Résumé :

One of the syllable structure changes that occur in rapid speech because of sounds influencing each other is elision. This paper provides an account of elision in Esahie, also known as Sehwi, a Kwa language spoken in the Western North region of Ghana. The paper discusses the processes involved in elision, and the context within which elision occurs in the language. The paper shows that sound segments, syllables and tones are affected by the elision process. It demonstrates that elision, though purely a phonological process, is influenced by morphological factors such as vowel juxtapositioning during compounding, and at word boundary. The evidence in this paper show that there is an interface between phonology and morphology when accounting for elision in Esahie. Data for this study were gathered from primary sources using ethnographic and stimuli methods.

Abstract The paper provides a comprehensive account of the derivation of action nominals in Esahie, a Ghanaian language of the Tano subgroup (Kwa, Niger-Congo) which has been undocumented thus far, especially as far as morphosyntactic phenomena are concerned. The aim of the research is threefold: to contribute to language documentation, to provide a systematic description and analysis of the morphosyntactic properties of Esahie action nominals, and to offer a typological assessment of these constructions. We argue that action nominalization in Esahie primarily involves a composite strategy: a morpho-syntactic operation, invariably involving affixation, and a concomitant prosodic operation in the form of a change in tonal melody. As far as the derivation of action nominals is concerned, it appears that in Esahie, tone raising is not simply a phonologically-conditioned prosodic effect, but plays a morphemic role. Further, depending on the arity of the base verb, nominalization may or may not be coupled with incorporation of the internal argument, which derives a form of synthetic compounding, as in the English truck-driving type. Abstract In the typology of West African languages, tone has been noted to play crucial grammatical and lexical roles, but its function in word formation has been less systematically explored and remains to be fully understood. Against this backdrop, the present study seeks to examine the form and function of tonal morphology in the formation of action nominals in four Kwa languages spoken in Ghana, namely Akan, Ga, Lεtε, and Esahie, a relatively unexplored language of the Central Tano subgroup. Relying on data from both secondary and primary sources, we argue that tone raising is an important component of Kwa action nominalization, as it is found across different languages and derivational strategies. Specifically, while across the Kwa languages considered, tone raising tends to be an epiphenomenon of phonological conditioning, sometimes tone is the sole component of the nominalization operation or, as in Esahie, it concurs with the affix to the derivation, hence playing a morphological function.

La teleradiologia può contribuire ad evitare ritardi nella trasmissione dei dati acquisiti con esami diagnostici computerizzati e a rendere più tempestiva ed efficace la terapia. Inoltre con la teleradiologia è possibile ottimizzare l'interscambio di informazioni fra il diagnosta e il curante, con l'obiettivo finale di rendere sempre migliore l'assistenza al malato ed al tempo stesso fornire migliori opportunità di tipo didattico. Dal Novembre 1986 è operativo presso l'Università degli Studi di Firenze un sistema per la trasmissione di immagini di risonanza magnetica tra la Sezione di RM del Dipartimento di Fisiopatologia Clinica e la Clinica Neurochirurgica. Nel periodo novembre 1986 - novembre 1991, 97 esami RM dell'encefalo o della colonna sono stati trasmessi in tempo reale dalla RM alia Clinica Neurochirurgica. L'esperienza nell'uso di tale collegamento ha dimostrato che esso ha avuto un rilevante impatto sulla gestione del paziente neurochirurgico. Infatti in caso di esame negativo, è stato possibile disporre immediatamente la dimissione del paziente, evitando l'esecuzione di altri esami. Nel caso di esito positivo, è stato possibile sfruttare immediatamente i risultati dell'esame per accelerare il successivo percorso diagnostico o chirurgico del paziente. Un importante vantaggio del collegamento è stato inoltre di tipo didattico: infatti gli esami che venivano effettuati presso la Radiologia universitaria venivano generalmente seguiti «pacchetto» dai medici specializzandi in neurochirurgia, i quali avevano in tal modo la possibilità di studiare, ancor prima che in sala operatoria, il quadro anatomico relativo ai casi in esame. Essi avevano inoltre la possibilità di acquisire esperienza diretta della RM ed al tempo stesso delle recenti tecnologie dell'informatica e delle comunicazioni.

