Abstract Background Defining ELF concentrations is desired for antibiotics developed for pneumonia. For ethical reasons, BAL sampling in humans is routinely done at a single time point, thereby creating ambiguity in the precise ELF profile. It is unknown if additional sampling of the ELF would lead to more accurate estimates of exposure. The swine pneumonia model was used to characterize the full ELF profiles (5-BAL) of two β-lactams for comparison with models employing 1-BAL (1B) and 2-BAL (2B) sampling time points only. Methods Sixteen ventilated swine were infected with Pseudomonas aeruginosa to establish pneumonia and then treated for 72 hours with ceftolozane/tazobactam (C/T) 50 mg/kg q8h (n = 8) or piperacillin/tazobactam (TZP) 200 mg/kg q8h (n = 8). Plasma and BAL concentrations were measured in each swine at 1, 2, 4, 6, and 8 hours after the first dose. Urea correction was used to calculate ELF values. Ceftolozane and piperacillin plasma and ELF data were fitted to a two compartment model using the nonparametric adaptive grid program in Pmetrics. Hypothetical models were refitted after randomly selecting either 1B or 2B sampling time points from each swine. A 5,000 subject Monte-Carlo simulation was performed for each model to define PTA (60% free time above the MIC) and ELF penetration [area under the curve in ELF (AUCELF) vs. free AUCplasma]. The KS-test was used to analyze distribution differences, reporting maximum vertical deviation (D) as percent difference; D < 20% was defined as negligible. Results Thirty-two C/T and 34 TZP plasma samples and 29 and 32 BAL samples were available for the full model, respectively; 1B and 2B sampling models used eight and 16 BAL samples. All models adequately fitted the data. C/T PTA at 4 mg/L was 94.8, 96.1, and 98.0%, for the full, 1B and 2B models. TZP PTA at 16 mg/L was 55.8, 46.8, and 46.7%, respectively. C/T median [interquartile range] penetration differences were negligible between the full (65% [25––109]) and 1B (72% [45––125], D = 15%) or 2B models (62% [32––111], D = 6%). TZP penetration differences were also minimal between the full (32% [9––67]) and 1B (17% [5––49], D = 18%) or 2B models (27% [9––44], D = 15%). Conclusion These data suggest that antibiotic ELF models constructed from a single BAL time point result in similar exposure estimates to full ELF profiles. Disclosures G. The safety of the unpowered mooring vessel was influenced by human factors, vessel condition, meteorological factors, hydrological and environmental factors. To ensure the safety of the unpowered mooring vessel during mooring, on the basis of unascertained measure evaluation model, the method of Monte Carlo and interval weight was put forward to assess the safety of the vessel. This method enables policymakers, in the treatment of evaluation, to avoid weighing the key factors from the traditional model. It also provides more valid intervals for policymakers while dealing with uncertain information of safety elements and errors caused by human preference. Through a random value of interval weight using Monte Carlo simulation method, the final average value of the simulation is taken as the final weight. Finally, a 16000t unpowered mooring cargo vessel is taken as an example to have a detailed discussion of the application of this method, and a safety result is obtained according to each key element. This method can provide new evaluation method and references to policymakers concerned.

In this study, a hybrid reliability methodology via Monte Carlo simulation techniques with radial basis function neural network (RBFNN) is presented. Monte Carlo simulation is a powerful tool, simple to implement, and capable of solving a broad range of reliability problems. However, its use for the evaluation of very low probabilities of failure implies a great number of analyses, and the computational time highly increases. In practice, the size of a design problem can be very large and the limit state functions (LSFs) are usually implicit in terms of the random variables. A hybrid method consisting of Monte Carlo simulation and RBFNN is proposed in the present study to approximate the LSF or failure function of the structure. Therefore, the computational burden of Monte Carlo simulation decreases significantly. A distinctive feature of this method is the introduction of an explicit approximate LSF. Using the parameters of the RBFNN, the explicit formulation of the LSF is derived. By introducing the derived approximate LSF, the failure probability can be easily estimated. In order to assess the effectiveness of the proposed methodology, some illustrative examples including frame structures are considered, and the numerical results are verified.

