Abstract In this paper, we present a conceptual design of an on-chip solder stack to connect silicon devices faster and more reliable. Almost all electronic devices rely on solder layers to provide electrical, mechanical, and thermal connections between components. We improve the solder connection with industry-standard solder parameters of 300°C and some minutes of soldering time. An ideal solder connection is composed of intermetallic phases (IMPs) at the interfaces between device and solder, and substrate and solder. Typically, a thin region of Sn-based solder remains between the two IMP layers at the interfaces. IMPs of copper (Cu) and tin (Sn) are Cu6Sn5 and Cu3Sn. The formation of IMPs is decisive for a good mechanical connection because of their high melting point and mechanical stability. To achieve these requirements, we implement the solder stack as a transient liquid phase bonding (TLPB) system. To realize durable interconnections, we use the diffusion of a high-melting first component in a second component, which is liquid at solder process temperature. Ongoing diffusion leads to the formation of IMPs with a melting point above process temperature, resulting in a solidification of the connection at constant temperature. By this isothermal solidification, the solder connection becomes more durable against mechanical and thermal load and is usable at temperatures exceeding 300°C.

Purpose This study aims to reveal more information and understanding on performance and failure mechanisms of high strength steel endplate connections after fire. Design/methodology/approach An experimental and numerical study on seven endplate connections after cooling down from fire temperature of 550°C has been carried out and reported herein. Moreover, the provisions of European design standard for steel structures, Eurocode 3, were validated with test results of high strength steel endplate connections. Findings In endplate connections, a proper design using a thinner high strength steel endplate can achieve the same failure mode, similar residual load bearing capacity and comparable or even higher rotation capacity after cooling down from fire. It is found that high strength steel endplate connection can regain more than 90 per cent of its original load bearing capacity after cooling down from fire temperature of 550°C. Originality/value The post-fire performance of high strength steel endplate connection has been reported. The accuracy of Eurocode 3 for endplate connections is validated against test results.

New detections of CCH have been made toward nine planetary nebulae (PNe), including K4-47, K3-58, K3-17, M3-28, and M4-14. Measurements of the N = 1 -> 0 and N = 3 -> 2 transitions of this radical near 87 and 262 GHz were carried out using the new 12 m and the Sub-Millimeter Telescope (SMT) of the Arizona Radio Observatory (ARO). The presence of fine and/or hyperfine structure in the spectra aided in the identification. CCH was not observed in two PNe which are sources of C-60. The planetary nebulae with positive detections represent a wide range of ages and morphologies, and all had previously been observed in HCN and HNC. Column densities for CCH in the PNe, determined from radiative transfer modeling, were N-tot(CCH) similar to 0.2-3.3 x 10(15) cm(-2), corresponding to fractional abundances with respect to H-2 of f similar to 0.2-47 x 10(-7). The abundance of CCH was found to not vary significantly with kinematic age across a time span of similar to 10,000 years, in contrast to predictions of chemical models. CCH appears to be a fairly common constituent of PNe that are carbon-rich, and its distribution may anti-correlate with that of C-60. These results suggest that CCH may be a product of C-60 photodestruction, which is known to create C-2 units. The molecule may subsequently survive the PN stage and populate diffuse clouds. The distinct, double-horned line profiles for CCH observed in K3-45 and M3-28 indicate the possible presence of a bipolar flow oriented at least partially toward the line of sight.

