The caspase recruitment domain (CARD) is a protein-binding module that mediates the assembly of CARD-containing proteins into apoptosis and NF-κB signaling complexes. We report here that CARD protein 11 (CARD11) and CARD protein 14 (CARD14) are novel CARD-containing proteins that belong to the membrane-associated guanylate kinase (MAGUK) family, a class of proteins that functions as molecular scaffolds for the assembly of multiprotein complexes at specialized regions of the plasma membrane. CARD11 and CARD14 have homologous structures consisting of an N-terminal CARD domain, a central coiled-coil domain, and a C-terminal tripartite domain comprised of a PDZ domain, an Src homology 3 domain, and a GUK domain with homology to guanylate kinase. The CARD domains of both CARD11 and CARD14 associate specifically with the CARD domain of BCL10, a signaling protein that activates NF-κB through the IκB kinase complex in response to upstream stimuli. When expressed in cells, CARD11 and CARD14 activate NF-κB and induce the phosphorylation of BCL10. These findings suggest that CARD11 and CARD14 are novel MAGUK family members that function as upstream activators of BCL10 and NF-κB signaling.

ABSTRACT Nod2 (CARD15) is a macrophage-specific protein containing two CARD domains, a large nucleotide binding domain and leucine-rich repeats. Human genetic studies have linked mutations in NOD2/CARD15 with Crohn's disease, although the mechanisms involved are unknown. However, Nod2 has been proposed to directly bind bacterial lipopolysaccharide (LPS) and subsequently act as an activator of NF-κB via the association of the CARD domains with Rip2/RICK/CARDIAK. This is hypothesized to constitute a pathogen recognition pathway distinct from Toll-like receptor 4-mediated recognition of LPS. Using targeted mutagenesis, we introduced a mutation to delete the CARD domains of mouse Nod2. Mice lacking Nod2 were indistinguishable from controls and showed no signs of intestinal pathology. Macrophages responded normally to multiple Toll-like receptor agonists in terms of NF-κB target activation, mitogen-activated protein kinase activation, and cytokine secretion. However, Nod2−/− mice were significantly protected in endotoxin challenge experiments, and Nod2−/− macrophages were refractory to muramyl dipeptide stimulation. These results argue that Nod2 does not play an essential, nonredundant role in the response of macrophages to bacterial products but rather plays unexpected roles in regulating systemic responses to pathogens.

Due to the special location and structure of transmission system on high-speed train named CRH5, dynamic unbalance state of the cardan shaft will pose a threat to the train servicing safety, so effective methods that test the cardan shaft operating information and estimate the performance state in real time are needed. In this study a useful estimation method based on ensemble empirical mode decomposition (EEMD) is presented. By using this method, time-frequency characteristic of cardan shaft can be extracted effectively by separating the gearbox vibration acceleration data. Preliminary analysis suggests that the pinions rotating vibration separated from gearbox vibration by EEMD can be used as important assessment basis to estimate cardan shaft state. With two sets gearbox vibration signals collected from the in-service train at different running speed, the comparative analysis verifies that the proposed method has high effectiveness for cardan-shaft state estimate. Of course, it needs further research to quantify the performance state of cardan shaft based on this method.

Analyze the deformation process detection cardan anti extruded deformation in advance and is extruded deformation. Meanwhile analysis showed that the cardan forgings have internal 59.929% area completely modification, have 4.7944% area did not happen dynamic recrystallization and didn't get forging modification, and forecasts the cardan internal grain size and distribution, the micro analysis for the prediction of forgings performance and design heat treatment process, provides the basis.

For this, it is necessary to develop a flexible informational system that would connect the constructive design of cardan transmissions to the technological design within a unitary structure. Due to the typodimensional diversity of cardan transmissions, the design and manufacturing of the various components for a certain application domain (medical aparature, dental and agricultural machines, lifting and transportation machines etc.) has been typified. In order to study the problems that occur in the practice of using cardan transmissions, there has been realised a questionnaire-based descriptive research. We insisted on calculation of the cardan transmission for Dacia 1304/1307.

