In questa tesi studiamo alcune proprietà delle "Varietà Doppie" o D-Varietà. In particolare studiamo la teoria delle deformazioni di D-Strutture e di D-Strutture CR, e troviamo una condizione che è equivalente alla classica condizione di Maurer-Cartan che descrive l'integrabilità di deformazioni di D-Strutture. Successivamente prestiamo attenzione alla coomologia delle D-Varietà, provando che una versione D-complessa del del-delbar-Lemma non può essere vera per D-varietà compatte. Inoltre sono stabilite alcune proprietà di sottogruppi speciali della coomologia di de-Rham, ottenute studiando il loro comportamento sotto l'azione di deformazioni. Infine, un risultato riguardante le sottovarietà Lagrangiane minimali dovuto ad Harvey e Lawson riguardante le varietà D-Kahler Ricci-Piatte è generalizzato a una classe di varietà simplettiche quasi D-complesse.<br>We study some properties of Double Manifold, or D-Manifolds. In particular, we study of deformations of D-structures and of CR D-structures, and we found a condition which is equivalent to the classical Maurer-Cartan equation describing the integrability of the deformations. We also focus on the cohomological properties of D-Manifold, showing that a del-delbar-Lemma can not hold for any compact D-Manifold. We also state some properties of special subgroups of de-Rham cohomology, studing also their behaviour under small deformations. Finally, a result by Harvey and Lawson about the minimal Lagrangian Submanifold of a D-Kahler Ricci-flat manifold is generalized to the case of a special almost D-complex symplectic manifold.

Résumé :

The finite-element method is based on the discretization of a distributed system. The system subdivision process is called mesh generation. The number, order, geometric characteristics (shape, size and orientation) and physical characteristics (material properties, boundary conditions and load conditions) of elements in the mesh will affect the accuracy of the finite-element solution. The objective of this project is to establish a number of criteria for evaluating 3-D mesh-generation software, and to select the 3-D mesh generator that is most suitable for use in our software pipeline for modelling and simulation of complex natural structures. The evaluation criteria for this project include: ability to preserve the surface mesh during 3-D mesh generation; mesh quality; robustness; time efficiency; and cost. The mesh quality is assessed by visualization methods; histograms of the shape qualities and sizes of elements; solution residuals; condition numbers; and closeness to the exact solution as estimated by a convergence curve. Four unstructured mesh-generation programmes (GiD, Gmsh, GRUMMP and TetGen) have been evaluated. A thin block and three structures (one ligament and two ossicles) of the middle ear were chosen to be the models for the evaluation. A programme was developed to convert the surface definitions describing the models to the native file formats of the mesh-generation programmes, to verify that the surface meshes of these models could be successfully imported into the mesh-generation programmes, and to verify that the resulting volume meshes are topologically correct. The results of the evaluation indicate that the mean value of the shape qualities of elements, the root mean square of the solution residuals, and the closeness to the exact solution are good indicators of the overall mesh quality. The Gmsh programme is finally selected as the best 3-D mesh generator for the purposes of our software pipeline.

Dans cette thèse, nous utilisons la théorie des espaces twistoriels afin de construire des structures complexes non standards (en un sens bien précis) sur des produits de 4-variétés réelles avec la sphère de dimension deux. Pour cela nous explicitons l'ensemble des surfaces complexes dont le fibré twistoriel est topologiquement trivial. Dans un deuxième temps nous déterminons parmi ces surfaces celles qui peuvent être munies d'une métrique riemannienne anti-autoduale. De ces résultats, nous déduisons une famille d'exemples simples de 4-variétés réelles parallélisables sans structure complexe. L'espace twistoriel associé à ces variétés admet une structure complexe. C'est notre première classe de 6-variétés munies d'une structure complexe non standard. Une deuxième classe d'exemple sera construite à partir de ces travaux. Enfin, et de façon indépendante, nous étudions brièvement les propriétés de connexités rationnelles des espaces twistoriels.

