A majority of commercial rose varieties bloom repeatedly throughout the year, as compared to most rose species, other woody ornamentals, and fruit crops that bloom once a year. This recurrent flowering feature of the commercial roses resulted from a flowering mutation named everblooming (evb). The mutation is recessive to once blooming and is found in the rose species Rosa chinensis. Although several molecular maps have been developed for rose, little is known about the evb gene, except for its classic genetics. The purpose of this study was to develop a large-insert bacterial artificial chromosome (BAC) library as a starting tool for molecular cloning and analysis of the evb gene by map-based cloning. To construct the large-insert BAC library, nuclear megabase-size DNA was isolated from the recurrent blooming diploid species, Rosa chinensis cv. Old Blush. The DNA was then partially digested with BamHI and separated on agarose gels by multi-phase pulsed-field gel electrophoresis. Size selected fragments estimated between 100 kb and 150 kb in size were cloned into the pECBAC1 BAC vector and the clones having rose DNA inserts were arrayed in 80 384-well microplates individually, with each clone being barcoded. The library contains 30,720 clones, has an average insert size of 108 kb and covers roughly 5.9x genome equivalents, with a >99% probability of isolating a single-copy clone from the library. The library is now available to be screened with the genes cloned from other species that control vernalization and floral development and will be used in mapbased cloning of the evb gene using a Rosa wichuraiana (‘Basye’s Thornless’) x ‘Old Blush’ backcross population.

Résumé :
The CHRNA5/A3/B4 genomic cluster encodes for the alpha5, alpha3 and beta4 subunits of the nicotinic acetylcholine receptors (nAChRs). Human genetic studies have revealed a significant association of variants in this genomic region with nicotine dependence. However, the mechanisms through which overexpression of these three subunits may influence smoking-related behaviours is not understood. To gain insight in the possible mechanisms, we used a BAC transgenic mouse model overexpressing this cluster containing the three genes together with their transcriptional regulatory elements. We found that overexpression of the cluster: i) increases sensitivity to the pharmacological effects of nicotine; ii) modifies particular cognitive domains associated to drug addiction and hippocampal neuronal complexity and synaptic plasticity; and iii) shifts the rewarding and aversive properties of nicotine and the manifestation of nicotine-withdrawal syndrome. Our study suggests that the genomic cluster CHRNA5/A3/B4 contributes to genetic vulnerability to nicotine addiction and promotes smoking-related behaviours possibly through hippocampal plasticity changes.
El cluster genòmic CHRNA5/A3/B4 codifica per les subunitats alfa5, alfa3 i beta4 dels receptors d’acetilcolina (nAChRs). Estudis de genètica humana han revelat que variants en aquesta regió genòmica estan significativament associats a la dependencia a nicotina. Malauradament, els mecanismes pels quals la sobreexpressió d’aquestes tres subunitats influencia comportaments relacionats amb el consum de tabac no són del tot coneguts. Per tal d’entendre els possibles mecanismes, hem utilitzat un model de ratolí transgènic que sobreexpressa aquest cluster amb els tres gens i les seus elements de regulació transcripcional. Hem trobat que la sobreexpressió del cluster: i) incrementa la sensibilitat als efectes farmacològics de la nicotina; ii) modifica determinats dominis cognitius associats a l’addicció a droges i la complexitat neuronal i plasticitat sinàpica de l’hipocamp; a més a més iii) canvia les propietats de recompensa i aversió de la nicotina i la manifestació del síndrome d’abstinència. El nostre estudi suggereix que el cluster genòmic CHRNA5/A3/B4 contribueix a la vulnerabilitat genètica a l’adicció a la nicotina i promou comportaments relacionats amb el consum de tabac possiblement a través de canvis de plasticitiat a l’hipocamp.