Cells release extracellular vesicles (EVs) to communicate in a paracrine manner with other cells, and thereby influence processes, such as angiogenesis. The conditioned medium of human cardiac-derived adherent proliferating (CardAP) cells was recently shown to enhance angiogenesis. To elucidate whether their released EVs are involved, we isolated them by differential centrifugation from the conditioned medium derived either in the presence or absence of a pro-inflammatory cytokine cocktail. Murine recipient cells internalized CardAP-EVs as determined by an intracellular detection of human proteins, such as CD63, by a novel flow cytometry method for studying EV–cell interaction. Moreover, endothelial cells treated for 24 h with either unstimulated or cytokine stimulated CardAP-EVs exhibited a higher tube formation capability on Matrigel. Interestingly, unstimulated CardAP-EVs caused endothelial cells to release significantly more vascular endothelial growth factor and interleukin (IL)-6, while cytokine stimulated CardAP-EVs significantly enhanced the release of IL-6 and IL-8. By nCounter® miRNA expression assay (NanoString Technologies) we identified microRNA 302d-3p to be enhanced in unstimulated CardAP-EVs compared to their cytokine stimulated counterparts, which was verified by quantitative polymerase chain reaction. This study demonstrates that both CardAP-EVs are pro-angiogenic by inducing different factors from endothelial cells. This would allow to select potent targets for a safe and efficient therapeutic application.

In this article, a nonlinear dynamic model with five degrees-of-freedom for a four-wheel-drive vehicle driveline connected by a cardan joint, including dynamic intersection angle, nonconstant velocity, and additional moment caused by the cardan joint, is established by using the Lagrange method to analyze the driveline coupling vibration in both torsional and lateral directions. High-order Runge–Kutta algorithm is applied to solve the differential equations and to calculate transient responses of the driveline rotors under acceleration condition. The color maps and second-order vibration of the driveline are acquired by frequency spectrum analysis and order tracking analysis, respectively. The second-order vibration and noise of the driveline and vehicle interior caused by the cardan joint is validated by vehicle experimental results and reduced effectively by decreasing intersection angle of the cardan joint under the operational condition. Moreover, application of a flexible coupling instead of the cardan joint significantly reduces the second-order vibration but simultaneously generates low-level third-order vibration.

An experiment has been conducted indicating invalidity of the assumption of negligible intermediate-joint axial forces in Cardan joint (universal joint) dynamic analysis. It has been observed that increased driveline torque induces the radially-inward deflection of the yoke arms of a Cardan joint, while increased speed will deflect the yoke arms outward. Some suggestions are given to optimize Cardan joints for either high torque or high speed.

The present study was undertaken to investigate the cardiovascular effect of three extracts from the root bark of Anthocleista schweinfurthii Gilg.: an aqueous extract (AE), a dichloromethane extract (DCMR) and a fraction enriched in cardiac glycoside type compounds (CARDAN). In isolated perfused frog heart, bolus injection of the extracts produced a positive inotropic effect. The responses to AE and DCMR, but not to CARDAN, were depressed by propranolol. In isolated rat aorta, DCMR produced a transient increase in contractile tension while AE and CARDAN induced a sustained constriction. AE vasoconstrictor effect was abolished by phentolamine, while contraction evoked by CARDAN was antagonized by verapamil. In aortic rings contracted in low K+ media, the addition of K+ evoked a relaxation, which was abolished by ouabain, depressed by DCMR but not affected by either AE or CARDAN. These observations indicate that Anthocleista schweinfurthii contains substances that promote vasoconstriction and increase cardiac contraction. The effect of DCMR was only partially mediated by inhibition of the Na+ pump while the mechanism of action of AE and CARDAN was distinct from the inhibition of the Na+,K+-ATPase pump, but could involve adrenergic receptors, or either direct or indirect activation of L-type calcium channels.

The paper briefly presents the geometry, structure, and kinematics of a universal joint, very commonly used in machine building, especially today for heavy and engine-driven vehicles and transmissions located in different areas as well as for all-wheel-drive vehicles. The universal joint, or the cardan cross, conveys the rotation movement from one bridge to the other (when the rotary shaft suffers both movements, upward and downward). The kinematic scheme of a cardan transmission is composed of two cardan shafts (one input and one output), both of which are equipped with a cardan cross (universal joint or universal joint). Between the two universal couplings, a further (additional) cardan shaft (axle) is mounted. This mechanism is designed to transmit the mechanical movement (within a vehicle) from one bridge to the other. If the vehicle's motor is on the front and with on the rear axle transmission, or vice versa when the vehicle's engine is on the rear and the transmission is on the front axle, or when we have multiple (multi-axle) transmission on heavy vehicles or 4x4 cars.

