Texte intégralThe tunneling splitting represents an important nuclear quantum effect that can be detected in a large number of molecular systems spectra. The work presented in this thesis aims to propose a novel approach to the computation of these quantities by harnessing, thanks to the isomorphic relation existing between the Born-Oppenheimer nuclear Hamiltonian and the symmetrized Fokker-Planck-Smoluchowski operator, the localization function approach, usually employed in the computation of kinetic constants of activated processes, in the field of tunneling splitting estimation. Following this strategy, an analytic approximation, characterized by exponentially better convergence in the limit of high barriers, has been obtained for the case of one-dimensional systems opening, as such, a new paradigm in the problem of tunneling splitting computation. Starting from this asymptotic approach, a broad range of theoretical tools has been developed allowing us to tackle, either with an approximated approach or a numerical solution, the ground and excited-state tunneling splitting estimation in one and many-dimensional model systems with a variable degree of accuracy.

Texte intégralQuesta tesi sviluppa gli aspetti teorici basilari delle proprietà elettroniche dei nanotubi di carbonio che sono necessari per una comprensione dettagliata delle misure di caratterizzazione ottica tramite fotoluminescenza e spettroscopia Raman. Partendo da una porzione finita del reticolo reale opportunamente scelta con condizioni periodiche al contorno appropriate, l’approccio del cristallo piccolo consente di trovare l’insieme più piccolo di punti della zona di Brillouin che sono sufficienti per calcolare il profilo essenziale degli spettri ottici di sistemi periodici, come i nanotubi a parete singola. La visione in spazio reale presente nell’approccio del cristallo piccolo consente di superare le limitazioni inerenti ai metodi in spazio reciproco, quando si debbano considerare effetti di rottura locale di simmetria nella struttura elettronica dei nanotubi di carbonio, come interazioni elettrone-elettrone, difetti puntuali e interazioni intertubo dipendenti dall’orientazione dei tubi costituenti, quest’ultimo nel caso particolare dei nanotubi a parete doppia. La tesi mostra l’applicazione dell’approccio del cristallo piccolo a questi problemi e i risultati ottenuti vengono discussi in relazione alle attuali conoscenze sperimentali e teoriche.

Texte intégralIn this Ph.D. thesis, centred around the study of lanthanide-based luminescent mo-lecular systems with applications in thermometry, several high-level quantistic calculation techniques have been explored; these have been applied to determine parameters and molecular characteristics, which are useful for the comprehension of the underlying mechanisms defining the temperature dependence of the optical properties. After a brief introduction in which the general nature of the systems is discussed, theoretical bases of numerical simulations are illustrated; scientific articles published on international journals with peer review and describing the theoretical modeling results are also included. Theoretical tools obtained from these studies allow not only to rationalise the optical characteristics of the investigated systems, but also to predict the behaviour of systems which have not yet been char-acterised.

Texte intégralNowadays, ripening of salami under natural conditions has been replaced by batch production in ventilated industrial chambers. Hence, the same product quality can be obtained regardless of local, environmental and climatic conditions. Nevertheless, carefully designed and monitored process conditions are necessary to achieve the targeted weight loss and quality of dry fermented sausages. Therefore, the availability of reliable mathematical models and manageable software codes for sausage drying, maturation and optimal production are highly welcome. This thesis deals with the development and the progressive improvement of a mathematical model (and its underlying software solving code) of sausage drying during actual industrial ripening conditions. The mathematical model considered as the start point had the following features: 1D, axi-symmetric, time-invariant cylindrical geometry; homogeneous and isotropic material; no distinction between the inner part (core) and the casing; water concentration as a distributed parameter; the internal water transfer rate as a concentration-dependent Fickian diffusion with the assumption of an effective diffusion coefficient depending on the local water content. Starting form this, different and more realistic improvements of the sausage geometrical representation have been pursued: cylindrical 2D axi-symmetric geometry, irregular 2D axi-symmetric geometry, irregular 3D geometry, cylindrical 2D axi-symmetric geometry with volume shrinking. From the comparison of the various model predictions with the experimental data that originated from external industrial-scale ripening tests, it was possible to deduce that a more realistic irregular geometry is influential on the computational load, but irrelevant to the prediction of the sausage weight loss, i.e., the most well-known process performance variable. For this reason, a simple cylindrical shape could be used to implement a more realistic variable-volume simulation model. Another assessment that was made with this study was that the ripening process can be considered isothermal because the temperature transients, e.g., occurring at startup or after a temperature set point change, have a characteristic time that is negligible when compared to that characteristic of mass transfer. A natural, but necessary development of the work has been a mathematical model of fermented sausage drying relying on an innovative description of the sausage as a heterogeneous material, i.e., separately made of lean meat and fat, and a porous medium. The fat is considered as an inert matter dispersed in the meat matrix, while the lean meat is considered as a porous medium in which the transport phenomena of water take place to the outside of the matrix during the ripening process. Porosimetry and image analysis were applied to the meat matrix to demonstrate the goodness of the porous media approach. However, the direct implementation of the above porous media approach in a calculation code has been prevented by the lack of correlations for the water diffusion in the porous meat matrix. For this reason, an experimental setup was thought that would allow an evaluation of the dependence of the diffusivity from the local water content in the meat matrix during the drying process. The experimental apparatus allows real-time monitoring of the sample weight and water concentration profiles that are established in the material under investigation. The sample is kept in a constant-humidity, controlled atmosphere during the whole experiment. By coupling the model simulation with porous media approach and the experimental data from such an apparatus, it is possible to simulate the drying process of a particular material for which the internal water diffusion characteristics are unavailable or unrealistic in the literature. In particular, two correlations for the water diffusion in the porous meat matrix (e.g., D=f(local moisture content, Temperature)) were found in literature, but their application or validation to the case of meat-based food was missing. So, it was possible to determine optimal parameters for them, just tailored for the lean meat under investigation here. Therefore, with a more reliable correlation for water diffusion and the calculation code based on the porous media approach, the prediction of the drying mechanism turned out more realistic and provided added value to the knowledge of the salami curing processes. Texte intégralThe contamination of aquatic sediments with metals is a widespread environmental problem. The marine sediment is a sink for contaminants in the water column, so that in areas with high level of anthropization the sediment can reach very high concentrations of metal contaminants. The biological strategies for remediation of contaminates environmental matrices is gaining increasing prominence and they are often considered as a very promising approach for the potential treatment of contaminated sediments. Unlike organic pollutants, metal contaminants cannot be degraded, neither the natural processes of decomposition can remove them. As a consequence, any bioremediation strategy can be just aimed at transforming metals and semi-metals in more soluble/insoluble compounds and/or in less toxic species. This dissertation deals with the potential of prokaryotes in the mobilization/ immobilization of (semi-)metals in marine contaminated sediments, with a particular focus on the biogeochemical interactions between sediment, bacteria and (semi-) metals. The research activities have addressed the following specific questions: 1. Can the bioaugmentation with acidophilic Fe/S oxidizing bacteria (Bioleaching) be applied on marine sediments to mobilize the metal contaminants? Which factors do influence the mobilization efficiency and the real applicability on contaminated marine sediments? 2. Do the geochemical properties of the sediment affect the bio-mobilization of metals? How do the interactions among abiotic and biotic processes affect the bio-mobilization of metals from contaminated marine sediments? 