L'ipnosi in campo medico puň avere molteplici applicazioni, molte delle quali possono migliorare la qualitŕ degli esami diagnostici. L'endoscopia puň rappresentare un campo di intervento dell'ipnosi, poiché si tratta di un esame invasivo, che necessita di una certa preparazione, di un supporto psicologico, e della attiva collaborazione del paziente. Verrŕ qui descritta brevemente la tecnica e l'impatto psicologico che questi esami hanno sul paziente. Segue poi una breve descrizione dell'uso dell'ipnosi in procedure ed interventi chirurgici. Rivisitando la letteratura attualmente presente, verrŕ esaminato come possa venire applicata l'ipnosi per diminuire la paura, il dolore ed altre fondamentali difficoltŕ da parte del paziente. In conclusione verranno descritte le tecniche ipnotiche che possono venire impiegate.

L'occlusione spontanea dell'arteria carotide comune è rara; lo sviluppo della circolazione collaterale recluta vasi intra- od extra-cranici. A livello extra-cranico la rivascolarizzazione avviene sfruttando i rami dell'arteria carotide esterna (aa. occipitale e tiroidea superiore) percorsi in via retrograda dal flusso ematico proveniente dai principali tronchi dell'a. succlavia: aa. vertebrale, cervicale ascendente e profonda, tiroidea inferiore. L'Angio-RM, pur dotata di minor risoluzione rispetto all'angiografia digitale, consente una buona visualizzazione del tipo e dell'entità dei circoli collaterali, talora invece non ben rilevabili – se non adeguatamente ricercati – con angiografia convenzionale. La RM, accoppiata all'Angio-RM, è esame fondamentale nel paziente affetto da insufficienza cerebro-vascolare ischemica; essa si colloca tra l'ecotomografia/Doppler e l'angiografia digitale; quest'ultima è da considerarsi esame pre-operatorio nei pazienti giudicati possibilmente chirurgici alla luce degli esami incruenti prima espletati; avvalendosi dei reperti Angio-RM, lo studio angiografico può essere eseguito in maniera mirata, più rapidamente e con maggior efficacia diagnostica.

Straipsnyje bandoma išryškinti L. Karsavino ontologijos ir kai kurių visuotinybės istorijos filosofijos fundamentalių kategorijų sąsajas. L. Karsavino filosofijos išeities taškas yra visuotinybės idėja, ją galima nagrinėti kaip svarbiausią gnoseologijos, antropologijos arba etikos principą, tačiau ypatingą reikšmę ji įgyja ontologijoje, kuri savo ruožtu sąlygojo ir istorijos filosofijos ypatybes. Visuotinybės idėja išreiškia visos esaties (neorganinio ir organinio pasaulio, visuomenės) principinę vienybę. Šios vienybės pagrindas ir centras yra Dievas, kurio esatis yra absoliuti būtis. L. Karsavinas skyrė istorijos teoriją ir istorijos filosofiją (siaurąja prasme): pirmoji nagrinėja istorinio proceso ir jo pažinimo esmę bei savybes, o istorijos filosofija – tuos pačius klausimus, tik susietus su absoliutu. Žmonijos istorija yra potencialybės (suprantamos kaip visuotinybė) virtimas santykine aktualia dabartimi. Istorijos subjektas yra visalaikis ir visaerdvis, nes erdvė ir laikas yra pačios būties savybės. L. Karsavino istorijos metafizikos mistinė tendencija išreiškia sistemos religinį orientuotumą, vidinį ryšį su stačiatikybės ortodoksija.

Neste trabalho, inicialmente, apresentamos a Dualidade de Esakia. Enfraquecendo os mor smos de reticulados e considerando os mor smo de Esakia como sendo Mor smos Parciais de Esakia, obtemos a Dualidade Generalizada de Esakia. Com esses resultados, estabelecemos uma dualidade entre as categorias de L ogicas Abstratas Distributivas e espa cos Priestley, e uma representa c~ao de L ogicas Abstratas Intuicionistas em Espa cos de Esakia. Por m, aplicamos os resultados na compara c~ao da Condi c~ao de Dom nios Fechados (CDF) com a Condi c~oes de Dom nios Fechados de Zakharyaschev (CDFZ).
In this work, initially, we present the Esakia duality. Weakening the morphisms of lattices and considering the Esakia morphism as Partial Esakia morphism, we obtain the Generalized Esakia Duality. With these results, we establish a duality between the categories of Distributive Abstract Logic and Priestley Spaces, and a representation of intuitionistic Abstract Logic and Esakia Spaces. Finally, we apply the results of the comparison closed domain condition (CDC) with the Zakharyaschev's closed domain condition (ZCDC).

