Background: We have previously reported that patients with Crohn’s disease (CD) have a very specific erythrocyte membrane phospholipid fatty acid profile. The findings of this study suggest that the activities of enzymes involved in the metabolism of linoleic acid (LA), that is, delta-6 desaturase, are higher in CD patients than in healthy individuals. Methods: We evaluated the utilities of various fatty acid compositions of the plasma (p-) as new serological markers for CD compared to those of erythrocyte membranes (e-). Results: Fifty CD patients and 50 healthy individuals were enrolled. In both plasma and erythrocyte membranes, the weight percentages of palmitic acid (PA) were significantly higher, while those of LA were significantly lower in CD patients than in controls. Fatty acids with high sensitivity and specificity were p-PA (0.86 and 0.74) and e-PA (0.80 and 0.74). With PA and LA as a CD fatty acid index (CDFAi), that is, CDFAi = (PA/LA), the sensitivity and specificity of plasma CDFAi (p-CDFAi) and e-CDFAi were 0.80 and 0.80; and 0.82 and 0.88 respectively. Conclusion: In CD patients, various fatty acids were specifically altered in both plasma and erythrocytes, and p-PA and p-CDFAi are potentially useful as new serological markers for CD.

Abstract In this work, we investigated the functions of structural modules within alginate lyase by truncating an endo-type alginate lyase into two successive catalytic modules. The effects of module deletion on biochemical characteristics and product distributions were further investigated. The N-terminal module (Aly7B-CDI) exhibited no activity toward alginate, polyM or polyG, but the C-terminal module (Aly7B-CDII) retained its activity. The full-length enzyme (Aly7B) and its truncated counterpart (Aly7B-CDII) had similar substrate specificities, but Aly7B-CDII had lower activity. Moreover, the activity of Aly7B was much higher than Aly7B-CDII at 30°C. Aly7B-CDII, however, possessed higher optimal pH and better pH stability than the full-length enzyme. The final degradation products for Aly7B were unsaturated di-, tri- and tetra-oligosaccharides, and those for Aly7B-CDII were unsaturated mono-, di-, tri-, tetra- and penta-oligosaccharides. Therefore, the potential impact of the noncatalytic module Aly7B-CDI on the catalytic module Aly7B-CDII was further elucidated by characterizing Aly7B and its truncations. These data contribute to the functional understanding of these differing modules.

We present a measurement of the dipion mass spectrum in the decay X(3872) [right arrow] J/ [psi] [pi]⁺ [pi]⁻ using a 360 pb-1 sample of pp collisions at av [square root]s = 1.96 TeV collected with the CDF II detector at the Fermnilab Tevatron Collider. As a benchmark, we also extract the dipion mass distribution for [psi] (2s) [right arrow] J/ [psi] [pi]⁺ [pi]⁻ decay. The X(3872) dipion mass spectrum is compared to QCD multipole expansion predictions for various charmonium states, as well as to the hypothesis [right arrow] J/ [psi] po. We find that the measured spectrum is compatible with 3S1 charmonium decaying to J/ [psi] [pi]⁺ [pi]⁻ - and with the X(3872) - J/po hypothesis. There is, however, no 3S1 charmonium state available for assignment to the X(3872). The multipole expansion calculations for 1P1 and 3DJ states are in clear disagreement with the X(3872) data. For the [psi] (2S) the data agrees well with previously published results and to multipole expansion calculations for 3S1 charmonium. Other, non-charmonium, models for the X(3872) are described too. We conclude that since the dipion mass spectrum for X(3872) is compatible with J/pO hypothesis, the X(3872) should be C-positive. This conclusion is supported by recent results from Belle Collaboration which observed X(3872) -+ J/7y decay. We argue that if X(3872) is a charmonium, then it should be either ... state, decaying into J/ [psi] [pi]⁺ [pi]⁻ in violation of isospin conservation. A non-charmonium assignment, such as DD* molecule, is also quite possible.