Durante o tempo em serviço, estruturas e componentes de equipamentos podem falhar e causar acidentes. Isto geralmente conduz a danos materiais podendo, ainda, causar perda de vidas humanas e danos ambientais. Frequentemente, os eventos danosos são originários de falhas de materiais e de fabricação, sobrecargas não previstas ou outros problemas oriundos do tempo de operação ou deficiência de manutenção de um componente. Neste estudo, com base numa análise de propagação de trinca superficial submetida a carregamento, se avaliou a integridade de um flange da conexão de topo de uma linha flexível, sob condições criogênicas de operação. Quando em serviço, a falha deste conector pode ocasionar interrupções na produção, dano a equipamentos e meio ambiente, bem como fatalidades em plataformas offshore de petróleo. As linhas (dutos) flexíveis utilizadas para injetar gás CO2 nos reservatórios de petróleo no país trabalham submetidas a elevadas pressões internas (da ordem de 10.000 psi), o que aumenta o risco de falha do componente. Além disto, a selagem da conexão flangeada pode falhar e provocar um vazamento de fluido gasoso, proporcionando uma queda acentuada da temperatura local e potencializando a fratura frágil do componente. O cenário proposto para o estudo foi relacionado a possível falha na vedação do anel BX, localizado entre a conexão da linha flexível e a plataforma de petróleo. A metodologia foi dividida em duas etapas: a primeira corresponde à análise global linear elástica do conector na ausência de trincas e baseada no método de elementos finitos (MEF). Na segunda etapa da análise, se introduziu uma trinca no conector na região com os maiores níveis de tensão e onde é possível realizar ensaio por líquido penetrante. Assim, o estudo se concentrou na conexão flangeada do conector, adotando como um aço com 9 por cento de níquel em sua composição química, liga ferrosa de larga aplicação em tanques de armazenagem de GNL. Se buscou avaliar a resistência à fratura do material através da associação de condições severas de carregamento, baixas temperaturas e tamanho de trinca em flanges de dutos flexíveis, conforme critério severizado da norma API 6A. Desta maneira, se espera contribuir para que a indústria offshore de petróleo tenha mais uma opção de material para ser adotado na fabricação de conectores de topo das linhas flexíveis que trabalhem em condição criogênica.
During the time in service, structures and equipment components can fail and cause accidents. This usually leads to damage and may also cause loss of human lives and environmental damages. Often, harmful events are originated from materials and manufacturing faults, unforeseen overloads or other problems arising from the operation time or component maintenance deficiencies. In this study, based on the analysis of surface crack propagation under loading, was assessed the integrity of a top connecting flange of a flexible line operation under cryogenic conditions. When in service, the failure of this connector may cause production disruptions, damages to the equipment and the environment, as well as human fatalities on offshore oil platforms. In Brazil, flexibles lines (ducts) used to inject CO2 gas in oil reservoirs work subjected to high internal pressures (of the order of 10,000 psi), which increases the risk of a component failure. Moreover, the damaged flange may cause leakage of gaseous fluid resulting in very low local temperatures and resulting in brittle fracture enhancing of the component. The scenario proposed in this study is related to a possible seal BX ring failure which is located between the flexible line connection and the oil platform. The methodology was divided into two stages: the first corresponds to the linear elastic global analysis of the connector in the absence of cracks and based on the finite element method (FEM). In the second stage of analysis, it introduced a crack on the connector in the region with the highest stress levels and where it is possible to test for penetrant. Thus, the study focused on the flange connection connector, adopting as a steel with 9 per cent nickel in its chemical composition, ferrous alloy wide application in LNG storage tanks. He sought to evaluate the fracture resistance of the material through the combination of severe loading conditions, low temperatures and 9 cracks in flanges of flexible ducts as severe criteria standard API 6A standard. Thus, it is expected to contribute to the offshore oil industry has one more material option to be adopted in the manufacture of top connectors of the flexible lines working in cryogenic condition.

We introduce a special set of relations on a finite set, called clausal relations. A restricted version of the Galois connection between polymorphisms and invariants, called Pol-CInv, is studied, where the invariant relations are clausal relations. Clones arising from this Galois connection, so-called C-clones, are investigated. Finally, we show that clausal relations meet a sufficient condition that is known to ensure polynomial time solvability of the corresponding CSP.

Among the available all-quadrilateral mesh generation algorithms progressive-front offers the best quality and robustness. The paving algorithm is a progressive-front technique that generates a two dimensional mesh of all quadrilaterals on planar surfaces. The technique is particularly suited to meshing irregular boundaries with interior holes. This thesis presents an extension of the paving algorithm to generalized three dimensional surfaces. The major problems in three-dimensional surface mesh generation are projection of nodes, calculation of interior angles, and connection of mesh fronts. The solutions to these problems are presented. This work is part of the ongoing CUBIT project at Sandia Labs. The application is written in object oriented style in C++. A description of object oriented programming is given, with the relative merits and drawbacks for engineering applications.

This MSc Thesis is focused on the multimodeling area, especially on the cooperation of the C++ language and the Prolog language. The recent research is established on my semester study, which also dealt with the multimodeling area. During this research a prototype of interconnection library for cooperation of above mentioned programming languages was developed. This prototype of the library was finished within the scope of this thesis and some case-studies were created, too, using also another simulation library - SIMLIB/C++. These case-studies have their focus in the problems of artificial intelligence. The main benefit of this thesis is the confrontation of methods with and without using artificial intelligence.

