Résumé :
Serine phosphorylation of insulin receptor substrate (IRS) proteins is a potential inhibitory mechanism in insulin signaling. Here we show that IRS-2 is phosphorylated by glycogen synthase kinase (GSK)-3. Phosphorylation by GSK-3 requires prior phosphorylation of its substrates, prompting us to identify the “priming kinase.” It was found that the stress activator anisomycin enhanced the ability of GSK-3 to phosphorylate IRS-2. Use of a selective c-Jun NH2-terminal kinase (JNK) inhibitor and cells overexpressing JNK implicated JNK as the priming kinase. This allowed us to narrow down the number of potential GSK-3 phosphorylation sites within IRS-2 to four regions that follow the motif SXXXSP. IRS-2 deletion mutants enabled us to localize the GSK-3 and JNK phosphorylation sites to serines 484 and 488, respectively. Mutation at serine 488 reduced JNK phosphorylation of IRS-2, and mutation of each site separately abolished GSK-3 phosphorylation of IRS-2. Treatment of H4IIE liver cells with anisomycin inhibited insulin-induced tyrosine phosphorylation of IRS-2; inhibition was reversed by pretreatment with the JNK and GSK-3 inhibitors. Moreover, overexpression of JNK and GSK-3 in H4IIE cells reduced insulin-induced tyrosine phosphorylation of IRS-2 and its association with the p85 regulatory subunit of phosphatidylinositol 3-kinase. Finally, both GSK-3 and JNK are abnormally upregulated in the diabetic livers of ob/obmice. Together, our data indicate that IRS-2 is sequentially phosphorylated by JNK and GSK-3 at serines 484/488 and provide evidence for their inhibitory role in hepatic insulin signaling.

Abstract Thrombin is a serine protease with multiple functions, including the conversion of fibrinogen to fibrin, platelet activation, activation of Factor VIII and Factor V. Although many low molecular weight substrates have been developed for the study of thrombin catalytic activity, our interest in analyzing thrombin activity in the blood of a living mouse required development of a new class of thrombin substrates of high affinity and high selectivity whose product upon hydrolysis could be visualized by intravital fluorescence microscopy. We have developed a novel substrate for thrombin using the fluorochrome Alexa 488 and the quencher QSY35. Alexa 488 is conjugated to the N-terminus of a 12 amino acid peptide based upon the thrombin cleavage site in the α-chain of fibrinogen and the quencher is coupled to the C-terminus of the peptide, yielding Alexa 488-KGGVR-GPRVVEA-QSY35. We term this substrate FBG-12. Through fluorescence energy resonance transfer, the emission from Alexa 488 is absorbed by the quencher QSY 35, thus minimizing fluorescence, since the two moieties have overlapping spectral properties and are separated by a Förster radius less than 44 Å. Thrombin hydrolyzes the peptide yielding the N-terminal fragment, Alexa 488-KGGVR-COOH, which is highly fluorescent, and the C-terminal fragment NH2-GPRVVEA-QSY35 peptide which, being physically separated from the Alexa 488, no longer quenches the fluorochrome. The peptide was synthesized by solid phase peptide synthesis, its N-terminus and C-terminus were modified with Alexa 488 and QSY35 respectively, and its identity confirmed by mass spectroscopy and protein sequencing. The kinetic properties of FBG-12 were determined in vitro. Hydrolysis of the substrate by thrombin resulted in a linear increase in fluorescence at 525 nm over time and was dependent on enzyme concentration. The fluorescence of the product of thrombin hydrolysis of FBG-12 was 120-fold greater than that of the substrate. Michaelis-Menten kinetic analysis of thrombin hydrolysis of the fluorogenic substrate FBG-12 revealed a Km of 2 μM, a kcat of 759 s−1, and a kcat/Km of 3795 x 106 M−1 s−1. To determine the selectivity of this substrate, other plasma serine proteases were analyzed for their ability to hydrolyze FBG-12. Factor Xa, Factor VIIa, and activated protein C did not hydrolyze FBG-12. Factor XIa (Km 11 μM, kcat 20s−1, kcat/Km = 20 x 106 M−1 s−1) did hydrolyze FBG-12, although the reaction was inefficient compared to thrombin. Our characterization of FBG-12 suggests that this substrate is hydrolyzed efficiently and selectively by thrombin. Its spectral properties and solubility in a physiologic environment make FBG-12 a suitable substrate for the detection of thrombin activity via intravital imaging during thrombus formation in vivo.