BACKGROUND:Brachypodium distachyon (Brachypodium) has been recognized as a new model species for comparative and functional genomics of cereal and bioenergy crops because it possesses many biological attributes desirable in a model, such as a small genome size, short stature, self-pollinating habit, and short generation cycle. To maximize the utility of Brachypodium as a model for basic and applied research it is necessary to develop genomic resources for it. A BAC-based physical map is one of them. A physical map will facilitate analysis of genome structure, comparative genomics, and assembly of the entire genome sequence.RESULTS:A total of 67,151 Brachypodium BAC clones were fingerprinted with the SNaPshot HICF fingerprinting method and a genome-wide physical map of the Brachypodium genome was constructed. The map consisted of 671 contigs and 2,161 clones remained as singletons. The contigs and singletons spanned 414 Mb. A total of 13,970 gene-related sequences were detected in the BAC end sequences (BES). These gene tags aligned 345 contigs with 336 Mb of rice genome sequence, showing that Brachypodium and rice genomes are generally highly colinear. Divergent regions were mainly in the rice centromeric regions. A dot-plot of Brachypodium contigs against the rice genome sequences revealed remnants of the whole-genome duplication caused by paleotetraploidy, which were previously found in rice and sorghum. Brachypodium contigs were anchored to the wheat deletion bin maps with the BES gene-tags, opening the door to Brachypodium-Triticeae comparative genomics.CONCLUSION:The construction of the Brachypodium physical map, and its comparison with the rice genome sequence demonstrated the utility of the SNaPshot-HICF method in the construction of BAC-based physical maps. The map represents an important genomic resource for the completion of Brachypodium genome sequence and grass comparative genomics. BACKGROUND:Sugarcane breeding has significantly progressed in the last 30 years, but achieving additional yield gains has been difficult because of the constraints imposed by the complex ploidy of this crop. Sugarcane cultivars are interspecific hybrids between Saccharum officinarum and Saccharum spontaneum. S. officinarum is an octoploid with 2n=80 chromosomes while S. spontaneum has 2n=40 to 128 chromosomes and ploidy varying from 5 to 16. The hybrid genome is composed of 70-80%S. officinaram and 5-20%S. spontaneum chromosomes and a small proportion of recombinants. Sequencing the genome of this complex crop may help identify useful genes, either per se or through comparative genomics using closely related grasses. The construction and sequencing of a bacterial artificial chromosome (BAC) library of an elite commercial variety of sugarcane could help assembly the sugarcane genome.RESULTS:A BAC library designated SS_SBa was constructed with DNA isolated from the commercial sugarcane variety SP80-3280. The library contains 36,864 clones with an average insert size of 125 Kb, 88% of which has inserts larger than 90 Kb. Based on the estimated genome size of 760-930 Mb, the library exhibits 5-6 times coverage the monoploid sugarcane genome. Bidirectional BAC end sequencing (BESs) from a random sample of 192 BAC clones sampled genes and repetitive elements of the sugarcane genome. Forty-five per cent of the total BES nucleotides represents repetitive elements, 83% of which belonging to LTR retrotransposons. Alignment of BESs corresponding to 42 BACs to the genome sequence of the 10 sorghum chromosomes revealed regions of microsynteny, with expansions and contractions of sorghum genome regions relative to the sugarcane BAC clones. In general, the sampled sorghum genome regions presented an average 29% expansion in relation to the sugarcane syntenic BACs.CONCLUSION:The SS_SBa BAC library represents a new resource for sugarcane genome sequencing. An analysis of insert size, genome coverage and orthologous alignment with the sorghum genome revealed that the library presents whole genome coverage. The comparison of syntenic regions of the sorghum genome to 42 SS_SBa BES pairs revealed that the sorghum genome is expanded in relation to the sugarcane genome.

Ratos adultos, machos, da linhagem Wistar foram distribuídos, por sorteio, em três grupos experimentais pareados: Grupo controle - constituído de 20 animais normais, sadios, não diabéticos; Grupo diabético - constituído de 20 animais diabéticos induzidos pela aloxana, não tratados; Grupo desnutrido - constituído de 20 animais desnutridos por restrição alimentar. Cada um destes três grupos experimentais foi subdividido, por sorteio, em dois subgrupos de ratos, compostos por 10 animais cada um, para serem avaliados e sacrificados em 2 momentos distintos do experimento, 1 mês e 6 meses de seguimento. As cirurgias de implantação do conjunto eletrodo/marcador magnético na serosa gástrica e do cólon, para obtenção dos registros elétricos e mecânicos, foram realizadas respectivamente após 1 mês e 6 meses de seguimento normal, diabético e de desnutrição. BACKGROUND:Molecular markers serve three important functions in physical map assembly. First, they provide anchor points to genetic maps facilitating functional genomic studies. Second, they reduce the overlap required for BAC contig assembly from 80 to 50 percent. Finally, they validate assemblies based solely on BAC fingerprints. We employed a six-dimensional BAC pooling strategy in combination with a high-throughput PCR-based screening method to anchor the maize genetic and physical maps.RESULTS:A total of 110,592 maize BAC clones (~ 6x haploid genome equivalents) were pooled into six different matrices, each containing 48 pools of BAC DNA. The quality of the BAC DNA pools and their utility for identifying BACs containing target genomic sequences was tested using 254 PCR-based STS markers. Five types of PCR-based STS markers were screened to assess potential uses for the BAC pools. An average of 4.68 BAC clones were identified per marker analyzed. These results were integrated with BAC fingerprint data generated by the Arizona Genomics Institute (AGI) and the Arizona Genomics Computational Laboratory (AGCoL) to assemble the BAC contigs using the FingerPrinted Contigs (FPC) software and contribute to the construction and anchoring of the physical map. A total of 234 markers (92.5%) anchored BAC contigs to their genetic map positions. The results can be viewed on the integrated map of maize 1,2].CONCLUSION:This BAC pooling strategy is a rapid, cost effective method for genome assembly and anchoring. The requirement for six replicate positive amplifications makes this a robust method for use in large genomes with high amounts of repetitive DNA such as maize. This strategy can be used to physically map duplicate loci, provide order information for loci in a small genetic interval or with no genetic recombination, and loci with conflicting hybridization-based information.

I sang the cantata " Ich will den Kreuzstab gerne tragen" BWV 56 at my graduate´s concert.