Traditionally, connection management applications (referred to as patchbays) for high-speed audio networking, are predominantly developed using third-generation languages such as C, C# and C++. Due to the rapid increase in distributed audio/video network usage in the world today, connection management applications that control signal routing over these networks have also evolved in complexity to accommodate more functionality. As the result, high-speed audio networking application developers require a tool that will enable them to develop complex connection management applications easily and within the shortest possible time. In addition, this tool should provide them with the reliability and flexibility required to develop applications controlling signal routing in networks carrying real-time data. High-speed audio networks are used for various purposes that include audio/video production and broadcasting. This investigation evaluates the possibility of using Adobe Flash Professional 8, using ActionScript 2.0, for developing connection management applications. Three patchbays, namely the Broadcast patchbay, the Project studio patchbay, and the Hospitality/Convention Centre patchbay were developed and tested for connection management in three sound installation networks, namely the Broadcast network, the Project studio network, and the Hospitality/Convention Centre network. Findings indicate that complex connection management applications can effectively be implemented using the Adobe Flash IDE and ActionScript 2.0.

The demand for computers is oriented toward faster computers and newer computers are being built with more than one CPU. These computers require sophisticated software to program them. One such approach to program the multiple CPU machines is through the use of object-oriented programming techniques. An example of such an approach is the use of C* on the Connection Machine. Though C* supports many of the object-oriented concepts, it does not support the concept of software reuse through inheritance. This thesis introduces a new language called C*±+ , an extension of C* language to support inheritance. We also discuss the issues invloved in the implementation of multiple inheritance in programming languages. This thesis describes the differences between C** and C* . It also discusses the various issues involved in the design and implementation of the translator from C** to C* . It also illustrates the advantages of programming in C*++ through an example. Since C*++ is designed to support software reuse which allows the users to create quality software in shorter time, it is anticipated that C*+ will have widespread use in programming the Connection Machine.

The collection of books assembled for Leofric, bishop of Exeter, during the twenty-two years of his episcopacy has been the subject of sustained scholarly attention, which has just started to uncover the importance that scriptorial materials have in illuminating the life and deeds of the bishop. Due to the lack of a hagiography dedicated to him, Leofric's importance in the eleventh century church has long remained obscure. The following thesis endeavours to shed new light on the activities that underpinned Leofric's episcopacy through an investigation into the books compiled at the bishop's behest. In particular, four homiliaries are here under scrutiny from a perspective that is both physical and textual: these manuscripts contain a selection of texts that Leofric requested specifically for performing his pastoral remit and that, for this reason, represent the bishop's interests. The analysis of some of these texts demonstrates that, when studied in their manuscript context, homilies yield crucial information on the way in which preaching materials were used and on the audiences to whom they were directed in times subsequent to their composition. The way in which Leofric exploited homiletic works dating to the late tenth century was innovative and original in so far as it reflected the administration policies that he adopted for his diocese and the reforms that he activated in restructuring a decaying episcopal see. His pastoral achievements were not only important in the context of the eleventh-century English episcopate but also in a broader, continental perspective and were attained as part of a reforming programme that would later culminate in the Gregorian Reform. Leofric's Lotharingian education triggered the activation of these reforms at Exeter at a time when Lotharingian prelates held prominent positions in the western Christendom.

AbstractIn this chapter, we consider the theoretical foundations for representing knowledge in the Internet of Things context. Specifically, we consider (1) the model-theoretic semantics (i.e., extensional semantics), (2) the possible-world semantics (i.e., intensional semantics), (3) the situation semantics, and (4) the cognitive/distributional semantics. Given the peculiarities of the Internet of Things, we pay particular attention to (a) perception (i.e., how to establish a connection to the world), (b) intersubjectivity (i.e., how to align world representations), and (c) the dynamics of world knowledge (i.e., how to model events). We come to the conclusion that each of the semantic theories helps in modeling specific aspects, but does not sufficiently address all three aspects simultaneously.

