Abstract ARTZ @ Jefferson positions people with dementia and care partners as authorities about their lived experiences; arts-based experiences assist them in communicating with and mentoring health-professions students about those lived experiences. Since Spring 2016, over 100 students have been mentored by people with dementia and their care partners. Their first encounters take place in museum galleries, through facilitated conversations about works of art. Over the next six to eight weeks, students and mentors build relationships through group meetings and individual conversations. Post course surveys demonstrated that nearly 100% of students indicated their increased ability to value listening and listen to others, enhanced the healthcare provider/patient relationship, and prioritized patients’ life experiences. The majority of mentors noted that student interactions added to their quality of life, social engagement and sense of purpose. Preliminary outcomes suggest that arts-based experiences establish mutual respect and empathy between people with dementia and students.

This is the first of two special issues based on materials presented at the FICE International Congress in Vienna, Austria in August 2016. The theme of the congress was “Together Towards a Better World for Children, Adolescents, and Families”. The same theme was chosen for these special issues. Child and youth care professionals who presented their quality material in the congress were invited to rework their presentations as formal papers meeting the norms of a scientific journal. FICE International is most thankful to the guest editors, Jim Anglin, Silke Gahleitner, and Emmanuel Grupper, who worked with all the contributors to prepare their materials for publication. ADP-ribosyltransferase activity was shown to be present on the surface of human monocytes. Incubating the cells in the presence of BSA leads to an increase in enzyme activity. The acceptor amino acid mainly responsible for the ADP-ribose bond was identified as a cysteine residue. An increase in ADP-ribosyltransferase activity was observed when cells were treated for 16h with bacterial lipopolysaccharide (LPS). Possible candidates for catalysing the reaction are mono-ADP-ribosyltransferases (ARTs). When measuring expression of the mRNA of ART1, 3, 4 and 5, only ART3 mRNA was detected in unstimulated monocytes. Upon stimulation for 16h with LPS, lipoteichoic acid or peptidoglycan, ART4 mRNA was found to be expressed. No ART4 signal appeared after a 4h exposure of the cells to LPS. Cell-surface proteins were labelled when incubating monocytes with [32P]NAD+. Their molecular masses were 29, 33, 43, 45, 60 and 82kDa. In response to LPS an additional protein of 31kDa was found to be labelled. The bound label was resistant to treatment with NH2OH but sensitive to HgCl2, characteristic of a cysteine-linked ADP-ribosylation.

We are grateful to Dr. Sibylle Artz, Editor of the International Journal of Child, Youth and Family Studies, for the invitation to produce a special issue on pain-based behaviour in child and youth care work. Since the term was created and published in 2002 (Anglin, 2002), the notion has entered the literature, the research, and, perhaps most importantly, the practice of child and youth care internationally. The eight articles in this issue come from Ireland, Australia, the United States, and Canada, and offer a broad range of perspectives.After receiving the invitation to compile this issue, we scanned the recent child and youth care literature and readily identified 13 publications — articles and books — using the term pain-based behaviour. There are undoubtedly many more, however we believed the authors of these publications would present a significant cross-section of perspectives on understanding and responding to pain and pain-based behaviour. We are excited and honoured that the authors represented here were able to contribute articles to this issue.

I am thankful for the opportunity to produce this special issue on refugees in Jordan, and grateful to Professor Sybille Artz, Editor of the International Journal of Child, Youth and Family Studies, who motivated and encouraged me. The issue includes four articles that focus in general on the problems and challenges that are encountered by Syrian youth and children who reside in Jordan either inside or outside the refugee camps there. Focusing on children and youth, both male and female, is important to the literature on Syrian refugees in Jordan because this age group has been neglected and overlooked by scholars since Syrian refugees first fled to Jordan in 2011. Furthermore, the challenges and problems of children are both culturally critical and morally sensitive, and that is why this special issue has extra importance. In Jordan’s conservative social and cultural environment, certain research topics relevant to the refugee camps are regarded as unlawful and prohibited: sexual harassment, underage marriage, child employment and exploitation, youth unemployment and deprivation, and other violations against children and youth.

