Natural antibodies (NAbs) are important regulators of tissue homeostasis and inflammation and are thought to have diverse protective roles in a variety of pathological states. E06 is a T15 idiotype IgM NAb exclusively produced by B-1 cells, which recognizes the phosphocholine (PC) head group in oxidized phospholipids on the surface of apoptotic cells and in oxidized LDL (OxLDL), and the PC present on the cell wall of Streptococcus pneumoniae. Here we report that titers of the E06 NAb are selectively increased several-fold in Cd1d-deficient mice, whereas total IgM and IgM antibodies recognizing other oxidation specific epitopes such as in malondialdehyde-modified LDL (MDA-LDL) and OxLDL were not increased. The high titers of E06 in Cd1d-deficient mice are not due to a global increase in IgM-secreting B-1 cells, but they are specifically due to an expansion of E06-secreting splenic B-1 cells. Thus, CD1d-mediated regulation appeared to be suppressive in nature and specific for E06 IgM-secreting cells. The CD1d-mediated regulation of the E06 NAb generation is a novel mechanism that regulates the production of this specific oxidation epitope recognizing NAb.

Abstract Oxidized phospholipids (OxPL), such as oxidized phosphatidylcholine, are generated by oxidative stress (OS)-induced lipid peroxidation. E06 IgM is a natural antibody that recognizes the phosphocholine (PC) moiety of OxPLs, but not native PLs. Generation of transgenic mice expressing a single chain (scFv) form of its antigen-binding domain, “E06-scFv” mice, protects against atherosclerosis, hepatic steatosis and high fat diet-induced loss of bone mass. In addition, E06-scFv increases cancellous and cortical bone mass in both male and female adult mice fed chow diet, by increasing bone formation. Age-related bone loss is associated with increased OS and lipid peroxidation, and is characterized by a reduction in osteoblast number and bone formation. Oxidative stress is involved also in the bone loss caused by sex-steroid deficiency and elevated OS markers are found in unloading-induced bone loss, raising the possibility that an increase of OxPLs induced by OS might be contributing to the pathogenesis of these conditions as well. We aged homozygous E06-scFv transgenic female and male mice and their wild-type littermates up to 22 and 24 months respectively. Serial DXA BMD every 3 months showed that overexpression of E06-scFv attenuated the age-associated bone loss in both sexes. In addition, male and female E06-scFv transgenic mice also accumulated less fat mass than WT littermates during aging. Micro-CT analysis revealed that E06-scFv attenuated the age-associated decline in cancellous, but not cortical, bone mass. The histological analysis of the vertebrae indicated that the aged E06-scFv transgenic mice had increased osteoblasts and decreased osteoclasts compared to the WT mice. To investigate whether the beneficial effect of the E06-scFv could be seen after ovariectomy, 4.5 month old E06-scFv homozygous females and WT controls were ovariectomized (OVX). DXA and micro-CT measurements 6 weeks post- surgery indicated that, unlike aging, E06-scFv did not protect against OVX-induced bone loss in either the cancellous or the cortical compartment. Lastly, we tail-suspended 5.5 month old male mice and sacrificed them 21 days later. E06-scFv transgenic mice had similar cortical bone loss compared to WT mice. In conclusion, the E06-scFV transgene attenuates the age-associated cancellous bone loss in both female and male mice, but has no effect on the OVX- or unloading-induced bone loss. These results fully support our hypothesis that an increase in PC-OxPLs with age, caused at least in part by a decrease in natural anti-PC antibodies, contributes to the age-associated bone loss. This evidence provides proof of concept that blocking PC-OxPLs represents a therapeutic approach to countering the increase of PC-OxPLs with age and their adverse effects on age-related bone loss as well as atherosclerosis and NASH. It also confirms that the mechanisms of cancellous and cortical bone loss are distinct.

