The effect of increasing temperatures on the duration of postharvest flower development was determined for three specialty crop species: marguerite (Argyranthemum frutescens Webb ex Schultz-Bip.) `Butterfly' and `Sugar Baby'; swan river daisy (Brachycome hybrid Cass.) `Ultra'; and bacopa (Sutera cordata Roth.) `Snowflake'. Plants were grown in a greenhouse at 18 °C (65 °F) until flowering, and then transferred into a phytotron to determine heat tolerance. Plants were stored for 8 weeks at constant temperatures of 18, 23, 28, and 33 °C (65, 73, 82, and 91 °F) for 2-week intervals. Flower bud and flower number were recorded weekly. Sutera cordata `Snowflake' and B. hybrid `Ultra' had the greatest flower number at the 23 °C temperature, decreasing in the 28 °C environment. Argyranthemum frutescens `Butterfly' and `Sugar Baby' had greatest flower number at 28 °C, but flowers were of lower quality thanat 23 °C. Flower development of all cultivars ceased at 33 °C, at the end of 8 weeks at increasing temperatures, but when plants were returned to the 18 °C production greenhouse, flower development resumed. High temperatures (28 °C) reduce the postharvest performance of S. cordata, B. hybrid, and A. frutescens plants grown in hanging baskets; therefore, these species should be marketed as spring-flowering products since summer performance may be unsatisfactory in warm climates.

Abstract Abstract 3221 Poster Board III-158 Introduction Umbilical cord blood (UCB) has become an easily, available and viable source of hematopoeitic stem cells for transplant. The main limitation factor for its wide use is cell dose. Previous studies have showed that certain physiological parameters pertaining to either the baby or the mother impact in the UCB cell yields. Objectives The aim of our study was to compare five physiological parameters pertaining to the mother or baby (mother`s age [MA], gestational age at delivery [GA], baby`s gender [G], baby`s birth weight [BW] and type of delivery [TD]) with total nuclear cells (TNC) and CD34 + cells recovery. We also evaluated the impact of time from collection to processing (TCP) on CD45+ cells viability. Methods UCB product collection was performed after the baby's delivery, while the placenta was still in uterus, either from vaginal or cesarean deliveries. Collection bags that were used contained 35 mL of CFDA-1 (CFD with Adenina) as anticoagulant. Cord blood units (CBU) were processed in our institution under local and international regulations regarding cord blood banking. Usual techniques with HES 6% for red cell depletion and 4°C centrifugation for plasma depletion were used. Twenty-five ml, EVA, two-compartment cord blood cells freezing bags (Pall Medical) were used for a final CBU volume of 20.5 ml combined with Dextran 40/40 and DMSO for cryopreservation. A sample was removed for flow cytometric analysis (BD FACSCan) and to determine the TNC (Cell-dyn 1200). Cultures pre and post CBU handling were done. Freezing took place in a controlled-rate freezer according to standard protocol before storage in liquid nytrogen. Results From May 2004 to Jun 2009, a total of 4,262 continous UCB collections were performed in our institution throught the Peruvian Republic. Seventy-eight percent of the CBU were collected by cesarean; median TCP was 30 hours 58 minutes. The mean CBU volume and TNC count were 81.8 ml and 8.63 × 108 respectively. The colected volume was greater in cesarean than vaginal delivery (85.3 ml vs 78.9, F=30.82, p<0.001). TNC counts collected were directly correlated with GA: in preterm delivery (<37sem) was 7.13×108, in term delivery (>=37sem) was 9.93×108. TNC counts were directly correlated with BW (F=325, p<0.001) while the MA had inverse correlation (F=8.05, p=0.005); regarding TD there was a significant mayor TNC count in the vaginal vs. cesarean group (10.49×108 vs. 9.22×108, F=48.207, p<0.001); while it was a trend for major TNC count in females vs. males babies (9.82×108 vs. 9.13×108, p=0.059). CD34+ cells count was directly correlated with BW (F=70.1, p<0.001). The strongest correlation was with GA (in preterm: 61.99 CD34+/ul and in term delivery: 84.67 CD34+/ul, F=27.62 p=<0.001); moreover, there was association between CD34+ cells count with TD (vaginal: 87.88 CD34+/ul vs cesarean: 80.26 CD34+/ul, F=5.327 p=0.02). There was not association either with G (females: 79.97 CD34+/ul vs males: 83.48 CD34+/ul, F=1.668 p=0.197) neither MA (F=1.82, p=0.177). There was a significant difference between CD34+ viability cells among CBU with less than 48 hours or more of TCP (99.6% vs. 99.4%, p=0.019); this difference was stronger when the CD45+ viability was evaluated (93.35% vs. 90.14%, p<0,001). Conclusions TNC and CD34+ UCB cells are influenced by many variables related to the mother and the baby. It looks like on time female babies with good weight, born to a younger mother and from a vaginal delivery reach highest TNC count. CD34+ cells count was directly correlated with BW and CD34+ viability is mainly influenced by the time from collection to processing Disclosures No relevant conflicts of interest to declare.

Fifty samples from five baby food products mainly made of cereal flour(s) were analyzed. The moisture contents of these products were between 11.14% and 11.9%, a level below 14.0%, the recommended level for safe storage of cereal grains and their products. The mycological analysis was carried out using the dilution plate method and two isolation media (DG18 for isolation of xerophilic fungi and DRBC for fungi in general). A total of 80 species related to 37 genera in addition to some unidentified fungal and yeast species were recorded on both media from the five products. The products were contaminated abundantly by xerophilic fungi which were occurring in 88% of food samples and accounting for 18.1% of the total CFU as recorded on DG18. The highest contamination level by xerophiles was registered in Mwebaza rice porridge (a component of rice flour) and the lowest in Mukuza (a product of maize, soyabean and sorghum flours). 11 xerophilic species were recorded of which <em>Aspergillus</em> and <em>Eurotium</em> (4 species each) were the predominant giving rise to 9.1% and 8.9% of the total CFU, with <em>A. wentii, A. candidus, E. cristatum</em> and E. repens were the most contaminating species. Of the fungi recorded other than xerophiles, species of Aspergillus (particularly <em>A. flavus</em> followed by <em>A. niger</em>), <em>Penicillium (P. citrinum, P. oxalicum), Fusarium (F. solani, F. tricinctum), Cladosporium (C. sphaerospermum)</em> and yeasts were the most predominant. Contamination of such foods is a matter of health hazard as these foods are for babies. So, the use of fresh, well-dried and uncontaminated flours for production of such foods is recommended.

