ABSTRACT Strains of Francisella tularensis secrete a siderophore in response to iron limitation. Siderophore production is dependent on fslA, the first gene in an operon that appears to encode biosynthetic and export functions for the siderophore. Transcription of the operon is induced under conditions of iron limitation. The fsl genes lie adjacent to the fur homolog on the chromosome, and there is a canonical Fur box sequence in the promoter region of fslA. We generated a Δfur mutant of the Schu S4 strain of F. tularensis tularensis and determined that siderophore production was now constitutive and no longer regulated by iron levels. Quantitative reverse transcriptase PCR analysis with RNA from Schu S4 and the mutant strain showed that Fur represses transcription of fslA under iron-replete conditions. We determined that fslE (locus FTT0025 in the Schu S4 genome), located downstream of the siderophore biosynthetic genes, is also under Fur regulation and is transcribed as part of the fslABCDEF operon. We generated a defined in-frame deletion of fslE and found that the mutant was defective for growth under iron limitation. Using a plate-based growth assay, we found that the mutant was able to secrete a siderophore but was defective in utilization of the siderophore. FslE belongs to a family of proteins that has no known homologs outside of the Francisella species, and the fslE gene product has been previously localized to the outer membrane of F. tularensis strains. Our data suggest that FslE may function as the siderophore receptor in F. tularensis.

The finite size Lyapunov exponent (FSLE) has been used extensively since the late 1990s to diagnose turbulent regimes from Lagrangian experiments and to detect Lagrangian coherent structures in geophysical flows and two-dimensional turbulence. Historically, the FSLE was defined in terms of its computational method rather than via a mathematical formulation, and the behavior of the FSLE in the turbulent inertial ranges is based primarily on scaling arguments. Here, we propose an exact definition of the FSLE based on conditional averaging of the finite amplitude growth rate (FAGR) of the particle pair separation. With this new definition, we show that the FSLE is a close proxy for the inverse structural time, a concept introduced a decade before the FSLE. The (in)dependence of the FSLE on initial conditions is also discussed, as well as the links between the FAGR and other relevant Lagrangian metrics, such as the finite time Lyapunov exponent and the second-order velocity structure function.

ABSTRACT The Gram-negative pathogen Francisella tularensis secretes a siderophore to obtain essential iron by a TonB-independent mechanism. The fslABCDE locus, encoding siderophore-related functions, is conserved among different Francisella strains. In the virulent strain Schu S4, fslE is essential for siderophore utilization and for growth under conditions of iron limitation. In contrast, we found that deletion of fslE did not affect siderophore utilization by the attenuated live vaccine strain (LVS). We found that one of the fslE paralogs encoded in the LVS genome, FTL_0439 (fupA/B), was able to partially complement a Schu S4 ΔfslE mutant for siderophore utilization. We generated a deletion of fupA/B in LVS and in the LVS ΔfslE background. The ΔfupA/B mutant showed reduced growth under conditions of iron limitation. It was able to secrete but was unable to utilize siderophore. Mutation of both fupA/B and fslE resulted in a growth defect of greater severity. The ΔfupA/B mutants showed a replication defect in J774.1A cells and decreased virulence following intraperitoneal infection in mice. Complementation of the ΔfupA/B mutation in cis restored the ability to utilize siderophore and concomitantly restored virulence. Our results indicate that fupA/B plays a significant role in the siderophore-mediated iron uptake mechanism of LVS whereas fslE appears to play a secondary role. Variation in iron acquisition mechanisms may contribute to virulence differences between the strains.

Abstract. Coastal transport in the Bay of Palma, a small region in the island of Mallorca, Spain, is characterized in terms of Lagrangian descriptors. The data sets used for this study are the output for two months (one in autumn and one in summer) of a high resolution numerical model, ROMS (Regional Ocean Model System), forced atmospherically and with a spatial resolution of 300 m. The two months were selected because of their different wind regime, which is the main driver of the sea dynamics in this area. Finite-size Lyapunov exponents (FSLEs) were used to locate semi-persistent Lagrangian coherent structures (LCS) and to understand the different flow regimes in the bay. The different wind directions and regularity in the two months have a clear impact on the surface bay dynamics, whereas only topographic features appear clearly in the bottom structures. The fluid interchange between the bay and the open ocean was studied by computing particle trajectories and residence time (RT) maps. The escape rate of particles out of the bay is qualitatively different, with a 32% greater escape rate of particles to the ocean in October than in July, owing to the different geometric characteristics of the flow. We show that LCSs separate regions with different transport properties by displaying spatial distributions of residence times on synoptic Lagrangian maps together with the location of the LCSs. Correlations between the time-dependent behavior of FSLE and RT are also investigated, showing a negative dependence when the stirring characterized by FSLE values moves particles in the direction of escape.