Résumé :

This work is focused on the recognition of natural lipopolysaccharide (LPS) by the innate immune protein human lung surfactant protein D (hSP-D) in the form of a biologically active recombinant fragment (rfhSP-D), containing the α-helical coiled-coil and three carbohydrate recognition domains (CRD). Intact LPS from two bacterial strains, S. minnesota (R5 mutant) and H. influenzae type b Eagan (CA7 mutant), were delipidated by means of mild acid hydrolysis, leaving the purified polysaccharide (PS) to be used in X-ray diffraction studies by means of co-crystallisation with rfhSP-D. S. minnesota R7 full LPS was also investigated following development of a suitable solubilisation method which also utilised the LPS from E. coli O111:B4. The structural studies of rfhSP-D bound to H. influenzae Eagan CA7 PS (solved and refined at 2.98 Å) and to S. minnesota rough mutant LPS/PS (solved and refined at 3.3 Å) reveal that rfhSP-D binds to LPS preferentially through the non-terminal inner core heptose HepI via the O6’ and O7’ hydroxyls. rfhSP-D recognition of S. minnesota HepI shows a similar bound heptose orientation to that previously reported for heptose binding by rfhSP-D in the literature with an indication of normal Kdo in the inner core Kdo-Hep-Hep trisaccharide. rfhSP-D recognition of the HepI of H. influenzae Eagan CA7 reveals a novel bound heptose orientation, with the heptose rotated by 180° about C5-C6, resulting in the O6’ and O7’ hydroxyls being interchanged with respect to coordination to Ca1 and protein. The novel orientation of HepI is accompanied by a salt bridge being formed between the flanking residue Arg343 and Glu347, both of which adopt a previously unseen conformation. The novel binding mechanism of rfhSP-D for Eagan CA7 suggests flexibility in recognition and offers evidence to explain why this mutant binds more weakly than the Eagan 4A mutant to both rfhSP-D and hSP-D.

Le récepteur de la vitamine D (VDR) est un facteur de transcription activé par la forme active de la vitamine D3. VDR est une cible thérapeutique potentielle pour de multiples pathologies telles que les maladies auto-immunes et neurodégénératives et certains cancers. VDR module l’expression de gènes par le recrutement sélectif de corégulateurs. Les données structurales disponibles à ce jour pour des complexes de récepteur nucléaire-corégulateurs sont très limitées. Cette étude se focalise sur l’architecture du complexe formé par VDR et un grand fragment du coactivateur MED1, une sous-unité du complexe Médiateur qui fait le lien entre les récepteurs nucléaires et la machinerie basale de transcription. Les résultats obtenus nous sont permis de caractériser l'interaction du récepteur avec le coactivateur et de révéler l'architecture globale du complexe. Ce travail fournit une base solide pour la détermination structurale d’autres complexes impliqués dans le contrôle de la transcription<br>The vitamin D nuclear receptor (VDR) is a transcription factor activated by the biologically active form of vitamin D3. VDR is a potential candidate to treat neurodegenerative and autoimmune disorders, and cancer. VDR modulates the expression of vitamin D3-regulated genes by selective recruitment of coregulators of transcription which are, in turn, attractive targets in epigenetic-oriented drug discovery. Available structural data for receptor-coregulator complexes are limited; investigation of such complexes is highly important. The present work focuses on the architecture of the complex between VDR and a large part of the coactivator MED1, a subunit of the Mediator complex linking nuclear receptors to the basal transcription machinery. Obtained results revealed important details of the interaction, as well as the overall organization of the complex. This work provides a solid background for the structural investigation of similar complexes involved in the transcriptional control