Background:Although caregiving is a normal part of being a parent of a young child, it is still unclear whether caregiving causes upper limb or neck disability in the caregiver.Objectives:The aim of this study was to investigate the relationship between caregiver burden and upper limb-neck disabilities and pain in baby caregivers.Methods:Sixty caregivers who are responsible for the caregiving of a 0-2 year old healthy baby, were included in this study. Physical characteristics and the gender of the caregivers were recorded. Caregiver burden was assessed by the Zarit Burden Interview; upper limb problems by DASH and neck problems by the Neck Disability Index and Neck Bournemouth Questionnaire. In addition, pain severity related to neck and upper limb was evaluated by using Visual Analog Scale over a 10 level scale.Results:The mean age of the caregivers was 30.96 ± 6.43 year. The mean body mass index of the caregivers was 23.34 ± 3.29 indicating normal body composition. Height and weight were 163.75 ± 6.01 and 62.55 ± 8.98, respectively. Ninety-five percent of the caregivers were mother and the others were the babysitter. A weak relationship was determined between the Zarit Burden Interview and DASH (p=0.02, r=0.2), while no relationship was found between caregiver burden and other assessments. Caregivers were found to have very low pain severity in the upper limb and neck.Conclusion:It has been previously reported that caregiving for a disabled child or adult causes care burden and various musculoskeletal disorders1-4. Bisphenol A (BPA), which is widely used for manufacturing polycarbonate plastics and epoxy resins, has been banned from use in plastic baby bottles because of concerns regarding endocrine disruption. Substances with similar chemical structures have been used as BPA alternatives; however, limited information is available on their toxic effects. In the present study, we reviewed the endocrine disrupting potential in the gonad and thyroid endocrine system in zebrafish after exposure to BPA and its alternatives (i.e., bisphenol AF, bisphenol C, bisphenol F, bisphenol S, bisphenol SIP, and bisphenol Z). Most BPA alternatives disturbed the endocrine system by altering the levels of genes and hormones involved in reproduction, development, and growth in zebrafish. Changes in gene expression related to steroidogenesis and sex hormone production were more prevalent in males than in females. Vitellogenin, an egg yolk precursor produced in females, was also detected in males, confirming that it could induce estrogenicity. Exposure to bisphenols in the parental generation induced a decrease in the hatchability associated with offspring generation. In zebrafish exposed to bisphenols, significant decreases in thyroxine concentrations and increases in thyroid-stimulating hormone concentrations were commonly observed. Alternative compounds used to replace a chemical of concern are believed to be less toxic than the original compound; however, several BPA alternatives appear to have similar or greater effects on the endocrine system in zebrafish. Since endocrine systems interact with each other, further studies are needed to assess the primary target of BPA alternatives among the endocrine axes.

