Résumé :
The surfactant system of the nose was examined biochemically in control cases and compared to cases of primary atrophic rhinitis. The study group included 25 cases with primary atrophic rhinitis compared to 10 normal volunteers. Biochemical analysis of the nasal aspirate in these cases revealed the presence of phospholipids constituting surfactant with phosphatidylcholine constituting 75.35 per cent of the total phospholipids. Biochemical analysis of the nasal aspirate in cases with primary atrophic rhinitis revealed a significant decrease in the total phospholipids compared to normal cases and also a significant change in the phospholipid profile. Thus significant biochemical changes in the surfactant system of the nose is an evident and early finding in cases of primary atrophic rhinitis. This suggests a possible role for surfactant deficiency in the aetiopathogenesis of cases of primary atrophic rhinitis.

Résumé :
Hexokinase 1 fromArabidopsis thaliana(AtHXK1) plays a dual role in glycolysis and sugar sensing for vital metabolic and physiological processes. The uncoupling of glucose signalling from glucose metabolism was demonstrated by the analysis of two mutants (AtHXK1G104DandAtHXK1S177A) that are catalytically inactive but still functional in signalling. In this study, substrate-binding experiments indicate that the two catalytically inactive mutants have a high affinity for glucose, and an ordered substrate-binding mechanism has been observed for wild-typeAtHXK1. The structure ofAtHXK1 was determined both in its inactive unliganded form and in its active glucose-bound form at resolutions of 1.8 and 2.0 Å, respectively. These structures reveal a domain rearrangement ofAtHXK1 upon glucose binding. The 2.1 Å resolution structure ofAtHXK1S177Ain the glucose-bound form shows similar glucose-binding interactions as the wild type. A glucose-sensing network has been proposed based on these structures. Taken together, the results provide a structural explanation for the dual functions ofAtHXK1.

Résumé :
The aim of this study was to evaluate the biologic outcome of endometrial carcinomas as compared to clinical and pathologic parameters and to identify multivariate independent prognostic factors. Charts were abstracted from patients with endometrial carcinoma from 1985 to 1995. Data on clinicopathologic variables, adjuvant treatment, site of recurrence, and survival were collected. χ2 test was used to test association between variables. Kaplan-Maier method was used for survival analysis and Cox proportional hazards model for multiple regression analysis. Univariate analysis revealed that FIGO stage, tumor grade, depth of myometrial invasion, biochemical analysis of progesterone receptor status, age, additional diabetes mellitus, lymph node metastasis, and type of tumor were significantly associated with the overall-survival. For disease-free interval, FIGO stage, tumor grade, depth of myometrial invasion, biochemical analysis of progesterone receptor status, lymph node metastasis, and type of tumor were also significantly associated. Multivariate analysis revealed that FIGO stage, tumor grading, tumor type, depth of myometrial invasion, and biochemically measured progesterone receptor status were associated significantly with overall survival. A significant correlation as independent prognostic factors were also seen for recurrence free interval for FIGO stage, tumor grade, and biochemical progesterone receptor status. In multivariate statistical analysis we identified FIGO stage, tumor type, tumor grade, biochemical analysis of progesterone receptor status, and depth of myometrial invasion as independent prognostic factors for overall survival, and FIGO stage, biochemical analysis of progesterone receptor status, and tumor grade as independent prognostic factors for recurrence-free interval.

Résumé :
Of 1240 outpatients referred to the Human Genetics Clinic between 1997 and 1998, 248 [20%]had inborn errors of metabolism, 36 [14%] of which were diagnosed as mucopolysaccharidoses. Parental consanguinity was present in 82% of these patients. Deficiency of alpha-L-iduronidase [IDUA] enzyme in leukocytes and increased urinary mucopolysaccharides excretion were detected in 17 patients. The urinary spot test for glucosaminoglycans was inconclusive in 4 of the 17 cases. Results showed a correlation between the biochemical enzyme activity in leukocytes, the amount of excreted mucopolysaccharides and the subtype and course of mucopolysaccharidosis type I. We conclude that estimation of IDUA enzyme activity in leukocytes can differentiate between clinically overlapping cases of MPS I and MPS II and given the clinical manifestations of MPS I is a definitive and unequivocal method of diagnosis while the urinary spot test is inconclusive

Résumé :
As it is well understood, in biological systems, small regulatory motifs are present at all scales, thus looking at simple negative feedback loops give us some information of how autocatalytic systems may be affected by regulation. For a single template self-replication, we consider a plausible mechanism, which we reduce to a 2-variable dimensionless set of ordinary differential equations, (ODE). The stability analysis of the steady states allows us to obtain exact relations to describe two-parameter bifurcation diagrams. We include a negative feedback to the reactants input to study the effect of regulation in biochemical self-replication. Surprisingly, the simpler regulation has the largest impact on the parameter space.

Résumé :
The present study deals with the introduction of an alteration in Legendre wavelets method by availing of the Picard iteration method for system of differential equations and named it Legendre wavelet-Picard method (LWPM). Convergence of the proposed method is also discussed. In order to check the competence of the proposed method, basic enzyme kinetics is considered. Systems of nonlinear ordinary differential equations are formed from the considered enzyme-substrate reaction. The results obtained by the proposed LWPM are compared with the numerical results obtained from Runge–Kutta method of order four (RK-4). Numerical results and those obtained by LWPM are in excellent conformance, which would be explained by the help of table and figures. The proposed method is easy and simple to implement as compared to the other existing analytical methods used for solving systems of differential equations arising in biology, physics and engineering.

