Texte intégralMorphogenesis is the process of reshaping single-cell zygotes to the final form of a developed animal. Embryonic gene patterning systems determine the body axes and lay down the spatiotemporal specification coordinates for cells. Gene patterning systems also affect the organization of cytoskeletal components in order to drive tissue morphogenesis. While much work was done to understand how AP and DV patterning independently control morphogenesis, little is known on how cross-patterning functions. We use the Drosophila embryo as a model system and focus on the process of tissue folding, a process that is vital for the animal since folding defects can impair neurulation in vertebrates and gastrulation in all animals which are organized into the three germ layers. Past work has shown that an actomyosin meshwork spanning the apical-medial side of prospective mesoderm cells and under the control of the embryo DV patterning plays a key role in mesoderm invagination. Nevertheless, both experimental and theoretical pieces of evidence have argued against apical constriction being the sole mechanism driving invagination. In this study, I have uncovered a lateral cell junctional network under the control of both AP and DV patterning. This contractile network generates tension along the apical-basal axis and within the tissue plane, 10-15 μm inside the mesoderm epithelium initiating lateral cell intercalation. Lateral forces in mesoderm cells seem to play a multivalent role in both driving mesoderm extension and invagination. Finally, by implementing 4D multi-view light-sheet imaging, infra-red femtosecond ablation to perturb the cytoskeleton, and optogenetics to synthetically control tissue morphology, this work shines new light on the origin and functions of a novel mechanism responsible for coordinated tissue elongation and folding