The article is devoted to modelling the duration of cargo operations at intermediate stations on the route of a modular express freight container train (MEFCT), that is, a train with a fixed schedule heading from the initial to the final station without remarshalling a rolling-stock, but performing the cargo handling operations for loading/unloading of containers at intermediate stops on the route. Cargo handling operations at intermediate stations of a MEFCT entail a non-productive run of a loader along the train from one container to be reloaded to the other. In the case of a random placement of containers on a train, the completion time of the complex of cargo handling operations will be a random variable. Modelling of this random variable is carried out on the basis of the Monte-Carlo method in order to determine the most probable time of stopping of a train to perform cargo handling operations, as well as possible deviations from the average value. At the same time, duration of the complex of cargo handling operations at the station is proposed to be estimated depending on the volume of loading/unloading operations of containers at each point and the distance travelled by a loader along a train, which is given in the model by a random evenly distributed value. Based on numerical experiments with the model, the regression dependence of the time of cargo handling operations on the volume of container loading was established, as well as variations of this value due to the random nature of the placement of containers on a train. The model allows you to form a reasonable timetable for a MEFCT and increase the reliability of departure time of a train.

Contemporarily, with the rapid development of the computer science, the calculation ability has been boosted a lot, which paves a path for carrying out simulations with random variable. In this case, Monte-Carlo simulations can be easily achieved with the state-of-art techniques. As one of the useful statistical tools, Monte-Calo simulation can be implemented in different areas including environment and Chemistry field. On this basis, this article searched several information to determine the usage of the simulation in the certain field. To be specific, the usage of Monte-Calo simulations of Chemistry and Environmental Science will be discussed with specific samples and data analysis. Moreover, relevant simulations results and corresponding applications will be demonstrated. In addition, this study will also summarize some limitations (e.g., the large range of the data). On this basis, it provides some suggests to elaborate the function in more digits and stating the conclusion. These results shed light on guiding further simulations in the context of the Monte-Carolo configuration.

Although the value of factor weight in an evaluation work is deterministic, the solving process is random, so connection between weight solution with digital characteristics or distribution functions of specific random variables or random process could be build. Using stochastic simulation method to get a lot of random solutions to the problem, expectation of the random solutions can be used as a estimation solution. On basis of idea of Monte Carlo simulation, this paper analyzed the probability process of calculating factor weight, and provided the procedures of estimating factor weight by means of Monte Carlo simulation. Through discussion and example in this paper, feasibility and validity of this method were proved, which may make foreshadowing for follow-up research work.

A Bayesian framework for the analysis of radial basis functions (RBF) is proposed that accommodates uncertainty in the dimension of the model. A distribution is defined over the space of all RBF models of a given basis function, and posterior densities are computed using reversible jump Markov chain Monte Carlo samplers (Green, 1995). This alleviates the need to select the architecture during the modeling process. The resulting networks are shown to adjust their size to the complexity of the data.

Neste artigo iremos buscar compreender as relações desenvolvidasentre a Universidade do Brasil e a Universidade de São Paulo (campus de São Carlos) por meio da atuação do matemático Achille Bassi e, desta forma, refletir sobre alguns elementos contribuintes ao processo de institucionalização da Matemática no Ensino Superior brasileiro. Para isso, nos deteremos resumidamente à biografia do personagem e ao contexto do processo de criação das duas Instituições de Ensino: aFaculdade Nacional de Filosofia (FNFi), integrante da Universidade do Brasil (UB), fundada em 1939 no Rio de Janeiro; e a Universidade de São Paulo (USP) criada em 1934 e expandida para o interior paulista, especificamente para São Carlos, com a criação da Escola de Engenharia de São Carlos (EESC) em 1953. A pesquisa baseada naanálise de fontes documentais do acervo de Achille Bassi e da EESC discutiu o processo inicial de institucionalização da Matemática em São Paulo, sinalizando para a importância da circulação de conhecimentos e matemáticos europeus na implantação destas instituições brasileiras.

