Résumé :

Objective Influenza season occurs every year in China, but its presentation was unusual in the period from December 2017 to early 2018. During this period, influenza activity was increasing across the country and was much greater than during the same period in previous years, with great harm to people’s health. Methods In this study, we isolated two human influenza virus strains—A/Hebei/F076/2018(H1N1) and B/Hebei/16275B/2018—from patients with severe influenza in Hebei, China, during the flu season in January 2018, and explored their genetic characteristics, pathogenicity, and transmissibility. Results A/Hebei/F076/2018(H1N1) belongs to the human-like H1N1 influenza virus lineage, whereas B/Hebei/16275B/2018 belongs to the Victoria lineage and is closely related to the World Health Organization reference strain B/Brisbane/60/2008. Pathogenicity tests revealed that A/Hebei/F076/2018(H1N1) replicated much more strongly in mice, with mice exhibiting 40% mortality, whereas B/Hebei/16275B/2018 was not lethal. Both viruses could be transmitted through direct contact and by the aerosol route between guinea pigs, but the H1N1 strain exhibited higher airborne transmissibility. Conclusions These results may contribute to the monitoring of influenza mutation and the prevention of an influenza outbreak.

Résumé :

The Escherichia coli YidC protein belongs to the Oxa1 family of membrane proteins that have been suggested to facilitate the insertion and assembly of membrane proteins either in cooperation with the Sec translocase or as a separate entity. Recently, we have shown that depletion of YidC causes a specific defect in the functional assembly of F1F0 ATP synthase and cytochrome o oxidase. We now demonstrate that the insertion of in vitro–synthesized F1F0 ATP synthase subunit c (F0c) into inner membrane vesicles requires YidC. Insertion is independent of the proton motive force, and proteoliposomes containing only YidC catalyze the membrane insertion of F0c in its native transmembrane topology whereupon it assembles into large oligomers. Co-reconstituted SecYEG has no significant effect on the insertion efficiency. Remarkably, signal recognition particle and its membrane-bound receptor FtsY are not required for the membrane insertion of F0c. In conclusion, a novel membrane protein insertion pathway in E. coli is described in which YidC plays an exclusive role.

Résumé :

Las plantas asintomáticas son reservorios de patógenos, ya que pueden permanecer infectadas la mayor parte de su ciclo de desarrollo, convirtiéndose en fuente de contaminación para el resto del cultivo. El objetivo de este estudio fue evaluar un método de detección y discriminación de dos cepas de Fusarium oxysporum en tomate usando espectroscopia. La enfermedad en las plantas de tomate inoculadas con la cepa aislada de gulupa (F05) fue mayor a la observada en la cepa aislada de tomate (F07), presentando valores de 60,0% (11 días) y 81,8% (22 días); la cepa F07 presentó incidencias de 30,0 y 64,3% en ambas mediciones. La planta infectada con la cepa F05 fue mejor discriminada en el periodo de incubación de la enfermedad en ambos periodos de tiempo en los Análisis de Componentes Principales (PCA) y Análisis Discriminantes Lineales (LDA) realizados con los controles en comparación con la cepa F07. Estos resultados sugieren que la espectroscopia de reflectancia VIS es un método sensible y confiable que puede ser adecuado para el diagnóstico temprano de enfermedades en plantas.

Latar Belakang. Laserasi perineum adalah salah satu akibat dari proses persalinan denganberbagai penyebab seperti kelahiran kepala secara tiba-tiba dan cepat, berat janin yang berlebihan, danjaringan kulit ibu yang mudah robek. Komplikasi dari laserasi perineum adalah penyembuhan lukayang tertunda, infeksi, nyeri, takut bergerak, dan terhambatnya pengeluaran lochea. Hambatan aliranlochea pada infeksi postpartum dapat menyebabkan kematian ibu. Binahong (Anredera Cordifolia(ten) Steenis) adalah zat alami yang membantu penyembuhan luka. Kandungan asam askorbat dalambinahong penting untuk mengaktifkan enzim hidroksilasi prolil yang mendukung tahap hidroksilasidalam pembentukan kolagen, yang mempercepat proses penyembuhan luka. Tujuan. Penelitian inibertujuan untuk mengetahui sifat fisik, homogenitas, uji pH, dan organoleptik ekstrak daun binahongsetelah diformulasikan ke dalam sabun cair dengan variasi konsentrasi 100%. Metode. Penelitianini menggunakan metode eksperimental, yang dilakukan di laboratorium farmasi. Ekstraksi daunbinahong dilakukan dengan merebus daun selama 30 menit dengan suhu 90°C menggunakanpelarut air. Sabun cair dibuat dengan dasar sabun PEG, carbopol (f0a), esaflor (f0b), HPMC (f0c),asam sitrat dan sodium hidrogen fosfat. Hasil f0c Formula dengan HPMC memiliki evaluasi terbaikdibandingkan dengan f0a dan f0b yang terlihat memiliki pH netral, organoleptik dan sifat fisikterbaik di antara formula lainnya. Kesimpulan. Sabun cair Binahong (Formula f0c) layak sebagaibahan sabun cair anti-septik dari hasil evaluasi stabilitas formula dan persyaratannya. Saran. Hasilpenelitian ini dapat digunakan sebagai referensi untuk tahap selanjutnya, yaitu optimalisasi rumusformulasi sabun cair sebagai sabun antiseptic