The cyclin-dependent kinases (CDKs) are a well-conserved family of serine/threonine protein kinases that drive mammalian cell cycle progression. The transition of cells through the early G1 stage of the cell cycle is co-ordinated by CDK4 and CDK6 following mitogen-dependent expression of D-type cyclins. Irregular CDK activity results in abnormal cell cycle control and is a significant indicator of poor cancer prognosis. The chaperone Hsp90, with the adaptor co-chaperone Cdc37, plays a key role in regulating the stability and function of a substantial proportion of the kinome including CDK4 and CDK6. However, the mechanism of CDK4(6)-cyclin D complex formation and regulation via the Hsp90 chaperone system remains poorly understood. A range of biophysical and biochemical techniques have been used to investigate the mechanism of CDK4(6)-cyclin D complex formation and regulation by the D-type cyclins and cyclin-dependent kinase inhibitors (CKIs) via the Hsp90 chaperone system. These have revealed that Cdc37 complexes can distinguish D-type cyclins, with CDK6 being more readily relinquished to cyclin D3 than cyclin D1, while the presence of p27KIP1 can stabilise the CDK-cyclin complexes against re-distribution into complexes with Cdc37, potentially providing an assembly role for CDK-cyclin complexes. In contrast to the D-type cyclins the CDKs were found to be readily relinquished by Cdc37 to members of the INK family, potent inhibitors of CDK4 and CDK6. However, this potency can be greatly reduced by the introduction of clinically significant INK point mutations, one of which differs greatly in its ability to bind and displace CDK4 and CDK6. In addition, co-crystal structures of selected CDK4/6 inhibitors bound to CDK2-cyclin A have been determined to gain a greater understanding of inhibitor selectivity and binding modes. Taken together these results suggest the Cdc37-Hsp90 chaperone system provides an opportunity to fine tune the regulation of CDK4 and CDK6 activities during G1.

Résumé :

