The human calcium- and integrin-binding protein 1 (CIB1) plays important roles in various cellular functions. In this study, three other members of this protein family (CIB2–4: CIB2, CIB3, and CIB4) were purified and subsequently characterized using biophysical and structural approaches. As expected from sequence alignments, CIB2–4 were shown to bind calcium (Ca2+) and magnesium (Mg2+) ions. Binding of Ca2+ or Mg2+ ions changes the secondary structure of CIB2–4 and the exposure of hydrophobic surface area. Ca2+ and Mg2+ ions also stabilize the tertiary structures for CIB2 and CIB3. Through in vitro binding experiments, we show that CIB2 can interact with the integrin αIIb cytoplasmic domain and the integrin α7b membrane-proximal fragment. Fluorescence experiments using a 7-azatryptophan labeled peptide demonstrate that CIB2, CIB3, and CIB4 are binding partners for the integrin αIIb subunit, which suggests that they are potentially involved in regulating integrin αIIb subunit activation. The distinct responses of αIIb to the different CIB3 and CIB4 metal (Ca2+ and Mg2+) binding states imply a potential connection between the calcium and integrin signaling pathways.

Abstract Spermatogenesis is a complex developmental process that involves the proliferation of diploid cells, meiotic division, and haploid differentiation. Many genes are shown to be essential for male fertility using knockout (KO) mice; however, there still remain genes to be analyzed to elucidate their molecular mechanism and their roles in spermatogenesis. Calcium- and integrin-binding protein 1 (CIB1) is a ubiquitously expressed protein that possesses three paralogs: CIB2, CIB3, and CIB4. It is reported that Cib1 KO male mice are sterile due to impaired haploid differentiation. In this study, we discovered that Cib4 is expressed strongly in mouse and human testis and begins expression during the haploid phase of spermatogenesis in mice. To analyze the function of CIB4 in vivo, we generated Cib4 KO mice using the CRISPR/Cas9 system. Cib4 KO male mice are sterile due to impaired haploid differentiation, phenocopying Cib1 KO male mice. Spermatogenic cells isolated from seminiferous tubules demonstrate an essential function of CIB4 in the formation of the apical region of the sperm head. Further analysis of CIB4 function may shed light on the etiology of male infertility caused by spermatogenesis defects, and CIB4 could be a target for male contraceptives because of its dominant expression in the testis.

Calcium- and integrin-binding protein 2 (CIB2) is a small EF-hand protein capable of binding Mg2+ and Ca2+ ions. While its biological function remains largely unclear, an increasing number of studies have shown that CIB2 is an essential component of the mechano-transduction machinery that operates in cochlear hair cells. Mutations in the gene encoding CIB2 have been associated with non-syndromic deafness. In addition to playing an important role in the physiology of hearing, CIB2 has been implicated in a multitude of very different processes, ranging from integrin signaling in platelets and skeletal muscle to autophagy, suggesting extensive functional plasticity. In this review, we summarize the current understanding of biochemical and biophysical properties of CIB2 and the biological roles that have been proposed for the protein in a variety of processes. We also highlight the many molecular aspects that remain unclarified and deserve further investigation.

SummaryPlatelet aggregation requires activation of the αIIbβ3 integrin,an event regulated by the integrin cytoplasmic tails. CIB1 binds to the cytoplasmic tail of the integrin αIIb subunit. Previous overexpression and knockdown studies in murine megakaryocytes demonstrated that CIB1 inhibits integrin αIIbβ3 activation.Here we analyzed Cib1-/- mice to determine the function of CIB1 in platelets in vitro and in vivo. We found that although these mice had no overt platelet phenotype, mRNA level of CIB1 homolog CIB3 was increased in Cib1-/- megakaryocytes. In vitro binding experiments showed that recombinant CIB1, -2 and -3 bound specifically to an αIIb cytoplasmic tail peptide. Subsequent protein modeling experiments indicated that CIBs 1–3 each have a highly conserved hydrophobic binding pocket. Therefore, the potential exists for compensation for the loss of CIB1 by these CIB family members, thereby preventing pathologic thrombus formation in Cib1-/- mice.

Abstract We studied the scientific literature and disease guidelines in order to summarize the clinical utility of genetic testing for Usher syndrome (USH). USH is mostly transmitted in an autosomal recessive manner and is caused by variations in the ADGRV1, CDH23, CIB2, CLRN1, HARS, MYO7A, PCDH15, PDZD7, USH1C, USH1G, USH2A, WHRN genes. Prevalence is estimated to be 1:30,000. Clinical diagnosis is based on audiogram, vestibular tests, visual acuity test, fundus examination, color test, optical coherence tomography and electroretinography. The genetic test is useful for confirming diagnosis, and for differential diagnosis, couple risk assessment and access to clinical trials.