We present a study of the charmless Bs to phiphi decay performed with the CDFII detector at the Fermilab Tevatron Collider. Charmless Bs meson decays currently can be studied only at the Tevatron and represent a field still to be fully explored that offers additional ways to test our present theoretical understanding. The Bs to phiphi belongs to a particular class of these decays: the Bs meson decays into a pair of vector particle and the final state is self-conjugate. It can be used to measure the Bs decay width difference (Delta Gamma_s), CKM studies, and tests of decay polarization predictions.

The Appalachian Regional Commission is currently working with a major foundation on the development of a new regional Community Development Financial Institution (CDFI), Appalachian Community Capital. By connecting Appalachia's small businesses to large external investors, will this CDFI bring more community development capital into the region, and help alleviate poverty? Or, as the neoliberal era deepens into the "Age of Austerity", is this but the latest use of market logic to attempt to solve public, political problems, fraught with the shortcomings of such an approach? I argue that the new CDFI may bring capital into the region. But because it does so using market logic, it cannot ensure that the money will go to the neediest areas, or that it will be invested in a manner which actually creates jobs for existing residents, in locally owned businesses (thereby keeping profit in the region), or in sustainable industries. It also cannot address the problems posed by a dysfunctional civic culture, in part the legacy of big coal's historic corporate paternalism and subsequent disinvestment, as corrupt local elites "other" the mostly white mountain poor as an intractable, permanent underclass. Further, even if the new entity could surmount these issues, I argue that it does not address the underlying challenge: the ongoing outflow of capital out of the region. Due to both regulatory barriers and industry economies of scale, institutional and individual investors ship most of Appalachia's capital out to major national financial centers, where it is disbursed around the world. These levels of exported capital stock dwarf the small volumes of community development capital that any CDFI might hope to reinvest locally. For the region's poverty level to decline, this challenge might be addressed through the removal of regulatory barriers and creation of local institutions and investment platforms to invest both community development capital specifically, and other forms of capital, as well. These institutions and platforms may not be most appropriately constructed at the geographic scale of the Appalachian region, given the economic diversity in the region, and given the value-laden history of the social construction of the term Appalachia itself.

This thesis explores Community Development Finance Institutions (CDFI) as an alternative vehicle for the supply of debt finance to financially excluded enterprises. CDFIs are part of a broader approach to addressing financial exclusion that is experienced by commercial and social enterprises in the US and UK. The thesis explores US and UK CDFI lending processes to develop an understanding of how financial and social objectives are balanced in the lending process and the ways in which CDFIs become embedded in local financial and business support networks. The analysis is based upon detailed comparative research of CDFIs located in the US and the UK; interviews were undertaken with CDFIs, their clients and a quantitative analysis of a CDFIs loan portfolio was undertaken. The research concludes that CDFIs are complex dynamic organizations as they have to balance a double or triple bottom line which has the potential to undermine the firm’s long term survival or mission. The danger is that over time a CDFI will reduce its exposure to risk and become more like a mainstream bank. The tensions with the CDFI business model implies that they will only ever provide a partial solution to the enterprise finance gap.

Congenital Dyserythropoietic Anemias (CDAs) are subtypes of bone marrow failure syndromes, hallmarked by ineffective erythropoiesis. The most common form is CDA type II (CDAII), showing moderate/severe anemia, relative reticulocytopenia, jaundice, splenomegaly, and iron overload. It is inherited as an autosomal recessive disorder due to loss-of-function mutations in the SEC23B gene. Molecular pathogenesis of CDA II still has to be investigated because the described animal models did not recapitulate the clinical features observed in humans. To date, treatments for CDAII patients consist of supportive therapy, such as erythrocyte transfusions, or bone marrow transplantation or splenectomy in transfusion-dependent cases. Recently, members of TGF-β superfamily have been studied as potential regulators of erythropoiesis, especially the growth differentiation factor 11 (GDF11). Through the binding of specific receptors, GDF11 leads to an inhibited late-stage erythropoiesis. Indeed, two GDF11 inhibitors, ACE-011 and ACE-536, have been associated with an improvement of hematologic parameters. Studies with the mouse counterpart of ACE-011, RAP-011, on a mouse model of β-thalassemia showed increased differentiation of erythroid cells, improvement of the anemic condition and reduced iron overload in treated mice. The first aim of our study was the establishment of a cellular model of CDA II, that could reproduce the main defects of the disease, such as the lack of the erythroid differentiation due to the low or absent expression of SEC23B gene. For this aim, we selected the K562 cell line and, through short-hairpin RNA-based strategy, we obtained two different clones of K562 showing a stable silencing of SEC23B. Then, we decided to assess the effects of RAP-011 on this CDA II model, by investigating the pathway involved in the GDF11 signaling. This treatment simulated the ligand trap function played by RAP-011 towards GDF11. The administration of RAP-011 resulted in a reduction of SMAD2 phosphorylation induced by GDF11 and, moreover, in an increase of different erythroid differentiation markers.