3. Can the biostimulation of autochthonous microbial assemblages affect the fate of metals in marine contaminated sediments? Which are the abiotic and abiotic constrains influencing the mobility of metals during biostimulation actions? 4. Which kind of interactions among abiotic and biotic factors affect metal mobilization/immobilization in marine sediments? Can the key findings from this research be generalized in order to describe the mobilization/immobilization of metals in the marine sediment? To answer these questions, the research activities were organized in four work-packages. The first experimental campaign was aimed at testing the potential of acidophilic bacteria as bioremediation strategy for the treatment of marine sediments contaminated with metals and semi-metals. Bioaugmentation experiments were carried out with a sediment concentration higher than usually investigated in the scientific literature. The potential of Fe-reducing bacteria in favoring the acidophilic Fe/S oxidizing bacteria was investigated. Additional factors investigated were the presence/absence of different growth substrates (ferrous iron, elemental sulfur, glucose). Our results revealed that the high concentration of sediment had a negative effect on the efficiency of metal solubilization. On the contrary, the main factor that favored the mobilization of metals was the presence of iron as growth substrate. The role of Fe/S oxidizing bacteria appeared to be limited at the production and regeneration of key chemical species, responsible for metal leaching (like H+ and Fe3+). On the basis of the results obtained during the first experimentation, a second experimental campaign was designed to investigate the effect of the geochemical properties of the sediment. In order to goal this objective, we performed new bioaugmentation experiments with the same acidophilic strains on three different sediment samples, coming from different seaports. The experimental conditions were similar to those in the first set of experiments. The results of this set of experiment confirmed the indirect role of the Fe/S oxidizing bacteria and the key role of iron. Nevertheless, the results pointed out also that the mobilization efficiencies were metal specific and site-specific. Indeed, geochemical properties of the sediment (e.g., the buffering capacity) influence the biological activity of Fe/S oxidizing bacteria and the ability of metals to be stable in the solution phase. However, the kind of metal contaminants involved and the partitioning among the geochemical fractions of the sediment, were found to play an additional role in determining the final bio-mobilization efficiencies. Thus, the maintenance of acid and oxidative conditions, the chemical behavior of each metal species in aqueous environment and the geochemical characteristics of sediment interact intimately to determine the metal mobilization efficiencies. Considering the need for effective and cost-feasible bioremediation strategies, all these evidences need to be taken into account for the definition of sustainable ex-situ treatments. Dissertation outline The last set of experiments addressed the potential of the autochthonous microbial community in influencing the fate of metals in marine contaminated sediments, in connection with eventual in-situ bioremediation strategies. Biostimulation experiments were performed in anoxic conditions, with different amendants. Contrary to what is usually expected, our results pointed out that the biostimulation in anoxic conditions can lead to an increase in metal mobility and that the latter is not necessary associated to solubilization events. Indeed, the microbial community does drive the fate of metals and semi-metals, but biotic factors, like the composition of the microbial assemblages and the kind of biostimulation, produce effects that interacts with geochemical processes driven by the properties of the sediment, the kind of metal contamination and the environmental conditions. As a consequence, the interactions among all these factors are the true vector to determine the fate of metals and semi-metals in contaminated marine sediments. The main results obtained from the whole research were used to outline two conceptual models that summarize and describe the main biogeochemical interactions affecting the fate of (semi-)metals during treatments of bioaugmentation with Fe/S oxidizing bacteria and during biostimulation actions. Although they are simplistic representations of a highly complex set of processes that occur in the reality, the definition of conceptual models can improve our understanding of factors involved in this field. Indeed, a conceptual model that describes the biogeochemical interactions occurring during bioleaching was completely lacking, while the conceptual model obtained by the biostimulation experiments fills the gaps of the existing models and represents an improvement in the understanding of the processes occurring.

Texte intégralThe purpose of this Ph.D. project was to investigate the potential of the torrefaction treatment for upgrading low-value agro-industrial residues into useable solid fuels to be employed as high-quality energy carriers. The first phase of the project involved a screening of the agro-industrial residues available in Campania region (Italy) with good potentiality for energy applications. As a result of this analysis, tomato processing residues and olive mill residues, which have stood out as those in need of a more sustainable and environmental friendly disposal solution, were at first selected as biomass feedstocks for this Ph.D. project. However, practical difficulties encountered in the pre-treatment of the virgin olive husk (i.e., specifically in reducing the size of olives stone fragments which compose olive mill residues together with the olive pulp) led afterwards to discard such residue as a potential feedstock for the subsequent lab-scale torrefaction tests... Texte intégralThis PhD project deals with the development of innovative systems for the abatement of odorous emissions from industrial plants, focusing on two industrial sectors developed in Marche Region: the wood industry and vegetable waste recycling. The odor impact of industrial activities is a serious problem for the production activities themselves; indeed, the competent authorities for control are often forced to take action. The main cause of such impacts is not always due to an overcoming of the concentrations imposed by the legal limits; indeed, an ineffective urban planning in the past has led to industrial plants located near residential areas: olfactory harassment becomes reason for citizens reports, that hinder the correct operation of companies. The problem of odorous emissions mainly concerns the following companies (Bertoni, 1993): - Companies in the industrial sector (wood industry, chemical industry, oil and gas refineries, foundries, plastic and paint production); - Agri-food companies (meat processing); - Companies that deal with waste and wastewater treatment (landfills and composting companies). In particular, the main odorous compounds are (Bertoni, 1993): - VOCs (Volatile Organic Compounds): aliphatic and aromatic hydrocarbons, phenols, aldehydes and ketons; - VSCs (Volatile Sulfur Compounds): hydrogen sulfide (H2S) and mercaptans (SH); - Nitrogen compounds: ammonia (NH3) and amines. The odorous emission from industrial facilities can be the cause of annoyance to the people living in the surrounding area. A long-term exposure may bring up serious damage to human health such as nausea, headaches and other related respiratory problems (Lebrero et al., 2011; Zarra et al., 2009b). In this context, the development of effective innovative systems is fundamental, aimed at containing odors from the production processes. The present PhD research project has been cofunded by a company (Pan Eco srl), whose mission is to support the various industries in the field compliance with environmental regulations; in this sector, it obtains authorizations and develops projects aimed at respecting all environmental constraints. Considering the scenario just described and the company mission, the general goal of the research program was the development of innovative systems for the containment of odorous emissions into the atmosphere by various industrial plants. The general goal of the project has been achieved through specific objectives and activities: - definition of the state of the art of the systems currently applied for the abatement of atmospheric emissions of organics compounds, including the legislative context. Through bibliographic research, a summary was made regarding the main techniques for sampling, analysis and abatement of odor emissions, also referring to the main laws at national and European level; - identification of companies in the Marche region (at least two) with problems associated with the odor impact of industrial production. Two companies have been identified, one operating in the wood industry and the other operating in the recycling of organic waste; - design and implementation of innovative emissions abatement system. In both sectors taken into consideration, an innovative odor abatement system was identified and implemented: UV painting for wood sector and vermicomposting for the recycling of organic waste; - sampling, characterization of emissions and assessment of the performance. In both companies, the sampling of emissions using consolidated techniques was carried out and the effective abatement of odor emissions was assessed.

