AbstractThis study reports on the chromosome number and karyological characteristics of the endangered species of European crayfish, Astacus astacus and A. leptodactylus (Decapoda, Astacidae), both native to Croatian freshwater habitats. The karyotype of A. astacus and A. leptodactylus consists of 2n = 176 and 2n = 180 chromosomes, respectively. The haploid chromosome complement of A. astacus consists of 52 metacentric, 35 metacentric-submetacentric, and 1 acrocentric chromosomes. Fluorochrome staining with 4,6-diamino-2-phenylindole (DAPI) has revealed that the karyotypes of A. astacus and A. leptodactylus are characterized by large heterochromatic blocks located at centromeric and intercalary positions on the chromosomes. Interstitial heterochromatic blocks were more frequent in A. astacus than in A. leptodactylus. In both species pairing of chromosomes in meiosis was regular with the majority of bivalents in a ring- and a dumbbell-form. Fluorescence in situ hybridization (FISH) has revealed that two 45S rDNA loci were present in the investigated species. In A. astacus one of the two 45S rDNA-bearing chromosome pairs was highly heteromorphic, exhibiting a three-fold size difference between 45S rDNA sites on homologous chromosomes. Such a size difference was significantly less pronounced in A. leptodactylus. The karyotype differences between A. astacus and A. leptodactylus suggest changes in chromosome number as well as position of repetitive DNAs have played a role in the karyotype evolution of the species of Astacus.

Abstract Morphology in general and developmental morphology in particular provide us with an insight into the evolution, phylogeny, ecological features, behavior, and life cycle of organisms. The present study describes the external morphology of the first three postembryonic developmental stages (Stage 1­3) of two prominent European crayfish species, the noble crayfish Astacus astacus (Linnaeus, 1758) and the narrow­clawed crayfish Pontastacus leptodactylus (Eschscholtz, 1823). External morphology and setation of appendages are similar, presence of re­curved tips for cheliped in Stage I and then the tip lost in Stage 2, long plumose setae on the telson in Stage 2 and then the setae disappearance in Stage 3 accompanied with the fully developed tail fan. The number of segments on antennal flagella differs between the species in Stage I and Stage II. Observed findings on early postembryonic development are consistent between European and North American (Pacifastacus sp.) astacid species, but differ in certain aspects from other crayfish families.

Pontastacus leptodactylus (Eschscholtz, 1823) (Decapoda:Astacidea:Astacidae) constitutes an ecologically and economically highly important species. In the present study, the mitochondrial genome of the freshwater crayfish P. leptodactylus from Greece is analyzed for the first time, using 15 newly designed primer pairs based on available sequences of closely related species. The analyzed coding part of the mitochondrial genome of P. leptodactylus consists of 15,050 base pairs including 13 protein-coding genes (PCGs), 2 ribosomal RNA gene (rRNAs), and 22 transfer RNA genes (tRNAs). These newly designed primers may be particularly useful in future studies for analyzing different mitochondrial DNA segments. Based on the entire mitochondrial genome sequence, compared to other haplotypes from related species belonging in the same family (Astacidae) available in the GenBank database, a phylogenetic tree was constructed depicting the phylogenetic relationships of P. leptodactylus. Based on the results, the genetic distance between Astacus astacus and P. leptodactylus is smaller than the genetic distance between Austropotamobius pallipes and Austropotamobius torrentium, despite the fact that the latter two are classified within the same genus, questioning the phylogenetic position of A. astacus as a different genus than P. leptodactylus. In addition, the sample from Greece seems genetically distant compared with a conspecific haplotype available in the GenBank database, possibly implying a genetic distinction of P. leptodactylus from Greece.

We studied the presence of possible Aphanomyces astaci infections in eight Turkish narrow-clawed crayfish (Astacus leptodactylus) populations by analyzing the prevalence and genotypes of the disease agent A. astaci. The qPCR analyses revealed A. astaci infection in seven of the studied eight populations, with the agent level A2 or higher. The agent levels among the infected populations varied from A0 to A5, i.e., from negative to high level of infection, based on qPCR ranking. Based on the sequencing of the chitinase gene and the mitochondrial ribosomal rnnS and rnnL subunits, we detected both A (As) and B (PsI) haplogroups of A. astaci in our samples, with each of the studied populations being carriers of only one haplotype. The results confirm previous detections of A. astaci in Turkish narrow-clawed crayfish populations and reveal, that both A and B haplogroup A. astaci carriers exist widely in A. leptodactylus populations of Turkey.