I och med framväxten av samhällsentreprenörer har ett nytt begrepp introducerats påden svenska marknaden. Det handlar om den nya generationen företagare som sermöjligheten i att tjäna pengar samtidigt som de driver verksamheter som skapar ettmervärde för samhället. Samhällsentreprenörerna i Sverige står dock framför ett problem,att på ett hållbart sätt finansiera sin verksamhet.Denna studie syftar till att kartlägga hur finansieringen av svenska samhällsentreprenörerser ut. Undersökningen bygger på en kvalitativ studie där vi har undersökt tresvenska verksamheter, som faller under definitionen ”samhällsentreprenör”. Ämnet ärfortfarande relativt nytt och okänt och det har därför inte bedrivits mycket forskningkring detta. Vi har därför, som underlag för vår undersökning, använt oss av en studiesom gjordes 2003 i Storbritannien.Då vi endast undersökt tre verksamheter har vi inte kunnat dra några generella slutsatserför hela den svenska marknaden. Dock indikerar resultatet av vår undersökning attden svenska marknaden, för att finansiera denna typ av verksamhet, är bristfällig och ibehov av utveckling. För såväl förutsättningarna som efterfrågan på samhällsentreprenörerfinns redan idag och denna grupp företagare ser ut att öka inom den närmasteframtiden.