In March of 2013, new symptoms were observed in more than seven million nursery-grown sweet pepper (Capsicum annuum) plants in El Ejido, Almería (southern Spain). Symptoms included wilting without yellowing of leaves and stunting of plants. Plant crowns exhibited necrosis that advanced through the main root along with slight root rot. Xylem was not affected above or below the crown. Symptoms were thought to be caused by the well-known pepper pathogen Phytophthora capsici. However, sporodochia of Fusarium oxysporum were observed on plant crowns. Symptomatic seedlings (n = 200) were sampled and analyzed. Tissue from roots and epidermal crowns were plated on PDA, PARP, and Komada media, as well as stem discs on PDA and Komada. No Phytophthora sp. were observed and F. oxyporum was exclusively isolated from all 200 samples, from roots and crowns, but not from xylem. Pathogenicity of 60 of these F. oxysporum isolates was studied by inoculation onto sweet pepper plants (cv. del Piquillo) at the 2-true-leaf stage. Twelve plants per isolate, grown on autoclaved vermiculite, were inoculated by drenching with 20 ml of a conidial suspension (1 × 105 CFU/ml) of each isolate per plant. Each suspension was obtained by blending one PDA petri dish fully covered with one isolate. Non-inoculated plants served as control. Plants were maintained for 30 days in a growth chamber with a 14-h photoperiod (1.6 ×·104 lux) and temperatures at 23 to 26°C. The assay was conducted twice. Symptoms described above were reproduced on crown and roots of the inoculated plants with no symptoms in stem discs. No symptoms were observed on controls after 48 days. Host specificity was tested for 13 isolates to tomato (Solanum lycopersicum) cv. San Pedro, eggplant (S. melongena) cv. Alegria, cucumber (Cucumis sativus) cv. Marketmore, watermelon (Citrullus lanatus) cv. Sugar Baby, and Chinese cabbage (Brassica campestris subsp. condensa) cv. Kasumi (4). These plants were inoculated as previously described for pathogenicity tests (12 plants per species, repeated twice). None of the plants exhibited the characteristic symptoms after 60 days. Five isolates of F. oxysporum f. sp. radicis-cucumerinum and four isolates of F. o. f. sp radicis-lycopersici were also inoculated without any symptoms in any of the inoculated sweet pepper plants. Morphological identity of all isolates corresponded to F. oxysporum. The fungi were identified following the morphological keys and methodology provided by (1) and (2). Three isolates from the 60 tested were selected for molecular identification. Molecular identification was performed by sequencing partial TEF-1α gene (3). Subsequent database searches by BLASTn indicated that the resulting sequence of 659-bp had 100% identity with the corresponding gene sequence of F. oxysporum. The sequences were identical for the three isolates and were deposited on the EMBL Sequence Database (HG916993, HG916994, and HG916995). Results suggest that the pathogenic ability of the isolates varies from a vascular Fusarium wilt. F. oxysporum f. sp. capsici is a reported pathogen to sweet pepper (5), but the symptoms we have found are closer to those manifested by the formae speciales that causes root and crown rot of other plants. Various measurement techniques suitable with the length scale of PEFC components have been explored to address the PEFC water management challenge. For the gas diffusion layer (GDL), technological advances have permitted operando water saturation quantification and elucidation of its mechanism with X-ray tomography with few microns resolution 1-4. Furthermore, sub-voxel information contained in the X-ray tomography data has demonstrated accurate water quantification in the nano-porous microporous layer (MPL)5. However, in the nanoscale regime of the catalyst layer, the quantification of water and the mechanism of the water filling the pores remain nontrivial. Studies involving imaging techniques dedicated to understand the water management in the catalyst layer have been carried out albeit with limited spatial resolution6. Nevertheless, the pore-scale information has remained unresolved. Small angle X-ray scattering (SAXS) technique is proposed as a non-destructive diagnostic tool to investigate the catalyst layer saturation in operating conditions. SAXS is well suited for diagnosing the presence of liquid water during operando experiments because of its sensitivity to electron density contrast, nanoscale observation window, high temporal resolution and adequate spatial resolution to distinguish the components in PEFCs. Herein, SAXS intensity profiles measured at cSAXS, Swiss Light Source, Paul Scherrer Institut, Switzerland, with a recently developed SAXS-compatible operando PEFC (Figure 1 middle and right) are interpreted using representative morphology models and assuming different water filling mechanisms (Figure 1 middle and left). The presentation will provide insights into the wetting phenomena in the catalyst layer of PEFC during operation using a representative morphology modelling approach. O objetivo do trabalho foi investigar os efeitos da salinidade da solução nutritiva (CEsol) sobre a produção, acúmulo de solutos orgânicos e inorgânicos e fluorescência da clorofila a em plantas de minimelancia cv. Sugar Baby. O delineamento experimental foi o inteiramente casualizado com quatro repetições. As plantas foram cultivadas em sistema hidropônico com cinco níveis de CEsol: 2,5; 3,5; 4,5; 5,5 e 6,5 dS m-1. Foram avaliados: massa do fruto, biomassa da parte aérea, teores de clorofilas a e b, fluorescência da clorofila a e teores de solutos orgânicos e inorgânicos. A massa do fruto e a biomassa seca da parte aérea foram influenciadas negativamente pela CEsol. Houve redução na concentração de carboidratos solúveis totais, aminoácidos livres, proteínas solúveis e prolina livre com o aumento da CEsol. Os íons sódio, cloreto e razão sódio/potássio aumentaram com a salinidade e os teores de potássio foram reduzidos. O incremento da salinidade aumentou linearmente o teor de clorofila b e reduziu linearmente o rendimento quântico da conversão de energia fotoquímica do FSII promovendo um ligeiro aumento no rendimento quântico da perda de energia regulada do FSII. This study investigated the effects of nutrient solution salinity (ECsol) on production, accumulation of organic and inorganic solutes and chlorophyl a fluorescence in mini watermelon plants cv. Sugar Baby. The experimental design was completely randomized with four replications. The plants were grown in a hydroponic system with five levels of ECsol: 2.5; 3.5; 4.5; 5.5 and 6.5 dS m-1. The variables evaluated were the mass of the fruit, the biomass of the aerial parts, contents of chlorophylls a and b, fluorescence of chlorophyll a, and contents of organic and inorganic solutes. The mass of the fruit and dry biomass of the aerial parts was negatively influenced by ECsol. There was a reduction in the concentration of total soluble carbohydrates, free amino acids, soluble proteins and free proline with the increase in ECsol. The sodium, chloride and sodium/potassium ratio increased with ECsol and potassium levels were reduced. The increase in ECsol linearly increased the chlorophyll b content and reduced the quantum yield of the photochemical energy conversion of the PSII promoting a slight increase in the quantum yield of the regulated energy loss of the PSII. The paper presents the results of a study of solid-phase equilibria in the Tb–Te system and the thermodynamic properties of terbium tellurides obtained by the methods of electromotive forces and X-ray diffraction analysis. Based on the experimental data, it was established that the TbTe, Tb2Te3, TbTe2 и TbTe3 compounds are formed in the system. For the investigations of the alloys from the two-phase regions TbTe3+Te, TbTe2+TbTe3, and Tb2Te3+TbTe2, the EMF of concentration cells relative to the TbTe electrode was measured. The EMF of concentration cells relative to the terbium electrode was measured for the TbTe+Tb2T3 region. The partial thermodynamic functions of TbTe and Tb in alloys were determined bycombining the EMF measurements of both types in the 300–450 K temperature range, based on which the standard thermodynamic functions of formation and standard entropies of the indicated terbium tellurides were calculated. Pour traiter cette question, nous avons construit notre argumentation en trois parties. Tout d'abord, après avoir défini le groupe des agents de conduite comme un groupe corporatiste, nous étudierons les modifications techniques et organisationnelles qui ont boulversé certaines références défendues par la profession. Nous tenterons ainsi de mesurer les conséquences sur la pratique quotidiene de la conduite. Nous ferons également le point sur les travaux menés antérieurement concernant les agents de conduite et sur les acquis de ces recherches. Dans une deuxième partie, nous nous intéresserons au processus de formation des agents de conduite. Quel est le modèle de formation utilisé par l'entreprise ? Les professionnels en poste jouent-ils un rôle important dans le processus de formation ? Pouvons-nous associer la formation des agents de conduite à un processus de socialisation ? Enfin, après avoir étudié les parcours biographiques des individus interrogés, nous tenterons de définir de manière opératoire le concept d'identité pour qu'il puisse rendre compte de notre travail. Enfin, nous proposerons un modèle des constructions identitaires mises en oeuvre par les membres de notre population.

Ce travail a ete consacre a l'etude de la reduction de l'oxygene sur des cobaltites de lanthane dopees au strontium. Ces materiaux sont deposes par pulverisation cathodique sur les deux faces polies d'une pastille de zircone yttriee stabilisee. Cette technique de depot nous a permis de fabriquer des electrodes de faible epaisseur, reproductibles, denses et couvrantes. La caracterisation electrochimique de ces systemes symetriques a ete realisee par spectroscopie d'impedance, sans polarisation continue. Les parametres de cette etude sont : l'epaisseur des electrodes (de 0,35 m a 2m), la temperature et la pression partielle d'oxygene. La resistance de polarisation des electrodes augmente lineairement avec l'epaisseur. La valeur de resistance specifique pour la plus faible epaisseur, extrapolee a 800c, est de 0,27. Cm#2, ce qui montre que ce materiau reste tres prometteur pour une application comme materiau de cathode pour pile a combustible a oxyde electrolyte solide. Quant a la reduction de l'oxygene, la diffusion de l'oxygene dans le volume de l'electrode et l'adsorption non dissociative constituent les processus dominants de cette reaction. Ils different avec la temperature et la pression partielle d'oxygene. Pour des epaisseurs plus elevees (1,6 m et 2m), une degradation des interfaces est mise en evidence. Elle se traduit en terme d'impedance par une augmentation de la resistance de polarisation au cours du temps. Elle est liee au developpement d'un demi-cercle haute frequence et d'un phenomene additionnel a tres basse frequence apres les recuits aux temperatures tres elevees.