This article studies the anisotropic particle dispersion in a continuously forced, two-dimensional turbulent flow on a β-plane. The flow is immersed in a large-scale closed basin with free-slip walls. The anisotropy is analyzed in two sets of numerical experiments characterized by the magnitude of the imposed, time-dependent forcing (weak and strong). Both experiments exhibit typical features of zonostrophic turbulence: eddy motions that, on average, form alternating east–west circulation bands due to the β-effect. The dispersion anisotropy is investigated through three Lagrangian statistics calculated by zonal and meridional components: (i) relative dispersion between pairs of particles; (ii) dispersion ellipses; and (iii) finite-scale Lyapunov exponents (FSLE), also measured with particle pairs. In the experiment with weak forcing, the relative dispersion and dispersion ellipses show anisotropy with a zonal preference toward the west; however, the FSLE did not reveal significant anisotropy. In the experiment with strong forcing, the relative dispersion and dispersion ellipses show zonal anisotropy toward the west when the particles are far from the boundaries. As the particles reach the western wall and are redistributed to fill the domain, the anisotropy ceases. The FSLE show zonal anisotropy for a wide range of particle separations. The results are examined further by using no-slip boundary conditions and a rectangular domain geometry.

Abstract To develop methodologies to maximize the information content of Lagrangian data subject to position errors, synthetic trajectories produced by both a large-eddy simulation (LES) of an idealized submesoscale flow field and a high-resolution Hybrid Coordinate Ocean Model simulation of the North Atlantic circulation are analyzed. Scale-dependent Lagrangian measures of two-particle dispersion, mainly the finite-scale Lyapunov exponent [FSLE; λ(δ)], are used as metrics to determine the effects of position uncertainty on the observed dispersion regimes. It is found that the cumulative effect of position uncertainty on λ(δ) may extend to scales 20–60 times larger than the position uncertainty. The range of separation scales affected by a given level of position uncertainty depends upon the slope of the true FSLE distribution at the scale of the uncertainty. Low-pass filtering or temporal subsampling of the trajectories reduces the effective noise amplitudes at the smallest spatial scales at the expense of limiting the maximum computable value of λ. An adaptive time-filtering approach is proposed as a means of extracting the true FSLE signal from data with uncertain position measurements. Application of this filtering process to the drifters with the Argos positioning system released during the LatMix: Studies of Submesoscale Stirring and Mixing (2011) indicates that the measurement noise dominates the dispersion regime in λ for separation scales δ < 3 km. An expression is provided to estimate position errors that can be afforded depending on the expected maximum λ in the submesoscale regime.

Abstract A detailed analysis of Lagrangian tracer dispersion is performed on datasets obtained from laboratory experiments that simulate rotating turbulence in the presence of a β effect. Compatible with the limitations of the experimental apparatus, a relatively wide range of the zonostrophy index Rβ, a parameter used to characterize flow regimes in β-plane turbulence, is explored. The considered range spans from values ~O(10−1), for which the flow is nearly isotropic, to values ~O(1), corresponding to the so-called transitional range in which the flow gradually leaves the friction-dominated regime to enter the full zonostrophic regime. The degree of anistropy and the characteristic scales of the flow have been estimated by means of a Lagrangian approach based on the reconstruction of tracer trajectories and on the measure of the finite-scale Lyapunov exponents (FSLE). The FSLE analysis allows one to identify the regimes of two-particle dispersion and to relate them to the physical parameters of the system. Moreover, a Lagrangian anisotropy index (LAI) is introduced and defined in terms of the FSLE zonal and radial components, in order to describe the onset of anisotropy and to check if it is consistent with the theoretical predictions. It is remarkable that the finite-scale dispersion rates are very sensitive to the degree of anistropy of turbulence, more so than other indicators defined in terms of Eulerian quantities. Furthermore, they offer an effective diagnostic tool of the degree of anisotropy that can be used even prior to attaining a fully developed regime of zonostrophic turbulence.