Several morphological variations in external morphology were detected, and the characters to discriminate between Astacus and Pontastacus have been used to update the diagnoses of these two genera.

Abstract Heterochely is an important feature in some marine decapod crustaceans, but it is seldom investigated in freshwater crayfish. In this study, we applied a biometrical analysis targeting wild populations of three European crayfish species, Astacus leptodactylus, Astacus astacus, and Austropotamobius torrentium, as well as one invasive North American species, Faxonius limosus. Field data were combined with video-recorded observations to understand the usage of chelae in laboratory experiments for A. leptodactylus and F. limosus. According to biometrical measurements, heterochely was evenly distributed between species and sexes in wild populations, leading to the assumption that there is no specific pattern in chela size. Moreover, we found that the ambidextrous usage of chelae is a commonly encountered behaviour in crayfish, since no significant relationship was found between their chelae and asymmetry. This behaviour could maximize the chances of survival for crayfish in general, since losing one or both chelae is often recorded in the wild.

The reaction of hydroxyurea with the oxyhaemocyanins of Astacus leptodactylus and Helix pomatia yielded methaemocyanins which could be regenerated with hydroxylamine. Hydroxyurea did not react with Astacus methaemocyanin, but quantitatively regenerated Helix methaemocyanin under N2. The reaction of hydroxyurea with Helix haemocyanin at pH 5.7 under air thus led to a steady state, with an oxyhaemocyanin/methaemocyanin ratio of 2.05:1.

One of the most critical challenges in species distribution modelling is testing and validating various digitally derived environmental predictors (e.g., remote-sensing variables, topographic variables) by field data. Therefore, here we aimed to explore the value of soil properties in the spatial distribution of four European indigenous crayfish species. A database with 473 presence and absence locations in Romania for Austropotamobius bihariensis, A. torrentium, Astacus astacus and Pontastacus leptodactylus was used in relation to eight digitalised soil properties. Using random forest modelling, we found a preference for dense soils with lower coarse fragments content together with deeper sediment cover and higher clay values for A. astacus and P. leptodactylus. These descriptors trigger the need for cohesive soil river banks as the microenvironment for building their burrows. Conversely, species that can use banks with higher coarse fragments content, the highland species A. bihariensis and A. torrentium, prefer soils with slightly thinner sediment cover and lower density while not influenced by clay/sand content. Of all species, A. astacus was found related with higher erosive soils. The value of these soil-related digital descriptors may reside in the improvement of approaches in crayfish species distribution modelling to gain adequate conservation measures.

Abstract We examined gills (podobranchs, arthrobranchs and pleurobranchs) from five European Astacidae (Austropotamobius pallipes, Austropotamobius torrentium, Astacus astacus, Pontastacus leptodactylus and Pontastacus pachypus) to assess the relationships between their phylogeny and gill morphology and formulae. In addition, we checked for the presence or absence of pleurocoxal lappets. The morphology of the gills and associated structures was similar in all examined European astacid species except for three rudimentary pleurobranchs found in P. pachypus. In European Astacidae and Cambaroididae, the pleurocoxal lappets are absent but are present in the American Astacidae, Cambaridae and Parastacidae. The number of pleurobranchs decreases from the most primitive Parastacidae to the most advanced Cambaridae. Nevertheless, although the most primitive parastacid crayfish and the most advanced Cambaridae all have pleurocoxal lappets, they are absent in European Astacidae and Cambaroididae, which suggests that a reversible gill morphology has occurred in crayfish evolution.

Astacus leptodactylusEschscholtz, 1823 is an important, marketable species of crustacean, consumed intensely as a luxury food in many countries. BecauseA. leptodactylusis less cannibalistic than other crayfish species, is more fecund, and has a fast growth rate, it is accepted as a considerably suitable crayfish for culturing in extensive pond systems. Information on the reproductive biology and anatomy of crayfish is important for developing suitable production methods. Sex recognition may also be essential in separating males and females in crayfish rearing (i.e., to prevent cannibalism, males grow faster than females). Although male reproductive system morphology and spermatophore formation in crayfish, includingA. leptodactylus, have been studied in detail, this study is the first to demonstrate the visibility of precursors of the gonopods and, thus, the actual presence of gonopods, which of course determine the male sex, in a freshwater crayfish. The present study shows that the first signs of secondary sexual characters (i.e., precursors of the gonopods) can be distinguished in the form of small tubercles on the first abdominal somite in male stage 4 juveniles, and the gonopods themselves are distinguishable in male stage 5 juveniles inA. leptodactylus. In conclusion, sex differentiation inA. leptodactyluscan be realized already in stage 4 juveniles. Further studies should be carried out on the presence and shape of precursors of gonopods in stage 4 and gonopods in stage 5 juveniles of species of astacid crayfish in order to facilitate solving taxonomic problems in the family Astacidae.

