Abstract For many lysosomal storage disorders, presymptomatic detection, before the onset of irreversible pathology, will greatly improve the efficacy of current and proposed therapies. In the absence of a family history, presymptomatic detection can be achieved only by a comprehensive newborn screening program. Recently we reported that the lysosome-associated membrane protein LAMP-1 was increased in the plasma from ∼70% of individuals with lysosomal storage disorders. Here we report on the evaluation of a second lysosome-associated membrane protein, LAMP-2, as a marker for this group of disorders. The median concentration of LAMP-2 in the plasma of healthy individuals was 1.21 mg/L, fourfold higher than the median LAMP-1 concentration (0.31 mg/L). LAMP-2 was increased in >66% of patients with lysosomal storage disorders, and the increases coincided with increased LAMP-1 concentrations. The reference intervals for LAMP-1 and LAMP-2 in blood spots taken from newborns were 0.20–0.54 mg/L (n = 1600) and 0.95–3.06 mg/L (n = 1600), respectively. A high correlation was observed between the concentrations of LAMP-1 and LAMP-2 in both control and affected individuals. The higher concentrations of LAMP-2, relative to LAMP-1, in plasma make LAMP-2 an attractive marker; however, the final selection will be dependent on the availability of new diagnostic markers and their ability to detect disorders currently not identified by LAMP-2.

LAMP-1 (lysosome-associated membrane protein), a major glycoprotein present in the lysosomal membrane, constitutes up to 50% of total membrane proteins. LAMP-1, expressed at the plasma membrane, is reported to be the major molecule expressing the sialyl-Lewis X antigen. Two forms of LAMP-1 exist; the full-length LAMP-1 [LAMP-1 (+Tail)] has a highly glycosylated lumenal domain, a membrane-spanning domain and a short cytoplasmic tail, and the truncated LAMP-1 [LAMP-1 (−Tail)] contains only the lumenal domain. Soluble LAMP-1 (±Tail) has been reported in circulation. LAMP-1 at the cell surface has been shown to interact with E-selectin and galectin and is proposed to function in cell–cell interactions. However, the functional role(s) of soluble LAMP-1 in circulation is unclear. To investigate the functional role of soluble LAMP-1 in circulation, recombinant LAMP-1 (−Tail) and LAMP-1 (+Tail) were produced in HT1080 cells. Two immune-quantification assays were developed to distinguish between the LAMP-1 forms. The interaction and aggregation properties of the different LAMP-1 forms were investigated using the immune-quantification assays. Only LAMP-1 (+Tail) was found to aggregate and interact with plasma proteins. Plasma proteins that interact with LAMP-1 were isolated by affinity chromatography with either the recombinant LAMP-1 (−Tail) or a synthesized peptide consisting of the 14 amino acids of the LAMP-1 cytoplasmic tail. Transthyretin was found to interact with the cytoplasmic tail of LAMP-1. Transthyretin exists as a homotetramer in plasma, as such may play a role in the aggregation of LAMP-1 in circulation.

The ability for motorcycle to be detected is an important aspect in preventing crash involving motorcycle which is the most dominant vehicle in emerging countries. Widely referred as conspicuity, the crash factor is appropriately addressed by the introduction of mandatory daytime running light (DRL) law and is usually a success story in many parts of the world. In 2011, there was a motion introduced in the 64thsession of the United Nations Working Party on Lighting and Light-Signalling (GRE) for amber position lamp (APL) to be made mandatory on motorcycle as additional measure to improve motorcycle conspicuity. An experiment was designed to evaluate conspicuity level of motorcycle headlamp and tail lamp equipped with APL over motorcycle with present DRL setting (baseline). 15 participants simultaneously rated both motorcycles which are placed in parallel, at different distances and times of day. Motorcycle with APL was noticeably better detected from rear than front at 50 meter and 100 meter distance, as well as during night time and twilight. Median conspicuity level between night time and daytime and between night time and twilight was also distinctly different for rear lamp. These findings suggest that APL introduction could enhance motorcycle conspicuity especially for rear lamp position.

