Queen Helen Nemanjić (1250–Brnjaci near Zubin Potok, February 8, 1314) was a Serbian medieval queen and consort of King Stefan Uroš I (r. 1243–1276), the fifth ruler of the Serbian Nemanide dynasty. She was the mother of the kings Stefan Dragutin and Stefan Uroš II Milutin. Today, she is known as Helen of Anjou (Jelena Anžujska in Serbian) although her real name was most probably Heleni Angelina (Ελένη Aγγελίνα). She was the founder of the Serbian Orthodox monastery of Gradac as well as four Franciscan abbeys in Kotor, Bar, Ulcinj, and Shkodër. Together with her sons, Kings Stefan Dragutin and Stefan Uroš II Milutin she helped renovation of Benedictine abbey of Sts. Sergius and Bacchus near Shkodër on Boyana river in present-day Albania. After the death of her husband, she ruled Zeta and Travunija until 1306. She was known for her religious tolerance and charitable and educational endeavors. She was elevated to sainthood by the Serbian Orthodox Church. Along with Empress Helen, the wife of Serbian Emperor Stefan Uroš IV Dušan, Queen Helen was the most frequently painted woman of Serbian medieval art. Six of her portraits can be found in the monumental painting ensembles of the Serbian medieval monasteries of Sopoćani, Gradac, Arilje, Đurđevi Stupovi (Pillars of St. George), and Gračanica, as well as on two icons and one seal. Queen Helen is also the only female Serbian medieval ruler whose vita was included in the famous collection of the “Lives of Serbian Kings and Archbishops” by Archbishop Danilo II, a prominent church leader, warrior, and writer.

This article explores Washington's annotated copy of the Constitution and the Acts of Congress (hereafter called the Acts of Congress to remain consist with the label Washington had placed on the front cover) to reveal new insights into his constitutional interpretation. Held in a private collection until 2012, this article is the first to examine Washington's notations in the Acts of Congress for their value as statements about political authority. Washington's comments in the margins of his volume suggest an evolving view of presidential power and constitutional limitations on the executive branch as early as January 1790. His margin notes on the Acts of Congress served as blueprint for his defense of presidential authority and the expansion of the executive branch in the 1790s. Finally, the annotated Acts of Congress inserts Washington's ideas about the presidency into the debate surrounding originalism by revealing how his analysis of the language evolved to meet the demands of governing, leading him to reject the delegates' intent for Article II, Section 2 of the Constitution.

The study examines the nexus of the ruling party's slogans in the state of Oceania, "war is peace, ignorance is strength, freedom is slavery" in George Orwell's novel 1984 to the present societies in developing countries, using the lens of Marxism. Sufyan Al-Dmour states; Three different classes are present in the depicted novel: the ruling class, which is echoed by the Inner Party; Outer Party represents the middle class, the Proles consist of the working class (SufyanAl-Dmour, 2020). The class system is dominating all over the world, specifically in developing countries. MacCartney and Zaidi argue; It was the military-bureaucratic control of a few influential figures, with the three classes which kept what can be called Pakistan's political setup. (McCartney, Zaidi, 2019). Common people in 1984 were severely penalized for who exerted to think or act differently. The pertinence of the slogan "freedom is slavery" in 1984 is still dominant in developing societies.

On January 15, 1993, President George Bush transmitted to the Senate for advice and consent to ratification the Treaty Between the United States of America and the Russian Federation on Further Reduction and Limitation of Strategic Offensive Arms (the START II Treaty), signed at Moscow on January 3, 1993. In his letter of transmittal, President Bush discussed the importance of the Treaty, in major part as follows:The START II Treaty is a milestone in the continuing effort by the United States and the Russian Federation to address the threat posed by strategic offensive nuclear weapons, especially multiple-warhead ICBMs [intercontinental ballistic missiles]. It builds upon and relies on the Treaty Between the United States of America and the Union of Soviet Socialist Republics on the Reduction and Limitation of Strategic Offensive Arms (the START Treaty) signed at Moscow on July 31, 1991. At the same time, the START II Treaty goes even further than the START Treaty.