AbstractAlthough sanitation is embedded in the socio-culture, the socio-cultural aspects of sanitation are not fully discussed. In this context, we focus on the socio-cultural aspects of sanitation, especially the connection between socio-culture with health (C), and with materials (B), as in the Sanitation Triangle model, from the perspective of humanities and social sciences in this part. To introduce this part, we first briefly review previous studies on sanitation by humanities and social sciences, and show the socio-cultural aspects of sanitation. Then, after providing an overview of this part, we discuss the theoretical points of each chapter such as “hard work” or “dirty work,” a social configuration of technology, and Menstrual Hygiene Management (MHM) in the local context, and indicate the relevant chapters in this book.

Abstract The introduction provides a rationale for this edited volume and presents its main topics: the emerging digital age and the development of electronic medical records (EMRs), the question of spirituality and documentation in a larger interprofessional context, as well as the sustainability of future spiritual care. In the second part, it gives an overview of the state of research on charting spiritual care. Five different but intertwined areas of research are defined: (a) evoking conceptual questions or fundamental debates like that of confidentiality and (b) highlighting the connection between spiritual assessment and documentation procedures, as well as (c) recent models and (d) actual practices of documentation. Lastly (e), we take a look on the integration of patients’ views and perspectives into documentation processes. We conclude this introduction with a short survey of the following chapters.

Flange rotation with internal pressure affects the sealing performance of pipe flange connections more with increasing the pipe nominal diameter. Therefore, it is hard to estimate the sealing performance of pipe flange connections with larger nominal diameter by using the test results of the sealing performance for pipe flange connections with small nominal diameter. In this study, the 20" diameter pipe flange connection was experimentally used as a large nominal diameter pipe flange connection and analyzed the stress in the connection. In the experiments, the sealing performance of the pipe flange connection was evaluated by measuring the amount of gas leakage at 50 °C and 100 °C under internal pressure. Non-asbestos graphite gaskets were used in the experiments. The bolts and nuts were tightened according to the Japanese method (HPIS Z103 TR). The gasket contact stress distributions of larger nominal diameter pipe flange connection were calculated by FEM under elevated temperature conditions as well as internal pressure application. The estimated results were compared with the experimental results. The results were also compared with the small nominal diameter test result and discussed.

Although a lot of pipe flange connections are exposed to elevated temperature during long-term plant operation, a sealing performance of the pipe flange connections at elevated temperature is not well understood because of the experimental difficulty and the analytical problems which result from the lack of the materials properties of gaskets at elevated temperature. The authors have been evaluating the effects of the material properties of spiral wound gaskets (SWG) and the sealing performance of the pipe flange connections at elevated temperature with numerical and experimental analysis. The gasket stress induced by the mismatch of the thermal expansion between the gasket and flange/bolt increased as increasing the temperature in the case that the thermal expansion coefficient of the gasket was larger than that of bolt/flange material and the sealing performance of the pipe flange connection was improved. However, the thermal expansion coefficient declines at temperatures higher than 210 °C. In addition, the binder compositions of SWG vaporize and the mechanical properties slightly change at high temperature such as higher than 200°C. Therefore, the sealing performance of pipe flange connection with SWG might decline more than 200°C. The sealing performance and long-term durability of pipe flange connection with SWG at elevated temperature were evaluated in this study. The long term leakage test with heat cycle revealed that the sealing performance of pipe flange connection didn’t show any degradation for 21 days. The compression tests for the new, the thermal aged and the used gaskets were done to investigate the effect of material characteristics on the long term sealing performance. In addition to the leakage test and the compression tests, the thermal characteristics of SWG were evaluated by the Thermogravimetry/Differential Thermal Analyzer (TG/DTA) measurement. The measurement showed the weight loss of SWG more than 130°C. According to those experimental results and FE analysis, the effect of temperature on the sealing performance was discussed.

It has been well known that a scatter in axial bolt forces of pipe flange connections tightened by the torque control method is substantial. It is necessary for evaluating the sealing performance of the pipe flange connections with the gaskets subjected to internal pressure to know the contact gasket stress distributions due to the scatter of the axial bolt forces in the connections tightened by the torque control method. Especially, when gases are used in the pipe flange connections with gaskets at a high temperature, it is necessary to evaluate the amount of gas leakage. In this study, when bolts and nuts in a pipe flange connection were tightened using a torque wrench, the axial bolt forces were measured and then the amount of gas leakage in the pipe flange connection at 50,100,150 and 200 °C was measured under internal pressure. Leakage tests were performed using Helium gases. In addition, the thermal stress distributions at the interfaces between pipe flanges and the gasket under heat condition (temperature 50,100,150 and 200 °C) and internal pressure were calculated by using the finite element method (FEM) taking into account hysteresis in the stress-strain curves of a spiral wound gasket. By using the calculated contact stress distributions and the results of the leakage tests, the sealing performance was evaluated.