Abstract Aims Myocardial ischaemia followed by reperfusion (IR) causes an oxidative burst resulting in cellular dysfunction. Little is known about the impact of oxidative stress on cardiomyocyte lipids and their role in cardiac cell death. Our goal was to identify oxidized phosphatidylcholine-containing phospholipids (OxPL) generated during IR, and to determine their impact on cell viability and myocardial infarct size. Methods and results OxPL were quantitated in isolated rat cardiomyocytes using mass spectrophotometry following 24 h of IR. Cardiomyocyte cell death was quantitated following exogenously added OxPL and in the absence or presence of E06, a ‘natural’ murine monoclonal antibody that binds to the PC headgroup of OxPL. The impact of OxPL on mitochondria in cardiomyocytes was also determined using cell fractionation and Bnip expression. Transgenic Ldlr−/− mice, overexpressing a single-chain variable fragment of E06 (Ldlr−/−-E06-scFv-Tg) were used to assess the effect of inactivating endogenously generated OxPL in vivo on myocardial infarct size. Following IR in vitro, isolated rat cardiomyocytes showed a significant increase in the specific OxPLs PONPC, POVPC, PAzPC, and PGPC (P < 0.05 to P < 0.001 for all). Exogenously added OxPLs resulted in significant death of rat cardiomyocytes, an effect inhibited by E06 (percent cell death with added POVPC was 22.6 ± 4.14% and with PONPC was 25.3 ± 3.4% compared to 8.0 ± 1.6% and 6.4 ± 1.0%, respectively, with the addition of E06, P < 0.05 for both). IR increased mitochondrial content of OxPL in rat cardiomyocytes and also increased expression of Bcl-2 death protein 3 (Bnip3), which was inhibited in presence of E06. Notably cardiomyocytes with Bnip3 knock-down were protected against cytotoxic effects of OxPL. In mice exposed to myocardial IR in vivo, compared to Ldlr−/− mice, Ldlr−/−-E06-scFv-Tg mice had significantly smaller myocardial infarct size normalized to area at risk (72.4 ± 21.9% vs. 47.7 ± 17.6%, P = 0.023). Conclusions OxPL are generated within cardiomyocytes during IR and have detrimental effects on cardiomyocyte viability. Inactivation of OxPL in vivo results in a reduction of infarct size.

Abstract Oxidative stress-induced lipid peroxidation generates oxidized phospholipids (OxPL), such as oxidized phosphatidylcholine, that are pro-inflammatory moieties present on oxidized low density lipoproteins and apoptotic cells. OxPL have been implicated in the pathogenesis of cardiovascular, neurological, autoimmune, liver, and lung diseases. It has been shown earlier that OxPL decrease bone formation. Natural antibodies (NAb) produced by B-1 lymphocytes, bind OxPL and prevent their inflammatory activity. The NAb E06 blocks oxidized phosphatidylcholine, and transgenic expression of a single chain (scFv) form of the antigen-binding domain of E06 IgM (E06-scFv) prevents many age-associated diseases, such as atherosclerosis, non-alcoholic steatohepatitis and age-related bone loss. OxPL are also implicated in the pathogenesis of the metabolic syndrome and diabetes by either causing insulin resistance, increasing macrophage infiltration and inflammation in white adipose tissue, decreasing insulin gene expression, and/or causing death of pancreatic beta cells through induction of oxidative stress and amyloid protein misfolding. Consistent with this evidence, NAb produced by B-1b lymphocytes are protective against obesity associated inflammation, glucose intolerance, and insulin resistance. In the work reported here, we have investigated the impact of oxidized phosphatidylcholine on glucose metabolism and age–related glucose intolerance. To this end, we aged chow-fed female and male C57BL/6 E06-scFv transgenic mice (and their WT littermates) up to 22 and 24 months. Both male and female E06-scFv transgenic mice accumulated less fat mass than WT littermates during aging, as determined by sequential body composition measurements by DXA. Intraperitoneal glucose tolerance test, performed at 10 and at 15 months of age, showed that glucose tolerance was greater in both male and female E06-ScFv transgenic mice than WT littermates and did not differ from the glucose tolerance of young mice. Insulin sensitivity, measured by insulin tolerance test, was increased in 4- and 10-month-old old transgenic females and in 15-month-old transgenic males. There was no difference in food consumption calculated over 5 days or the production of glucose after pyruvate infusion, indicating similar liver gluconeogenesis in both strains. Physical activity or energy expenditure, measured by indirect calorimetry in 11-month-old females, was indistinguishable between transgenic and WT mice. However, E06-scFv transgenic mice exhibited a significant increase in the respiratory exchange ratio, indicating that utilization of glucose was the main metabolic source under basal conditions. Collectively, these results show that blocking oxidized phosphatidylcholine improves age–related glucose intolerance in mice fed a normal diet; hence, OxPL are major culprits of the aging-associated glucose intolerance. Therefore, targeting OxPL with a neutralizing antibody represents a novel therapeutic intervention that may simultaneously ameliorate several age-associated diseases, including the metabolic syndrome and diabetes. Definitive hematopoietic stem and progenitor cells (HSPCs) are generated de novo from a subset of endothelium, the hemogenic endothelium (HE), via a process called the endothelial-to-hematopoietic transition (EHT). To facilitate the study of HE throughout this process, our lab generated a Runx1 enhancer-reporter mouse model (23GFP) to dissect the cellular and molecular events that underlie EHT. Previous work in the lab highlighted that HE undergoes dynamic fate changes already early in development characterized by two distinct states, a competent state (cHE) and a specifying stage (sHE). In the context of EHT, this study rewrote the timeline of hematopoietic commitment during EHT and showed this is initiated in sHE before the proHSC and type I and type II preHSC stages. My aim was to identify and characterize the gene regulatory interactions (GRIs) underlying these cell fate transitions. We reasoned that integration of expression (RNA-seq) and chromatin profiles (ATAC-seq) would facilitate identification of these GRIs. After validating and characterizing the data sets, we focused on TFs where the timing and appearance of the motif coincided with expression of the TF and identified these as putative regulators of accessibility and subsequent changes in gene expression. Focusing on two key cell fate transitions: (1) cHE to sHE and (2) sHE to proHSC, we generated GRI maps. Analysis of these regulatory interactions identified putative causative regulators, including Bcl11a, Mef2c, Ikzf1, and Ikzf2. Looking more closely at Ikzf1, we identified a -28 element required for proper Ikzf1 expression. Interestingly, -28 activity was Runx1-dependent. Further investigation identified Notch signaling as a downstream target of the novel the Runx1-Ikzf1 regulatory axis. Finally, interrogation of this regulatory axis using genetic perturbation revealed a complex interaction whereby Runx1 and Ikzf1 work both individually and synergistically to regulate Notch during in vitro EHT.