The use of Data Assimilation methodologies, known also as a data adjustment, liaises the results of theoretical and experimental studies improving an accuracy of simulation models and giving a confidence to designers and regulation bodies. From the mathematical point of view, it approaches an optimized fit to experimental data revealing unknown causes by known consequences that would be crucial for data calibration and validation. Data assimilation adds value in a ND evaluation process, adjusting nuclear data to particular application providing so-called optimized design-oriented library, calibrating nuclear data involving IEs since all theories and differential experiments provide the only relative values, and providing an evidence-based background for validation of Nuclear data libraries substantiating the UQ process. Similarly, it valorizes experimental data and the experiments, as such involving them in a scientific turnover extracting essential information inherently contained in legacy and newly set up experiments, and prioritizing dedicated basic experimental programs. Given that a number of popular algorithms, including deterministic like Generalized Linear Least Square methodology and stochastic ones like Backward and Hierarchic or Total Monte-Carlo, Hierarchic Monte-Carlo, etc., being different in terms of particular numerical formalism are, though, commonly grounded on the Bayesian theoretical basis. They demonstrated sufficient maturity, providing optimized design-oriented data libraries or evidence-based backgrounds for a science-driven validation of general-purpose libraries in a wide range of practical applications.

Quantum Monte Carlo (QMC) methods are the gold standard for studying equilibrium properties of quantum many-body systems. However, in many interesting situations, QMC methods are faced with a sign problem, causing the severe limitation of an exponential increase in the runtime of the QMC algorithm. In this work, we develop a systematic, generally applicable, and practically feasible methodology for easing the sign problem by efficiently computable basis changes and use it to rigorously assess the sign problem. Our framework introduces measures of non-stoquasticity that—as we demonstrate analytically and numerically—at the same time provide a practically relevant and efficiently computable figure of merit for the severity of the sign problem. Complementing this pragmatic mindset, we prove that easing the sign problem in terms of those measures is generally an NP-complete task for nearest-neighbor Hamiltonians and simple basis choices by a reduction to the MAXCUT-problem.

In a case study, the exact solution of a self-consistent Langevin equation associated with a nonlinear Fokker–Planck equation is derived. On the basis of this solution, a Monte Carlo simulation scheme for the Langevin equation is proposed. The case study addresses a generalized geometric Brownian walk that describes the collective dynamics of a large set of interacting stocks. Numerical results obtained from the Monte Carlo simulation are compared with analytical solutions derived from the nonlinear Fokker–Planck equation. The power of the Monte Carlo simulation is demonstrated for situations in which analytical solutions are not available.

The FSUE Central Research Institute of Machine Building (TsNIIMash), Rocket and Space Corporation “Energia”, and Institute of Solar-Terrestrial Physics of Siberian Branch of Russian Academy of Sciences (ISTP SB RAS) jointly conducted the active space experiment “Radar-Progress” in 2007–2015. During this experiment, we used the Irkutsk Incoherent Scatter Ra-dar to study space-time characteristics of ionospheric disturbances generated by exhaust products of “Pro-gress” cargo spacecraft engines. As the basic effect dur-ing exhaust product injection we consider the formation of new centers for recombination of ambient iono-spheric ions O+ on molecules of water and carbon diox-ide. This produces an ionization “hole” in the region of injection. In nighttime conditions when the majority of experiments were performed, this hole was filled by hydrogen ions from the plasmasphere, thus the ion composition in the vicinity of the hole and incoherent scatter spectra were changed. For successful obser-vation of the ionization hole dynamics, the critical fac-tors are the degree of radar antenna diagram filling by exhaust products and the velocity of the thermospheric neutral wind, which makes exhaust gases move from the antenna diagram. These two factors lead to poor repeatability of successful experiments. Successful experiments recorded a decrease in electron density up to 35 % in the hole that existed for 30 min. The lifetime of the region with high concentration of H+ ions can be as long as one hour.

This paper presents a probabilistic algorithm for segmenting and recognizing text embedded in video sequences based on adaptive thresholding using a Bayes filtering method. The algorithm approximates the posterior distribution of segmentation thresholds of video text by a set of weighted samples. The set of samples is initialized by applying a classical segmentation algorithm on the first video frame and further refined by random sampling under a temporal Bayesian framework. This framework allows us to evaluate a text image segmentor on the basis of recognition result instead of visual segmentation result, which is directly relevant to our character recognition task. Results on a database of 6944 images demonstrate the validity of the algorithm.

