Résumé :
Staphylococcus aureus is a major public health concern. Methicillin resistant Staphylococcus aureus is not only resistant to methicillin and other β-lactam antibacterial agents but also to other antibacterial agents. Therefore, new agents are needed to treat Methicillin Resistant Staphylococcus aureus (MRSA). The main aim of the present research was to study the antibacterial activity of four plants extract against clinical isolates of Staphylococcus aureus at the purpose of overcoming its infection. The current investigation was carried out at Pokhara Bigyan tatha Prabidhi Campus based on the evaluation of traditional plants on its antibacterial activity against MRSA. Antibacterial activity of the medicinal plant extract was observed by mixing 100 g powder of collected leaves in 70% methanol making 1000 sml. After performed processes, extract was filtered and methanol was evaporated. Antibacterial activity of the medicinal plant extract was determined using agar well diffusion assay method. Methanol leaf extracts may have the potential to act against MRSA and could be a possible source to obtain new and effective herbal medicines to treat infections caused by methicillin resistant strains of microorganisms from community as well as hospital settings. The synergistic effect was clearly observed among all four medicinal herbs.

Résumé :
Beetroot is notable for its sweetness, high sugar content, but is very low in calories. Despite its nutritional value, many people refuse to consume it because of its earthy flavor. So, the study was conducted in the laboratory of Pokhara Bigyan Tatha Prabidhi Campus from September 2020 to December 2020 to prepare sugar and honey-based beetroot candy and make a comparative study on their qualities. By following the preliminary operations, fresh beetroot was candied by using honey and sugar syrup of 30 °Brix. Within certain days of boiling and draining of syrup, a gradual increment of TSS was obtained up to 70 °Brix and was finally dried at 55±3 ℃. The moisture content (26.18±0.58), crude fat (0.52±0.01), crude protein (1.60±0.14), crude fiber (2.39±0.19), titratable acidity (0.23±0.05), pH (5.82±0.02), reducing sugar (38.07±0.28), color (6.7±0.24), flavor (6.3±0.18), texture (6.65±0.25), taste (7.10±0.22), appearance (7.17±0.27) and overall acceptability (7.10±0.24) was recorded more in honey-based candy while carbohydrate (70.01±0.76) and ash (1.75±0.13) was recorded more in sugar-based candy. The total soluble solid was obtained equally in both sugar and honey-based candy which was 70 °Brix. The honey-based candy was better in nutritional, chemical and sensory attributes so it is suggested to use honey as a syrup instead of sugar syrup.

Résumé :
AbstractThe widespread availability of large data-sets poses both an opportunity and a challenge to logic programming. A first approach is to couple a relational database with logic programming, say, a Prolog system with MySQL. While this approach does pay off in cases where the data cannot reside in main memory, it is known to introduce substantial overheads. Ideally, we would like the Prolog system to deal with large data-sets in an efficient way both in terms of memory and of processing time. Just In Time Indexing (JITI) was mainly motivated by this challenge, and can work quite well in many application.Exo-compilation, designed to deal with large tables, is a next step that achieves very interesting results, reducing the memory footprint over two thirds. We show that combining exo-compilation with Just In Time Indexing can have significant advantages both in terms of memory usage and in terms of execution time.An alternative path that is relevant for many applications is User-Defined Indexing (UDI). This allows the use of specialized indexing for specific applications, say the spatial indexing crucial to any spatial system. The UDI sees indexing as pluggable modules, and can naturally be combined with Exo-compilation. We do so by using UDI with exo-data, and incorporating ideas from the UDI into high-performance indexers for specific tasks.