This project used a variety of genetic markers to investigate the evolution and population genetics of hydrothermal vent fauna that were recovered from the Scotia Sea, in the Atlantic sector of the Southern Ocean. The origins of one of these species, an undescribed species of Kiwa sp. found on the East Scotia Ridge (ESR) and its constituent family Kiwaidae, a group of vent and seep-associated decapod squat lobsters (infraorder Anomura) was investigated using a concatenated nine-gene dataset and key divergences were dated using fossil calibrations. These results confirm earlier research showing Kiwaidae reside in the superfamily Chirostyloidea, but form a monophyletic clade with the non-chemosynthetic family Chirostylidae and not Eumunididae. Chirostyloid families diverged in the Cretaceous, although extant Kiwaidae radiated in the Eocene, consistent with many other chemosynthetic taxa that appear recently derived. The basal tree position of Pacific species (and the Alaska location of a likely stem-lineage kiwaid fossil) suggests kiwaids originated in the East Pacific. Within a Southern Hemisphere clade, the divergence between the southeastern Pacific K. hirsuta and a non-Pacific lineage (Kiwa sp. ESR and Southwest Indian Ridge kiwaids) is no earlier than 25.9 Ma, consistent with a spread from the Pacific into the Scotia Sea and beyond via now-extinct active ridge connections or mediated by a Miocene onset of the Antarctic Circumpolar Current (ACC) through a newly-opened Drake Passage. This project also investigated the population genetics of three undescribed species found at two vent fields ~ 440 km apart at either end of the ESR: Kiwa sp., a peltospirid gastropod and Lepetodrilus sp. limpets. Lepetodrilus sp. was also found at the Kemp Caldera, a submerged part of the South Sandwich Islands (SSI). Analyses of cytochrome c oxidase subunit 1 (COI) as well as microsatellite loci developed from Roche 454 sequence libraries revealed no differentiation along the ESR for all three species consistent with panmixia, or the dominance of non-equilibrium processes between vent field colonies within a metapopulation, possibly enhanced further by cold-induced arrested larval development. Despite apparent connectivity along the ESR, both COI and microsatellites revealed differentiation between ESR limpets and Kemp Caldera limpets ~ 95 km to the east, possibly owing to the hydrographic isolation of the caldera. Both COI and microsatellite diversity patterns were consistent with recent (< 1 Ma) demographic expansions for all three species (although the influence of selection sweeps on COI cannot be discounted); a pattern observed worldwide at vent communities and may reflect demographic instability over time as a consequence of the stochastic birth and death of vent colonies within a metapopulation. Different COI bottleneck ages between the three species (excluding the influence of possible selection) as well as the absence of kiwaids and peltospirids at Kemp, have been attributed to differences in life history, in particular larval morphology and presumed dispersal strategy. These results highlight the role of larval dispersal of vent fauna along active spreading ridges, both in maintaining vent metapopulations across vent colonies prone to stochastic birth and extinction in the short term, but also in the spread of taxa globally and the formation of biogeographic provinces. The likelihood that the three species presented here exist at vents east of the ESR and SSI, prompts further exploration along ridges in the South Atlantic, in order to investigate the effect of the ACC in enhancing gene flow and delineating biogeographic provinces.