Le travail présenté dans cette thèse est une contribution au dimensionnement et à la commande d’un générateur de type Cascade de Machines Asynchrones à Double Alimentation. Le modèle de la machine pour les simulations est basé sur une représentation modulaire qui découle du modèle de Park de deux machines asynchrones doublement alimentées distinctes, en respectant deux systèmes de référence. Le modèle dynamique est ensuite étendu pour décrire la machine dans un repère unifié. Cette représentation est cruciale pour le dimensionnement de la machine et la synthèse de la commande. Le domaine de fonctionnement de la cascade en régime permanent est exploré. Une méthode analytique générique est proposée pour établir les plages de puissances active-réactive. Les courbes limites sont définies par rapport aux grandeurs nominales de la machine. L’étude prend en compte l’effet de saturation du circuit magnétique. Il est prouvé que la capacité en puissance de la machine est déterminée par les valeurs maximales des courants statoriques et peut être soumise à plusieurs limitations. L’approche analytique est testée et validée par des mesures expérimentales. Deux cas sont considérés pour la commande de la cascade : un générateur raccordé au réseau et un générateur autonome. Pour la connexion au réseau l’application visée est les éoliennes de grande puissance. Une nouvelle méthode pour l’extraction de la puissance maximale d’une éolienne à vitesse variable est proposée. Le système est contrôlé dans le but de fournir une puissance maximale quasi-constante indépendamment des fluctuations du vent. En plus de l’optimisation de la puissance, le facteur de puissance est également ajusté selon les normes de raccordement standard imposées par les services système, et les limites de fonctionnement de l’unité intégrée. Une commande vectorielle sans capteur de tension, basée sur une orientation suivant un flux virtuel, est appliquée pour la régulation découplée des puissances active et réactive. Pour le fonctionnement en mode isolé, l’application visée est la génération électrique pour système avionique embarqué. Deux réseaux de distribution sont traités : réseau AC à fréquence fixe et réseau DC. Dans les deux cas, la commande est élaborée dans le but de maintenir une tension de sortie constante. Le fonctionnement de la cascade alimentant une charge triphasée déséquilibrée est également étudié. Des schémas de contrôle servant à la compensation du déséquilibre des tensions statoriques sont développés. Deux méthodes de compensation sont proposées : l’une basée sur le principe de deux repères tournants et l’autre sur un correcteur répétitif. Les approches présentées sont validées par simulation avec Matlab/Simulink et par expérimentation
This thesis is a contribution to the power sizing and the control of a Cascaded Doubly Fed Induction Generator (CDFIG). The machine model for simulation is based on a modular representation, derived from the Park model of two distinct Doubly Fed Induction Machines expressed in their own reference frames. The dynamic model of the cascaded machine is then extended to be described in a unified reference frame. This representation is convenient for the sizing of the machine and for the design of the controller. The steady state power operating margins of the cascaded machine are investigated. A generic analytic method is suggested to derive the active-reactive power domain. The limit curves are defined in terms of the rated quantities of the machine. The study takes into account the magnetic circuit saturation effect. It is proven that the power capability of the machine is determined by the stator current maximum values and is subject to several limitations. The analytical approach is tested and validated by experimental measurements. The CDFIG is controlled in grid-connected and standalone operation modes. In grid-connected application, the attention is paid to high power wind generation systems. A new maximum power tracking of a variable speed wind turbine is suggested. The generating plant is carried to provide a quasi-constant maximum power regardless wind fluctuations. In addition to active power optimization the power factor is adjusted according to the grid code requirements and the operating domain of the integrated unit. A virtual flux oriented vector control is applied for the decoupled regulation of active and reactive powers, leading to grid voltage sensorless operation. In standalone operating mode, the study treats in particular the embedded aircraft power system generation. Two distribution networks are considered: constant frequency AC network and DC network. In both cases, the control aims to maintain a constant output voltage. The operation of the CDFIG supplying unbalanced three-phase load is further explored. Control schemes to cope with unbalanced stator voltage conditions are developed. Two compensation methods are elaborated: the first dealing with dual rotating frames and the second is based on repetitive controller. Simulations with Matlab/Simulink software and experiments validate the control approaches