Alcohol use and abuse among college/university students continues as a major public health concern.  One potential source of alcohol-related harm is the inability of students to estimate their current level of intoxication (gBAC).  The current four field studies use breathalyzers to investigate student gBAC and the efficacy of a variety of BAC-feedback tools at promoting greater awareness of driving risks while under the influence of alcohol.     The research was conducted across 89 nights spanning seven academic semesters from Fall 2009 to Fall 2012.  Research tables were setup between the hours of 6:00pm and 2:00am at three locations near downtown bar establishments and one on-campus location near a late-night dining facility.  Unique subject codes were created to track participants across multiple nights of participation.  In total, 12,432 blood alcohol concentration (BAC) readings were collected from 10,225 unique individuals. Study 1 examined general epidemiology across all nights.  The average BAC of drinking participants was .100 mL/L.  Results revealed significant differences in BAC as a function of demographic and environmental factors.  Additionally, it was found the average student was incorrect in estimating his or her BAC by .034 mL/L. Studies 2 and 3 examined the accuracy of BAC-estimation tools (i.e., nomograms, sobriety tests, and phone applications) and the efficacy of these tools to increase awareness of driving-related risks.  On average, both nomograms and BAC-estimation phone applications  
were incorrect in estimating BAC by over .05 mL/L.  Sobriety tests performed slightly better than chance at discriminating BACs of .08 mL/L.  
Participants receiving BAC-feedback had increased awareness of driving risk across levels of intoxication. Nomogram and breathalyzer feedback tended to promote healthier perceptions of external risk.  Sobriety testing shifted the internal perception of feeling less safe to drive.  No effect was observed for BAC-estimation phone applications. Study 4 found individuals who received breathalyzer feedback across multiple nights of the research were significantly more accurate at estimating their BAC.  Specifically, individuals on the fifth night of participation were .017 mL/L more accurate at estimating their BAC as compared to the first night.  Future research areas and policy implications are discussed.

PURPOSE: To examine differences in mean power output between high bar (HBS) and low bar back squats (LBS). METHODS: Six trained males (25.0 ± 3.1 years, 1.78 ± 0.04 m, 87.6 ± 7.5 kg) with previous squatting experience (experience: 7.5 ± 4.1 years, HBS 1RM: 157.0 ± 15.3 kg, squat/bodyweight: 1.8 ± 0.18) completed the study using a crossover design. Subjects completed a 4-week familiarization phase with both conditions. Mean power data was collected over 2 sessions using dual uniplanar force plates and 4 linear position transducers sampling at 1,000 Hz. Subjects were randomly assigned to the HBS or LBS for 1 set of 3 repetitions at 20, 30, 40, 50, 60, 70, 80, and 90% of their most recent HBS training 1RM with 3 to 5 minutes’ rest between sets and 2-7 days between testing conditions. A 2x8 repeated measures analysis of variance was used to determine interactions and main effects for condition and load with post-hoc tests conducted for statistical main effects. RESULTS: Analysis revealed significant main effects for load (p < 0.01) but not for condition. CONCLUSIONS: According to this pilot data, athletes seeking to increase power production ability should choose a squatting style in which they feel most proficient and comfortable. Furthermore, either the HBS or LBS can be used as the primary squatting movement, or as a secondary movement to provide variation. However, based on previous research it is likely that sport specific biomechanical parameters will influence the squatting style selection for the majority of athletes who participate in sports that involve jumping, sprinting, and change of direction.

