Texte intégralThe current PhD work brings together various studies linked to bacterial protein synthesis. The first chapter is about the origins of protein synthesis at the time of the RNA world. This theoretical work continues with the presentation of a high-resolution structure of the elongation factor G (EF-G) in complex with the ribosome by cryo-electron transmission microscopy (cryo-TEM). We describe for the first time EF-G bound to the ribosome in the absence of any inhibitor. This particular structure of EF-G displays a yet unseen positioning of its third domain, which becomes very flexible. This study helps to understand the way the antibiotic fusidic acid blocks translation. The work then switches to a study of trans-translation, the main rescuing system of stalled ribosomes in bacteria. Trans-translation is generally vital or at least necessary for bacterial virulence. We conducted a preliminary structural study on the way faulty mRNAs are degraded during this process. This is why we present a study of trans-translation as a target for the development of new antibiotics. For this we developed and validated a reporter system for trans-translation, which is used to screen molecules targeting trans-translation

Texte intégralThe residual strength after low velocity impact is a sizing criterion of thin composite structures in aeronautics. The majority of work on the subject is focused on the analysis of plain laminates. This study expands the field of interest to tapered area. The objective is to study the impact damage tolerance of a laminate with ply drop-off using an experiment/ modelling dialogue: on one hand carrying experimental test campaign and on other hand adapting numerical modelling of composite damage. The experimental part successively examines the impact, compression and fatigue R=-l after impact (CAI and FAI) of specimens with ply drop-off. Analysis of the impact results has enabled the identification of impact resistance and damage mechanism. Then, a testing tool for CAI and FAI was specially designed to suit the geometry of the specimens. Compression tests show a static residual strength after impact similar to plain laminates. Fatigue tests carried at 60% of CAI strength show a propagation of impact delamination (below B VID). Failure of specimens occurs after a relatively small number of cycles. While the impact damage tolerance of tapered laminates is comparable to plain laminates under static loading, high vulnerability is observed under fatigue loading. The numerical part allowed to test the Discrete Ply Model (DPM), developed in previous works, on a particular configuration. A specific meshing was realized to account for the discontinuities in the ply drop-off area. Regarding the material law, a unified formulation of the fiber breakage in tension/ compression is implemented. Impact simulation results correlated well with the experimental data. The model is able to predict the overall response of the specimen and internal damage. The modeling of the CAI enabled to confirm the failure mechanisms identified during tests. These numerical results are remarkable since no particular local modeling has been realized for the ply drop-off area. DPM approach has proven robust enough and well suited to damage modeling of unidirectional laminates

Texte intégralThis research work regards the development of a complex structure model with non-linear viscoelastic components. The purpose of this study is to simulate the response of this structure submitted to a random vibration excitation based on a power spectral density definition (PSD). The industrial applicative case is the vibratory insulation of a automotive engine cooling module supported by elastomer mounts. A brief review of elastomers behavior depending on solicitations types enables to identify the parameters of the different investigated models. Preliminary tests have been conducted to define the range of amplitudes of excitations and evaluate the internal warming of rubbers during the full structure validation test. The experimental characterization of the suspension is based on rubbers mounts and their interfaces with the cooling module, in order to take into account in a unique model all nonlinearities due to the viscoelastic behavior, the slidings, and the friction. Measured force-deflection hysteretic cycles in axial and radial direction are post-processed with an expert system developed to obtain the parameters of the retained model: the modified Dahl’s model, generalized to viscoleastic aspect. This process has been developed with Octave/Matlab code. Interpolation and extrapolation methods enable to obtain a good model response on the global operating range. These methods have been coded in an Abaqus UserSubroutine. Imposing random vibration excitation of a non linear mechanical system based on PSD imposes to take into account signal processing aspects. To evaluate response levels versus norms requirements, it’s mandatory to consider the time-frequency transfer. In addition, the size and the complexity of the total finite element model of the industrial structure don’t allow a global resolution in the time domain for all the degrees of freedom. Homogenization and dynamic reduction techniques are used to evaluate the response of the system submitted to large frequency range excitations, and to analyse the behavior of the suspension

Texte intégralThis dissertation approaches the event and argument structure of resultative construction (e.g., Peter wiped the table clean) from the perspective of two understudied and endangered Oceanic languages, Daakaka and Samoan, in which both the manner and result components are realized by verbal predicates, i.e. resultative serial verb constructions (RSVCs). This observation contrasts with non-serializing languages, such as English, in which only one of the two meaning components is expressed by the main verb. By examining the morphosyntactic and semantic properties of two types of resultative construc-tions, namely resultative secondary predication and means constructions, I develop a novel configurational analysis within the generative framework of Distributed Morphology that models cross-linguistic variation in terms of the morphosyntactic size and the semantic composition of the respective meaning components and their interaction with idiosyncratic requirements on roots and argument structure. Based on original fieldwork, I demonstrate that despite the superficial differences in categorial status, Oceanic RSVCs are an instance of the means construction, in which the manner verb directly adjoins to a causative verb modifying the underspecified causing event entailed in the event structure of the causa-tive predicate. Consequently, serializing and non-serializing languages do not vary significantly in their morphosyntactic and semantic composition with further implications for the typology of resultatives in the world’s languages.