Le complexe d'initiation de la traduction 3 des eucaryotes (eIF3) joue un role central dans le réseau d'interaction des divers facteurs d'initiation de la traduction qui s'assemblent sur les ribosomes 40S, et participent aux différentes réactions au cours de la voie d'initiation de la traduction. Chez Saccharomyces cerevisiaea, ce complexe est composé de cinq sous-unités, qui sont toutes homologues avec les sous-unités de cœur du complexe eIF3 des mammifères, composé de 13 sous-unités. Un objectif majeur actuellement est d'obtenir une structure tridimensionnelle de ce complexe. Dans une première étape vers la résolution de cette structure, les efforts portent sur la détermination des régions de liaisons entre sous-unités, dont assez peu sont actuellement connues. La région d'interaction entre la sous-unité 3i et le domaine C-terminal extrémal de 3b a récemment été résolu par RMN et structure cristallographique. D'un autre côté, la région d'interaction entre 3i et 3g, bien que localisée à l'extrémité N-terminale de 3g, doit encore être précisée. Les échanges hydrogène/deutérium (HDX) se sont développés depuis les années 1990 comme outil d'analyse structurale de protéines et de complexes multiprotéiques. La spectrométrie de masse est couramment utilisée pour réaliser la mesure de l'échange. L'approche HDX la plus usuelle repose sur une mesure de masse de peptide marqués issus d'une digestion enzymatique des protéines d'intérêt afin de déterminer le contenu en deutérium ainsi que leur vitesse d'incorporation. Pour ce travail, un spectromètre de masse à ultra-haute résolution, de type FT-ICR 7T, a été utilisé conjointement avec une séparation nano-LC pour générer des données HDX-MS de haute qualité. La précision de mesure de masse d'un spectromètre de masse FT-ICR n'est pas suffisante en elle-même pour identifier de façon certaine les peptides issus d'une digestion à la pepsine, du fait de l'absence de spécificité de la pepsine. Nous avons en conséquence développé une approche statistique pour l'identification des peptides, en se basant sur la définition d'une valeur de probabilité d'occurrence pour un peptide donné dans une digestion à la pepsine. En combinaison avec la précision élevée sur la mesure de masse, ce seul critère permet une identification efficace des peptides, sans devoir recourir à une validation par MS/MS systématique. Cette méthode a été mise en application pour l'étude des régions de liaison dans les complexes eIF3i :b et eIF3i :g. Dans les deux cas, 3i a été surexprimée sous sa forme complète. Au contraire, pour 3b et 3g, seul un segment partiel de la protéine native, contenant le domaine présumé d'interaction a été surexprimé. La liste de référence des peptides présents permet une excellente couverture de séquence et une forte superposition entre séquences adjacentes, ce qui assure une élucidation de la structure avec une bonne résolution spatiale. Pour la liaison entre 3i et 3b, les régions d'interactions qui sont apparues dans des conditions en solution proches des conditions physiologiques sont cohérentes avec la structure proposée par une autre équipe au cours de la thèse, apportant des informations complémentaires à celles issues de la structure cristallographique dérivée de la phase solide. Pour la liaison entre 3i et 3g, la région d'interaction a été étudiée en l'absence de toute donnée structurale à l'échelle atomique pour 3g. Les résultats apportent une vision nouvelle sur la formation du complexe entre 3g et 3i, et pour la première fois les régions de liaisons exacts ont été mis en évidence<br>The eukaryotic initiation factor 3 (eIF3) complex plays a core role in the interaction network among several eIFs that assemble on the 40S ribosomes and participate in the different reactions throughout the translation initiation pathway. The Saccharomyces cerevisiaea eIF3 complex comprises five subunits, all of which are the core subunits of the mammalian eIF3 complex consisting of 13 subunits. Attempts to decipher its tridimensionnal structure are under way. A first path to study the structure of this complex is to complete the identification of binding regions, few of which are currently known. Recently, the interaction region between eIF3i and extreme C-terminal domain of eIF3b has been obtained through NMR and crystal structure. On the other hand, the interaction region between 3i and 3g, although located to the N-terminal domain of 3g still remains to be defined. Hydrogen/deuterium exchanges (HDX) have been developed for a long time and are widely used for structural studies of proteins and multiprotein complexes. It is commonly analyzed using mass spectrometry. The most classic standard HDX-MS approach consists in making a mass measurement of deuterium-labelled peptides from an enzymatic digestion of the protein of interest to determine the level and rate of deuterium incorporation. In this study, a high performance 7 T FT-ICR mass spectrometer was used in combination with nanoLC separation to acquire highly accurate HDX-MS data. The precision on the mass measurement of FT-ICR MS is by itself not sufficient to unambiguously identify peptides from a pepsin digest due to the lack of pepsin specificity. We therefore developed a statistical approach for peptide identification, based on a probability of occurrence value of a given peptide within a pepsin digest. In combination with high mass accuracy, this method allows efficient identification of the peptides, without additional need of MS/MS verification. This method has been applied on the study of the binding regions in the complexes of eIF3i:bC3 and eIF3i:gC1ΔC. Peptide reference lists with high sequence coverage and rich sequence superposition ensured structure elucidation with high spatial resolution. For the binding of 3i and 3b, the detailed interaction regions were unveiled for proteins in the solution phase which resembled the physiological condition and were coherent with the reported protein structure, thus provided complimentary information to the crystallographic structure in solid phase. For the binding of 3i and 3g, the interaction regions were studied with the absence of any atomic structural information of 3g. This provides significant insights of the complex formation of 3i and 3g, and for the first time the precise binding regions were successfully revealed

