Résumé :
Fatigue damage plays an increasingly important role in the design of various safety critical components that are exposed simultaneously to thermal and mechanical loads. Non-isothermal conditions, as these are to be found in turbine components, rocket engines, but also in high-speed machining tools makes the understanding even more complex. As the nature of those loading histories is mostly multiaxial, design engineers are interested in material models, which take into account the complexity of stress state and temperature history as well. The experimental validations of those models require specially designed test set-ups. The basic concepts of experimental techniques to perform non-isothermal, uniaxial fatigue tests will be described in general. Test systems, capable to simulate non-isothermal multiaxial stress states are presented. A new miniaturised electrothermalmechanical test rig, which allows testing of small specimens under complex thermomechanical loading conditions, will be discussed.

Résumé :
Film adaptation is a popular approach to game design, but it prioritizes blockbuster films and conventional “game-like” qualities of those films, such as shooting, racing, or spatial exploration. This leads to adaptations that tend to use the aesthetics and narratives of films, but which miss out on potential design explorations of more complex cinematic qualities. In this article, I propose an experimental game design method that prioritizes an unconventional selection of films alongside strict game design constraints to explore tensions and affinities between cinema and videogames. By applying this design method and documenting the process and results, I am able both to present an experimental set of videogame film adaptations, along with potentially generative design and development themes. In the end, the project serves as an illustration of the nature of adaptation itself: a series of pointed compromises between the source and the new work.

Résumé :
Purpose – The purpose of this paper is to validate methodologies for expedited multi-objective design optimization of complex antenna structures both numerically and experimentally. Design/methodology/approach – The task of identifying the best possible trade-offs between the antenna size and its electrical performance is formulated as multi-objective optimization problem. Algorithmic frameworks are described for finding Pareto-optimal designs using auxiliary surrogate models and two alternative approaches to design refinement: response correction techniques and co-kriging. Numerical and experimental case studies are provided to demonstrate feasibility of solving real-world and complex antenna design tasks. Findings – It is possible, through appropriate combination of the surrogate modeling techniques (both data driven and physics based) and response correction methods, to find the set of alternative designs representing the best possible trade-offs between conflicting design objectives, here, electrical performance and size. Design optimization can be performed at practically feasible computational costs. Research limitations/implications – The study demonstrates feasibility of automated multi-objective design optimization of antennas at low computational cost. The presented techniques reach beyond the commonly used design approaches based on parameter sweeps and similar hands-on methods, particularly in terms of automation, reliability and reduction of the computational costs of the design processes. Originality/value – Multi-objective design of antenna structures is very challenging when high-fidelity electromagnetic simulations are utilized for performance evaluation of the structure at hand. The proposed design framework permits rapid optimization of complex structures (here, MIMO antenna), which is hardly possible using conventional methods.

Abstract: The present work had as the main the objective, the isolation and selection of potential B-vitamin producer yeast and the study of fermentative process for B-vitamin production for feed application. Among 267 isolated strains, one was selected as extracellular thiamine producer and the strain Candida sp. LEB 130 as riboflavin and biotin producer. In the study of different carbon and nitrogen sources effect on the production of riboflavin by the screened yeast, including complex compounds, considerable results were obtained by using molasses, flaxseed oil and xylose. As for the study of pH, temperature and agitation of flasks for riboflavin production by Candida sp. LEB 130 yeast, using statistical design 23 as a tool, the highest riboflavin production occurred at pH 5.0 after 48 h of fermentation at 400C and 160 rpm of stirring. The highest productivity of riboflavin production (0.31 mg/mL.h) was obtained in the fermentation of yeast at pH 7.7, 400C and 100 rpm. As for the study of medium components for yeast fermentation for riboflavin production using factorial design 25, it was observed significant effect of sucrose, yeast extract Prodex LacÒ and the interaction between ZnSO4 and flaxseed oil in the limit of confidence of 90%. As for factorial design 24, without the addition of yeast extract Prodex LacÒ, the most significant factors were ZnSO4 and flaxseed oil in the limit of confidence of 90%. Taking into account that Candida sp. LEB 130 strain produces both riboflavin and biotin, a study of the simultaneous production of both vitamins was done. In the study of the medium compounds in the fermentation process of the strain for both vitamins production, sucrose and mineral salts were significant for riboflavin production, while sucrose, ammonium tartrate, ferric citrate and magnesium sulphate were significant for biotin production in the confidence level of 95%. Statistical designs experiments to evaluate the effect of culture medium components and fermentation conditions were important for the study of fermentation process of screened yeast Candida sp. LEB 130 and production of riboflavin and biotin. Riboflavin and biotin production by yeast were 155.7 mg/mL and 5,6mg/mL, respectively, which is 77.8- and 2.9-fold higher than at the moment of isolation, respectively. Nonetheless, it should be considered that this microorganism is a wild type, without any genetic engineering modification

