Ceramide kinase (CerK) is a lipid kinase that converts the proapoptotic ceramide to ceramide 1-phosphate, which has been proposed to have pro-malignant properties and regulate cell responses such as proliferation, migration, and inflammation. We used the parental human breast cancer cell line MDA-MB-231 and two single cell progenies derived from lung and bone metastasis upon injection of the parental cells into immuno-deficient mice. The lung and the bone metastatic cell lines showed a marked upregulation of CerK mRNA and activity when compared to the parental cell line. The metastatic cells also had increased migratory and invasive activity, which was dose-dependently reduced by the selective CerK inhibitor NVP-231. A similar reduction of migration was seen when CerK was stably downregulated with small hairpin RNA (shRNA). Conversely, overexpression of CerK in parental MDA-MB-231 cells enhanced migration, and this effect was also observed in the non-metastatic cell line MCF7 upon CerK overexpression. On the molecular level, CerK overexpression increased the activation of protein kinase Akt. The increased migration of CerK overexpressing cells was mitigated by the CerK inhibitor NVP-231, by inhibition of the phosphoinositide 3-kinase (PI3K)/Akt pathway and the Rho kinase, but not by inhibition of the classical extracellular signal-regulated kinase (ERK) pathway. Altogether, our data demonstrate for the first time that CerK promotes migration and invasion of metastatic breast cancer cells and that targeting of CerK has potential to counteract metastasis in breast cancer.

Prostate cancer (PCa) is one of the most prevalent cancers in men. Androgen receptor signaling plays a major role in this disease, and androgen deprivation therapy is a common therapeutic strategy in recurrent disease. Sphingolipid metabolism plays a central role in cell death, survival, and therapy resistance in cancer. Ceramide kinase (CERK) catalyzes the phosphorylation of ceramide to ceramide 1-phosphate, which regulates various cellular functions including cell growth and migration. Here we show that activated androgen receptor (AR) is a repressor of CERK expression. We undertook a bioinformatics strategy using PCa transcriptomics datasets to ascertain the metabolic alterations associated with AR activity. CERK was among the most prominent negatively correlated genes in our analysis. Interestingly, we demonstrated through various experimental approaches that activated AR reduces the mRNA expression of CERK: (i) expression of CERK is predominant in cell lines with low or negative AR activity; (ii) AR agonist and antagonist repress and induce CERK mRNA expression, respectively; (iii) orchiectomy in wildtype mice or mice with PCa (harboring prostate-specific Pten deletion) results in elevated Cerk mRNA levels in prostate tissue. Mechanistically, we found that AR represses CERK through interaction with its regulatory elements and that the transcriptional repressor EZH2 contributes to this process. In summary, we identify a repressive mode of AR that influences the expression of CERK in PCa.

Ceramide kinase (CerK) plays a critical role in the regulation of cell growth and survival and has been implicated in proinflammatory responses. In this work, we demonstrate that CerK regulates adipocyte differentiation, a process associated with obesity, which causes chronic low-grade inflammation. CerK was upregulated during differentiation of 3T3-L1 preadipocytes into mature adipocytes. Noteworthy, knockdown of CerK using specific siRNA to silence the gene encoding this kinase resulted in substantial decrease of lipid droplet formation and potent depletion in the content of triacylglycerols in the adipocytes. Additionally, CerK knockdown caused blockade of leptin secretion, an adipokine that is crucial for regulation of energy balance in the organism and that is increased in the obese state. Moreover, CerK gene silencing decreased the expression of peroxisome proliferator-activated receptor gamma (PPARγ), which is considered the master regulator of adipogenesis. It can be concluded that CerK is a novel regulator of adipogenesis, an action that may have potential implications in the development of obesity, and that targeting this kinase may be beneficial for treatment of obesity-associated diseases.