Abstract Background Clostridioides difficile is an anaerobic spore-forming, toxin-producing Gram-positive bacillus listed by the CDC as an “urgent threat” pathogen. Epidemiologic studies using whole-genome sequencing (WGS) have found that genetically distinct lineages infections occur in hospitalized patients, in addition to the fact that C. difficile spores persist on hospital surfaces after disinfection. The purposes of this study were to isolate and characterize C. difficile from the environment of a large Texas hospital. Methods We collected 330 swab samples of hospital environmental surfaces using sterile cotton gauze. The samples were then anaerobically enriched in brain heart infusion broth for 48–72 hours and plated onto cycloserine–cefoxitin fructose agar (CCFA). Suspected colonies were then genetically characterized using PCR (for tcdA, tcdB, cdtA, cdtB and tpi genes) and genotyped using fluorescent PCR ribotyping techniques. Results A total of 90/330 (27.3%) environmental samples were culture positive for C. difficile, of which 75/90 (82.1%) tested were toxigenic C. difficile by the presence of tcdA, tcdB, cdtA or cdtB. A total of 16 distinct ribotypes were identified from 41 C. difficile isolates tested using a fluorescent-ribotyping method. The predominant ribotypes isolated were F078–126 (8/41), F002 (5/41), F106 (4/41), F255 (4/41), and F014-020 (3/41). Conclusion We found a diversity of C. difficile strain types in various hospital high-touch surface environment in addition to ribotype F027 and F078, suggesting the hospital environment a reservoir and significant source C. difficile infections. Résumé :

This research explores the link between pupils’ perceived control and their level of attendance in school. Whilst there is research into the link between perceived control and disaffection in pupils it has not been possible to identify any research which links school non-attendance to perceived control. Research into pupils’ reasons for non-attendance identified a range of different factors, many of which the author felt could be attributed to perceived control. Forty-one participants took part in the study. Participants were male and female year nine pupils at two secondary schools in the north west of England. Participants were grouped according to their levels of attendance as high (98%+) attenders, mid-range attenders (90-94%) and low (below 80%) levels of attendance. Participants’ levels of perceived control were measured using the Multi-dimensional Measure of Children’s Perceptions of Control (MMCPC) (Connell 1985). This research also explored the pupils’ experiences of school using appreciative inquiry. Responses on the MMCPC were analysed using a one way ANOVA and descriptive statistics. No significant differences were found between scores for each of the attendance groups and the reasons for this are discussed. Thematic analysis of focus groups with an appreciative inquiry structure identified key themes raised by pupils in regard to positive experiences in school. The findings for both parts of the research are discussed and suggestions for the implications for future research and the practice of teachers and educational psychologists supporting attendance in school are made.

In this thesis, the 18F-labelling of new "chemotypes" will be described with the aim of application in pharmaceutically interesting targets. In Chapter 1, a general introduction to the effect of fluorine substituents on molecular properties and reactivity is provided. This includes the application of organofluorine in both medicinal chemistry and positron emission tomography. Chapter 2 describes a novel silver-mediated 18F-labelling of Ar-SCF3, Ar OCF3 and Ar-OCF2H, including [18F]Riluzole, the 18F labelled version of a drug for treatment of amyotrophic lateral sclerosis (ALS). This work demonstrates that AgOTf can induce halogen exchange nucleophilic 18F-fluorination under mild reaction conditions with a wide range of substrates. In addition, the 18F-labelled Ar-SCF3 substrate is further transformed into the [18F]Umemoto reagent in a single step. The Umemoto reagent has been extensively used for electrophilic trifluoromethylation of various functional groups. Therefore the labelling of this reagent could potentially expand the radiochemical space available for PET applications. To augment the utility of this reagent, we developed a late-stage stereoselective trifluoromethylation-thiocyclisation of alkenes using electrophilic trifluoromethylating reagents such as Umemoto reagent or Togni reagent, which will be discussed in Chapter 3. In this process, thiourea acts as both S-nucleophile and CF3 radical initiator; therefore no metal or photoredox catalyst is required. The reaction affords novel trifluoromethylated 2-amino-thiazolines and 2-amino-thiazines, important scaffolds in the development of aspartate beta-secretase enzyme (BACE-1) inhibitors, a therapeutic target for Alzheimer's disease. Chapter 4 investigates the 18F-labelling of a,a-difluoro-a-aryloxyacetic acid, a class of substrate that can serve as a versatile intermediate which can undergo various decarboxylative functionalisation reactions to afford a wide range of novel 18F labelled a fluorinated aryl ethers. Finally, Chapter 5 gives full experimental procedures and characterisation data for all compounds.