Nous avons choisi certains ligands suivant des objectifs très précis et développé leur chimie de coordination afin d'obtenir des propriétés magnétiques spécifiques.<br /> La voie de synthèse par méthode solvothermale a permis de caractériser des complexes originaux à base de macrocycles. Deux complexes dinucléaires à base de CoII et FeIII où les deux centres métalliques sont pris en « sandwich » entre deux unités calixarènes ont été ainsi isolés. Ces dimères ont été utilisés comme brique moléculaire de départ pour l'élaboration de cluster à plus haute nucléarité. Un complexe associant six ions CoII a pu être isolé et caractérisé.<br /> La synthèse de nouveaux de ligands de type base de Schiff résultant de la condensation de l'acetylacétone avec différents acides aminés a été développée et leur complexation avec différents éléments de la première série de transition a été étudiée. Ainsi, un trimère à base de CuII a été caractérisé et ses propriétés magnétiques ont été modélisées par une série de calculs ab initio. Cette étude théorique a été complétée par un ensemble de mesures RPE en solution en fonction de la température.<br />Enfin, la chimie de coordination de ligands de type β-dicétone, en particulier l'acétylacetonate, avec différents métaux (3d et 4f ) a été revisitée. Ce ligand et certains de ses dérivés permettent de contrôler la nucléarité d'édifices moléculaires à base d'ions 3d et 4f. L'utilisation d'une brique moléculaire préformée de type cubane a permis d'isoler un complexe heptanucléaire de type « double cubane » à base de NiII qui présente les caractéristiques d'une molécule-aimant. En jouant sur l'effet de l'encombrement stérique du ligand, trois familles de complexes polynucléaires Ln5, Ln8 et Ln9 ont été obtenues par auto-assemblage suivant les méthodes de la chimie douce à base d'ions 4f. Des études structurales, magnétiques et une analyse approfondie des caractéristiques spectroscopiques des complexes à base d'europium(III) ont été réalisées. Une étude complète a été menée sur une famille de complexes Y9 dopés en europium (Y9-xEux).

During this work, several ligands with intrinsic properties have been chosen and their chemistry has been developed in order to get specific magnetic behavior. Solvothermal syntheses allowed the characterization of several complexes based on macrocycles. Two dinuclear CoII and FeIII complexes were isolated. Their molecular structures are built from metal ions located like in a "sandwich" between two calixarenes units. These complexes considered as molecular starting blocks were used for the synthesis of clusters with higher nuclearity. An original complex associating six CoII ions has been then isolated. The complexation of Schiff base ligands resulting from the condensation of the acetylacetone with various amino acids has been performed. From one of these original ligands, an almost perfect triangular trinuclear CuII complex has been then isolated and fully characterized. Ab initio calculations have been carried out in order to explain its magnetic properties. Finally, the complexation of β-diketone based ligands with various metals was explored. It has been then possible to control the nuclearity of molecular buildings block. The use of a preformed brick (cubane type) gave an access to a heptanuclear NiII complex. This latter presents the characteristics of a single molecule magnet. The last stage of these researches was to modify the steric effect of these β-diketone ligands in order to control the final cluster size. Three families of polynuclear 4f complexes (Ln5, Ln8 and Ln9) were obtained. Structural, magnetic studies and detailed analysis of the spectroscopic characteristics of these europium(III) complexes were achieved. A completely original work has been carried out on Y9 clusters doped with Eu (Y9-xEux) by linking molecular structures with luminescence properties

Résumé :

