Dual-modal imaging by combining magnetic resonance (MR) and near-infrared (NIR) fluorescence can integrate the advantages of high-resolution anatomical imaging with high sensitivity in vivo fluorescent imaging, which is expected to play a significant role in biomedical researches. Here we report a dual-modality imaging probe (NIR/MR-MSNs) fabricated by conjugating NIR fluorescent heptamethine dyes (IR-808) and MR contrast agents (Gd-DTPA) within highly aminated mesoporous silica nanoparticles (MSNs-NH2). The dual-modality imaging probes NIR/MR-MSNs possess a size of ca. 120 nm. The NIR/MR-MSNs show not only near-infrared fluorescence imaging property with an emission peak at 794 nm, but also highly MRT1relaxivity of 14.54 mM−1 s−1, which is three times more than Gd-DTPA. In vitro experiment reveals high uptake and retention abilities of the nanoprobes, while cell viability assay demonstrates excellent cytocompatibility of the dual-modality imaging probe. After intratumor injection with the NIR/MR-MSNs, MR imaging shows clear anatomical border of the enhanced tumor region while NIR fluorescence exhibits high sensitive tumor detection ability. These intriguing features suggest that this newly developed dual-modality imaging probes have great potential in biomedical imaging.

Gadolinium chelate and Quantum Dots (QDs) decorated single walled carbon nanotubes (SWNTs) nanohybrids (SWNT-Gd-QDs) were designed for dual-modal cellular imaging via magnetic resonance imaging (MRI) and optical imaging. The nanohybrids were prepared via a simple and novel layer-by-layer (LBL) assembly in combination with covalent connection strategy. The SWNT-Gd-QDs nanohybrids showed a strong near-infrared photoluminescence (PL) emission at room temperature and enhanced T1-weighted and T2-weighted MRI signals in water. Thus, the nanohybrids combine magnetic and fluorescent properties and can serve as dual-modal MRI/optical imaging contrast agent.

The design of near-infrared fluorescent (NIRF) probes that are activated by specific proteases has, for the first time, allowed enzyme activity to be imaged in vivo. In the current study, we report on a method of imaging enzyme activity using two fluorescent probes that, together, provide improved quantitation of enzymatic activity. The method employs two chemically similar probes that differ in their degradability by cathepsin B. One probe consists of the NIRF dye Cy5.5 attached to a particulate carrier, a crosslinked iron oxide nanoparticle (CLIO), through cathepsin B cleavable l-arginyl peptides. A second probe consists of Cy3.5 attached to a CLIO through proteolytically resistant d-arginyl peptides. Using mixtures of the two probes, we have shown that the ratio of Cy5.5 to Cy3.5 fluorescence can be used to determine levels of cathepsin B in the environment of nanoparticles with macrophages in suspension. After intravenous injection, tissue fluorescence from the nondegradable Cy3.5–d-arginyl probe reflected nanoparticle accumulation, while fluorescence of the Cy5.5–l-arginyl probe was dependent on both accumulation and activation by cathepsin B. Dual wavelength ratio imaging can be used for the quantitative imaging of a variety of enzymes in clinically important settings, while the magnetic properties of the probes allow their detection by MR imaging.

Dual or multimodal imaging probes have emerged as powerful tools that improve detection sensitivity and accuracy in disease diagnosis by imaging techniques. Two imaging techniques that are complementary and do not use ionizing radiation are magnetic resonance imaging (MRI) and optical fluorescence imaging (OFI). Herein, we prepared metal-free organic species based on dendrimers with magnetic and fluorescent properties as proof-of-concept of bimodal probes for potential MRI and OFI applications. We used oligo(styryl)benzene (OSB) dendrimers core that are fluorescent on their own, and TEMPO organic radicals anchored on their surfaces, as the magnetic component. In this way, we synthesized six radical dendrimers and characterized them by FT-IR, 1H NMR, UV-Vis, MALDI-TOF, SEC, EPR, fluorimetry, and in vitro MRI. Importantly, it was demonstrated that the new dendrimers present two properties: on one hand, they are paramagnetic and show the ability to generate contrast by MRI in vitro, and, on the other hand, they also show fluoresce emission. This is a remarkable result since it is one of the very few cases of macromolecules with bimodal magnetic and fluorescent properties using organic radicals as the magnetic probe.