Introdu??o: Em pacientes com insufici?ncia renal cr?nica, s?o comuns altera??es nos n?veis s?ricos de pot?ssio, f?sforo e de c?lcio. Este estudo verificou a associa??o entre a ingest?o de pot?ssio, f?sforo e c?lcio e os n?veis s?ricos desses elementos em pacientes com insufici?ncia renal cr?nica em tratamento hemodial?tico. M?todos: Estudo transversal e contempor?neo que incluiu pacientes com insufici?ncia renal cr?nica em tratamento hemodial?tico h? pelo menos tr?s meses, de ambos os sexos, maiores de 18 anos de idade, n?o hospitalizados e pass?veis de avalia??o antropom?trica de peso e altura, que aceitassem participar do estudo. Realizou-se a avalia??o diet?tica e bioqu?mica de pot?ssio, f?sforo e c?lcio. Resultados: Foram avaliados 74 pacientes (47 masculinos). Utilizando-se o coeficiente de correla??o de Pearson, com signific?ncia determinada pelo teste t, houve correla??o positiva de regular intensidade (r = 0,44; p < 0,001) em an?lise bivariada entre a ingest?o de pot?ssio e o n?vel s?rico desse elemento. Ocorreu aus?ncia de correla??o (r = 0,14; p = 0,25) entre a ingest?o de f?sforo e o n?vel s?rico desse elemento, como tamb?m a aus?ncia de correla??o (r = 0,062; p = 0,60) entre a ingest?o de c?lcio e o n?vel s?rico desse elemento. Dois (3%) pacientes do estudo foram classificados como desnutridos, 40 (54%) pacientes como eutr?ficos e 32 (43%) pacientes como pr?-obesos ou obesidade. Conclus?es: Houve correla??o positiva entre a ingest?o de pot?ssio e seus n?veis s?ricos, assim como a aus?ncia de correla??o entre a ingest?o de f?sforo e c?lcio e seus n?veis s?ricos no grupo estudado.

A Psicologia do Desporto tem como preocupação tentar identificar os efeitos psicológicos do comportamento humano em contextos desportivos e os efeitos psicológicos que a participação desportiva poderá ter nos seus participantes (Cruz, 1996a). O presente estudo visa identificar a orientação cognitiva e o perfil psicológico de prestação de jogadores masculinos de futebol de 11 profissional, em equipas de elite – Chelsea e Benfica – avaliando os skill psicológicos desses jogadores, procurando entender se há algum efeito Mourinho, distinguindo a amostra entre os que foram (ou não) treinados pelo treinador português, pesquisando assim sobre o efeito concreto de um treinador da actualidade – José Mourinho – reconhecido mundialmente pelos resultados positivos e consistentes obtidos nos últimos anos em qualquer clube e país por onde tem trabalhado. Acompanhando o emergir de novas valências conceptuais e metodológicas decorrentes de distintas áreas, procedemos a uma recolha de matéria bibliográfica, visando aprofundar conhecimentos e reflectir sobre o PPP e as OC em contexto desportivo. Procurando enriquecer o conteúdo desta obra, decidimos, além de uma abordagem a conteúdos já existentes na literatura, tentar seguir, inquestionavelmente, um “caminho” mais específico e qualitativo, através de entrevistas a pessoas ligadas ao futebol (Drogba, Petr Cech, Sá Pinto etc) e da aplicação directa de questionários a 29 jogadores profissionais, (21 jogadores do Chelsea e 8 jogadores do Benfica). Os questionários utilizados foram o Perfil Psicológico de Prestação, desenvolvido e validado por Loehr (1986) e traduzido para português por Vasconcelos-Raposo (1993) e o Teste de Orientação Cognitiva para a tarefa e para o ego, teste elaborado por Duda e Nicholls (1989), traduzido e adaptado para Português por Fonseca (1999). Os procedimentos estatísticos consideraram análises descritivas, comparativas e correlacionais. Os valores obtidos nas variáveis para o total da amostra foram: Motivação (27.55+2.097), Auto- Confiança (26.83+2.494), Atitude Competitiva (26.03+2.758), Pensamentos Positivos (25.10+2.160), Atenção (25.07+3.046), Visualização (23.55+3.429), Pensamentos Negativos (22.24+2.824), Tarefa (4.23+0.493) e Ego (2.79+0.741).No que concerne à comparação entre Chelsea e Benfica, não se verificaram diferenças estatisticamente significativas sendo que também não foram verificadas evidências significativas entre os jogadores que foram e os que não foram treinados por Mourinho. Uma das variáveis que permitiu maior diferenciação entre os factores psicológicos foi continente de origem. Essas diferenças foram encontradas ao nível da atenção. No que concerne aos grupos de idade, verificamos diferenças estatísticas significativas ao nível da atenção e visualização. Constatamos que conforme a idade vai aumentando, aumenta também os níveis de atenção. A visualização apresenta os valores mais elevados nos jogadores mais novos e nos mais velhos, existindo assim uma fase intercalar onde foram observados os valores são baixos. Ao nível da experiência competitiva, verificamos evidências estatisticamente significativas que nos levam a constatar que quanto maior é a experiência competitiva, mais elevados são os índices de atenção e auto-confiança dos jogadores. Com base na presente investigação, sugerimos a necessidade de integrar mais sistematicamente o treino mental na preparação dos jogadores de elite do Chelsea e do Benfica, visando a maximização do rendimento e igualmente contribui para a identificação do estado ideal de prestação desportiva na modalidade de Futebol.
The Sport Psychology has its eyes on trying to identify the psychological effects of human behavior in sports contexts and psychological effects that sport participation can have on its participants (Cruz, 1996a). This study aims to identify the cognitive orientation and psychological profile of the provision of male soccer players, 11 professional teams in the elite - Chelsea and Benfica - evaluating the psychological skill of these players, trying to understand if there are any Mourinho, distinguishing between the sample those who were (or not) trained by the Portuguese coach, so searching on the net effect of a coach today - José Mourinho - recognized worldwide for their consistent and positive results obtained in recent years at any club and country for which it has worked. Following the emergence of new conceptual and methodological valences arising from different areas, we made a collection of bibliographic material, to deepen knowledge and reflect on the PPP and the OC in the sporting context. Seeking to enrich the contents of this work, we decided, and an approach to existing content in literature, try to follow unquestionably a "way" more specific and qualitatively, through interviews with people linked to football (Drogba, Petr Cech, Sá Pinto etc.) and direct application of questionnaires to 29 professional players (21 players from Chelsea and eight players from Benfica). The questionnaires used were the Psychological Performance, developed and validated by Loehr (1986) and translated into Portuguese by Vasconcelos-Raposo (1993) and the Test of Cognitive Orientation to the task and ego test, developed by Duda and Nicholls (1989 ), translated and adapted to Portuguese by Fonseca (1999). The statistical procedures considered descriptive analysis, comparative, and correlational. The values obtained in the variables for the total sample were: Motivation (27.55+2.097), Self-Reliance (26.83+2.494), Competitive Attitude (26.03+2.758), Positive Thoughts (25.10+2160), Attention (25.7+3.046), Visualization (23.55+3.429 ), Negative Thoughts (22.24+2824), Task (4.23+0.493) and Ego (2.79+0.741). Regarding the comparison between Chelsea and Benfica, there were no statistically significant differences were also not significant evidence was found among the players who were and those who were not coached by Mourinho. One of the variables that allowed for greater differentiation between the psychological factors was continent of origin. These differences were found at the level of attention. Regarding the age groups, we found statistically significant differences at the level of attention and visualization. We found that as age increases, also increases levels of attention. The display shows the highest values in younger players and older, so there is an intermediate stage where they were observed values are low. At the level of competitive experience, we found statistically significant evidence that leads us to see that the greater the competitive experience, the higher rates of attention and self-confidence of the players. Based on this research, we suggest the need to integrate further the mental training in preparation of the elite players from Chelsea and Benfica, aiming to maximize income and also helps to identify the ideal state of sporting provision in the form of Football.