Texte intégralThe phylogenetic relationships of the higher arthropod taxa are still of special interest. Especially the interrelationships of the different Crustacea taxa have long been debated. The focus of this investigation is to make a contribution to the phylogenies of two Crustacea taxa using molecular markers: The Phyllopoda which belong together with the Anostraca to the branchiopods, and of the Astacoidea, one of the two higher crayfish taxa (Astacida). The following molecular markers were used: 1) Phyllopoda: the 3rd domain of the mitochondrial encoded 12S rRNA taking into account informations of the secondary structure, the nuclear encoded proteingene EF-1 alpha and the positions of introns found in the coding region of EF-1 alpha. 2) Astacoidea: the 3rd domain of the 12S rRNA and the mitochondrial encoded proteingene cox1. The choice of the mentioned markers in combination with different computercladistical methods allowed to give a satisfying, and in the case of the Astacoidea a more surprising answer to most addressed phylogenetic questions. The gained hypotheses are then discussed in detail in the light of morphological features and hypotheses.

Texte intégralPreviously, simulations were sequentially carried out and only intervene downstream from design activity in a validation goal. The current approaches move towards a concurrent simulation with integration of multiple parameters such as stochastic analyses (statistical), multi-physics simulations of optimization of multi-scenarios analyses. The direct impact of these approaches is the increase in the number of simulation and alternatives of simulation, the growth of the complexity of the models of simulation, and the increase in the volume of data. These evolutions created a strong need for management, traceability, exchange and synchronization of the simulation data to guarantee obtaining of the decision-makings results. These concerns are at the origin of the work presented in this memory. This work specifies an environment of simulation data management in a PLM approach which was defined in optics to support integration of CAD data and finite element analysis. This environment, named EGDS, proposes an innovative modelling of simulation cycles through the adaptation and the enrichment of data management methods of the PLM approach. In order to guarantee its interoperability, EGDS also implements a neutral format dedicated to simulation data management exchanges based on STEP standard

Texte intégralA multi-scale FE2 approach based on the periodic homogenization theory is developed to predict the overall response of nonlinear mechanical and fully coupled thermomechanical 3D composite structures. The computational strategy integrates the periodic microstructure effects by introducing the architecture of the reinforcement and the local constitutive laws.The considered constituents' constitutive laws obey generalized standard materials laws and are formulated within the framework of thermodynamics of irreversible processes. The characteristic equations (equilibrium and thermodynamics laws) are formulated under the assumption of small strains and rotations. They are solved simultaneously at both scales (microscopic and macroscopic) using an incremental scheme. For the numerical implementation, an advanced Meta-UMAT subroutine is developed and combined with a parallelization technique in the finite element commercial software Abaqus/Standard. The multi-scale computational strategy is applied to simulate the overall response of 3D composite structures under complex thermomechanical loading paths. The composite structures consist of thermoplastic polymer matrix with viscoelastic-viscoplastic behavior and ductile damage, reinforced by different types of reinforcements (short fibers or woven fabrics). The anisotropic damage within the yarns is modeled through a micromechanical approach to follow the transverse micro-cracks density evolution. This computational strategy is deployed on composite structures having periodic microstructure, whose phases exhibit different types of nonlinear behavior laws (rheology, damage mechanisms and thermomechanical coupling). The capabilities of the multi-scale approach are demonstrated (i) by comparing numerical predictions with experimental results in terms of global response, macroscopic and microscopic strain fields, and (ii) through the access to spatio-temporal distributions of internal variables at the microstructure scale as well as the intrinsic dissipation in the constitutive phases

The goal for these developments is to provide an answer to the problems encountered when analyzing large nonlinear transformations of monolithic and sandwich thin, thick and 3D structures. The common point to various elements is the use of the ULF, while considering the effects of TS and the normal stress to properly take into account the different phenomena across the thickness. Changes and effective techniques have been used to improve performance and avoid locking. The robustness and effectiveness of different models are demonstrated through a large number of monolithic and laminated structures, in either static implicit or dynamic explicit in 2D and 3D