We report a reliable and facile procedure to prepare near infrared fluorescence (NIRF)-magnetic resonance dual-modal imaging probes (DMIPs) using Cy5-labeled, Gd–Al co-doped mesoporous silica nanoparticles. High intensity NIRF and a relaxation rate (17.7 mM−1s−1) can be observed. Furthermore, the DMIPs in saline were injected into mice through the tail vein for imaging.

We have developed a novel approach to fabricate single nucleotide polymorphisms (SNPs) library on magnetic nanoparticles (MNPs) based on adaptor PCR. Each SNP locus in the library was interrogated by hybridization with a pair of allele specific dual-color fluorescence (Cy3, Cy5) probes to determine SNP. Two SNPs loci (M235T and A-6G) associated with essential hypertension in the angiotensinogen (AGT) gene were detected by this method and their fluorescent signals were quantified. The fluorescent ratios (match probe: mismatch probe signal) of homozygous genotypes were over 3.0, whereas heterozygous genotypes had ratios near to 1.0. Without any complex multiplex PCR procedure, it is a simple, efficient and reliable method for the multiplex SNPs detection using limited amount of DNA samples from individuals.

Both live and dead Salmonella typhimurium (S.T) are harmful to human health, but there are differences in pathological mechanism, dosage, and security. It is crucial to develop a rapid and simultaneous assay to distinguish and quantify live and dead S.T in foods. Herein, one dual-mode biosensor for simultaneous detection of live and dead S.T was fabricated based on two phage probes, using portable bioluminescence and fluorescent meter as detectors, respectively. Firstly, a magnetic phage capture probe (M-P1) and a phage signal tag (P2-S) labeled with SYTO 13 fluorescent dye were prepared, respectively. Both M-P1 and P2-S can specifically conjugate with S.T to form a magnetic sandwich complex. After magnetic separation, the isolated complex can emit a fluorescent signal under an excited 365 nm laser, which can reflect the total amount of S.T. Afterwards, the lysozyme was added to decompose the captured live S.T, which can release ATP and produce a bioluminescent signal corresponding to the live S.T amount. The dead S.T concentration can be deduced by the difference between total and live examples. The detection limit of 55 CFU/mL for total S.T and 9 CFU/mL for live ones was within 20 min. The assay was successfully employed in milk samples and prospectively for on-site screening of other dead and live bacteria, while changing the phages for the targets.

It is of great importance to integrate two kinds of fluorescent probes in one system to construct ratiometric sensing platform. Inspired by the efficient overlapped spectrum of glutathione-stabilized copper nanoclusters (GSH-CuNCs) and 2, 3-diaminophenazine (DAP), a novel and label-free ratiometric fluorescent sensor has been developed for Cu2+ and kojic acid (KA) detection on the basis of in situ converting o-phenylenediamine (OPD) into DAP by the catalytic oxidation ability of Cu2+. The Cu2+ catalytically generated DAP was served as an energy acceptor to inhibit the fluorescence emission of GSH-CuNCs (energy donor). As a result, the sensitivity and accuracy of the strategy were greatly improved due to the dual-emission reverse change ratiometric profile. The sensing system showed high sensitivity toward Cu2+ in a range of 0.1–50 μM and with a limit of detection of 20 nM. Furthermore, based on the specific coordination interaction between Cu2+ and KA, the ratiometric sensor displayed the range of detection of KA from 0.2 μM to 300 μM with a limit of detection of 50 nM. Thus, the ratiometric method paved a new way for the effective quantification of Cu2+ and KA in real samples, which were closely concerned with human health.