Utiliser des milieux nanostructurés pour confiner la lumière permet d'augmenter l'interaction entre un émetteur et le rayonnement électromagnétique. Dans cette thèse, nous utilisons un formalisme classique (présenté au Chap. 1) pour décrire cette interaction dans différents contextes, qui peuvent être regroupés en deux parties (respectivement Parties II et III). <br /> Dans un premier temps, nous étudions l'apparition de modes localisés en champ proche de structures complexes. Nous nous intéressons à deux différents types de structures: des nanoantennes d'or et des films d'or désordonnés. Nos résultats nous permettent de discerner les modes radiatifs et non-radiatifs. Nous introduisons le concept de Cross Density Of States (CDOS) pour décrire quantitativement la cohérence spatiale intrinsèque associée à la structure modale d'un milieu complexe. Nous démontrons ainsi une réduction de l'extention spatiale des modes au voisinage de la percolation électrique des films d'or désordonnés. <br /> Nous nous intéressons ensuite à des milieux fortement diffusants. En éclairant de telles structures par une source cohérente, on obtient une figure d'intensité complexe appelée speckle. Nous utilisons une méthode diagrammatique pour démontrer une corrélation négative entre les figures de speckle réfléchie et transmise à travers une tranche dans le régime mésoscopique. Nous nous intéressons ensuite à la corrélation C0, qui apparait lorsque la source est enfouie dans le milieu. Nous proposons une démonstration générale de l'égalité entre la corrélation C0 et les fluctuations normalisées de la LDOS, et soulignons le rôle fondamental des interactions de champ proche. Finalement, nous observons numériquement le régime de couplage fort entre un diffuseur résonnant et un mode localisé d'Anderson au sein d'un milieu désordonné 2D.

Les protéines de la famille L7Ae sont les constituants de nombreuses RNP essentielles. Chez les vertébrés, les particules snoRNP C/D et H/ACA sont impliquées dans la biogenèse des ribosomes, la UsnRNP U4 dans l'épissage des pré-ARNm, le complexe télomérase dans la réplication des télomères, et les mRNP SECIS dans la traduction des sélénoprotéines. Comme c'est le cas pour la majorité des RNP eucaryotes, leur assemblage, sous forme d'entités fonctionnelles, ne constitue pas un processus autonome et requiert l'intervention de facteurs spécialisés. En basant notre étude sur l'assemblage des snoRNP C/D, dans l'organisme modèle Saccharomyces cerevisiae, et en utilisant des approches de biologie moléculaire, de biochimie et de génétique, nous avons entrepris de caractériser ces événements. Nos travaux ont contribué à identifier un ensemble de protéines, agissant de façon coordonnée au sein d'une machinerie conservée entre la levure et l'homme. Cette dernière est composée de deux principales sous-unités : (i) Rsa1p/NUFIP, une protéine plate-forme, qui interagit avec certaines protéines de la famille L7Ae et facilite l'assemblage des RNP, (ii) le complexe R2TP (Rvb1p/TIP49, Rvb2p/TIP48, Pih1p/PIH1, Tah1p/SPAGH), qui pourrait opérer des remodelages conformationnels nécessaires à la formation des RNP matures. En plus de ces acteurs centraux, d'autres facteurs sont apparus intimement liés à ce mécanisme. La protéine Hit1p/TRIP3, interagit notamment avec Rsa1p/NUFIP et s'est avéré requise pour assurer sa stabilité chez la levure. La chaperonne HSP90, dont le rôle est prédominant chez l'homme, exerce son activité sur certains constituants des RNP. Enfin, la protéine Bcd1p/BCD1 pourrait être associée à cette machinerie dans le cadre spécifique de l'assemblage des snoRNP C/D<br>The L7Ae family proteins are essential components of many RNPs. In vertebrates, C/D and H/ACA snoRNPs are involved in ribosome biogenesis, the U4 snRNP in pre-mRNA splicing, the telomerase complex in telomeres replication, and mRNP SECIS in selenoproteins translation. Like most eukaryotic RNPs, assembly in functional entities is not an autonomous process and requires the intervention of specialized factors. Basing our study on the assembly of C/D snoRNP in the model organism Saccharomyces cerevisiae, and using approaches of molecular biology, biochemistry and genetics, we undertook to decipher these mechanisms. Our work has helped to identify a set of proteins, acting in a coordinated manner within a machinery conserved between yeast and human. This machinery consists of two major subunits: (i) Rsa1p/NUFIP, a platform protein that interacts with some proteins of the L7Ae family and facilitates the RNPs assembly, (ii) the R2TP complex (Rvb1p/TIP49, Rvb2p/TIP48, Pih1p/PIH1, Tah1p/SPAGH), which could induce conformational remodeling necessary for the formation of mature RNPs. In addition to these key players, other factors appeared closely linked to this mechanism. The Hit1p/TRIP3 protein interacts with Rsa1p/NUFIP and is required to ensure its stability in yeast. HSP90 chaperone, whose role is predominant in human, operates on some components of the RNPs. Finally, the Bcd1p/BCD1 protein is associated specifically with this machinery during C/D snoRNPs assembly

