The turn of the 21st century heralded in the semiconductor age alongside the Anthropocene epoch, characterised by the ever-increasing human impact on the environment. The ecological consequences of semiconductor chip manufacturing are the most predominant within the electronics industry. This is due to current reliance upon large amounts of solvents, acids and gases that have numerous toxicological impacts. Management and assessment of hazardous chemicals is complicated by trade secrets and continual rapid change in the electronic manufacturing process. Of the many subprocesses involved in chip manufacturing, lithographic processes are of particular concern. Current developments in bottom-up lithography, such as directed self-assembly (DSA) of block copolymers (BCPs), are being considered as a next-generation technology for semiconductor chip production. These nanofabrication techniques present a novel opportunity for improving the sustainability of lithography by reducing the number of processing steps, energy and chemical waste products involved. At present, to the extent of our knowledge, there is no published life cycle assessment (LCA) evaluating the environmental impact of new bottom-up lithography versus conventional lithographic techniques. Quantification of this impact is central to verifying whether these new nanofabrication routes can replace conventional deposition techniques in industry as a more environmentally friendly option.

This study aims to describe the optimization of village fund processing to realize the dsa mandini community in the Cileles sub-district of Lebak Banten. The research method used is descriptive with a qualitative approach. The result that will be achieved is to create the concept of a village head apparatus who has a strong will to build his village into a mandini village. The main factor is building human resources and utilizing a friendly and hard-working community culture in people's lives. This is a capital that can be developed by the village head in building his village in mutual cooperation. From the results of this study it was concluded that the head of the village in utilizing village funds, could not be implemented optimally by the village, because the village planning proposal had not been fully approved by the district government which made changes to the village proposal, the change was not necessarily in accordance with the needs of the development his village, as a result village must accept changes even if it is not as expected by the village. Such a change certainly does not allow the village to respond quickly, resulting in delays in receiving village funds and automatically the village development program will be too late. besides the lack of resources (HR) that have the skills needed by the village to go to the village of Mandani.

AbstractThe desire to combine advanced user-friendly interfaces with a product personality communicating environmental friendliness to customers poses new challenges for car interior designers, as little research has been carried out in this field to date. In this paper, the creation of three personas aimed at defining key German car users with pro-environmental behaviour is presented. After collecting ethnographic data of potential drivers through literature review, information about generation and Euro car segment led to the definition of three key user groups. The resulting personas were applied to determine the most important interaction points in car interior. Finally, present design cues of eco-friendly product personality developed in the field of automotive design were explored. Our work presents three strategic directions for the design development of future in-car user interfaces named as a) foster multimodal mobility; b) emphasize the interlinkage economy - sustainable driving; and c) highlight new technological developments. The presented results are meant as an impulse for developers to fit the needs of green customers and drivers when designing user-friendly HMI components.

<p><em>Kota Yogyakarta adalah kota yang mulai menerapkan Age Friendly City. Puskesmas adalah salah satu fasilitas penting dalam kosep Age Friendly City. Namun dari segi aksesibilitas fisik, puskesmas masih terkendala pada beberapa aspek. Oleh karena itu,</em><em> </em><em>tujuan penelitian ini adalah mengetahui aksesibilitas fisik pada puskesmas ramah lansia dalam mencapai Age Friendly City di Kota Yogyakarta. Penelitian ini menggunakan analisis skoring dan pemetaan ArcGIS 10.2. Aksesibilitas dibagi menjadi 3, yakni tinggi, sedang, dan rendah. Kajian aksesibilitas fisik pada puskesmas ramah lansia menunjukkan tingkat jangkauan pelayanan puskesmas bagi lansia adalah tinggi, sedangkan pelayanan sistem jaringan transportasi ramah lansia adalah rendah. Berdasarkan hasil analisis aspek aksesibilitas, empat dari enam skor total aspek adalah tinggi, sehingga tingkat aksesibilitas fisik pada fasilitas puskesmas ramah lansia secara keseluruhan dianggap memiliki tingkat sedang dalam mendukung penerapan Age Friendly City di Kota Yogyakarta. Penelitian ini merekomendasikan untuk memperbaiki kondisi aksesibilitas fisik puskesmas bagi lansia terutama dengan melakukan evaluasi akses pelayanan rute angkutan umum puskesmas, evaluasi letak lokasi halte terhadap puskesmas, dan perbaikan kualitas kondisi jalur pejalan kaki yang belum ramah lansia, serta penyediaan ruang parkir prioritas pada setiap sarana puskesmas agar dapat meningkatkan aksesibilitas fisik puskesmas bagi lansia.</em></p>

AbstractProcess engineering industries are now facing growing economic pressure and societies' demands to improve their production technologies and equipment, making them more efficient and environmentally friendly. However unexpected additional technical and ecological drawbacks may appear as negative side effects of the new environmentally-friendly technologies. Thus, in their efforts to intensify upstream and downstream processes, industrial companies require a systematic aid to avoid compromising of ecological impact. The paper conceptualises a comprehensive approach for eco-innovation and eco- design in process engineering. The approach combines the advantages of Process Intensification as Knowledge-Based Engineering (KBE), inventive tools of Knowledge-Based Innovation (KBI), and main principles and best-practices of Eco-Design and Sustainable Manufacturing. It includes a correlation matrix for identification of eco-engineering contradictions and a process mapping technique for problem definition, database of Process Intensification methods and equipment, as well as a set of strongest inventive operators for eco-ideation.