L’omeostasi del ferro svolge un ruolo cruciale nella normale eritropoiesi e in anemie da sovraccarico di ferro, come ad esempio la β-talassemia intermedia e le anemie congenite diseritropoietiche (CDAs), che rappresentano interessanti modelli umani per studiare l'eritropoiesi patologica. Le CDAs sono malattie genetiche che provocano la distruzione midollare delle cellule eritroidi, ossia l’eritropoiesi inefficace. Ciò causa un’iperplasia eritroide nel midollo e l’anemia con una risposta sub-ottimale dei reticolociti. Quattro le forme di CDAs ben note, il tipo II è la forma più frequente, il cui gene causativo, SEC23B, è coinvolto nel traffico vescicolare tra reticolo endoplasmatico e il Golgi. La cirrosi, il diabete e i calcoli biliari sono le complicazioni più comuni causate dall’anomalo accumulo di ferro. I meccanismi responsabili di tali complicazioni, dovute principalmente al sovraccarico di ferro, restano da chiarire. Lo scopo di questo studio sta proprio nel chiarire il ruolo del gene SEC23B nel sovraccarico di ferro attraverso studi in vivo e in vitro. Negli studi in vivo abbiamo valutato i livelli dei marcatori del sovraccarico di ferro, quali il fattore di crescita differenziale 15 (GDF15), il recettore solubile della transferrina (sTfR), l’emojuvelina solubile (sHJV), la ferritina e l’epcidina nel siero o plasma dei pazienti CDAII. I livelli sierici o plasmatici di queste molecole sono stati correlati con i parametri biochimici del ferro ed i parametri ematologici per identificare i marcatori prognostici in grado di predire l'insorgenza del sovraccarico di ferro. Quindi, questi dosaggi, se fatti durante l'infanzia o l'adolescenza, possono essere analizzati come marker di gravità della malattia per lo sviluppo di strategie di trattamento preventive per i bambini CDAII o altri affetti da eritropoiesi inefficace. Con gli studi in vitro, invece, abbiamo effettuato una caratterizzazione del sovraccarico di ferro nella linea cellulare di carcinoma epatocellulare umano (Huh7) e nella linea cellulare umana di leucemia eritromieloblastoide (K562) silenziate per il gene SEC23B per analizzare l'espressione genica dei geni del metabolismo del ferro. Inoltre abbiamo caratterizzato il nuovo regolatore eritroide, Eritroferrone (Erfe), sia in vivo nei pazienti CDAII, che in vitro nelle linee cellulari silenziate per l'espressione genica di SEC23B. I livelli sTfR indicano il numero di precursori dei globuli rossi e la richiesta di ferro da parte delle cellule. Nel nostro studio, sTfR è aumentato nei pazienti e correla negativamente con l’epcidina e la ferritina sierica, nonostante il suo ruolo di inibitore della sintesi di epcidina. Quindi, sTfR non influenza la sintesi di epcidina nella CDAII e per questo non può esserne un importante regolatore negativo nella patologia in esame. Anche sHJV è un inibitore dell’epcidina, quindi per la prima volta i suoi livelli sono stati analizzati nella CDAII. Noi abbiamo osservato che i pazienti rispetto ai controlli sani hanno livelli di sHJV più alti e che questa molecola correla con la gravità del fenotipo ematologico e con il sovraccarico di ferro nella CDAII. L'eritropoietina media l'eritropoiesi e aiuta la maturazione e lo sviluppo degli eritroblasti nelle fasi di sviluppo successive. I livelli di EPO nei pazienti CDAII sono più alti rispetto ai controlli sani. Questi dati confermano che l'anemia nella CDAII potrebbe essere dovuta a un deficit della funzione ematopoietica del midollo osseo, poiché il livello di EPO è aumentato nel paziente anemico che ha un numero insufficiente di nuovi globuli rossi. I nostri dati dimostrano, inoltre, che il GDF15 è coinvolto nell’accumulo di ferro nei pazienti CDAII. Le sue concentrazioni correlano con la gravità dell'anemia e la disponibilità di ferro sierico nei pazienti, rispecchiando la gravità dell’eritropoiesi inefficace. Il GDF15 modula l'espressione dell’epcidina in risposta al sovraccarico di ferro e aumenta l'assorbimento di ferro in condizioni di stress eritropoietico. Quindi, per predire la gravità della malattia nei pazienti CDAII è possibile combinare le informazioni sulle concentrazioni del GDF15 con gli altri marcatori del sovraccarico di ferro.
Iron homeostasis plays a crucial role in normal erythropoiesis as well as in iron loading anemias, such as β-thalassemia intermedia and CDAs (congenital dyserythropoietic anemias), which represent interesting human models to study abnormal erythropoiesis. CDAs are genetic disorders which result in intramedullary destruction of developing erythroid cells (ineffective erythropoiesis). The disorder of erythropoiesis results in the paradoxical association of a bone marrow showing marked erythroid hyperplasia and anemia with a suboptimal reticulocyte response. Three forms of CDAs are well known: the type II is the most frequent, whose causative gene, SEC23B, is involved in ER-Golgi network. Cirrhosis and hemochromatosis, diabetes and gallstones are common complications due to abnormal iron accumulation. The mechanisms responsible for such complications mainly due to iron overload remain to be elucidated. In this work we investigated the role of SEC23B gene in iron overload through studies in vivo e in vitro. In in vivo studies we evaluated growth differentiation factor 15 (GDF15), soluble transferrin receptor (sTfR), soluble hemojuvenil (sHJV), ferritin and hepcidin levels in serum or plasma samples of CDAII patients as iron overload markers. Serum or plasma levels of these molecules were correlated with iron biochemical and hematological parameters to identify prognostic markers to predict the onset of iron overload in CDAII. So these measurements in infancy or childhood may be explored as a disease severity marker for development of preventative treatment strategies for CDAII children or others with ineffective erythropoiesis. In in vitro studies we performed a characterization of CDAII iron overload in human hepatocarcinoma (Huh7) cell line and in human erythromyeloblastoid leukemia (K562) cell line silenced for SEC23B to analyse gene expression of iron metabolism genes. We also characterized the new erythroid regulator erythroferrone (Erfe) both in vivo in CDAII patients and also in vitro in cell lines silenced for SEC23B gene expression. The levels sTfR are indicative of the number of red blood cell precursors and their demand for iron. In our study, sTfR was increased in CDAII patients and negatively correlated with hepcidin and serum ferritin, despite the predicted role of sTfR as an inhibitor of hepcidin. Together with previous findings, sTfR failed to affect hepcidin concentrations, suggesting that sTfR may not be a major suppressor of hepcidin in CDAII patients. In order to investigate whether sHJV down-regulates hepcidin contributing to iron loading pathology, for the first time its levels was analysed in CDAII. We reported high levels of sHJV in CDAII patients compared to healthy controls and its correlation with the severity of hematological phenotype and also iron overload in CDAII. Erythropoietin (EPO) is the main signalling molecules which mediates erythropoiesis and helps the maturation and development of erythroblasts in the later developmental stages. EPO levels in CDAII patients are increased. This data confirms that anemia in CDAII could be due to a deficiency of hematopoietic function of bone marrow, because the level of EPO is increased in anemic patient who has an insufficient number of new red blood cells. Our results reveals that EPO is involved in iron overload decreasing hepcidin expression. Also GDF15 is involved in the iron accumulation in CDAII patients. Its concentrations correlate with the severity of anaemia and serum iron availability in patients, so it reflects the magnitude of the ineffective erythropoiesis. Our data support a role for GDF15 in blunting hepcidin expression in response to iron overload and increasing iron absorption under erythropoietic stress conditions. So, it may be possible to combine information about GDF15 concentrations with other indicators to predict disease severity in CDAII patients.