Résumé :
The complexity of statistical models that are used to describe biological processes poses significant computational challenges in design of experiments. To address such challenges, in this thesis, new methods are developed in optimisation and approximate inference, and are applied in real-world experiments. The proposed methods enable practitioners to gain greater insight and understanding into the biological processes they are studying, and this is demonstrated by designing experiments to understand important biological processes in epidemiology and ecology such as the spread of infectious diseases and interactions between predator and prey in environmental systems.

Where and how does the brain represent complex rule sets? This thesis presents a series of three empirical studies that decompose representations of complex rule sets to directly address this question. An additional methodological study investigates the employed analysis method and the experimental design. The empirical studies employ multivariate pattern analysis (MVPA) of functional magnetic resonance imaging (fMRI) data from healthy human participants. The methodological study has been inspired by the empirical work. Its impact and application range, however, extend well beyond the empirical studies of this thesis. Questions of the empirical studies (Studies 1-3) include: Where are cues and rules represented, and are these represented independently? Where are compound rules (rules consisting of multiple rules) represented, and are these composed from their single rule representations? Where are rules from different hierarchical levels represented, and is there a hierarchy-dependent functional gradient along ventro-lateral prefrontal cortex (VLPFC)? Where is the order of rule-execution represented, and is it represented as a separate higher-level rule? All empirical studies employ information-based functional mapping ("searchlight" approach) to localise representations of rule set features brain-wide and spatially unbiased. Key findings include: compositional coding of compound rules in VLPFC; no order information in VLPFC, suggesting VLPFC is not a general controller for task set; evidence against the hypothesis of a hierarchy-dependent functional gradient along VLPFC. The methodological study (Study 4) introduces "The Same Analysis Approach (SAA)". SAA allows to detect, avoid, and eliminate confounds and other errors in experimental design and analysis, especially mistakes caused by malicious experiment-specific design-analysis interactions. SAA is relevant for MVPA, but can also be applied in other fields, both within and outside of neuroscience.

Résumé :
Process development of new complex reactions in the pharmaceutical and fine chemicals industries is challenging, and expensive. The field is beginning to see a bridging between fundamental first-principles investigations, and utilisation of data-driven statistical methods, such as machine learning. Nonetheless, process development and optimisation in these industries is mostly driven by trial-and-error, and experience. Approaches that move beyond these are limited to the well-developed optimisation of continuous variables, and often do not yield physical insights. This thesis describes several new methods developed to address research questions related to this challenge. First, we investigated whether utilising physical knowledge could aid statistics-guided self-optimisation of a C-H activation reaction, in which the optimisation variables were continuous. We then considered algorithmic treatment of the more challenging discrete variables, focussing on solvents. We parametrised a library of 459 solvents with physically meaningful molecular descriptors. Our case study was a homogeneous Rh-catalysed asymmetric hydrogenation to produce a chiral γ-lactam, with conversion and diastereoselectivity as objectives. We adapted a state-of-the-art multi-objective machine learning algorithm, based on Gaussian processes, to utilise the descriptors as inputs, and to create a surrogate model for each objective. The aim of the algorithm was to determine a set of Pareto solutions with a minimum experimental budget, whilst simultaneously addressing model uncertainty. We found that descriptors are a valuable tool for Design of Experiments, and can produce predictive and interpretable surrogate models. Subsequently, a physical investigation of this reaction led to the discovery of an efficient catalyst-ligand system, which we studied by operando NMR, and identified a parametrised kinetic model. Turning the focus then to ligands for asymmetric hydrogenation, we calculated versatile empirical descriptors based on the similarity of atomic environments, for 102 chiral ligands, to predict diastereoselectivity. Whilst the model fit was good, it failed to accurately predict the performance of an unseen ligand family, due to analogue bias. Physical knowledge has then guided the selection of symmetrised physico-chemical descriptors. This produced more accurate predictive models for diastereoselectivity, including for an unseen ligand family. The contribution of this thesis is a development of novel and effective workflows and methodologies for process development. These open the door for process chemists to save time and resources, freeing them up from routine work, to focus instead on creatively designing new chemistry for future real-world applications.

