Résumé :
Abstract Background and Aims: Squint surgery is a risk factor for postoperative vomiting (POV) in children. This study was designed to compare the incidence of POV in children undergoing strabismus surgery under balanced anesthesia with sevoflurane versus intravenous anesthesia with propofol. Material and Methods: In this prospective randomized controlled study conducted in a tertiary care ophthalmology hospital, 70 ASA I-II children aged 1-12 years undergoing strabismus surgery were randomized to two groups –Group S (sevoflurane-based anesthesia) and Group P (propofol-based anesthesia) for maintenance. The surgical details, intraoperative hemodynamic parameters, recovery characteristics, and emergence delirium were recorded. Any episode of postoperative vomiting in the 0-2 hours, 2-6 hours, and 6-24 hours period was noted. Rescue antiemetic was administered if there was more than one episode of vomiting. Results: Both the groups were similar with respect to demographic and surgical details. The average duration of surgery was 118.2 ± 41.88 min in group S and 137.32 ± 39.09 min in group P (P = .05). Four children in group S (11.4%) and one child in group P (2.9%) had POV in the first 24 hours but this was not statistically significant (P = .36). The median time to discharge from post anesthesia care unit was significantly less (P = .02) in the P group (50 min) than in the S group (60 min). Conclusion: Propofol-based anesthesia does not offer advantage over sevoflurane, in reducing POV after squint surgery, when dual prophylaxis with dexamethasone and ondansetron is administered. It, however, reduces the duration of stay in the post anesthesia care unit.

Résumé :
Despite the upsurge in mushroom consumption in Nigeria, breeding of new strains for improved quality, early maturity and biological efficiency is a new concept. This study investigated cross-compatibility among three mushroom parental strains (Pleurotus ostreatus, P. tuber-regium and Lentinus edodes) and assessed yield attributes in parents and the progenies of compatible strains – (P. ostreatus and L. edodes). The crosses were carried out at the Department of Biology, Kogi State University, Anyigba, Kogi State, Nigeria using the dual culture technique. Among the twenty-seven combinations, only five (5) hybridized successfully. Crosses between P. tuber-regium and the other two strains (P. ostreatus and L. edodes) failed to yield viable progenies while the cross between P. ostreatus (po) and L. edodes (le) yielded five (5) new strains. Data collected on morphological and yield attributes of the compatible parents and the new strains revealed that three (3) strains (po1 x le1, po3 x le2 and po3 x le3) produced white fluffy colony growth morphology which represented the characters of P. ostreatus while the remaining two strains (po2 x le1 and po2 x le3) had milky, appressed morphology, representing the attributes of L. edodes. The strain from cross combination po3 x le3 completed spawn run in 37 days which was the fastest (ρ < 0.05) among the strains and also exhibited superiority (ρ < 0.05 or ρ < 0.01) for all other attributes including fresh yield and biological efficiency over other strains and the two parents. Two other strains viz. po2 x le3 and po3 x le2 also exhibited significantly (ρ < 0.05) higher yield and biological efficiency than either of the parental strains. These new strains can be exploited for commercial production of improved mushrooms in Nigeria. Key words: Cross Compatibility, Pleurotus, Dual Culture, Yield, Biology efficiency

Résumé :
A novel single-cavity laser system with flexible pulsing schemes suitable for particle image velocimetry (PIV) measurements is presented. The system is based on the concept of picking selected pulse pairs from an ultra-high repetition rate fiber laser which enables time-resolved and conventional dual-pulse PIV at highly flexible recording frame rates. Pulse picking PIV laser systems allow for the use of high-quality low speed dual-frame PIV cameras as well as highspeed cameras in both dual-frame and continuous recording modes, enabling the investigation of highly-transient phenomena at limited fields of view using time-resolved PIV as well as statistical PIV measurements with larger fields of view and higher spatial resolutions. The single-cavity output additionally avoids the alignment of laser beams of laser pulses, which is otherwise necessary in conventional dual-cavity PIV laser systems. Two pulse picking systems were set up and tested: An acousto-optic deflector selecting pulses using the diffraction across an acoustic wave and an electro-optic modulator rotating the polarization state of selected pulses in a Pockels cell. The performance of both systems was evaluated in terms of achievable inter-pulse durations, beam profile alterations, and pulse energy losses caused by the introduction of additional optical elements in the beam path.