In this study, the sealing performance of pipe flange connection was experimentally evaluated by measuring the amount of gas leakage at room temperature (RT), 100 °C and 200 °C in addition to internal pressure. Non-asbestos graphite gaskets were used in the experiment. The nominal size of the pipe flange used during the experimental study was 3 inch. The eight bolts and nuts were tightened according to ASME PCC-1 and the Japanese method (HPIS). During assembly, the axial bolt force was individually measured with strain gauges attached to each bolt. The scatter in the axial bolt force during the assembling process was examined and compared between the ASME and the HPIS. In addition, the gasket contact stress distribution of each assembly process was calculated by finite element method under elevated temperature and internal pressure. In the FE study, experimentally measured physical properties such as elastic modulus and thermal expansion were used. ηhe amount of leakage was estimated from the contact gasket stress distribution obtained by FE analysis. The estimated axial bolt forces were compared with those of experimental ones.

Abstract In a global context aiming to unlock a low carbon future by industry decarbonization, developing the infrastructure for capturing and storing CO2 emissions is a key target of countries, energy companies and regulatory bodies. Injection for geological storage in suitable reservoirs is an advantageous option which presents challenges related to the completion accessories and string exposed to the injected fluid and the thermodynamical loads during injection and the well life. The purpose of this work is to simulate by numerical analysis and full-scale test, the behavior of a gas-tight Metal-to-Metal OCTG premium dope-free connection when subjected to low temperatures and loads generated by the effect of a sudden CO2 high pressure drop during injection in depleted reservoirs. Extreme temperature drop down caused by the Joule-Thompson (J-T) effect between injection conditions (P-T) inside the tubular and those in the annulus, may expose tubing connections to a thermal shock reaching a temperature near the theoretical figure of -78.5°C. This temperature drop assumed as worst-case scenario is also explored. The analysis is performed considering estimated loads for a CO2 injection case study. The numerical analysis and full-scale test performed confirm the structural and sealability performance of the connection is not affected by the exposure to such low temperatures. Additionally, transient thermal loads, with a drop of approximately 100°C, appears to be not critical for the metal-to-metal dope-free seal integrity and also not affecting the structural integrity of the connection. The challenges setting up of a prototype testing frame, simulating the cooling by thermal shock, lead to a methodology for assessing CCS projects premium connection able to define a robust testing protocol for cryogenic temperatures. The numerical and full-scale results collected on the tested connection size, together with the ones previously tested, allow extrapolation to near sizes of the same premium thread family. The results achieved by testing a premium connection which has been subjected to a thermal shock approaching -78.5°C represent a forefront in the industry, demonstrating the reliability of the product not only in operative conditions during CO2 injection, but also after an extreme event, assessing performance for the CCUS storage projects.

Abstract It is important to complete thermal and high-pressure/high-temperature (HPHT) wells with tubular connections that possess adequate structural integrity and sealing capacity under the severe load conditions typically experienced by these wells throughout their life cycle. Individual premium connection designs are required to be evaluated and qualified through physical tests to broadly adopted industry protocols, such as ISO/PAS 12835: 2013 for thermal wells that experience temperatures from 180°C to 350°C, and ISO 13679:2019 and API RP 5C5:2017 for HPHT wells which experience peak temperatures up to 180°C and pressures greater than 70 MPa. Recognizing the time and capital expenses associated with completing full-scale physical testing of product lines with multiple connection designs of different tubular diameter, weight, and grade, industry is developing a hybrid approach that supplements results from physical qualification tests with numerical/analytical simulation, such as Finite Element Analysis (FEA). The key challenges associated with analytical evaluation are the lack of evaluation criteria and suitable guidelines for analysis methodologies. This paper provides a review of recent work related to the development of sealability evaluation criteria; and presents guidelines to facilitate performance evaluation of tubular connections in thermal and HPHT wells through advanced FEA. For thermal well applications, this paper presents a methodology for quantitative evaluation of sealability of casing connections, as a supplement to the determination of a biased test population using FEA following ISO/PAS 12835:2013 requirement. For HPHT wells, this paper presents considerations for analyzing various testing loads, such as Test Series A (internal and external pressure cycles), Test Series B (internal pressure with bending), Test Series C (thermal and mechanical cycles), and Limit Load Cases. Analysis examples with generic premium connections are presented to demonstrate the use of the proposed analysis methodologies.