Tower structure is sensitive to hurricane and earthquake, and it is easy to generate large deflection and dynamic response. The multiple cardan gyroscope has two rotational degrees of freedom, which can generate strong moments to constrain the two horizontal orthogonal deflections if the rotor operates in high speeds, so the structural dynamic responses can be decreased. Hence, the method of dynamic control of the tower structure under wind load and earthquake action is proposed by using the multiple cardan gyroscopes as the dampers. The dynamic mechanism and the fixed axis principle of the multiple cardan gyroscope are introduced, and the dynamic equation of the gyroscope is established. The damping mechanism of the gyroscope is also described. For the tower structure equipped with the multiple cardan gyroscope dampers, the multidimensional control equation considering torsion effect is established, and the equivalent state space equation is presented. Taking a TV Tower with a number of gyroscope dampers as an analysis example, the structural dynamic responses and damping performance under fluctuating wind loads and earthquake action is studied. The results show that the multiple cardan gyroscope dampers with suitable parameters can effectively decrease the structural vibration in horizontal directions and torsional direction.

The input-output displacement relations of two Cardan joints arranged in series on a driveline has been investigated in detail, including the effects of unequal joint angles, the phase angle between the two Cardan joints and also such manufacturing tolerance errors as nonright angle moving link lengths and offset joint axes. A combined Newton-Raphson method and Davidon-Fletcher-Powell optimization algorithm using dual-number coordinate-transformation matrices was employed to perform the analysis. An experiment was conducted to validate the results of the analysis. The apparatus consisted of a double-Cardan-joint driveline whose rotations were measured by optical shaft encoders that were sampled by a computer data-acquisition system. The equipment was arranged so that the phase angle between the joints and the offset angles between the shafts at each of the two joints could be readily varied. The “relative phase angle,” the difference between the phase angle of the two joints and the angle between the planes defined by the input and intermediate and the intermediate and output shafts, was found to be the significant factor. If the offset angles at both Cardan joints are equal, the double-Cardan-joint driveline functions as a constant-velocity coupling when the magnitude of the relative phase angle is zero. If the offset angles at the two Cardan joints are unequal, a condition prevailing in the important front-wheel-drive automobile steering-column application, then fluctuation in output velocity for a constant input velocity is minimized although not eliminated for zero relative phase angle.

Problem. An analysis of the literature has shown that some issues of the kinematics and dynamics of the Hooke’s joint are not fully covered. In particular, this applies to the determination of the kinematic characteristics of the relative movements of the links in the movable joints of the crosspieces and cardan yokes, as well as the acceleration of the crosspiece in the study of the hinge. These kinematic characteristics are the basis for dynamic analysis of the cardan transmission; they are used at research of operation of the bearings of a cardan joint cross; they are necessary to determine the energy expended to overcome the forces of friction in these kinematic pairs; they form the basis for evaluating transmission efficiency. Goal. The goal of this work is to determine the kinematic characteristics of the cross in its absolute and relative motions. Methodology. The research is based on the graphical and analytical method of kinematic analysis of spherical motion of a rigid body. At the initial stage, the calculated dependences for determining the angular velocity of the cross in its absolute and relative motions were obtained. The relative angles of rotation of the links in the movable joints of the crosspiece and cardan yokes were determined by integrating the corresponding differential equations. To determine the angular acceleration of the crosspiece, the theorem on the addition of angular accelerations of a rigid body rotating around two non-parallel axes was used. Results. The study of the kinematics of the cardan joint made it possible to identify and eliminate ambiguities in determining the relative angular velocities of the elements of the kinematic pairs of the cross and cardan yokes. Functional dependences for determination of angular acceleration of a crosspiece in absolute and relative movements, and also relative angles of rotation of links in kinematic pairs of a crosspiece and cardan yokes were received. It was shown that the angular acceleration of the crosspiece is a periodic function of the angle of rotation of the input link. During the operation of the hinge, the crosspiece creates insignificant periodic dynamic loads, which should be taken into account in accurate calculations of high-speed cardan transmissions. Practical value. The work is devoted to the study of the kinematics of the asynchronous hinge, which is the foundation for its dynamic analysis and ultimately aimed at improving the output performance of the cardan transmission. The research method combines mathematical calculations with simplicity and clarity of graphic constructions, which contributes to a deeper understanding of the kinematic features of a hinge of uneven angular velocities. The obtained results can be used in the design and study of motor-transmission units, which include the Hooke’s joint.