L'étude du livre d'Esaïe s'est focalisée pendant plus d'un siècle sur la question de savoir s'il avait un, deux ou trois auteurs ou rédacteurs. Puis l'on a pris conscience de l'intérêt de l'étudier dans son unité, car son organisation est porteuse de sens. Cela a mis en relief l'importance de la métrique et des strophes et une certaine cohérence de la pensée de l'œuvre. Placé entre deux blocs de chapitres (40-55 et 56-66, deux parties majeures de l'œuvre), et au milieu des débats qu'ils suscitent, Esaïe 55 interpelle par cette position et par son message. C'est un appel à revenir à Yahvé, Dieu qui se veut proche, tout en tenant compte de la distance séparant la terre des cieux. En fait, l'approche du prophète atteste les états d'âme de la communauté en exil et son propre désir d'accompagner et de sauver son peuple. La distance exprime une différence salutaire, c'est une garantie : Dieu peut pardonner et sauver car il est profondément différent et infiniment plus grand. Le salut et la vie de l'homme sont liés à l'acceptation de cette différence. A cause du pêché, la distance entre les cieux de la Terre devient l'expression d'une muraille entre Dieu et l'homme. L'intervention divine dans l'histoire du peuple d'Israël et des hommes est alors mal comprise, voire imperméable à la pensée humaine. Cela explique l'absence apparente de la présence divine et de la libération tant attendue, thème majeur d'Es 40-55. Le retour de l'exil n'est alors plus un simple retour au pays mais à Yahvé. La transformation du désert montre cette volonté créatrice, restauratrice de Yahvé envers l'humanité et la nature et dépasse le désir de sauver ou libérer Israël
For over a century then study of Isaiah was focused on whether it was the work of one, two or three authors. Then some researches began to realise the advantage of studying the book as a unit, for its structure is in itself meaningful. This highlighted the importance of its metrics, stanzas and a certain conceptual coherence. Located as it is between two blocks of chapters (40-55 and 56-66, two major sections of the work), and in the midst of the debates that provoke, Isaiah 55 raises questions via both its message and its position. It is a call to return to Yahwe, the God who wants to be close to man while taking into account the gap between heaven and hearth. Indeed, the prophet's approach attests to the feelings of the community in exile and his own desire to accompany and save his people. Distance reveals a healthy difference, a guarantee: God can pardon and save because he is profoundly different and infinitely greater. Man's life and salvation are linked to the acceptance of this difference. Because of sin, the distance between heaven and earth becomes the expression of the wall between man and God. Divine intervention in the history of the people of Israel and of mankind, is then misunderstood by, and even incomprehensible to, the human mind. This explains the apparent absence of the divine presence and of the divine presence and of the eagerly awaited liberation-a major theme of Isaiah 40-55. The return from exile is no longer a simple return to the homeland, but to Yahwe also. The transformation of the dersert shows this creative, restorative will of Yahwe towards humanity and nature and goes beyond the desire to save or liberate Israel