Abstract Platelets undergo biochemical and morphologic changes when stimulated that greatly alter their function and contribute to their role in thrombosis and hemostasis. We recently identified and cloned the cDNA for a platelet surface glycoprotein expressed on activated, not resting cells. We found that this protein, lysome-associated membrane protein-1 (LAMP-1), is an integral membrane protein of the lysosome that translocated to the surface membrane when platelets were stimulated by a strong agonist. We now show with immunofluorescence flow cytometry that LAMP-2, a lysosomal membrane protein that shares approximately 30% homology with LAMP-1, is also expressed preferentially on the surface of activated platelets. Equilibrium binding studies with 125I-anti-LAMP- 2 IgG showed approximately 1,100 binding sites per thrombin-stimulated platelet and less than 50 per resting platelet. Sucrose gradient ultracentrifugation fractionation of resting platelet sonicates showed that LAMP-2 colocalized with LAMP-1 and with lysosomal enzymes, and not with thrombospondin or serotonin, which are markers of the two other platelet granule compartments, alpha-granules and dense granules. LAMP- 2 surface expression was minimal in response to platelet stimulation by weak agonists such as epinephrine and ADP. These data show that LAMP-2, like LAMP-1, translocates from the lysosomal membrane compartment to the surface membrane when platelets are activated. Regulated surface expression of these heavily glycosylated proteins may play a role in the adhesive, prothrombotic phenotype of these cells.

Acute myeloid leukemias with t(8;21) or inv(16)/t(16;16) are commonly referred to as Core-Binding Factor AML and represent 5-8% of all AMLs . These genomic rearrangements are characterized by disruption of the CBF and CBF genes respectively. Both genes encode a subunit of core-binding factor complex, an important transcriptional regulator of hematopoiesis involved in myeloid differentiation. CBF AML are considered a favorable AML risk group based on high remission rate and survival probabilities. However, the presence of the cKIT D816V mutation has been reported to harbor an unfavorable prognostic implication in CBF-AML. The proto-oncogene cKIT encodes a receptor for SCF. It belongs to the type-III receptor of the tyrosine kinase subfamily, that is characterized by five extracellular immunoglobulin-like domains, a single transmembrane helix, a cytoplasmic juxtamembrane domain, and a kinase domain. The cKIT D816V mutation is found in 20–25% of patients with t(8;21) and in about 30% with inv(16)/t(16;16). cKIT D816V causes the indipendent-ligand activation of the tyrosine kinase receptor cKIT, and the activation of downstream pathways supporting cell proliferation and survival. CBF AML patients exhibiting D816V mutation in cKIT gene, are classified in the intermediate risk category, since they present an high incidence of relapse and a low overall survival. Moreover the replacement of the aspartic acid residue with valine at codon 816 causes resistence to Imatinib, the tyrosine kinase inhibitor usually used in the treatment of CBF AML to suppress the overexpressed cKIT. In this case, patients are treated with TKIs of second generation, such as Dasatinib or Nilotinib. So, the single point mutation D816V in cKIT gene, represents a critical prognostic factor for CBF AML patients and an important indicator to define the appropriate treatment. Several PCR-based methods are developed for the detection of D816V mutation, such as PCR followed by direct DNA sequencing, enzymatic digestion with HinfI enzyme (RFLP), or dHPLC. However, most of the PCR-based methods result expensive, time-consuming and labor-intensive. At the moment Sanger sequencing is still the gold-standard for D816V detection, even if it presents a low sensitivity (10–15%). The aim of this work is to improve the detection of cKIT D816V mutation in CBF AML patients by the development of rapid, user-friendly and cost-effective assay based on Allele-Specifc Loop mediated isothermal AMPlification (AS-LAMP) technology. The AS-LAMP assay is based on LAMP technology that amplifies target DNA at costant temperature thanks to the employment of the DNA polymerase with strand-displacement activity. AS-LAMP assay is able to detect the single point mutation thanks to the cooperation of two elements: a loop primer specific for the mutated cKIT sequence, and a blocker that binds only the wild type gene. While the blocker suppresses the aspecific amplification of wild type alleles, the LB primer recognizes the mutated codon and allows its amplification. The AS-LAMP assay includes also the amplification of the housekeeping ABL gene as internal control, that is informative to exclude false-negative results due to the failure of DNA extraction procedure. The target and the IC are detected in real-time in 500 nm and 530 nm channels respectively, thanks the use of two labeled probes specifically binding cKIT or ABL genes. The AS-LAMP assay was optimized on synthetic plasmid containing the cKIT sequence and was successfully validated on negative and positive clinical samples, confirming in all cases the results obtained with PCR-based methods. In conclusion the AS-LAMP assay is able to detect the D816V mutation with high sensitivity and specificity in only 40 minutes and it is easy-to-use. For these reasons the AS-LAMP assay represents a powerful tool for molecular diagnostics applications in clinical routine.

