs 14.6 ± 3.8 mmol/L cell x h, respectively). However, the maximal response of the Gardos channel activity to ET was significantly blunted in ARG supplemented vs non-supplemented mice (15.7 ± 2.8 vs 22.1 ± 2.2 mmol/L cell x h, p<0.03, n=5). We also performed QT-RT PCR using Taqman assays on aortas from these mice and observed that mRNA expression of both ET and ET receptor A levels were significantly decreased in ARG supplemented vs non-supplemented mice (p<0.05, n=5) while ET receptor B levels were not significantly affected (n=5). Thus, our results suggest that reduced ET is in part responsible for the blunted Gardos channel activity in arginine supplemented mice and raises the possibility that ET metabolism is modulated by arginine supplementation. We conclude that arginine supplementation of sickle cell patients may have multiple benefits including reduced polymer formation, reduced vasoconstriction, and reduced inflammatory response.

So, Ayurveda needs a deep study and research for proving all the facts established by Acharya in ancient time. In Ayurveda, the concept of Garbha & Garbhotpatti is described systematically. Acharya have explained four important factors as Garbha Sambhav Samagri that is Ritu, Kshetra, Ambu and Beeja. These factors are very essential for conception. Infertility may occur if there is any abnormality in Garbha Sambhav Samagri. So, for a healthy and best offspring the factors concerned with conception must be in good qualities. The knowledge of Garbhotpatti helps to create suitable conditions for conception and healthy foetus also. This study will help to understand the normal and abnormal features of Garbha Sambhav Samagri in production of a healthy child.

Résumé :
Cell dehydration mediated by Ca2+-activated K+channels plays an important role in the pathogenesis of sickle cell disease. CD-1 mouse erythrocytes possess a Ca2+-activated K+channel (Gardos channel) with maximal velocity ( Vmax) of 0.154 ± 0.02 mmol ⋅ l cells−1⋅ min−1and an affinity constant ( K0.5) for Ca2+of 286 ± 83 nM in the presence of A-23187. Cells pretreated with 500 nM endothelin-1 (ET-1) increased their Vmaxby 88 ± 9% ( n = 8) and decreased their K0.5for Ca2+to 139 ± 63 nM ( P < 0.05; n = 4). Activation of the Gardos channel resulted in an EC50of 75 ± 20 nM for ET-1 and 374 ± 97 nM for ET-3. Analysis of the affinity of unlabeled ET-1 for its receptor showed two classes of binding sites with apparent dissociation constants of 167 ± 51 and 785 ± 143 nM and with capacity of binding sites of 298 ± 38 and 1,568 ± 211 sites/cell, respectively. The Gardos channel was activated by the endothelin B (ETB) receptor agonist IRL 1620 and inhibited by BQ-788, demonstrating the involvement of ETBreceptors. Calphostin C inhibited 73% of ET-1-induced Gardos activation and 84% of the ET-1-induced membrane protein kinase C activity. Thus endothelins regulate erythrocyte Gardos channels via ETBreceptors and a calphostin-sensitive mechanism.

Résumé :
Pregnancy is a state of high nutritional demand. The diet taken by the pregnant women is utilised in three ways as said by the classics - For the nourishment of pregnant women, for the production of breast milk and the proper nourishment of the foetus. So, the health and growth of the foetus depend mainly on the diet and nutrition of the mother. With this viewpoint, garbhini paricharya is explained. A condition in our classics has been explained as ‘Garbha Shosha’ where there is retarded growth of the foetus in the womb. ‘Aaharamaapnothi yathaa na garbhaha’ is said as the nidana of garbha shosha by Acharya Charaka which states that this condition arises where the foetus is not provided with a nutritious diet through the mother. Acharya Vagbhata points out that vitiation of ‘Rasavaha Srotas’ happens in the same. Acharya Bhavamishra advised brimhana drugs with mamsarasa in this condition for nutrition. Strict adherence to masanumasika pathya may help in preventing garbha shosha by promoting the growth of the foetus. Keywords: Garbha shosha, Diet, Nutrition, Brimhana, Masanumasika pathya