Recent studies demonstrate that energetic sub-mesoscale fronts (10–50 km width) extend in the ocean interior, driving large vertical velocities and associated fluxes. However, diagnosing the dynamics of these deep-reaching fronts from in situ observations remains challenging because of the lack of information on the 3-D structure of the horizontal velocity. Here, a realistic numerical simulation in the Antarctic Circumpolar Current (ACC) is used to study the dynamics of submesocale fronts in relation to velocity gradients, responsible for the formation of these fronts. Results highlight that the stirring properties of the flow at depth, which are related to the velocity gradients, can be inferred from finite-size Lyapunov exponent (FSLE) at the surface. Satellite altimetry observations of FSLE and velocity gradients are then used in combination with recent in situ observations collected by an elephant seal in the ACC to reconstruct frontal dynamics and their associated vertical velocities down to 500 m. The approach proposed here is well suited for the analysis of sub-mesoscale-resolving datasets and the design of future sub-mesoscale field campaigns.

Abstract The theoretical and practical understanding of projected changes in rainfall is desirable for planning and adapting to climate change. In this study, finite size Lyapunov exponents (FSLE) are used to study error growth rates of the system at different timescales. This is done to quantify the impact of enhanced anthropogenic greenhouse gas emissions on the predictability of fast and slow varying components of central Indian rainfall (CIR). The CIR time series for this purpose is constructed using the daily gridded high-resolution India Meteorological Department (IMD) dataset and Coupled Model Inter-comparison Project phase 5 (CMIP5) output for historical run and three representative concentration pathways (RCP2.6, RCP4.5, and RCP8.5) from the HadGEM2-ES, IPSL-CM5A-LR, CCSM4, BCC-CSM1.1, and MPI-ESM-LR models. The analyzed CIR dataset reveals a low dimensional chaotic attractor, suggesting that CIR requires a minimum of 5 and maximum of 11 variables to describe the state of the system. FSLE analysis shows a rapid decrease in the Lyapunov exponent with increasing timescales. This analysis suggests a predictability of about 2–3 weeks for fast varying components at short timescale of the CIR and about 5–9 years for slow varying components at long timescales.

Many species of Zingiber have great ornamental potential, due to durability and exotic appearance of the inflorescences. Despite its large phenotypic variability, they are scarcely exploited or not yet exploited regarding the ornamental potential. To conserve potential ornamental genotypes, and subsidize breeding program, the Agronomic Institute (IAC) maintain a Germoplasm Collection of Ornamental Zingiberales with promising accessions, including Zingiber. The aim was the morphophenological characterization of ten Zingiber accessions and the indication for landscape purposes. A large variation was observed to the evaluated characters: Clump height (CH); Inflorescence visualization (IV); Clump area (CA); Clump density (CD); Leaf stem Firmness (LSF); Number of leaf stems per clump (NLSC); Number of leaves per stem (NLS); Leaf color (LCol); Evergreen tendency (ET); Flower stem growth (FSG); Flower stem length (FSLe); Flower stem diameter (FSD); Flower stem per clump (FSC); Color sensorial perception (CSP); Flower stem weight (FSW); Inflorescence length (IL); Inflorescence diameter (ID); Bracts aspects (BAs); and Flowering season (FSe). The accessions very suitable and with the best performance to use for landscape purpose were Z. spectabile, IAC Anchieta (Z. spectabile), Z. newmanii.