Résumé :
Super-resolution image reconstruction has always been a popular research direction in the field of computer vision, which aims to recover high-resolution clear images from low-resolution images. Traditional super-resolution reconstruction algorithms mainly rely on the construction of constraints and the accuracy of registration between images to achieve the reconstruction effect, but their accuracy cannot meet the needs of practical applications with large multiples. Thanks to the rapid development of deep learning field, super-resolution image reconstruction based on deep learning has become the mainstream, and has achieved great success in reconstruction accuracy and speed. According to the different generative models used, the existing super-resolution image reconstruction methods mainly include two categories: GAN-based and VAEs-based. To quantitatively compare the limits of the two approaches' performance, this study selects two representative algorithms, BigGAN and VQ-VAE-2, and introduces the theoretical details and training process of these two methods, respectively. Furthermore, the reconstruction results of BigGAN and VQ-VAE-2 are further compared. Finally, this paper discusses the future development trend of super-resolution picture reconstruction with the current potential problems of BigGAN and VQ-VAE-2.

Résumé :
Background: Vascular injury induces the exposure of subendothelial extracellular matrix (ECM) important to serve as substrate for platelets to adhere to the injured vessel wall to avoid massive blood loss. Different ECM proteins are known to initiate platelet adhesion and activation. In atherosclerotic mice, the small, leucine-rich proteoglycan biglycan is important for the regulation of thrombin activity via heparin cofactor II. However, nothing is known about the role of biglycan for hemostasis and thrombosis under nonatherosclerotic conditions. Methods: The role of biglycan for platelet adhesion and thrombus formation was investigated using a recombinant protein and biglycan knockout mice. Results: The present study identified biglycan as important ECM protein for the adhesion and activation of platelets, and the formation of three-dimensional thrombi under flow conditions. Platelet adhesion to immobilized biglycan induces the reorganization of the platelet cytoskeleton. Mechanistically, biglycan binds and activates the major collagen receptor glycoprotein (GP)VI, because reduced platelet adhesion to recombinant biglycan was observed when GPVI was blocked and enhanced tyrosine phosphorylation in a GPVI-dependent manner was observed when platelets were stimulated with biglycan. In vivo, the deficiency of biglycan resulted in reduced platelet adhesion to the injured carotid artery and prolonged bleeding times. Conclusions: Loss of biglycan in the vessel wall of mice but not in platelets led to reduced platelet adhesion at the injured carotid artery and prolonged bleeding times, suggesting a crucial role for biglycan as ECM protein that binds and activates platelets via GPVI upon vessel injury.

Résumé :
Biglycan, a member of the small leucine-rich proteoglycan family, has been implicated in the development and progression of human cancers. However, the clinical significance of biglycan expression in gastric cancer has not been determined. In the present study, biglycan mRNA and protein concentrations were analyzed using quantitative realtime reverse transcription polymerase chain reaction and Western blot in 69 gastric cancer and adjacent non-tumorous tissues, respectively. Biglycan expression was further assessed using immunohistochemistry in tissue microarrays that contained 264 cases of gastric cancer, and others containing normal or metastasized lymph node tumor tissues. Biglycan was upregulated at the transcriptional and translational levels and there was a correlation between the expression of biglycan mRNA and protein ( P = 0.000, κ = 0.769). Over-expression of biglycan was strongly associated with lymph node metastasis, tumor (T) classification, metastasis (M) classification, vascular invasion and Union for International Cancer Control (UICC) stage. Patients with biglycan-positive tumors had a significantly higher disease recurrence rate and poorer survival than patients with biglycan-negative tumors after the radical surgery. Multivariate analysis revealed that biglycan expression is an independent prognostic indicator for survival of patients with gastric cancer. The data from the current study demonstrate that elevated expression of biglycan may play an important role in the development and progression of gastric cancer, and could be further evaluated as a biomarker for predication of a poor clinical outcome.

Johnson, Lisa Dawn. « Faculty Teaching Goals at Senior Research Universities ». Diss., Virginia Tech, 1997. http://hdl.handle.net/10919/30701.