Super-resolution microscopy is an umbrella term for a family of powerful imaging techniques that examine the organization and dynamics of biomolecules at the nanoscale. However, an increasing number of super-resolution techniques along with their distinct limitations make it a nontrivial task to select the right technique to answer a specific question. Moreover, effective integration and interpretation of the multimodal, often conflicting data present another unique challenge, underscoring the need for experimental validation of super-resolved membrane protein organization. To shed light on the discrepancy between different super-resolution techniques, the impact of dopamine transporter labeling strategy and cell processing on its dynamic behavior was examined in catecholaminergic CAD cells. To this end, dopamine transporter fused to yellow fluorescent protein at the amino terminus was expressed in CAD cells and externally labeled with antagonist-conjugated quantum dot probes. The dual labeling strategy allowed direct comparison of transporter nanoscopic organization in live and fixed cells obtained via either yellow fluorescent protein tracking or quantum dot tracking in the same field of view. Our data indicated that the labeling strategy along with fixation had a significant impact on observed transporter surface dynamics and highlighted the importance of careful experimental vetting of the results obtained via super-resolution imaging.

Molecular imaging is an emerging non-invasive method to qualitatively and quantitively visualize and characterize biological processes. Among the imaging modalities, PET/SPECT and near-infrared (NIR) imaging provide synergistic properties that result in deep tissue penetration and up to cell-level resolution. Dual-modal PET/SPECT-NIR agents are commonly combined with a targeting ligand (e.g., antibody or small molecule) to engage biomolecules overexpressed in cancer, thereby enabling selective multimodal visualization of primary and metastatic tumors. The use of such agents for (i) preoperative patient selection and surgical planning and (ii) intraoperative FGS could improve surgical workflow and patient outcomes. However, the development of targeted dual-modal agents is a chemical challenge and a topic of ongoing research. In this review, we define key design considerations of targeted dual-modal imaging from a topological perspective, list targeted dual-modal probes disclosed in the last decade, review recent progress in the field of NIR fluorescent probe development, and highlight future directions in this rapidly developing field.

A small organic molecule P was synthesized and characterized as a fluorometric and colorimetric dual-modal probe for Hg2+. The sensing characteristics of the proposed probe for Hg2+ were studied in detail. A fluorescent enhancing property at 583 nm (>30 fold) accompanied with a visible colorimetric change, from colorless to pink, was observed with the addition of Hg2+ to P in an ethanol-water solution (8:2, v/v, 20 mM HEPES, pH 7.0), which would be helpful to fabricate Hg2+-selective probes with “naked-eye” and fluorescent detection. Meanwhile, cellular experimental results demonstrated its low cytotoxicity and good biocompatibility, and the application of P for imaging of Hg2+ in living cells was satisfactory.

In this paper, we demonstrate multifunctional upconversion nanoparticles with intense near-infrared emission and unique magnetic properties for dual-modal upconversion luminescent bioimaging and T2-weighted magnetic resonance imaging. High-quality BaYbF5:Tm3+ nanoparticles are synthesized via a hydrophobic method and then converted to be hydrophilic via a hydrochloric acid treatment. The as-synthesized nanoparticles are cubic phase and about 6 nm in diameter with narrow size distribution. The intense near-infrared emission makes these nanoparticles can be acted as bio-probes in upconversion luminescent bioimaging with deep tissue penetration. Besides, these nanoparticles can also be used as T2-weighted contrast agents in magnetic resonance imaging due to the high value of relaxation rate (r2 = 4.05) in 0.55 T. This finding may have further bio-applications in the future due to the high performance of these BaYbF5:Tm3+ nanoparticles in dual-modal bioimaging.