Analysis of Croatian freshwater crayfish populations, aiming to gather new distributional data and complement previous surveys (2005–2011), was performed during 2014–2016, within the frame of Natura 2000 Integration Project. The research included different waterbodies across the whole of Croatia, harbouring both indigenous and non-indigenous crayfish species (ICS and NICS, respectively). Field work was conducted in 117 grid squares with dimensions 10 × 10 km, and up to four waterbodies were surveyed per grid square, making a total of 450 studied sites. Out of those, crayfish were not recorded in 368 sites. In the sites with crayfish presence (i.e., 82), the most frequently observed ICS was Astacus astacus (registered in 33 sites) followed by Austropotamobius pallipes (16 sites), Austropotamobius torrentium (12 sites), and Astacus leptodactylus (5 sites). Concerning NICS, the majority of records were for Orconectes limosus (13 sites), followed by Pacifastacus leniusculus (2 sites), whereas Procambarus fallax f. virginalis was registered in only one locality. Comparisons of obtained data with those from previous surveys showed that NICS are progressively spreading and displacing ICS, as recorded for A. leptodactylus that was almost completely displaced by O. limosus in waterbodies of the east Croatia. Existing ICS populations are under growing anthropogenic preassure, frequently fragmented and isolated. Moreover, this survey showed that in the last decade 55% of A. astacus and 67% of A. pallipes populations disappeared mainly as a consequence of anthropogenic influence onto their habitats. Further monitoring and conservation actions for ICS should be urgently applied to mitigate negative impacts of both NICS and anthropogenic influence.

The narrow-clawed crayfish (Pontastacus leptodactylus) is an alien species in Finland with only a few populations reported from the southeastern region during the last century. We discovered a productive population in the lake Jängynjärvi, which is upstream from the previously reported wild narrow-clawed crayfish population in that region. Preliminary studies indicated that this population is not infected with Aphanomyces astaci. We collected narrow-clawed crayfish samples from the lake Jängynjärvi population for both infection challenge and genetic studies, in order to investigate possible A. astaci resistance among this Finnish population and to evaluate their phylogenetic position that would enable us to speculate different scenarios of distribution pathways or origin of the population. The infection studies indicated that the narrow-clawed crayfish in this population were more resistant against A. astaci infection (B haplogroup A. astaci) compared to the noble crayfish (Astacus astacus) from the lake Rytky in North Savo, while all crayfish of both species in the B haplogroup A. astaci challenged groups died within 58 days post-infection. Results of the phylogenetic reconstruction indicate that the lake Jängynjärvi narrow-clawed crayfish are closely related to narrow-clawed crayfish from the lake Bolshoye near Krasnoye, located on the White Sea island of Solovestky and also populations from Tyumen region, both in Russia. This could confirm previous speculations about introduction of the narrow-clawed crayfish from Russia into Finland or could indicate previous hydrological connection.

Five species of crayfish, comprising two native species Austropotamobius torrentium and Astacus astacus, one introduced European species, Astacus leptodactylus and two alien American species, Orconectes limosus and Pacifastacus leniusculus, occur in open waters of the Czech Republic. The stone crayfish is highly endangered species in the country, known only from several recent records. Four localities have been confirmed for the presence of the stone crayfish in the Czech Republic. Basic characteristics of the following stone crayfish occurring localities are described: Míza and Upořský brook in Křivoklát protected area, Zubřina brook near Domažlice, Luční brook near Trutnov and Klabava brook in Brdy Military Training Area. The present localities have been investigated for chemical, structural and terrestrial environments. Properties of these waters (morphology, supply of substrate, current, aquatic and terrestrial environment) are compared with literature data. Strong conservation actions are necessary to save the endangered stone crayfish on most north-eastern limit of its distribution.