Résumé :
A dual synthetic jets actuator driven by different electrical factors was investigated using particle image velocimetry (PIV). A transfer-phase and sub-frequency technique was provided to capture the arbitrary phase of the dual synthetic jets, and a transfer-phase to equal technique was provided to determine the phase of the dual synthetic jets. The results show that both the amplitude and frequency of the electrical forcing voltages vastly affect the flow-field of the dual synthetic jets actuator. Both the forcing frequency and the driving voltage amplitudes contribute to the pressure difference and the area of the lower pressure, which determines the interactions of dual synthetic jets. The dual synthetic jet actuator exits a circumscription of electrical factors in which the actuator works efficiently.

Résumé :
Abstract Research towards aging of migrant populations has so far mostly focused on (bio)medical and psychological aspects. Here we explore to what extent second language ability is a factor influencing the healthy aging process of older Turkish migrants in the Netherlands. To get a first understanding of the linguistic situation of older migrants within this setting, interviews with ten healthcare consultants1 with insight into their local Turkish community were conducted, forming a ‘needs-analysis’ of this group. It emerged that older migrants rely heavily on their ethnically close-knit networks for information and care. In communicating their healthcare needs to others outside the family, the older migrants experience language barriers, directly resulting from low L2 proficiency and L2 anxiety. These barriers prevent access to health information and induce dependence. This initial exploration of the interplay between language and aging in a migrant population can be used as input into more advanced, empirical investigations.

Résumé :
In chronic thromboembolic pulmonary hypertension (CTEPH), assessment of severity requires right heart catheterization (RHC) through cardiac index (CI). Previous studies have shown that dual-energy CT allows a quantitative assessment of the lung perfusion blood volume (PBV). Therefore, the objective was to evaluate the quantitative PBV as a marker of severity in CTEPH. In the present study, thirty-three patients with CTEPH (22 women, 68.2 ± 14.8 years) were included from May 2017 to September 2021. Mean quantitative PBV was 7.6% ± 3.1 and correlated with CI (r = 0.519, p = 0.002). Mean qualitative PBV was 41.1 ± 13.4 and did not correlate with CI. Quantitative PBV AUC values were 0.795 (95% CI: 0.637–0.953, p = 0.013) for a CI ≥ 2 L/min/m2 and 0.752 (95% CI: 0.575–0.929, p = 0.020) for a CI ≥ 2.5 L/min/m2. In conclusion, quantitative lung PBV outperformed qualitative PBV for its correlation with the cardiac index and may be used as a non-invasive marker of severity in CTPEH patients.

Résumé :
Pulmonary arterial dual-energy (aDE) CT is an established technique for evaluating pulmonary perfusion blood volume (PBV). As DECT protocols are increasingly used for thoraco-abdominal CT, this study assessed image quality and clinical findings in portal–venous phase dual-energy (vDE) CT and compared it to aDE. In 95 patients, vDE-CT was performed using a dual-source scanner (70/Sn150 kV, 560/140 ref.mAs). Pulmonary triggered aDE-CT served as reference (n = 94). PBV was reconstructed using a dedicated algorithm. Mean relative attenuation was measured in the pulmonary trunk, aorta, and segmented lung parenchyma. A distribution ratio (DL) between vessels and parenchyma was calculated to assess the iodine uptake of the lung parenchyma. Subjective overall diagnostic image quality was assessed for PBV images on a five-point Likert scale. Image artifacts were classified into five groups based on scale rating and compared between vDE and aDE. Pathological findings were correlated with the anatomical image datasets. Mean relative attenuation of the lung parenchyma was comparable in both groups (vDE: 23 ± 6 HU and aDE: 22 ± 7 HU), but significantly lower in the vessels of vDE. Therefore, iodine uptake of the lung parenchyma was significantly higher in vDE (DL: 10% vs. 8%, p < 0.01). The subjective overall image quality of the PBV images was comparable (p = 0.5). Rotation and streak artifacts were found in most of the patients (>86%, both p > 0.6). Dual-source artifacts were found in only a few patients in both groups (vDE 5%, aDE 7%, p = 0.5). Recess and subpleural artifacts were increased in vDE (vDE 53/27%, aDE 24/7%, both p < 0.001). Pathological findings were found in 19% of the vDE patients and 59% of the aDE patients. Comparable objective and subjective image quality of lung perfusion can be obtained in vDE and aDE. Iodine uptake of the lung parenchyma is increased in vDE compared to aDE, suggesting an interstitial pooling effect. Knowledge of the different appearances of artifacts will aid in the interpretation of the images. Additional clinical information about the lung parenchyma can be provided by PBV evaluation in vDE.

