Abstract The chr 5p15.33 genomic locus encoding telomerase reverse transcriptase (TERT) is structurally complex, with several highly polymorphic intronic variable number tandem repeats (VNTRs). A germline VNTR located in TERT intron 6 (VNTR6-1, 38 bp repeat unit) has previously been associated with TERT alternative splicing but its potential role in regulating cellular functions and disease risk is unclear. To functionally characterize VNTR6-1, we deleted this region (1-2 kb) using CRISPR/Cas9 in UMUC3 (bladder cancer cell line). Cell count growth tracking of parental and VNTR6-1 knockout (VNTR6-1 KO) cells via automated live-cell monitoring revealed different relative growth rates under serum starvation vs. normal conditions, suggesting that VNTR6-1 might be sensitive to growth factors present in culture media. Evaluation of growth in media with full vs. charcoal-stripped (CS) serum, showed significant differences, with higher cell counts in parental vs. VNTR6-1 KO in CS-serum (P=4.1e-3), parental vs. parental in full (P=1.57e-4) or CS-serum (P=4.76e-2), but not in parental vs. VNTR6-1 KO in full serum (P=0.378). After 5-6 days of growth, cells in CS-serum also rapidly began outgrowing cells in full serum. We observed that VNTR6-1 KO dampened the outgrowth response in cells lacking growth factors in CS-serum. Proliferation-specific growth assays tracking the dilution of an incorporated dye in cells by flow cytometry further showed that VNTR6-1 KO leads to more pronounced proliferation differences due to the presence or absence of serum-supplied factors. Overall, cell growth patterns appear to be sensitive to serum in interaction with VNTR6-1, suggesting a potential VNTR6-1-dependent response to extracellular signaling. In human populations, we have identified two VNTR6-1 groups: Short (24-27 copies) and Long (40.5-66.5 copies). In the PLCO dataset (n=100,445 cancer cases and controls), logistic regression showed that VNTR6-1-Long group was associated with a decreased risk in male-prevalent cancers such as bladder and prostate, but an increased risk in female-prevalent cancers such as breast, endometrial and ovarian. These results support a link between the TERT region, and potentially VNTR6-1, with hormone signaling and cancer risk. Motif analysis within VNTR6-1 predicted binding sites for several transcription factors, including those associated with development and cell growth. Together, these results suggest that VNTR6-1 may be involved in regulating cell growth, potentially by facilitating responses to extracellular signaling pathways. Additionally, given the location of VNTR6-1 within a multi-cancer GWAS region, our findings provide functional context for the role of this polymorphic repeat in differential risk and outcomes in certain cancers. In traffic flow analysis several regimes are distinguished, such as congested and noncongested flow conditions. Indications exist that driving behavior differs by regime and that it may change discontinuously between regimes. In contrast most traffic flow models used today basically assume the same car-following behavior irrespective of the traffic flow regime. It is hypothesized that, because of this deficiency, these models do not always perform satisfactorily. To clarify this issue, differences in car-following between congested and noncongested flow are analyzed with data from two sites on Dutch freeways. It is shown that, at the same speeds, passenger car drivers follow with smaller headways in noncongested than in congested flow. Car-following of truck drivers does not show differences between regimes. Microscopic distance gap-speed models are established for several road-user classes, valid for each of the two flow regimes. To show the improvements resulting from these new microscopic relationships, the latter are implemented in a microscopic simulation model with which macroscopic patterns in traffic flow are modeled. The macroscopic findings produced with the regime-specific car-following rules show a considerable improvement in modeling performance.

A linear acceleration car-following model has been developed for realistic simulation of traffic flow in intelligent transportation systems (ITS) applications. The new model provides continuous acceleration profiles instead of the stepwise profiles that are currently used. The brake reaction times of the drivers are simulated effectively and are independent of the simulation time steps. Chain-reaction times of the drivers are also simulated and perception thresholds are incorporated in the model. The preferred time headways are utilized to determine the simulated drivers’ separation during car-following. The features of the model and the realistic vehicle simulation in car-following and in stop-and-go conditions make this model suitable to ITS, especially to autonomous intelligent cruise-control systems. The car-following algorithm is validated at microscopic and macroscopic levels by using field data. Simulated versus field trajectories and statistical tests show very strong agreement between simulation results and field data.