The surface transport in the Northeastern Adriatic Sea has been investigated by evaluating, for the first time, the Finite-Size Lyapunov Exponent (FSLE) on the current field detected by the High Frequency (HF) coastal radar network active in the period August 2007 - August 2008. A similar analysis has been carried out on the MITgcm (Massachusetts Institute of Technology general circulation model) current field in order to have a perspective on the various results given by the application of the same FSLE evaluation procedure to different velocity fields. This work includes also the development, testing and calibration of the FSLE evaluation algorithm for the specific Adriatic area considered. The aim of this thesis is to study the surface dynamics of the Northeastern Adriatic current field associated with the typical wind regimes, namely Bora and Sirocco. The strongest and most persistent wind events, coinciding with the radar network activity, have been identified in the first instance from ALADIN (Aire Limitée Adaptation dynamique Développement InterNational) model meteorological data and then confirmed by the in situ meteo-mareographic time series in Trieste and Venice. For comparison purposes, the transport structures developing throughout the longest calm wind periods have also been investigated. In this thesis, the combination of the FSLE resulting from model and radar current fields contributes to interpret the surface transport dynamics in the studied area. In fact, it allows: i) to determine the strongest dynamical features, ii) to extend the transport information beyond the radar coverage and iii) to bring to the light the possible ambiguity of some structures originated from model currents. The FSLE analysis evidenced along the northern Adriatic margin an attractive transport structure with a filament-shaped conformation. The dynamics of this structure is driven by the water exchange between the Gulf of Trieste (GoT) and the North Adriatic Sea. The spatial location of this filament and the advective direction associated with it, vary according to the wind patterns. In fact, in calm wind periods this attractive filament is found right in front of the gulf entrance indicating the pattern of the GoT outflow (east-west direction). During Bora episodes this attractive filament is advected westward and it elongates following the northern Adriatic coast. Differently, Sirocco wind pushes this filament closer to the northeastern coastline reversing the transport direction along it (from west to east). Previous studies evidenced that the current signal in the southern part of the domain is less correlated to the wind pattern with respect to what observed in the northern area of the current field. Therefore also from the FSLE analysis no recurrent transport dynamics is observed in the southern area except for the Bora cases, when a repulsive structure originates from the Istrian coast and it looses strength while it is advected northwestward.
Il trasporto superficiale nella parte nord-orientale del Mar Adriatico è stato studiato calcolando, per la prima volta, i Finite-Size Lyapunov Exponent (FSLE) sui campi di corrente misurati dalla rete di radar ad alta frequenza attiva durante il periodo Agosto 2007 - Agosto 2008. Uno studio analogo è stato effettuato sui campi di corrente del modello MITgcm (Massachusetts Institute of Technology general circulation model), per avere una panoramica sulla varietà dei risultati prodotti dalla valutazione degli FSLE su diversi campi di velocità. Il lavoro include anche lo sviluppo, i test e la calibrazione dell'algoritmo di calcolo degli FSLE per la specifica area dell'Adriatico considerata. L'obiettivo è analizzare la dinamica superficiale del campo di corrente associata ai regimi di vento tipici dell'area nord adriatica, ovvero Bora e Scirocco. Gli episodi di vento più intensi e persistenti, verificatisi durante il periodo di attività dei radar, sono stati identificati in primo luogo dai dati meteorologici prodotti dal modello ALADIN (Aire Limitée Adaptation dynamique Développement InterNational) e successivamente sono stati confermati dalle serie temporali dei dati meteo-mareografici delle stazioni di Trieste e Venezia. A scopo comparativo, sono state analizzate anche le strutture di trasporto sviluppatesi durante i prolungati periodi di calma di vento. In questa tesi, la combinazione degli FSLE risultanti dai campi di corrente dei radar e del modello contribuisce ad una più completa interpretazione della dinamica del trasporto superficiale nell'area di interesse. I risultati ottenuti permettono: i) di determinare le strutture dinamiche più intense, ii) di estendere l'informazione sul trasporto al di fuori della copertura della rete dei radar, nonché iii) di portare alla luce eventuali ambiguità di alcune strutture originatesi dalle correnti del modello. L'analisi degli FSLE ha evidenziato una struttura di trasporto attrattiva che assume l'aspetto di un filamento lungo il margine settentrionale del Mar Adriatico. La dinamica di questa struttura è regolata dagli scambi di acqua tra il Golfo di Trieste ed il Nord Adriatico. La posizione del filamento e la direzione avvettiva associata ad esso dipendono dal regime di vento. Infatti, nei periodi di calma di vento il filamento attrattivo si trova di fronte all'entrata del golfo, ad indicare la direzione del flusso d'acqua in uscita dal golfo stesso (da est ad ovest). Durante gli episodi di Bora il filamento attrattivo è trasportato verso ovest e si allunga seguendo la costa nord adriatica. Invece, lo Scirocco spinge questo filamento verso la costa nord-orientale invertendo la direzione del trasporto associata ad esso (da ovest ad est). Precedenti lavori hanno evidenziato che le correnti nella parte meridionale del dominio sono meno correlate al segnale di vento, rispetto a quanto osservato nell'area settentrionale del campo delle correnti. Quindi anche dall'analisi degli FSLE non si osserva una dinamica di trasporto ricorrente ad eccezione dei casi di Bora, durante i quali si sviluppa una struttura repulsiva lungo la costa Istriana, la cui intensità diminuisce mentre viene trasportata verso nordovest.