Résumé :
The aim of this in vitro study was to investigate cytotoxic effects of dual-cure bulk-fill resin materials polymerized with a third-generation LED light-curing unit (LCU) on L929 fibroblast cells in terms of morphology and viability. Three novel dual-cure, flowable bulk-fill materials (Fill-Up!™), a bioactive material (ACTIVA™ BioACTIVE-RESTORATIVE™), and a dual-cure bulk-fill composite material (HyperFIL® HAp) polymerized by LED LCU (VALO™ Cordless) were tested. Each material was placed in plastic rings (4 mm × 5 mm) in a single layer. Unpolymerized rings filled with each material were placed in direct contact with cells and then polymerized. After polymerization, the removed medium was readded to wells. In this study, four control groups were performed: the medium-free control group, medium control group, physical control group, and light applied control group. Three samples were prepared from each group. After 24 h, the morphology of cells was examined and a WST-1 test was performed. The percentage of cell viability (PCV) of each group was calculated. The experiment was repeated three times. Data were analyzed by a Kruskal–Wallis Test and a Mann–Whitney U test. p < 0.05 was considered significant. The PCV of all groups were found to be significantly lower than the medium control group (p < 0.05). The lowest PCV was obtained in HyperFIL® Hap, while highest was in the Fill-Up!™. In the morphology of cells related to the experimental groups, it was observed that the spindle structures of cells were disrupted due to cytotoxicity; cells became rounded and intercellular space increased. There were no significant differences between the control groups (p > 0.05). All control groups showed acceptable PCV (>70%) and cells were spindle-like, similar to the original fibroblast cells. It can be suggested that clinicians should pay attention when applying dual-cure bulk-fill materials in deep cavities, or they should use a liner material under these materials.

Résumé :
<p>Nowadays, POI recommendation has been a hot research area, which are almost based on incomplete social relationships and geographical influence. However, few research simultaneously focuses on the refined social relationship and the user deep implicit topic similarity under a reachable region. Under this background, a novel Dual Geo-Social Relationship and Deep Implicit Interest Topic Similarity mining under a Reachable Region for POI Recommendation (DDR-PR) is proposed. DDR-PR first adopts kernel density estimation to compute the user checking-in reachable area. Under the reachable area, the combined relationship similarity based on the link relationship and common check-in social relationship is computed out. Then, the deep implicit interest topic similarity between users is mined out adopting the proposed topic model RTAU-TCP. We formulate the combined relationship similarity and implicit interest topic similarity as two regularization terms to incorporate into matrix factorization, which can recommend new POIs for a user under his or her reachable area. Extensive experiments prove the superiority of DDR-PR.</p> <p>&nbsp;</p>

Résumé :
Dual-energy computed tomography (DECT) can improve the differentiation of material by using two different X-ray energy spectra, and may provide new imaging techniques to diagnostic radiology to overcome the limitations of conventional CT in characterizing tissue. Some techniques have used dual-energy imaging, which mainly includes dual-sourced, rapid kVp switching, dual-layer detectors, and split-filter imaging. In iodine images, images of the lung’s perfused blood volume (PBV) based on DECT have been applied in patients with pulmonary embolism to obtain both images of the PE occluding the pulmonary artery and the consequent perfusion defects in the lung’s parenchyma. PBV images of the lung also have the potential to indicate the severity of PE, including chronic thromboembolic pulmonary hypertension. Virtual monochromatic imaging can improve the accuracy of diagnosing pulmonary vascular diseases by optimizing kiloelectronvolt settings for various purposes. Iodine images also could provide a new approach in the area of thoracic oncology, for example, for the characterization of pulmonary nodules and mediastinal lymph nodes. DECT-based lung ventilation imaging is also available with noble gases with high atomic numbers, such as xenon, which is similar to iodine. A ventilation map of the lung can be used to image various pulmonary diseases such as chronic obstructive pulmonary disease.