This master thesis is based on the study of the relationship between literature and society and discusses the literature role in the representation of Brazil’s cultural identity. In this study, we understood that some literary works can evidence the influences of sociocultural factors in their plots; and it is from this perspective that will be analyzed the literary works Memórias de um Sargento de Milícias (1854), written by Manuel Antonio de Almeida, O homem que sabia Javanês (1911), by Lima Barreto, and Macunaíma, (1928), by Mário de Andrade. We understand that these works present Brazilian cultural traits configured as elements of cultural identity that literature symbolically recreates, particularly patrimonialism, friendly and rogueness. Based on studies and empirical knowledge about this relationship, some questions emerged as guiding factors for this investigation: How do the mentioned works present Brazilian cultural traits like cultural identity elements symbolically recreated by literature and how the concepts of friendly man and rogue can be associated with the protagonists of each of them, while as categories of analysis? What meanings of ‘Brazilianism’ evokes? How does the plot of such works consent us to indicate the ideas of patrimonialism and Brazilian way? To find answers to these questions, we define as general objective to analyze the marks of friendly, patrimonialism, rogueness and Brasilian way in the novel, the story and the rhapsody and study the protagonists of each one as friendly man’s literary examples. Moreover, this purpose allows us to study these analysis categories as citizen Brazilian cultural characteristics and of the own Brazil’s cultural formation. Thus, this research is guided by comparative literature, based on the literary sociocritical, which assumes that literature must be analyzed as a phenomenon belonging to the sociocultural context of its arise. Therefore, this is a basic and qualitative research that aims at an interpretative analysis based on the bibliographical review about the categories listed above, articulated by sociological critique studies intertwined with comparative literature. Because of this research process, we reached the conclusion that friendly, rogueness and patrimonialism had been preponderant cultural characteristics at the time works publication, and even considering the culture dynamic character, these brands preponderated until the contemporaneousness as Brazilian cultural and social aspects.
A dissertação ora apresentada se pauta no estudo da relação entre literatura e sociedade e do papel da literatura na representação da identidade cultural do Brasil. Parte-se do princípio de que algumas obras literárias são capazes de evidenciar em suas tramas as influências concretas dos fatores socioculturais e é sob essa perspectiva que se toma como foco analítico as obras literárias Memórias de um Sargento de Milícias (1854), de Manuel Antonio de Almeida, O homem que sabia Javanês (1911), de Lima Barreto, e Macunaíma, (1928), de Mário de Andrade. Entende-se que tais obras apresentam traços culturais brasileiros que operam como elementos de identidade cultural recriados simbolicamente na literatura, dentre os quais, destaca-se, o patrimonialismo, a cordialidade e a malandragem. A partir de estudos e do conhecimento empírico acerca dessa relação, alguns questionamentos despontaram como norteadores dessa investigação: de que modo as obras supracitadas apresentam traços culturais brasileiros que operam como elementos de identidade cultural recriados simbolicamente pela literatura e em que medida os conceitos de homem cordial e malandro podem ser encaixados enquanto categorias de análise nos protagonistas de cada uma delas? Que sentidos de brasilidade isso evoca? De que forma a trama de tais obras deixam entrever as ideias de patrimonialismo e jeitinho brasileiro? Com vistas a encontrar respostas a tais problematizações, traçou-se, como objetivo geral, analisar as marcas da cordialidade, do patrimonialismo, da malandragem e do jeitinho no romance, no conto e na rapsódia e estudar os protagonistas de cada um como exemplos literários do homem cordial. Não obstante, tal propósito favorece o estudo dessas categorias de análise como características culturais do brasileiro e da própria formação cultural do Brasil. Nessa perspectiva, sustentou-se essa pesquisa na literatura comparada, fundamentada na sociocrítica da literatura, a qual parte do princípio de que a literatura deve ser analisada enquanto fenômeno circunscrito ao contexto sociocultural de seu surgimento. Trata-se, portanto, de uma pesquisa de tipo básica e qualitativa, com a qual se busca construir uma análise interpretativista pautada na revisão bibliográfica das categorias acima elencadas, articuladas pelas vias da crítica sociológica entrecruzada com a literatura comparada. Como resultado desse processo de investigação, chegou-se a constatação de que cordialidade, malandragem e patrimonialismo se constituíram como marcas culturais preponderantes à época da publicação das obras em análise e, mesmo em face do caráter dinâmico da cultura, preponderaram até a contemporaneidade, como elementos culturais que perpassam o tecido social brasileiro.