O nematóide de galhas, Meloidogyne exigua, parasita o cafeeiro (Coffea sp.) e sua erradicação das regiões infestadas é impraticável. Estudos anteriores mostraram que a resistência a M. exigua é controlada por um gene de herança simples, designado Mex-1, presente em C. canephora. Linhagens de café arábica apresentando resistência têm sido desenvolvidas em programas de melhoramento genético por meio da introgressão deste gene. Uma biblioteca BAC de café foi avaliada inicialmente utilizando-se três marcadores (Exi-2, Exi-3 e Exi-4) ligados a Mex-1, obtidos a partir de marcadores AFLP previamente clonados. Os clones positivos associados à região Mex-1 foram identificados e isolados. O DNA destes clones foi então analisado por hibridização e amplificação com marcadores SCAR (Exi-2 e Exi- 3) e 9 combinações de primers das extremidades BAC. Os clones BAC positivos, relativos aos marcadores Exi-2, Exi-3 e Exi-4, foram reunidos em 5 contigs. O primeiro contig apareceu composto de dois clones BAC identificados com o marcador Exi-4. Dos cinco clones BAC identificados com o marcador Exi-2, dois contigs designados Exi-2 I e Exi-2 II foram distintos. Os clones BAC Exi-3 foram organizados em dois contigs distintos chamados Exi-3 I e Exi-3 II. No total, nove sítios de restrição ou de PCR, correspondendo a sete marcadores físicos, foram localizados na região Exi-3. As três regiões cromossômicas correspondentes ao contig Exi-4, aos dois contigs Exi-2 e aos dois contigs Exi-3 não apresentaram sobreposição. A presença de clones BAC contendo análogos de gene de resistência (RGA) na região Mex-1 foi investigado por meio de amplificação com primers degenerados e hibridização com sondas RGA de café. Alguns clones BAC contendo RGAs foram identificados. Para confirmar a introgressão do gene Mex-1, vinte e uma linhagens de Icatu foram comparadas a três testemunhas resistentes, Iapar 59, Híbrido de Timor e Sarchimor e a uma testemunha susceptível, Catuaí. Foi avaliada, por meio de marcadores AFLP, a presença do fragmento introgredido associado ao locus Mex-1. Nove marcadores AFLP foram amplificados. Destes, os marcadores Exi-1, Exi-2, Exi-3, Exi-11 e Exi-13 confirmaram a presença do fragmento associado à resistência a M. exigua, mostrando que esta marca pode ser utilizada em programas de seleção assistida. Uma biblioteca de cDNA de Caturra foi utilizada para se isolar fragmentos de genes associados à resistência, através da hibridização com sondas RGA. Após a utilização das nove famílias RGA de café, 32 colônias foram isoladas e seqüenciadas. Destas, 25 seqüências não apresentaram similaridade com outras seqüências disponíveis na base de dados do NCBI ou não puderam ser seqüenciadas. Seis amostras continham seqüências muito pequenas e não confiáveis, e apenas uma amostra resultou em uma seqüência de 428 nucleotídeos (142 aminoácidos). Esta seqüência apresentou alta homologia a catalases de outras plantas como Nicotiana plumbaginifolia, Glycine max, Phaseolus vulgaris, entre outras. A esta catalase descoberta em café foi dado o nome de Cat-C. A catalase de ligação ao ácido salicílico (SR1) e a seqüência parcial de aminoácidos da proteína de ligação ao ácido salicílico (SABP), ambos de N. tabacum, apresentaram 82% e 77% de identidade, respectivamente, quando comparadas a Cat- C. Na região C-terminal desta catalase foi detectada a presença de uma seqüência peroxissomal. No entanto, ao invés de uma seqüência SRL (Serina-Arginina- Leucina), foi encontrada uma seqüência TRL (Treonina-Arginina-Leucina). Esta diferença é devido à troca de uma timina por uma adenina. Estes dados sugerem que Cat-C pode estar associado ao mecanismo de resistência em café.
The root-knot nematode Meloidogyne exigua parasites the arabica coffee tree (Coffea sp.) and its eradication of the infested areas is impracticable. Previous studies showed that the resistance to M. exigua is controlled by a simple inheritance gene, designated Mex-1, present in C. canephora. Lineages of resistant Arabica coffee have been developed in some genetic breeding programs through the introgression of this gene. The BAC library of coffee was initially evaluated by three markers (Exi-2, Exi-3 and Exi-4) linked to Mex-1, previously obtained from cloned AFLP markers. The positive clones associated with Mex-1 region were identified and isolated. DNA clones were then analyzed by hybridization and amplification with SCAR (Exi-2 and Exi-3) markers and 9 combinations of primers of the BAC-end. The positive BAC clones, relative to the Exi-2, Exi-3 and Exi-4 markers, were assembled in 5 contigs. The first contig identified with the Exi-4 marker is composed of two positive BAC clones. Of the five BAC clones identified with Exi-2 markers, two contigs designated Exi-2 I and Exi-2 II were different from each other. The BAC clones Exi-3 were organized in two different contigs called Exi-3 I and Exi-3 II. In the total, nine restriction sites or PCR, corresponding to seven physical markers were located in the Exi-3 region. The three chromosomal regions that corresponded to the Exi-4 contig, the two Exi-2 contigs and the two Exi-3 contigs did not present any superposition. The presence of BAC clones containing resistance gene analogs (RGA) in the Mex-1 region was investigated by both PCR amplification with degenerate primers, and hybridization with coffee RGA probes. Some BAC clones containing RGA were identified. To confirm the introgression of Mex-1 gene, twenty-one Icatu lineages xiiiwere compared to three resistant cultivars, Iapar 59, Hibrido de Timor and Sarchimor and to a susceptible cultivar Catuaí, through AFLP markers, evaluating the presence or not of the introgressed fragment associated with the Mex-1 locus. Nine AFLP markers were amplified. Among these, Exi-1, Exi-2, Exi-3, Exi-11 and Exi-13 markers confirmed the presence of the fragment associated to the resistance to Meloidogyne exigua, showing that this marker can be used in selected assistent programs. A cDNA library of Caturra was used to isolate fragments of genes associated with the resistance, through the hybridization using RGA probes. After the use of the nine coffee RGA families, 32 colonies were isolated and sequenciated. Among these, 25 sequences either did not present similarity with other available sequences in the NCBI data base or could not be sequenced. Six samples contained very small and not trustable sequences, and just one sample resulted in a sequence of 428 nucleotides (142 aminoacids). This sequence presented high homology to catalases of other plants such as Nicotiana plumbaginifolia, Glycine max, Phaseolus vulgaris, etc. To this discovered coffee catalase was given the name of Cat-C. The salicylic acid receptor (SR1) and the partial sequence of aminoacids of the salicylic acid binding protein (SABP), both in Nicotinana tabacum, showed 82% and 77% of identities, respectively, when compared with Cat-C. In the C-terminal region of this catalase was detected the presence of a peroxisomal sequence. However, instead of a SRL sequence (Serine-Arginine-Leucine), a TRL sequence was found (Treonine- Arginine-Leucine). This difference is due to the change of a timine by an adenine. These data suggest that Cat-C can be associated with resistance mecanism in coffee.