Fluorinated human serum albumin conjugates were prepared and tested as potential metal-free probes for 19F magnetic resonance imaging (MRI). Each protein molecule was modified by several fluorine-containing compounds via the N-substituted natural acylating reagent homocysteine thiolactone. Albumin conjugates retain the protein’s physical and biological properties, such as its 3D dimensional structure, aggregation ability, good solubility, proteolysis efficiency, biocompatibility, and low cytotoxicity. A dual-labeled with cyanine 7 fluorescence dye and fluorine reporter group albumin were synthesized for simultaneous fluorescence imaging and 19F MRI. The preliminary in vitro studies show the prospects of albumin carriers for multimodal imaging.

Our project investigated a new method for the cryopreservation of honey bee (Apis mellifera) sperm cells (SC). Few methods have been developed and none achieve normal sex ratios in progeny. Recently, honey bee colonies have been decimated by colony collapse disorder and infestation by varroa bee mites. A bank of preserved SC might enable the creation of a seed stock for restoration of genetic diversity through AI (Cobey 1983 Am. Bee J. 123, 389–395). We investigated two freezing rates using two diluents and their effect on post-thaw survival of the SC. The slower freezing rate was chosen from a report with the highest success to date (Harbo 1983 Ann. Entomol. Soc. Am. 76, 890–891). The rapid freezing rate was a method developed by us. We reasoned that the small volumes of ejaculate made it potentially suitable for vitrification. The SC were frozen either in 40% Harbo's DMSO diluent containing 25% DMSO, 25% egg yolk, 50% buffer (1.1% NaH2PO4�H2O (w/v) and 0.845% Na2HPO4�2H2O (w/v)), and 60% semen; or in 50% glycerol-based diluent containing 9% glycerol, 24% egg yolk, 67% buffer (5.9% Tris (w/v), 0.8% glucose (w/v), and 3.2% citric acid (w/v)), and 50% semen. Ejaculates were collected by applying bilateral pressure to the abdomens of the drones causing endophallus eversion. About 1 µL (8 � 106 SC) of ejaculate was drawn into siliconized 50-µL capillary tubes fitted to a Hamilton threaded-plunger syringe preloaded with Fluorinert (Sigma, St. Louis, MO, USA). Micro-glass cryostraws (µC) were constructed by pulling Pasteur pipettes to a 230-µm internal diameter and keeping what was the tip end of the pipette as the large end of the µC. The large end was fitted with Silastic tubing to act as a bulb for drawing and expelling fluid. Three µC per treatment were filled with 5 µL of diluted ejaculate and sealed with Critoseal. The µC were inserted into 500-µL Cassou straws (IMV Technologies, L'Aigle, France), immersed in a water bath, and cooled from 21�C to 5�C over 2 h. A Freeze Control� programmable cryochamber (CryoLogic Pty. Ltd., Mulgrave, Victoria, Australia) was used to cool samples slowly from 5�C to –40�C at 3�C min–1. At –40�C, the cryostraws were plunged into liquid N2 (LN2). Rapid freezing was done by plunging µC into a LN2 vortex, created using a magnetic stir bar. The µC were reinserted into the Cassou straws, while still under LN2, for storage in an LN2 tank. The µC were thawed by removal from the Cassou straws and immediate immersion in a 35�C H2O bath. Survival rates were evaluated using a dual fluorescent staining system (Molecular Probes, Eugene, OR, USA) and fluorescent microscopy. The largest portion of live staining cells (93.18%) were treated with DMSO diluent using the rapid freezing. The remaining treatments ranked as follows: slow freezing with DMSO (78.84%), rapid freezing with glycerol (38.9%), and slow freezing with glycerol (26%). All treatments differed significantly (P < 0.01). Other studies state that queens inseminated with greater than 50% viable SC have a good probability of producing normally throughout a season. Therefore, our technique of rapid freezing in DMSO diluent might be useful to apiculturists.