Gastrointestinal parasites Haemonchus contortus and Fasciola hepatica are major impediments to livestock production worldwide. Faecal egg counts remain the most commonly used and widely accepted diagnostic tool for these parasites; however, there is a need for improved, field-applicable diagnostics. In this study, loop-mediated isothermal amplification (LAMP) was optimised and evaluated for the detection of H. contortus (in sheep) and F. hepatica (in cattle) infection. LAMP assays were optimised to enable visual detection using calcein dye. DNA extraction techniques were developed that have the potential for on-farm application. Faeces suspended in water, heated, then centrifuged, with two cheap and stable chemicals, enabled detection of H. contortus at clinically relevant infection burdens. For F. hepatica, a faeces-water suspension was sieved to remove particulate matter, then physical disruption (bead-beating) was applied. LAMP was conducted under laboratory conditions and in the field; compared to FEC (the most commonly used diagnostic for the target parasites) and PCR. LAMP was conducted using three incubation methods: a commercially available thermocycler; a field-friendly low-cost portable styrofoam esky; and a dedicated field applicable LAMP incubator. The general trend was for LAMP to have high sensitivity but only moderate specificity when compared to FEC. However, the use of PCR (both pathogens) and a highly sensitive amended FEC (F. hepatica only) suggested that the apparent low specificity was the result of LAMP being able to detect low-level parasite infection when conventional FEC could not. A LAMP assay paired with a potentially field-applicable DNA extraction was able to adequately detect haemonchosis at ‘clinically relevant’ parasite burdens in a laboratory study. A field study for the detection of F. hepatica demonstrated the potential utility of LAMP on-farm. The studies conducted in this thesis demonstrate the potential of LAMP for parasitic disease diagnosis in agriculture.

Current diagnostic tools for malaria lack the sensitivity to identify individuals with low-density infections. Asymptomatic low-density infections are common in malaria endemic regions and these individuals provide an important reservoir of infection that enables transmission to mosquitoes. Failure to detect these individuals threatens the global health goal of malaria elimination. Loop-mediated isothermal amplification (LAMP) is a technique to amplify DNA and has the potential to diagnose these individuals. The LAMP assay was assessed in a field study in rural Vietnam. 5421 samples were collected and tested with a commercially available LAMP assay in Commune Health Care Centres in Binh Phuoc and Dak Nong Provinces. 101 positive LAMP cases (asymptomatic, smear, and RDT negative) were identified, with the proportion of positives ranging from 0.18% and 3.25% across five communes. In order for LAMP to be used as a screening tool, it must be cost effective and have a workflow suitable for minimally trained end users. To achieve this, an in-house LAMP assay was developed and compared to PCR. The assay was combined with instrument detection to simplify decision making for the end user and improve sensitivity. The in-house assay was as sensitive as the PCR assay and cost US$0.60 per reaction compared to US$3.57 for PCR and US$8.23 for the commercial LAMP. An integrated single cartridge, called T1, was assessed to further simplify this workflow of sample preparation, LAMP amplification and detection. Further development of the cartridge and the assay will be required for future deployment. The LAMP assay is suitable for detection of low density infections in asymptomatically infected individuals in field settings and has the potential for cost effective population based screening.

Windows Azure Platform is product of the Microsoft company aimed to host web applications. With this product, Microsoft is reacting on the new trend in IT -- cloud computing. This diploma thesis deals with all the aspects of developing and running PHP applications on Windows Azure Platform. Thesis is divided into two parts -- theoretical and practical. In the theoretical part, Windows Azure Platform is described in detail. The description consists of it's main parts, position in the world of cloud computing, price and main competitors. In the practical part, migration of school PHP application developed during 4IT445 course from LAMP platform to Windows Azure Platform is described. Except the migration itself, the benefits of new platform to application are described. In the thesis conclusion, development and running applications on both platforms is compared. The target of the thesis is to write first complete Czech text about running PHP applications on Windows Azure Platform and to help PHP developers to chose which way to go.