Apoptosis, or programmed cell death, is an essential process in all multi-cellular organisms. It is indispensable to an organismâ s survival, preventing the malicious propagation of DNA damage and pathogenic alterations, through the clean disposal of afflicted cells. The BAD/tBID/BAK pathway is a portion of the apoptosis molecular pathway, albeit an important pathway since it is known to be deregulated and lead to pathological ailments such as cancer. Using chemical kinetics the BAD/tBID/BAK signaling pathway is modeled as a set of (nonlinear) ordinary differential equations. A first-cut numerical analysis reveals a mechanism where BAD sensitizes a switch from tBID activation to BAK activation. The phosphorylation of BAD is shown to inhibit this sensitizing effect. All behaviors are supported by experimental data, thereby validating the model of the BAD/tBID/BAK pathway. Moreover, modeling the phosphorylation of BAD as one of two modes, some conflicting experimental data about BADâ s phosphorylation can be disentangled. Parameter values (in this case the kinetic rate constants) are prone to error or missing altogether. Chemical reaction network theory, however, provides a theoretical approach to complement the initial numerical analysis without having to specify rate constant values. We extend the global asymptotic stability and robustness results in [92] to include any complex-balanced mass-action network. This enables us to study the BAD/tBID/BAK signaling network by breaking it into two sub-networks: one with BAD and tBID, and the other with tBID and BAK. The complex-balanced BAD/tBID sub-network is shown to possess a unique steady state which is globally asymptotically stable. This verifies the simple and dynamically well-behaved exchange of BAD for Bcl-2 proteins which guard against tBID activation. The second sub-network, tBID/BAK, is formulated as a complex-balanced network with a perturbation representing the reaction of tBID catalyzing the activation of BAK. Our theoretical results produce a non-conservative, though state-dependent, condition which can be used to prove global convergence to a neighborhood of the unperturbed steady state. We then illustrate the biological importance of the result for tBID/BAK sub-network with an example design for a drug delivery system.

Este trabalho teve como objetivo principal implementar um algoritmo empírico para o monitoramento do processo de eutrofização da Baía de Guanabara (BG), Rio de Janeiro (RJ), utilizando dados de clorofila-a coletados in situ e imagens de satélite coletadas pelo sensor MERIS, a bordo do satélite ENVISAT, da Agência Espacial Européia (ESA). Para a elaboração do algoritmo foi utilizada uma série histórica de clorofila-a (Out/2002 a Jan/2012) fornecida pelo Laboratório de Biologia Marinha da UFRJ, que, acoplada aos dados radiométricos coletados pelo sensor MERIS em datas concomitantes com as coletas in situ de clorofila-a, permitiu a determinação das curvas de regressão que deram origem aos algorítmos. Diversas combinações de bandas foram utilizadas, com ênfase nos comprimentos de onda do verde, vermelho e infra-vermelho próximo. O algoritmo escolhido (R = 0,66 e MRE = 77,5%) fez uso dos comprimentos de onda entre o verde e o vermelho (665, 680, 560 e 620 nm) e apresentou resultado satisfatório, apesar das limitações devido à complexidade da área de estudo e problemas no algoritmo de correção atmosférica . Algorítmos típicos de água do Caso I (OC3 e OC4) também foram testados, assim como os algoritmos FLH e MCI, aconselhados para águas com concentrações elevadas de Chl-a, todos com resultados insatisfatório. Como observado por estudos pretéritos, a Baia de Guanabara possui alta variabilidade espacial e temporal de concentrações de clorofila-a, com as maiores concentrações no período úmido (meses: 01, 02, 03, 10, 11 12) e nas porções marginais (~ 100 mg.m-3), particularmente na borda Oeste da baia, e menores concentrações no período seco e no canal principal de circulação (~ 20 mg.m-3). O presente trabalho é pioneiro na construção e aplicação de algoritmos bio-óptico para a região da BG utilizando imagens MERIS. Apesar dos bons resultados, o presente algorítmo não deve ser considerado definitivo, e recomenda-se para trabalhos futuros testar os diferentes modelos de correção atmosférico para as imagens MERIS.
This work aimed to implement an empirical algorithm for monitoring the process of eutrophication at Guanabara Bay (BG), Rio de Janeiro (RJ), using in situ chlorophyll-a data and satellite images by MERIS sensor, onboard ENVISAT satellite, from European Space Agency (ESA). A time series of chlorophyll-a (Dec / Jan 2002/2012) provided by Marine Biological Laboratory from UFRJ, was used to elaborate the algorithm, coupled with the radiometric data collected by MERIS sensor on concurrent dates with the collections, what allowed the determination of the regression curves that gave rise to algorithms. Several band combinations were used, with emphasis on wavelengths of green, red and near infrared. The algorithm chosen (R = 0.66 and SRM = 77.5%) made use of wavelengths between green and red (665, 680, 560 and 620 nm) and showed satisfactory results, despite the limitations, due to the complexity of the study area and problems in atmospheric correction algorithm. Typical algorithms water Case I (OC3 and OC4) were also tested, as well as FLH MCI and algorithms suggested for water with high concentrations of Chl-a, all with unsatisfactory results. As noted by past studies, Guanabara Bay has high spatial and temporal variability of chlorophyll-a concentrations, with the highest concentrations in the rainy seasons (months: 01, 02, 03, 10, 11, 12) and in the marginal portions (~ 100 mg.m-3), particularly in the western edge of the bay, and lower concentrations in the dry season and in the main circulation channel (~ 20 mg.m-3). This study is a pioneer in the construction and application of bio-optical algorithms for the region of BG using MERIS images. Despite the good results, the algorithm should not be considered definitive, and it is recommended for future work to test different models of atmospheric correction for MERIS images.