Foot-and-mouth disease virus (FMDV) is a highly contagious disease that affects cloven-hoofed animals. The traditional diagnostic methods for FMDV have several drawbacks such as cross-reactivity, low sensitivity, and low selectivity. To overcome these drawbacks, we present an optical and electrochemical dual-modal approach for the specific detection of FMDV serotypes O and A by utilizing a magnetic nanoparticle labeling technique with resorufin β-d-glucopyranoside (res-β-glc) and β-glucosidase (β-glc), without the use of typical lateral flow assay or polymerase chain reaction. FMDV serotypes O and A were reacted with pan-FMDV antibodies that recognize all seven FMDV serotypes (O, A, C, Asia 1, SAT 1, SAT 2, and SAT 3). The antigen–antibody complex was then immobilized on magnetic nanoparticles and reacted with β-glc-conjugated FMDV type O or type A antibodies. Subsequently, the addition of res-β-glc resulted in the release of fluorescent resorufin and glucose owing to catalytic hydrolysis by β-glc. The detection limit of fluorescent signals using a fluorescence spectrophotometer was estimated to be log(6.7) and log(5.9) copies/mL for FMDV type O and A, respectively, while that of electrochemical signals using a glucometer was estimated to be log(6.9) and log(6.1) copies/mL for FMDV type O and A, respectively. Compared with a commercially available lateral flow assay diagnostic kit for immunochromatographic detection of FMDV type O and A, this dual-modal detection platform offers approximately four-fold greater sensitivity. This highly sensitive and accurate dual-modal detection method can be used for effective disease diagnosis and treatment, and will find application in the early-stage diagnosis of viral diseases and next-generation diagnostic platforms.

Over the past decade, iron (Fe)-based hollow nanoplatforms (Fe-HNPs) have attracted increasing attention for cancer theranostics, due to their high safety and superior diagnostic/therapeutic features. Specifically, Fe-involved components can serve as magnetic resonance imaging (MRI) contrast agents (CAs) and Fenton-like/photothermal/magnetic hyperthermia (MTH) therapy agents, while the cavities are able to load various small molecules (e.g., fluorescent dyes, chemotherapeutic drugs, photosensitizers, etc.) to allow multifunctional all-in-one theranostics. In this review, the recent advances of Fe-HNPs for cancer imaging and treatment are summarized. Firstly, the use of Fe-HNPs in single T1-weighted MRI and T2-weighted MRI, T1-/T2-weighted dual-modal MRI as well as other dual-modal imaging modalities are presented. Secondly, diverse Fe-HNPs, including hollow iron oxide (IO) nanoparticles (NPs), hollow matrix-supported IO NPs, hollow Fe-complex NPs and hollow Prussian blue (PB) NPs are described for MRI-guided therapies. Lastly, the potential clinical obstacles and implications for future research of these hollow Fe-based nanotheranostics are discussed.

Tumor accurate imaging can effectively guide tumor resection and accurate follow-up targeted therapy. The development of imaging-stable, safe, and metabolizable contrast agents is key to accurate tumor imaging. Herein, ultra-small and metabolizable dual-mode imaging probe Au/Gd@FA NCs is rationally engineered by a simple hydrothermal method to achieve accurate FL/MRI imaging of tumors. The probes exhibit ultra-small size (2.5–3.0 nm), near-infrared fluorescence (690 nm), high quantum yield (4.4%), and a better T1 nuclear magnetic signal compared to commercial MRI contrast agents. By modifying the folic acid (FA) molecules, the uptake and targeting of the probes are effectively improved, enabling specific fluorescence imaging of breast cancer. Au/Gd@FA NCs with good biosafety were found to be excreted in the feces after imaging without affecting the normal physiological metabolism of mice. Intracellular reactive oxygen species (ROS) increased significantly after incubation of Au/Gd@FA NCs with tumor cells under 660 nm laser irradiation, indicating that Au/Gd@FA NCs can promote intracellular ROS production and effectively induce cell apoptosis. Thus, metabolizable Au/Gd@FA NCs provide a potential candidate probe for multimodal imaging and tumor diagnosis in clinical basic research. Meanwhile, Au/Gd@FA NCs mediated excessive intracellular production of ROS that could help promote tumor cell death.