A total of 8869 shots were registered (167,40±21,45). The crystal structure of the perovskite phase KCaF3 has been redetermined at 4.2 and 300 K using powder neutron diffraction collected at the highest resolution. At both temperatures the phase was found to be orthorhombic in space group Pnma, with lattice parameters a=0.622 879(5) nm, b=0.870 031(7) nm, c=0.611 210(5) nm at 4.2 K, and a=0.621 488(6) nm, b=0.876 360(8) nm, c=0.616 481(6) nm at 300 K. The CaF6 octahedron is regular at both temperatures with octahedral rotations of 9.6° and 13.2° for the in-phase and anti-phase tilts, respectively, at 4.2 K. No evidence was found to support the recent revision of the space group from Pnma to the monoclinic space group B21∕m.

ABSTRACT Mutant forms of herpesvirus saimiri (HVS) subgroup C strain 488 with deletions in either STP-C488 or Tip were constructed. The transforming potentials of the HVS mutants were tested in cell culture and in common marmosets. Parental HVS subgroup C strain 488 immortalized common marmoset T lymphocytes in vitro to interleukin-2-independent growth, but neither of the deletion mutants produced such growth transformation. Wild-type HVS produced fatal lymphoma within 19 to 20 days of experimental infection of common marmosets, while HVS ΔSTP-C488 and HVS ΔTip were nononcogenic. Virus was repeatedly isolated from the peripheral blood of marmosets infected with mutant virus for more than 5 months. These results demonstrate that STP-C488 and Tip are not required for replication or persistence, but each is essential for transformation in cell culture and for lymphoma induction in common marmosets.

Abstract Introduction: Atrial fibrillation (AF) is the most common sustained arrhythmia in clinical practice, contributing directly to morbidity and mortality largely through thromboembolism events such as stroke. The risk of stroke in patients with AF is five times higher than in patients without AF. The pathophysiological of thromboembolism in AF is multifactorial and is not only related to stasis in a poorly contractile left atrium. Indeed, there is an increasing body of evidence to support that Virchow triad, including abnormal blood stasis, endothelial damage, and changes in blood constituents, is the main cause of thromboembolism or prothrombotic state in AF patients. However, relatively little is known about the precise role of phosphatidylserine (PS), one of the blood constituents, in the pro-thrombotic state or hypercoagulability of non-valvular AF (NVAF). Our objectives were to study the increased PS exposure on microparticles (MPs) and the outer membrane of MP-origin blood cells in NVAF patients, and to evaluate their procoagulant activity (PCA). Methods: Our study included NVAF patients without (n = 60) and with left atrial thrombosis (n = 30) and healthy controls (n = 30). PS exposure on MPs and blood cells was analyzed with flow cytometry and confocal microscopy. PCA was evaluated using clotting time, extrinsic/intrinsic FXa and prothrombinase production, and fibrin formation assays. Inhibition assays of PCA of PS+ MPs and blood cells were performed using lactadherin. Results: The number of PS+ MPs was significantly higher in NVAF patients (1.8-fold, P < 0.001) and NVAF patients with left atrial thrombosis (2.7-fold, P < 0.001) compared with healthy subjects, respectively. Furthermore, the number of PS+ MPs in NVAF patients with left atrial thrombosis was significantly higher than that in NVAF patients without (1.4-fold, P < 0.01, Figure 1A). Moreover, the PS+ MPs were originated from platelets (CD41a+, PMPs), neutrophils (CD66b+, NMPs), mononuclear cells (CD14+, MMPs), lymphocytes (CD3/19+, LMPs), erythrocytes (CD235a+, ErMPs), and endothelial cells (CD31+ CD41a-, EMPs) (Figure 1B). The levels of PS+ platelets, neutrophils, mononuclear cells, lymphocytes, and erythrocytes were significantly higher (all P < 0.001) in each NVAF group than those in healthy controls. And the levels of PS+ platelets, neutrophils, and erythrocytes in NVAF patients with left atrial thrombosis were significantly higher than those in NVAF patients (all P < 0.01, Figure 1C). In addition, circulating PS+ MPs cooperated with PS+ blood cells, contributing to markedly shortened coagulation time and dramatically increased FXa/thrombin generation and fibrin formation in each NVAF group (all P < 0.01). Moreover, blockade of exposed PS on MPs and blood cells with lactadherin inhibited PCA by approximately 80%. Conclusions: Our results suggest that increased PS exposure on MPs and blood cells play a procoagulant role in NVAF patients with or without left atrial thrombosis. Blockade of PS prior to left atrial thrombosis could become a novel therapeutic strategy to prevent thrombosis in these patients. Relations between aristocratic landowners and agriculturalists in the late Roman period were part of systems of vertical alliance that were fluid, ambiguous and flexible. This thesis moves beyond theories of Patrocinium and the 'colonate of the late Roman Empire' to reconstruct those alliance systems. Three basic types of vertical interaction in rural contexts may be identified: tenancy, patronage and slavery. Peasants also interacted with aristocratic landowners in relations of casual or periodic labour, estate management and legal agency. It is argued that patronage for the purposes of tax evasion and registered tenancy were merely elements in these systems of vertical alliance. Chapter 1 addresses the attitudes of late Roman legislators and authors to patronage and registered tenancy through the language they employed in describing these phenomena. It will be demonstrated that these concepts continued to be analytically fluid, and did not crystallise into the institutions that have been labelled Patrocinium and the 'colonate' in the historiographical literature. In Chapter 2, late Roman rural labour relations are explored. It is argued that the analytical distinctions between the three categories of vertical alliance identified above mask a reality of blurred boundaries and elision of various relationships of dependence. In Chapter 3, those systems of vertical alliance are further contextualised within the social and economic world of the fourth and fifth centuries. It is argued that that world was characterised by 'landscapes of diversity', within which particular systems of interaction between peasants and aristocrats were necessarily adaptable and disparate. Those systems of interaction complemented and competed with patterns of horizontal organisation within individual rural communities, and between communities. It is upon these patterns of competition and complementarity that the tax system of the late Roman period was imposed in the late third century. Chapter 4 focuses upon the tax system, and the elements of it that contributed to the high visibility of structures of rural social organisation in the period.