Nuclear Medicine enables, among other things, the visualization, characterization and quantification of biochemical processes, metabolism, biomarkers and receptors in vivo by molecular imaging. One of the methods of functional imaging available in this area is Positron Emission Tomography (PET), which depends on the administration of a radiopharmaceutical in the studied organism. One disease that can be studied by PET scan is multiple sclerosis (MS) through the access to the TSPO (Translocator protein) receptors density. The radiopharmaceutical [11C]-(R) -PK11195 has high affinity and selectivity for these receptors, which are increased in activated microglia cells in the case of MS. Therefore, the objective of this study was to develop and to validate the production process (synthesis, purification and quality control) of the radiotracer [11C]-(R)-PK11195 in the Radiopharmaceutical Production Center of the Brain Institute of Rio Grande do Sul (BraIns), allowing performing PET scans for the study of MS. The radioisotope was produced in the 16 MeV PETrace cyclotron (GE Healthcare) as [11C]CO2 via the nuclear reaction 14N (p, ?)11C after the bombardment of a gaseous target containing N2 and 0.5% of O2 with protons. The production was performed in the equipment TRACERlab FX-C Pro (GE Healthcare) to meet the requirements of Good Manufacturing Practices (GMP). The radiopharmaceutical quality control was carried out at the BraIns Quality Control Laboratory and the final product was tested for Identity and Radionuclidic Purity, Radiochemical Purity, Specific Activity, Residual Solvents, pH, Sterility, the Filtration Membrane Integrity and Bacterial Endotoxins. The synthesis was performed through the methylation of the precursor (R)-[N-desmethyl]PK11195 using [11C]CH3I (methyl iodide labeled with carbon-11) under the defined optimal conditions: 400 uL of dimethyl sulfoxide (DMSO), approximately 1 mg of precursor, approximately 10 mg of potassium hydroxide (KOH) at 40 ?C for 1 minute. The reaction mixture was then neutralized with 1 mol.L-1 hydrochloric acid and diluted with mobile phase (76% acetonitrile and 24% Milli-Q water). The diluted mixture was injected into a semi-preparative HPLC system for purification. Solid phase extraction (through Sep-Pak C18 cartridges - Waters) was used to remove the solvent. Product elution and formulation was conducted with 0.7 mL of ethanol and 6.3 ml of 0.9% saline (sterile and pyrogen free). Sterilization was performed by filtration on a 0.22 ?m pore membrane and, as well as the filling process, was carried out under laminar flow. The process steps could be optimized and fitted to the equipments and laboratories structure. Radiochemical yield of the process in relation to the [11C ]CH3I start activity was 15.5 ? 2.4 % ( 7471.5 ? 1283.8 MBq ; n = 39 ) . All batches were approved according to the specifications defined and tested in the quality control, reaching Radiochemical Purity levels greater than 99% and a Specific Activity of 39.7 ? 11.1 GBq / micromol , 10 minutes before the injection. All analytical methods were validated and met the requirements established by current regulations. Three consecutive batches were produced and approved by the quality control, so the process could be considered validated. The developed process was suitable for production of adequate doses of the radiopharmaceutical [11C]-(R)-PK11195 for clinical use at BraIns. The production of the radiopharmaceutical labeled with carbon-11 at the institute provides an additional tool for improving health services in the country and to improve the life quality of the patients.
A medicina nuclear possibilita, dentre outras coisas, a visualiza??o, caracteriza??o e quantifica??o de processos bioqu?micos, metab?licos, biomarcadores e receptores, in vivo, atrav?s da imagem molecular. Dentre as modalidades de imagem funcional dispon?veis nesta ?rea est? a Tomografia por Emiss?o de P?sitrons (PET), que depende da administra??o de um radiof?rmaco no organismo estudado. Uma das doen?as pass?veis de estudo atrav?s do exame PET ? a esclerose m?ltipla (EM), atrav?s do acesso ? densidade de receptores TSPO (Prote?na Translocadora). O radiof?rmaco [11C]-(R)-PK11195 possui alta afinidade e seletividade por estes receptores, aumentados nas c?lulas ativadas da micr?glia nos casos de EM. O controle de qualidade do radiof?rmaco foi realizado no Laborat?rio de Controle de Qualidade do InsCer e o produto acabado foi submetido ?s an?lises de Identidade e Pureza Radionucl?dica, Pureza Radioqu?mica, Atividade Espec?fica, Solventes Residuais, pH, Esterilidade, Integridade da Membrana Filtrante e Teor de Endotoxinas. A s?ntese foi realizada atrav?s da metila??o do precursor (R)-[N-desmetil]PK11195 utilizando o [11C]CH3I (iodeto de metila marcado com carbono-11) sob as condi??es ?timas definidas: 400 ?L de dimetilsulf?xido (DMSO), aproximadamente 1 mg de precursor, aproximadamente 10 mg de hidr?xido de pot?ssio (KOH), a 40 ?C durante 1 minuto. A mistura de rea??o foi posteriormente neutralizada com ?cido clor?drico 1 mol.L-1 e dilu?da com fase m?vel (acetonitrila 76 % e ?gua Milli-Q 24 %). A mistura dilu?da foi injetada em sistema CLAE semi-preparativo para purifica??o. Extra??o de fase s?lida (atrav?s de cartuchos SepPak C18 ? Waters) foi utilizada para remo??o do solvente. A elui??o e formula??o do produto foi realizada com 0,7 mL de etanol e 6,3 mL de solu??o salina 0,9 % (est?ril e livre de pirog?nios). A esteriliza??o foi realizada atrav?s de filtra??o em membrana de poro de 0,22 ?m e, assim como o envase, ocorreu em fluxo laminar. As etapas do processo foram otimizadas e adequadas aos equipamentos e estrutura dos laborat?rios. O rendimento radioqu?mico do processo em rela??o ? atividade de [11C]CH3I de partida foi de 15,5?2,4 % (7471,5?1283,8 MBq; n=39). Todos os lotes produzidos foram aprovados quanto ?s especifica??es definidas e testadas no controle de qualidade, atingindo Pureza Radioqu?mica maior que 99 % e uma Atividade Espec?fica de 39,7?11,1 GBq/?mol, 10 minutos antes da inje??o. Todas as metodologias anal?ticas foram validadas e atenderam os crit?rios estabelecidos pela norma vigente. AbstractIn the recent years, awareness has risen by an increasing number of researchers that we need studies that appropriately model the complexity of school improvement if we want to increase our knowledge about school improvement substantially and to close the identified research gaps within this field (Feldhoff T, Radisch F, Klieme E, J Educ Admin 52(5):565–736, 2014; Hallinger P, Heck RH, School Effect School Improv 22(2):149–173, 2011; Sammons P, Davis S, Day C, Gu Q, J Educ Admin 52(5):565–589, 2014). So far, respective quantitative studies, that appropriately consider those complexities, have hardly been realized because of the high efforts of current methods and costs involved (Feldhoff T, Radisch F, Bischof LM, J Educ Admin 2(54):209–240, 2016). It is, therefore, apparent to look for new, innovative methods that can adequately take into account the complexity of school improvement. For a reasonable search and selection of innovative methods, it is necessary to describe the systematic complexity of school improvement and the resulting requirements for the methods in more detail. This is a central goal in this chapter. For this reason, we present our framework of complexity. We then formulate questions that will prompt the reader to reflect critically upon the methods in general and especially in this volume.