Abstract In the vast majority of technical applications, there is a necessity to transmit the torque from drive to driven machine together with the demand to overcome great distance between given devices. One of the solutions of this problem is the use of a cardan shaft. Operations of connecting shaft and drivetrain are accompanied by oscillations negatively influencing the driver comfort. Main subject of this article is the measurement of vibrations on different parts of a heavy truck and its evaluation by FFT analysis. Measurements are performed with two kinds of suspension bushing which are compared with to each other. Based on the previous measuring, influence of cardan shaft length was also investigated. First of all, dependency on the length of the cardan shaft on deflection angles for V arrangement was evaluated by analytical method. The theoretical introduction is followed by research of eigen frequency depending on the length of the cardan shaft. The results obtained by modal analysis in FEM are verified by experimental modal analysis [1].

This study sought to compare developmental controls in breeding a within two winter wheat improvement programs, one Australian and a CIMMYT/ICARDA program based in West Asia. Developmental controls considered were intrinsic earliness, and responses to photoperiod and to vernalization. The reliability with which each control on development had been measured was tested in separate experiments using the wheats utilized in the Australian program. Measures of intrinsic earliness showed significant agreement between experiments, better agreement being found for response to photoperiod and between integrated response to vernalization and time to double ridge after late summer sowings. The wheats utilized in the CIMMYTI/CARDA programs were found to be quick in intrinsic earliness, and to possess little response to photoperiod. While these controls varied more for the wheats utilized in the Australian program, commercial Australian winter wheats were similar to the CIMMYTI/CARDA lines. Lines utilized by both programs were represented by types with spring, facultative and winter habit. The clearest differences between programs were that CIMMYTI/CARDA winter wheats appeared to have much stronger response to vernalization than the Australian winter wheats. These findings suggest breeders would find a good proportion of segregates, from crosses between the Australian and the CIMMYTI/CARDA winter wheats, to be developmentally adapted to south-central New South Wales. This suggests CIMMYTI/CARDA winter wheats provide a matching pool from which to access germplasm to introduce new characters into Australian winter wheats.

The results of studying operational destruction of a high-loaded cardan shaft of the propeller engine made of steel 38KhN3MFA are presented to elucidate the cause of damage and develop a set of recommendations and measures aimed at elimination of adverse factors. Methods of scanning electron and optical microscopy, as well as X-ray spectral microanalysis are used to determine the mechanical properties, chemical composition, microstructure, and fracture pattern of cardan shaft fragments. It is shown that the mechanical properties and chemical composition of the material correspond to the requirements of the regulatory documentation, defects of metallurgical origin both in the shaft metal and in the fractures are absent. The microstructure of the studied shaft fragments is tempered martensite. Fractographic analysis revealed that the destruction of cardan shaft occurred by a static mechanism. The fracture surface is coated with corrosion products. The revealed cracks developed by the mechanism of corrosion cracking due to violation of the protective coating on the shaft. The results of the study showed that the destruction of the cardan shaft of a propeller engine made of steel 38Kh3MFA occurred due to formation and development of spiral cracks by the mechanism of stress corrosion cracking under loads below the yield point of steel. The reason for «neck» formation upon destruction of the shaft fragment is attributed to the yield point of steel attained during operation. Regular preventive inspections are recommended to assess the safety of the protective coating on the shaft surface to exclude formation and development of corrosion cracks.

Abstract CARD11 mediates lymphocyte activation by functioning as a scaffold in antigen receptor signaling to NF-κB, JNK, and mTOR. Antigen-induced CARD11 signaling proceeds through an ordered series of steps, including Opening, Cofactor Recruitment, Enzyme Activation, and Complex Disassembly. Recently, several forms of Primary Immunodeficiency have been described that are due to germline CARD11 mutations. One of these syndromes, Combined Immunodeficiency (CID) with atopy, is inherited in an autosomal dominant manner and caused by loss-of-function (LOF) CARD11 alleles, which display dominant negative activity that poisons signaling from co-expressed wild-type CARD11. To investigate the mechanistic basis for disease-associated dominant negative activity we reconstituted CARD11-deficient T cells with a panel of LOF CARD11 variants in the absence and presence of co-expressed wild-type CARD11. In the absence of wild-type CARD11, the LOF variants were indistinguishable, such that each variant failed to promote the signal-induced 1) recruitment of the cofactors Bcl10, HOIP, and MALT1 to CARD11, 2) Bcl10 polyubiquitinylation, 3) MALT1 protease activation, 4) NF-κB activation, and 5) JNK2 activation. In the presence of co-expressed wild-type CARD11, the LOF variants surprisingly displayed a range of dominant negative activity from strong to intermediate to weak, indicating a specific potency of some mutations to disrupt signaling when heterozygous. Our results suggest that disease-associated dominant negative CARD11 mutations interfere with the Opening Step in the CARD11 signaling cycle and provide a mechanistic basis to explain why some LOF CARD11 mutations can cause disease when heterozygous while others do so when homozygous.