ATmega16 and FT232R were used as the most important parts of hardware solution. Software for control of the position system was designed in C++ Builder and it can run under MS Windows XP. This work is based on semestral thesis and in future can be used in other applications or modified. 2D position system will be later used by UBMI in education and scientific applications.

The historical evolution of the heptanese during the classical and hellenistic periods has been little studied until now. The main aim of this thesis is consequently, thanks to a geopolitical approach, to show the richness and the complexity of the international life of the four principal ionian islands forming this archipelago, namely, corcyra (corfu), leucas, cephallenia, and zacynthos. Located between the balkanic and aegean greek world on the east, and the italian-sicilian world on the west, these islands were geopolitical positions of great importance, coveted by all the imperialists powers anxious, between 480 and 167 b. C. , to take control of the ionian sea and the corinthian gulf, and therefore, of the maritime links between the two basins of the mediterranean sea : corinth, athens, sparta, macedonia, syracuse, carthage and rome were among these states that, either simply manifested their presence, or firmly established their domination there

RESUMO Serão investigadas, nesta dissertação, as origens da democracia ateniense através da análise de quatro epigramas de Simônides de Ceos e da contextualização de alguns outros. Partindo do assassinato de Hiparco por Harmódio e Aristogíton e traçando suas conseqüências, tanto do ponto de vista mítico quando político, tentaremos demonstrar como o povo, agora identificado como mesmo ideal de igualdade perante a lei (isonomia), foi capaz de afastar tanto o perigo de uma helotização da Ática, quanto o de uma submissão ao império aquemênida, ao vencer duas batalhas decisivas, cujo relato foi preservado em inscrições que chegaram até nós sob o nome de Simônides por transmissão literária ou epigráfica ou ambas. Também iremos lidar com todos os aspectos relativos à composição, caráter e transmissão de cada um dos quatro epigramas comentados, além de fornecer uma tradução de todos os outros - incluindo os novos fragmentos elegíacos recentemente descobertos - que possam ter alguma relevância para o assunto em questão.
The origins of Athenian democracy are herein surveyed through the analysis of four Simonidean epigrams and the canvassing of several others. Starting from the murder of Hipparchus by Harmodius and Aristogeiton, and following its consequences, both mythic and political, we shall proceed to demonstrate how the people, now identified with the same democratic ideal, was able to eschew both the danger of helotization by Sparta and submission by the Persian Empire by its winning two decisive battles recorded in inscriptions that came down to us either through literary or epigraphical transmission or both. We shall also deal with all aspects surrounding the composition, nature and transmission of each of the four epigrams, besides providing a translation of all other epigrams - including the newly discovered elegiac fragments - that may bear any relevancy to the subject under appreciation.

Abstract In Language 64: 3 (1988), Lyle Campbell contributed a review article concerning my book Language in the Americas (henceforth C and LIA, respectively) that was uniformly negative. I believe that readers of Language should be made aware that one year before my book appeared in print, on the basis of a brief exposition of its contents in Current Anthropology (Greenberg, Turner, and Zegura 1986), Campbell wrote that my classification of American Indian languages ‘should be shouted down’ (1986: 488). Under these circumstances, an objective review could hardly be expected. More importantly, the reader of C’s review article gets no real notion of the main arguments and contents of the book.