Résumé :
This study presents a dual-wavelength phosphorimeter developed to measure microvascular Po2 (μPo2) in different depths in tissue and demonstrates its use in rat kidney. The used phosphorescent dye is Oxyphor G2 with excitation bands at 440 and 632 nm. The broad spectral gap between the excitation bands combined with a relatively low light absorption of 632 nm light by tissue results in a marked difference in penetration depths of both excitation wavelengths. In rat kidney, we determine the catchments depth of the 440-nm excitation to be 700 μm, whereas the catchments depth of 632 nm is as much as 4 mm. Therefore, the measurements differentiate between cortex and outer medulla, respectively. In vitro, no difference in Po2 readings between both channels was found. On the rat kidney in vivo, the measured cortical μPo2 was on average 20 Torr higher than the medullary μPo2 over a wide Po2 range induced by variations in inspired oxygen fraction. Examples provided from endotoxemia and resuscitation show differences in responses of mean cortical and medullary Po2 readings as well as in the shape of the Po2 histograms. It can be concluded that oxygen-dependent quenching of phosphorescence of Oxyphor G2 allows quantitative measurement of μPo2 noninvasively in two different depths in vivo. Oxygen levels measured by this technique in the rat renal cortex and outer medulla are consistent with previously published values detected by Clark-type oxygen electrodes. Dual-wavelength phosphorimetry is excellently suited for monitoring μPo2 changes in two different anatomical layers under pathophysiological conditions with the characteristics of providing oxygen histograms from two depths and having a penetration depth of several millimeters.

Abstract. A novel dual-plane dye laser particle image velocimetry (PIV) technique used to analyze helicity and energy dissipation in an unexcited turbulent swirling jet of pressurized cold air has established that regions within the flow field of the jet exhibiting high helicity are correlated regions of high turbulent kinetic energy dissipation. This PIV configuration provides estimates of all components of the velocity gradient tensor, facilitating calculation of the helicity from the vorticity components. Application of this novel dual-plane PIV technique is extended in this study to investigate helical structures in a turbulent swirling jet where the underlying shear flow is subjected to external acoustic sinusoidal forcing in a plane perpendicular to the central axis of the jet. It was found that acoustic excitation had a significant effect on the mean velocity profile parallel to the direction of the jet. The horizontal forcing resulted in the generation of vorticity that was skewed with a pitch that favored a distribution of angles around 90° with respect to the velocity vector. The distribution of the time-averaged helicity angle indicated organized helical activity, but such activity is not dominated by large-scale coherent structures of maximal helicity.

Résumé :
To support system-wide synchronization accuracy and precision in the sub-microsecond range without using GPS technique, the precise time protocol (PTP) standard IEEE-1588 v2 is chosen. Recently, a new clock skew estimation technique was proposed for the slave based on a dual slave clock method that assumes that the packet delay variation (PDV) in the Ethernet network is a constant delay. However, papers dealing with the Ethernet network have shown that this PDV is a long range dependency (LRD) process which may be modeled as a fractional Gaussian noise (fGn) with Hurst exponent (H) in the range of0.5<H<1. In this paper, we propose a new clock skew estimator based on the maximum likelihood (ML) technique and derive an approximated expression for the Cramer-Rao lower bound (CRLB) both valid for the case where the PDV is modeled as fGn (0.5<H<1). Simulation results indicate that our new clock skew method outperforms the dual slave clock approach and that the simulated mean square error (MSE) obtained by our new proposed clock skew estimator approaches asymptotically the developed CRLB.