Protein C (PC) deficiency associated with hereditary venous thromboembolic disease was first reported in 1981 and is inherited as an autosomal dominant disorder. The prevalence of heterozygous PC deficiency is estimated to be 1 to 4% in venous thrombotic diseases. The homozygous PC deficiency is even rare, and has been reported in only about 10 families througout the world. It usually presents in newborn infants as purpura fulminans or severe thrombotic disease. We herein report two newborn brothers in a Chinese family, who manifested with purpura fulminans soon after birth and died at age of 21 days and 27 days respectively. Vitamin K was administered to the second baby after birth. Both parents are not consanguineous and there were no family histories of thromboembolism on paternal and maternal sides. Blood sample was not available for specific studies in the first baby. PC antigen level by electroimmunoassay was <6% in the second baby and 49% and 60% respectively in their mother and father. Antithrombin III activity by amidolytic method was 49% in the second baby, and 90% and 97% respectively in their mother and father. Vitamin K-dependent coagulation factors and factor V were within the expected range for a newborn. Factor VIII and fibrinogen level were notably decreased. Autopsy findings of the two newborns demonstrated the similar pictures characterized by fibrin thrombi in blood vessels causing extensive hemorrhagic infarts of skin, lung, liver, kidneys, testis, urinary bladder, esophagus and brain. Our Data indicate that neonatal purpura fulminans can be familial and caused by severe homozygous PC deficiency.

Since hypertension is generally accepted as a risk factor for atherosclerosis and, on the other hand, alterations in haemostasis were reported in the disease, we focused our interest on two plasma inhibitors-protein C and antithrombin III, which play critical roles in the regulation of blood clotting process. We studied 19 patients with newly diagnosed essential hypertension at ist and 2ncl degreeof the disease (acc.to WHO criteria). Protein C antigen level was measured by means of ELISA Protein C test (Boehringer Mannheim GmbH).AT III and Factor VIII R:Ag concentrations were measured by rocket immunoelectrophoresis acc.to Laurell using monospecific antisera (Behringwerke AG, Marburg). In comparison with healthy subjects the patients with essential hypertension revealed statistically significant decrease in protein C antigen level (79 t 21% vs 101 ± 16; p≺0.01) and substantially increased F. VIII R: Ag values (174 ± 72% vs 103 ± 33%; p p≺0.01). No significant correlation was found between F. VIII R:Ag and protein C level. The patients had also elevation of AT III concentration ofno statistical significance. Since F. VIII R:Ag is produced and released by endothelial cells its elevated level in patients with hypertension may reflect injuryto the vascular wall. Decreased levels of protein C probably due to its increased consumption observed in the patients may contribute to imbalance in the haemostatic system and promote vascular complications.

A concentrate of factor VII obtained by adsorption onto an inorganic adsorbent using a by-product of routine fractionation as starting material This fraction contains only small amounts of the other constituents of the prothrombin complex ( PC ) but it is enriched in proteins C and S. It contains much less VIII CAg than the usual even activated PC concentrates. The amounts of isoagglutinins anti-A and B have also been checked. The concentrate of partially versus completely activated factor VII ( by factor Xlla ) has been submitted to animal experiences including those effected on hemophilic dogs. The concentrate and highly purified factor VII prepared from it were checked on a serie of chromogenic substrates and inhibitors of proteolytic enzymes of plasmatic and other origin. The results obtained are compared with those given by the two most commonly used activated PCCs. The correlation between active F VII and by-passing activity has been studied. This concentrate might thus be indicated for the treatment of hemophilia A-patients with inhibitors.

Effective repair of vascular injury requires assembly of a secure fibrin patch at the injury site. A secure fibrin patch must be anchored to the injury site, else it becomes an embolus. Using enzyme linked immunoassay (ELISA), we have evaluated the binding of fibrinogen (F) to single as well as combinations of the subendothelial BM components including laminin (L), type IV collagen (C), and dermatan sulfate (D). Tests were performed using plates coated over night at either 4°C or 37°C for one hour with individual BM or combinations or BM components. Results indicate that F strongly binds to both D and type IV collagen. Binding of F to L was approximately 50% compared to C or D. Combinations of C, L and D were studied with mole ratios of L and D normalized to C. Thus, when the mole ratio of L was increased from 0.1 to 10 relative to C, the amount of F bound to the plate coated with C and L decreased by 30%. When both D and L were added to C and the binding of F to the coated plate evaluated, increasing the mole ratio of D from 1 to 10 relative to C and L (C:D:L=1:1:0-10), increased the binding of F two fold. If the mole ratio of L was increased by a factor of 10 compare to C, and D varied form 0 to a 10 mole excess compared to C, (C:L:D=1:10:0-10) the increase in F binding was only increased by 1/3. Therefore, L decreases the binding of F by C, while D strongly increases the binding of F. These data suggest that an important interaction between the endothelial basement membrane and fibrin exists and the mole ratio between CLD is critical in determining the extent of the interaction. The interaction between F and C,L,D may underlie the mechanism by which a fibrin clot is anchored and stabilized to the injury site.