CerK (ceramide kinase) produces ceramide 1-phosphate, a sphingophospholipid with recognized signalling properties. It localizes to the Golgi complex and fractionates essentially between detergent-soluble and -insoluble fractions; however, the determinants are unknown. Here, we made a detailed mutagenesis study of the N-terminal PH domain (pleckstrin homology domain) of CerK, based on modelling, and identified key positively charged amino acid residues within an unusual motif in the loop interconnecting β-strands 6 and 7. These residues are critical for CerK membrane association and polyphosphoinositide binding and activity. Their mutagenesis results in increased thermolability, sensitivity to proteolysis, reduced apparent molecular mass as well as propensity of the recombinant mutant protein to aggregate, indicating that this loop impacts the overall conformation of the CerK protein. This is in contrast with most PH domains whose function strongly relies on charges located in the β1–β2 loop.

Abstract We showed that ceramide-1-phosphate (C1P) is formed by ceramide kinase (CERK). In this study the mechanism by which C1P enhanced phagocytosis and phagolysosomal formation in COS-1 cells expressing hCERK was evaluated. We hypothesized that CERK has a unique role in inducing Ca2+ signaling during Fc-mediated phagocytosis via its generation of C1P. Furthermore, we also hypothesize that CERK mediated changes in Ca2+ levels were not direct but required the participation of a store operated channels (SOC). In some systems, evidence suggests that SOC might be related to transient receptor potential channel (TRP) homologues. This hypothesis was tested using cell lines with different levels of expression of CERK and by using series of pharmacological inhibitors. To monitor subcellular localization of hCERK, red fluorescent protein (RFP)-tagged hCERK was created and confocal microscopy was performed. To study dynamic changes in Ca2+ during phagocytosis, imaging microscopy was used. Phagolysosome formation was evaluated by labeling the cells with lysotracker red DND-99 at 37oC and followed by imaging. When challenged with EIgG, hCERK transfected cells increased phagocytosis and simultaneously increased C1P. During subcellular fractionation of hCERK transfected cells, CERK translocates during activation from the cytosol to a lipid raft fraction. We also observed that TRP-1 accumulated at the site of CERK translocation in lipid rafts. Microfluorimetry of indo-1 revealed that Ca2+ oscillations in COS-1 cells, stably transfected with Fcγ RIIA/hCERK, were much higher compared to Fcγ RIIA transfected cells. The enhanced Ca2+ signals were accompanied by enhanced phagolysosome formation. hCERK transfected cells had a mean rate of fusion of 90%, compare to 76% ( catalytic inactive mutant of CERK-G198DhCERK), 65% (Fcγ RIIA), and 70% (Vector) transfected cells. The indo-1 intensity spikes demonstrated significantly higher Ca2+ intensities for phagosomes associated with the hCERK transfectants. The SOC inhibitor SKF96365 blocked enhanced Ca2+ signaling in the hCERK transfected cells. Similarly SOC inhibitors reduced the phagocytic index and phagolysosomal fusion in hCERK transfected cells. Our data showed that there is a co-localization of EIgG and CERK as well as caveolin-1 and CERK on the surface of the phagocytic cells. Site directed mutagenesis demonstrated that the kinase activity of hCERK was required for higher Ca2+ signals. Pharmacological inhibitors revealed that this Ca2+ signal operates through SOC. Our results show that introduction of the hCERK gene in cells alters the functional behavior of intracellular signals and supports the role of C1P in promoting membrane fusion by modulating Ca2+ signaling through activation of SOC

Sphingolipids are key signaling biomolecules that play a distinct role in cell proliferation, migration, invasion, drug resistance, metastasis, and apoptosis. Triple-negative (ER−PR−HER2−) and triple-positive (ER+PR+HER2+) breast cancer (called TNBC and TPBC, respectively) subtypes reveal distinct phenotypic characteristics and responses to therapy. Here, we present the sphingolipid profiles of BT-474 and MDA-MB-231 breast cancer cell lines representing the TPBC and TNBC subtypes. We correlated the level of different classes of sphingolipids and the expression of their corresponding metabolizing enzymes with the cell proliferation and cell migration properties of BT-474 and MDA-MB-231 cells. Our results showed that each cell type exhibits a unique sphingolipid profile, and common enzymes such as ceramide kinase (CERK, responsible for the synthesis of ceramide-1-phosphates) are deregulated in these cell types. We showed that siRNA/small molecule-mediated inhibition of CERK can alleviate cell proliferation in BT-474 and MDA-MB-231 cells, and cell migration in MDA-MB-231 cells. We further demonstrated that nanoparticle-mediated delivery of CERK siRNA and hydrogel-mediated sustained delivery of CERK inhibitor to the tumor site can inhibit tumor progression in BT-474 and MDA-MB-231 tumor models. In summary, distinct sphingolipid profiles of TPBC and TNBC representing cell lines provide potential therapeutic targets such as CERK, and nanoparticle/hydrogel mediated pharmacological manipulations of such targets can be explored for future cancer therapeutics.