This work presents an efficient lighting system using high intensity light emitting diodes (LEDs). The goal is to develop a compact and low cost electronic circuit to drive and control the current of LEDs disposed in a single enclosure. The proposal aims energy saving, high luminous efficacy and high useful life when replacing traditional lighting systems, as dichroic, incandescent or compact fluorescent lamps by LEDs without any change in electrical system installation. Furthermore, an emergency lighting system integrated into the lamp is proposed, allowing the use of the same equipment in the daily activities, supplied by mains, and under a possible break in the electrical network. To supply the LEDs through the mains with universal input voltage and charge the battery, a Buck converter was employed. To supply the LEDs with a continuous current through the battery, a Boost converter was used. The converters operate at high frequency and control the current through the LEDs independently. The battery can be composed of three rechargeable Ni-MH batteries (1,2 V) or a Li-ion battery (3,6 V). The design attends the Brazilian standard and international standard for emergency lighting systems. To validate the proposed idea, the circuits were designed and implemented. The experimental results are shown with the intention to prove the satisfactory behavior of the lamps.
Este trabalho apresenta um sistema de iluminação eficiente empregando diodos emissores de luz (LEDs) de alta intensidade. O objetivo é desenvolver um circuito eletrônico, compacto e de baixo custo, para acionamento e controle da corrente dos LEDs contidos em um único invólucro. A proposta visa a racionalização no uso da energia elétrica através de uma elevada eficácia luminosa e longa vida útil na substituição dos sistemas de iluminação tradicionais, como lâmpadas dicróicas, incandescentes ou fluorescentes compactas, por LEDs sem qualquer alteração na instalação elétrica. Além disso, é proposto um sistema de iluminação de emergência integrado a esta lâmpada, permitindo a utilização de um único equipamento para iluminação convencional, alimentada pela rede elétrica, e sob uma possível falha no fornecimento de energia. Para alimentar os LEDs, através da rede elétrica, com tensão de entrada universal e ainda carregar a bateria, um conversor Buck é empregado. Para alimentar os LEDs em corrente contínua através da bateria, um conversor Boost é utilizado. Os conversores operam em alta freqüência e controlam a corrente nos LEDs de forma independente. A bateria pode ser composta por três pilhas recarregáveis de Ni-MH (1,2 V) ou por uma bateria de Li-Ion (3,6 V). O projeto atende às exigências da norma brasileira (NBR 10898) e da norma internacional (IEEE Std 446-1995) para sistemas de iluminação de emergência. Para analisar a idéia proposta, os circuitos são projetados e implementados. Os resultados experimentais são apresentados de forma a comprovar o comportamento satisfatório das lâmpadas.

During the fourteenth century, with the rise of Dol po pa Shes rab rgyal mtshan (1292-1361), the gzhan stong philosophical tradition became a source of great controversy in Tibet. Dol po pa taught this new philosophical tradition for the first time to the wider Tibetan intellectual community. As Dol po pa's Jo nang teachings attracted an audience, many other philosophical giants of the day, such as Bu ston Rin chen grub (1290-1364), Red mda' ba Gzhon nu blo gros (1349-1412/13), and their students composed polemical works to refute Jo nang tradition. Lamp that Illuminates the Expanse of Reality was composed in the midst of this controversy to defend the Jo nang point of view. In it, its author, Gha rung pa Lha'i rgyal mtshan (1319-1402/03), attempts to prove that the Jo nang philosophical tradition is the definitive teaching and the quickest path to achieve the Buddhahood.

Résumé :

The aim of this thesis project was to migrate an existing excel-based application, used

to analyze energy use and carbon dioxide emission of companies, to a web-based application.

Special development questions were put around which software development process,

solution stack and user interface to be used according to the company needs. The spiral

lifecycle model has been chosen because it provides a clear view of the process and has the

concept of early prototypes. A solution stack based on Linux, Apache, PHP and MySQL has

been chosen because such approach has met the company requirements in terms of cost,

security, support, and maintenance. As a result, the developed web-based system overcomes

the problems normally found in excel-based applications, such as application deployment and

maintenance, and provides a more usable and richer user interface.