Positive and negative regulation of platelet receptors for adhesive proteins, fibrinogen (F) and von Willebrand Factor (vWF) determines whether binding of these ligands will or will not take place. We have shown previously that ADP stimulates and cyclic AMP inhibits binding of F and vWF to human platelets. Now we show that positive regulation of F and vWF binding to platelets via the glycoprotein 11b/1111a complex is dependent on platelet Protein Kinase C, a calcium- and phospholipid-dependent enzyme. A potent activator of Protein Kinase C, phorbol-12-myristoyl-13-acetate (PMA) induced saturable and specific binding of F and vWF which was inhibited by synthetic peptides, gamma chain .dodecapeptide (gamma 400-411) and RGDS. The phosphorylation of 47kDa protein (P47), a marker of Protein Kinase C activation in platelets, preceded binding of F and vWF induced with PMA as well as with ADP and thrombin. Sphingosine, an inhibitor of Protein Kinase C, blocked binding of F and vWF to platelets stimulated with PMA, ADP, and thrombin. Inhibition of binding was concentration-dependent and it was accompanied by inhibition of platelet aggregation. Thus, stimulation of Protein Kinase C is required for exposure of platelet receptors for adhesive proteins whereas inhibition of Protein Kinase C prevents receptorexposure. Protein Kinase C fulfills the role of an intraplatelet signal transducer, regulating receptors for adhesive proteins, and constitutes a target for pharmacologic modulation of the adhesive interactions of platelets.

Endotoxic Lipid A is the biologically active principle of lipopolysaccharide of Gram-negative bacteria, a most frequent cause of sepsis underlying Disseminated Intravascular Coagulation (DIC) and shock. We have shown that endotoxic Lipid A activates Protein Kinase C in human platelets. Phosphorylation of a 47kDa protein (P47), a marker for Protein Kinase C activation, was observed within the first minute of interaction of Lipid A with platelets. This was accompanied by gradual exposure of the receptor for 125I-labeled fibrinogen (F). Binding of 125I-F was saturable and specific. When Lipid X, a precursor of endotoxic Lipid A and its competitive inhibitor, was used, the binding of 125I-F was blocked with 50% inhibition at a 1:1 stoichiometry between Lipid X and Lipid A. At the same time, phosphorylation of P47 was prevented. Since Lipid X constitutes a "half molecule" of Lipid A, we interpret these results as indicative of competitive blocking of endotoxic Lipid A in terms of Protein Kinase C activation and exposure of platelet receptors for fibrinogen. Binding of fibrinogen is necessary for platelet aggregation and endotoxic Lipid A-induced aggregation was also blocked by Lipid X. Endotoxic Lipid A-induced exposure of fibrinogen receptors via the Protein Kinase C pathway can contribute to involvement of platelets in microcirculatory thrombosis observed in patients with DIC and Gram-negative sepsis

Protein C (PC) and Factor VII (F VII) are both vitamino-K dependent factors with similar short half-life. Differential behaviour of both factors in their two expressions was assessed on :- 16 resolutive liver toxicity due to reinforced chemotherapy,- 7 specific liver toxicity of Asparaginase or Bisanthrene- 6 veno-occlusive disease complicating an allogenic bone marrow transplantation- 5 cirrhotic liver insufficiencies- 10 patients submitted to stabilised treatment by oral anticoagulant- 3 families of congenital deficiencies in F VII.In case of congenital deficiency in F VII, both expressions of this factor were parallely decreased and PC was normal. During anti-vitamin K therapy, functional levels of PC and F VII were parallely decreased, more profondly than antigenic dosages. In case of severe liver insufficiencies both expressions of both factors were parallely decreased. Evoluated V0D behaved as severe liver insufficiency. Onset of therapy by asparaginase and bisanthrene, resolutive toxicity due to reinforced chemotherapy and early phase of the V0D were characterized by a decrease of the functional activities of these factors before molecular decrease and despite a systematic preventive treatment by injection of vitamin K. Often one of these factors was more affected than the other (in these cases : PC more often than F VII). The ratio activity/antigenicity of these factors proved to be an early and sensitive marker of liver involvement. Changes occured earlier than modifications in prealbumin or in liver enzymes. Considerations of these tests to follow liver functions and specially to assess early diagnosis of VOD must be considered.

In January 2020, the US Ocean Carbon & Biogeochemistry (OCB) Project Office funded the Ocean Nucleic Acids 'omics Intercalibration and Standardization workshop held at the University of North Carolina in Chapel Hill. Thirty-two participants from across the US, along with guests from Canada and France, met to develop a framework for standardization and intercalibration (S&I) of ocean nucleic acid ‘omics (na’omics) approaches (i.e., amplicon sequencing, metagenomics and metatranscriptomics). During the three-day workshop, participants discussed numerous topics, including: a) sample biomass collection and nucleic acid preservation for downstream analysis, b) extraction protocols for nucleic acids, c) addition of standard reference material to nucleic acid isolation protocols, d) isolation methods unique to RNA, e) sequence library construction, and f ) integration of bioinformatic considerations. This report provides a summary of these and other topics covered during the workshop and a series of recommendations for future S&I activities for na’omics approaches.

Altering milk composition via transgenesis depends on three main factors. (1) The availability of an efficient regulatory sequences for targeting transgene(s) to the mammary gland; (2) a reliable in vitro model to test the expression of transgenes prior to their introduction to the animal genome; and (3) better understanding of the major factors which determine the rate of gene expression and protein synthesis. The current studies provide the necessary means and knowledge to alter milk protein composition via transgenesis. The following specific goals were achieved: a: Identifying regulatory regions in the b-lactoglobulin (BLG) gene and the cross-talk between elements which enabled us to construct an efficient vector for the expression of desirable cDNA's in the mammary gland. b: The establishment of a sheep mammary cell line that serves as a model for the analysis of endogenous and exogenous milk protein synthesis in the mammary gland of livestock. c: An accurate comparison of the potency of the 5' regulatory sequences from the BLG and whey acidic protein (WAP) promoters in directing the expression of human serum albumin (HSA) to the mammary gland in vitro and in vivo. In this study we have also shown that sequences within the coding region may determine a specific pattern of expression for the transgene, distinct from that of the native milk protein genes. d: Characterizing the dominant role of ECM in transgene expression in mammary epithelial cells. e: Further characterization of the BCE-1 enhancer element in the promoter of the b-casein gene as a binding site for the c/EBP-b and Stat5. Identifying its interaction with chromatin and its up regulation by inhibitors of histone deacetylation. f: Identifying a mechanism of translational control as a mediator for the synergistic effect of insulin and prolactin on protein synthesis in the mammary gland.