Frutas são alimentos perecíveis, portanto, sua desidratação contribui para o aumento de sua vida útil. A liofilização vem recebendo destaque dentre as operações de secagem, já que os produtos não são submetidos a altas temperaturas. O objetivo do presente trabalho foi modelar a cinética de liofilização de tangerinas e uvas e avaliar sensorialmente as frutas desidratadas. A liofilização foi realizada com frutas congeladas à pressão de 70 Pa e aquecimento a 60 °C por 48 h. Os dados experimentais foram ajustados à lei de Fick e a modelos empíricos/semi empíricos. Foram avaliados sensorialmente cor, sabor, aroma, textura e impressão global dos consumidores com base em uma escala hedônica de nove pontos. A lei de Fick representou satisfatoriamente os dados experimentais de desidratação das frutas. O modelo de Page foi o que melhor representou a liofilização das uvas e o Aproximação da difusão, a das tangerinas. Aproximadamente 65 % das notas atribuídas pelos provadores às frutas desidratadas situaramse entre os termos “gostei ligeiramente” e “gostei muito” para todos os quesitos avaliados. A textura das frutas desidratadas apresentou média superior a 7,0. Os resultados mostraram que há mercado para produtos inovadores, como uvas e tangerinas liofilizadas.

A program has been developed to fabricate silver film-diffused glass, optical waveguides for use as optical couplers in optical signal processing systems. The diffusion process was assisted by applying electric fields ranging from 40 to 200 V/mm and at elevated temperatures ranging from 270 to 300°C. The graded-index profiles were analyzed using the WKB method and the effective index of refraction values measured with the prism coupling technique. Results of the WKB analysis showed that a Gaussian profile fits the effective index data best. Loss measurements of around 1 dB/cm were found for wavelengths of 633 nm or longer. However, at shorter wavelengths of 488 and 514 nm losses are substantially higher and could reach several dB/cm. One mechanism proposed for this loss is the wavelength-dependent absorption by silver aggregates or silver complexes which are present in the guiding wave layer. A strong red incoherent emission is produced in the guiding-wave layer, and this radiation is emitted normal to the waveguide surface when guided wave modes are excited at 488 nm. When 514 nm is used, this emission takes on a brownish red appearance. This emission consists of broad bands with band centers that are shifted from the excitation line by wavenumber shifts ranging from 3190 to 6475 cm-1, with 488-nm excitation and with 514-nm excitation, the shifts range from 4250 to 5320 cm-1.

The phenomena of self-pulsation, oscillation chaos, and frequency shift in selfpumped photorefractive BaTiO3 crystal have been recently studied.1,2 In this paper we present the pulsation and chaotic phenomena in self-pumped BaTiO3 for both divergent and convergent beams focused at various locations near the crystal surfaces. An argon laser (Innva model 70) operating at 488 nm pumps a 6- × 3- × 6-mm BaTiO3 crystal which was mounted on a translation stage to control the incident beam focusing location. The front c-face of the crystal was taken as the reference plane with the top right corner coordinate defined as (x, y) = (0,0). The convergent and divergent beams (within the crystal) are defined by focal points at (3, 6) and (3, –6) mm, respectively, where the c-axis is along the 6-mm length. A focal length f = 200 mm was used to keep the incident beam the same spot size of 1 mm on the crystal surfaces for both convergent and divergent cases.

Abstract This paper is aimed to an experimental study on the flow patterns formed by heavy crude oil (488 mPa.s, 925.5 kg/m3 at 20 °C) and water inside vertical and horizontal 1 in. pipes. The interfacial tension was 29 dynes/cm. Effort is concentrated into flow pattern characterization, which was visually defined. The similarities with gas-liquid flow patterns are explored and the results are expressed in flow maps of the superficial velocities. In contrast with other studies, the annular flow pattern (‘core annular flow’) was observed in both horizontal and vertical test sections. In fact this flow pattern typically occurs in heavy oil-water flows at low water input fractions. Because of the practical importance of core flow in providing an effective means for heavy oil production and transportation, this paper discusses two criteria that favor its occurrence in pipes.

Abstract The Fluorescent Microthermal Imaging (FMI) technique was developed as a new failure analysis tool for hot spot and leakage site localization in order to complement the Liquid Crystal and Light Emission Microscopy techniques. The goal of this work was to produce a user-friendly and automated FMI system which is simple to use by the laboratory personnel. The instrumentation was set up on a probe station. It involved a uv light source, a dark box, a motorized optical microscope, a slow scan cooled CCD camera, a temperature controller, electrical test equipment, and a PowerMacintosh computer with IPLab software for instrumentation control, image acquisition and image processing. Software extensions for IPLab were developed with Think C to add automated functions to the system using the GPIB-IEEE 488 bus. IPLab scripts were written to fully automate instrumentation control, image acquisition and processing. A study of EuTTA compound fluorescence intensity variation with temperature, exposure time, and uv excitation was performed in order to characterize the system. The FMI technique was then applied to hot spot identification on microelectronic devices and failure analysis. With a few simple modifications, the same system was also employed for Light Emission Microscopy.