Abstract CARD11 is an oligomeric, multidomain scaffold that mediates lymphocyte activation. Upon antigen receptor engagement, CARD11 becomes active and signals through NF-κB, JNK, and mTOR to elicit the adaptive immune response. The CARD11 signaling cycle consists of at least four stages beginning with the Opening step, followed by Cofactor Recruitment, Enzyme Activation, and Complex Disassembly. Primary Immunodeficiency Diseases (PIDDs) have been attributed to both autosomal dominant and recessive CARD11 variants. Combined Immunodeficiency with atopy is caused by the inheritance of a single dominant negative CARD11 allele, but the mechanistic basis for the dominant negative effects is not well understood. In this study, we investigated the mechanistic basis for dominant negative CARD11 mutations in T cells by assessing the activity of loss-of-function variants in the presence and absence of co-expressed wild-type CARD11. In the absence of wild-type CARD11, the variants perturbed the Opening step and downstream signaling steps, including recruitment of Bcl10, HOIP, and MALT1 to CARD11, Bcl10 polyubiquitinylation, MALT1 protease activity, NF-κB and JNK2 activation, and IL-2 production. When the variants were co-expressed with wild-type CARD11, a range of dominant negative effects on NF-κB activation was observed, which correlated with the ability of the CARD11 oligomer to open, recruit cofactors, and mediate IL-2 production. We conclude that dominant negative mutations interfere with the CARD11 Opening step and provide insights into the manifestation of PIDDs caused by the inheritance of dominant and recessive CARD11 alleles.

Pathogenic CARD11 mutations cause aberrant nuclear factor κB (NF-κB) activation, which is presumably responsible for multiple immunological disorders. However, whether there is an NF-κB–independent regulatory mechanism contributing to CARD11 mutations related to pathogenesis remains undefined. Using three distinct genetic mouse models, the Card11 knockout (KO) mouse model mimicking primary immunodeficiency, the CARD11 E134G point mutation mouse model representing BENTA (B cell expansion with NF-κB and T cell anergy) disease, and the mouse model bearing oncogenic K215M mutation, we show that CARD11 has a noncanonical function as a negative regulator of the AKT-FOXO1 signal axis, independent of NF-κB activation. Although BENTA disease–related E134G mutant elevates NF-κB activation, we find that E134G mutant mice phenotypically copy Card11 KO mice, in which NF-κB activation is disrupted. Mechanistically, the E134G mutant causes exacerbated AKT activation and reduced FOXO1 protein in B cells similar to that in Card11 KO cells. Moreover, the oncogenic CARD11 mutant K215M reinforces the importance of the noncanonical function of CARD11. In contrast to the E134G mutant, K215M shows a stronger inhibitory effect on AKT activation and more stabilized FOXO1. Likewise, E134G and K215M mutants have converse impacts on B cell development and differentiation. Our results demonstrate that, besides NF-κB, CARD11 also governs the AKT/FOXO1 signaling pathway in B cells. The critical role of CARD11 is further revealed by the effects of pathogenic CARD11 mutants on this noncanonical regulatory function on the AKT-FOXO1 signaling axis.

The work presents the aproach of the finite element investigation of the cardan transmission joint. We treat the problem of structural behaviour, such as analyses that target the strength-related behaviour. It is thus sought to determine certain parameters (nodal displacements, stresses, strains) in the conditions of applying various types of loads - forces, pressures or moments. For this, we used a software for finite element analysis – Cosmos - a product integrated in the Solidworks package. We insisted on calculation of the cardan transmission for Dacia 1307 but they can be easily transposed also to other typodimensions of cars, machines or medical devices from the same category.