Esta pesquisa se propÃe a compreender as relaÃÃes participativas e de convivÃncia entre os moradores da comunidade SÃo Vicente de Paulo (Quadra) e a rÃdio poste Centro de ComunicaÃÃo Alternativa, localizada no bairro Aldeota, em Fortaleza (CearÃ). A partir da pesquisa participante (BrandÃo, 1984; 2006) como metodologia, foi possÃvel investigar a histÃria de criaÃÃo da rÃdio poste, refletir sobre as diversas formas de participaÃÃo (Bordenave, 1983) de dois grupos de moradores da Quadra, jovens catÃlicos e mulheres evangÃlicas, e compreender a relaÃÃo deles com a rÃdio dentro da convivÃncia com as paisagens sonoras (Schafer, 2011) da comunidade. Entre as consideraÃÃes finais, foi possÃvel entender que a participaÃÃo de jovens e mulheres religiosos na rÃdio deu-se atravÃs da mobilizaÃÃo dos prÃprios grupos religiosos. Por fim, o campo ainda proporcionou momentos interventivos, durante o aniversÃrio do programa âRecordar Ã Viverâ e participaÃÃo no projeto de reforma da rÃdio. Este percurso de pesquisa tambÃm me fez perceber que a intervenÃÃo esteve presente desde minhas primeiras aproximaÃÃes com a Quadra. PorÃm, antes, pesquisa e intervenÃÃo pareciam se distanciar. Agora, concluo que as metodologias participativas unem esses processos.
This research aims to understand the participatory coexistence and relations between the residents of SÃo Vicente de Paulo community (Quadra) and the radio post Centro de ComunicaÃÃo Alternativa, located in Aldeota district, in Fortaleza (CearÃ). From the participatory research (BrandÃo, 1984; 2006) as a methodology, it was possible to investigate the history of creation of the post radio, consider about the various forms of participation (Bordenave, 1983) of two groups of residents of Quadra, young Catholics and evangelical women, and understand their relationship within the radio in the living with soundscapes (Schafer, 2011) community. Among the final comments, it was possible to understand that the participation of young women and religious radio was given by mobilizing of their own religious groups. Finally, the course of this research also provided interventional times during the anniversary of the program "Remembering is living" and participation in the radio reform. This search path also made me realize that the intervention has been present since my first contact with Quadraâs community. However at first, research and intervention seemed to be distance. I concluded that the participatory methodologies unite these processes.

Résumé :

The pre-symptomatic stage of neurodegenerative diseases such as Alzheimer’s disease (AD) and Parkinson’s disease (PD) occurs several decades before the clinical onset. Changes in the lysosomal network, i.e. the autophagosomal, endosomal and lysosomal vesicular system, are among the first alterations observed. There are currently no treatments to slow or cure neurodegenerative diseases, and there is a great need for discovery of treatment targets in cellular pathways where pathology pre-dates the neuronal death. It is also crucial to be able to diagnose neurodegenerative diseases earlier, both to enable early intervention treatment and aid in selecting clinical trial populations before the patient has widespread pathology. This thesis aims at investigating the potential of lysosomal network proteins as biomarkers and therapeutic targets in neurodegenerative disease. A targeted search for lysosomal network proteins was performed in cerebrospinal fluid (CSF) from AD patients, and seven proteins: early endosomal antigen 1 (EEA1), lysosomal-associated membrane proteins 1 and 2 (LAMP-1, LAMP-2), lysozyme, microtubule-associated protein 1 light chain 3 (LC3), Rab3 and Rab7, were elevated. The levels of EEA1, LAMP-1, LAMP-2, LC3, lysozyme and Rab3 were also measured in CSF from parkinsonian syndrome patients: PD, clinically diagnosed 4-repeat tauopathy, pathologically confirmed corticobasal degeneration (CBD) and pathologically confirmed progressive supranuclear palsy (PSP) patients. LAMP-1 and LAMP-2 were decreased in PD. LC3 and lysozyme levels were increased in 4-repeat tauopathy patients. EEA1 was decreased and lysozyme increased in PSP, and LAMP-1, LAMP-2, LC3 and lysozyme were increased in CBD. The lysosomal network proteins had different CSF protein profiles in all the parkinsonian syndromes, as well as in AD. It should be emphasized that only a select few of the lysosomal network proteins were observed to be changed, rather than a general change in lysosomal network proteins, which implicates the involvement of these seven proteins in specific pathological processes. The most interesting candidates, LAMP-2 and lysozyme, were selected for further study for their involvement in the pathology of AD. Lysozyme was found to co-localise with Aβ plaques in AD patients and overexpression prolonged survival and improved the activity in a Drosophila model of AD. Lysozyme was found to alter the aggregation pathway of Aβ1-42, to counteract the formation of toxic Aβ species and to protect from Aβ1-42 induced cell toxicity. Aβ1-42 in turn was found to increase the expression of lysozyme in both neuronal and glial cells. These data suggest that lysozyme levels rise in AD as a compensatory response which is protective against Aβ associated toxicity. LAMP-2 mRNA and protein were found increased in brain areas relevant for AD pathology and various cellular models showed complex involvement of LAMP-2 in Aβ related pathology, with extensive crosstalk between LAMP-2 and Aβ. Exposure to oligomeric Aβ1-42 caused an upregulation of LAMP-2 and in turn, overexpression of LAMP-2 caused a reduction in secreted levels of Aβ1-42, as well as changing the generation pattern of Aβ and affecting clearance and secretion of Aβ1-42. These data indicate that the increased levels of LAMP-2 in AD could be an attempt to regulate Aβ generation and secretion. In summary, this thesis reports that utilising lysosomal network proteins as biomarkers and novel therapeutic targets for neurodegenerative diseases holds great promise.