In current bridge design specifications and evaluation manuals from the American Association of State Highway and Transportation Officials (AASHTO LRFD) (AASHTO, 2018), the detail category for base metal at the toe of transverse stiffener-to-flange fillet welds and transverse stiffener-to-web fillet welds to the direction of the web and hence, the primary stress) is Category C′. In skewed bridges or various other applications, there is sometimes a need to place the stiffener or a connection plate at an angle that is not at 90 degrees to the web. As the plate is rotated away from being 90 degrees to the web, the effective “length” of the stiffener in the longitudinal direction increases. However, AASHTO is currently silent on how to address the possible effects on fatigue performance for other angles in between these two extremes. This report summarizes an FEA study that was conducted in order to investigate and determine the fatigue category for welded attachments that are placed at angles other than 0 or 90 degrees for various stiffener geometries and thicknesses. Recommendations on how to incorporate the results into the AASHTO LRFD Bridge Design Specifications are included in this report.

This report is one of a series of reports documenting the methods and findings of a multi-year, multi-disciplinary project coordinated by the Pacific Earthquake Engineering Research Center (PEER and funded by the California Earthquake Authority (CEA). The overall project is titled “Quantifying the Performance of Retrofit of Cripple Walls and Sill Anchorage in Single-Family Wood-Frame Buildings,” henceforth referred to as the “PEER–CEA Project.” The overall objective of the PEER–CEA Project is to provide scientifically based information (e.g., testing, analysis, and resulting loss models) that measure and assess the effectiveness of seismic retrofit to reduce the risk of damage and associated losses (repair costs) of wood-frame houses with cripple wall and sill anchorage deficiencies as well as retrofitted conditions that address those deficiencies. Tasks that support and inform the loss-modeling effort are: (1) collecting and summarizing existing information and results of previous research on the performance of wood-frame houses; (2) identifying construction features to characterize alternative variants of wood-frame houses; (3) characterizing earthquake hazard and ground motions at representative sites in California; (4) developing cyclic loading protocols and conducting laboratory tests of cripple wall panels, wood-frame wall subassemblies, and sill anchorages to measure and document their response (strength and stiffness) under cyclic loading; and (5) the computer modeling, simulations, and the development of loss models as informed by a workshop with claims adjustors. Quantifying the difference of seismic performance of un-retrofitted and retrofitted single-family wood-frame houses has become increasingly important in California due to the high seismicity of the state. Inadequate lateral bracing of cripple walls and inadequate sill bolting are the primary reasons for damage to residential homes, even in the event of moderate earthquakes. Physical testing tasks were conducted by Working Group 4 (WG4), with testing carried out at the University of California San Diego (UCSD) and University of California Berkeley (UCB). The primary objectives of the testing were as follows: (1) development of descriptions of load-deflection behavior of components and connections for use by Working Group 5 in development of numerical modeling; and (2) collection of descriptions of damage at varying levels of peak transient drift for use by Working Group 6 in development of fragility functions. Both UCSD and UCB testing included companion specimens tested with and without retrofit. This report documents the portions of the WG4 testing conducted at UCB: two large-component cripple wall tests (Tests AL-1 and AL-2), one test of cripple wall load-path connections (Test B-1), and two tests of dwelling superstructure construction (Tests C-1 and C-2). Included in this report are details of specimen design and construction, instrumentation, loading protocols, test data, testing observations, discussion, and conclusions.