This study investigated the utility of the MMPI-A short form described by Archer, Tirrell, and Elkins (2001) for detecting the presence of emotionality in adolescents in the school setting. Students were placed in one of three groups based on their performance on an established and frequently used self-report measure of personality, the Behavior Assessment System for Children-II (BASC- 2). Subjects who had significant elevations on one or more of the scales in Internalizing Index on the BASC-2 were placed in the Clinical group and subjects who had significant elevations on one or more of the scales the School Problems Index or Personal Adjustment Index were placed in the Adjustment group. Those without significant elevations on the BASC-2 were placed in the Nonclinical group. Differences between the three groups on each of the MMPI-A short form clinical scales were reported. The results indicated that the students in the Clinical group scored higher than students in the Non-clinical group on each of the MMPI-A short form scales. Adjustment group scores tended to be higher than Non-clinical group scores but not all scales were significantly higher. Discriminant analysis correctly classified 75% of the non-clinical group, 52% of the Clinical group, but only 37% of the Adjustment group. These findings, combined with additional analysis of clinical relevant data, provided positive indicators supporting the use of the short form in clinical settings.

Polyhedral oligomeric silsesquioxanes (POSS) have attracted substantial academic interest for many years as hybrid materials and nanofillers for controlling thermal and mechanical properties, and for providing thermal and chemical resistance while retaining ease of processing. A natural extension of these studies has been working on POSS-based amphiphiles and thin film coatings. Studies at the air/water (A/W) interface have shown that trisilanol-POSS derivatives are amphiphilic and form uniform Langmuir films, whereas closed-cage POSS derivatives are hydrophobic and aggregate. In this study, two novel POSS derivatives were synthesized from PSS-(3-hydroxypropyl)-heptaisobutyl substituted (POSS-OH) and completely characterized. Weisocyanate reacted with POSS-OH, and a POSS-based triester (POSS-triester) was obtained. Trifluoroacidolysis of the POSS-triester at room temperature afforded the corresponding triacid (POSS-triacid). Purified POSS-OH, POSS-triester, and POSS-triacid were studied by using surface pressure - area per molecule (? -A) isotherms as well as Brewster angle microscopy (BAM) at the air/water (A/W) interface. Compared with previous work on trisilanol-POSS derivatives, the results indicated that POSS-triester was surface active and formed a liquid-expanded (LE) monolayer. In contrast, POSS-triacid monolayers were more condensed (LC) and were not dramatically affected by changes in pH. Results for the lift-off areas (Alift-off), limiting areas (A0), collapse areas (Ac), and collapse pressures (? c) of POSS-OH, POSS-triester, and POSS-triacid were compared to trisilanolisobutyl-POSS (TiBP) and were interpreted in terms of possible molecular conformations. Whereas, TiBP has been hypothesized to exist in a vertex-on conformation, POSS-OH and POSS-triacid packing at the A/W interface was consistent with face-on conformations. For POSS-triester, the isotherm was consistent with a vertex-on conformation at low ? , but a face-on conformation at high ? .

The work presents the main concepts of the wind power system and wind turbine technology based on synchronous generators connected to the grid through a Back-to-Back converter. A mathematical modeling of the main components of the generation system is developed in this work as well as design of the controllers used to operate the system. The proposed wind generation system is constructed in the laboratory as well as new control platform based on a oating-point hardware to allow experimental testing. The wind turbine is emulated by an induction motor mechanically coupled to the synchronous generator. In this paper we used classical control techniques PI type in synchronous reference and non-linear based on passivity. A theoretical study on the control laws Interconnection and Damping Assignment Passivity-based Control - IDA-PBC in the application context is held and the controller design is presented. The in uence of the IDA-PBC controller parameters is also discussed. Maximum power tracking of the wind generator techniques are developed for simulation and practical system. Finally the simulated and experimental results for the developed system are presented and discussed.
O trabalho de dissertação apresenta as principais conceitos do sistema de geração eólica e a tecnologia de aerogerador baseada no geradores síncronos conectados á rede elétrica através de um conversor Back-to-Back. Uma modelagem matemática dos principais componentes do sistema de geração é desenvolvida neste trabalho assim como o projeto dos controladores utilizados para comandar o sistema. O sistema de geração eólica proposto é construído em laboratório assim como a nova plataforma de controle baseado em uma hardware de ponto utuante para permitir a realização de teste experimentais. A turbina eólica é representada por um motor de indução mecanicamente acoplado ao gerador síncrono. Neste trabalho são utilizadas técnicas de controle clássicas do tipo PI no referencial síncrono e n~ao-linear baseado em passividade. Um estudo téorico sobre as leis de controle Interconnection and Damping Assignment Passivity-based Control IDA-PBC no contexto da aplicação é realizado e o projeto dos controlador é apresentado e a in u^encia dos parâmetros do controlador IDA-PBC é discutida. Técnicas de rastreamento de máxima potência do gerador eólico são desenvolvidas para o sistema simulado e prático. Por m os resultados simulados e experimentais para o sistema desenvolvido são apresentados e discutidos.