Neste presente estudo pretendeu-se compreender o impacto de um trabalho colaborativo na evolução de conceções e práticas de avaliação das aprendizagens na Educação Pré-Escolar. O trabalho colaborativo consistiu num processo de reflexão entre uma Educadora de Infância (EI) e uma Amiga Critica (critical friend), esta também EI e autora deste estudo. O estudo desenvolveu-se, assim, em torno da seguinte questão de investigação: “Que conceções, práticas e orientações emergem de um trabalho colaborativo entre uma Educadora de Infância e uma Amiga Critica para a renovação da avaliação das aprendizagens na Educação Pré-Escolar?”. O estudo desenvolveu-se em três fases. A primeira consistiu na construção de um referencial teórico que enquadrasse e sustentasse o estudo empírico a realizar. Procedeu-se, para tal, a uma revisão bibliográfica através da leitura de estudos considerados relevantes para a nossa temática. A segunda e terceira fase dizem respeito ao estudo empírico. Este seguiu uma abordagem de estudo de caso, sendo o caso as conceções e práticas de avaliação das aprendizagens da Educadora de Infância participante e sua evolução face ao percurso reflexivo com a Amiga Critica. Esse percurso consistiu na realização de cinco encontros reflexivos entre a Educadora de Infância e a sua Amiga Critica, de aproximadamente duas horas cada, com um formato que se assemelha ao de uma entrevista semi-estruturada. Foram ainda solicitados à Educadora de Infância duas reflexões por escrito, a meio e no final do percurso. Todos os encontros reflexivos foram áudio-gravados e validados pela Educadora participante. Mais especificamente na segunda fase, e num primeiro momento, foi negociado o percurso investigativo com a investigação participante e a Educadora de Infância. Num segundo momento, a que corresponderam os dois primeiros encontros reflexivos, procurou-se desocultar conceções e práticas de avaliação das aprendizagens da Educadora de Infância participante. A terceira fase do estudo traduziu-se, por um lado, num maior aprofundamento da reflexão sobre a temática da avaliação utilizando como suporte dois documentos de autores de referência e, por outro, na avaliação do impacto do percurso experienciado nas conceções e práticas de avaliação das aprendizagens da Educadora de Infância. Nesta fase ocorreram três encontros reflexivos e foram recolhidas duas reflexões escritas da Educadora participante. A análise dos dados foi realizada de forma contínua durante o percurso reflexivo, organizando e interpretando a informação recolhida, fundamentalmente no sentido de orientar a ação em curso. Contudo, procedeu-se a uma análise mais detalhada no final do estudo empírico. Os resultados obtidos evidenciam: (a) a importância do trabalho colaborativo através de encontros reflexivos entre duas Educadoras de Infância, em que uma assume o papel de Amiga Critica, constituindo-se esses encontros como oportunidades de pensar e questionar conceções e práticas, neste caso sobre avaliação de aprendizagens na Educação Pré-Escolar e, consequentemente, um espaço propiciador de desenvolvimento profissional das participantes da investigação; (b) a ocorrência de alguma evolução na compreensão do conceito de avaliação das aprendizagens no discurso educativo da Educadora de Infância participante. Atendendo à reduzida abrangência da presente investigação, e também a algumas das suas limitações, considera-se importante prosseguir com investigações centradas no mesmo objeto de estudo mas incluindo (a) uma maior duração no tempo dos momento reflexivos, (b) a participação de outras Educadoras de Infância e (c) a observação de práticas de avaliação pela Amiga Critica e sua posterior discussão critica.