This paper examines which basis functions are efficient to employ a combined method of Hull and White (1990) with the Monte Carlo simulation when we price a callable range note or a callable bond. We use the Huge and Rom-Poulsen (2007) method which has modified the least squared Monte Carlo simulation proposed by Longstaff and Schwartz (2001) to reduce the estimation errors of the continuation value or the underlying assets. To use Monte carlo Simulation for pricing the early exercise premium, it is essential to accurately estimate the continuation value, because the investors will choose the higher value between the exercise and the continuation value at the possible early exercise dates. The main purpose of this paper is to analyze the estimation errors originating from the choice of the basis functions for the underlying asset and the continuation value estimation. We choose the callable bond and the callable range accrual note to show which basis functions are reliable to reduce the estimation errors. For this purpose, we replicate the callable range accrual note with a portfolio of a fixed rate bond and a delayed digital option. We use several basis functions such as a constant, the instantaneous interest rates, and the range in order to see which basis function is efficient for our purpose. We examine several combinations of the basis functions depending on which basis functions will be used for the underlying asset or the continuation value estimation. We show that the range which is an important determinant of the callable range accrual note is an effective basis function to accurately determine the underlying asset and the continuation value for the pricing of the callable range accrual note.

Water-quality models that are simple yet sound and reliable and that correspond to water-quality criteria that include magnitude, frequency, and duration components are needed. Monte Carlo models are developed on the basis of available flow and water-quality data and a deterministic water-quality model appropriate for the problem at hand and the data available. Monte Carlo models yield time series and probability distributions of constituents of interest in conformance with water-quality criteria. The application of a Monte Carlo model to the probabilistic evaluation of ammonia toxicity in Milwaukee's Outer Harbor is presented here. Under typical operating conditions for the Jones Island Wastewater Treatment Plant, ammonia toxicity was found to not be a problem for the Outer Harbor. The Monte Carlo model then was used to determine effluent limits that would meet the ammonia toxicity criteria.

Abstract The paper considers the theoretical foundations of the Monte Carlo method based on the limit theorems of probability theory. Simulations of a coin toss and the number of determination are given and described. The authors proved experimentally that sufficiently high accuracy of solving problems with the Monte Carlo method application is guaranteed only when a large number of experiments are carried out. Therefore, the Monte Carlo method is practically implemented only with the computer. The procedure for generating random numbers is a fundamental basis of the statistical test method. The average values tend to some constant with an unlimited increase in the number of experiments. It was found on the basis of a comparative analysis of the obtained results when studying the laws of large numbers by the method of simulation statistical modeling in MathCad and MS Excel. The Limit theorems guarantee the high quality of statistical estimates obtained by the Monte Carlo method for a large model dimension n. However, practically acceptable estimates of the simulated system’s characteristics can often be obtained even at relatively small values of n (of a few tens and hundreds). It has been proven that averaging the values obtained over several runs of the model significantly improves the quality of statistical estimates.

The nuclear pore complex regulates cargo transport between the cytoplasm and the nucleus. We set out to correlate the governing biochemical interactions to the nanoscopic responses of the phenylalanineglycine (FG)–rich nucleoporin domains, which are involved in attenuating or promoting cargo translocation. We found that binding interactions with the transport receptor karyopherin-β1 caused the FG domains of the human nucleoporin Nup153 to collapse into compact molecular conformations. This effect was reversed by the action of Ran guanosine triphosphate, which returned the FG domains into a polymer brush-like, entropic barrier conformation. Similar effects were observed in Xenopus oocyte nuclei in situ. Thus, the reversible collapse of the FG domains may play an important role in regulating nucleocytoplasmic transport.

Using bold diagrammatic Monte Carlo method, we solve the Schwinger–Dyson equations and find the spectral density function of the theory beyond the weak coupling regime. We then compare our result with the one obtained from the perturbation theory. In order to analyze the effectiveness of interaction of the cargo terminal operator with customs authorities and participants in foreign economic activity, a four-sector model of the network diagram of the interaction stages between participants in the transport and logistics process at the sea cargo terminal is presented in the paper; the model allows visualizing and controlling the process of aggregated relationships during cargo operations and the provision of customs services in the area of seaports activity. Calculations of works time parameters are performed on the basis of a four-sector model, in which the event is divided into four sectors, where the number of the event is indicated on the top, the early start of work is indicated on the left, the late end of work is marked in the right sector, and the number of the initial event of the previous work is indicated in the lower sector, according to which the critical path passes. At the same time, simulation modeling of the system of relations between the participants in the transport and logistics process at the sea cargo terminal is carried out. When modeling by the Monte Carlo method, a set of parametric values presented in the form of a probability distribution is used. It is noted that the operations performed in the simulation are subject to the normal distribution law, since a large number of participants are involved in the transport and logistics system. The results of the algorithm operation are presented in the form of a data array, which is a sequence of works performed during the interaction of participants in the transport and logistics process at the sea cargo terminal. One of the methods for modeling randomly distributed variables is the Box-Muller generation method. The use of the Box-Muller method in the work makes it possible to apply the fundamental law of probability transformation to obtain two independent normal variables that have a standard normal distribution. Thus, the chosen method gives a set of random numbers that have a normal distribution with zero mean and unit standard deviation. The results of simulation modeling allow us to conclude that the aggregated system of relationship between participants in the transport and logistics process, which uses the technology of preliminary declaration in order to make a decision on the early placement of goods in the inspection zone, is effective.