Texte intégralThe management of a huge quantity of waste from electric and electronic equipment (WEEE) represents a critical issue for the modern society. The negative environmental and health effects due to the improperly management are combined with the loss of valuable materials. The present work focused on the recovery of metals from WEEE with particular attention to indium from end-of-life liquid crystal displays (LCD). The experimental section allowed the optimization of a process that includes an acid leaching characterized by an innovative cross-current design, followed by a cementation with zinc powder. Considering the satisfying efficiencies obtained on the lab scale, higher than 90%, the whole process was studied from an environmental point of view comparing its emissions with those produced by the current management strategies (disposal in landfilling sites, incineration and traditional recycling). A life cycle assessment (LCA) of the different scenarios proved the significant advantage of recycling ways. Moreover, the traditional recycling resulted to be the most favorable, due for both the relevant water consumption of the innovative treatment and to the low indium content in the LCD. Nevertheless, a simple water recirculation system, combined with a physical indium upgrading in the waste, make the innovative option the best choice. The simple design of the optimized process allows its implementation in a mobile plant, built within the European project, HydroWEEE. The plant mobility prevents the impacts due to the waste transport, that contributes to the 30-40% of the currently treatments. Furthermore, this advantage is combined with the possibility to treat several WEEE for the recovery of different metals. The sustainability of this approach was proved by a LCA that highlighted the positive effect also in the comparison with the primary production, with a benefit between 20 and 80%. Last, but not least, the risk for workers in the real mobile plant was assessed.

Texte intégralThe deepened analysis of both European and Italian norms represented the starting point to evaluate the current implementation of legislation in Italy, with a particular focus on Ancona Province. This approach allowed the assessment of the current performance of MW collection and recycling system, highlighting the main criticalities connected to the biodegradable residue in the undifferentiated waste, intended for the disposal. A doubt emerged about the obligation of a treatment of undifferentiated waste, before the disposal in landfilling sites. Therefore, a real scale analysis was carried out selecting the Corinaldo landfilling site (which collects the MW of Ancona Province) as case study. The possibility of the real scale study represents a strength of the present research, compared to the available scientific literature, often referred to lab scale. The analysis evaluated the effect of the mechanical biological treatment (MBT) on MW, sent to Corinaldo facility. With this aim, the characteristics of disposal in two reference periods (before and after the MBT facility starting) were studied. The results did not identify significant improvements after the implementation of the MBT, considering the high efficiency of collection and recycling system in Ancona. The further environmental and economic assessments suggest that the best strategy of MW management should invest on recycling system, rather than the WM stabilization, in agreement with the circular economy principles. Starting from these observations, the manuscript presents a critical study of the technical criteria of the Italian norms suggesting a more deepened evaluation of parameters to assess the cases in which the MBT (before the disposal) produces real benefits on MW management. These evaluations should be able to balance the benefit with the MBT facility impact, both in environmental and economic terms, in the scenarios characterized by a virtuous recycling system.

Texte intégralThe increase of waste from electric and electronic equipment has pushed the research towards the development of high sustainability treatments for their exploitation. The end-of-life printed circuit boards (PCBs) represent one of the most significant waste in this class. The interest for these scraps is due to the high Cu and Zn concentrations, around 25% and 2% respectively. Biohydrometallurgical strategies are gaining increasing prominence, for the possibility to decrease the environmental costs. Nevertheless, these techniques show the main limit due to the low treated PCB amount, which makes unsustainable the further scale-up. To overcome this criticality, the present research introduces an innovative bioleaching process carried out by At. ferrooxidans. The developed technology allows to reach high PCB concentration thanks to a high efficiency two-step design, able to reduce the metal toxicity on the bacteria metabolism. The treatment uses the Fe3+ generated by bacterial oxidation, as oxidant, to leach Cu and Zn from PCBs. The possibility to overcome the solid concentration criticality is combined with high yield of 95% for Cu and Zn. The best selected conditions are: 30°C, pulp density of 5% (w/v), 10 g/L of Fe2+, time of treatment 11 days. A mathematical model to study the kinetic of the chemical reaction for the Cu leaching from PCBs, by Fe3+, is reported and an activation energy of 18-25 kJ/mol is estimated. The mechanism of the bioleaching process is studied in detail, integrating the previous model results with two differential equation to describe the bacteria growth and metabolism rate. The developed model is consistent with a R2 higher than 0.97. The results confirm the positive effect of the new innovative process. The determined mathematical model, suitable for the implementation at industrial scale, could be an important tool to predict the bioleaching mechanism. In the next step, the research is focused on the selective metal recovery from the leaching solution. The best identified conditions include: the Fe precipitation with NaOH, followed by the Cu cementation with Zn and a final Zn precipitation with oxalic acid. The metals show recovery efficiencies and purities higher than 95%. The experimental results are further enhanced by the carbon footprint assessment which proved the environmental advantage, compared to both the reference chemical treatment through Fe3+ and literature processes (hydrometallurgical and bioleaching approaches). The environmental impact assessment drives the optimization and improvement of the developed process in laboratory scale. It is used to identify the best conditions for the bioleaching process able to make the treatment actually sustainable. Overall, the environmental assessment identifies, as main criticalities to solve: the high energy demand of bioleaching process and the raw materials demand of the following recovery steps. These preliminary results have determined the choices in the further experiments. More in detail, the bioleaching process is improved by the increase of treated PCB concentration from 5% to 10% (w/v) maintaining a leaching efficiency higher than 95% for Cu and Zn. This technical improvement is translated into the halved of energy demand. Furthermore, the environmental load of recovery can be reduced by a double strategy: avoiding Zn recovery or substituting the hydrometallurgical techniques by electrochemical approaches. In the last chapter, a sustainable process, which uses the fungal strain Aspergillus niger for metal extraction from PCBs, is described. The best identified conditions are PCB addition after 14 days, Fe3+ as oxidant agent, and a pulp density of 2.5% (w/v). Extraction efficiencies of 60% and 40% for Cu and Zn, respectively, are achieved after 21 days of fermentation. The ecodesign of the process is further enhanced by using milk whey as substrate for the fungal growth and the consequent citric acid production.