This study aimed to understand the impact of a collaborative work on the development of conceptions and practices of assessment in Pre-school Education. The collaborative work consisted of a process of reflection between a Childhood Educator (CE) and a Critical Friend, the last also a CE and the author of this study. The study was carried out, then, centered on the following research question: "What conceptions, practices and guidelines emerge from a collaborative work between a Childhood Educator and a Critical Friend, for the renewal of the assessment of learning in Pre-school Education?”. The study was developed in three phases. The first consisted in building a theoretical framework that would justify and sustain the empirical study to be undertaken. A literature review of studies considered relevant for our research was then done. The second and third phases correspond to the empirical study. This followed a case study approach, where the case is the conceptions and practices of assessment of the participant Childhood Educator and their evolution taken into account the reflective process with the Critical friend. This process consisted of five reflective meetings between the Childhood Educator and her Critical Friend, each one with duration of approximately two hours, with a format that resembles a semi-structured interview. The Childhood Educator was also requested to do two written reflections, at the middle and the end of the process. All meetings were audio-recorded and validated by the participant Childhood Educator. More specifically in the second phase, and at a first moment, the collaborative process was negotiated with the participant Institution and the Childhood Educator. In a second moment that corresponded to the two first reflective meetings, we attempted to uncover the conceptions and practices of assessment of the participant Educator. The third phase of the study consisted, in one hand, on a further deepening of the reflection on assessment, with the support of two documents of reference, and, on the other, in the evaluation of the impact of the reflective process on the conceptions and practices of assessment of the participant Educator. At this phase three other reflective meetings occurred and two written reflections were made by the participant Educator. The analysis of the data was done continuously during the reflective process, by organizing and interpreting the information collected, in order to guide the ongoing action. However, we proceeded to a more detailed analysis at the end of the empirical study. The results found support: (a) the importance of the collaborative work through the reflective meetings between the two Childhood Educators, where one assumes the role of critical friend. These meetings constituted opportunities to think about and question conceptions and practices, in this case of assessment in Pre-school education and, consequently, a space which provided professional development of the research participants; (b) the occurrence of same progress in the understand of the concept of assessment in the participant Educator discourse. Given the small scope of this investigation, and also some of its limitations, considered as important to continue with research focusing on the same object of study but including: (a) a longer time duration of the reflective process, (b) participation of other educators and (c) the observation of assessment practices by the critical friend followed by their critical analysis.

The goals of this project were to develop essential genomic tools for modern chickpea genetics and genomics research, map the genes and quantitative traits of importance to chickpea production and generate DNA markers that are well-suited for enhanced chickpea germplasm analysis and breeding. To achieve these research goals, we proposed the following research objectives in this period of the project: 1) Develop an ordered BAC library with an average insert size of 150 - 200 kb (USA); 2) Develop 300 simple sequence repeat (SSR) markers with an aid of the BAC library (USA); 3) Develop SSR marker tags for Ascochyta response, flowering date and grain weight (USA); 4) Develop a molecular genetic map consisting of at least 200 SSR markers (Israel and USA); 5) Map genes and QTLs most important to chickpea production in the U.S. and Israel: Ascochyta response, flowering and seed set date, grain weight, and grain yield under extreme dryland conditions (Israel); and 6) Determine the genetic correlation between the above four traits (Israel). Chickpea is the third most important pulse crop in the world and ranks the first in the Middle East. Chickpea seeds are a good source of plant protein (12.4-31.5%) and carbohydrates (52.4-70.9%). Although it has been demonstrated in other major crops that the modern genetics and genomics research is essential to enhance our capacity for crop genetic improvement and breeding, little work was pursued in these research areas for chickpea. It was absent in resources, tools and infrastructure that are essential for chickpea genomics and modern genetics research. For instance, there were no large-insert BAC and BIBAC libraries, no sufficient and user- friendly DNA markers, and no intraspecific genetic map. Grain sizes, flowering time and Ascochyta response are three main constraints to chickpea production in drylands. Combination