The present study documents the fragmented publication history of Malacostraca Podophthalmata Britanniæ by William Elford Leach, illustrated with coloured figures of all the species by James Sowerby. This work was originally proposed to consist of 12 or 14 numbers. One number was to be published every two months between the years 1815 and 1818. Although this was increased to 19 numbers its publication by James Sowerby halted at number 17 in 1820. In that year Leach had a complete nervous breakdown and, although he eventually recovered, he was retired from his post at the British Museum in 1822. Although Leach was optimistic and set out plans to complete Malacostraca, he died from cholera in 1836 near Genoa, Italy, with the work unfinished. During the early 1870s fortuitous events occurred that would lead to the publication of numbers 18 and 19 and the completion of the work. At that time William Sowerby began negotiating with Bernard Quaritch, a London publisher, for disposal of old stock from the Sowerby publishing house including Malacostraca. George Brettingham Sowerby the younger, an established naturalist and highly skilled illustrator, proposed that the Malacostraca should be updated and he prepared Nos. 18 and 19 for publication. These last two volumes of Malacostraca were finally made available by Quaritch in November 1875. The authorship of Nos. 1–17 has never been in doubt and this is the work of Leach with illustrations by James Sowerby. Among the taxa illustrated in Nos. 18 & 19 however, are species which were not known to occur in British waters when Leach was working and as such the choice of Malacostraca illustrated in 1875 differed significantly from the original proposals. Consequently the 1875 supplement should therefore correctly be credited entirely to G.B. Sowerby II and cited as Sowerby, G.B. II in Leach, W.E. (1875). Finally, because copies of the Malacostraca are not generally available, all the magnificent plates illustrated by James Sowerby and George Sowerby II are reproduced here in full colour.

Highlights. In recent decades, the number of patients with diffuse, multivessel coronary artery disease has increased due to widespread use of endovascular methods of treatment, long-term conservative management of patients, and the expansion of indications for surgical revascularization. In order to improve the results of treatment, in 2007 academician Yu. L. Shevchenko developed and began using the method of stimulation of extracardiac angiogenesis – «YurLeon». The present article demonstrates the long-term results of supplementing coronary bypass surgery with this method in patients with diffuse coronary artery disease.Aim. To evaluate long-term results of coronary bypass surgery supplemented by extracardiac myocardial revascularization.Methods. The prospective, randomized study included 205 patients with diffuse, multivessel coronary artery disease who were treated at the St. George Thoracic and Cardiovascular Surgery Clinic, National Medical and Surgical Center named after N.I. Pirogov. The majority of patients were men – 75% (n = 153), whereas women accounted for 25% (n = 52), all patients were 60 to 78 years old (67.3±5.74 years). The group I consisted of patients with coronary bypass surgery supplemented with the «YurLeon» technique, the group II consisted of patients with coronary bypass grafting only. All patients provided consent to participate in the study. The clinical condition of patients, echocardiography, myocardial scintigraphy and coronary angiography data, quality of life indicators (the 36-Item Short Form Health Survey) were assessed.Results. In the long-term postoperative period (36 months), the group I, compared with the group II, presented with a statistically significant decrease in the functional class of angina pectoris (1[1–2] vs. 2[1–2], p = 0.02); fewer adverse events: myocardial infarction (4.5% vs. 11.6%, p = 0.011), recurrent angina pectoris (10% vs. 17.9%, p = 0.038); an increase in the left ventricular ejection fraction (59.0±5.15% vs. 53.2±6.13%, p = 0.001); a decrease in the Summed Rest Score (SRS) (4.5±3.5 vs. 8.4±5.7, p = 0.012) according to myocardial scintigraphy with technetril; a decrease in the Summed Thickening Score (STS) (6.1±3.3 vs. 10.7±2.9, p = 0.012); a decrease in the hibernating myocardium (3.5±2.2% vs. 7.4±5.1%, p = 0.03); additional extracardiac sources were found in the group I according to coronary graft angiography, there were no statistically significant differences between the groups by occluded autogenous venous bypass grafts (in the group I – 23 out of 46 venous conduits (49%), in the group II – 30 out of 49 venous conduits (51%), p = 0.089); The groups differed in terms of quality of life (the 36-Item Short Form Health Survey): physical functioning (87.3±10.32 vs. 75.3±14.15 points, p = 0.001); role-based functioning due to physical condition (90.1±10.50 vs. 79.7±7.01 points, p = 0.012); pain (93.10±6.37 vs. 80.5±5.21 points, p = 0.03); the total physical component of health (57.9±5.4 vs. 45.8±6.1 points, p = 0.012).Conclusion. We have accumulated 15 years of experience in conducting complex myocardial revascularization in patients with coronary artery disease and diffuse coronary atherosclerosis, which allowed us to prove the effectiveness and safety of the YurLeon technique.