Résumé :
The monograph is devoted to improving the methods of directed and controlled regulation of the C — S — H-gel structure by varying the doses, sizes, physical and chemical characteristics of the surface, and the nanoparticles used. The authors have developed an additive that additionally contains a superplasticizer to reduce the water demand of the concrete mixture and stabilize the nanoparticles. The dependences of the strength growth of cement stone and structural heavy concrete on the components of the complex additive are revealed. Experimental confirmation of the mechanism of action of a combined nano—additive with a reduced consumption of nanoparticles on the structure of C — S - H-gel was obtained based on the results of the application of a set of methods. It is revealed that the use of a complex additive contributes to a proportional increase in the reduced modulus of elasticity, hardness, and mechanical characteristics of Portland cement stone and concrete. The study of the additive in the conditions of the construction site showed the prospects of its application for construction, ensuring a reduction in the cost of the technology of nanomodification of concrete relative to the effect of improving performance. For specialists of research, construction and design organizations dealing with the modification of concrete with nanomaterials, as well as for students, undergraduates, postgraduates, teachers who work on the problems of building materials science.

The planetary instantaneity that digital technologies have enabled is leading to an effacement of the divisions that separate the past from the future, ensuring that the present is ubiquitous. While contemporary architecture seems to have lost the capacity to conceive of the past as a transformative force, this book stresses the need to rethink today's complex temporal mechanisms through the notion of the untimely. This concept opens up a whole spectrum of possibilities to go beyond what seems predictable. The contributors to this book employ critical concepts and architectural design tools in order to offer experimental and speculative approaches for unknown futures of architecture.

Résumé :
The monograph discusses the application of nonparametric criteria of agreement (Kolmogorov, Cooper, Kramer-Mises -Smirnov, Watson, Anderson -Darling, Zhang) when testing simple and complex hypotheses. The appendix contains tables containing percentage points and statistical distribution models necessary for the correct use of criteria when testing simple and, most importantly, various complex hypotheses. In comparison with the first edition, more attention is paid to the application of criteria in non-standard application conditions, in particular for the analysis of large samples. It is shown that in applications, the properties of criteria can change significantly due to the presence of rounding errors, and this must be taken into account when forming statistical conclusions. Following the recommendations in data analysis will ensure the correctness of statistical conclusions and increase their validity. It is designed for specialists who, in one way or another, face issues of statistical data analysis, processing of experimental results, the use of statistical methods to analyze various aspects and trends of the surrounding reality in their activities. It will be useful for engineers, researchers, specialists of various profiles (physicians, biologists, sociologists, economists, etc.), university teachers, graduate students and students.

The volume concludes a multi-year study conceived and carried out by the Museo del Tessuto of Prato, which involved public bodies and cultural institutions in the city motivated by the desire to see one of the identifying products of the Prato textile district returned to contemporary life: its cloth. The idea to experimentally reconstruct the cloth arose from the study of some documents of the Datini Fund, a unique archival complex in the world for historical and economic studies on the Middle Ages. The collective and interdisciplinary project is documented in a volume created in collaboration with the International Institute of Economic History “F. Datini”. The historical contributions on the manufacturing and commercial management of the Datini cloth are followed by a second part that documents the study and experimentation phases of its reconstruction.

Résumé :
AbstractThe mapping of flows in a city is essential for understanding urban systems and enabling the transition of the city into a circular economy. However, while tools for the virtual representations of physical volumes and spaces in urban environments have proliferated, effective tools for modeling the underlying flows are still missing. This paper discusses the development of Parametric Sankey, a tool for the trans-scalar representation of flows, and demonstrates its instrumentation in a research design process. The proposed Parametric Sankey tool overlays different flow categories (material, energy, labor, customer, waste, etc.) into one dynamic trans-scalar system, enabling interactive processes between analysis and intervention, and providing thereby an experimental interface to envision future circular cities and architectures.