Model for market of Uzbekistan cargo transportation is proposed. Positive sides of proposed scheme are listed. Practical significance: Improvement of market model of rail cargo transportation makes it possible to renew cargo car and container park, to create concurrence in cargo car and container provision for loading and to reduce transport constituent on account of concurrence.

The workspace is one of the most important parameters for evaluating robot flexibility and is important for optimizing robotic configuration, motion planning and control. Firstly, a kinematic model of the manipulator based on its basic structure was put forward. The systems of connection coordinates are established and the direct kinematic solution derived using DH methods. On its basis, the working space of the manipulator analyzed by the Monte Carlo method, based on random probability and software simulation MATLAB for the structural parameters of the robot. A cloud of workspace points has been compiled. Considering the problem of insufficient accuracy of the traditional Monte Carlo method in calculating the working space of the robot, an improved Monte Carlo method using the Beta distribution proposed.

Configurations of the polymer state in rubbers, such as so-called isotropic (random) and anisotropic (almost aligned) states, are symmetric/asymmetric under space rotations. In this paper, we present numerical data obtained by Monte Carlo simulations of a model for rubber formulations to compare these predictions with the reported experimental stress–strain curves. The model is defined by extending the two-dimensional surface model of Helfrich–Polyakov based on the Finsler geometry description. In the Finsler geometry model, the directional degree of freedom σ → of the polymers and the polymer position r are assumed to be the dynamical variables, and these two variables play an important role in the modeling of rubber elasticity. We find that the simulated stresses τ sim are in good agreement with the reported experimental stresses τ exp for large strains of up to 1200 % . It should be emphasized that the stress–strain curves are directly calculated from the Finsler geometry model Hamiltonian and its partition function, and this technique is in sharp contrast to the standard technique in which affine deformation is assumed. It is also shown that the obtained results are qualitatively consistent with the experimental data as influenced by strain-induced crystallization and the presence of fillers, though the real strain-induced crystallization is a time-dependent phenomenon in general.

Congelation accident in pipeline of crude oil is always being a serious problem in china, which is mainly determined by the properties of domestic crude oil such as high content of wax, high pour point and viscosity. When oil pipeline operates at low throughput or shutdown, congelation accident always happen as the temperature of oil drops, which causes malignant accidents and significant financial losses. Using improved Monte-Carlo method of reliability theory, the failure possibility of congelation was numerically calculated. Whereas, the consequences of congelation accident were also fully analyzed, these can provide evidences for accident treatment, planned maintenance, security operation and scientific management.

We discuss a method called quasi-sparse eigenvector diagonalization which finds the most important basis vectors of the low energy eigenstates of a quantum Hamiltonian. It can operate using any basis, either orthogonal or non-orthogonal, and any sparse Hamiltonian, either Hermitian, non-Hermitian, finite-dimensional, or infinite-dimensional. The method is part of a new computational approach which combines both diagonalization and Monte Carlo techniques.

Autophagy is an evolutionarily conserved intracellular degradation process, through which large cellular cargos are sequestered into double-membrane vesicles (autophagosomes) and delivered to the lysosome. Starvation-induced autophagy represents a general non-selective degradation pathway that breaks down cellular components for energy replenishment. Alternatively, selective autophagy targets specific organelles, protein aggregates or invading pathogens that need to be precisely removed from the cell during development or pathogenic infection. Selective autophagy receptors noncovalently bind to the family of ATG8 modifiers and can be controlled by post-translational protein modifications, including ubiquitination and phosphorylation. In this article, we review recent advances in our understanding of the molecular basis of cargo selection in autophagy.