Texte intégral
Résumé :
The purpose of this study was to identify faculty teaching goals at senior research universities and to determine variables which could be used to predict these goals. The eight disciplines used in this study were: accounting, chemistry, computer science, economics, English, math, mechanical engineering, and psychology. The independent variables were: gender, academic rank, course level, tenure, and four Biglan categories (pure-hard, pure-soft, applied-hard, and applied-soft). Using the Teaching Goals Inventory (Angelo & Cross, 1993), the teaching goals and primary teaching role of faculty at senior research universities across eight academic disciplines were identified (N = 352). Through a principal axis factor analysis, seven factors emerged for the data gathered from the Teaching Goals Inventory, results of which were slightly different from that of previous research. Further analyses indicated differences between the teaching goals and primary teaching role of faculty at senior research universities, community colleges, and four-year colleges. Seven one-way ANOVAs and subsequent post-hoc comparisons were conducted which indicated significant differences (p < .05) among the factor means across the eight academic disciplines. Comparisons of the primary teaching role across the eight academic disciplines also indicated significant differences (p < .05) in the percentage of faculty selecting each of the six roles. Furthermore, this study provides additional evidence to support the theory that the Biglan categories help explain the differences in teaching goals across academic disciplines. Significant differences (p < .05) were detected in the teaching goals and primary teaching role of senior research university faculty across the four Biglan categories. Through regression analyses, three of the four Biglan categories, gender, and level of course entered as predictors of teaching goals. Academic rank and tenure did not enter into any of the regression equations; however, further analyses indicated these variables were intercorrelated with several other independent variables. Implications for these findings are discussed.

Born in 1771 and died in 1841, Édouard Bignon embodies the generation who has live, with some trouble, through the French Revolution, the Directory, the Consulate, the First Empire, the Restoration and the July Monarchy. His career reflects all the French political changes. A private tutor, he enlisted in 1793 while trying to avoid being arrested. During the Directory, in 1797, he joined the Foreign Office and stayed in this departement until 1815 : he was constantly promoted, from the position of 2nd secretary in 1797 to minister plenipotentiary in Poland in 1813. As he joined the ranks of Napoleon during The Hundred Days, he had to retire from public life once Louis XVIII came back. Two years after Waterloo, he became a member of Parliament and sat among the liberals until the Bourbons fall. Separately from his political activities, he published several historical books which rose his popularity. In his will, Napoleon asked him to write the diplomatic history of his reign. Begun in 1821, the Histoire de France was issued from 1829 to 1850 and split into 14 volumes. In 1830, he contributed to the July Monarchy’s birth and became one of its support in Parliament : Louis-Philippe rewarded him and appointed him as peer of France in 1837, as a public recognition for his duties. This study which tackles all Bignon’s activities, aims to emphasize a medium scale diplomatic experience, parliamentarian’s occupations during the Restoration and the July Monarchy and Napoleon historian’s originality

Ramirez, Aviles Camila Alexandra. « P-bigon right-veeringness and overtwisted contact structures ». Diss., University of Iowa, 2017. https://ir.uiowa.edu/etd/5609.

Texte intégral
Résumé :
A contact structure is a maximally non-integrable hyperplane field $\xi$ on an odd-dimensional manifold $M$. In $3$-dimensional contact geometry, there is a fundamental dichotomy, where a contact structure is either tight or overtwisted. Making use of Giroux's correspondence between contact structures and open books for $3$-dimensional manifolds, Honda, Kazez, and Mat\'{i}c proved that verifying whether a mapping class is right-veering or not gives a way of detecting tightness of the compatible contact structure. As a counter-part to right-veering mapping classes, right-veering closed braids have been studied by Baldwin and others. Ito and Kawamuro have shown how various results on open books can be translated to results on closed braids; introducing the notion of quasi right-veering closed braids to provide a sufficient condition which guarantees tightness. We use the related concept of $P$-bigon right-veeringness for closed braids to show that given a $3$-dimensional contact manifold $(M, \xi)$ supported by an open book $(S, \phi)$, if $L \subset (M, \xi)$ is a non-$P$-bigon right-veering transverse link in pure braid position with respect to $(S, \phi)$, performing $0$-surgery along $L$ produces an overtwisted contact manifold $(M', \xi')$. Furthermore, if we suppose $L \subset (M, \xi)$ is a pure and non-quasi right-veering braid with respect to $(S, \phi)$, performing $p$-surgery along $L$, for $p \geq 0$, gives rise to an open book $(S', \phi')$ which supports an overtwisted contact manifold $(M', \xi')$.