Abstract The Eocene nephelinite from Księginki quarry (SW Poland) contains five types of clinopyroxene phenocrysts varying by texture and chemical composition. Type I phenocrysts are formed of Mg-rich (mg# = 0.93–0.88) homogenous cores, patchy mantle and zoned rims. Abundant type II is less magnesian (mg# = 0.65–0.88) and consists of spongy or spongy-patchy core surrounded by zoned rims, whilst in type III (mg# = 0.69–0.84), the cores are massive but patchy. The mg# of cores of type IV phenocrysts is slightly lower than that of type I (0.79–0.89), but its cores are either massive or patchy. Type V is very scarce and consist of relatively Mg-poor (mg# = 0.75–0.77) core enveloped by nonpatchy, sometimes zoned mantle and zoned outer rim. Chemical composition of type I and type IV cores suggests that they are xenocrysts introduced into the nephelinite from disintegrated peridotite and clinopyroxenitic xenoliths, respectively. Type V is also of xenocrystic nature, but its source rock was significantly more evolved than mantlederived ones. Types II and III are possibly cognates from the host nephelinite or a melt related to the nephelinite. All the types of phenocrysts suffered from disequilibrium with the nephelinitic (or proto-nephelinitic) melt or dissolution during adiabatic uplift. Linear variation in chemical composition of phenocrysts of Księginki nephelinite suggests its evolution because of fractional crystallisation, without significant influence of other differentiation processes.

Objective: Phase II/III randomized clinical trials (RCTs) are vulnerable to many types of bias beyond randomization. Insights into the reporting quality of RCTs involving migraine patients treated with monoclonal antibodies targeting the calcitonin gene-related peptide system (anti-CGRP MAbs) are currently lacking. Our aim was to analyze the reporting quality of phase II/III RCTs involving migraine patients treated with anti-CGRP MAbs. Methods: A systematic search was performed on the PubMed and EMBASE databases, according to PRISMA guidelines, for relevant RCTs in either episodic or chronic migraine prevention. Additionally, an adapted version of the 2010 CONSORT statement checklist was utilized. The ROBvis online tool was used to document the risk of bias. Results: From the initially identified 179 articles, we finally found 31 RCTs that were eligible for evaluation. The average CONSORT compliance was 88.7% (69.7–100%), while 93.5% (N = 29) of the articles had a compliance greater than 75%. Twenty-eight CONSORT items were reported in more than 75% of the articles. The average compliance of the analyzed RCTs was 93.9% for Galcanezumab, 91.3% for Fremanezumab, followed by 85.4% for Erenumab and Eptinezumab studies. Implementation of the ROB2 tool showed some concerning “missing information” arising from the inadequate reporting. Specifically, 50% of the studies (N = 16) were categorized as having inadequate information regarding the randomization process. Conclusions: Adequate reporting quality was disclosed in the evaluated RCTs with anti-CGRP MAbs in migraine prevention. However, some methodological issues need to be highlighted to be addressed in future studies assessing the efficacy of new molecules targeting CGRP or other candidate pathways implicated in migraine pathophysiology.