Texte intégralNonostante i processi batch siano relativamente semplici da configurare e da gestire anche con un livello limitato di automazione e una conoscenza ridotta dei meccanismi che ne stanno alla base, spesso è difficile assicurare una qualità del prodotto finito riproducibile ed elevata. La strumentazione comunemente utilizzata nella pratica industriale riesce solo raramente a fornire misure in tempo reale della qualità di un prodotto. Inoltre, molte complicazioni nascono dalla natura multivariata della qualità, la quale dipende da una serie di parametri fisici, operativi o addirittura soggettivi. Sebbene le informazioni sulla qualità del prodotto non siano facilmente accessibili, esse sono racchiuse nelle variabili di processo abitualmente registrate dai calcolatori di processo e memorizzate in banche di dati storici. I metodi statistici multivariati permettono di ridurre la dimensione del problema proiettando le variabili di processo in uno spazio di dimensioni ridotte costituito di variabili fittizie che sono in grado di mantenere tutto il contenuto informativo sulla qualità, superando i problemi del rumore di misura delle variabili, della ridondanza e dell’elevato grado di correlazione. Inoltre, questi metodi sono in grado di trattare dati anomali o dati mancanti. Lo scopo di questa Tesi di Dottorato è di sviluppare dei sistemi innovativi per il monitoraggio della qualità di prodotti dall’alto valore aggiunto mediante tecniche statistiche multivariate. In particolare, i contributi scientifici di questo progetto di Dottorato sono: • l’elaborazione di tecniche per lo sviluppo di sensori virtuali per la stima in tempo reale della qualità del prodotto in sistemi produttivi di tipo batch; • l’applicazione non convenzionale di tecniche di proiezione su sottospazi latenti al fine di prevedere la durata di un batch o delle relative fasi operative; • lo sviluppo di metodiche innovative per il monitoraggio multirisoluzione e multivariato della qualità mediante l’analisi di immagini di un prodotto dall’alto valore aggiunto. Innanzi tutto, in questa Tesi vengono proposti sensori virtuali per la stima in linea della qualità del prodotto. Essi sono stati sviluppati e implementati prendendo in considerazione il caso di studio un processo industriale reale per la produzione di resine mediante polimerizzazione batch. I sensori virtuali proposti sono basati sulla tecnica statistica multivariate della proiezione su strutture latenti (PLS), che opera una regressione delle misure di processo usualmente disponibili in linea in tempo reale. Questo sistema riesce a garantire una accuratezza delle stime della qualità che è dello stesso ordine di grandezza delle misure di qualità fatte in laboratorio, col vantaggio che le stime in linea sono disponibili con altissima frequenza (sull’ordine di grandezza di s-1), cioè una frequenza centinaia di volte superiore delle misure che possono essere fatte in laboratorio (sull’ordine di grandezza di h-1). Inoltre, le stime sono accessibili in tempo reale e senza il ritardo che è tipico delle misure di laboratorio. Al fine di compensare le non linearità dei dati e i cambiamenti nella struttura di correlazione fra le variabili, la procedura adottata divide il batch in una sequenza di un numero limitato di fasi di stima, all’interno delle quali lo stimatore virtuale è in grado di dare stime molto accurate per mezzo di modelli PLS lineari. Il passaggio da una fase a quella successiva avviene in corrispondenza di alcuni “eventi” facilmente riconoscibili nelle stesse variabili di processo. La caratteristica principale del sensore virtuale proposto è che esso tiene conto di informazioni sulla dinamica del processo per mezzo di modelli a “variabili ritardate” (i quali aggiungono informazioni sulla dinamica del processo da valori passati delle variabili di processo) o modelli a media mobile. Il filtro a media mobile aggiunge una “memoria temporale” al sensore virtuale che migliora l’accuratezza di stima e, mediando le variabili di processo all’interno di una finestra temporale di dimensione fissata, riesce ad eliminare il rumore di misura, attenuare il rumore di processo, appiattire valori anomali e compensare l’effetto di temporanee mancanze di dati. L’ampiezza della finestra deve comunque essere scelta con cautela, dato che una finestra temporale troppo larga potrebbe ritardare gli allarmi sull’attendibilità della stima. Da un punto di vista operativo, il sistema proposto aiuta il personale che opera nell’impianto a rilevare delle derive sulla qualità del prodotto, suggerisce tempestivamente le correzioni da apportare alla ricetta del processo, e aiuta a minimizzare i fuori specifica del prodotto finale. Inoltre, il numero di campioni per la misura della qualità in laboratorio può essere ridotto drasticamente, la qual cosa determina un guadagno sia sul tempo totale del batch, sia sui costi relativi al laboratorio che alla manodopera e alla sua organizzazione. Anche una seconda tipologia di sensori virtuali è stata sviluppata per assistere il monitoraggio in linea della qualità del prodotto e per fornire informazioni utili per una programmazione efficace della produzione: un sensore virtuale per la previsione in tempo reale della durata del batch. Questa strategia di monitoraggio si basa su modelli PLS evolutivi che sfruttano le informazioni progressivamente raccolte nel tempo durante il batch per prevedere la durata del batch o di ciascuno dei relativi stadi operativi. Anche l’accuratezza ottenuta dalle previsioni ottenute con questo sensore virtuale è del tutto soddisfacente, dato che l’errore di previsione è molto inferiore sia alla variabilità delle durata del batch che alla durata dei turni di lavoro degli operatori. Inoltre, la parte iniziale del batch conferma di essere di importanza fondamentale per la durata, in quanto le condizioni iniziali delle attrezzature, lo stato delle materie prime, e la fase di riscaldamento iniziale del reattore esercitano una grandissima influenza sulle prestazioni del batch stesso. Le informazioni che si ricavano sulla durata con grande anticipo rispetto alla fine del batch permettono una migliore organizzazione degli interventi sull’impianto, degli operatori d’impianto e dell’utilizzazione delle apparecchiature. L’efficacia dei sensori per la stima della qualità e per la previsione della durata del batch è stata verificata applicandoli ed implementandoli in linea nel caso della produzione di resine mediante polimerizzazione batch. Infine, i metodi statistici multivariati sono stati utilizzati anche nel campo dell’analisi dell’immagine. Abitualmente, nella pratica industriale, le ispezioni di un prodotto mediante analisi dell’immagine vengono svolte con semplici misurazioni dei più importanti parametri fisici opportunamente messi in evidenza per mezzo di tecniche di filtrazione. Inoltre, queste misure vengono ottenute in modo non sistematico. Molte informazioni utili restano però “nascoste” nelle immagini. Queste permettono di identificare la natura complessa della qualità del prodotto finale. Per questo è stato sviluppato un sistema totalmente automatizzato per il monitoraggio in tempo reale da immagini di un manufatto dall’alto valore aggiunto. Questo sistema di monitoraggio basato su tecniche multirisoluzione e multivariate è stato applicato al caso della caratterizzazione della superficie di un semiconduttore dopo fotolitografia, un’operazione fra le più importanti nella fabbricazione di circuiti integrati. Tecniche avanzate di analisi multivariata dell’immagine estraggono le tracce che il processo lascia sul prodotto, aiutando sia il rilevamento di situazioni critiche nel processo che l’intervento con azioni correttive a neutralizzare eventuali problemi. L’approccio proposto in questa Tesi si basa su un filtraggio preliminare multirisoluzione dell’immagine mediante wavelet, seguito da uno schema di monitoraggio che conduce in parallelo un’analisi della rugosità superficiale e della forma della superficie di un prodotto. Ad esempio, la rugosità della superficie può essere esaminata