In an exploratory study, techniques were developed for isolating bulk plant proteins and measuring the 13C isotopic compositions of their constituent amino acids by HPLC-IRMS. Samples of plants expected to be of potential palaeodietary significance in northwestern Europe were selected for investigation. Different tissues of plants, leaves and seeds, may be distinguished from each other by the relative 13C isotopic compositions (‘isotopic signatures’) of the amino acids of their constituent proteins. For each tissue type, different plant types may be distinguished in the same way. These signatures can vary slightly according to environment and season, but the variation among types is greater than this. For leaves, isotopic signatures can be used to differentiate (i) nettles, (ii) true grasses, (iii) reeds etc, (iv) trees, (v) legumes, (vi) maize, (vii) freshwater plants and (viii) marine algae. For seeds, these signatures are able to differentiate (i) wheat-type cereals, (ii) barley-type cereals, (iii) C4 cereals, (iv) pseudocereals, (v) legumes and (vi) tree nuts. From investigations using a mixing model, it appears that these signals, particularly those of essential amino acids, are reflected in the tissues of their consumers. Freshwater plants are identified as the base of the food chain for dragonfly larvae, marine algae as the diet of marine molluscs and grass as the diet of archaeological cattle and aurochs. Isotopic ‘marine signals’ identified by previous researchers have been refined using these data and the isotopic signatures of fish muscle. These findings are expected to be of particular value in the study of palaeodiets using proteins from archaeological tissues, especially bone and hair. This approach will also find application in the fields of plant physiology and biochemistry.

Chez le rat, la tolérance des allogreffes rénales CMH-incompatibles peut être induite par l'administration le jour de la greffe d'anticorps anti-CMH II du donneur. Au cours de ce travail, nous avons étudié les mécanismes impliqués dans la phase de maintien de cette tolérance, grâce à l'analyse par microarray des profils d'expression des gènes de greffons rénaux tolérés de longue date. La comparaison des patterns d'expression entre les greffons tolérés et les greffons syngéniques a permis de révéler 5954 gènes différentiellement exprimés (p<0. 05). Une analyse plus approfondie de ce panel de gènes a montré un rôle clé de la voie de signalisation WNT, l'une des voies principales impliquées dans la régulation cellulaire, et qui, jusqu'à présent, n'avait pas été décrite comme impliquée dans le cadre de la transplantation. Plusieurs gènes appartenant à cette voie ont été significativement surexprimés dans les greffons tolérés, en particulier la protéine MMP7 (Matrix Metalloproteinase 7). L'analyse de plusieurs autres molécules relatives à cette voie a pu montrer que la surexpression de MMP7 était la conséquence d'une activation de la voie de signalisation non-classique WNT. L'expression de MMP7 était restreinte aux cellules musculaires lisses des vaisseaux (SMC). En effet, MMP7 n'a pas été détecté ni dans d'autres modèles de tolérance de greffons rénaux ou cardiaques, ni dans les modèles de rejet chronique obtenus dans les mêmes souches de rats. Déterminer les mécanismes de tolérance par lesquels MMP7 contribue au maintien de la tolérance pourrait permettre le développement de nouvelles stratégies visant à améliorer la tolérance du greffon à long terme
In rats, tolerance to MHC incompatible renal allografts can be induced by administration of anti-donor class II antibodies on the day of transplantation. Here we explored the mechanisms involved in the maintenance phase of this tolerance by analyzing intra-graft gene expression profiles by microarray in long-term accepted kidneys, Comparison of the gene expression patterns of tolerated to syngeneic kidneys revealed 5954 differentially expressed genes (p<0. 05). Further analysis of this gene set revealed a key role for the WNT signaling pathway, one of the pivotal pathways involved in cell regulation that bas not yet been implicated in transplantation. Several genes within this pathway were significantly up-regulated in the tolerated grafts, particularly Matrix Metalloproteinase 7 (Fold Change>40). Analysis of several other pathway-related molecules indicated that MMP7 over-expression was the result of the non-canonical WNT signaling pathway. MMP7 expression was restricted to vascular smooth muscle cells (SMC) and was specific to anti-class II antibody-induced tolerance as it was undetectable in other models of renal and heart transplant tolerance and chronic rejection induced across the same strain combination. These results suggest a novel role, for non-canonical WNT signaling in maintaining kidney transplant tolerance in this model, with MMP7 being a key target. Determining the mechanisms whereby MMP7 contributes to transplant tolerance may help in the development of new strategies to improve long-term graft outcome