Objectives. To evaluate the reporting quality of published randomized clinical trials (RCTs) in the Tai Chi literature following the publication of the CONSORT guidelines in 2001.Data Sources. The OVID MEDLINE and PUBMED databases.Review Methods. To survey the general characteristics of Tai Chi RCTs in the literature, we included any report if (i) it was an original report of the trial; (ii) its design was RCT; (iii) one of the treatments being tested was Tai Chi; and (iv) it was in English. In addition, we assessed the reporting quality of RCTs that were published between 2002 and 2007, using a modified CONSORT checklist of 40 items. The adequate description of Tai Chi interventions in these trials was examined against a 10-item checklist adapted from previous reviews.Results. The search yielded 31 Tai Chi RCTs published from 2002 to 2007 and only 11 for 1992–2001. Among trials published during 2002–2007, the most adequately reported criteria were related to background, participant eligibility and interpretation of the study results. Nonetheless, the most poorly reported items were associated with randomization allocation concealment, implementation of randomization and the definitions of period of recruitment and follow-up. In addition, only 23% of RCTs provided adequate details of Tai Chi intervention used in the trials.Conclusion. The findings in this review indicated that the reporting quality of Tai Chi intervention trials is sub-optimal. Substantial improvement is required to meet the CONSORT guidelines and allow assessment of the quality of evidence. We believe that not only investigators, but also journal editors, reviewers and funding agencies need to follow the CONSORT guidelines to improve the standards of research and strengthen the evidence base for Tai Chi and for complementary and alternative medicine.

BackgroundThe findings of the Clinical Antipsychotic Trials of Intervention Effectiveness (CATIE) study and the Cost Utility of the Latest Antipsychotic Drugs in Schizophrenia Study (CUtLASS) called previous trials of antipsychotics into question, including pre-licensing trials. Concerns regarding methodological robustness and quality of reporting increased. This systematic review aimed to examine the quality of reporting of phase II and III trials for new antipsychotics in the aftermath of the CATIE and CUtLASS studies.MethodElectronic searches were conducted in EMBASE, Medline and Cochrane databases and also ClinicalTrials.gov for antipsychotic trials (published between January 2006 and February 2012). Phase II and III randomized controlled trials (RCTs) for iloperidone, asenapine, paliperidone, olanzapine, lurasidone and pomaglumetad methionil were selected for schizophrenia and schizoaffective disorder. The reporting of the methodology was evaluated in accordance with Consolidated Standards of Reporting Trials (CONSORT) guidelines.ResultsThirty-one articles regarding 32 studies were included. There was insufficient reporting of design in 47% of studies and only 13% explicitly stated a primary hypothesis. Exclusion criteria were poorly reported for diagnosis in 22% of studies. Detail regarding comparators, particularly placebos, was suboptimal for 56% of studies, and permitted concomitant medication was often not reported (19%). Randomization methods were poorly described in 56% of studies and reporting on blinding was insufficient in 84% of studies. Sample size calculations were insufficiently reported in 59% of studies.ConclusionsThe quality of reporting of phase II and III trials for new antipsychotics does not reach the standards outlined in the CONSORT guidelines. Authors often fail to adequately report design and methodological processes, potentially impeding the progress of research on antipsychotic efficacy. Both policymakers and clinicians require high quality reporting before decisions are made regarding licensing and prescribing of new antipsychotics.

Background and objectives: Our research aimed to evaluate the quality of reporting of randomized controlled trials (RCTs) linked to rotator cuff (RC) tears. The present study analyzed factors connected to the quality of the RCTs and trends in the quality of reporting through time. Materials and Methods: The online databases used to search all RCTs on the topic of RC surgery completed until March 2020 were PubMed and Ovid (MEDLINE). The quality of reporting was evaluated using the modified Coleman methodology score (MCMS) and the consolidated standards of reporting trials (CONSORT). Results: The online search found 957 articles. Finally, 183 studies were included in the quantitative synthesis. A total of 97 (53%) of 183 studies had a level of evidence I and 86 (47%) of 183 studies had a level of evidence II, according to the Oxford Center of Evidence Based Medicine (EBM). A statistically significant difference in MCMS between articles written before 2010 and articles written after 2010 was found. Articles written after 2010 had, on average, the highest Coleman score. The average number of CONSORT checklist items for each article across all analyzed RCTs was 21.67. The 37 studies completed up to 2010 averaged a number of checklist items of 19.97 and the studies completed between 2011 and 2019 averaged a number of checklist items of 22.10. A statistically significant difference in the number of checklist items between articles written before 2010 and articles written after 2010 was found. Articles written after 2010 had on average more checklist items. However, low correlation (0.26) between the number of checklist items for each article and the respective Coleman score was found. On the other hand, articles with the CONSORT diagram had a significantly high Coleman score. Conclusions: An improvement in the quantity and quality of RCTs relating to RC surgery over the analyzed period was found.