Polyhedral oligomeric silsesquioxanes (POSS) have attracted substantial academic interest for many years as hybrid materials and nanofillers for controlling thermal and mechanical properties, and providing thermal and chemical resistance while retaining ease of processing. A natural extension of these studies has been POSS-based amphiphiles and thin film coatings. Studies at the air/water (A/W) interface have shown that trisilanol-POSS derivatives are amphiphilic and form uniform Langmuir films, whereas closed-cage POSS derivatives are hydrophobic and aggregate. In previous work, a triester (POSS-triester) and a triacid (POSS-triacid) were synthesized from PSS-(3-hydroxypropyl)-heptaisobutyl (POSS-OH) and Weisocyanate and fully characterized by surface pressure – area per molecule (Π-A) isotherm and Brewster angle microscopy (BAM) studies at the A/W interface. The results indicated that POSS-triester is surface active forming a liquid expanded (LE) monolayer, whereas POSS-triacid forms a liquid condensed (LC) monolayer that is only weakly affected by pH. A face-on conformation was proposed and examined to understand the packing of POSS-based amphiphilic molecules at the A/W interface. The face-on/vertex-on comparison is rarely discussed for Langmuir monolayers at the A/W interface. In this thesis, three other POSS-based esters were synthesized from POSS-OH and aminopropylisobutyl-POSS (POSS-NH₂) using Weisocyanate and a similar isocyanate containing two tert-butyl protected carboxylic acids. The synthesized materials are characterized by Π-A isotherm and BAM. For POSS-OH based diester (PAlDE) and POSS-NH2 based diester (PAmDE), LE/LC phase transitions were observed in Π-A isotherms over part of the experimentally accessible temperature range and were attributed to a change from a vertex-on to face-on conformation. Apparent BAM images confirmed LC islands coexisted with the LE phase. The experimentally observed dynamic estimates of the critical temperatures (Tc) were estimated from a two-dimensional Clausius-Clapeyron analysis and were consistent with the temperature dependence of the Π-A isotherms. These LE/LC phase transitions are the first observed for POSS amphiphiles.

This paper investigates the nature of the attraction of XPs to clauseinitial position in German (and other languages). It argues that there are two different types of preposing. First, an XP can move when it is attracted by an EPP-like feature of Comp. Comp can, however, also attract elements that bear the formal marker of some semantic or pragmatic (information theoretic) function. This second type of movement is driven by the attraction of a formal property of the moved element. It has often been misanalysed as “operator” movement in the past. Japanese wh-questions always exhibit focus intonation (FI). Furthermore, the domain of FI exhibits a correspondence to the wh-scope. I propose that this phonology-semantics correspondence is a result of the cyclic computation of FI, which is explained under the notion of Multiple Spell-Out in the recent Minimalist framework. The proposed analysis makes two predictions: (1) embedding of an FI into another is possible; (2) (overt) movement of a wh-phrase to a phase edge position causes a mismatch between FI and wh-scope. Both predictions are tested experimentally, and shown to be borne out.

Le but de ce travail était de tester l'effet nutritionnel de la vitamine A et des AGPI (dont l'EPA et le DHA) sur le développement des larves de bar. Un excès de vitamine A dans l'aliment provoquait des malformations au niveau de la tête reliées à une hausse de la transcription de RAR? et la perte d'une vertèbre lorsque les larves étaient nourries avec cet aliment de J8 à J13. Un aliment contenant un excès d'AGPI perturbait le développement de la colonne vertébrale des bars, les malformations étaient corrélées à une chute de l'expression de RARa, RAR?, RXRa et BMP4 et une vertèbre surnuméraire apparaissait sur les larves nourries de J8 à J13 avec cet aliment. Un aliment contenant 31 mg/kg de matière sèche de vitamine A et entre 1. 1 et 2. 3% d'EPA + DHA permet un développement harmonieux des larves. Il apparaît que la période J8-J13 est cruciale pour le développement, période pendant laquelle les larves sont très sensibles à la nature et à la teneur des nutriments