Up to 65% of children who undergo surgery may experience high levels of preoperative anxiety (Dreger & Tremback, 2006). Preoperative anxiety, poor compliance during induction of anaesthesia, and other maladaptive preoperative behaviours have been shown to predict postoperative maladaptive behaviours (e.g. Stargatt, Davidson, Huang, Czarnecki, Gibson, Stewart, & Jamsen, 2006). Research has illustrated that parent’s anxiety about their child’s operation is related to children’s preoperative anxiety, yet the relationship remains unclear (Kain, Maclaren, Weinberg, Huszti, Anderson & Mayes 2009). With better understanding of parent-child factors associated with increased levels of children’s preoperative anxiety, it is hoped that children at-risk can be identified in advance. The study aimed to investigate the association between certain child, parental and in-hospital setting factors with children’s preoperative anxiety. The study also investigated the association between child self report and parental proxy report of children’s preoperative anxiety. This study employed a cross-sectional correlational design in 90 children aged 3-12 years who underwent elective surgery at the Evelina London Children’s Hospital, and their parents. It was designed to test for relationships between parent’s anxiety, parental coping, parenting style and children’s preoperative anxiety. Correlational analyses showed that parent proxy report of children’s anxiety was significantly associated with observational measures of children’s preoperative anxiety. A multiple linear regression model revealed that child’s age, parent’s state anxiety and parental coping style predicted higher levels of children’s preoperative anxiety. Overall the findings suggest that older children, high levels of parental preoperative anxiety and parents’ emotion focussed coping are risk factors for children’s preoperative anxiety. Assessment of these factors needs to be incorporated into routine preoperative clinics and for interventions to be appropriately tailored and targeted to those parent-child dyads ‘at risk’. Theoretical and clinical implications are discussed as well as limitations and avenues for future research.

This study provides two empirical studies in market-based accounting research. One study focuses on using out-of-sample valuation errors to evaluate various estimation approaches for firm-valuation models. The second empirical study uses portfolio analysis to evaluate an empirical accounting-based firm valuation model developed in the UK context.The first study uses out-of-sample valuation errors as an alternative metric capturing the effectiveness of various estimation approaches in generating reliable estimates of coefficients in accounting-based valuation models and, accordingly, less valuation bias and higher valuation accuracy. Valuation bias is expressed as the mean proportional valuation error, where estimated market value less the actually observed market value divided by the actual market value is the proportional valuation error, and valuation accuracy is measured by both the mean absolute and the mean squared proportional valuation error. We find that deflating the full equation including the constant term of the undeflated model and, hence, estimating without a constant term in the deflated model provides less bias and more accurate value estimates relative to including a constant term in the regression equation. Also estimating the valuation model on high- and low-intangible asset firms separately, instead of pooling the full sample for estimation, provides better performance in all cases. As expected, the results suggest that an extended model including the main accounting variables found to be associated with market value in the UK is better specified than a benchmark model, widely adopted in prior research, where market value is regressed on book value and earnings alone. Inclusion of 'other information' also seems to improve the performance of the models. However, there is no clear evidence that one particular deflator out of the five we investigate outperforms the others, although book value and opening and closing market value appear to generally perform better than sales and number of shares.The second empirical study tests for the existence of a 'mispricing' effect associated with accounting-based valuation models in the UK. It investigates a specific firm valuation model where market value is expressed as a linear combination of book value, earnings, research and development expenditures, dividends, capital contributions, capital expenditures and other information. All these accounting variables have been found value-relevant in prior studies in the UK. Firms are ranked by in-sample proportional valuation errors. Results show that although firms in the higher rank deciles tend to have higher abnormal returns than firms in the lower rank deciles, the difference between the two extreme portfolios (or the hedge returns) is statistically insignificant. As a consequence, accounting-based valuation models do not seem to provide superior estimates of intrinsic value to market values. We can conclude that the UK stock market is semi-strong form efficient, in the sense that it does not appear to be possible to generate positive abnormal returns based upon publicly available accounting information embedded in the valuation models studied.

Abstract Two phase flow pressure drop characteristics observed during condensation and boiling of azeotropic refrigerant mixtures R-404A (R125/R134a/R143a:44/4/52), R-407B (R32/R125/ R134a:10/70/20), R407C (R32/R125/R134a:23/25/52) and R408A (R22/R125/R143a:46/7/47) are presented in this paper. Experiments showed that for liquid Reynolds numbers higher than 3.00 E06, R-408A appears to have greater heat transfer rates than the other blends under investigation. Furthermore, it is quite evident from this data that R-407C has the highest specific pressure drop among the refrigerants under investigation.