Cephalopods are a remarkable group for studies on sexual selection, due to their complex reproductive behavior, pronounced sexual dimorphism, polygamous behavior, as well as cumulative evidence of pre- and postcopulatory male-male competition and female choice in the group. is paper provides a review of sexual selection studies on cephalopods, with an emphasis on the loliginid squid mating system. In this system, males may adopt alternative reproductive tactics (ART) and display male intrasexual dimorphism, with each morph generally exhibiting distinct behaviors and ejaculate traits. Consort males typically transfer ejaculates to the female mantle cavity, while sneaker males typically transfer ejaculates to the female buccal region, where a sperm storage organ is located. Each site provides distinct fertilization environments for the ejaculates of each morph, differing in the availability of eggs, gamete protection, and timing between mating and fertilization. Accordingly, the ejaculates of each morph show putative adaptations for each site, differing in size, morphology, sperm release duration, and sperm behavior. Although solid evidence is lacking for most loliginids, cryptic female choice may occur in this system through, for example, spermatangia removal or egg string manipulation during fertilization. In addition, recent evidence for one loliginid species indicates that adoption of male ART is ontogenetic and may represent two alternative pathways: from immature males (i) directly to consort phenotype or (ii) going through a sneaker phase, then switching to consort morph. The second pathway would, therefore, imply striking change in behavior, physiology, and ejaculates. Moreover, data on the age of this species suggests that the adoption of sneaker tactic early in life may be associated with extending the male mating period by 25%. With such a peculiar system, loliginids should prove useful not only for comprehending the evolution of female promiscuity and ART in cephalopods, but also as an emerging nonvertebrate and nonarthropod model for testing sperm competition models.

Ladislaus II Jagiełło (1386–1434). Ladislaus II Jagiełło is the founder of the Jagiellonian dynasty that had ruled over Poland and the Grand Duchy of Lithuania (until 1572), Bohemia (1471–1526) and Hungary (1440–1444, 1490–1526). A Grand Duke of Lithuania from 1377, and from 1386 a king of Poland and lord of Lithuania, which he ruled jointly with his cousin Witold (Vytautas), the son of Kęstutis. Five medieval portraits of Jagiełło survive, four of which date from the period of his reign in the Polish–Lithuanian state and one was executed posthumously. The earliest image, on Jagiełło’s Great Seal, was made in connection with his coronation as king of Poland (1386). Two portraits in the Holy Trinity Chapel at the Castle of Lublin (1418) are part of a wall paintings scheme commissioned by the monarch and executed by a team of painters brought from Ruthenia. Furthermore, the sumptuous tomb (before 1430) in Cracow was commissioned by the king. Its top slab bears an effigy of Jagiełło with his suggestively rendered countenance, which undoubtedly reflects the actual facial features of the elderly monarch. An image of the king represented as one of the Three Magi in a panel of an altarpiece in the tomb chapel of Casimir IV Jagiellonian, Jagiełło’s son and his successor on the Polish throne, dates from 1470. The chapel dedicated to the Holy Cross, erected at Cracow Cathedral, was in all likelihood commissioned by Casimir himself and his consort Elizabeth of Austria.

A small tile panel created by the Italian ceramic painter Francisco Niculoso (active 1498-1529 from the private collection of Ferdinand Saxe-Coburg-Gotha (1816-1885), king consort to Queen Maria II of Portugal (1819-1853), was in 1902 acquired by the Rijksmuseum. Although the ownership history of this Visitation panel is not fully documented, this essay provides a more comprehensive understanding of its historical narrative, by exploring the various contexts of acquisition and collecting. The panel’s journey from Lisbon to Amsterdam vividly illustrates the remarkable fluidity and permeability that defined the boundaries of art markets and museums around the turn of the twentieth century.