Apart from cytokines and chemokines, sphingolipid mediators, particularly sphingosine-1-phosphate (S1P) and ceramide 1-phosphate (C1P), contribute to cancer and inflammation. Cancer, as well as other inflammatory conditions, are associated with skeletal muscle (SkM) atrophy, which is characterized by the unbalance between protein synthesis and degradation. Although the signaling pathways involved in SkM mass wasting are multiple, the regulatory role of simple sphingolipids is limited. Here, we report the impairment of ceramide kinase (CerK), the enzyme responsible for the phosphorylation of ceramide to C1P, associated with the accomplishment of atrophic phenotype in various experimental models of SkM atrophy: in vivo animal model bearing the C26 adenocarcinoma or Lewis lung carcinoma tumors, in human and murine SkM cells treated with the conditioned medium obtained from cancer cells or with the glucocorticoid dexamethasone. Notably, we demonstrate in all the three experimental approaches a drastic decrease of CerK expression. Gene silencing of CerK promotes the up-regulation of atrogin-1/MAFbx expression, which was also observed after cell treatment with C8-ceramide, a biologically active ceramide analogue. Conversely, C1P treatment significantly reduced the corticosteroid’s effects. Altogether, these findings provide evidence that CerK, acting as a molecular modulator, may be a new possible target for SkM mass regulation associated with cancer or corticosteroids.

Cancer cells rewire their metabolic programs to favor biological processes that promote cell survival, proliferation, and dissemination. Among this relevant reprogramming, sphingolipid metabolism provides metabolites that can favor or oppose these hallmarks of cancer. The sphingolipid ceramide 1-phosphate (C1P) and the enzyme responsible for its biosynthesis, ceramide kinase (CERK), are well established regulators of cell growth and survival in normal, as well as malignant cells through stress-regulated signaling pathways. This metabolite also promotes cell survival, which has been associated with the feedback regulation of other antitumoral sphingolipids or second messengers. C1P also regulates cancer cell invasion and migration of different types of cancer, including lung, breast, pancreas, prostate, or leukemia cells. More recently, CERK and C1P have been implicated in the control of inflammatory responses. The present review provides an updated view on the important role of CERK/C1P in the regulation of cancer cell growth, survival, and dissemination.

ET resistance is a critical problem for estrogen receptor-positive (ER+) breast cancer. In this study, we have investigated how alterations in sphingolipids promote cell survival in ET-resistant breast cancer. We have performed LC-MS-based targeted sphingolipidomics of tamoxifen-sensitive and -resistant MCF-7 breast cancer cell lines. Follow-up studies included treatments of cell lines and patient-derived xenograft organoids (PDxO) with small molecule inhibitors; cytometric analyses to measure cell death, proliferation, and apoptosis; siRNA-mediated knockdown; RT-qPCR and Western blot for gene and protein expression; targeted lipid analysis; and lipid addback experiments. We found that tamoxifen-resistant cells have lower levels of ceramides and hexosylceramides compared to their tamoxifen-sensitive counterpart. Upon perturbing the sphingolipid pathway with small molecule inhibitors of key enzymes, we identified that CERK is essential for tamoxifen-resistant breast cancer cell survival, as well as a fulvestrant-resistant PDxO. CERK inhibition induces ceramide-mediated cell death in tamoxifen-resistant cells. Ceramide-1-phosphate (C1P) partially reverses CERK inhibition-induced cell death in tamoxifen-resistant cells, likely through lowering endogenous ceramide levels. Our findings suggest that ET-resistant breast cancer cells maintain lower ceramide levels as an essential pro-survival mechanism. Consequently, ET-resistant breast cancer models have a unique dependence on CERK as its activity can inhibit de novo ceramide production.