Résumé :
AbstractIn recent years, the research on digital design and perceptual evaluation has gradually become a hot topic in the field of digital design. Based on digital space optimization theory and perceptual evaluation tools, this study attempts to establish an optimization method to optimize built space elements in real-time using human psychological indicators. This method takes the specific indicators of the Meditation value and Attention value in the human EEG signal analyzed by the TGAM module as the optimization objective, the architectural space color and the window size as the optimization object, and the multi-objective genetic algorithm as the optimization tool. To realize this optimization method, this research combines virtual reality scene and parametric linkage model to establish tool platform and workflow. Taking the optimization of typical residential space as an example by recruiting 50 volunteers to participate in the experiment, this study concludes that this method is effective and feasible through experiment and quantitative analysis of experimental results and lays the foundation for more EEG indicators and more complex spatial element optimization research in the future.

Résumé :
The complex modes of an end-damped cantilevered beam are studied as an experimental example of a non-modally damped continuous system. An eddy-current damper was applied considering its noncontact and linear properties. The state-variable modal decomposition (SVMD) is applied to extract the modes from the impact responses in the cantilevered beam experiments. Characteristics of the mode shapes and modal damping are examined for various values of the damping coefficient. The modal frequencies and mode shapes obtained from the experiments have a good consistency with the results of the finite-element model. The variation of damping ratio and modal nonsynchronicity with varying damping coefficient also follow the prediction of the model. Over the range of damping coefficients studied in the experiments, we observe a maximum damping ratio in the lowest underdamped mode, which correlates with the maximum modal nonsynchronicity. Complex orthogonal decomposition (COD) is applied in comparison to the modal idenfication results obtained from SVMD.

The present paper addresses the identification of a real-life spacecraft structure possessing an impact-type nonlinear component. The complete identification procedure, i.e. from nonlinearity detection to parameter estimation, is carried out using experimental data collected during a typical spacecraft qualification test campaign. The complementary use of several techniques reveals particularly interesting and complex phenomena such as nonlinear jumps, nonlinear modal interactions, internal force relaxation and chattering during impacts.

Résumé :
Stability and non-linear dynamics in a complex aircraft brake model are investigated. The non-linear contact between the rotors ands the stators, and mechanisms between components of the brake system are considered. The stability analysis is performed by determining the eigenvalues of the jacobian matrix of the linearized system at the equilibrium point. Parametric studies with linear stability theory is conducted in order to determine the effect of system parameters on stability. In order to obtain time-history responses, the complete set of nonlinear dynamic equations may be integrated numerically. But this procedure is both time consuming and costly to perform when parametric design studies are needed. So it is necessary to use nonlinear analysis : the center manifold approach and the rational approximants are used to obtain the limit cycle of the non-linear system and to study the behaviour of the system in the unstable region. Results from these nonlinear methods are compared with results obtained by integrating the full original system. These non-linear methods appear very interesting in regard to computational time and also necessitate very few computer resources.

Résumé :
Aim: This study aimed to explore how qualitative data about rehabilitation for patients with intermittent claudication do provide an enhanced understanding of the quantitative experimental results. Background: The study was a randomized clinical trial comparing a rehabilitation intervention with usual care. A statistically significant difference between rehabilitation and usual care was found in walking distance, physical activity, quality of life and diet. The findings from the quantitative and qualitative analyses were analysed separately on their own tradition. In this study, mixed methods address whether the qualitative results could help explain the quantitative results and bring forward additional information. Design: Complex mixed-method intervention design with a convergent questionnaire variant. Methods: From April 2017- May 2019, patients diagnosed with intermittent claudication were included in a randomized clinical trial (N=118). In addition, qualitative interview informants from the intervention group were sampled from the quantitative study population for a survey (N=43) and focus group interviews (N=10). Interviews were conducted from April 2018-August 2019. Results: Integrated analyses identified how improvement in walking distance, physical activity, diet and quality of life was affected by team spirit, pedometer, education and fun exercise in a local setting. Quantitative and qualitative findings primarily confirmed and expanded each other; however, two discordant results were also evident. Conclusion: Our study adds empirical evidence regarding how a mixed-methods study can be used to obtain a more nuanced understanding of complex healthcare problems. The study provides new knowledge concerning how to set up a rehabilitation programme for patients with intermittent claudication.