La tesi di dottorato in oggetto tratta dell’esame incrociato. In particolare nel primo capitolo ho affrontato il tema delle attività preliminari all’esame incrociato, nel secondo la sua disciplina e nel terzo la sua tecnica. Tale tema di ricerca è stato affrontato facendo costante riferimento alla prassi dei tribunali, alla giurisprudenza e dottrina di riferimento evidenziandone i profili problematici. Questo istituto può essere definito come quell’insieme di regole con le quali le parti pongono direttamente le domande alla persona esaminata. Le regole sono state elaborate da una prassi secolare che si è formata negli ordinamenti angloamericani. L’esame incrociato è stato definito nei paesi anglosassoni il miglior “strumento” per conoscere la verità. Esso si basa sul principio secondo cui la verità si conosce tanto meglio, quanto, più spazio è lasciato alla dialettica tra le parti in conflitto. Rispetto a tale modello, il codice italiano si caratterizza per il fatto di riprodurre soltanto alcune di quelle regole e per la scelta di attribuire al giudice un controllo penetrante sullo svolgimento dell'esame. In particolare, il presidente dell'organo giudicante ha la funzione di “assicurare la pertinenza delle domande, la genuinità delle risposte, la lealtà dell'esame e la correttezza delle contestazioni ( ... )” (art. 499, comma 6). L’esame incrociato si articola nei tre momenti fondamentali dell'esame diretto, del controesame e del riesame (art. 498): i soggetti che pongono le domande sono il pubblico ministero ed i difensori delle parti private. L’esame diretto è condotto dalla parte che ha chiesto di interrogare il testimone (o altro soggetto che rende dichiarazioni). L'esame diretto tende ad ottenere la rappresentazione dei fatti conosciuti dal testimone; tali fatti dovrebbero essere utili a dimostrare la tesi di colui che lo ha citato; il suo scopo è quello di dimostrare che il teste è attendibile e credibile. Per tale motivo sono vietate le “domande suggerimento” (art. 499, comma 3) Il controesame è eventuale, nel senso che le parti, che non hanno chiesto l’ammissione di quel teste, possono a loro volta porre domande. Il controesame è condotto dalla parte che ha un interesse contrario a quella che ha chiesto l'esame del testimone (o altro dichiarante). Il controesame può avvenire sui fatti ovvero sulla credibilità del testimone, o ancora su entrambi gli oggetti. Nel controesame sono ammesse le domande-suggerimento.. Il riesame è doppiamente eventuale perché si svolge soltanto se vi è stato il controesame; nel riesame la parte che ha condotto l’esame diretto può “proporre nuove domande”. La funzione del riesame è quella di consentire, a chi ha introdotto la prova, il “recupero” della sequenza dei fatti, dopo che il controesame ha cercato di mettere in dubbio la loro esistenza; oppure consente di esporre la ragione di contraddizioni nelle quali il testimone è caduto; pertanto tende a corroborare la validità della dichiarazione inizialmente resa. In questa tesi ho affrontato molti profili problematici. Particolare attenzione è stata dedicata al problema delle domande nocive, di quelle suggestive di quelle trabocchetto e del teste c.d. ostile. Infine sono state evidenziate le regole che disciplinano l’esame incrociato con particolare riferimento alla loro interpretazione giurisprudenziale e alle sanzioni eventualmente previste in caso di trasgressione.
This doctoral thesis deals with cross examination. In particular I have dealt with the topic of the activities preliminary to cross examination in the first chapter, its discipline in the second and its technique in the third. The research topic has been dealt with making constant reference to the court’s praxis and to the jurisprudence and doctrine of reference, and therefore evidencing some of the problematic profiles. This institute can be defined as the rules by with which the parties ask the questions directly to the examined witness. The rules were drawn from a secular praxis that has formed in Anglo-American orderings. The cross examination has been defined in the Anglo-Saxon countries as the best instrument for discovering the truth. It is based on the principle that the truth is better known when more space is left to the dialectic between the parties in conflict. Compared to that model, the Italian code is characterized for the fact that it reproduces some of those rules and by the choice of giving the judge a penetrating control on the development of the cross examination. In particular, the purpose of is to assure the question’s relevancy, the answer’s sincerity, the examination’s loyalty and the accuracy of the objections (…)” (art. 499, c.p.p.). The cross examination is structured in three fundamental stagesmoments 1) direct examination, 2) cross examination and 3) re-examination (art. 498): the subjects that ask the questions are the public minister and the private party’s advocates. The direct examination is conduct by the party that asked to interrogate the witness. The direct examination aim aims to obtain a representation of facts as known to the witness; such facts should be useful in demonstrating the examiner’s thesis. The final purpose of the direct examination is to demonstrate that the witness is reliable and credible. Therefore leading questions are prohibited (art. 499, c.p.p.). Cross-examination is optional in the sense that parties that didn’t ask for admission of that witness, can ask questions. Cross-examination is conducted by the party that has a contrary interest. Cross-examination can be regarding facts or credibility of the witness, or both subjects. Leading questions are permitted during the cross-examination. The re-examination is also optional because it is possible only after the cross-examination. In the re-examination the party that conducted the direct examination can propose new questions. The function of the re-examination is to allow to the examiner to recover the sequence of facts after the cross-examination tried to put in doubt its existence; it also allows explanation of any contradictions into which the witness has fallen; therefore it aims to corroborate the validity of the initially rendered declaration. In this thesis I have dealt with many problematic aspects. In particular, detailed attention has been dedicated to the problem of injurious questions, leading questions, trick questions and the hostile witness. Finally I have evidenced the rules that regulate cross examination with particular reference to their jurisprudential interpretation and to the sanctions eventually predicted in transgression cases.