Résumé :
The study of emotions is complicated by a number of factors, including the biological bases of emotional experience and expression. Although biochemical changes have long been thought to be part of the evocation of emotions, research has not consistently included this level of measurement, and theories of emotion have not systematically specified the role of these biochemical bases. In this paper, we briefly review relevant biochemical principles and measures that can be integrated into research and theory on emotions. Applications of these principles in extant studies of emotion are also discussed.

Résumé :
Hematological diseases in childhood occur frequently. Analyses of hematological status as well as standard biochemical analysis of blood, as a result of hematopoetic system function, are of great importance in detection and recognition of these diseases. All hematological diseases appear as disturbance of hematopoetic system function at the level of the production and viability in blood flow; this disturbance is reflected on the hemogram and biochemical analyses of blood plasma and serum. The basic hematological parameters of blood diseases (except disturbance of blood coagulation) are Total blood count cells (erythrocytes, leukocytes with differential leukocytes formulae, trombocytes) hemoglobin concentration, amount of hematocrit (PCV, packed cell volume), erythrocytes sedimentation rate erythrocytes osmotic fragility. Among the standard biochemical analysis the determination of total bilirubin in the calculation fraction of indirect bilirubin, the amount of serum iron and transferrine (TIBC), serum copper and ceruloplasmin are in use in these diseases. Hemoglobin electrophoresis is also use. The measurement of lactate dehydrogenase activity and other enzymological analyses are rarely performed. The basic idea of this report is the evaluation of another biochemical possibility with the aim to understand the nature of hematological disorders by the use a large spectrum of enzyme analyses, which are present in different blood cells and with direct reflection on the serum enzyme activities.

Résumé :
CSF from unique groups of Parkinson’s disease (PD) patients was biochemically profiled to identify previously unreported metabolic pathways linked to PD pathogenesis, and novel biochemical biomarkers of the disease were characterized. Utilizing both 1H NMR and DI-LC-MS/MS we quantitatively profiled CSF from patients with sporadic PD (n = 20) and those who are genetically predisposed (LRRK2) to the disease (n = 20), and compared those results with age and gender-matched controls (n = 20). Further, we systematically evaluated the utility of several machine learning techniques for the diagnosis of PD. 1H NMR and mass spectrometry-based metabolomics, in combination with bioinformatic analyses, provided useful information highlighting previously unreported biochemical pathways and CSF-based biomarkers associated with both sporadic PD (sPD) and LRRK2 PD. Results of this metabolomics study further support our group’s previous findings identifying bile acid metabolism as one of the major aberrant biochemical pathways in PD patients. This study demonstrates that a combination of two complimentary techniques can provide a much more holistic view of the CSF metabolome, and by association, the brain metabolome. Future studies for the prediction of those at risk of developing PD should investigate the clinical utility of these CSF-based biomarkers in more accessible biomatrices. Further, it is essential that we determine whether the biochemical pathways highlighted here are recapitulated in the brains of PD patients with the aim of identifying potential therapeutic targets.

Résumé :
The article presents the results of biochemical studies of the total antioxidant activity of the hepatoprotective collection of plant origin and its individual components, which show that it consists of birch leaves (Betulae folia), tansy flowers (Tanaceti flores), phytohepatol No. 2 (choleretic collection No. 2) and individual its components have pronounced antioxidant activity. More antioxidant activity founded in tansy and true hepatoprotective collection. Comparative tests of the flowers of meadowsweet, with the highest antioxidant activity, showed the promise of their use in the collections of medicinal plants as effective natural metabolic regulators. The total antioxidant activity of the water extract collection at 13.56% rel. exceeded the calculated value obtained by summing the activities of the individual components of the collection, which indicates the presence of synergism in their action, which may be of value for use in the treatment and prevention of liver diseases. When all components of the drug collection are finished, the indicator increases by 11-24 % rel., which indicates the thermal stability of the antioxidant substances that make up their composition. In the oxidation of aqueous extracts of the components of hepatoprotective collection with hydrogen peroxide, the most stable were antioxidants of birch leaves. It was found that collection only at a ratio of 7.7 g per liter of water shows synergism (15 % rel.), and the rest of all the samples we tested are antagonistic. The detected increase in the antioxidant activity of the new collection compared with the effects of its individual components allows us to consider it as a promising dosage form for use in the treatment and prevention of liver diseases.

Résumé :
In the unstable conditions of the Anapo-Taman zone of the Krasnodar Kray, the issues of grape resistance to low tempera-tures in the winter period are of particular relevance. The results of a physiological and biochemical assessment of the winter hardiness of grape varieties (interspecific hybrids) of various ecological and geo-graphical origin are presented: Kristall (control – Euro-Amur-American origin; Dostoyny, Krasnostop AZOS – Euro-American origin; Vostorg – Amur-American origin; Aligote – Western-European origin, Zarif – East European origin. The studies were carried out on the basis of the ampelographic collection of the "North Caucasian Federal Scientific Center for Horticulture, Viticulture, Winemaking" (NCFSCHVW), located in the city of Anapa. Differences in the water content of the buds, the content starch, soluble sugars, proline, associated with the formation of a protective response to the meteorological conditions of the autumn-winter period 2020–2022. According to the degree of decrease in water content of the buds, the degree of starch hydrolysis, the dynamics of the accumulation of water soluble sugars in response to low temperatures in the win-ter period varieties Kristall, Krasnostop AZOS, Vostorg showed the greatest po-tential for resistance to low temperatures in the autumn-winter period of 2020–2022 compared to other varieties studied. Varie-ties Kristall, Krasnostop AZOS, Vostorg are recommended for cultivation in the Anapo-Taman zone of the Krasnodar Kray and for breeding purposes as sources of winter hardiness.