Résumé :
Surveys often require monitoring during data collection to ensure progress in meeting goals or to evaluate the interim results of an embedded experiment. Under complex designs, the amount of data available to monitor may be overwhelming and the production of reports and charts can be costly and time consuming. This is especially true in the case of longitudinal surveys, where data may originate from multiple waves. Other such complex scenarios include adaptive and responsive designs, which were developed to act on the results of such monitoring to implement prespecified options or alternatives in protocols. This paper discusses the development of an interactive web-based data visualization tool, the Adaptive Total Design (ATD) Dashboard, which we designed to provide a wide array of survey staff with the information needed to monitor data collection daily. The dashboard was built using the R programming language and Shiny framework and provides users with a wide range of functionality to quickly assess trends. We present the structure of the data used to populate the dashboard, its design, and the process for hosting it on the web. Furthermore, we provide guidance on graphic design, data taxonomy, and software decisions that can help guide others in the process of developing their own data collection monitoring systems. To illustrate the benefits of the dashboard, we present examples from the National Longitudinal Study of Adolescent to Adult Health (Add Health). We also discuss features of the dashboard to be developed for future waves of Add Health.

Regulatory experiments can be useful to guide complex transitions in the field of sustainable development. They help to understand the effects of policies and regulations and offer insights into the dynamics of social processes. Empirical studies analyzing heterogeneous samples of regulatory experiments are missing. This paper uses a qualitative content analysis to examine 26 international cases of regulatory experiments in the field of sustainable development. The results show the diversity of existing regulatory experiments in terms of their design. We use the results to formulate implications on how to use regulatory experiments that facilitate learning processes.

Résumé :
The purpose of this project was to complete a handbook with practical design procedures for submarine pipeline on-bottom stability. The remaining part of the handbook was primarily a description of the interaction between non-trenched pipelines and the seabed where the pipelines were free to move under environmental loading. The objective of this project was to determine the lateral soil resistance forces on a pipeline moving cyclically during hydro-dynamic loading. To meet the goal, full-scale pipe-soil interaction tests were conducted. The models presented in this report are based on the results and general understanding obtained from 110 experimental tests of pipe-soil interaction on loose and dense sand, and soft clay. Raw data from 29 experimental tests on stiff clay in the PIPESTAB project have been qualitatively considered.

Résumé :
Diseases are often the consequence of proteins or protein complexes that are non-functional or that function improperly. An active area of research has focused on the identification of molecules that can interact with defective proteins and restore their function. While 22% percent of human proteins are estimated to be druggable, less than fifteen percent are targeted by FDA-approved drugs, and the vast majority of untargeted proteins are understudied or so-called "dark" proteins. Elucidation of the function of these dark proteins, particularly those in commonly drug-targeted protein families, may offer therapeutic opportunities for many diseases. Reactome is the most comprehensive, open-access pathway knowledgebase covering 2585 pathways and including 14246 reactions, 11088 proteins, 13984 complexes, and 1093 drugs. Placing dark proteins in the context of Reactome pathways provides a framework of reference for these proteins facilitating the generation of hypotheses for experimental biologists to develop targeted experiments, unravel the potential functions of these proteins, and then design drugs to manipulate them. To this end, we have trained a random forest with 106 protein/gene pairwise features collected from multiple resources to predict functional interactions between dark proteins and proteins annotated in Reactome and then developed three scores to measure the interactions between dark proteins and Reactome pathways based on enrichment analysis and fuzzy logic simulations. Literature evidence via manual checking and systematic NLP-based analysis support predicted interacting pathways for dark proteins. To visualize dark proteins in the context of Reactome pathways, we have also developed a new website, idg.reactome.org, by extending the Reactome web application with new features illustrating these proteins together with tissue-specific protein and gene expression levels and drug interactions.