A program was undertaken to determine potential emission benefits of use of bio-based fuels and lubricants in snowmobile engines. Candidate fuels and lubricants were evaluated using a fan-cooled 488-cc Polaris engine, and a liquid-cooled 440-cc Arctco engine. Fuels tested include a reference gasoline, gasohol (10% ethanol), and an aliphatic gasoline. Lubricants evaluated include a bio-based lubricant, a fully synthetic lubricant, a high polyisobutylene (PIB) lubricant, as well as a conventional, mineral-based lubricant. Emissions and fuel consumption were measured using a five-mode test cycle that was developed from analysis of snowmobile field operating data. Emissions measured include total hydrocarbons (THC), carbon monoxide (CO), nitrogen oxides (NO<sub>x</sub>), carbon dioxide (CO<sub>2</sub>), particulate matter (PM), polycyclic aromatic hydrocarbons (PAH, both particulate bound and vapor-phase), and individual hydrocarbon species (C<sub>1</sub>-C<sub>12</sub>). Emissions and fuel consumption using bio-based fuels and lubricants were compared to results using a conventional fuel and lubricant. Promising candidates were identified and recommended for further study in a field demonstration in Yellowstone National Park.</div> <div class="htmlview paragraph"> </div>

<div class="section abstract"><div class="htmlview paragraph">In the present work, five surrogate components (n-Hexadecane, n-Tetradecane, Heptamethylnonane, Decalin, 1-Methylnaphthalene) are proposed to represent liquid phase of diesel fuel, and another different five surrogate components (n-Decane, n-Heptane, iso-Octane, MCH (methylcyclohexane), Toluene) are proposed to represent vapor phase of diesel fuel. For the vapor phase, a 5-component surrogate chemical kinetic mechanism has been developed and validated. In the mechanism, a recently updated H<sub>2</sub>/O<sub>2</sub>/CO/C<sub>1</sub> detailed sub-mechanism is adopted for accurately predicting the laminar flame speeds over a wide range of operating conditions, also a recently updated C<sub>2</sub>-C<sub>3</sub> detailed sub-mechanism is used due to its potential benefit on accurate flame propagation simulation. For each of the five diesel vapor surrogate components, a skeletal sub-mechanism, which determines the simulation of ignition delay times, is constructed for species C<sub>4</sub>-C<sub>n</sub>. The five skeletal sub-mechanisms are coupled with the new C<sub>2</sub>-C<sub>3</sub> and H<sub>2</sub>/O<sub>2</sub>/CO/C<sub>1</sub> detailed sub-mechanisms. In order to simulate soot, a semi-detailed soot model is coupled with the 5-component diesel vapor surrogate chemical kinetic mechanism. Together with a reduced NOx (oxides of nitrogen) sub-mechanism, the final version of the diesel mechanism has 119 species and 488 reactions, which are feasible currently for simulating diesel engine combustion and emissions. The reaction rate constants of the five skeletal sub-mechanisms were optimized in this work to match available experimental data of either pure fuels or fuel blends, including ignition delay times, laminar flame speeds, and important species profiles in the literature. Using the new models developed in this work, simulations of the combustion and emissions of several diesel engines under typical operating conditions were carried out. The simulated in-cylinder pressures and emissions including unburned hydrocarbons, nitrogen oxides, carbon monoxide, and soot were compared with the experimental data, showing good agreements between simulations and experiments.</div></div>

In recent years much work has been done to determine the cause of photosensitive grating effects in germanium-doped silica fibers. Past research has suggested a link between the formation of defect centers and the occurrence of photosensitivity in these materials. For the first time, we observe a correlation between photosensitivity and the Ge E' center, characterized by a singly charged oxygen vacancy, as measured with electron-spin resonance (ESR) techniques. We irradiated a length of single-mode optical fiber (n = 0.028, c = 438 nm) with argon laser radiation in the usual manner. A grating was written in the fiber, and optical transmission and reflection data were recorded. Subsequently, the fiber end was dipped in index-matching fluid, prohibiting back reflection, and the sample was re-exposed with a high-power Ar+ laser beam for an extended time to induce sufficient defects to be detectable with ESR. Analysis of optical data at a wavelength of 488 nm showed a 50% decrease in fiber transmission after 3.5 s. ESR analysis of the same sample showed the Ge E' defect concentration more than doubled over that of an identical, but unexposed, fiber. We will discuss the significance of these results in the context of the current theory of two-photon absorption to these E' centers.

Continuous-wave (cw), narrow-bandwidth lasers that are tunable in the mid-IR wavelength range are important for analytical applications. The primary laser sources for this wavelength region include color-center as well as lead-salt diode lasers. Wide tunability can be achieved by difference-frequency mixing. Good results are obtained with LiNbO3 by using collinear 90° phase matching and temperature tuning. We achieved tuning between 2.4 µm and 3.6 µm and cw output powers of several microwatts by mixing cw dye- (Rh 6G) and 488 nm Ar+-laser radiation in LiNbO3. The required range of temperature was 230–400°C. The wavelength region beyond 4 µm can be accessed with new solid-state lasers and optical nonlinear materials. The design options of novel tunable cw IR laser systems based on difference-frequency mixing at near-room temperature in AgGaS2 or AgGaSe2 will be discussed. For AgGaS2, the mixing of Ti:sapphire- and dye-laser radiation permits tunability of 3.85–9.1 µm with a higher figure of merit than for LiNbO3. Furthermore, the wavelength can be extended to 12 µm by using AgGaSe2, which exhibits the highest figure of merit in this region. This can be accomplished, for example, by mixing cw single-frequency 1.32 µm Nd:YAG and NaCl:OH color-center laser radiation. The temperature-stability and damage-threshold issues were examined carefully for both new nonlinear optical materials.