Résumé :
Single-phase inverters are widely used in different renewable energy applications. Although the full-bridge inverter istypically used, dual-buck inverters provide an important advantage, since they eliminate the shoot-through problems. However, solutions proposed in the literature require additional inductors, use linear controllersdesigned around an operation point, or cannot be used in grid-connected applications. This paper presents a hysteresis current control of a single-phase dual-buck full-bridge inverter for grid-connected active power injection. Includes the dynamical modelin state variables, as well as analytical conditions to guarantee the evolution of the error dynamics in a set with boundaries defined by the designer. Moreover, the paper provides guidelines for the design of the dead band required for the transitions between the positive and negative semi-cycles (and vice-versa) of the grid voltage. Finally, simulation results validate the main features of the controller as well as the design of the dead band

Résumé :
This paper deals with spatial interpolation of instantaneous Particle Tracking Velocimetry (PTV) data. Such a task can be handled using generic signal processing tools like B-splines, radial basis functions (RBF) or Kriging. We deal with non time-resolved PTV as gathered by dual frame particle image velocimetry (PIV) systems followed by dual frame PTV algorithms. In such a dual frame context, Physics-based interpolation techniques typically rely on enforcement of divergence free constraints on the interpolated field. The aim of this communication is to explore the wider field of Physics-based PTV interpolators based on learning a deep neural network. These interpolators are learned on a database of flow examples that may be generated by high resolution PIV or DNS. We propose an instance of such a Physics-based PTV interpolator based on the machine learning concept of unrolled optimization. This concept is extended in order to deal with random measurement location, characteristic of PTV data. We also show how to use this concept to perform pressure estimation from PTV data, along with velocity interpolation. The concept is illustrated on a 2D laminar flow, DNS data is used to train and benchmark the proposed method against RBF interpolation. The proposed method is shown to be much more robust than RBF interpolation against low density seeding and noise.

Résumé :
Viral glycoprotein-mediated membrane fusion is an essential step for productive infection of host cells by enveloped viruses; however, due to its rarity and challenges in detection, little is known about the details of fusion events at the single particle level. Here, we have developed dual-color foamy viruses (FVs) composed of eGFP-tagged prototype FV (PFV) Gag and mCherry-tagged Env of either PFV or macaque simian FV (SFVmac) origin that have been optimized for detection of the fusion process. Using our recently developed tracking imaging correlation (TrIC) analysis, we were able to detect the fusion process for both PFV and SFVmac Env containing virions. PFV Env-mediated fusion was observed both at the plasma membrane as well as from endosomes, whereas SFVmac Env-mediated fusion was only observed from endosomes. PFV Env-mediated fusion was observed to happen more often and more rapidly than as for SFVmac Env. Strikingly, using the TrIC method, we detected a novel intermediate state where the envelope and capsids are still tethered but separated by up to 400 nm before final separation of Env and Gag occurred.

PurposeTourism spot or point-of-interest (POI) recommendation has become a common service in people's daily life. The purpose of this paper is to model users' check-in history in order to predict a set of locations that a user may soon visit.Design/methodology/approachThe authors proposed a novel learning-based method, the pattern-based dual learning POI recommendation system as a solution to consider users' interests and the uniformity of popular POI patterns when making recommendations. Differing from traditional long short-term memory (LSTM), a new users’ regularity–POIs’ popularity patterns long short-term memory (UP-LSTM) model was developed to concurrently combine the behaviors of a specific user and common users.FindingsThe authors introduced the concept of dual learning for POI recommendation. Several performance evaluations were conducted on real-life mobility data sets to demonstrate the effectiveness and practicability of POI recommendations. The metrics such as hit rate, precision, recall and F-measure were used to measure the capability of ranking and precise prediction of the proposed model over all baselines. The experimental results indicated that the proposed UP-LSTM model consistently outperformed the state-of-the-art models in all metrics by a large margin.Originality/valueThis study contributes to the existing literature by incorporating a novel pattern–based technique to analyze how the popularity of POIs affects the next move of a particular user. Also, the authors have proposed an effective fusing scheme to boost the prediction performance in the proposed UP-LSTM model. The experimental results and discussions indicate that the combination of the user's regularity and the POIs’ popularity patterns in PDLRec could significantly enhance the performance of POI recommendation.