The reaction of nitrite at pH 5.7 with deoxyhaemocyanin of Astacus leptodactylus yielded methaemocyanin in two one-electron steps, as nitrite was reduced to NO. This methaemocyanin could be almost fully regenerated by an anaerobic treatment with HONH2, in contrast with the methaemocyanin prepared with H2O2. A destruction of active sites on treating oxyhaemocyanin with HONH2 explains the partial regeneration of methaemocyanin under air, as traces of H2O2 are formed in the autoxidation of HONH2. The reaction rate of nitrite with deoxyhaemocyanin is almost 15 times that with oxyhaemocyanin. The slope of -1.0 for the logarithm of the pseudo-first-order rate constants plotted against pH indicates that HNO2 is the reacting species. Methaemocyanin was e.p.r.-undetectable, but a binuclear signal was observed at g = 2 on binding nitrite to methaemocyanin. This signal disappeared with a pKa of 6.50, suggesting that a mu-aquo bridging ligand, which can be replaced by nitrite, is deprotonated to a mu-hydroxo bridging ligand, which resists substitution by nitrite. The intensity of this triplet e.p.r. signal allowed the determination of the association constant of nitrite to the active site of Astacus methaemocyanin and yielded a value of 237 M-1 at pH 5.7. The interpretation by some authors of nitrosylhaemocyanin as a nitrite derivative of semimethaemocyanin is contradicted by this rapid reaction of nitrite with copper(I) in deoxyhaemocyanin and in semi-methaemocyanin and by the low binding constant of nitrite to the active site of methaemocyanin.

In this study, it was aimed to determine there and fecundity by adapting crayfish (Astacus leptodactylus) obtained from Bafra Fish Lakes (Ulugöl-Samsun) in culture conditions. Crayfish obtained in November 2016 were stocked in tanks and egg and morphometric characteristics of female crayfish with carrying eggs were determined. Correlation analysis were used for the relationship between the determined parameters. The mean total weight and total length of crayfish were 41.79±3.00g, 10.72±0.22cm, respectively whereas the mean number and diameter of eggs were 156.40±33.24 and 2.50±0.12mm. There were positive strong relationship between total length and total weight of crayfish negative relationship was found between egg diameter and total number of eggs, egg diameter and total weight of eggs and egg diameter and number of eggs for per unit of live weight.

Abstract Due to the increasing anthropogenic pressures and the introduction of invasive alien species, indigenous crayfish species are rarely found in the urban areas of big cities. The aim of this research was to study, for the first time, presence and distribution of freshwater crayfish in artificial lakes of the Maksimir Park, located in the centre of Zagreb, the capital of Croatia. Additionally, the goal was to estimate population size and structure, and analyse morphometric characteristics, the proportion of injuries and crayfish body condition. In 4 out of 5 lakes within the park, two crayfish species, Astacus astacus and Astacus leptodactylus, were recorded with no overlap in their distribution. Each species was established in two lakes in relatively abundant populations. Differences in body condition, sex ratio and proportion of injuries were recorded between species. Recorded differences are most likely the result of different trapping techniques applied, predation of alien invasive species of turtles and intraspecific competition. Obtained results will serve as a baseline for further research and monitoring of the crayfish fauna in the Maksimir lakes.

Thiamethoxam (Thmx) is a globally used neonicotinoid pesticide contaminated in freshwater ecosystems with residues detected in fishery products. Astacus leptodactylus is a popular freshwater crustacean that is cultivated and exported in many countries. In this study, we investigated the acute toxic effects of Thmx on A. leptodactylus using various biomarkers (acetylcholinesterase, carboxylesterase, glutathione S-transferase, glutathione, superoxide dismutase, glutathione peroxidase, glutathione reductase, and adenosinetriphosphatases). The 96-h LC50 value of Thmx was calculated as 8.95 mg active ingredient L–1. As the dose of Thmx increased, oxidative stress was induced by the inhibition/activation of antioxidant enzymes, while the activities of acetylcholinesterase, carboxylesterase and adenosinetriphosphatases were inhibited. As a result, it can be said that Thmx has highly toxic effects on crayfish, therefore they are under threat in the areas where this pesticide is used.