Many economic interactions are characterized by imperfect information and imperfect enforcement. This thesis analyzes the use and the effectiveness of incentives, reputation and learning and how economic agents use them to mitigate the problems caused by both. Chapter 1 presents experimental evidence on the use of price incentives and reputational incentives from gift-exchange game lab experiments in Ghana and the United Kingdom. While in the United Kingdom, like in other experiments, participants assigned the role of employers reward high effort by increasing wages and punish low effort by decreasing wages, it appears that Ghanaian participants refrain from doing so. This results in lower average effort in Ghana compared to the United Kingdom. Introducing competition for workers or a reputation mechanism does not improve effort. Chapter 2 further examines low worker productivity in Ghana by empirically testing two other incentive mechanisms: the employer giving feedback and the worker making a pledge in advance. While these incentives on average do not affect effort, they do increase conditional reciprocity of the worker's behalf. Workers receiving a high wage are more likely to exert high effort when feedback and promises are possible. Chapter 3 presents a partial-equilibrium model showing how business networks can lead to higher quality firms overall. The presence of such networks leads to a filtering effect, which means that fewer low-quality firms produce, but also to a lock-out effect, implying that networks redistribute demand to their own members and make production outside the network less viable. Chapter 4 presents a model and experimental evidence on learning and strategic experimentation in the context of a repeated ultimatum game. The model predicts that Proposers make low offers in early periods, hoping that learning will benefit them in later periods. However, when Responders also have strategic considerations, the Responders will strategically reject low offers, which will optimally lead to higher offers. A series of online experiments finds evidence that Responders reject low offers, but cannot disentangle this from distributional preferences and reciprocity concerns.

Population displacement has long been a controversial companion of development. The central tension has been between the position challenging the kind of development that removes people from their homes, livelihoods and communities, and a managerial position that the impoverishment risks of displacement can be mitigated through an effective intervention. Whereas recent research has been devoted to unpacking a rather unsuccessful performance of involuntary resettlement as a mitigation measure, this study aims to question the assumption of mitigation itself by expanding the concept of development impacts beyond the realm of displacement. Through an empirical study of a railway project in Metro Manila, the Philippines, I examine how urban residents are affected by a large-scale demolition and displacement that took place in their locality. Semi-structured interviews were conducted along the railway tracks after the land was cleared of informal settlements since the study placed particular focus on residents who were not physically displaced. They are identified in my research as non-displaced people. Few studies have addressed the possibility that other people might have been adversely affected in situ and this is particularly so in urban areas. Empirical findings reveal that the physical environment and socio-economic relationships in the locality were significantly transformed through the clearance; impacting the tenure status, livelihoods and social milieu of non-displaced people. Tenure security was important for avoiding displacement but was not a definitive factor as a number of people are still informal settlers who continue to be faced with other eviction threats. For the non-displaced, the physical change of the locality became relevant when their productive capital, notably, a second house or business space, was affected. The loss or erosion of physical capital had a secondary impact on livelihoods, which was compounded by the rupture in the local livelihood network following a mass population outflow. Whereas the income of locally-based businesses decreased substantially, livelihoods that operate beyond the locality remain relatively resilient. Differentiated experiences of a local change are also reflected in a range of evaluations that describe local social ambiance before and after the event. Diverse ways in which non-displaced people were affected underline that the current conceptualisation of impacts is limited to one dimension of displacement. This raises the need to adopt a more holistic and disaggregated approach to understanding the complexities of development impacts. A discussion on whether and how they can be mitigated would benefit further from such a comprehensive study.