MIT Motorsports is a FSAE Electric team at MIT that designs, manufactures, and tests electric formula style racecars to compete in an annual international collegiate design competition. The braking system for the MY18 vehicle developed by MIT Motorsports must enable the driver to consistently and reliably decelerate the vehicle at the maximum rate allowed by the traction limit of the tires. Crucial engineering data needed for a well-informed design, such as the coefficient of friction of the brake pads, motivated the development of a custom brake dynamometer to empirically test for the required data. The brake dynamometer became a very valuable tool eventually being used to select an appropriate brake rotor material and the most effective cooling geometry for the brake rotors. The braking system also integrates a regenerative braking system that works in parallel with the hydraulic braking system for the purpose of recovering braking energy and thus increasing vehicle efficiency. The MY18 braking system, will increase the maximum vehicle deceleration by 50% compared to MY17 and allow for up to 1kW-Hr more of energy recovery compared to MY17 without compromising reliability or consistency.

MIT Motorsports is a FSAE Electric team at MIT that designs, manufactures, and tests electric formula-style racecars to compete in an annual collegiate competition. The cooling system for the Model Year 19 vehicle must provide adequate cooling to the motors and motor controllers, meet power consumption requirements dictated by the low voltage battery, and minimize system mass. Fluid modelling and benchtop testing were used to select the most suitable pump, fan and radiator. Tube routing and mounting design also have to allow for serviceability. With the use of a cooling test bench, a flow architecture was developed, and components were modified and tested to meet specifications.

Reducing weight while maintaining structural integrity is one of the key challenges Formula SAE teams face as they try and design the suspension of the formula car. The purpose of this paper is to present experimental data on designing and optimizing a carbon fiber suspension system for formula cars. The reason carbon fiber suspensions are favored over the current steel suspensions is because of they can reduce the weight of the suspension by 50%. Pull tests on an Instron machine were performed on over 15 specimens composed of a carbon fiber tube with an aluminum insert bonded to each end. Loctite E-120HP epoxy was used and the surface preparation, bond gap, and bond length were varied to find the optimal bond strength. An average bond strength of 2,382.6 pounds per square inch was determined for specimens with surface preparation. Furthermore a bond gap of 0.0065 to 0.008 inches was found to give the strongest bond.

MIT Motorsports is the Formula SAE team at MIT that has competed in Lincoln, NE since 2013. Last year's racecar performed exceptionally in all dynamic events but was disqualified from the endurance event due to a power violation. In order to prevent a violation in the following year, a power limiter was developed to ensure that the power consumption of the vehicle is reliably under the limit. Since weight and space come at a premium when it comes to an electric racecar, the power limiting approaches were implemented through C code in the car's Vehicle Control Unit. The power limiter makes use of a feed-forward mechanical power limiter combined with an electrical power limiter that makes use of the battery's power consumption with the use of a PI closed-loop controller. Powertrain efficiency calculations were performed to best model the power consumption of the vehicle. Testing became an essential tool used to develop, debug, and validate the power limiter. This year's power limiter will reliably keep the team under the power limit while minimally affecting performance to increase our potential at the next competition.