Ceramide kinase (CERK) has been implicated in important cellular processes such as inflammation and apoptosis. Its activity is usually measured using radiolabeled ceramide or [γ-32P]-ATP, followed by extraction, thin-layer chromatography, and detection of the formed labeled ceramide-1-phosphate. To eliminate the use of radioactivity, we developed similarly but independently from the approach by Don and Rosen (2008), a fluorescence-based ceramide kinase assay, using N-[7-(4-nitrobenz-2-oxa-1,3-diazole)]-6-aminohexanoyl-sphingenine (NBD-C6-ceramide) as substrate. ItsKmvalue (4 μM) was comparable to that of N-hexanoyl-sphingenine (C6-ceramide). The produced fluorescent NBD-C6-ceramide-1-phosphate was captured by means of solid-phase extraction on an aminopropyl phase, resulting in a fast and sensitive CERK measurement. By performing this assay in a 96-well format, it is also suitable for high-throughput screening (HTS) to search for CERK modulators. A limited screen revealed that some protein kinase inhibitors (e.g., U-0126;IC504 μM) and ceramide analogues (e.g., fenretinide, AMG-9810;IC501.1 μM) affect CERKin vitro.

La investigación analiza las propiedades psicométricas del Cuestionario de Experiencias Relacionadas al Internet (CERI) y del Cuestionario de Experiencias Relacionadas al Móvil (CERM) en 508 estudiantes de una universidad privada de Lima. Se aplicó la Escala de Adicción a Internet de Lima (EAIL), la Escala de Zung para la depresión (SDS), el Inventario de Ansiedad Rasgo-Estado (IDARE) y la Escala de Alexitimia de Toronto (TAS-20) para la validez convergente y discriminante. Para validez de criterio se hicieron comparaciones con las horas de uso de internet y celulares. Para el CERI, se obtuvo una estructura bifactorial con un 33.47% de varianza explicada. La confiabilidad mostró una consistencia interna adecuada para la prueba, siendo de .73, y cuestionable para las áreas siendo de .69 y .62. En el CERM, se obtuvo una estructura unifactorial con un 37.76% de varianza explicada. La confiabilidad mostró una consistencia interna adecuada, siendo de .81. Las relaciones del CERI y CERM con el EAIL resultaron significativas, directas y grandes; con el SDS, IDARE y TAS-20 resultaron significativas, directas y medianas. La comparación del CERI y sus dos áreas con las horas de conexión a internet fue significativa y pequeña para el CERI y su primera área. La comparación del CERM con las horas de conexión al celular fue significativa y pequeña. Con ello, el CERI no evidencia contar con propiedades psicométricas adecuadas para medir el uso problemático de internet, pero se confirma que el CERM cuenta con propiedades psicométricas adecuadas para evaluar el uso problemático del celular.
The study focuses on the psychometric properties of the Internet Experiences Questionnaire (CERI) and the Cell pone Experiences Questionnaire (CERM) in 508 students of a private college in Lima. The instruments to determine convergent and discriminant validity of the scales were the Internet addiction scale of Lima (EAIL), The Zung Self-Rating Depression Scale (SDS), The State-Trait Anxiety Inventory (STAI), and the Toronto Alexithymia Scale (TAS-20). For evidence of criterion validity, comparisons were made with the daily hours of Internet and cell phone use. Results from the factorial analysis of CERI reveal a two dimensional structure with a 33.47% of explained variation. In addition, the reliability analysis shows a suitable internal consistency of .73 for the total score of the scale, and questionable for the areas with values of .69 and .62. The results from the factorial analysis of CERM reveal one dimensional structure with a 37.76% of explained variation. The reliability analysis shows a suitable internal consistency of .81. Significant, direct and large correlations of CERI and CERM with EAIL and significant, direct and medium correlations of CERI and CERM with SDS, STAI and TAS-20. The comparison of CERI, its two areas and CERM with the hours of Internet connection and cell phone use was significant and small for CERI, its first área and CERM. With this, CERI can’t show that has adequate psychometric properties the measure the Internet problematic use, but it is confirmed that CERM is an instrument with adequate psychometric properties to evaluate cell phone’s problematic use.