All'interno di questo elaborato, facendo riferimento ad una sentenza civile, emanata dalla Seconda Corte Intermedia del Popolo di Tianjin, vengono trattate le seguenti tematiche: lavoro e licenziamento in Italia e contratti nella RPC. In particolare, al CAPITOLO I, analizzo le normative italiane in materia di lavoro e licenziamento; al CAPITOLO II, viene effettuato un sunto delle principali caratteristiche dei contratti in Cina. Al CAPITOLO III, si riporta la traduzione della sentenza giuridica con relativo commento. L'obiettivo di questo elaborato è quello di fornire, attraverso l'analisi della sentenza, un'ampia panoramica in materia di lavoro per quanto concerne entrambi i paesi.

Résumé :

La ricerca è incentrata sulla valutazione dell’applicabilità dell’esame ecocontrastografico (CEUS) nello studio delle patologie dell’apparato gastroenterico (GE) dei piccoli animali ed in particolare dell’Inflammatory Bowel Disease (IBD) e dell’ischemia gastrointestinale. Per l’IBD sono stati condotti due studi: uno per determinare l’applicabilità della metodica CEUS nella valutazione della perfusione parietale del duodeno in cani con IBD e l’altro per valutare l’applicabilità della CEUS nel monitoraggio della risposta terapeutica. E’ stato arruolato un gruppo di cani con IBD e uno di cani sani sui quali sono stati formulati e confrontati score clinici, ecografici, ecocontrastografici, endoscopici e istologici del duodeno. Inoltre in un gruppo di soggetti con IBD tali score sono stati confrontati sia al momento della diagnosi che dopo un tempo standardizzato dall’inizio della terapia. Sono state osservate differenze nella modalità di distribuzione del mezzo di contrasto nella parete duodenale tra soggetti sani e quelli con IBD, e tra soggetti con IBD per e post terapia. La quantificazione delle variabili perfusive CEUS ha invece evidenziato differenze statisticamente significative tra soggetti sani e malati per quanto riguarda il Peak Intensity (PI) e Mean Transit Time (MTT). Nessuna differenza è stata evidenziata nelle variabili CEUS dei soggetti con IBD pre- e post-terapia. Su questi risultati è possibile affermare che la CEUS è utile nel distinguere i cani malati dai sani ma non è utili per il monitoraggio terapeutico. Nello studio delle patologie ischemiche GE è stato incluso un piccolo gruppo di cani e gatti (5 pazienti) con lesioni ischemiche di diversa natura. La CEUS ha mostrato una severa riduzione o una completa assenza della perfusione parietale del tratto gastroenterico colpito in confronto alla normale perfusione dei tratti intestinali circostanti dimostrando l’applicabilità e l’utilità di questa metodica nel determinare un danno perfusivo parietale in analogia con quanto già dimostrato in Medicina Umana.
May research project was focused on contrast-enhanced ultrasonography (CEUS) for the evaluation of two different small animal gastrointestinal (GI) diseases: Inflammatory Bowel Disease (IBD) and GI ischemia. Concerning the first disease, 42 dogs with IBD and 20 healthy dogs were included in the study. CEUS of the duodenum was performed to characterize the perfusion of the wall both in healthy and affected dogs. For 12 patients with IBD we performed comparisons of the CEUS parameters before and after two months of a standardized treatment. The duodenal wall exhibited a typical perfusion pattern characterized by a radial and simultaneous enhancement of wall. No differences were observed on the contrast medium distribution between healthy and affected dogs, and between dogs with IBD before and after treatment. Statistically significant differences occurred in the peak intensity and mean transit time between healthy and affected dogs. No significant differences were found for the perfusion parameters before and after treatment. In conclusion, this preliminary study demonstrated that CEUS is useful in distinguishing sick dogs from healthy ones, but it was not able to provide useful information to monitor therapeutic response. Concerning the second disease, the aim of this study was to describe the use of CEUS in a case series of small animal with confirmed GI ischemia. On conventional US all GI lesions were characterized by a focal hypoechoic thickening of the gastric/intestinal wall with loss of normal layering associated with hyperechoic mesentery surrounding the affected GI tract. CEUS showed a severe reduced or absent enhancement of the GI lesions compared to the normally perfused surrounding gastric/intestinal wall. The CEUS features observed in our patients with GI ischemia are similar to those reported in humans. These preliminary findings demonstrated the feasibility and usefulness of CEUS for the detection of canine and feline GI wall impaired perfusion