Il cibo, nella società contemporanea, invade le nostre vite a 360°. Ad essere così avvolgente non è solamente la presenza stessa del cibo quanto, in realtà, la sua comunicazione. Nel presente elaborato si prende in analisi una figura che dedica la sua quotidianità alla comunicazione della cucina, Massimo Bottura. Il celebre chef parla attraverso l’alta cucina dell’Osteria Francescana e attraverso la Onlus Food For Soul, nata ai margini di Expo 2015 per riutilizzare gli avanzi alimentari della manifestazione e trasformarli in piatti da servire a chi non può permettersi un pasto caldo. La tesi si concentra sul modo di comunicare la cucina e il cibo da parte di Bottura. L’analisi in particolare si propone di descrivere ed evidenziare eventuali differenze nel modo di trattare il cibo nei due mondi, quello stellato della Francescana e quello, frugale ma ricco di umanità, dei refettori.

Il lavoro di ricerca ha a oggetto l’analisi del modello europeo (UE) di regolamentazione delle nanotecnologie nel settore agroalimentare, con riferimento agli aspetti etico-giuridici e sociali, ai fini della definizione del quadro normativo di riferimento nella sua relazione con la dimensione di complessità e incertezza intrinseca nel sapere scientifico-tecnologico. La prospettiva teorica da cui muove l’analisi è la co-produzione tra i linguaggi della scienza e del diritto proposta dagli STS (Science & Technologies Studies). Partendo dalla descrizione degli aspetti scientifici dei nanomateriali e delle applicazioni nanotecnologiche nel settore alimentare, il lavoro analizza, in primo luogo, le problematiche correlate alle procedure di gestione del rischio – dalle prospettive più riduzionistiche della cd. “scienza del rischio” nell'innovazione alle più complesse modalità di valutazione integrata del rischio. L’indagine s’incentra, quindi, sulle forme che la normazione sta assumendo nell'intreccio con i saperi delle nanotecnologie, attraverso un approccio comparatistico delle esperienze normative europea e statunitense. L’ultima parte del lavoro s’indirizza, infine, all’analisi delle esigenze di democraticità sottese alle suddette scelte scientifico-giuridiche, problematizzando il concetto di governance anticipatoria e responsabile delle nanotecnologie (concetto correlato all’idea di riuscire a guidare i processi di innovazione attivamente), alla luce del rapporto tra conoscenze scientifiche, politiche agroalimentari e diritto.
In recent decades technoscientific innovation has pushed the food boundaries to a new frontier of nanofood. Such a term refers to an array of food products, whose processes of growing, production and packaging involve nanoscale (nanotechnology and nanosciences) knowledges and applications. This research focuses on the analysis of the European (EU) regulatory framework in the field of agrofood nanotechnology. The analysis considers the salient features of emerging applications of nanotechnologies in the agrofood sector and compares the legal framework on nanofood in the EU with the USA’s regulatory approach. It also develops an interpretation of the normative evolution in the EU, by trying to understand what is the role of science in governing technological risks in nanofood safety, and assessing how adequate the regulatory instruments are in achieving the goal of responsible research and innovation as proposed within the process of rethinking European governance.

Background: Cancer-induced bone pain (CIBP) is a major clinical problem and a considerable therapeutic challenge. Radiotherapy (XRT) is the gold standard treatment for CIBP, but only half of patients achieve adequate analgesia. Patients have increased morbidity, anxiety and depression and reduced performance and quality of life. Despite these issues, CIBP is a neglected area of clinical research. Animal models have increased current knowledge of the pathophysiology, but clinical research is needed to translate these findings from bench to bedside. Also lacking is a standardised, comprehensive tool to assess CIBP and clinical biomarkers to predict analgesic response to treatment. Aims: 1) To summarise current understanding of the pathophysiology, epidemiology, clinical features, assessment and management of malignant bone disease and CIBP. 2) To characterise CIBP using quantitative sensory testing as a measure of altered sensory processing. 3) To establish systematically the sensory, cognitive, affective and functional components of CIBP to develop a comprehensive assessment tool. 4) To explore whether clinical biomarkers can be developed to aid prediction of response to treatment for CIBP, in particular XRT. Results: Assessment of CIBP, characterising the multi-dimensional components, was clinically practical and acceptable to patients. Using objective measures of function, patients with CIBP were a frailer, less active population compared with healthy adults. Prior to treatment, pain was severe with relationships seen between CIBP and sensation, mood, fear avoidance, catastrophizing and function. Patients who dropped out prior to follow up were significantly less active, with higher levels of depression and fear avoidance behaviour. Sixty-nine percent of evaluable patients who completed two assessments (48% of all patients on an intention-to-treat basis), achieved an analgesic response to XRT for CIBP, as defined as an improvement of ≥ 30% in the Brief Pain Inventory worst pain score two months after treatment. All dimensions of pain, fear avoidance and catastrophizing improved significantly in responders, but not non-responders. Anxiety, depression and emotional distress fell by a greater degree in responders. No objective functional differences were seen after XRT. Clear evidence of altered sensory processing was seen at the site of CIBP with abnormalities in both mechanical and thermal parameters. XRT resulted in alterations in response to evoked stimuli in responders with a greater number of patients in whom sensation normalised after XRT compared with non-responders. Patients with a combination of altered sensation to thermal, pin prick and wind up stimuli showed the largest reduction in worst pain after XRT. Abnormal cool sensation at the site of CIBP was an independent predictor of analgesic response to treatment. Conclusion: Strong associations exist between CIBP, sensation, cognition, mood and function. Multi-dimensional assessment should be performed to improve quality of life. Translational research to provide targeted individualised treatment should be high on the research agenda. Future work should focus on thermal sensory processing as a potential clinical biomarker of response to palliative XRT for CIBP.