The paper analyzes the well-known historical works of George Akropolites and George Pachymeres from the viewpoint of their literary composition structure, lexica and narrative techniques used for describing events and personalities from Bulgaria and Serbia. An attempt is made to surpass the 'traditional' methodology and the usually applied standard approach that focuses almost exclusively to the search for the 'strong' historical facts in these literary works par excellence, in that way overlooking their strong literary character, careful expressions, sometimes evident and sometimes more subtle style differences whose purpose was to underline the focal points and the conclusions of the content of the stories - in short, the information and the stories about Bulgaria and Serbia by Akropolites and Pachymeres are analyzed in their entirety, viewed as the integral parts of the complex literary works, and not as isolated episodes, divided from the rest of the narrative. The ways in which George Akropolites depicted and described deaths of the Bulgarian rulers were studied as one of the best examples of his conscious endeavors to bring the literary style of his History in accord with his own judgments about the rulers of the neighboring country. Scenes of death, conspicuously frequent in Akropolites' relatively short historical work, play an important role in characterizing personalities, and the same principle which Akropolites uses for judging the Byzantines is applied to the Bulgarians as well: generally positive opinion about somebody's life and achievements is emphasized with a depiction of his death in a positive way (Ivan II Asen), and vice versa, Akropolites' 'villains' receive their horrible deaths as a deserved punishment for their bad deeds (Kalojan's death). On the other hand, Akropolites' scarce information about Serbs is supplemented with his depiction of the Serbian king Uros (1243-1276), and his stance after the death of John Batatzes from the funerary oration to the same emperor, which has been completely overlooked by the scholars until now. Akropolites confirms that the Empire of Nicaea and king Uros's Serbia were strongly connected, and that only after the death of the 'mighty' John Batatzes Serbian king allied himself with the ruler of Epiros, despotes Michael II Angelos. More complex in structure, language, narrative techniques and expressions than Akropolites' is without doubt the voluminous History by George Pachymeres. In the context of Pachymeres' depiction of the Bulgarians and Serbs, the chapters that describe Byzantine marital diplomacy regarding Bulgaria and Serbia are studied in details, with the emphasis placed equally on the literary analysis of the corresponding chapters, and on the concordance between the content and style, author's attitudes and expressions used, Pachymeres' intention and the form he had chosen, which all contribute in the end to the better understanding of the historical circumstances, as well. The second part deals with the testimonies of three Byzantine historians: George Acropolites, George Pachymeres and Nikephoros Gregoras, who provide us with different information concerning Theodore II Lascaris as co-emperor. While Acropolites refers to Theodore II as basileuj even before he actually came to the throne, Pachymeres and Gregoras claim that Theodore II was never his father?s coruler. Thus, it is the aim of this study to offer an explanation for the differences in the testimonies of the mentioned historians.

A life of service to others ended on March 26, 2009, when professor emeritus George A. Warp of the Humphrey Institute of Public Affairs of the University of Minnesota passed away at age 95. George was born on June 12, 1913, in Northfield, Ohio, and graduated from Bedford High School in Ohio. Prior to being associated with the University of Minnesota for the past 60 years, he graduated from Oberlin College, Case Western University, and Columbia University, earning degrees in political science, public administration, international administration, as well as law. George served briefly as a political science faculty member at the University of Minnesota, where he met and married his late wife, Lois, in 1940 before entering the U.S. Navy following the entry of the United States into World War II. His service in the Pacific theater led to his postwar appointment as a civilian advisor under General MacArthur in Japan from 1946–1948. Upon completion of that assignment, George returned to the University of Minnesota in 1948 as a professor of political science and served first as associate director and then director of the graduate program in public administration in the department's Public Administration Center until 1965 when the center became a self-standing unit of the College of Liberal Arts. He remained director through 1968 when the center was succeeded by the School of Public Affairs and recreated as the Humphrey Institute of Public Affairs in 1978 as a collegiate unit named as a memorial honoring the late vice president and Minnesota's senator. George served as a professor and chair of graduate admissions until his retirement in 1982.