of large seeds, early flowering time and Ascochyta blight resistance is desirable and of significance for further genetic improvement of chickpea. However, it was unknown how many genes and/or loci contribute to each of the traits and what correlations occur among them, making breeders difficult to combine these desirable traits. In this period of the project, we developed the resources, tools and infrastructure that are essential for chickpea genomics and modern genetics research. In particular, we constructed the proposed large-insert BAC library and an additional plant-transformation-competent BIBAC library from an Israeli advanced chickpea cultivar, Hadas. The BAC library contains 30,720 clones and has an average insert size of 151 kb, equivalent to 6.3 x chickpea haploid genomes. The BIBAC library contains 18,432 clones and has an average insert size of 135 kb, equivalent to 3.4 x chickpea haploid genomes. The combined libraries contain 49,152 clones, equivalent to 10.7 x chickpea haploid genomes. We identified all SSR loci-containing clones from the chickpea BAC library, generated sequences for 536 SSR loci from a part of the SSR-containing BACs and developed 310 new SSR markers. From the new SSR markers and selected existing SSR markers, we developed a SSR marker-based molecular genetic map of the chickpea genome. The BAC and BIBAC libraries, SSR markers and the molecular genetic map have provided essential resources and tools for modern genetic and genomic analyses of the chickpea genome. Using the SSR markers and genetic map, we mapped the genes and loci for flowering time and Ascochyta responses; one major QTL and a few minor QTLs have been identified for Ascochyta response and one major QTL has been identified for flowering time. The genetic correlations between flowering time, grain weight and Ascochyta response have been established. These results have provided essential tools and knowledge for effective manipulation and enhanced breeding of the traits in chickpea.

Senescence is an agriculturally significant process due to its negative impact to crop yield and postharvest quality. The genetic regulatory systems controlling senescence induction and progress respond to both developmental and environmental stress signals and involve numerous gene expression changes. Knowledge about the key molecular factors which control senescence is very limited. MicroRNAs (miRNAs) are a class of small RNAs which typically function by guiding cleavage of target messenger RNAs. They have been shown to play major roles in a variety of plant processes including development, responses to environmental stresses, and senescence. The long-term goal of this work is to elucidate roles of small RNAs associated with plant senescence. The hypothesis underlying this research is that miRNA-mediated regulation makes important contributions to the senescence process in plants. Specific, original research objectives included: 1) Profiling of small RNAs from senescing plants; 2) Data Analysis and public access via a user-friendly web interface; 3) Validation of senescence-associated miRNAs and target RNAs; 4) Development of transgenic plants for functional analysis of miRNAs in Arabidopsis. Major revisions made in the research compared to the original work plan included 1) Exclusion of the planned work with tomato as recommended by the BARD review panel; 2) Performing miRNA study also in senescing Arabidopsis siliques, in addition to senescing leaves. To identify senescenceregulation of miRNAs in Arabidopsis thaliana, eight small RNA libraries were constructed and sequenced at four different stages of development and senescence from both leaves and siliques, resulting in more than 200 million genome-matched sequences. Parallel Analysis of RNA Ends (PARE) libraries, which enable the large-scale examination of miRNA-guided cleavage products, were also constructed and sequenced, resulting in over 750 million genome-matched sequences. These massive datasets lead to the identification of new miRNAs, as well as new regulation of known miRNAs and their target genes during senescence, many of which have established roles in nutrient responsiveness and cell structural integrity. In keeping with remobilization of nutrients thought to occur during senescence, many miRNAs and targets had opposite expression pattern changes between leaf and silique tissues during the progression of senescence. Taken together, these findings highlight the integral role that miRNAs may play in the remobilization of resources and alteration of cellular structure that is known to occur in senescence. Experiments were initiated for functional analysis of specific senescence-associated miRNAs and respective target genes. Transgenic Arabidopsis plants were generated in which miR408, found in this study to be significantly induced in leaf senescence, was over-expressed either constitutively or under a senescence-specific promoter. These plants are currently being characterized for any altered phenotypes. In addition T-DNA knock out mutants for various target genes identified in this research are being analyzed. This work provides insights about specific miRNAs that contribute to leaf and silique senescence. The knowledge generated may suggest new strategies to monitor and alter the progression of senescence in crops for agricultural improvement.