Abstract Mutation breeding induced by irradiation with highly energetic photons and ion beams is one of the important methods to improve plant varieties, but the mutagenic effects and molecular mechanisms are often not entirely clear. Traditional research is focused on phenotype screening, chromosome aberration tests and genetic variation analysis of specific genes. The whole genome sequencing technique provides a new method to understand and undertake the comprehensive identification of mutations caused by irradiations with different linear energy transfer (LET). In this study, ten Arabidopsis thaliana M3 lines induced by carbon-ion beams (CIB) and ten M3 lines induced by gamma-rays were re-sequenced by using the Illumina HiSeq sequencing platform, and the single base substitutions (SBSs) and small insertions or deletions (indels) were analysed comparatively. It was found that the ratio of SBSs to small indels for M3 lines induced by CIB was 2.57:1, whereas the ratio was 1.78:1 for gamma-rays. The ratios of deletions to insertions for carbon ions and gamma-rays were 4.8:1 and 2.8:1, respectively. The single-base indels were more prevalent than those equal to or greater than 2 bp in both CIB and gamma-ray induced M3 lines. Among the detected SBSs, the ratio of transitions to transversions induced by carbon-ion irradiation was 1.01 and 1.42 for gamma-rays; these values differ greatly from the 2.41 reported for spontaneous substitutions. This study provides novel data on molecular characteristics of CIB and gamma-ray induced mutations at the genome-wide scale. It can also provide valuable clues for explaining the potential mechanism of plant mutation breeding by irradiations with different LETs.

In order for a resin to be suitable for stereolithographic applications, it must have a high curing speed, high build resolution and low shrinkage during the liquid to solid polymerisation process. (1). We performed nondegenerate four-wave mixing on two UV curable resins, the DuPont Somos™ 6100 and the Ciba-Geigy Ciba- tool™ SL 5180. Gratings with spatial wavelength Λ=0.3 - 1.1 µm were written using an Argon ion laser operating on the λ0=351.1 nm line and for a range of irradiance 0.5-3 W/cm2. The gratings were then read using a He-Ne laser, operating at 632.8 nm. These experiments were carried out to prove the suitability of the photopoly, nerisation systems for microstereolithography.

At the start of the last decade, United Cities and Local Governments’ (UCLG) policy paper on Decentralised Cooperation and Local Government laid out a clear rationale for decentralised cooperation and set out recommendations to the prevalent tackle weaknesses of international development cooperation and to strengthen development effectiveness. In many ways, the paper was a forerunner in calling for stronger sustained support for South-South development cooperation particularly among countries that have undergone similar socio-economic challenges so that learnings can be shared across partners. It laid emphasis on professional structures and programme-based approaches, with clear monitoring and evaluation tools and indicators on impact and called for a sharing of objectives across local and regional governments, and their associations, committed to continuing improvement, learning and exchange. These recommendations have helped strengthen international decentralised cooperation over the past decade, and their core principles continue to be highly relevant today. In 2021, the Institute of Development Studies, UK, with support of the UCLG Capacity and Institution Building Working Group (CIB), has engaged a wide range of member governments, associations, and networks, alongside a range of external commentators and experts, to assess UCLG principles, priorities, and actions in the context of contemporary global challenges and the resulting landscape of decentralised development cooperation. Following a series of survey-based, individual, case study, and workshop interactions, the study presents key points and recommendations.