Abstract The activated B-cell-like (ABC) molecular subtype of diffuse large B cell lymphoma (DLBCL) is characterized by constitutive activation of the nuclear factor-kB (NF-kB) pathway, which it requires for survival. By contrast, the NF-kB pathway is infrequently activated in the germinal center B cell-like (GCB) subtype of DLBCL. A crucial protein involved in activation of the NF-kB pathway by antigen receptor stimulation in normal B and T lymphocytes is CARD11. A previous loss-of-function genetic screen using RNA interference identified the signaling pathway from CARD11 to IkB kinase b as essential for the constitutive NF-kB activity in ABC DLBCL. However, the oncogenic mechanisms underlying the activity of CARD11 in this lymphoma subtype have yet to be elucidated. To this end, we resequenced some or all of the CARD11 exons in 157 primary DLBCL patient samples and cell lines and, as a control, in 19 MALT lymphoma patient samples. Various missense mutations were discovered in 10.4% of ABC DLBCL samples (8/77), and all mutations were located in exons encoding the coiled-coil domain of CARD11. In contrast, CARD11 coiled-coil domain mutations were detected in only 3.8% of the GCB DLBCL patient samples (3/80) and in no MALT lymphoma samples. ABC and GCB DLBCLs with CARD11 mutations were characterized by high expression of an NF-kB gene expression signature. Experimental introduction of each CARD11 mutant form in lymphoma cell lines altered their NF-kB signaling properties. Six of the CARD11 mutants caused strong constitutive NF-kB activation in the absence of any exogenous stimulus, a phenotype that was not elicited by wild type CARD11. For other CARD11 mutants, the enhancement of NF-kB signaling was most apparent upon antigen receptor stimulation. Fluorescence microscopy of cells bearing GFP-tagged CARD11 mutants revealed one or more prominent cytosolic aggregates in most cells whereas GFP-tagged wild type CARD11 was distributed diffusely in the cytoplasm. Multispectral imaging flow cytometry (Imagestream, Amnis) was used to quantitate the degree of aggregate formation by the CARD11 mutants. This analysis revealed a positive correlation between aggregate formation and NF-kB pathway activation. Immunofluorescent staining revealed that endogenous IkB kinase proteins were co-localized with the CARD11 mutants in the cytosolic aggregates, supporting the hypothesis that aggregate formation plays a central role in the activation of NF-kB by the CARD11 mutants. These results demonstrate that CARD11 is a bone fide oncogene in DLBCL. Mutations in the CARD11 coiled-coil domain caused spontaneous multimerization of the protein leading to IkB kinase recruitment and NF-kB signaling, thereby preventing cell death. These findings provide a strong genetic rationale for the development of CARD11 pathway inhibitors for the therapy of DLBCL.

RESUMO O objetivo do presente estudo foi analisar a dissimilaridade fenotípica de amostras populacionais de três linhagens de éguas Crioulas, uruguaia (La Invernada), argentina (Cardal) e chilena, para caracteres morfológicos, apontando as características de maior variância intra e entre as diferentes linhagens que são passíveis de seleção. Foram avaliadas 22 características morfológicas de 113 éguas da linhagem uruguaia “La Invernada”, 38 argentinas “Cardal” e 73 chilenas puras, totalizando 224 éguas de cria pertencentes à raça Crioula. Para determinação das variáveis lineares e de perímetro, foi utilizada fita métrica, prumo, paquímetro e hipômetro. Verificou-se variabilidade nas características morfológicas entre as três linhagens (P<0,05), incluindo as características lineares altura, perímetro de tórax e perímetro de canela (P<0,01), consideradas de caráter impositivo para fins de registro. Maior variação fenotípica foi observada nas éguas da linhagem chilena em relação aos outros dois grupos genéticos (P<0,01). As características morfológicas altura garupa, perímetro rostral pescoço, comprimento metatarso, largura garupa, profundidade do tórax, comprimento dorsal pescoço e comprimento do corpo foram passíveis de seleção entre as éguas Crioulas pertencentes às linhagens argentina (Cardal), uruguaia (La Invernada) e chilena. Em conclusão, não há homogeneidade entre as linhagens estudadas, o que identifica a diversidade entre as linhagens de éguas Crioulas estudadas.

CARD11 is a crucial scaffold protein that controls antigen-induced activation of lymphocytes. Upon antigen receptor signaling, CARD11 engages several signaling pathways, leading to the activation of NF-κB, mTOR, and JNK. CARD11 mutations are frequently found in patients with non-Hodgkin lymphoma and their ability to induce aberrant lymphocyte proliferation may be enhanced by mutations in regulators of CARD11 signal transduction. Here we describe how dysregulated CARD11 activity can promote lymphomagenesis through branched signaling pathways whose components and intermediates provide targets for novel diagnostic and therapeutic approaches. Statement of novelty: This review discusses how gain-of-function CARD11 mutations promote lymphomagenesis by engaging branching signaling pathways and how these different pathways provide multiple targets for therapies.