Although the pro-apoptotic BCL-2 family proteins BAK and BAX play a key role in mitochondrial perturbation their transition from an inactive closed conformation to a membrane permeabilising pore remains unclear. I found that BAK in viable cells existed in a primed state which was characterized by an occluded N-terminus and an exposed BH3 domain. This conformation facilitated binding to the hydrophobic groove of BCL-XL and served as a checkpoint maintaining cell survival by preventing its further activation. Isolation of BAK by immunoprecipitation suggests that only a discrete portion is present in this primed conformation. Reconstitution of the BCL-XL BAK complex into a BAK/BAX null background rendered cells more sensitive to the BAD BH3 mimetic ABT-737 indicating that primed BAK is primarily involved in ABT-737 induced apoptosis. Primed BAK was displaced from BCL-XL by ABT-737 followed by an N-terminal conformational change and subsequent formation of dimers and higher molecular weight complexes. These sequential BAK activation steps occurred independently of cell fate and did not represent the rate limiting steps in BAK activation as a BAK BH3 mutant L78A lost proapoptotic function but still oligomerised as efficiently as wt BAK. Thus the transition from inactive BAK to a membrane permeabilising pore requires an additional activation step. I demonstrate that after 30 min of ABT-737 exposure primed BAK, after its displacement from BCL-XL, interacts with BIMEL reflecting the transient nature of this interaction. This interaction represented an additional step in BAK activation as BAK pro-apoptotic function was enhanced when BIMEL and BAK were co-expressed. However BIMEL did not induce the N-terminal conformational change nor oligomerisation of BAK and its interaction occurred downstream of both these events. In addition the pool of BIMEL involved in the further activation of BAK did not represent that sequestered by the antiapoptotic proteins BCL-2 and BCL-XL. These data suggest that BAK activation occurs in multiple steps in which a further activation event is required after the exposure of the BH3 domain, the N-terminal conformational change and the formation of high molecular weight complexes but prior to cytochrome c release. I propose that this event may be represented by interaction of N-terminal conformational changed/oligomerised BAK with BIMEL.

It must be a bad child I build rooms, installations, where I combine objects into scenes.  This scene is an attempt to portray feelings and mental states surrounding sexual abuse. I'm building a girls' room, but it's more of a psychological place than a real room. I want to let the girl take place and show a trauma that lives in the hidden, depending on the environment not seeing, in the public space.  It's about living with memories of a trauma in a traumatized body. I place two larger sculptures in the installation, a girl and a woman, who share the (mental) room. Selection of materials is an important part of my practice. In my installations there is almost always a feeling of discomfort, that things are not what they seem at first. I use the expectations that exist around different materials and what they symbolize and create uncertainty in the room by, for example, giving them new places and functions. I use film to get movement into the installation. The sculptures come to life through the film. The film contains no sound when I see the state I portray as silent.

However, it does not mean that businesses will offshore production activities abroad no more. Instead, the two phenomena, respectively offshoring and back-reshoring, are destined to coexist.

A low-cost power router (PR), capable of dynamic, independent control of active- and reactive-power flows on meshed grids is presented. The operating principle, detailed schematics, and various possible implementations of the proposed power router are discussed. Various operating modes are identified and a control algorithm has been proposed and verified through simulations. Small-signal and frequency-domain models of the power router from basic time-domain equations are developed. A three-tier protection system based on the fail-normal switch to avoid single point-of-failure is presented. The operation of proposed protection system in isolating the converter and the grid in the event of faults is verified through simulation. An analytical method to evaluate the stability of a system with multiple power routers is proposed. Necessary conditions for the PR-controller design to ensure stable operation of a system with multiple power routers is proposed. These necessary conditions are verified through simulation studies. Potential applications of proposed power router in distribution system and the associated challenges in implementation are presented. The functionality and advantages of the proposed power router are experimentally demonstrated at 13 kV, 1 MVA. The proposed power router can result in a low cost power routing solution that can reduce electric grid congestion and efficient implementation of RPS mandates.

The web browser has become one of the most recognizable software applications on consumer desktops. Yet its utilization and capabilities are often misunderstood. Recent innovations in the web have evolved the Internet into a network of sophisticated applications that defy historical uses of the “browser”; a term that itself has become somewhat of a misnomer. This research studies the evolving set of user expectations for the browser as an application platform and challenges certain anachronistic features, specifically the “back” button, that are unnecessary and confusing given the new environment that browsers are used in. Because of this shift, implicit new user requirements arise around the browser’s user interface. The back button, like other elements in the browser have already demonstrated, should be de-emphasized in modern iterations of web browsers. The study is qualified by an analysis of user behavior within a popular, modern, web application.

The thesis deals with the development of systems for intelligent building, focused on wiring system, division of them and comparison of their differences. The advantages of these systems like comfort, energy reduction, etc. are also mentioned. Nowadays several standards of systems for intelligent building exist as KNX/EIB or LON, therefore thesis concerns to trade-off these standards too. Main part of the thesis is focused on system Egon made by company ABB where the system concept as well as bus modules are described that allow different functions, mutual connection of modules and their programming. Tutorial panel with ABB Ego-n wiring system, which will be used for study purpose, is designed in the practical part of the work. It includes scheme of the panel, electric linkage of components and photographs of constructed panel as documentation. In the final part of the work two examples of laboratory exercise for installation and setting are listed, including programming description.
