To increase the knowledge about the biological properties of tomatoes, the antioxidant and antiproliferative activities of extracts of nine cultivars of Lycopersicon esculentum, as well as their chemical composition, were studied. The highest antioxidant capacity has been revealed in San Marzano Cirio 3 and Pomodoro Giallo cultivars, both in hydrophilic and lipophilic fractions, while San Marzano, Corbarino di Corbara, and Pomodoro Giallo exhibited the best radical scavenger activity in methanolic fraction. As regards the antiproliferative activity, the median inhibition concentrations of the lipophilic extracts ranged from 272.3 (Pomodoro Giallo) to 364.4 (Corbarino di Corbara) mg dried extract L-1 on Hep-G2 and from 324.6 (San Marzano Cirio 3) to 455.4 (Nero di Sicilia) on Caco-2. The methanolic extracts were more active on Caco-2 than Hep-G2, while the hydrophilic extracts were not active. These biological properties could be ascribed to the identified carotenoids and phenolic acids as well as to a pull of minor compounds exerting their synergistic effect.

The use of ecofriendly strategies, such as the use of Plant Growth Promoting Bacteria, to improve the yield and quality of crops has become necessary to satisfy the growing demand of food and to avoid the use of chemical fertilizers and pesticides. In this study, we report the effects of an innovative microbial inoculation technique, namely Effective Microorganisms (EM), compared with traditional approaches, on productivity and nutritional aspect of four tomato varieties: Brandywine, Corbarino Giallo, S. Marzano Cirio 3, S. Marzano Antico. Results showed an increase of plant productivity as well as an enhanced antioxidant activity mainly in San Marzano Antico and Brandywine varieties treated with EM technology. Moreover, the polyphenol and carotenoid contents also changed, in response to the plant treatments. In conclusion, the application of EM® technology in agriculture could represent a very promising strategy in agricultural sustainability.

There is a strong need to develop eco-sustainable agricultural techniques to improve crop yields while preserving biomolecule contents and reducing the adverse environmental impact of agro-chemicals. The use of microorganisms in agriculture represents an attractive and innovative solution. Herein, a chemical study on the nutritional and sensory qualities of San Marzano Cirio 3 (SMC3), Corbarino (CO) and Brandywine (BW) tomato varieties cultivated with and without effective microorganisms (EM) is reported. LC–MS analysis of the methanolic extracts allowed for the identification of 21 polyphenol derivatives. In different proportions among the studied varieties, the two main polyphenols were rutin and naringenin chalcone; the latter was isolated and chemically identified by complementary HR-ESIMS/MS and NMR methods. SMC3 and CO were richer in naringenin chalcone. Conversely, BW showed higher proportions of rutin; however, in all cases, the relative amounts of the two polyphenols considered together increased over the other minor components after the EM treatment. The qualitative and quantitative HPLC analyses of taste-active compounds (aspartic acid, glutamic acid, AMP and GMP) revealed a significant difference in aspartic and glutamic acids and ribonucleotide contents according to the cultivation condition (±EM), particularly in BW. This study provides chemical data in support of the use of EM green technology for the cultivation of edible agricultural products, such as tomato preserves, and may even improve nutritional and sensory qualities while safeguarding the environment.