Abstract We recently described BENTA (B cell Expansion with NF-kB and T cell Anergy), a novel lymphoproliferative disorder caused by germline, gain-of-function mutations in the lymphocyte-specific scaffolding protein CARD11. Normally, antigen receptor (AgR) stimulation induces CARD11 phosphorylation to evoke an active conformation that nucleates a signalosome incorporating BCL10, MALT1, and other proteins required for IkB kinase (IKK) activation and NF-κB nuclear translocation. However, ectopic expression of CARD11 mutants triggers constitutive NF-kB activity in B and T cell lines without AgR stimulation. Here we show that these CARD11 mutants form large, cytosolic protein aggregates with a novel substructure, dubbed mCADS (Mutant CARD11 Dependent Shells). Endogenous mCADS are also detected in B cell lymphomas harboring similar CARD11 mutations. In contrast, WT CARD11 cannot form mCADS -/+ AgR stimulation. Only MALT1 and phospho-IKK are stably colocalized with mCADS, indicative of active, sustained signaling. Surprisingly, mCADS formation requires interaction with BCL10 but not NEMO (IKKg). Our results suggest that gain-of-function CARD11 mutants spontaneously assemble into an active, highly-ordered signalosome that is structurally and functionally distinct from WT CARD11, regardless of AgR ligation. The unique structure and composition of mCADS may represent a novel therapeutic target for treating BENTA or lymphoma without disrupting normal AgR-induced NF-kB activation.

We investigated the role of miR-181a in diffuse large B-cell lymphoma (DLBCL) and its potential target genes. miR-181a levels were lower in activated B-cell- (ABC-) like DLBCL cells than that in germinal center B-cell- (GCB-) like DLBCL cells. Overexpression of miR-181a in ABC-like DLBCL cell lines (OCI-LY10 and U2932) resulted in G0/G1 cell cycle arrest, increased apoptosis, and decreased invasiveness. miRNA target prediction programs (miRanda, TargetScan, and miRDB) identified caspase recruitment domain-containing protein 11 (CARD11) as a putative miR-181a target. CARD11 mRNA and protein levels were higher in the ABC-like DLBCL than that in GCB-like DLBCL. Moreover, CARD11 mRNA and protein levels were downregulated in the OCI-LY10 and U2932 cell lines overexpressing miR-181a. Dual luciferase reporter assays confirmed the miR-181a binding site in the CARD11 3′UTR region. OCI-LY10 and U2932 cells transfected with a CARD11 expression vector encoding miR-181a with a mutated binding site showed higher CARD11 protein levels, cell viability, G2/M phase cells, and invasiveness compared to those transfected with a wild-type CARD11 expression vector. Nude mice xenografted with OCI-LY10 cells with overexpressed wild-type miR-181a generated smaller tumors compared to those with overexpressed mutated binding site of CARD11 3′UTR and miR-181a. These results indicate that miR-181a inhibits ABC-like DLBCL by repressing CARD11.

Abstract: A rear-wheel-drive vehicle's Cardan shaft, a rotating shaft that transfers power from the engine to the differential gear, must work via continually shifting angles between the gearbox and axle. Due to the constant rotation, there is significant vibration on it. Due to this issue, the shaft frequently bends or deforms. Vibration in the Cardan shaft is a significant issue. In the present work, three distinct geometries—circular cut, rectangular cut, and no-cut—were modelled using SolidWorks software. Modal analysis was then carried out to obtain the deformation and natural frequency of all the CAD models for comparison. In the current investigation, the rectangular cut model, which produces the least deformation and natural frequency, was designated as the superior model.

Abstract The present study aims to measure vibrations in machine parts, perform the corresponding spectral analysis and, finally, obtain a diagnosis. The test was performed on a cardan shaft, measuring the vibrations with a uni-axial accelerometer, and follow-up, with the use of the Adash acquisition board, data were recorded and analyzed. It is known that each mechanical part has its own vibration frequency and that the operation at critical speed determines the occurrence of the resonance phenomenon. When this phenomenon appears, damage occurs, which is difficult to remedy and in some cases could endanger the safety of operators. Therefore, it was necessary to perform modal analysis in the Ansys program in order to be able to analyze the own vibration modes of the cardan shaft.

Transmission with cardan joints and non­linear gearbox elements generates vibrations caused by spinning motion of transmission. This affects the pressure and productivity of centrifugal pump. The momentum of centrifugal pump in turn affects dynamic characteristics of transmission. In order to find the effect of transmission elements on centrifugal pump we investigate transmission composed of asynchronous electrical engine, gear box, cardan joint and Ziegler Ultra centrifugal pump. We create mathematical models of transmission and centrifugal pumps and investigate how the dynamic effects of transmission influence productivity centrifugal pump. This model can be applied to study the transmission parameters, effect of intake and outflow parameters on productivity of centrifugal pump and presence of cavitation. The paper includes results and conclusions of mathematical model.