There is increased demand for high quality fruit produced and marketed with reduced chemical inputs to minimize toxic effects on human health and the environment. Granny Smith (GS) apple quality is reduced by two major physiological disorders, superficial scald and bitter pit (BP). These disorders cause great loss to apple growers worldwide. Superficial scald is commonly controlled by chemical treatments, mainly the antioxidant diphenylamine (DPA) and/or the ethylene action inhibitor, 1-methylcyclopropene (1–MCP). Both chemicals are ineffective in controlling bitter pit incidence. We proposed to investigate the beneficial use of non-chemical, abiotic stress with low O2 (LO2) applied for 10d at 20°C on GS apple fruit. During the project we expanded the treatment to more apple cultivars, Golden Delicious (GD) and Starking Delicious (SD) and another pome fruit, the pear. Apple and pear have similar physiological disorders that develop during cold storage and we examined if the LO2 treatment would also be effective on pear. Application of 0.5% LO2 atmosphere for 10d at 20°C or 500ppb 1-MCP at 20°C prior to cold storage at 0°C, was effective in reducing superficial scald in GS apple. Moreover, LO2 pretreatment was also effective in reducing bitter pit (BP) development in California GS and Israeli GD and SD apples The BP symptoms in GS from California were much more prominent, so the effect of LO2 was more dramatic than the effect on the Israeli cvs. GD and SD, nevertheless the LO2 treatment showed the same trend in all cultivars in reducing BP. The LO2 and 1-MCP -treated fruit exhibited lower levels of ethylene, - farnesene and its oxidation product, 6-methyl-5-hepten-2-one (MHO), as determined by SPME/GC-MS analysis. In addition, LO2 pretreatment applied to California Bartlett or Israeli Spadona pears was effective in reducing superficial scald, senescent scald and internal breakdown after 4 m of cold storage at 0°C. For GS apple, low-temperature storage resulted in oxidative stress and chilling injury, caused by increased production of superoxide anions which in turn led to the generation of other dangerous reactive oxygen species (ROS). Using confocal laser-scanning microscopy and H2O2 measurements of apple peel, we observed ROS accumulation in control fruit, while negligible amounts were found in LO2 and 1-MCP treated fruit. Gene-expression levels of ROS-scavenging enzymes were induced by the various pretreatments: catalase was induced by LO2 treatment, whereas Mn superoxide dismutase was induced by 1-MCP treatment. We assume that LO2 and 1-MCP pretreated fruit remained healthier due to reduced production of ethylene and reactive oxygen substances, such as MHO, during cold storage. The LO2-treated apple exhibited greener peel and firmer fruit after 6 m of cold storage, and the fruit had high crispiness leading to high taste preference. In both pear cultivars, the LO2 treatment led to a reduction in internal breakdown and browning around the seed cavity. We tested the LO2 pre-storage treatment on a semi-commercial scale that would be applicable to a small organic grower by sealing the fruit within the plastic field bins. The treatment was most effective with a continuous flow of nitrogen through the bins; however, a single 6 hour flush of nitrogen was also fairly effective. In addition, we determined that it was very important to have the oxygen levels below 0.5% for approximately 10 days to achieve good scald control, not counting the time required to reduce the oxygen concentration. Our LO2 technology has been proven in this project to be effective in reducing several physiological disorders developed in pome fruit during cold storage. We hope that our non-chemical treatment which is friendly to the environment will be used in the near future for the organic apple and pear industry. The next step should be an analysis of the cost-benefits and commercial feasibility.