The paper reports the methods for the synthesis of new cluster compounds of trans-tetrachlorodi--carboxylates of dirhenium(III) with dipeptides of the glycyl series. The compositions and the structures were determined by electron absorption and IR spectroscopies as well as by elemental analysis for the following newly synthesized compounds: trans-[Re2(H3N+–CH2–СO–NH–CH(COO)–CH2–CH(CH3)–CH3)2Cl42СН3CN]Cl2 (І) and trans-[Re2(H3N+–CH2–СO–NH–CH(COO)–CH2–C6H5)2Cl42СН3CN]Cl2 (ІІ). Analysis of the electronic absorption spectra of the solutions of the synthesized substances showed the presence of a doublet pattern (12500 and 16129 сm–1), which corresponds to the *-electronic transition of the Re–Re quadruple bond characteristic of solutions of trans-tetrachlorodi--carboxylates of dirhenium(III). The appearance of oscillations at 1485 сm–1 for І and 1486 сm–1 for ІІ, characteristic of the s(CO) of the coordinated carboxylate group, was detected on the IR spectra, which indicates the bridging coordination of this group to the Re26+ binuclear fragment. The protonation of the amino group is indicated by the appearance of a wide band of valence oscillations (NH3+) in the range of 3400–3350 сm–1 and deformation oscillations (NH3+) at 1557 сm–1 and 1614 сm–1 for I and II, respectively. The stability of the obtained complex compounds in aqueous solutions was determined. It was shown that the hydrolysis of the synthesized substances takes place in 4–5 days, which is accompanied by a decrease in the pH of the reaction solution.

Increasing demand for road freight has lead to a widespread adoption of more-productive large freight vehicles (LFVs), such as B-Doubles, by Australia's road freight industry. Individual LFVs have a greater potential to impact traffic efficiency through their greater length and poorer longitudinal performance. However, this is offset to an extent as fewer vehicles are required to perform a given freight task on a tonne-km basis. This research has developed a means of characterising the effects that large freight vehicles have on the performance of an urban arterial corridor managed by signalised intersections. A corridor-level microsimulation model was developed from first principles, which modelled the longitudinal performance of individual vehicles to a greater accuracy than most existing traffic simulation software does. The model was calibrated from traffic counts and GPS-equipped chase car surveys conducted on an urban arterial corridor in Brisbane's southern suburbs. The model was applied to various freight policy and traffic management scenarios, including freight vehicle mode choice, lane utilisation and traffic signal settings; as well as the effectiveness of green time extension for approaching heavy vehicles. Benefits were able to be quantified in terms of reduced travel times and stop rates for both heavy and light vehicles in urban arterial corridors.

Older Australians walk for many reasons: health, recreation and transport. However, road safety statistics show that pedestrians over 65 represent one-third of Australia's pedestrian deaths. As Australia's population ages in place and older people take up a walking regime for health and transportation reasons, they need a supportive suburban setting. Urban design theories discuss such "pedestrian-friendly" concepts as sense of place, sense of community, responsive environments, traditional neighbourhood design, transit-oriented development, and crime prevention through environmental design. To investigate these concepts in relation to older pedestrians, this study brings together two areas of literature - research into older pedestrians in relation to urban design theories. Qualitative research methods were used in two case studies, to reveal how older people's interpretation of their local walking environment relates to urban design theories concerning walkable suburbs. The two Brisbane suburbs of Bulimba and Forest Lake were chosen for study, as they have different histories, topographies, street patterns, and other variations. Analysis of key themes gathered from two focus group discussions, one from each suburb, revealed the significance for participants of social interaction when walking for health. A photographic exercise performed by the Forest Lake focus group provided pictorial information for analysis, and revealed participants' interest in the lake's fauna and flora, and in its ongoing maintenance. The study was limited by an unforeseen failure to obtain the cooperation of the Bulimba group in the photographic exercise. In support of the claims made in the literature review, it seems that when older pedestrians walk through suburban streets, they avoid steep hills, busy roads, and intersections where possible, and require footpaths with even surfaces and shelters. When walking for health reasons, participants in this study did not favour local streets, but preferred "natural" places designed exclusively for walkers. Forest Lake participants stated a preference for driving to places they deemed suitable for walking, which suggests a need for more detailed design attention to the urban design qualities of local streets, so that those older people without cars are not disadvantaged.