According to an account which had until recently gone unnoticed, at the beginning of the 14th century and despite belonging to the Kingdom of Navarre, the village of Goizueta was ascribed to the Kingdom of Castile intus Ypuzcoam. Within the context of the shaping of the border between the northwest of the Kingdom of Navarre and the Kingdom of Castile, this paper aims to track down historical information about the village of Goizueta and to compare its situation with that of other villages in the same mountainous area to reveal the impact of factors such as population, geography, communications, the dissolution of the noble manor of Juan Corbarán de Leet and the expansion of the domain of the Collegiate Church of Roncesvalles. RESUMEN La villa de Goizueta, a tenor de una noticia que hasta la fecha había pasado desapercibida, se atribuía a comienzos del siglo xiv al reino de Castilla «intus Ypuzcoam» a pesar de su pertenencia al reino de Navarra. En el contexto de la configuración de la frontera entre los reinos de Navarra y Castilla en su tramo noroccidental, en este trabajo se plantea rastrear las noticias históricas sobre la localidad de Goizueta y comparar su situación con la de otras villas de ese mismo espacio montañoso para conocer la incidencia de factores como el poblamiento, la geografía, las comunicaciones, la disolución del señorío nobiliario de Juan Corbarán de Leet y la ampliación del dominio de la colegiata de Roncesvalles. Colorectal cancer represents the third most common cancer and the fourth cause for cancer death in both sexes worldwide. Different epidemiological and observational evidences strictly correlated the risk of colorectal cancer to lifestyle, especially to diet. Numerous studies have investigated the effect of antioxidant substances derived from food but data on lycopene or tomato extracts are still rare. This project is based on comparing the effects of extracts (total and lipophilic fractions) obtained from fresh tomato of one tomato cultivar (corbarino) versus another tomato variety (tangerino), this latter known to have lycopene already in bioactive isoform. Using two colorectal carcinoma cell lines we previously investigated the ability of tomatoes to inhibit cancer cell growth and proliferation and hypothesized a selective action on cancer cells and a lack of an effect on non-cancer cells (normal human fibroblasts). We noticed a major effect of tangerino tomato extracts, particularly of total fraction. These data made us hypothesize a possible effect of tomato extract on cell cycle. We analyzed cell cycle progression by flow cytometry. Data obtained showed that there are not great differences between treatment and control, but we noticed different peaks in sub G0/G1 phase, suggesting a possible cellular death via apoptosis. At the molecular level, we found variation in the expression of different proteins (RBL1, RBL2, pAKT, p21cip1, p27kip1, etc) involved in different cellular mechanisms. Based on the known anti-inflammatory effect of lycopene, we also performed a western blot for IL6 and IL10 to understand if tomato extracts have an impact on the inflammation process. Data showed a reduction of IL6 and a small increase of IL10 levels, compatible with an anti-inflammatory action.

Colorectal cancer is one of the most common causes of cancer death in the Western world. Most of the colon tumors are sporadic and develop somatically in epithelial cells. Apart from genetic factors, nutritional factors can markedly affect tumor development. In particular, while a high intake of red meat and animal fat is considered key points predisposing to colorectal cancer development, epidemiological studies often observed an inverse correlation between tomato and tomato product (sauce, paste) consumption and colon cancer risk. Tomato antioxidant bioactive molecules such as carotenoids and polyphenols could be responsible, at least in part, for the healthy effect. Here we analyzed the effect of total lipophilic extracts of a Southern Italy tomato variety, Corbarino and its processed form, Corbarino sauce, on two in vitro model of human colorectal adenocarcinoma cell lines, Colo–320 and SW-480, characterized by different aggressiveness. Our results support the hypothesis of a role for this variety of tomato in the inhibition of some features involved in the neoplastic advancement. The treatment with tomato extracts affected cancer cell ability to grow both in adherence and in semisolid medium, reducing also cell migration ability as highlighted after 24 hrs and more relevant results were gained after 72 hrs of incubation. Moreover, the most effective results were obtained with Corbarino sauce extract. No toxic effects were observed on non-tumoral cells, Human Skin Fibroblasts (HSFs). The observed inhibition of cancer cell growth and aggressiveness is associated with a negative regulation of cell cycle progression as pointed out by the increased expression levels of pRb/p105, p107 and pRb2/p130 while p21-Cip1 and p27-Kip1 expression levels decreased. The extent of antineoplastic effects, furthermore, seem to be correlated with the antioxidant activity of the two tomatoes form. Our data indicate that Corbarino tomato and its processed form intake might be further considered as nutritional support not only in cancer prevention, but also for cancer patient diet