Photorefractive materials such as LiNbOa3, BaTiOa3, KNbO3, Sr1- x Ba x Nb2O6 (SBN), Ba2- x Sr x K1- y Na y Nb5O15 (KNSBN), and Bi12SiO20 (BSO) are attractive candidates for optical data processing applications. The real time holographic gratings formed by the photorefractive effect produce energy coupling between the two writing beams (two-wave mixing). This energy transfer from one optical to another has potential applications in image amplification and correlation convolution operations. In this work, investigation of two-wave mixing (beam coupling) has been performed for the first time in Fe-doped (K x Na1- x )0.4(Sr y Ba1- y )0.8Nb2O6 (KNSBN) crystals. These crystals have photorefractive sensitivity and time response comparable with those of BaTiO3, KNbO3, and SBN. Large size crystals of relatively good optical quality have been grow. The composition of the crystal used is (K0.5Na0.5)0.4 (Sr0.61Ba0.39)0.8Nb2O6 with 0.05-wt. % Fe doping. The crystal dimension is 10 × 6.7 × 3.5 mm with the c-axis normal to the 10 × 3.5-mm face. Beam coupling and object beam amplification have been investigated as a function of light intensity, light wavelength, object beam to reference beam ratio, and the ange between the two beams. Beam coupling at 488-nm wavelength is observed to be stronger than that at 514.5 nm. The object beam gain is observed to be less dependent on the total beams' intensity and on the ratio between the object and reference beams.

Techniques for fabrication of large, uniform arrays of semiconductor structures with nanometer-scale dimensions are of great interest for investigating quantum size effects. We report on a simple technique for fabricating arrays of Si wires with transverse dimensions ≤ 10 nm. This technique takes advantage of the capabilities of optical interferometric lithography to produce very high spatial frequency gratings and the strongly directional KOH etching of Si. A <110> Si wafer is prepared with thin (50-nm) Si3N4 and standard photoresist (0.15 mm) layers. A submicrometer period grating is interferometrically exposed in the photoresist (0.2-1 mm pitch) by using a cw Ar-ion laser source (361 or 488 nm). Following resist development, the grating pattern is transferred into the Si3N4 by CF4 plasma etching. Large area patterns with ~500-1000 nm lines are readily produced. For lines aligned in the <110> direction, a 40% KOH solution provides a highly selective (500:1) wet etch that produces very high aspect ratio Si lines. The etch selectivity also serves to reduce any nonuniformities or edge roughness induced in the lithography and etching steps. At this stage, Si structures with widths of 30-100 nm and depths of 0.5- 1 mm are obtained. Finally, a controlled dry oxidation (~800°C) is used to further reduce the Si dimensions. Large areas of uniform nanometer scale structures, including Si wires isolated by SiO2 from the substrate, are produced. Photoluminescence and Raman scattering studies will be presented.

Avec l'augmentation attendue des activités minières dans les régions nordiques du Canada en raison de la demande mondiale de minéraux critiques, il est impératif de concevoir des méthodes nouvelles et fiables pour identifier les lacs qui peuvent être situés au-dessus des sections de sol non gelées (c.-à-d. les taliks) dans les environnements de pergélisol continu. Lorsque les taliks pénètrent complètement le pergélisol, ils peuvent permettre des interactions entre les eaux de surface et les eaux souterraines sous le pergélisol, ce qui présente des défis importants pour les opérations minières. Ce rapport fournit des données de référence détaillées sur la température de l'eau, la conductivité et parfois des mesures de pH pour les lacs près de Rankin Inlet, au Nunavut, effectuées en avril et en septembre 2023. Cet ensemble de données alimentera nos modèles thermiques et géophysiques pour l'identification des taliks et les modèles conceptuels cryo-hydrogéologiques pour la simulation de l'écoulement hydrogéologique souterrain dans les régions de pergélisol continu. Nos résultats révèlent notamment qu'en avril, les températures de la colonne d'eau dans les bassins lentiques sélectionnés variaient de 0,1 à 2°C directement sous la couverture de glace et de 1,7 à 3,5°C à l'interface eau-sédiment. Inversement, en septembre, la colonne d'eau des bassins étudiés était isotherme. Ces profils de température suggèrent que les lacs étudiés présentent des caractéristiques indiquant des bassins polymictiques froids, la stratification hivernale étant plus prononcée avec l'augmentation de la profondeur du bassin. En outre, nos mesures de conductivité indiquent une relation salinité-densité dans la colonne d'eau des lacs. Plus précisément, les mesures effectuées directement sous la couche de glace et près du fond du lac variaient respectivement de 74 à 168 µS cm-1 et de 77 à 352 µS cm-1 en avril 2023, tandis que les conductivités de la surface et du fond du lac variaient respectivement de 44 à 111 µS cm-1 et de 73 à 488 µS cm-1 en septembre. En outre, les évaluations préliminaires des effets de la cryoconcentration sur la charge en solutés pendant les saisons de prise et de retrait des glaces indiquent une contribution potentielle aux écarts observés dans les conductivités de la colonne d'eau entre les deux saisons d'échantillonnage sur le terrain. Toutefois, il est essentiel de procéder à des analyses géochimiques supplémentaires pour valider cette hypothèse et explorer la possibilité d'autres phénomènes naturels qui pourraient expliquer ces différences.