The goal of this Master’s thesis is to modernize the CERN library public terminal system. With modernization the aim was set to minimize the administrative burden on library staff from the new system. Modernization focuses on replacing the currently used Linux based thin client system that suffers from hardware ageing with a system that resides in the CERN-IT operated private cloud. Another aspect in modernization was to utilize the provided network infrastructure more to simplify system complexity and the amount of remote components that require library staff maintenance and administration. The new system differentiates from a traditional LTSP-system by dividing every thin client into a different virtual machine. By distributing every thin client in our own virtual host, we can distribute resource allocation, like CPU-time and physical memory, between different physical hosts. The system also contains monitoring features that informs administrative problems that have been noted. Monitoring decreases the time required for dealing with system maintenance. Linux-based thin client remote connection efficiencies are compared using UI-latency and bandwidth efficiency as metrics. A plain X11 connection is measured against an NX-connection and an SSH-tunneled X11 connection. Results show NX bandwidth efficiency that comes from utilizing an extra caching layer. Pääsuunta modernisaatiossa on rakentaa uusi asiakaspäätejärjestelmä vanhan LTSP järjestelmän tilalle, joka kärsii palvelinraudan vanhenemisesta, hyödyntäen CERN-IT:n operoimaa sisäistä pilvipalvelua sekä jo olemassa olevaa verkkoinfrastruktuuria. Rakennettu järjestelmä poikkeaa perinteisestä LTSP-järjestelmästä määrittelemällä jokaiselle päätteelle erillisen virtuaalikoneen johon kytkeytyä. Erillisillä virtuaalikoneilla saadaan käyttäjien varaamia resursseja kuten CPU-aikaa ja fyysistä muistia hajautettua useammalle fyysiselle palvelimelle. Järjestelmä sisältää myös proaktiivisen monitoroinnin joka ilmoittaa havaituista ongelmista ylläpitovastuussa olevia henkilöitä eikä näin vaadi järjestelmän aktiivista tarkkailua ylläpitovastuussa olevilta henkilöiltä. Linux-pohjaisten tyhmien päätteiden etäyhteys-protokollien tehokkuutta vertaillaan käyttämällä käyttöliittymän latenssia ja kaistankulutusta mittareina. Normaalia X11 yhteyttä verrataan yleisesti käytettyyn SSH-tunneloituun X11 yhteyteen sekä NX-yhteyteen. Tulokset osoittavat NX-yhteyden ylimääräisen välimuistin tehostavan kaistankäyttöä. Cílem této magisterské práce je navrhnout a implementovat IDE (integrované vývojové prostředí), které zvýší efektivitu a bezpečnost vývoje pro SIMATIC WinCC Open Architecture. Tato práce je založena na výzkumu provedeném týmem z Technické univerzity v Eindhovenu a splňuje požadavky pocházející ze SCD sekce v CERN (Evropské organizace pro jaderný výzkum). Vyvinuté IDE je postaveno na platformě Eclipse, přičemž pro syntaktickou analýzu, linkování a sémantickou analýzu kódu používá Xtext framework. IDE nabízí také podporu pro nově vytvořený programovací jazyk, který umožňuje programátorům jednoduše nadefinovat šablonu pro konfigurační soubory používané WinCC OA. Interpret tohoto nového jazyka je schopen provést syntaktickou analýzu šablony a konfiguračního souboru a rozhodnout, zdali konfigurační soubor odpovídá šabloně. Praktickým výstupem této práce je integrované vývojové prostředí, které podporuje vývoj WinCC OA aplikací v CERN a periodicky provádí analýzu kódu těchto aplikací napsaného v jazyce Control script.