Graph representation learning has attracted tremendous attention due to its remarkable performance in many real-world applications. However, prevailing supervised graph representation learning models for specific tasks often suffer from label sparsity issue as data labeling is always time and resource consuming. In light of this, few-shot learning on graphs (FSLG), which combines the strengths of graph representation learning and few-shot learning together, has been proposed to tackle the performance degradation in face of limited annotated data challenge. There have been many studies working on FSLG recently. In this paper, we comprehensively survey these work in the form of a series of methods and applications. Specifically, we first introduce FSLG challenges and bases, then categorize and summarize existing work of FSLG in terms of three major graph mining tasks at different granularity levels, i.e., node, edge, and graph. Finally, we share our thoughts on some future research directions of FSLG. The authors of this survey have contributed significantly to the AI literature on FSLG over the last few years.

Formula SAE (FSAE) is a design competition organized each year by the Society of Automotive Engineers (SAE). The objective of the competition is to bring the best and brightest future engineers from each participating school to present a small scale race car. Although this sounds like a relatively simple concept, the actual execution is rather challenging and rewarding for the team. For almost three years Tennessee Tech University (TTU) has had a FSAE team. The first year was a planning year, so Tennessee Tech University has participated in the competition for the last two years. Both years have been extreme learning experiences since TTU was not prepared for the level of competition brought by participating schools. However TTU FSAE team is beginning to implement modern design tools such as FEA, Virtual Manufacturing, and Rapid Prototyping to help streamline the design efforts so that one day Golden Eagle FSAE will be one of the top competing teams. In this publication, authors will report on one Golden Eagle FSAE component (the rear upright) development stages and its accomplishments.

A new manufacturing approach in which the friction stirring phenomenon is used as a vehicle for facilitating the deformation of bulk lightweight materials into manufactured components has been recently introduced under the general concept of “bulk friction stir forming”. In a preliminary work, the concept was applied to the back extrusion process, in what was referred to as “friction stir back extrusion” or FSBE; it was shown that the concept is practically valid and FSBE is capable of producing lightweight tubular specimens. Nevertheless, the FSBE process was claimed to have several merits over conventional processes, mainly: (i) unique process capabilities and energy efficiency (ii) significant grain refinement, and thus (iii) favorable mechanical properties in the formed tubes. None of these claims were adequately addressed nor quantified in earlier work. Therefore, this work presents a comprehensive study that aims to reveal the true merits of FSBE, validate the claims, and quantify its effects on the microstructure and mechanical properties of the deformed material. The outcomes of the study are presented in three major parts, each addressing one of the abovementioned claims. Force, torque and power measurements during FSBE experiments are used to address the first claim. Detailed optical microscopy and electron back scatter diffraction work is carried out in the second part to quantify the changes to the grain structure and texture of the material. Finally, the third part presents detailed mechanical characterization using digital image correlation to quantify the effects of FSBE on the performance of the produced tubes.

Para demostrar a funcionalidade e utilidade da FS3E, implementamos e disponibilizamos também cinco métodos sofisticados de seleção de características para o domínio de detecção de malwares Android.

Abstract The paper addresses the modelling, validation, and analysis of an FSAE car. The aim is to devise a method for enhancing its dynamic performance. The validation of the model is carried out in acceleration, skidpad and slalom tests, against the experimental data recorded within the 2018 FSAE competition in Varano de’ Melegari (Italy). It is shown that the skidpad and slalom performance can be significantly enhanced by varying the trim of the car, namely toe, camber, steering geometry and anti-roll bars. Implications on steering characteristics are also discussed, for investigating the effects on handling.

La caracterización de las prioridades presupuestarias en la pesca y acuicultura son importantes para optimizar la asignación y ejecución de sus recursos presupuestarios en Perú en beneficio de sus actores. Con base en la metodología del Estimado del Apoyo a la Pesca (FSE, por sus siglas en inglés), este documento caracteriza las prioridades de inversión y gasto público y analiza su consistencia con los instrumentos de gestión en el sector. Se identificó que el valor del apoyo a este sector se incrementó de PEN 235,2 millones en 2016 a PEN 376,4 millones en 2020. Además, a pesar de la volatilidad en la inversión pública, se destacó los esfuerzos orientados a investigación y desarrollo, ordenamiento pesquero e infraestructura. No obstante, todavía persisten varios problemas en el sector, a pesar de que el apoyo de Perú en relación con su PBI no es bajo en comparación a otros países que son potencias pesqueras. Ante estos hallazgos, parece ser que hay un problema respecto al uso poco eficiente de los recursos presupuestarios.