Community development financial institutions (CDFI) are grassroots entities that guide sustainable economic growth by prioritizing the needs of marginalized populations. Traditionally, CDFIs have been evaluated by the extent to which their activities foster local development or lessen economic inequities. Our analysis suggests this phenomenon is the result of deeply entrenched alliances between the public sector and the interests of CDFI stakeholders. These institutions are major resource providers and thus have set the narrative around CDFI impact measurement and evaluation. We propose an alternative framework that prioritizes changes in individual welfare which foster economic autonomy and improve relationships with community institutions. Our framework is admittedly theoretical, and further iteration will be required to operationalize it into a workable concept. Ultimately though, the existing framework around these issues is broken – and we find scant evidence that it can be salvaged.

Community Development Financial Institutions (CDFIs) are grassroots organizations that provide equitable access to financial capital. While a robust body of evidence supports the ability of CDFIs to promote holistic and sustainable development, attempts to systematically evaluate the industry have yielded disparate and often confounding results. We apply an institutional theory lens to examine challenges to meaningful data collection, impact measurement, and program evaluation. Our data show how regulators, major funders, and third-party rating organizations have applied indirect and direct pressures that have systematically lowered the capacity of nonprofit CDFI loan funds. This combination of coercive, mimetic, and normative isomorphic forces has (1) hampered meaningful data collection, (2) created a lack of staff expertise in these areas, (3) raised the cost and complexity of utilizing technology systems to improve evaluation processes, and (4) fostered industry norms which de-prioritize meaningful evaluation. The data suggest several ways for stakeholders to improve these trends. For example, funders might consider providing support which builds organizational capacity via unrestricted operating grants and recurring financial commitments.