con una analisi delle componenti principali “nidificata”. Questa è una strategia che si articola su due differenti livelli: il livello esterno che permette di discriminare parti differenti della superficie per mezzo di una analisi dei gruppi con PCA; il livello interno esegue il monitoraggio della rugosità superficiale con PCA. La forma della superficie viene analizzata per mezzo di un approccio PCA a “finestra mobile nello spazio”, il quale coglie l’informazione dell’immagine secondo il relativo ordine nello spazio e riesce anche a tener conto sia delle non linearità che delle differenze strutturali della superficie. Questo sistema è in grado di rilevare alcune delle caratteristiche qualitative del prodotto che abitualmente non sono accessibili senza richiedere l’intervento dell’uomo. Inoltre, il monitoraggio risulta essere veloce, attendibile e non ambiguo, ed esegue una scansione di un’immagine del prodotto localizzando in modo preciso difetti e anomalie e rilevando eventuali derive del processo. In conclusione, nonostante le metodologie proposte siano state testate su specifici casi di studio, esse hanno dimostrato di essere generali e vantano un grande potenziale. Per questo si ritiene sia possibile estenderle a differenti campi di ricerca e a diverse applicazioni industriali (ad esempio: ingegneria alimentare; industria farmaceutica; biotecnologie; etc…), nonché a differenti scale di indagine, dalla scala macroscopica alla microscopica o nanoscopica.

Texte intégralThe intensification of extreme natural events, caused by climate change, together with strong urbanization, makes it essential for society to be prepared to cope with these emergency situations. The main priority after a natural event, that hit an anthropized area, is to save all human lives involved in the disaster. Subsequently, it will be necessary to restore a normal situation. Events such as floods, landslides and earthquakes generate a huge amount of debris and rubbles, creating numerous problems during emergency activities. To remedy this, it will be necessary to clean up the affected area as soon as possible, from this material. To ensure that the disaster waste management is efficient, it is necessary to have a protocol or at least a guideline to follow. In the world, a lot of protocols exist to manage the emergency situation after the natural event, but there is not enough information for the disaster waste management. This lack drives local authorities to use empirical management methods. The empirical method is based on experience in the field and does not take into account a scientific methodological approach, that analyses the environmental and economic impact generated by the disaster waste management. The guiding principles should be the reduction of the environmental footprint and of management costs; however, the emergency does not allow to carry out such analyses before choosing a strategy for the management of waste, due to the lack of time.

Texte intégralLa recente introduzione del concetto di Quality-by-Design (QbD) da parte della Food and Drug Administration e delle altre agenzie di regolamentazione farmaceutica ha l’obiettivo di migliorare e modernizzare gli approcci tradizionalmente utilizzati dalle industrie farmaceutiche per lo sviluppo di nuovi prodotti e dei relativi processi produttivi. Scopo dell’iniziativa è di incoraggiare le industrie stesse all’utilizzo di procedure sistematiche e basate su presupposti scientifici sia nella fase di sviluppo di prodotto e processo, che nella fase di conduzione del processo produttivo stesso. A tal proposito, le Agenzie hanno definito paradigmi e linee guida per agevolare l’implementazione di queste procedure in ambito industriale, favorendo una migliore comprensione dei fenomeni alla base dei processi produttivi, in maniera da assicurare un controllo stringente sulla qualità dei prodotti finali, in termini di proprietà fisiche, ma soprattutto di efficacia e sicurezza per i pazienti. Da un punto di vista ingegneristico, il Quality-by-Design può essere visto come il tentativo di introdurre principi di modellazione in ambiti di sviluppo e di produzione farmaceutica. Questo offre enormi opportunità all’industria farmaceutica, che può beneficiare di metodologie e strumenti ormai maturi, già sperimentati in altri settori industriali maggiormente inclini all’innovazione tecnologica. Allo stesso tempo, non va tralasciato il fatto che l’industria farmaceutica presenta caratteristiche uniche, come la complessità dei prodotti, le produzioni tipicamente discontinue, diversificate e in bassi volumi e, soprattutto, lo stretto controllo regolatorio, che richiedono strumenti dedicati per affrontare i problemi specifici che possono sorgere in tale ambiente. Per questi motivi, vi è l’esigenza di concepire metodologie che siano adeguate alle peculiarità dell’industria farmaceutica, ma al tempo stesso abbastanza generali da poter essere applicate in un’ampia gamma di situazioni. L’obiettivo di questa Dissertazione è dimostrare come la modellazione statistica, e in particolar modo i modelli a variabili latenti (LVM, latent variable models), possano essere utilizzati per guidare l’implementazione pratica dei principi fondamentali del Quality-by-Design in fase di sviluppo di prodotto e di processo e in fase di produzione in ambito farmaceutico. In particolare, vengono proposte metodologie generali per l’impiego di modelli a variabili latenti nelle tre aree principali sulle quali l’iniziativa del Quality-by-Design si fonda: il miglioramento della comprensione sui processi, la progettazione di nuovi prodotti e processi produttivi, e il monitoraggio e controllo di processo. Per ciascuna di queste aree, l’efficacia della modellazione a variabili latenti viene dimostrata applicando i modelli in diversi casi studio di tipo industriale, di laboratorio, o simulati. Per quanto riguarda il miglioramento della comprensione sui processi, nel Capitolo 3 è proposta una strategia generale per applicare LVM nello sviluppo di sistemi di produzione in continuo. L’analisi è applicata a supporto dello sviluppo di un processo industriale continuo di produzione di compresse su scala pilota. La procedura si basa su tre fasi fondamentali: i) una fase di gestione dei dati; ii) una fase di analisi esplorativa; iii) una fase di analisi globale. Viene mostrato come i parametri dei modelli costruiti a partire dai dati del processo possano essere interpretati sulla base di principi fisici, permettendo di identificare le principali forze motrici che agiscono sul sistema e di ordinarle a seconda della loro importanza. Questo può essere utile per supportare una valutazione dei rischi necessaria a definire una strategia di controllo per il processo e per guidare la sperimentazione fin dalle prime fasi dello sviluppo. In particolare, nel caso studio considerato, la metodologia proposta individua nel processo utilizzato per macinare le particelle di principio attivo e nella sezione nella quale il principio attivo è formulato le principali fonti di variabilità entranti nel sistema con effetto sulle proprietà fisiche del prodotto finale. Dall’analisi globale, è mostrato come l’utilizzo di modelli a variabili latenti a blocchi multipli permetta di individuare le unità del processo più critiche e, all’interno di ciascuna di esse, le variabili più critiche per la qualità del prodotto. Inoltre questi modelli si dimostrano particolarmente utili nell’identificare le traiettorie lungo le quali il processo si muove, a seconda delle proprietà delle materie prime e dei parametri di processo utilizzati, fornendo così uno strumento per garantire che l’operazione segua la traiettoria designata. Nell’ambito della progettazione di nuovi prodotti e processi, l’efficacia dei modelli a variabili latenti è dimostrata nel Capitolo 4, dove è proposta una procedura generale basata sull’inversione di LVM per supportare lo sviluppo di nuovi prodotti e la determinazione delle condizioni operative dei rispettivi processi di produzione. L’obiettivo della procedura proposta è quello di