Abstract: The long-term goals of our research are to understand the regulation of sclerotial development and pathogenicity in S. sclerotior11111. The focus in this project was on the elucidation of the signaling events and environmental cues involved in the regulation of these processes, utilizing and continuously developing tools our research groups have established and/or adapted for analysis of S. sclerotiorum, Our stated objectives: To take advantage of the recent conceptual (ROS/PPs signaling) and technical (amenability of S. sclerotiorumto manipulations coupled with chemical genomics and next generation sequencing) developments to address and extend our fundamental and potentially applicable knowledge of the following questions concerning the involvement of REDOX signaling and protein dephosphorylation in the regulation of hyphal/sclerotial development and pathogenicity of S. sclerotiorum: (i) How do defects in genes involved in ROS signaling affect S. sclerotiorumdevelopment and pathogenicity? (ii) In what manner do phosphotyrosinephosphatases affect S. sclerotiorumdevelopment and pathogenicity and how are they linked with ROS and other signaling pathways? And (iii) What is the nature of activity of newly identified compounds that affect S. sclerotiori,111 growth? What are the fungal targets and do they interfere with ROS signaling? We have met a significant portion of the specific goals set in our research project. Much of our work has been published. Briefly. we can summarize that: (a) Silencing of SsNox1(NADPHoxidase) expression indicated a central role for this enzyme in both virulence and pathogenic development, while inactivation of the SsNox2 gene resulted in limited sclerotial development, but the organism remained fully pathogenic. (b) A catalase gene (Scatl), whose expression was highly induced during host infection is involved in hyphal growth, branching, sclerotia formation and infection. (c) Protein tyrosine phosphatase l (ptpl) is required for sclerotial development and is involved in fungal infection. (d) Deletion of a superoxidedismutase gene (Sssodl) significantly reduced in virulence on both tomato and tobacco plants yet pathogenicity was mostly restored following supplementation with oxalate. (e) We have participated in comparative genome sequence analysis of S. sclerotiorumand B. cinerea. (f) S. sclerotiorumexhibits a potential switch between biotrophic and necrotrophic lifestyles (g) During plant­ microbe interactions cell death can occur in both resistant and susceptible events. Non­ pathogenic fungal mutants S. sclerotior111n also cause a cell death but with opposing results. We investigated PCD in more detail and showed that, although PCD occurs in both circumstances they exhibit distinctly different features. The mutants trigger a restricted cell death phenotype in the host that unexpectedly exhibits markers associated with the plant hypersensitive (resistant) response. Using electron and fluorescence microscopy, chemical effectors and reverse genetics, we have established that this restricted cell death is autophagic. Inhibition of autophagy rescued the non-pathogenic mutant phenotype. These findings indicate that autophagy is a defense response in this interaction Thus the control of cell death, dictated by the plant (autophagy) סr the fungus (apoptosis), is decisive to the outcome of certain plant­ microbe interactions. In addition to the time and efforts invested towards reaching the specific goals mentioned, both Pls have initiated utilizing (as stated as an objective in our proposal) state of the art RNA-seq tools in order to harness this technology for the study of S. sclerotiorum. The Pls have met twice (in Israel and in the US), in order to discuss .נחd coordinate the research efforts. This included a working visit at the US Pls laboratory for performing RNA-seq experiments and data analysis as well as working on a joint publication (now published). The work we have performed expands our understanding of the fundamental biology (developmental and pathogenic) of S. sclerotioז111וז. Furthermore, based on our results we have now reached the conclusion that this fungus is not a bona fide necrotroph, but can also display a biotrophic lifestyle at the early phases of infection. The data obtained can eventually serve .נ basis of rational intervention with the disease cycle of this pathogen.

At the beginning of this project, it was clear that R. albus adhered tightly to cellulose and its efficient degradation of this polysaccharide was dependent on micromolar concentrations of phenylacetic acid (PAA) and phenylpropionic acid (PPA). The objectives for our research were: i) to identify how many different kinds of cellulose binding proteins are produced by Ruminococcus albus; ii) to isolate and clone the genes encoding some of these proteins from the same bacterium; iii) to determine where these various proteins were located and; iv) quantify the relative importance of these proteins in affecting the rate and extent to which the bacterium becomes attached to cellulose. BARD support has facilitated a number of breakthroughs relevant to our fundamental understanding of the adhesion process. First, R. albus possesses multiple mechanisms for adhesion to cellulose. The P.I.'s laboratory has discovered a novel cellulose-binding protein (CbpC) that belongs to the Pil-protein family, and in particular, the type 4 fimbrial proteins. We have also obtained genetic and biochemical evidence demonstrating that, in addition to CbpC-mediated adhesion, R. albus also produces a cellulosome-like complex for adhesion. These breakthroughs resulted from the isolation (in Israel and the US) of spontaneously arising mutants of R. albus strains SY3 and 8, which were completely or partially defective in adhesion to cellulose, respectively. While the SY3 mutant strain was incapable of growth with cellulose as the sole carbon source, the strain 8 mutants showed varying abilities to degrade and grow with cellulose. Biochemical and gene cloning experiments have been used in Israel and the US, respectively, to identify what are believed to be key components of a cellulosome. This combination of cellulose adhesion mechanisms has not been identified previously in any bacterium. Second, differential display, reverse transcription polymerase chain reaction (DD RT-PCR) has been developed for use with R. albus. A major limitation to cellulose research has been the intractability of cellulolytic bacteria to genetic manipulation by techniques such as transposon mutagenesis and gene displacement. The P.I.'s successfully developed DD RT- PCR, which expanded the scope of our research beyond the original objectives of the project, and a subset of the transcripts conditionally expressed in response to PAA and PPA have been identified and characterized. Third, proteins immunochemically related to the CbpC protein of R. albus 8 are present in other R. albus strains and F. intestinalis, Western immunoblots have been used to examine additional strains of R. albus, as well as other cellulolytic bacteria of ruminant origin, for production of proteins immunochemically related to the CbpC protein. The results of these experiments showed that R. albus strains SY3, 7 and B199 all possess a protein of ~25 kDa which cross-reacts with polyclonal anti-CbpC antiserum. Several strains of Butyrivibrio fibrisolvens, Ruminococcus flavefaciens strains C- 94 and FD-1, and Fibrobacter succinogenes S85 produced no proteins that cross-react with the same antiserum. Surprisingly though, F. intestinalis strain DR7 does possess a protein(s) of relatively large molecular mass (~200 kDa) that was strongly cross-reactive with the anti- CbpC antiserum. Scientifically, our studies have helped expand the scope of our fundamental understanding of adhesion mechanisms in cellulose-degrading bacteria, and validated the use of RNA-based techniques to examine physiological responses in bacteria that are nor amenable to genetic manipulations. Because efficient fiber hydrolysis by many anaerobic bacteria requires both tight adhesion to substrate and a stable cellulosome, we believe our findings are also the first step in providing the resources needed to achieve our long-term goal of increasing fiber digestibility in animals.