The general objective of this study was to develop rationally designed mimetic antagonists (and agonists) of the PK/PBAN Np class with enhanced bio-stability and bioavailability as prototypes for effective and environmentally friendly pest insect management agents. The PK/PBAN family is a multifunctional group of Nps that mediates key functions in insects (sex pheromone biosynthesis, cuticular melanization, myotropic activity, diapause and pupal development) and is, therefore, of high scientific and applied interest. The objectives of the current study were: (i) to identify an antagonist biophores (ii) to develop an arsenal of amphiphilic topically active PK/PBAN antagonists with an array of different time-release profiles based on the previously developed prototype analog; (iii) to develop rationally designed non-peptide SMLs based on the antagonist biophore determined in (i) and evaluate them in cloned receptor microplate binding assays and by pheromonotropic, melanotropic and pupariation in vivo assays. (iv) to clone PK/PBAN receptors (PK/PBAN-Rs) for further understanding of receptor-ligand interactions; (v) to develop microplate binding assays for screening the above SMLs. In the course of the granting period A series of amphiphilic PK/PBAN analogs based on a linear lead antagonist from the previous BARD grant was synthesized that incorporated a diverse array of hydrophobic groups (HR-Suc-A[dF]PRLa). Others were synthesized via the attachment of polyethylene glycol (PEG) polymers. A hydrophobic, biostablePK/PBAN/DH analog DH-2Abf-K prevented the onset of the protective state of diapause in H. zea pupae [EC50=7 pmol/larva] following injection into the preceding larval stage. It effectively induces the crop pest to commit a form of ‘ecological suicide’. Evaluation of a set of amphiphilic PK analogs with a diverse array of hydrophobic groups of the formula HR-Suc-FTPRLa led to the identification of analog T-63 (HR=Decyl) that increased the extent of diapause termination by a factor of 70% when applied topically to newly emerged pupae. Another biostablePK analog PK-Oic-1 featured anti-feedant and aphicidal properties that matched the potency of some commercial aphicides. Native PK showed no significant activity. The aphicidal effects were blocked by a new PEGylated PK antagonist analog PK-dF-PEG4, suggesting that the activity is mediated by a PK/PBAN receptor and therefore indicative of a novel and selective mode-of-action. Using a novel transPro mimetic motif (dihydroimidazole; ‘Jones’) developed in previous BARD-sponsored work, the first antagonist for the diapause hormone (DH), DH-Jo, was developed and shown to block over 50% of H. zea pupal diapause termination activity of native DH. This novel antagonist development strategy may be applicable to other invertebrate and vertebrate hormones that feature a transPro in the active core. The research identifies a critical component of the antagonist biophore for this PK/PBAN receptor subtype, i.e. a trans-oriented Pro. Additional work led to the molecular cloning and functional characterization of the DH receptor from H. zea, allowing for the discovery of three other DH antagonist analogs: Drosophila ETH, a β-AA analog, and a dF analog. The receptor experiments identified an agonist (DH-2Abf-dA) with a maximal response greater than native DH. ‘Deconvolution’ of a rationally-designed nonpeptide heterocyclic combinatorial library with a cyclic bis-guanidino (BG) scaffold led to discovery of several members that elicited activity in a pupariation acceleration assay, and one that also showed activity in an H. zea diapause termination assay, eliciting a maximal response of 90%. Molecular cloning and functional characterization of a CAP2b antidiuretic receptor from the kissing bug (R. prolixus) as well as the first CAP2b and PK receptors from a tick was also achieved. Notably, the PK/PBAN-like receptor from the cattle fever tick is unique among known PK/PBAN and CAP2b receptors in that it can interact with both ligand types, providing further evidence for an evolutionary relationship between these two NP families. In the course of the granting period we also managed to clone the PK/PBAN-R of H. peltigera, to express it and the S. littoralis-R Sf-9 cells and to evaluate their interaction with a variety of PK/PBAN ligands. In addition, three functional microplate assays in a HTS format have been developed: a cell-membrane competitive ligand binding assay; a Ca flux assay and a whole cell cAMP ELISA. The Ca flux assay has been used for receptor characterization due to its extremely high sensitivity. Computer homology studies were carried out to predict both receptor’s SAR and based on this analysis 8 mutants have been generated. The bioavailability of small linear antagonistic peptides has been evaluated and was found to be highly effective as sex pheromone biosynthesis inhibitors. The activity of 11 new amphiphilic analogs has also been evaluated. Unfortunately, due to a problem with the Heliothis moth colony we were unable to select those with pheromonotropic antagonistic activity and further check their bioavailability. Six peptides exhibited some melanotropic antagonistic activity but due to the low inhibitory effect the peptides were not further tested for bioavailability in S. littoralis larvae. Despite the fact that no new antagonistic peptides were discovered in the course of this granting period the results contribute to a better understanding of the interaction of the PK/PBAN family of Nps with their receptors, provided several HT assays for screening of libraries of various origin for presence of PK/PBAN-Ragonists and antagonists and provided important practical information for the further design of new, peptide-based insecticide prototypes aimed at the disruption of key neuroendocrine physiological functions in pest insects.