Im Rahmen eines grossangelegten Modernisierungsprogramms des CERN Beschleunigerkomplexes zur Steigerung der Intensität der Protonenstrahlen wird in einer ersten Phase der Protonenbeschleuniger Linac2 durch einen H− Ionen Beschleuniger Linac4 ersetzt. Um den Ionenstrahl an die einzelnen Beschleunigerelemente des Linac4 anzupassen sind drei Strahltransportsektionen notwendig. Diese sind aufgrund ihrer strahldynamischen Eigenschaften sensitive Stellen für ungewolltes Emittanzwachstum und Teilchenverluste und sind im Schwerpunkt dieser Thesis. Die erste Strahltransportsektion, „Low Energy Beam Transport”(LEBT), befindet sich zwischen der Teilchenquelle und dem ersten Beschleunigerelement, den Radiofrequency Quadrupole (RFQ). Im Rahmen dieser Thesis wurde das LEBT in Betrieb genommen, dessen Strahldynamik rekonstruiert und die Leistungslimitierungen experimentell bestimmt. Der zweite Teil der Thesis ist auf die Auslegung und Strahldynamik der Transfer Linie zwischen Linac4 und dem Proton Synchrotron Booster(PS-Booster) konzentriert. Die Strahldynamik der Transferlinie wurde optimiert und teilweise komplett überarbeitet. Die neue Transferlinie zeichnet sich durch eine verbesserte Emittanzerhaltung, höhere Stabilität der Strahldynamik gegenüber Aufstellungsfehlern und Feldjittern der Magnete und durch eine optimale Anpassung der Strahlparameter an die verschiedenen Injektionsschemata des PS-Boosters aus. Für eine abschließenden „Start-To-End” Simulationen wurden die Strahlcharakteristiken, die bei der Inbetriebnahme des LEBTs bestimmt worden sind, als Anfangsbedingungen für die strahldynamische Simulation von Linac4 einschließlich Transferlinie genutzt. Mit Hilfe dieser „Start-To-End” Simulationen werden kritische Positionen der Strahldynamik identifiziert, die durch Teilchenverluste oder Emittanzwachstum gekennzeichnet sind. Eine besonders kritische Sektion stellt das „Medium Energy Beam Transport” da, dessen Optik auf die neuen Strahlparameter angepasst werden musste.
In the first phase of the upgrade program of the CERN accelerator complex the proton injector Linac2 will be replaced by a new, normal-conducting H− ion Linac, Linac4, allowing a significant increase of the proton flux intensity along the downstream accelerator complex. In the design of Linac4 three beam transport sections are implemented to match the beam between the different accelerator elements and to model the longitudinal pulse structure. These three beam transport sections, which are the most critical locations in terms of beam quality preservation, are in the focus of this thesis. During the work of this thesis the Low Energy Beam Transport (LEBT), which is required to match the source beam to the radiofrequency quadrupole (RFQ), has been commissioned and its beam dynamics re-constructed. The measurement campaign used to reconstruct the LEBT beam dynamics was performed with the aim to prepare the RFQ commissioning and to maximise the LEBT performance. Downstream of the Linac4 accelerator the beam is transported along a 180m long transfer line to the Proton Synchrotron Booster (PS-Booster). The transfer line optics was studied, optimised and sections were completely re-designed. The new transfer line optics is characterised by an improved preservation of the beam emittance, higher stability of the optical solution with respect to alignment errors and field jitters of the transfer line magnets and it is matched to each of the PS-Booster injection schemes. In a concluding ”Start-To-End” simulation based on the measured beam characteristics at the LEBT exit the beam dynamics of the downstream Linac, including the transfer line, was calculated. To minimise particle losses within acceptable emittance preservation the beam optics of the Medium Energy Beam Transport (MEBT) was adapted to the measured beam parameters. This ”Start-To-End” simulation was performed to identify critical sections of the Linac4 beam dynamics and to adjust the commissioning strategies.

As an immediate response to the economic impact of COVID, in 2020 regional UK governments promoted short-term funded university courses to enable individuals that were redundant or furloughed to reskill in the area of software development. One of the courses developed was a novel Post Graduate Certificate in Software Development (Cert), based on a well-established MSc in Software Development (conversion) course (MSc SD) delivered at a local University. The course attracted over 700 applications, enabling the application of higher entry criteria than the MSc SD. Differences in the entry criteria and delivery format (entirely online for Cert) presented a unique Action Based research opportunity. This paper initially reports on course performance of the Cert in comparison to the MSc SD. It then provides an analysis of impact and differences within the enhanced entry criteria for the Cert. Providing discussion and recommendations for entry criteria and delivery mechanisms for similar courses.