This research is motivated by the existence of a vocal instructor, who does not have a musical education background, at Purwa Caraka Music Studio Padang. However, he is able to bring his students to get achievement at national level. This study aims to determine and describe the implementation of vocal training at Purwa Caraka Music Studio Padang. The aspects explored in this study are: 1) The program of vocal training at Purwa Caraka Music Studio Padang, 2) The implementation of vocal training at Purwa Caraka Music Studio Padang, 3) Obstacles to the implementation of vocal training at Purwa Caraka Music Studio Padang.This research useda descriptive analytic method. The data were collected through observation, interviews, documentation, and literature study. The results show that the implementation of vocal training at Purwa Caraka Music Studio Padang is based on the curriculum and handbooks. The methods used in vocal training are lectures, individual approachesand exercises, as well as voice production exercises which include: posture and diaphragmatic breathing, articulation techniques, intonation techniques, resonance techniques, and phrasering techniques. The obstacles in the implementation of the vocal training at Purwa Caraka Music Studio Padang are: 1) Mood of the students; 2) tone and tempo blind of the students.Keywords: Implementation, BinaVokalia, Purwa Caraka

Background/Aims: Spinal cord glioma is a highly aggressive malignancy that commonly results in high mortality due to metastasis, high recurrence and limited treatment regimens. This study aims to elucidate the effects of long non-coding RNA LINC01260 (LINC01260) on the proliferation, migration and invasion of spinal cord glioma cells by targeting Caspase recruitment domain family, member 11 (CARD11) via nuclear factor kappa B (NF-κB) signaling. Methods: The Multi Experiment Matrix (MEM) website was used for target gene prediction, and the DAVID database was used for analysis of the relationship between CARD11 and the NF-κB pathway. In total, 60 cases of glioma tissues and adjacent normal tissues were collected. Human U251 glioma cells were grouped into blank, negative control (NC), LINC01260 vector, CARD11 vector, siRNA-LINC01260, siRNA-CARD11, LINC01260 vector + CARD11 vector and LINC01260 + siRNA-CARD11 groups. A dual-luciferase reporter assay was conducted to verify the target relationship between LINC01260 and CARD11. Reverse transcription quantitative polymerase chain reaction (RT-qPCR) and western blot analysis were employed to assess expression of LINC01260, E-cadherin, p53, CARD11, Ki67, N-cadherin, matrix metalloproteinase (MMP)-9, NF-κBp65 and NF-κBp50. MTT, flow cytometry, wound-healing and Transwell assays were performed to examine cell viability, the cell cycle, apoptosis, invasion and migration. Tumor growth was assessed through xenografts in nude mice. Results: CARD11 was confirmed to be a target gene of LINC01260 and was found to be involved in regulating the NF-κB pathway. Compared with adjacent normal tissues, glioma tissues showed reduced expression of LINC01260 and elevated expression of CARD11 and genes related to apoptosis, invasion and migration; activation of NF-κB signaling was also observed. In contrast to the blank and NC groups, an elevated number of cells arrested in G1 phase, increased apoptosis and reduced cell proliferation, invasion and number of cells arrested in S and G2 phases, as well as tumor growth were found for the LINC01260 vector and siRNA-CARD11 groups. Conclusions: Our findings demonstrate that overexpression of LINC01260 inhibits spinal cord glioma cell proliferation, migration and invasion by targeting CARD11 via NF-κB signaling suppression.

The concepts of kinetic energy and complementary kinetic energy permit to distinguish between two different formulations of what happens to be the same quantity in Newtonian mechanics. These formulations turn out to play a significant role in gyrodynamics in that they can be used very effectively to establish fundamental equations. It is relatively straightforward to establish a complementary kinetic energy expression. The establishment of the kinetic energy expression is, in spite of its unambiguous definition, more complicated, and a way is presented how to obtain it without too much difficulty. Equations are presented for the more common angle systems, i.e., for Euler angles of the first kind, Cardan angles of the first kind, both because of their fundamental importance, and Cardan angles of the fifth kind because of its prevalence in aeronautics.

We have found that psoriasis-associated CARD14 mutations release the CARD14 protein from autoinhibition, inducing constitutive formation of CARD14-containing signalling complexes. These activate NF-κB and MAP kinase signalling pathways, which drive the pathogenic expression of pro-inflammatory genes in skin cells.