Abstract Background: The addition of neoadjuvant platinums to standard chemotherapy regimens for triple negative breast cancer (TNBC) improves rates of pathologic complete response (pCR). Our prior trial combining carboplatin (CAR) with liposomal doxorubicin (DOX) for metastatic TNBC showed good response rates with minimal side effects and allows for higher platinum dosing. We hypothesized that the doublet of DOX+CAR is effective and tolerable in the neoadjuvant setting for TNBC and tumor genomics may identify biomarkers of complete response and actionable targets. Methods: A phase II single arm trial was conducted for patients (pts) diagnosed with stage II-III TNBC. Patients received 4 cycles of neoadjuvant carboplatin (AUC 5) and liposomal doxorubicin (30mg/m2) administered every 28 days, then underwent definitive breast surgery followed by 12 weeks of adjuvant paclitaxel 80 mg/m2 administered weekly. Primary and secondary clinical endpoints were rate of pCR and two year recurrence free survival (RFS) and overall survival (OS), respectively. Cardiac safety of the combination was assessed. Fresh residual tumor samples were obtained at time of surgery for generation of patient derived xenografts (PDX). Whole genome sequencing (WGS) and RNA sequencing will be performed to evaluate specific patterns of small-variant mutations and patterns of structural variants to identify which patients are likely to have pCR. Results: From 2/2015 to 6/2021, 62 pts were enrolled, 8 pts withdrew consent either prior to treatment or completion of neoadjuvant therapy and/or surgery. Median age of the cohort was 55.4 years. There was high participation by under-represented groups: 17.0% African American, 20.8% Asian, 13.2% Hispanic. Most histologies were invasive ductal carcinoma, but included apocrine, pleomorphic lobular, and metaplastic subtypes. 53 pts completed all 4 cycles of neoadjuvant DOX+CAR, followed by definitive surgery; 1 pt progressed after 1 cycle and proceeded to surgery thereafter. 30.2% (16) pts achieved pCR, and residual cancer burden (RCB) will be reported in patients with residual disease. Of the 53 pts who completed 4 cycles of treatment, 5 pts progressed (recurrence or death) within two years from time of surgery (4 distant and 1 local recurrence), yielding the 2-year recurrence-free-survival rate of 90.3% (95%CI 81.0%, 99.6%). The most common toxicities during DOX+CAR were grade 1 fatigue in 50 pts (92.6%), grade 1 anemia in 44 pts (81.5%), and grade 3/4 neutropenia in 16 pts (29.6%); these pts received GCSF support with subsequent cycles; febrile neutropenia occurred in 1 pt (1.9%) in this group. Only 10 pts (18.5%) had grade 1 alopecia, 5 pts (9.3%) had grade 2 alopecia, 2 pts (3.7%) had grade 3 thrombocytopenia. There were no delays in treatment due to cardiotoxicity or complications from surgical healing. Of the 37 pts who had residual disease, PDX was attempted in 21 pts, and 16 (76%) PDX were established, including those for 4 patients experiencing recurrence. WGS and RNA sequencing data from pre-treatment and residual disease samples will be analyzed and reported.Conclusion: 4 cycles of neoadjuvant DOX+CAR achieved rate of pCR similar to standard regimens and has good tolerability. Post chemotherapy PDX is feasible and may help identify targeted approaches for patients with resistant disease. These results warrant further evaluation of this combination for early stage TNBC. The arrival in Madrid in 1690 of the future queen consort, Maria Anna of Palatinate-Neuburg (1667–1740), second wife of the Spanish king Carlos II (1661–1700), also brought about the establishment of an unofficial palace faction known as the German Chamber. Since the members of this faction were part of Maria Anna’s household, the analysis of their social activities gives insight into court practices during the last decade of Carlos II’s reign. This article intends to shed light on the new networks and agency of members of the German Chamber at the Madrid court, in particular, that of the queen’s confessor, Gabriel Pontifeser di Chiusa.