La tesis aborda el problema de la conservación de la biodiversidad acuática en la Península Ibérica e Islas Baleares desde una doble perspectiva, estableciendo prioridades de conservación para espacios y especies utilizando inventarios de coleópteros acuáticos. En primer lugar, se evalúa si los coleópteros acuáticos pueden ser utilizados como buenos indicadores de biodiversidad en ecosistemas acuáticos mediterráneos. Posteriormente, se seleccionan áreas prioritarias para la conservación de la biodiversidad acuática usando coleópteros como indicadores. Se crea una base de datos de coleópteros acuáticos ibéricos y se evalúa tanto el esfuerzo de muestreo, como la cantidad y naturaleza de los posibles sesgos de los datos. Se ha obtenido una función basada en variables ambientales y espaciales capaz de predecir la distribución de la riqueza de especies en la Península Ibérica. Por último se identifican las especies endémicas mas amenazadas, y se evalúa la efectividad de la Red Natura 2000 en su protección.
This thesis comprises the problem of freshwater biodiversity conservation in the Iberian Peninsula and Balearic Islands using inventories of water beetles as surrogates of biodiversity. The thesis involves different approaches and methods from the double perspective of setting conservation priorities for species and areas. Firstly, the use of water beetles as reliable biodiversity indicators in Maditerranean aquatic ecosystems was assessed. Then, areas for freshwater biodiversity conservation at regional scale were selected using water beetles as biodiversity surrogates. Afterwards, a database of Iberian water beetles was compiled. The survey effort, the degree of geographical coverage and the amount and nature of bias in this database were assessed. Subsequently, a function based on environmental and spatial variables able to predict species richness distribution in the Iberian Peninsula and Balearic Islands were obtained. Lastly, the most threatened endemic water beetles were identified and the effectiveness of existing reserve network (Natura 2000) in protecting them was evaluated.

Lo sviluppo tecnologico e la nascita di Internet ha portato alla costituzione di un nuovo canale di comunicazione il quale costituisce, insieme all'uso dei sistemi informatici, un veicolo per la realizzazione di illeciti. Lo scopo di questa tesi di laurea è di presentare e analizzare in modo critico i principali reati informatici. Le ricerche sono state effettuate attraverso diverse banche dati, lettura di libri e vari articoli. La tesi espone al primo capitolo una panoramica storica e generale sui reati informatici. Nel secondo capitolo viene esaminata la Legge 23 dicembre 1993 n. 547. Il terzo capitolo è il cuore della tesi e analizza i principali reati informatici insieme a relativi casi giuridici. Il quarto ed ultimo capitolo, esamina l'esigenza di avere una normativa comune tra i vari stati del mondo. La trattazione fornisce un quadro d'insieme sul dilagante fenomeno dei crimini informatici, seguito da un'analisi approfondita dei singoli reati. La ricerca e lo studio dei casi reali hanno consentito di concretizzare la normativa teorica.