Knowing the factors that affect the composition and quality of milk in Dairy Production Systems (SPL) is of utmost importance to the producer. The objective was to evaluate the characteristics of 32 SPL in the city of Toledo, together in partnership with a dairy. We performed the data collection of properties, with the aid of a semi-structured questionnaire, which collected information on the production traits, milk composition and quality, besides the technical and management practices carried out, mainly related to hygiene and management health of milking. Milk samples were analyzed for their composition (fat, protein, lactose and total solids), Somatic Cell Count (SCC), Total Bacterial Count (TBC), and also to the presence of total coliforms and fecal coliforms. Subsequently, data collection, the variables were selected and analyzed using multivariate techniques, using the principal component analysis (PCA) and analysis of Ascendant Hierarchical Classification (AHC). We analyzed variables related to production and milk quality, techniques and practices used at the time of milking, feeding management, milking system and socio-cultural variables related to the producer. The variables that best explained the differences in the PCA were related to the hygienic and sanitary practices (pre and post-dipping, use of detergents in alkaline and acid cleaning, testing for mastitis) held for milking, with the composition of milk and linked to the producer (age, education, length of service). It was found that the use of these practices can help to improve production and milk quality, ensuring a raw material with high commercial value. The grouping of production systems has shown that there is great diversity among the forms of production, allowing identifying the factors that can influence it, in order to form strategies that may result from gains in production and milk quality
Conhecer os fatores que afetam a composição e qualidade do leite em Sistemas de Produção Leiteiros (SPL) é de extrema importância ao produtor. Objetivou-se avaliar as características de 32 SPL, na cidade de Toledo, juntamente em parceria com um laticínio. Foi realizada a coleta de dados das propriedades, com o auxílio de um questionário semi-estruturado, onde se colheu informações acerca das características de produção, composição e qualidade do leite, além das técnicas e práticas de manejo realizadas, principalmente relacionadas ao manejo higiênico-sanitário de ordenha. As amostras de leite foram analisadas em relação a sua composição (gordura, proteína, lactose e sólidos totais), Contagem de Células Somáticas (CCS), Contagem Bacteriana Total (CBT), e também para presença de coliformes totais e coliformes termotolerantes. Posteriormente, a coleta de dados, as variáveis foram selecionadas e analisadas por meio de técnicas multivariadas, sendo utilizada a análise de Componentes Principais (ACP) e a análise de Classificação Hierárquica Ascendente (CHA). Foram analisadas variáveis relacionadas à produção e qualidade do leite, técnicas e práticas utilizadas no momento da ordenha, manejo alimentar, sistema de ordenha e variáveis sócio-culturais relacionadas ao produtor. As variáveis que melhor explicaram as diversidades das ACP foram as relacionadas com as práticas higiênico-sanitária (pré e pós-dipping, utilização dos detergentes alcalino e ácido na higienização, testes para detecção de mastite) realizadas na ordenha, com a composição de leite e vinculadas ao produtor (idade, escolaridade, tempo na atividade). Foi verificado que o uso destas práticas podem auxiliar para melhorar a produção e qualidade do leite, assegurando uma matéria-prima com maior valor comercial. O agrupamento dos sistemas de produção permitiu verificar a existência de grande diversidade entre as formas de produção, possibilitando identificar os fatores que podem influenciá-lo, a fim de formar estratégias que possam resultar ganhos em produção e qualidade do leite

We have 3G on the doorstep but nothing seems to attract ordinary people to this technology. To attract the mass market the telecom industry must show something beyond high bit rates. They must show how ordinary people can take advantage of this new technology. This is done by showing the possibilities of the new technology and by demonstrating applications that it will handle. The telecom industry must convince the telecom operators to invest in this technology and the only thing that matters to them is how much revenue they can make by adopting the upcoming technology.