Résumé :
Capacitive deionization is an emerging desalination technology with mild operation conditions and high energy efficiency. However, its application is limited due to the low deionization capacity of traditional capacitive electrodes. Herein, we report a novel dual-ion capacitive deionization system with a lithium-ion battery cathode LiMn2O4/C and a sodium-ion battery anode NaTi2(PO4)3/C. Lithium ions could enhance the charge transfer during CDI desalination, while NaTi2(PO4)3/C provided direct intercalation sites for sodium ions. The electrochemical capacities of the battery electrodes fitted well, which was favorable for the optimization of the desalination capacity. The low potential of the redox couple Ti3+/Ti4+ (−0.8 V versus Ag/AgCl) and intercalation/deintercalation behaviors of sodium ions that suppressed hydrogen evolution could enlarge the voltage window of the CDI process to 1.8 V. The novel CDI cell achieved an ultrahigh desalination capacity of 140.03 mg·g−1 at 1.8 V with an initial salinity of 20 mM, revealing a new direction for the CDI performance enhancement.

Résumé :
Pure electric vehicle (PEV) equipped with a dual-motor coupling drive system can make full use of the high efficiency working range of the motor in order to improve vehicle efficiency. In order to further expand the application range of the system and to improve its practical application, this paper designs and proposes a new dynamic coupling drive system of three axis-double working modes, which is based on the Simpson planetary gear train. The new system adopts two planetary gears (P1 and P2), and the two sun gears of double rows, planetary carrier of P1 and gear ring of P2 are bunded. The power output of the P1 gear ring (mode 1) and P2 planetary carrier (mode 2) is realized by a controlling wet clutch. This paper adopts the linear interpolation method, least square method and 5-fold CV cross validation method to establish the full load speed characteristics and efficiency characteristics models of two motors (13 and 30 kW). This paper proposes an optimization design method based on an improved simulated annealing (I-SA) algorithm for new system parameter matching and working mode switching strategy determination. The results show that the modeling accuracy of the two motors is high, and the mean value of MAPE is 4.337%. The proposed optimization design method achieves the demand goal of PEV effectively. The I-SA algorithm has good effectiveness and fast convergence, the mean efficiency of the optimized PEV is 83.91% under all working conditions, the maximum speed is 142.56 km/h and the power utilization rate of the dual-motor is 100%. This study proposes a new hardware system and a design optimization method on software and provides a direct reference for the research of PEV drive systems by combining hardware with software.

Résumé :
Purpose. This study was conducted to determine if physical activity volume (PAv) and intensity (PAi) at baseline influence the likelihood of gaining hip bone mineral density (BMD) over 6 years. Design and Sample. In a prospective study, the sample was limited to 244 female nonsmokers, ages 35 to 45 years, and was approximately 90% white. Setting and Measures. PAv and PAi were measured in daily living conditions using accelerometers at baseline. BMD, measured by dual energy x-ray absorptiometry, and several confounding factors were measured in the lab. Analysis. On the basis of BMD change scores, participants were divided into three categories: BMD loss, minimal change, and BMD gain. Risk ratios were used to show the likelihood of BMD gains over time across different levels of PAv and PAi at baseline. Results. Women with higher PAv were more likely to show improvements in hip BMD from baseline to follow-up than their counterparts, as indicated by the Mantel-Haenszel chi-square (χ2mh = 6.1, p = .01). Women with high PAv were 2.50 times (95% CI, 1.19–5.24) more likely to experience hip BMD gains than women with low PAv, and women with moderate PAv were 2.20 times (95% CI, 1.08–4.45) more likely. PAi was not predictive of gains in hip BMD. Adjusting for potential confounders had little influence on the results. Conclusions. Middle-aged women with moderate or high levels of PAv are more likely to experience BMD gains at the hip over time compared with those who have low levels of PAv. However, PAi does not appear to influence the likelihood of gaining BMD at the hip over 6 years.

Résumé :
Peanut (Arachis hypogaea) plants dually infected with tomato spotted wilt tospovirus (TSWV) and peanut mottle potyvirus (PMV) exhibited a wide variety of symptoms, ranging from PMV-like symptoms of transient mild leaf mottle to TSWV-like symptoms of severe leaf distortion and stunting of the plant. Dual infection did not cause greater symptom severity than infection with either virus alone. In the early stages of disease development, PMV symptoms were similar to the first leaf symptoms of TSWV infection, suggesting that identification of TSWV in field-grown peanuts should not depend on visual observation. The virus titer, determined by enzyme-linked immunosorbent assay, indicated a delay in TSWV disease progress in doubleinfected plants, compared to plants infected with TSWV alone. In the later phase of disease progress, the virus titer in dually infected plants was not significantly different from that of singly infected plants. Infection with TSWV and PMV alone and with both viruses in combination was consistent among commercially grown peanut cultivars. In plants inoculated with TSWV or PMV alone or with both viruses in combination, the length of the latent period and final disease incidence, as measured by the number of plants showing symptoms, did not depend on the cultivar.