Novel electronic properties of superlattices, double barrier diodes and other semiconductor microstructures have generated considerable current interest from fundamental as well as device points of views. One of the driving forces behind this interest is the possibility of novel high speed devices; e.g. Sollner et al [1] have shown very high frequency response for double barrier diodes. There are also a number of very interesting fundamental issues as proposed in the original work of Esaki and Tsu [2]. Some of this work has been recently reviewed by Esaki [3] and Capasso et al [4].

Over the last two years, the Esade Center for Social Impact, which is part of Esade Business School in Spain, and its partner BBK, a banking foundation in Bilbao, have been at the center of a web of committed European foundation professionals sharing their thoughts, learnings, practices, frustrations, and eureka moments related to impact measurement and management (IMM). This paper is not a practical guide to Impact Measurement and Management (IMM) – we believe that there are other publications which may help with implementation. Rather, it recounts how foundations at different stages of development and with a range of profiles (corporate, family, operating, and grant-making) are going about IMM on a day-to-day basis and grappling with some of the challenges and philosophical issues arising. For the impact geeks, this will no doubt be interesting grist for the mill. For those not so deep into this space, it might provide an overview of where foundations are in Europe and how they are focusing their efforts on IMM. Our aim is that by synthesizing and sharing what we have heard in this safe space we can inform and inspire others.

Résumé :

Many bacterial pathogens of plants can inject pathogenicity proteins into host cells using a specialized type III secretion system encoded by hrpgenes. This system deliver effector proteins, into plant cells that function in both susceptible and resistant interactions. We have found that the virulence of Erwinia stewartii(Es; syn. Pantoea stewartii) and Erwinia herbicola pv. gypsophilae (Ehg, syn. Pantoea agglomerans), which cause Stewart's wilt of corn and galls on Gypsophila, respectively, depends on hrpgenes. The major objectives of this project were: To increase expression of hrpgenes in order to identify secreted proteins; to identify genes for proteins secreted by the type-III systems and determine if they are required for pathogenicity; and to determine if the secreted proteins can function within eukaryotic cells. We found that transcription of the hrp and effector genes in Es and Ehg is controlled by at least four genes that constitute a regulatory cascade. Environmental and/or physiological signaling appears to be mediated by the HrpX/HrpY two component system, with HrpX functioning as a sensor-kinase and HrpY as a response regulator. HrpYupregulateshrpS, which encodes a transcriptional enhancer. HrpS then activates hrpL, which encodes an alternate sigma factor that recognizes "hrp boxes". All of the regulatory genes are essential for pathogenicity, except HrpX, which appears only to be required for induction of the HR in tobacco by Es. In elucidating this regulatory pathway in both species, we made a number of significant new discoveries. HrpX is unusual for a sensor-kinase because it is cytoplasmic and contains PAS domains, which may sense the redox state of the bacterium. In Es, a novel methyl-accepting protein may function upstream of hrpY and repress hrp gene expression in planta. The esaIR quorum sensing system in Es represses hrp gene expression in Es in response to cell-density. We have discovered six new type III effector proteins in these species, one of which (DspE in Ehg and WtsE in Es) is common to both pathogens. In addition, Es wtsG, which is a homolog of an avrPpiB from P. syringae pv. pisi, and an Ehg ORF, which is a homolog of P. syringae pv. phaseolicola AvrPphD, were both demonstrated to encode virulence proteins. Two plasmidborne, Ehg Hop proteins, HsvG and PthG, are required for infection of gypsophilia, but interestingly, PthG also acts as an Avr elicitor in beets. Using a calmodulin-dependent adenylate cyclase (cyaA) reporter gene, we were successful in demonstrating that an HsvG-CyaA fusion protein can be transferred into human HeLa cells by the type-III system of enteropathogenic E. coli. We believe that our basic research on the characterization of type III virulence effectors should enable future identification of their receptors in plant cells. This may lead to novel approaches for genetically engineering resistant plants by modifying their receptors or inactivating effectors and thus blocking the induction of the susceptible response. Alternatively, hrp gene regulation might also provide a target for plant produced compounds that interfere with recognition of the host by the pathogen. Such strategies would be broadly applicable to a wide range of serious bacterial diseases on many crops throughout the USA and Israel.