Buchanan, Samuel Andrew. « The Effect of Biglycan on TNF-alpha Signaling and Innervation in the Intervertebral Disc ». Thesis, Rush University, 2016. http://pqdtopen.proquest.com/#viewpdf?dispub=1581897.

Résumé :

Biglycan is a small, leucine-rich repeat proteoglycan that has been shown to be elevated in the intervertebral discs (IVDs) of older adults (Cs-Szabo, 2002) and can interact with many different signaling molecules and structural proteins. TNF-alpha (TNF) is a general modulator of inflammation and may be a trigger in the chronic inflammatory response that leads to disc degeneration. Since biglycan has been shown to interact with TNFalpha (Tufvesson and Westergren-Thorsson, 2002), it may play a role in modulating its response in the IVD. Western (NFkB) and RT-PCR (MMP-1, -13, NGF and VEGF) assays were performed on both human and bovine cultured cells from IVD tissue. Treatment with biglycan alone was found to be almost indistinguishable from control groups; while biglycan in combination with TNF-alpha was able to significantly reduce transcription and expression response when compared with just TNF-alpha. Biglycan may help modulate the effects of TNF-alpha in the IVD.

D'Antoni, Michelle. « The effect of decorin and biglycan on human airway smooth muscle cell proliferation, apoptosis and attachment ». Thesis, McGill University, 2011. http://digitool.Library.McGill.CA:80/R/?func=dbin-jump-full&object_id=103624.

Texte intégral
Résumé :
Changes in extracellular matrix (ECM) deposition along with increases in airway smooth muscle (ASM) mass are characteristic features of airway remodeling which contribute to asthma pathophysiology. Modifications in ECM include increased collagen deposition and altered proteoglycans (PGs) levels. While biglycan, a small leucine-rich PG (SLRP), deposition is generally increased, changes in decorin, another SLRP, are more variable. The interaction between the cell and the surrounding ECM triggers numerous cellular responses, which modulate functions such as survival, proliferation and differentiation. Since decorin and biglycan are known to influence cell behaviour in various types of cells, we hypothesized that these molecules would affect human airway smooth muscle cell (HASMC) function. We sought to determine the effects of decorin and biglycan on HASMC proliferation, apoptosis and attachment, and to explore the putative mechanisms responsible for these effects. Collagen type I was used as a control.Culture on a decorin matrix caused a decrease in HASMC number, which was attributed to a decrease in proliferation and an increase in apoptosis. Culture on biglycan caused an initial decrease in cell number that was not sustained. Collagen caused a significant increase in cell number. Neither biglycan nor collagen caused changes in proliferation or apoptosis. Subsequent experiments showed that cell anchorage to decorin and biglycan matrices caused abnormal focal adhesion and cytoskeletal organization, as well as, increases in alpha2 integrin protein levels as compared to cells seeded on collagen or plastic. To assess integrin activation, focal adhesion kinase (FAK) protein levels and activation were measured. FAK levels were significantly decreased compared to cells cultured on plastic or collagen I, but activation levels were unchanged. These studies demonstrate that PGs have differential effects on HASMC growth. Whereas decorin reduces ASM proliferation and increases apoptosis, biglycan has no effect. Cell-matrix adhesions were irregular in HASMCs seeded on decorin or biglycan compared to cells on collagen I or plastic. Decreases in decorin in the asthmatic airway wall, as observed in fatal asthma cases, may permit more exuberant ASM hyperplasia, suggesting that decorin's presence could serve a protective role by modulating the increase in ASM mass in asthma.