Résumé :

La chimie de coordination des éléments de transition (d ou f) permet de synthétiser de nombreux matériaux magnétiques moléculaires aux propriétés physiques variées (aimants moléculaires, molécules aimants, ou complexes à transition de spin). Nous avons ainsi développé la chimie de coordination de différents ligands afin d'obtenir des propriétés magnétiques spécifiques. La complexation de ligands fluorés du type bases de Schiff avec différents éléments de la première série de transition a été étudiée. Des complexes tétranucléaires de type cubane de cuivre (II) ou de nickel (II) ont été synthétisés afin d'accéder à des systèmes moléculaires étendus. Ces mêmes ligands ont conduit à l'obtention d'une roue neutre heptanucléaire [Co4III CO3II]. La complexation de lanthanides et de cuivre (II) par ces ligands a conduit à la formation de système nonanucléaires [Ln3Cu6] avec dans le cas du dysprosium (III) une molécule aimant à l'énergie d'activation de 25K. Des systèmes tétranucléaires [Ln1Cu3] ont également été caractérisés grâce à l'utilisation de tels ligands. Enfin, une étude magnétique complète par diffraction des neutrons polarisés et par calculs théoriques a été menée sur un complexe dinucléaire de cuivre (II). La voie de synthèse par méthode solvothermale nous a permis de caractériser des complexes de macrocycles originaux. Un complexe tétranucléaire de manganèse (II) et d'azacalis[4]arène a été étudié. Chaque unité à quatre manganèse est pontée par quatre ions chlorure conduisant à la formation de chaînes infinies dans le composé. Un complexe associant un calix[4]arène et un polyoxométallate de vanadium de type Lindqvist présente de façon exceptionnelle une valcence mixte (ViiiV5iv]. Enfin un complexe trinucléaire associant un platine (II) et deux manganèse (II) pontés par des thiocyanates a été carctérisé spectroscopiquement. Ce travail mené en collaboration avec l'Université de Montréal a permis d'effectuer ces mesures sous différentes contraintes (Pression, température). Un comportement tout à fait nouveau a ainsi été mis en évidence sur la luminescence du platine (II) sous l'effet de la pression

The human vitamin D receptor (hVDR) is a member of the nuclear receptor superfamily, involved in calcium and phosphate homeostasis; hence implicated in a number of diseases, such as Rickets and Osteoporosis. This receptor binds 1α,25-dihydroxyvitamin D3 (also referred to as 1,25(OH)2D3) and other known ligands, such as lithocholic acid. Specific interactions between the receptor and ligand are crucial for the function and activation of this receptor, as implied by the single point mutation, H305Q, causing symptoms of Type II Rickets. In this work, further understanding of the significant and essential interactions between the ligand and the receptor were deciphered, through a combination of rational and random mutagenesis. A hVDR mutant, H305F, was engineered with increased sensitivity towards lithocholic acid, with an EC50 value of 10 µM and 40 + 14 fold activation in mammalian cell assays, while maintaining wild-type activity with 1,25(OH)2D3. Furthermore, via random mutagenesis, a hVDR mutant, H305F/H397Y, was discovered to bind a novel small molecule, cholecalciferol, a precursor in the 1α,25-dihydroxyvitamin D3 biosynthetic pathway, which does not activate wild-type hVDR. This variant, H305F/H397Y, binds and activates in response to cholecalciferol concentrations as low as 100 nM, with an EC50 value of 300 nM and 70 + 11 fold activation in mammalian cell assays.