Texte intégralPhosphorylase kinase (PhK) is a key enzyme in glycogenolysis. PhK is a hexadecameric complex, made of four different subunits (alpha, beta, gamma, delta)4. Catalytic activity is conferred by the gamma subunit and is modified by the two related regulatory subunits alpha and beta which together account for ~ 4/5 of the PhK mass. Mutations in PhK lead to Glycogen Storage Disease (GSD) type IX, which is the most frequently encountered disorder of glycogen metabolism. The structural features of the quaternary structure of the holoenzyme and the PhK subunits, except for the catalytic domain of the PhK gamma and the PhK delta subunit are poorly understood. Here we have used sensitive methods of sequence analysis to unravel hidden structural and functional features of the PhK alpha and beta subunits. We confirm that the first domain (A) belongs to the glucoamylase family (Glycosyl Hydrolase 15) and show domains C and D are related to calcineurin B-like proteins, which are EF-hand family members involved in the Ca2+-dependent regulation of kinases. Mutations leading PhK deficiency, mostly missense mutations in PhK alpha, are located within the predicted active sites of these domains, suggesting that they may have a direct impact on their predicted functions. Furthermore, we docked the 3D structures of the different PhK domains into the volume obtained by cryo-electron microscopy at 9. 9 Angstrom resolution constraining with various interaction data reported in the literature. Altogether, our findings open new perspectives to understand how the different PhK subunits may regulate the holoenzyme activity

Texte intégralThis work deals with a new doubly curved and C1 finite shell element for linear and non linear analysis of multilayered structures. Based on a conforming finite element method, a new triangular finite shell element is developped :-with an exact geometric description using an explicit map,-based on a refined model to satisfy interlayer continuity for both displacements and transverse stresses, and free edges conditions for the shell, avoiding the use of correction factors, and characterized by : its simplicity to be used, its robustness according to classical numerical pathologies like locking, spurious modes and so on, its capability to correctly predict both displacements, strains and stresses. The previous qualities are demonstrated through numerical tests issued from litterature and by comparing numerical and experimental results. The end of this work is devoted to the programming environment used for our finite element developments

Texte intégralOver the past few decades, several investigations have shown interest in research and understanding of the executive system. The term executive functions is used as a general construct to characterize a set of cognitive functions that comprise several distinct subcomponents, and whose efficient performance is crucial for the control of thoughts, behaviors and emotions. Executive functions depend on the maturation of the frontal lobes of the brain, especially the prefrontal cortex, although they also involve the parietals, knowing that the front parietal network is strongly associated with it.This exploratory study had as main purpose to contribute to the validation of an executive functions inventory for the Portuguese population, namely, The Behavior Rating Inventory of Executive Function – Second Edition: Teacher Form (BRIEF-2; Gioia, Isquith, Guy, & Kenworthy, 2015).Precision analyzes were performed through the study of internal consistency (Cronbach's alpha) and construct validity (exploratory and confirmatory factor analysis), based on a sample of 114 children and adolescents.The results obtained revealed the good psychometric properties of this instrument in terms of internal consistency, presenting very good values. Regarding the structure of the instrument, the original study´s structure was not replicated, and it was found that the factorial model that is more adequate is constituted, not by three, but by two factors: Cognitive Regulation Index (Factor 1) and Behavior Regulation Index + Emotion Regulation Index (Factor 2), which explains 79.19% of the total variance.In conclusion, the results found in this exploratory study, especially in regards to internal consistency, reveal psychometric robustness and indicate that BRIEF-2, Teacher Form, may be a good option for the study of executive functions in Portuguese children and young people, between 6 and 18 years old.
Ao longo das últimas décadas, diversas investigações têm demonstrado interesse pela pesquisa e compreensão do sistema executivo. O termo funções executivas é utilizado como um construto geral para caraterizar um conjunto de funções cognitivas que comportam vários subcomponentes distintos, e cujo desempenho eficiente é crucial para o controlo de pensamentos, comportamentos e emoções. As funções executivas dependem da maturação dos lobos frontais do cérebro, sobretudo, do córtex pré-frontal, embora também envolvam os lobos parietais, sabendo-se que a rede frontoparietal lhe está fortemente associada.Este estudo exploratório teve como principal objetivo contribuir para a validação de um inventário de funções executivas para a população Portuguesa, designadamente, o Inventário de Avaliação Comportamental de Funções Executivas - 2ª Edição: versão para Professores (BRIEF-2, The Behavior Rating Inventory of Executive Function - Second Edition; Gioia, Isquith, Guy, & Kenworthy, 2015).Foram realizadas análises no âmbito da precisão através do estudo da consistência interna (alfa de Cronbach) e da validade de construto (análise fatorial exploratória e confirmatória), com base numa amostra composta por 114 crianças e adolescentes.Os resultados obtidos evidenciaram as boas propriedades psicométricas deste instrumento ao nível da consistência interna, observando-se valores muito bons. Relativamente à estrutura do instrumento, não foi replicada a do estudo original, tendo-se constatado que o modelo fatorial que melhor se adequa é constituído, não por três, mas sim por dois fatores: Índice de Regulação Cognitiva (Fator 1) e Índice de Regulação Comportamental + Índice de Regulação Emocional (Fator 2), que explicam 79.19% da variância total.Concluindo, os resultados observados neste estudo exploratório, principalmente no que se refere à consistência interna, revelam robustez psicométrica e indicam que o BRIEF-2, versão para Professores, poderá ser uma boa escolha para o estudo das funções executivas nas crianças e jovens portugueses, com idades compreendidas entre os 6 e os 18 anos.