Résumé :
The experiments of artificial heart valves were divided into in vivo and in vitro experiments; in vivo experiments provide accurate experimental parameters serving in vitro research. Simulation experiment used in vitro usually goes like this, firstly design a similar model or prototype phenomenon, then analysis the model working out the regular parameters related to the process, ruled out the possibility of impact on the study of individual exist in vivo experiment. In vitro experiments are likely designed; performance can be simplified and prominently concerned about contents, even designed some extreme conditions to test. A number of means related to fluid experimental measurement are included, such as the Particle Image Velocimetry(PIV)[1], Dual Catheter Method [2],and ultrasonic method[3] and so on. However, these methods have different kinds of limitations, for example the Dual Catheter Method cannot be used as a routine determination for clinic due to its destructiveness, and PIV test requires expensive equipment. This study was designed by the image processing technology of high-speed photography aiming at the production of a reliable, simple, economic, injury-free and non-contact measurement method.

Résumé :
Time-dependent ventilatory responses to hypoxic and hypercapnic challenges, such as posthypoxic frequency decline (PHxFD) and posthypercapnic frequency decline (PHcFD), could profoundly affect breathing stability. However, little is known about the mechanisms that mediate these phenomena. To determine the contribution of specific carotid body chemostimuli to PHxFD and PHcFD, we developed a novel in situ arterially perfused, vagotomized, decerebrate rat preparation in which central and peripheral chemoreceptors are perfused separately (i.e., a nonanesthetized in situ dual perfused preparation). We confirmed that 1) the perfusion of central and peripheral chemoreceptor compartments was independent by applying specific carotid body hypoxia and hypercapnia before and after carotid sinus nerve transection, 2) the Pco2 chemoresponse of the dual perfused preparation was similar to other decerebrate preparations, and 3) the phrenic output was stable enough to allow investigation of time-dependent phenomena. We then applied four 5-min bouts (separated by 5 min) of specific carotid body hypoxia (40 Torr Po2 and 40 Torr Pco2) or hypercapnia (100 Torr Po2 and 60 Torr Pco2) while holding the brain stem Po2 and Pco2 constant. We report the novel finding that specific carotid body chemostimuli were sufficient to elicit several phrenic time-dependent phenomena in the rat. Hypoxic challenges elicited PHxFD that increased with bout, leading to progressive augmentation of the phrenic response. Conversely, hypercapnia elicited short-term depression and PHcFD, neither of which was bout dependent. These results, placed in the context of previous findings, suggest multiple physiological mechanisms are responsible for PHxFD and PHcFD, a redundancy that may illustrate that these phenomena have significant adaptive advantages.

Résumé :
The propagation of the obligate intracellular parasite Toxoplasma gondii is tightly regulated by calcium signaling. However, the mechanisms by which calcium homeostasis and fluxes are regulated in this human pathogen are not fully understood. To identify Toxoplasma’s calcium homeostasis network, we have characterized a novel EF-hand domain-containing protein, which we have named TgEFP1. We have determined that TgEFP1 localizes to a previously described compartment known as the plant-like vacuole or the endosomal-like compartment (PLV/ELC), which harbors several proteins related to ionic regulation. Interestingly, partial permeabilization techniques showed that TgEFP1 is also secreted into the parasitophorous vacuole (PV), within which the parasite divides. Ultrastructure expansion microscopy confirmed the unusual dual localization of TgEFP1 at the PLV/ELC and the PV. Furthermore, we determined that the localization of TgEFP1 to the PV, but not to the PLV/ELC, is affected by disruption of Golgi-dependent transport with Brefeldin A. Knockout of TgEFP1 results in faster propagation in tissue culture, hypersensitivity to calcium ionophore-induced egress, and premature natural egress. Thus, our work has revealed an interplay between the PV and the PLV/ELC and a role for TgEFP1 in the regulation of calcium-dependent events.
