Texte intégralThe work of this thesis responds to the context of the evolution of engine depollution norms together with the increase of the clientrequirements. It proposes a complete methodology of engine calibration considering dynamic effects with the aim of an efficient control in terms of emissions and performances. The method is divided into four steps: the dynamic design of experiments generating a set of RDE (Real Driving Emissions) cycles and dynamic variations of engine parameters using low discrepancy sequences: test results are used to train a dynamical model using LSTM neural network to predict output dynamic variations(CO, HC, NOx, Exhaust flow and temperature). The trained model is used in an optimization loop to calibrate the engine parameters using a genetic algorithm. The catalyst warm-up phase is the chosen phase for the development of the method. It is the phase occuring from engine start until the catalyst is the most efficient. It is indeed the phase with the most important emissions which is coherent with the aim of the engine calibration. The results showed noticeable improvements of CO, HC and Nox reduction compared to the steady state (baseline) method

Texte intégralThe classical and hierarchical view of the motor system has been challenged since the discovery of other structures able to modulate the motor output in the framework of a hodotopic model. The aim of this work was to study the motor control network thanks to direct electrostimulations performed during awake surgery for brain tumors. This method has shown its effectiveness to preserve motor functions while giving new highlights about the organization of the motor system. In our work, motor control has been studied through the negative motor phenomenon, which consists in a complete arrest of movement without loss of tonus or consciousness during electrostimulations. Initially described at a cortical level, our work demonstrated the possibility to elicit negative motor phenomenon in both hemispheres at a subcortical level. Moreover, we identified a bilateral modulatory motor pathway able to inhibit both upper limbs during unilateral subcortical stimulations. We also shown that fibers driving negative motor responses are organized in a somatotopic manner, like the pyramidal pathway. Resection of these fibers lead to a supplementary motor area syndrome with permanent deficit in fine motor skills and bimanual coordination. These results explain the neurological deficits which might occur after surgery in premotor areas when no active brain mapping is performed, that is, when only primary motor structures are sought. They emphasize the necessity to perform a motor mapping during awake surgery whatever the side and hemispheric dominance. These subcortical results led us to define the concept of negative motor networks and their involvement in motor control networks. Evidences of this network allowed us to explore the cortical level and to report a well-defined organization of the negative motor area, different from the random or somatotopic distributions previously described. This effector-dependent and redundant organization in several areas defined by direct electrostimulations has been helpful to confirm the rostro-caudal and dorso-ventral segregation of the precentral gyrus. Consequently, it was possible to propose several hypothesis about the role of these networks. We presume that they are constituted by several large-scale interconnected networks, based on internal inhibitor mechanisms, whose role goes from modulation of the motor output in a competitive model of decision-making integrated in the negative motor area to real inhibition of motor behaviors thanks to cortico-basal ganglia circuitry. The probabilistic map created with these works will be helpful to plan surgery but could also provide regions of interest for brain stimulations therapies as well as neuroscientific research

Texte intégralMotor cortex stimulation (MCS) is a neurosurgical technique developed on empirical basis and currently used as last solution for patients suffering from refractory neuropathic pain. The present work is a new attempt among other contemporary studies aiming to understand the mechanisms of action of MCS, which remain incompletely elucidated at that time. The main objective of this thesis is to study the electrophysiological effect of MCS at the thalamic level, in a cat model. The first part of this work aims to establish the stereotactic somatotopic map of the cat motor cortex (MC), not available so far in the literature. Based on this mapping, we created and validated a cat model of MCS, using a mini-invasive electrode implantation. The second part of this study included a recording and analysis of the potential changes of the unitary extracellular activity of cells located in the thalamic ventro-postero-lateral (VPL) nucleus, induced by different MCS protocols. Our results indicate a modulation of the VPL cells activity after MCS, depending on the nociceptive or non-nociceptive nature of the recorded thalamic cell. MCS increases the activity of non-nociceptive cells and decreases that of nociceptive cells. For a given cell the matching between the somatotopy of the MC stimulated region and the receptive field localization of the recorded thalamic cell is not a prerequisite for obtaining such a modulation. In conclusion, the present work has proven a neuro-modulatory differential effect of MCS on nociceptive and non-nociceptive cells in the thalamic VPL nucleus

Texte intégralThe aim of the thesis was to investigate the mapping of the anatomofunctional organization of the human sensorimotor system and how volutional movements of human are produced and controlled. Neuroimaging and especially DTI, fine anatomo-functional observation in patient and direct electrical stimulation were considered. This multi-modal approach permitted to improve our understanding of sensorimotor organization in humans. In the first study, we showed that awake brain surgery with the use of direct electrical stimulation is a safe and efficient procedure in children in order to decrease post-operative neurological deficit. It improves the accuracy of detecting eloquent area, with a good tolerance from a neuropsychological and psychological aspect. Age-adapted neuropsychologic preparation may enable offering ABS even to younger children on an individual basis. In a second series of two studies, we showed that the dorso-posterior part of the parietal cortex is a key structure in the complex organization of movement in human with a S1-DPPr-M1 loop. In the first study, direct electrical stimulation of focal cortical site in the dorso-posterior part of the parietal cortex triggered inhibition of movement production and blocked ongoing movement without producing muscle contraction or conscious movement sensation. In the second study, we aimed to find a direct projection from the PRR, defined in the first study (Desmurget et al., 2018), to the primary motor cortex and the primary somatosensory cortex. Thanks to the DTI state-of-the-art tractography, we succeeded in finding such major ipsilateral streamlines projecting in the well-known hand knob region giving new insights of the white matter structures involved in the inhibition of volitional hand movements. These observations confirm clinical per-operative data showing that stimulating the counterpart of PRR in humans can disrupt hand movements ipsilaterally, irrespective of the hemisphere. Moreover, our results shed light on the implication of the PRR for the volitional hand sensorimotor operating behavior. In the last study, we investigate the impact of early cerebellar damage on long-term functional recovery in 3 groups of posterior fossa survivors, comparable with respect to their tumoural characteristics but operated at different ages: young (≤ 7 years), middle (> 7 years and ≤ 13 years) and old (> 13 years). Daily (Health-related Quality of Life -hrQol-, Performance Status -PS-), motor (International Cooperative Ataxia Rating Scale -ICARS-, Pegboard Purdue Test -PegBoard-) and cognitive (Full Scale Intelligence Quotient -FSIQ-) functioning were measured. Early age at surgery, lesion of deep cerebellar nuclei and post-operative radiotherapy had a significant, independent negative influence on long term recovery. These results support the existence of an early critical period of development during which the cerebellar "learning machine" is of critical importance