Our project focused on genes for high affinity iron acquisition in Cochliobolus heterostrophus, a necrotrophic pathogen of maize, and their intertwined relationship to oxidative stress status and virulence of the fungus on the host. An intriguing question was why mutants lacking the nonribosomal peptide synthetase (NRPS) gene (NPS6) responsible for synthesis of the extracellular siderophore, coprogen, are sensitive to oxidative stress. Our overall objective was to understand the mechanistic connection between iron stress and oxidative stress as related to virulence of a plant pathogen to its host. The first objective was to examine the interface where small molecule peptide and reactive oxygen species (ROS) mechanisms overlap. The second objective was to determine if the molecular explanation for common function is common signal transduction pathways. These pathways, built around sensor kinases, response regulators, and transcription factors may link sequestering of iron, production of antioxidants, resistance to oxidative stress, and virulence. We tested these hypotheses by genetic manipulation of the pathogen, virulence assays on the host plant, and by following the expression of key fungal genes. An addition to the original program, made in the first year, was to develop, for fungi, a genetically encoded indicator of redox state based on the commercially available Gfp-based probe pHyper, designed for animal cell biology. We implemented several tools including a genetically encoded indicator of redox state, a procedure to grow iron-depleted plants, and constructed a number of new mutants in regulatory genes. Lack of the major Fe acquisition pathways results in an almost completely avirulent phenotype, showing how critical Fe acquisition is for the pathogen to cause disease. Mutants in conserved signaling pathways have normal ability to regulate NPS6 in response to Fe levels, as do mutants in Lae1 and Vel1, two master regulators of gene expression. Vel1 mutants are sensitive to oxidative stress, and the reason may be underexpression of a catalase gene. In nps6 mutants, CAT3 is also underexpressed, perhaps explaining the sensitivity to oxidative stress. We constructed a deletion mutant for the Fe sensor-regulator SreA and found that it is required for down regulation of NPS6 under Fe-replete conditions. Lack of SreA, though, did not make the fungus over-sensitive to ROS, though the mutant had a slow growth rate. This suggests that overproduction of siderophore under Fe-replete conditions is not very damaging. On the other hand, increasing Fe levels protected nps6 mutants from inhibition by ROS, implying that Fe-catalyzed Fenton reactions are not the main factor in its sensitivity to ROS. We have made some progress in understanding why siderophore mutants are sensitive to oxidative stress, and in doing so, defined some novel regulatory relationships. Catalase genes, which are not directly related to siderophore biosynthesis, are underexpressed in nps6 mutants, suggesting that the siderophore product (with or without bound Fe) may act as a signal. Siderophores, therefore, could be a target for intervention in the field, either by supplying an incorrect signal or blocking a signal normally provided during infection. We already know that nps6 mutants cause smaller lesions and have difficulty establishing invasive growth in the host. Lae1 and Vel1 are the first factors shown to regulate both super virulence conferred by T-toxin, and basic pathogenicity, due to unknown factors. The mutants are also altered in oxidative stress responses, key to success in the infection court, asexual and sexual development, essential for fungal dissemination in the field, aerial hyphal growth, and pigment biosynthesis, essential for survival in the field. Mutants in genes encoding NADPH oxidase (Nox) are compromised in development and virulence. Indeed the triple mutant, which should lack all Nox activity, was nearly avirulent. Again, gene expression experiments provided us with initial evidence that superoxide produced by the fungus may be most important as a signal. Blocking oxidant production by the pathogen may be a way to protect the plant host, in interactions with necrotrophs such as C. heterostrophus which seem to thrive in an oxidant environment.

Female genital mutilation/cutting (FGM/C) threatens the health and well-being of girls in a variety of ways. Some experts have suggested that human immunodeficiency virus (HIV) may be among the practice’s adverse health outcomes. They theorize that because women with FGM/C risk genital trauma and bleeding during intercourse, their chances of acquiring HIV from an infected male partner are increased. While evidence that FGM/C increases susceptibility to HIV could bolster FGM/C eradication efforts, this connection has received limited research attention. This brief highlights the Evidence to End FGM/C program’s findings on the current state of evidence on the association between FGM/C and HIV, including the quality of the existing research and recommendations for further investigation.

Policymakers and advocates often cite fistula—a hole between the vagina and rectum or bladder that leaves a woman leaking urine or excrement or both—among the many adverse health conditions thought to be related to female genital mutilation/cutting (FGM/C). But high-quality research supporting a connection between the two conditions is limited. Health workers, program managers, and health advocates need a clear understanding of the evidence linking FGM/C and fistula and more accurate information on the dynamics between them. To fill this gap, the Evidence to End FGM/C program examined the association between FGM/C and fistula. This brief highlights the program’s findings on the relationship between the two conditions, focusing on what we know and what we still need to investigate.