AbstractWhat role can board of directors play in the strategy process of an SME? The case of “Light-Tech” (a luxury lamp producer) shows how. The chairman and several of the other directors had bad feelings about the issue of technology replacement. Contrary to the top management team, they were not convinced that the breakthrough of the new LED technology in the market was still far away. To address this issue, the board of directors—together with the executive team—performed a scenario analysis for about 3 hours. Then, the board requested the top management team to formulate a precautionary strategy which much better prepares the SME for the pessimistic scenario. Result: 2 years later, the pessimistic scenario reveals itself as reality. By performing a scenario analysis together with the top management team, the board of directors were able to play an important role as constructive sparring partners.

AbstractUrban Internet of Things (IoT) is in an early speculative phase. Often linked to the smart city movement, it provides a way of sensing and collecting data—environmental, societal, and transitional—both automatically, remotely, and with increasing levels of spatial and temporal detail. From city-wide data collection down to the scale of individual buildings and rooms, this chapter details the technology behind the rise of IoT in urban areas and explores the challenges (societal and technical) behind city-wide deployments. Drawing from a series of deployments at the Queen Elizabeth Olympic Park, London, it details the challenges and opportunities for mass data collection. Widening out the view, it looks at what is becoming known as “the humble lamp post” in Urban IoT fields to detail the potential of Urban IoT with the objects that already form part of the urban fabric. Finally, it examines the potential of Urban IoT for input into urban modeling and how we are on the edge of a shift in the collection, analysis, and communication of urban data.

AbstractWith the globalization of seed trade and transgenic variety development, the application of molecular technologies for seed quality gained more significance in both the internal and international markets. Besides germination, genetic purity and seed health are the two most important seed quality parameters that determine the planting value of a seed lot. Compared to the conventional methods of testing, molecular marker technologies are more efficient for quality analysis as these are more accurate, robust, abundant, and faster. Among the various markers, simple sequence repeats (SSRs), due to their genome-wide presence, reproducibility, multi-allelic nature, and co-dominant inheritance, have emerged as the best markers, for establishing varietal distinctness, identity, and variety/hybrid seed purity testing. With the advent of the next-generation sequencing (NGS) technology, single nucleotide polymorphic (SNP) markers also became widely popular, and the closest to being an ideal marker besides SSRs, in seed genetic purity testing. With large-scale GM crop cultivation, testing for the adventitious presence and trait purity are two added components of seed quality testing. The methods of GM seed quality testing include DNA-based (conventional and real-time PCR), protein-based (lateral flow test and ELISA), and bioassay-based technologies. DNA-based methods including PCR/real-time PCR assays have been successfully employed to detect the adventitious presence of transgenic seeds in seed trade especially at international level, as well as in the national gene banks for germplasm conservation. ISTA plays a prominent role in international harmonization and providing universal guidelines on use of different methods to detect GM seeds. The BMT group of UPOV and the Working Group on DNA Methods of the Variety Committee of ISTA, work in tandem to standardize suitable molecular techniques for establishing variety identity and purity testing, respectively. In the area of seed health testing also, molecular detection assays such as, PCR (nested PCR, multiplex PCR, real-time PCR), loop-mediated isothermal amplification (LAMP), and DNA microarray with many advantages over the conventional assays have been proven highly useful. However, there is a need to validate the usefulness of molecular markers through stringent multi-laboratory tests for their reproducibility before recommending them in routine seed purity and health testing.