fornire uno strumento atto a dare un’adeguata formalizzazione matematica, in termini di inversione di LVM, al problema di progettazione, secondo gli obiettivi e i vincoli che il problema stesso può presentare. Dal momento che l’inversione di LVM può avere soluzioni multiple, vengono individuati quattro possibili problemi di ottimizzazione, tramite i quali effettuare l’inversione. L’obiettivo dell’inversione del modello è di stimare le condizioni ottimali in ingresso al sistema (in termini, per esempio, di caratteristiche delle materie prime o di parametri di processo) che assicurino di raggiungere la qualità desiderata per il prodotto in uscita. La procedura è applicata con successo in un caso studio industriale, per la determinazione delle proprietà delle materie prime in ingresso a un processo di granulazione a umido, con l’obiettivo di ottenere in uscita granuli con determinate caratteristiche di qualità. È inoltre esaminato il concetto di spazio nullo, lo spazio cioè cui appartengono tutte le soluzioni di un problema di inversione di LVM, che corrispondono ad uno stesso insieme di variabili desiderate (proprietà del prodotto) in uscita. In particolare, si dimostra come la definizione di spazio nullo presenti diverse caratteristiche comuni alla definizione di spazio di progetto (design space) di un processo, stabilita dalle linee guida delle Agenzie di regolamentazione, e come lo spazio nullo possa essere utilizzato al fine di una identificazione preliminare dello spazio di progetto. Al fine di avere una misura sull’affidabilità delle soluzioni del problema di inversione, viene proposta una strategia per stimarne le incertezze. Sono inoltre presentate alcune soluzioni per affrontare questioni specifiche relative all’inversione di LVM. In particolare, si propone una nuova statistica da utilizzare per la selezione del numero di variabili latenti da includere in un modello utilizzato per l’inversione, in modo tale da descrivere adeguatamente l’insieme dei regressori, oltre a quello delle variabili in uscita. In aggiunta, dato che a causa delle possibili incertezze del modello non è assicurato che la sua inversione fornisca una soluzione che consenta di ottenere le proprietà desiderate per il prodotto, è proposta una strategia per sfruttare la struttura di covarianza dei dati storici per selezionare nuovi profili di qualità per il prodotto, in modo da facilitare l’inversione del modello. Gli approcci proposti sfruttano i parametri del modello e i vincoli imposti per la qualità del prodotto per stimare nuovi insiemi di proprietà, per i quali l’errore di predizione del modello è minimo. Questo agevola l’inversione del modello nel fornire le proprietà del prodotto desiderate, dal momento che queste possono essere assegnate come vincoli rigidi al problema di ottimizzazione. Nel Capitolo 5 la procedura presentata al Capitolo 4 per l’inversione di LVM è applicata per progettare la formulazione di nuovi prodotti farmaceutici, in cui l’obiettivo è di stimare i migliori eccipienti da miscelare con un dato principio attivo e la loro quantità in modo da ottenere una miscela di proprietà adeguate per la fase di compressione. La procedura proposta al Capitolo 4 è ampliata al fine di includere i vincoli per la selezione dei materiali e di considerare gli specifici obiettivi che un problema di formulazione può presentare (per esempio, massimizzare la dose di principio attivo, o minimizzare il peso della compressa finale). L’inversione del modello è risolta come problema di programmazione non lineare misto-intera, per il quale è sviluppata un’interfaccia utente che consenta ai formulatori di specificare gli obiettivi e i vincoli che il problema di formulazione da risolvere può presentare. La metodologia proposta è testata in un caso studio industriale per progettare nuove formulazioni per un dato principio attivo. Le formulazioni progettate in-silico sono preparate e verificate sperimentalmente, fornendo risultati in linea con le predizioni del modello. Nel Capitolo 6 è presentata una diversa applicazione della procedura generale per l’inversione di LVM presentata al Capitolo 4. Il caso studio riguarda un problema di trasferimento di prodotto, in cui l’obiettivo è di ottenere nanoparticelle di diametro medio predefinito, tramite un processo di precipitazione con anti-solvente in un dispositivo obiettivo. La metodologia sfrutta i dati storici disponibili da esperimenti effettuati su un dispositivo di riferimento di diversa dimensione da quello obiettivo, e sullo stesso dispositivo obiettivo ma con una diversa configurazione sperimentale. Un modello di tipo joint-Y PLS (JY-PLS) è inizialmente utilizzato per correlare dati di diversa origine (per dispositivo e configurazione sperimentale). Quindi, la procedura presentata al Capitolo 4 viene impiegata per invertire il modello JY-PLS al fine di determinare le condizioni operative nel dispositivo obiettivo, che assicurino l’ottenimento di nanoparticelle di diametro medio desiderato. La convalida sperimentale conferma i risultati ottenuti dall’inversione del modello. Inoltre gli esperimenti consentono di convalidare sperimentalmente il concetto di spazio nullo, dimostrando come diverse condizioni di processo stimate lungo lo spazio nullo consentano effettivamente di ottenere nanoparticelle con le medesime dimensioni medie. La sezione finale di questa Dissertazione propone l’applicazione di LVM a supporto del monitoraggio e controllo di processo in operazioni farmaceutiche. In particolare, nel Capitolo 7, è affrontato il problema del trasferimento di modelli per il monitoraggio di processo tra impianti diversi. In questo caso il problema è di assicurare che l’operazione in un impianto obiettivo sia sotto controllo statistico fin dai primi istanti di funzionamento dell’impianto, sfruttando la conoscenza disponibile (in termini di dati) da altri impianti. È proposta una procedura generale basata su LVM per far fronte a questo tipo di problemi. La procedura identifica cinque diversi scenari, a seconda del tipo di informazioni disponibili (solo dati di processo o sia dati di processo sia conoscenza di base sul processo), della provenienza dei dati disponibili (solo dall’impianto di riferimento o sia dall’impianto di riferimento sia dall’impianto obiettivo) e dal tipo di variabili di processo considerate per la costruzione del modello (solo variabili comuni tra gli impianti o sia variabili comuni sia altre variabili). Per modellare in maniera congiunta i dati disponibili da impianti diversi, sono utilizzate analisi delle componenti principali (PCA) o modelli di tipo JY-PLS, a seconda che, per la costruzione del modello di monitoraggio, si considerino solo variabili comuni tra gli impianti (nel caso PCA), o sia variabili comuni sia altre variabili (nel caso JY-PLS). Le metodologie proposte sono verificate nel trasferimento di modello per il monitoraggio di un processo industriale di atomizzazione, dove il riferimento è un impianto su scala pilota, mentre l’impianto obiettivo è un’unità produttiva su scala industriale. Le prestazioni in fase di monitoraggio del processo su scala industriale sono soddisfacenti per tutti gli scenari proposti. In particolare, è dimostrato come il trasferimento di informazioni dall’impianto di riferimento migliori le prestazioni del modello per il monitoraggio dell’impianto obiettivo. Le procedure proposte sono inoltre applicate in uno studio preliminare per il trasferimento di sistemi di monitoraggio in processi discontinui, considerando come caso studio un processo simulato di fermentazione per la produzione di penicillina, in cui sono simulati due impianti differenti per scala e configurazione. Le prestazioni del sistema di monitoraggio indicano che, anche in questo caso, considerare nella costruzione del modello i dati disponibili dalle operazioni nell’impianto di riferimento rende il sistema più efficiente nella rilevazione delle anomalie simulate nell’impianto obiettivo, rispetto a considerare nel modello di monitoraggio i soli (pochi) dati disponibili dall’impianto obiettivo stesso.