To convince the operators industry must show how the operators can charge for the new types of applications that will be introduced soon. This is the main reason why this Master's Thesis has been conducted. The purpose of this thesis is to provide a demonstration to Ericsson's 3G lab in Katrineholm in the form of an IP application with a billing solution. This thesis describes the migration from 1G to 3G and examines existing and future billing strategies as well.

The IP application is an application that uses progressive streaming in order to stream multimedia content to a PDA connected to a 3G phone. This application is platform independent because it is placed on leading Web servers, Apache and IIS.

The billing application consists of a number of steps. The first step is logging, which is performed by the Web server on which the streaming application is placed. The second step, processing and billing, is performed in the BGw, which is Ericsson's mediation tool, and the SQL server.The third step is displaying the bill, which is done by using ASP to create an active HTML page.

A photographic exercise performed by the Forest Lake focus group provided pictorial information for analysis, and revealed participants' interest in the lake's fauna and flora, and in its ongoing maintenance. The study was limited by an unforeseen failure to obtain the cooperation of the Bulimba group in the photographic exercise. In support of the claims made in the literature review, it seems that when older pedestrians walk through suburban streets, they avoid steep hills, busy roads, and intersections where possible, and require footpaths with even surfaces and shelters. When walking for health reasons, participants in this study did not favour local streets, but preferred "natural" places designed exclusively for walkers. Forest Lake participants stated a preference for driving to places they deemed suitable for walking, which suggests a need for more detailed design attention to the urban design qualities of local streets, so that those older people without cars are not disadvantaged.

Il commento alle Lettere e Rime che Chiara Matraini (1515- 1604), ottantaduenne, pubblicò a Venezia, presso Moretti, nel 1597, intende fornire un’analisi organica della composizione, del rapporto tra i generi, delle modalità dell’imitatio e del sostrato filosofico, sociale e culturale che fanno da sfondo all’ultimo canzoniere. Lo studio si concentra anche sulle revisioni occorse lungo l’arco di un quarantennio (1555-1597), dall’adesione al canone petrarchista e bembiano, al Manierismo di fine secolo, nonché sulla torsione in senso controriformistico caratterizzante a parallela scrivania filosofico-devozionale e qui ravvisabile in filigrana. L’indagine considera inoltre l’intero liber di Lettere e Rime come un insieme volutamente organico e strutturato nel raccordo tra le due parti. Ad emergere è un’opera complessa, anche in virtù della sua ‘marginalità’, restituita insieme ai molteplici ripensamenti che animarono la lunga, quasi secolare, esistenza dell’autrice lucchese.

The Washington Metropolitan Area Transit Authority (WMATA) contracted with Booz Allen Hamilton to conduct a non-destructive structural assessment of the 2000 and 3000 series rapid transit rail cars. The main goals of the assessment were to identify any significant changes that have occurred to key elements of the car body structure since their date of manufacture and to confirm that no significant and obvious structural deterioration or damage exists. The methodology behind selecting a small representative sample of cars is discussed as an introduction to this work. The paper then discusses the process by which several critical areas were selected for non-destructive inspection. One of the biggest challenges in successfully completing this project was not interfering with WMATA’s fleet operating requirements. The use of WMATA facilities had to be coordinated to create minimum interference with WMATA’s daily maintenance activities. Also, the inspection work had to be planned in such a way as to minimize the amount of vehicle component disassembly in order to return the vehicle to revenue service as soon as possible. The inspections produced valuable results regarding the construction and condition of these cars. The structural welds of the car appear to have performed satisfactorily through the operational life of the cars to date without any significant deterioration. Some corrosion was noted in the door opening areas of the cars, particularly at the door thresholds. The findings of this report will be used to target specific areas of the car during the upcoming mid-life rehabilitation project.