In summer 2021, drought and low humidity combined with record-breaking heat of up to 48.8°C (119.8°F) led to fire outbreaks across the Mediterranean countries, killing more than 100 people and burning more than 620,000 ha of land in July and August. As forests are often fiercely protected areas, the common approach to wildfire management in the Mediterranean is to extinguish every fire as it appears, but counterintuitively this supports the formation of “mega-fires” that burn beyond our capacity to control. Fires are not yet seen as integral to ecosystem functioning, and the management of fires and fires are often disconnected. This technical background report for the 2021/2022 edition of the Interconnected Disaster Risks report analyses the root causes, drivers, impacts and potential solutions for the Mediterranean wildfires through a forensic analysis of academic literature, media articles and expert interviews.

This report describes the demonstration of technologies and procedures developed during April 2002 and May 2004 under the Joint Rapid Airfield Construction (JRAC) Program. The demonstration took place at Sicily Landing Zone (LZ) at Fort Bragg, NC, in July of 2004. The objective of the exercise was to demonstrate the procedures and technologies developed under the JRAC Program by rapidly building two parking aprons capable of supporting C-130 transport aircraft taxiing and parking operations. The exercise was conducted under continuous 24-hr operations to simulate a real-world rapid construction environment. Apron 1 (north apron) was constructed using two technologies, one-half being ACE™ Matting and the other half being a cement-polymer stabilized soil surface. Apron 2 (south apron) was constructed solely of a fiber-cement-stabilized soil system. Both aprons were treated with a polymer emulsion surface application to form a sealed surface against abrasion and water infiltration. The entire construction of both aprons required 76 hr, with Apron 1 finished in 48 hr. The construction of Apron 1 was validated by operation of a C-130 aircraft approximately 31 hr after completion with success and high praises from the aircraft flight crew on the stability and surface of the apron, as well as its dust-abating characteristics.

: The necessity to improve broiler thermotolerance and performance led to the following hypothesis: (a) thethermoregulatory-response threshold for heat production can be altered by thermal manipulation (TM) during incubation so as to improve the acquisition of thermotolerance in the post-hatch broiler;and (b) TM during embryogenesis will improve myoblast proliferation during the embryonic and post-hatch periods with subsequent enhanced muscle growth and meat production. The original objectives of this study were as follow: 1. to assess the timing, temperature, duration, and turning frequency required for optimal TM during embryogenesis; 2. to evaluate the effect of TM during embryogenesis on thermoregulation (heat production and heat dissipation) during four phases: (1) embryogenesis, (2) at hatch, (3) during growth, and (4) during heat challenge near marketing age; 3. to investigate the stimulatory effect of thermotolerance on hormones that regulate thermogenesis and stress (T₄, T₃, corticosterone, glucagon); 4. to determine the effect of TM on performance (BW gain, feed intake, feed efficiency, carcass yield, breast muscle yield) of broiler chickens; and 5. to study the effect of TM during embryogenesis on skeletal muscle growth, including myoblast proliferation and fiber development, in the embryo and post-hatch chicks.This study has achieved all the original objectives. Only the plasma glucagon concentration (objective 3) was not measured as a result of technical obstacles. Background to the topic: Rapid growth rate has presented broiler chickens with seriousdifficulties when called upon to efficiently thermoregulate in hot environmental conditions. Being homeotherms, birds are able to maintain their body temperature (Tb) within a narrow range. An increase in Tb above the regulated range, as a result of exposure to environmental conditions and/or excessive metabolic heat production that often characterize broiler chickens, may lead to a potentially lethal cascade of irreversible thermoregulatory events. Exposure to temperature fluctuations during the perinatal period has been shown to lead to epigenetic temperature adaptation. The mechanism for this adaptation was based on the assumption that environmental factors, especially ambient temperature, have a strong influence on the determination of the “set-point” for physiological control systems during “critical developmental phases.” In order to sustain or even improve broiler performance, TM during the period of embryogenesis when satellite cell population normally expand should increase absolute pectoralis muscle weight in broilers post-hatch. Major conclusions: Intermittent TM (39.5°C for 12 h/day) during embryogenesis when the thyroid and adrenal axis was developing and maturing (E7 to E16 inclusive) had a long lasting thermoregulatory effect that improved thermotolerance of broiler chickens exposed to acute thermal stress at market age by lowering their functional Tb set point, thus lowering metabolic rate at hatch, improving sensible heat loss, and significantly decreasing the level of stress. Increased machine ventilation rate was required during TM so as to supply the oxygen required for the periods of increased embryonic development. Enhancing embryonic development was found to be accomplished by a combination of pre-incubation heating of embryos for 12 h at 30°C, followed by increasing incubation temperature to 38°C during the first 3 days of incubation. It was further facilitated by increasing turning frequency of the eggs to 48 or 96 times daily. TM during critical phases of muscle development in the late-term chick embryo (E16 to E18) for 3 or 6 hours (39.5°C) had an immediate stimulatory effect on myoblast proliferation that lasted for up to two weeks post-hatch; this was followed by increased hypertrophy at later ages. The various incubation temperatures and TM durations focused on the fine-tuning of muscle development and growth processes during late-term embryogenesis as well as in post-hatch chickens.