Texte intégralIn un sistema economico globalizzato come quello in cui viviamo, garantire elevati standard in termini di qualità di prodotto costituisce per ogni azienda produttiva un fattore di successo. Monitorare in modo accurato la qualità del prodotto lungo tutti gli stadi della filiera produttiva, tuttavia, è divenuto progressivamente più complesso a causa della dimensione globale che quest’ultima ha assunto. É questo un effetto dei fenomeni di delocalizzazione della produzione, legati alla necessità delle aziende di non perdere quote di mercato a discapito di paesi emergenti caratterizzati da costi di produzione inferiori. In un tal sistema, aumenta anche il rischio di frodi, adulterazioni e contraffazioni del prodotto. Per certe categorie di prodotti, come quelli alimentari e farmaceutici, tali attività non solo danneggiano i consumatori dal punto di vista economico, ma possono anche causare seri problemi di salute. Nonostante la grande importanza del monitorare la qualità di prodotto, a livello industriale si è ancora lontani da un sistema che permetta di caratterizzarla in modo rapido, economico, non invasivo (e quindi utilizzabile in tempo reale), riproducibile e multivariato (cioè in grado di quantificare contemporaneamente più parametri di qualità). Le tecniche che si sono dimostrate più promettenti in tal senso sono la spettroscopia d’assorbimento nella regione del visibile e del vicino infrarosso e l’analisi d’immagine. Per analizzare la moltitudine di dati (spettri e immagini) caratterizzati da forti correlazioni che queste generano, è necessario ricorrere a tecniche statistiche apposite, in particolare i modelli a variabili latenti (LVM, latent variable models). Tali tecniche, che sono pensate per trattare tali tipologie di dati, nascono dall’assunto che un sistema possa essere descritto mediante pochi fattori (detti anche variabili latenti) esprimibili come combinazione lineare delle variabili originali e interpretabili sulla base dei fenomeni chimico/fisici che interessano il sistema. Il numero di applicazioni di LVM nel campo della caratterizzazione di prodotti alimentari e farmaceutici è cresciuto rapidamente negli ultimi anni. La maggior parte dei contributi pubblicati, tuttavia, offre soluzioni a specifici problemi anziché fornire approcci generali. L’obiettivo di questa Dissertazione è quello di superare alcune delle limitazioni esistenti al fine di favorire la diffusione di questi strumenti nella comune pratica industriale. La ricerca presentata si suddivide in tre macro aree di applicazione, che si differenziano a seconda della tecnica utilizzata per caratterizzare la qualità di prodotto, e cioè l’analisi d’immagine, la spettroscopia, e la fusione di dati (data fusion), cioè la combinazione delle informazioni provenienti da più strumenti analitici. Per ciascuna di queste aree, l’efficacia della modellazione a variabili latenti viene dimostrata applicando i modelli in diversi casi studio di tipo industriale o di laboratorio. Con riferimento all’analisi d’immagine, vengono proposte applicazioni nel campo farmaceutico. Nel Capitolo 3, l’analisi d’immagine viene utilizzata per il miglioramento della comprensione di un processo industriale di rivestimento di compresse. In tale processo la qualità finale del prodotto, che è legata all’omogeneità del rivestimento e al grado di erosione superficiale, viene tradizionalmente valutata da un panel di esperti, che necessariamente fornisce un giudizio soggettivo e poco riproducibile. Inizialmente, il Capitolo discute come, a partire da immagini del prodotto finito, sia possibile valutare in modo quantitativo e riproducibile i parametri di qualità. Le metriche sviluppate vengono quindi utilizzate per il troubleshooting del processo stesso, con il fine di indagare il meccanismo che porta all’erosione superficiale. A tal scopo, le metriche vengono correlate ai parametri di processo tramite un modello a variabili latenti, e i parametri del modello vengono utilizzati per definire le condizioni operative ottimali da utilizzare per garantire un prodotto in specifica. Il Capitolo 4, usando ancora come pretesto un processo di rivestimento di compresse, discute in modo critico il problema della riproducibilità dei risultati ottenuti tramite analisi d’immagine. Tale riproducibilità, infatti, è garantita solamente se le condizioni sperimentali utilizzate per raccogliere le immagini destinate alla calibrazione del modello di stima della qualità vengono mantenute inalterate. Tali condizioni includono il sistema di illuminazione e la fotocamera stessa. Viene proposto innanzitutto un modello per il monitoraggio dello stato dell’apparato sperimentale, da utilizzare ogniqualvolta viene avviata una campagna di controllo qualità e basato semplicemente sull’acquisizione di un’immagine di standard colorati. In caso venga rilevato un cambiamento, viene proposta una strategia per adattare il modello di stima della qualità alle nuove condizioni. I risultati dimostrano l’efficacia della strategia proposta, che si basa su una tecnica già nota nel contesto della sincronizzazione vocale e dell’allineamento di traiettorie temporali in processi produttivi di tipo batch. Con riferimento alla spettroscopia d’assorbimento, le applicazioni presentate riguardano prodotti alimentari, con particolare attenzione alle tecnologie per la rilevazione rapida di frodi di sostituzione (di un prodotto avente un certo valore di mercato con uno a valore di mercato inferiore). Nel Capitolo 5, viene presentata una nuova tecnica per la classificazione di dati spettrali, che ha l’obiettivo di razionalizzare il pretrattamento cui i dati stessi sono generalmente sottoposti. I risultati dimostrano come la tecnica proposta garantisca di ottenere la stessa accuratezza di altri metodi, senza tuttavia ricorrere a procedure di tipo trial-and-error, onerose in termini computazionali, per la scelta del miglior pretrattamento. Nel Capitolo 6, accanto a due applicazioni di autenticazione di prodotti alimentari (filetti pescati di branzino e formaggio Asiago d’allevo) tramite spettroscopia, viene presentata una tecnica multi specie per la rilevazione di una tipica frode del settore ittico, cioè la sostituzione di filetti freschi con filetti decongelati. Rispetto al tradizionale approccio di costruire un modello di rilevazione della frode per ciascuna specie, lavorare con un modello multi specie (e, magari, indipendentemente dalla specie) riduce notevolmente i tempi e i costi necessari nella fase di calibrazione. Delle tre strategie proposte, quella che fornisce risultati migliori lavora decomponendo l’informazione contenuta nei dati spettrali in due componenti, una legata alla specie e una legata allo stato fresco o decongelato. La tecnica, convalidata su un numero di spettri molto maggiore rispetto alla applicazioni riportate in letteratura, si è dimostrata efficace anche nell’autenticazione di campioni di specie non utilizzate nella fase di calibrazione. Infine, con riferimento alla fusione di dati, il Capitolo 7 dimostra, attraverso due applicazioni in campo alimentare, come unire le informazioni ottenute da più strumenti analitici permetta di migliorare la caratterizzazione della qualità di un prodotto. La combinazione dei segnali a disposizione (detta low level, per distinguerla da altre tecniche di fusione di dati), opportunamente pesati, permette di ottenere risultati migliori rispetto all’utilizzo dei singoli segnali.