The CHF in PWR fuel assemblies is usually predicted by the local flow correlation approach based on subchannel analysis while the effects of spacer grids, cold walls, non-uniform heat flux, etc are investigated. By using the subchannel code ATHAS to calculate each set of bundle CHF data, the local thermal-hydraulic parameters at DNB occurrence point were obtained. In present study, the minimum DNBR point method was applied to develop a new CHF correlation for PWR fuel assemblies. The so-called “three-step method” and “magnitude analysis method” were used to determine the shape and the expression of each item, respectively and the least square method was applied to determine the coefficients of the correlation. Based on the large database of CHF tests, the CHF correlation named ACC correlation has been developed to calculate the risk of DNB. The analysis and assessment results indicate that the ACC correlation can fit the experimental data well with high prediction accuracy and correct parametric trends. Coupled with subchannel code ATHAS, this correlation can simulate the thermal-hydraulics performances of PWR fuel assemblies exactly.

Since July 2002 a pump & treat remedial action has been in operation for the extraction of Uranium- and “CHC-compounds” contaminated ground-water flowing below an industrial area used before as production site for nuclear fuel elements. So far about 17 kg Uranium and 22 kg CHC-compounds have been eliminated. As described earlier [1] two ground-water plumes have transported the Uranium downstream to locations where two main remediation wells are placed. They show characteristic differences with respect to their Uranium-nuclide vector. More than 220.000 m3 ground-water of the whole 350.000 m3 have been pumped therefrom. Preferred ground-water paths cause blending the two plumes’ waters. The first plume’s water characterized by decreasing Uranium-235 ratios is meanwhile dominated by the second plume’s water of up to 85% in case of well W10. The overall activity of 122 MBq Uranium-235 has been extracted from the saturated zone with decreasing quantities. Only a small area remains where the first plume’s pure ground water will be extracted in future. Both target values of 20 μg/l and 10 μg/l for dissolved Uranium and CHC-compounds, respectively, could not yet be reached for all remediation wells. In accordance to the 10μSv-concept of radiation protection the improvement of specific parameters will still be necessary in order to release the complete site from the German Atomic Law.

This paper describes measurement of the aerodynamic pressures produced by a Bombardier High-Speed Non-Electric Locomotive (HSNEL) on an adjacent stationary double-stack freight car. Static pressures are measured on the near and far-side faces of the freight containers over a range of locomotive speeds from 60 to 130 mph. This data is also compared with the pressures predicted by computational fluid dynamics (CFD) simulations.

Vibration signatures of defective roller bearings on railroad freight cars were analyzed in an effort to develop an algorithm for detecting bearing defects. The effort is part of a project to develop an on-board condition monitoring system for freight trains. The Office of Research and Development of the Federal Railroad Administration (FRA) is sponsoring the project. The measurements were made at the Transportation Technology Center (TTC) in Pueblo, CO on July 26 – 29, 1999 during the Phase III Field Test of the Improved Wayside Freight Car Roller Bearing Inspection Research Program sponsored by FRA and the Association of American Railroads (AAR). Wheel sets with specific roller bearing defects were installed on a test train consisting of 8 freight cars designed to simulate revenue service. The consist also contained non-defective roller bearings. Accelerometers were installed on the inboard side of the bearing adapters to measure the vibration signatures during the test. Signatures of both defective and non-defective bearings were recorded. The data were recorded on Sony Digital Audio Tape (DAT) Recorders sampling at a rate of 48 K samples per second. We used both ordinary and envelope spectral analysis to analyze the data in an effort to detect features that could be related to known defects. The spectra of non-defective bearings show no remarkable features at bearing defect frequencies. In general, the ordinary spectra of defective bearings do not exhibit remarkable features at the bearing defect frequencies. In contrast, the envelope spectra of defective bearings contain a number of highly resolved spectral lines at these frequencies. In several cases the spectral lines could be related to specific bearing defects. Based on the analysis performed to date, the envelope spectrum technique provides a promising method for detecting defects in freight car roller bearings using an on-board condition monitoring system.