The purpose of this study is to establish a detailed three-dimensional (3D) model of containment safety verification via integral test (CERT) using the containment code GOTHIC 8.0. This paper presents the model construction and a typical CERT case for the model evaluation. In the typical CERT case, steam with high mass and energy is released to the test vessel to simulate the passive containment response during main steamline break (MSLB) accident. Heat removal process is accomplished primarily by absorption of energy by the gas volume and structures inside vessel, by condensation of steam on the inside shell surface, by heat conduction through the steel shell, and by evaporation of water film covered on the outer vessel shell surface. The main results of the typical CERT case are qualitatively compared with the results obtained from simulations with GOTHIC 8.0 code. From comparison, a verification of the code in terms of pressurization, temperature response, steam condensation and water film evaporation are carried out. The code analysis results are of significance on the research of thermal hydraulic phenomena which occur in the passive containment cooling system (PCS) during accidental sequences.

A utilização do Compensador Estático de Reativos (CER) no sistema elétrico de potência (SEP) permite uma maior flexibilidade no controle das variáveis do sistema e, como consequência, pode-se postergar os investimentos de expansão do SEP. A inserção do CER na linha de transmissão pode alterar os parâmetros do fluxo de potência e, consequentemente, os módulos e ângulos das tensões e correntes no sistema em que estão conectados. Portanto, neste trabalho analisa-se a influência do CER na alteração dos tempos de atuação e nos alcances das zonas de proteção das linhas de transmissão que compõem o SEP. O software PSCAD/ EMTDC é utilizado para realizar toda a modelagem de um SEP. Após a modelagem do SEP, perturbações são causadas no sistema e avalia-se a função de proteção de impedância (ANSI 21) frente a essas anormalidades no cenário com e sem a presença do CER. Os resultados indicam que houve um subalcance da função ANSI 21 frente a distúrbios na perspectiva da presença do CER.

A laser interferometer was used to monitor thermally induced length changes in samples of Cer-Vit 101 and 142 between discrete temperatures in the 250–450 K range. The Cer-Vit 101 was then irradiated with a γ-ray 132Cs source for 37 days, and the Cer-Vit 142 with a 60Co source for 58 days. Both samples received a radiation dose of 105 rad which caused them to become opaque.

The increasing cost of offshore developments involving floating structures has resulted in the need to ensure accurate cost estimates are developed very early in the project lifecycle. A number of conceptual costs estimating approaches exist in the industry however, there are concerns about their accuracy and applicability in the early stages of the project. Cost Estimating Relationships (CERs) that can be applied at the concept development phase of floating structures have therefore been developed. They are derived from the reported cost and weight of existing Floating Production, Storage and Off take (FPSOs) vessels and have been applied to ship based offshore development concepts. These CERs are risk based, to produce varying results on a cost (weight) and schedule basis, such that overestimates/ underestimates are offset by risk ranking, to yield a relatively small error in the cost estimates. The uncertainties identified in the risk ranking of components of the development concepts (and resulting cost estimates) are accounted for by applying fuzzy logic to the CERs. This paper presents some of the results of ongoing research that is expected to contribute significantly to the development of floating offshore structures while improving confidence levels in conceptual cost estimates, thus facilitating a comprehensive front-end evaluation of offshore developments.

Em um Ecossistema de Software Móvel (MSECO), as organizações de software passaram a reestruturar seus processos para desenvolvedores visando atingir metas, como o aumento do número de aplicações móveis (apps). No entanto, a “barreira” de qualidade provida pela Loja de Apps, não define critérios que garantem que as apps certificadas a partir deles atinjam as metas. É necessário considerar a qualidade de suporte oferecido aos desenvolvedores por meio dos processos (certificação), para atingir o desempenho esperado para as apps. Neste trabalho, foi definida uma abordagem baseada em processos para certificação de apps (MSECO-CERT). O uso da MSECO-CERT para desenvolvimento de apps gerou um coeficiente de crescimento de downloads 363% maior e média de avaliações dos usuários 28% maior em comparação a uma abordagem ad hoc.