Texte intégralThe catalytic chemistry of gold has had a relatively belated development with respect to other late transition metals, and this has been attributed to the preconception that gold is expensive and unreactive. The interest in gold has grown over the last thirty years, because both of these conceptions have been proven false, and successful applications of gold catalysis have emerged in chemical processing, pollution control, fuel cells design, and many others fields. These evidences have sparked a veritable “gold rush” in the field of catalysis, both homogeneous and heterogeneous. We investigated the role of gold in both homogeneous and heterogeneous catalytic processes. In fact, the theoretical study of mechanistic details for reactions, that involves and underline the characteristics of gold, have been the subjects of this thesis. Density functional theory (DFT) is the method of choice in this kind of studies. Regarding heterogeneous catalysis the synthesis of vinylacetate is the reaction on which we have focused our attention. In particular, a bimetallic catalyst containing low Pd coverage on Au surface (100 and 111) has been selected to outline the reaction mechanism of VAM formation. We have studied in detail both mechanisms proposed in literature, in order to selected the more active surface and the more likely mechanism. The homogeneous catalytic process that has been selected to point out the catalytic activity of gold is the hydration of 1,2-diphenylacetylene to yield benzyl phenyl ketone, catalyzed by a complex of Au(I) with triphenylposphine. This cationic complex coordinates to the alkyne in the first step of the catalytic cycle, thus rendering it more susceptible for a nucleophilic attack. That reaction is a relatively new synthetic strategy that have recently studied experimentally. Our aim is to elucidate the mechanism of the whole reaction.

Texte intégralTo design advanced dosage forms, suitable carrier materials are used to overcome the undesirable properties of drug molecules. Hence various kinds of high-performance biomaterials are being constantly developed. From the viewpoint of the optimization of pharmacotherapy, drug release should be controlled in accordance with the therapeutic purpose and pharmacological properties of active substances. The main objective of the present thesis was to characterize the interactions between drugs and drug carriers by using combined molecular dynamics, molecular mechanics, and docking computational techniques. These simulations are likely to benefit the study of materials by increasing our understanding of their chemical and physical properties at a molecular level and by assisting us in the design of new materials and predicting their properties. Simulations are usually considerably cheaper and faster than experiments. Molecular simulations also offer a unique perspective on the molecular level processes controlling structural, physical, optical, chemical, mechanical, and transport properties. In particular the attention was put on cyclodextrinic carriers supported on membrane and molecularly imprinted polymers. Thus, structural information, such as the geometries of the cyclodextrinic complexes, and thermodynamic data, i.e. the variation of the enthalpy, were considered to draw a complete picture of the βCD-drug interactions. The results were in good agreement with the experimental data found in the measurement of stability constants. Finally the molecular dynamics on the polymeric system formed by adding on the surface of PEEK-WC the βCD-drug complex showed the release of the included drug in a water solution. The docking and molecular mechanics techniques provided also informations on the geometry and the energy of complexation of a β-cyclodextrin derivative with naringin showing that the driving force for the host-guest complexation is due to the van der Waals interaction. Moreover the molecular dynamics calculations provided details on the complexation of naringin on the PEEK-WC surface containing the β-cyclodextrin derivative. The binding affinity and selectivity of MIP towards drug template were calculated from the interaction energy between the ligand and the monomers and from docking simulations, respectively, as also the number of hydrogen bonds was determined. Our computational results shown a higher interaction energy between the drug template and monomers and justified the experimental data of selective recognition and rebinding of the template in terms of MIP performance confirming the reliability of our computational method. Moreover the diffusion coefficient of 5-FU into a PMAA matrix on the release step was determined.Thus, atomistic modelling of material structure was a tool for understanding the mechanisms of physical processes on atomic and molecular levels, gaining insights into the molecular origins of behaviour of bulk polymers.

Texte intégralDensity functional theory calculations were performed to study the ability of thorium (Th+, Th2+) and uranium (U+, U2+ The potential energy surfaces were explored taking into consideration different spin states. A close description of the reaction pathways leading to different reaction products is presented, and the obtained results are compared with experimental data. ) cations to activate the C-H and C-C bonds of methane, ethane and propane in the gas-phase. Th+ activates the C-H bonds of methane and ethane, in contrast, U+ is inert in both reactions. Th2+ reacts with all three alkanes, whereas U2+ reacts with C2H6 and C3H8, with product distributions different than those of Th2+ The computed potential energy profiles, which all proceed by insertion, were used to evaluate the relationship between the energetics of the bare Th . + (2+) and U+ (2+) ions and the energies for C-H and C-C bond activation. It was found that the computed energetics for insertion are entirely consistent with the empirical model which relates insertion efficiency to the energy needed to promote the An+ (2+) ion from its ground state to a prepared divalent state with two non 5f valence electrons suitable for bond formation in {C-An+ (2+)-H} and {C-An+ (2+)-C} activated intermediates.