This thesis articulates the resonances between J.M. Coetzee's lifelong engagement with mathematics and his practice as a novelist, critic, and poet. Though the critical discourse surrounding Coetzee's literary work continues to flourish, and though the basic details of his background in mathematics are now widely acknowledged, his inheritance from that background has not yet been the subject of a comprehensive and mathematically- literate account. In providing such an account, I propose that these two strands of his intellectual trajectory not only developed in parallel, but together engendered several of the characteristic qualities of his finest work. The structure of the thesis is essentially thematic, but is also broadly chronological. Chapter 1 focuses on Coetzee's poetry, charting the increasing involvement of mathematical concepts and methods in his practice and poetics between 1958 and 1979. Chapter 2 situates his master's thesis alongside archival materials from the early stages of his academic career, and thus traces the development of his philosophical interest in the migration of quantificatory metaphors into other conceptual domains. Concentrating on his doctoral thesis and a series of contemporaneous reviews, essays, and lecture notes, Chapter 3 details the calculated ambivalence with which he therein articulates, adopts, and challenges various statistical methods designed to disclose objective truth. Chapter 4 explores the thematisation of several mathematical concepts in Dusklands and In the Heart of the Country. Chapter Five considers Waiting for the Barbarians and Foe in the context provided by Coetzee's interest in the attempts of Isaac Newton to bridge the gap between natural language and the supposedly transparent language of mathematics. Finally, Chapter 6 locates in Elizabeth Costello and Diary of a Bad Year a cognitive approach to the use of mathematical concepts in ethics, politics, and aesthetics, and, by analogy, a central aspect of the challenge Coetzee's late fiction poses to the contemporary literary landscape.

Résumé :

Ion-binding organocatalysis is an emerging field that has the potential to control the stereochemical outcome of any transformation that goes via charged intermediates. The aim of this project was to explore how this concept could be applied to an asymmetric allylation reaction. Chapter 2 of this thesis discusses anion-binding catalysis and investigates a chiral cooperative thiourea catalyst that could bind to fluoride to control allylation using an allylsilane. Optimization using a non-chiral thiourea (Schreiner’s catalyst) demonstrated that the reaction proceeded in high yield with TBAT onto an N-benzoylhydrazone. A chiral cooperative thiourea catalyst library was then synthesized but unfortunately, although the allylation using these catalysts proceeded in excellent yield, the product was isolated as the racemate (Scheme 1). Scheme 1: Anion-binding catalysis gave allylated products in high yields but gave no stereocontrol. Chapter 3 examines a chiral quaternary ammonium fluoride as an example of chiral cation-directed catalysis. We hypothesized that an allylsilane activated by fluoride would generate an allyl anion species that would associate with the chiral quaternary ammonium cation through electrostatic interactions. Extensive optimization found that the allylation reaction proceeded in good yield in chloroform at reflux with N-benzoylhydrazones. Different fluoride catalysts were prepared using an ion-exchange resin, and cinchonidine-derived catalysts performed the best. This methodology was extended to a phase-transfer catalyzed process, where solid cesium fluoride exchanged with chloride in situ, removing the need to synthesize and isolate ammonium fluoride catalysts (Scheme 2). Scheme 2: Cation-directed asymmetric allylation. In Chapter 4 cation-directed asymmetric catalysis was extended to an intramolecular allylation reaction. Substrate synthesis was attempted by cross metathesis but the reaction was capricious and yields were low. Intramolecular allylation with these materials gave promising results (Scheme 3) but a lack of material prevented optimization. Scheme 3: Intramolecular allylation results.

This thesis develops efficient optimization methods for Model Predictive Control (MPC) to enable its application to constrained systems with fast and uncertain dynamics. The key contribution is an active set method which exploits the parametric nature of the sequential optimization problem and is obtained from a dynamic programming formulation of the MPC problem. This method is first applied to the nominal linear MPC problem and is successively extended to linear systems with additive uncertainty and input constraints or state/input constraints. The thesis discusses both offline (projection-based) and online (active set) methods for the solution of controllability problems for linear systems with additive uncertainty. The active set method uses first-order necessary conditions for optimality to construct parametric programming regions for a particular given active set locally along a line of search in the space of feasible initial conditions. Along this line of search the homotopy of optimal solutions is exploited: a known solution at some given plant state is continuously deformed into the solution at the actual measured current plant state by performing the required active set changes whenever a boundary of a parametric programming region is crossed during the line search operation. The sequence of solutions for the finite horizon optimal control problem is therefore obtained locally for the given plant state. This method overcomes the main limitation of parametric programming methods that have been applied in the MPC context which usually require the offline precomputation of all possible regions. In contrast to this the proposed approach is an online method with very low computational demands which efficiently exploits the parametric nature of the solution and returns exact local DP solutions. The final chapter of this thesis discusses an application of robust tube-based MPC to the nonlinear MPC problem based on successive linearization.