The girth weld defect acceptance standards based on good workmanship reflect quality levels that can be reasonably expected from a qualified welder. Workmanship (WM) or weld quality standards specify a maximum allowable length whilst the percentage loss of cross-sectional area is used for porosities. This approach to defect acceptance philosophy has arisen from the use of radiography as the NDE technique for detecting and quantifyingweld discontinuities. The first WM standard for inspection and acceptance of finished girth welds was implemented by API in 1953. The specific requirements of the 1953-standard were largely based on the Unfired Pressure Vessel Code which was first adopted by ASME in 1931. Since then, a number of slightly revised standards were issued to reflect what should be attainable with normal good welding practices. The failure behaviour of defective girth welds in large diameter pipe lines was assessed using radiographic and mechanized ultrasonic inspection, small scale (tensile, hardness, Charpy and CTOD) and wide plate tests. The specimens were taken from girth welds in API 5LX70 pipe of 1219 mm (48 inches) in diameter by 8,0 mm (0,323 inch) and 13,3 mm (0,524 inch) wall. The test welds were made with the SMAW (8 welds) and GMAW (9 welds) welding processes. Upon completion of the non-destructive tests, 96 curved wide plate specimens were tested to destruction under tensile load. Testing was performed at low temperature (-50�C/-58�F). Defect type, defect position and size were determined from photographs of the fracture face and macro sections (defect characterization and sizing). In total, 290 typical surface breaking and embedded defects in SMAW or GMAW girth welds have been evaluated.

Following preliminary observations by one of the collaborating scientists on this project and the completion of a 1-year, BARD-supported feasibility study (IS-1908-90F), this 3-year BARD project has been executed. The main objectives of the research were to elucidate biochemical and pathological aspects of UV-induced resistance in fruits and vegetables, to characterize physical and biological variables of induced resistance and delay of ripening, and to explore the application of the treatment as a control practice of postharvest diseases and shelf-life extension of fruits and vegetables. Our findings, which are detailed in numerous joint publications, have shown that the effect of UV-C light on induction of resistance and delay of ripening is a general one and of wide oddurrence. Apart from surface sterilization of the commodity, the reduction of decay of different fungi has been associated with and induced resistance phenomenon which gradually builds up within 24 to 48 hours after the UV treatment and can be reversed by visible light. In citrus, induced resistance has been associated with increased activity of the enzymes phenylalanine ammonia-lyase and peroxidase, and with the levels of endglucanase and chitinase. In tomato, resistance was correlated with the production of high levels of tomatine. Our study of some molecular aspects of the induced resistance in grapefruit has revealed the induction of a cDNA which represents a gene encoding for an isoflavone reductase-like protein that, in legumes, has been associated with phytoalexin biosynthesis. This gene was cloned and sequenced. Delay of ripening was associated in tomato with inhibition of ethylene production, carotenoid synthesis, and chlorophyll degradation and with the presence of high levels of polyamines. In peach fruit epiphytic populations of a yeast increased following the UV treatment. Pilot-size treatment and packing lines were constructed in the US and Israel to test the application of the UV treatment on a semi-commercial scale. Although effective in reduction of decay and delay of ripening, a number of problems will have to be addressed before practical application of this methodology can be realized. The main issues are associated with the temporal and variable response to the treatment, and its relationship to the maturity and date of harvest of the commodity.