The thick–clawed crayfish Pontastacus pachypus (Rathke, 1837) is an endangered, and the least known, crayfish species in Europe. Currently, detailed information regarding the morphology, ecology and reproduction of thick–clawed crayfish is outdated. This study presents, for the first time, detailed photographs of the thick-clawed crayfish, and updated information on distinctive morphological characters and morphometric analysis. New specifications of the carapace and appendage morphological characteristics were established as: 1) the rostrum is long, sharply pointed and has three pairs of distinctive sub-apical lateral spines, 2) two well–developed pairs of post–orbital ridge on the carapace are ended by prominent spine; 3) each finger of chelae ends with a black sharp tip. Among the 18 morphometric indices, carapace width to the total length (CPW/TL), abdomen width to the total length (ABW/TL) and claw height to the claw width (CLH/CLW) clearly differentiate P. pachypus from the other representatives of Astacus genus (A. colchicus and A. astacus) and P. leptodactylus (P<0.05). Comparison of individual indices between P. pachypus and P. leptodactylus revealed that almost all indices differed significantly except head length to the total length (HEL/TL) and rostrum length to the total length (ROL/TL). This study contributes to the identification of the thick–clawed crayfish for the purpose of conservation and protection of its localities.

The rate of the reaction of Astacus leptodactylus methaemocyanin with NO follows the Henderson-Hasselbalch equation with a pKa of 5.85, suggesting that one imidazole ligand of Cu was exchanged for NO. The reaction is blocked by F- as a bridging ligand. The same imidazole residue might be responsible for the decomposition of nitrosylhaemocyanin, [Cu1NO+CuII], with an unlocated binding site for NO, into methaemocyanin and NO, as the rate increase with pH. NO could react preferentially with CuA of Helix pomatia methaemocyanin, CuA′IICuBII, as it possibly has only two histidine ligands instead of the three of CuA in Astacus haemocyanin. This difference might explain the higher formation rate and the much greater stability of Helix nitrosylhaemocyanin. The fast reaction is governed by a pKa of 6.80, probably of a bridging mu-aquo ligand. With F- or a mu-hydroxo bridging ligand a low reaction rate was still observed, in contrast with Astacus methaemocyanin. Helix nitrosylhaemocyanin was transformed by N3- into methaemocyanin with the liberation of N2 and N2O. This methaemocyanin could almost quantitatively be regenerated with H2O2. Helix nitrosylhaemocyanin was only partially regenerated by a direct treatment with H2O2 and almost quantitatively by HONH2 in a similar fairly fast reaction, followed by a much slower one.

Строительство и последующее использование водохранилищ стало ведущим фактором проникновения и распространения чужеродных видов в Обь-Иртышском бассейне. Фауна чужеродных видов Новосибирского водохранилища представлена пятью видами, три из которых (Gmelinoides fasciatus Steb., Micruropus possolskii Sowinsky и Neomysis intermedia (Czerniavsky)) были преднамеренно вселены для повышения кормовой базы водоема, еще два (Viviparus viviparus (L.) и Astacus leptodactylus Eschscholtz) появились в результате непреднамеренных интродукций. Прослежена история проникновения и натурализации каждого вида, проанализированы экологические факторы их обитания, рассмотрены особенности современного пространственного распределения вселенцев. Отмечено, что несмотря на невысокое видовое богатство, вселенцы существенно влияют на структурные характеристики донных сообществ водохранилища.

Narrow-clawed crayfish (Astacus leptodactylus Eschscholtz, 1823 species-complex) is one of five European freshwater crayfish species. Even though widely distributed, it hasn’t been frequently studied and its taxonomy and systematics are unresolved. The results of a recent comparative morphometric character study revealed that morphometry of Asian and European populations differ significantly. In this research, for the first time, mitochondrial molecular markers (16S rRNA and COI) were used with the aim of elucidating the phylogenetic relationship between European and Asian populations of the narrow-clawed crayfish. Analyses included crayfish from Croatia, Bulgaria, Armenia, Russia, Poland and Turkey, and three different optimality criteria were applied. Phylogenetic relationships were reconstructed using the COI dataset, as well as the concatenated one (COI + 16S rRNA). For both datasets, congruent topologies were obtained and trees were characterised by the existence of two well supported phylogroups, one that included European populations, and the other Asian. Results indicate the presence of distinct evolutionary lineages within the A. leptodactylus species-complex, and corroborate previous results obtained using morphometric analyses.

Midgut gland and hypodermis of the crayfish Astacus leptodactylus have been cultured in a serum-free medium for several days. The medium consists of 1 part of van Harreveld solution and 1 part of an amino acid mixture supplemented with 1.2 mᴍ Na2HP04, 12 mᴍ Hepes and 80 mᴍ glucose. The antibiotics penicillin (15 mg/l) and streptomycin (25 mg/1) were added for long term culturing. This medium, called TG medium, allows the maintainance of the tissues for more than 100 h without any loss of their viability with respect to protein synthesis and secretion.