Vascular endothelial cells express muscarinic acetylcholine receptors, which lead to NO- and endothelium derived hyperpolarization responses that constitute major pathways for vasodilation. As evidence suggests that perivascular nerves may not represent a viable source of innervating ACh, we investigated whether suggestions of a 'non-neuronal cholinergic system' within the vasculature might act as a source of ACh that is able to influence the endothelium. Rat mesenteric arteries were cut open and held as an en-face preparation with the endothelial cell (EC) layer exposed to measure EC [Ca2+]i. Furthermore we present the first example of EC tubes isolated from rat mesenteric arteries, used to measure EC [Ca2+]i. Immunohistochemistry was performed on intact arteries to demonstrate localization of important cholinergic proteins. Direct application of rat whole blood induced increased Ca2+ activity in ECs that did not appear to rely on shear stress or other mechanical interaction of the blood with the endothelial cells. This response, shown to rely on IP3 mediated Ca2+ release, was attenuated by the muscarinic antagonist atropine; it was concluded that blood contains or induces the release of non-neuronal ACh. The localization of the ACh synthesizing protein, choline acetyltransferase by immunohistochemistry provides evidence for a source of ACh originating within the endothelium. In accordance with this, the blocker of high affinity choline uptake, hemicholinium-3, had a significant effect in reversing choline potentiated responses in ECs. Taken together, we proposed that ACh is released potentially via organic cation transporter proteins located in the endothelium, upon stimulation by blood in response to an as yet undetermined signal. The ability to influence EC Ca2+ highlights a role for blood in the local control of these cells and the data presented suggest ACh produced locally may provide physiological relevance by exerting influence on endothelial M3 muscarinic acetylcholine receptors.

Résumé :

Traditionally, creative activities have been used in occupational therapy practice as an intervention with people who have a mental illness. Recent research has explored engagement in creative activities with people living with disabilities, mental illness, retired people and people with cognitive impairment. The findings centred on positive emotions, increase in self-confidence and respite from worrying thoughts. Anecdotal evidence from local occupational therapy practitioners and arts organisations suggested that people in remission of mental illness seemed to use creative activities in their daily lives as a way to keep well, but deeper understanding was missing about this. The first aim of this study was to gain a deeper understanding of the phenomenon of voluntary participation in creative activities by people in remission of mental illness. The second aim was to explore and provide evidence of any relationship between well-being and creative activities as it was perceived by the participants. The research utilised a hermeneutic phenomenological approach. Qualitative data were collected through interviews with participants, transcribed verbatim and analysed within the hermeneutic tradition. Ten participants volunteered and were recruited by mental health NHS staff in central England. All participants had a diagnosis of a mental illness but were in remission from between six months and seven years at the time of the interviews. The data were analysed via my own interpretation of Cohen, Kahn and Steeves’ (2000) approach to hermeneutic phenomenological data analysis. The findings revealed that for all participants, engagement in their chosen creative activity evoked enjoyment and was of particular personal significance and meaning. Their subjective experiences highlighted the possible therapeutic potential of creative occupation, in particular, temporary mental relief from self-referential thoughts through deep immersion into the creative process. Deep engagement in creative occupation encompassed different types of optimal experience and might have a soothing effect on the Default Mode Network. The deliberate engagement in creative activities supports Wilcock’s theory of the use of occupation for self-restoration and keeping healthy. Additionally, the real contact with people and places facilitated a more active lifestyle which impacted also positively on the participants’ sense of well-being. The findings of the study are discussed from an occupational science perspective to extend our understanding of the solitary and group nature and effects of participation in creative activities.

Résumé :

Leaks in fuel storage tanks and distribution piping systems have been identified as a mission-critical problem by the Department of Defense and the U.S. Army. Fuel system leaks are often hard to locate and virtually inaccessible for efficient repair because the piping is often installed under a concrete pad or tarmac. Leak repair could cost up to $2,000, and the cost of cleanup and re-mediation for fuel spills can exceed $50,000. In this project an acoustic remote sensing system was installed to monitor an Army heliport refueling system to determine whether it could detect and accurately locate fuel leaks using computer software technolo-gies to distinguish acoustic leakage signatures from normal fuel system operational noise. Demonstration and validation efforts were disadvantaged by the fact that no fuel leaks occurred in the monitored system for the duration of the project. However, the monitoring system did identify several unusual acoustic events within the fueling system and interpret them as indications of intermittent malfunctions of a check valve and a fuel pump. The 30-year ROI is about 6.42. Further work is required before the technology can be fully implemented: its ability to detect fluid leaks must be proven, and the system specifications must be certified through an EPA third party.