Tuberculosis (TB) is an airborne infectious disease caused by the bacterium Mycobacterium Tuberculosis (MTB) and is a persistent problem in developing countries. Present methods for its detection include normal or nested Polymerase Chain Reaction (PCR) followed by electrophoresis, real-time PCR, Ziehl-Neelsen staining, and culture assay. These techniques entail various disadvantages such as high cost, long assay time and use of toxic substances. Novel loop-mediated isothermal amplification (LAMP) permits DNA to be amplified rapidly under constant temperature. The combination of LAMP and chromatographic Lateral Flow Dipstick (LAMP-LFD) by using biotinylated LAMP amplicon hybridized with Fluorescein Isothiocyanate (FITC)-labeled probes are allowed to detect MTB without electrophoresis and interpreted within 3-5 min. LAMP-LFD is as highly sensitive as PCR-electrophoresis method. Based on its sensitivity, specificity, rapidity, cost effectiveness, ease of use, and convenience, LAMP-LFD could be suitable for use in early MTB detection.

Slit lamp examination is an important component of a pediatric eye exam. The slit lamp instrument, also known as a biomicroscope, is used to provide a stereoscopic magnified view of the ocular structures. The slit lamp facilitates the examination of both the anterior segment as well as the posterior segment of the eyes. For posterior segment examination, handheld lenses are used in conjunction with the slit lamp. Different tools and examination techniques are often needed to examine infants, toddlers, and other pediatric patients who cannot be positioned in the slit lamp. This chapter discusses the techniques and equipment used to facilitate the ocular health examination, including anterior segment structures, posterior segment structures, and intraocular pressure measurements in the pediatric population.

Fluorescent lamp wastes are one of the discarded electrical devices. These lamp wastes pose serious health concerns, and require extreme care in its disposal to avoid any adverse impacts. Also, with the widespread use of energy efficient LED lamps for lighting purposes, environmental concerns related to disposal of used mercury containing fluorescent lamps have become progressively important. This paper explores the possibility of replacement of fine aggregates in mortar with different proportions of fluorescent lamp wastes in order to find the optimum percentage of replacement to get better properties. Fluorescent lamp wastes were collected and crushed to the grading of fine aggregates. Compressive strength and bulk density of mortar cubes with 0%, 25%, 50%, 75% and 100% replacement of fine aggregates with fluorescent lamp wastes were evaluated after 7 days of initial curing. The compressive strength and bulk density of mortar was found to be decreasing with increasing percentage of lamp waste added. The replacement of fine aggregate by 25% lamp waste has greater strength compared to other proportions. Bulk density was found to be lesser at higher replacement levels and can be tried in producing light weight mortar. The utilization of lamp waste in mortar/concrete is a noble attempt in reducing the negative impact of lamp wastes on the environment

The National Historic Preservation Act of 1966 (NHPA) established the National Register of Historic Places (NRHP), which requires federal agencies to address their cultural resources, defined as any prehistoric or historic district, site, building, structure, or object. NHPA Section 110 requires federal agencies to inventory and evaluate their cultural resources. Section 106 requires them to determine the effect of federal undertakings on properties deemed eligible or potentially eligible for the NRHP. Camp Perry Joint Training Center (Camp Perry) is located near Port Clinton, Ohio, and serves as an Ohio Army National Guard (OHARNG) training site. It served as an induction center during federal draft periods and as a prisoner of war camp during World War II. Previous work established boundaries for a historic district and recommended the district eligible for the NRHP. This project inventoried and analyzed the character-defining features of the seven contributing buildings and one grouping of objects (brick lamp posts) at Camp Perry. The analysis is to aid future Section 106 processes and/or the development of a programmatic agreement in consultation with the Ohio State Historic Preservation Office (SHPO).