The journey from reporting rape to convicting rapists is complex, leading to high attrition and non-conviction rates. After wide consultation, the law in England and Wales was revised in 2003 to try to secure more convictions. In South Africa, a similar process occurred to produce a new law in 2007. Nonetheless, reported rapes have risen and conviction rates have fallen in both jurisdictions and it has been suggested that the failure of the criminal justice system to deal with ‘rape…encapsulates the sheer inadequacy of the law’ (Wykes and Welsh, 2009: 111) and offers little hope of justice to victims and little deterrence to perpetrators. In South Africa little has changed, except more is known about ‘the lived experiences of sexual violence’ (Artz and Smythe, 2007: 17) and more support is offered to victims after the event. This article explores the part played by law in dealing with rape, through a comparison of the UK and South Africa. Critical gendered analysis of their respective rape laws leads to the conclusion that that law cannot work effectively to deter or convict rapists: only men’s willingness to change can stop rape.

ABSTRACT In the thermophile Geobacillus stearothermophilus, the uptake of basic amino acids is mediated by an ABC transporter composed of the substrate binding protein (receptor) ArtJ and a homodimer each of the pore-forming subunit, ArtM, and the nucleotide-binding subunit, ArtP. We recently identified two putative binding sites in ArtJ that might interact with the Art(MP)2 complex, thereby initiating the transport cycle (A. Vahedi-Faridi et al., J. Mol. Biol. 375:448-459, 2008). Here we investigated the contribution of charged amino acid residues in the second extracellular loop of ArtM to contact with ArtJ. Our results demonstrate a crucial role for residues K177, R185, and E188, since mutations to oppositely charged amino acids or glutamine led to a complete loss of ArtJ-stimulated ATPase activity of the complex variants in proteoliposomes. The defects could not be suppressed by ArtJ variants carrying mutations in site I (K39E and K152E) or II (E163K and D170K), suggesting a more complex interplay than that by a single salt bridge. These findings were supported by cross-linking assays demonstrating physical proximity between ArtJ(N166C) and ArtM(E182C). The importance of positively charged residues for receptor-transporter interaction was underscored by mutational analysis of the closely related transporter HisJ/LAO-HisQMP2 of Salmonella enterica serovar Typhimurium. While transporter variants with mutated positively charged residues in HisQ displayed residual ATPase activities, corresponding mutants of HisM could no longer be stimulated by HisJ/LAO. Interestingly, the ATPase activity of the HisQM(K187E)P2 variant was inhibited by l- and d-histidine in detergent, suggesting a role of the residue in preventing free histidine from gaining access to the substrate binding site within HisQM.

Mono-ADP-ribosylation is a post-translational modification that regulates the functions of target proteins or peptides by attaching an ADP-ribose moiety. Here we report the purification, molecular cloning, characterization and tissue-specific distribution of novel arginine-specific Arts (ADP-ribosyltransferases) from chicken. Arts were detected in various chicken tissues as GPI (glycosylphosphatidylinositol)-anchored forms, and purified from the lung membrane fraction. By molecular cloning based on the partial amino acid sequence using 5′- and 3′-RACE (rapid amplification of cDNA ends), two full-length cDNAs of chicken GPI-anchored Arts, cgArt1 (chicken GPI-anchored Art1) and cgArt2, were obtained. The cDNA of cgArt1 encoded a novel polypeptide of 298 amino acids which shows a high degree of identity with cgArt2 (82.9%), Art6.1 (50.2%) and rabbit Art1 (42.1%). In contrast, the nucleotide sequence of cgArt2 was identical with that of Art7 cloned previously from chicken erythroblasts. cgArt1 and cgArt2 proteins expressed in DT40 cells were shown to be GPI-anchored Arts with a molecular mass of 45 kDa, and these Arts showed different enzymatic properties from the soluble chicken Art, Art6.1. RNase protection assays and real-time quantitative PCR revealed distinct expression patterns of the two Arts; cgArt1 was expressed predominantly in the lung, spleen and bone marrow, followed by the heart, kidney and muscle, while cgArt2 was expressed only in the heart and skeletal muscle. Thus GPI-anchored Arts encoded by the genes cgArt1 and cgArt2 are expressed extensively in chicken tissues. It may be worthwhile determining the functional roles of ADP-ribosylation in each tissue.

FICE International is an international organization focused on children and youth who are in need of out-of-home care. One of its major activities is organizing a world congress every three years. In these congresses, professionals from all over the world who are involved with out-of-home care can meet, exchange knowledge, and learn from each other. The most recent FICE congress, held in October 2019 in Tel Aviv, Israel, was dedicated to the theme “Better Future Opportunities for Children and Young People in Multicultural Societies”. The editor of this journal has been kind enough to offer this platform for the publication of selected material presented at the congress. FICE-International, and we as guest editors, are most thankful to Prof. Sibylle Artz, editor of the IJCYFS, for this opportunity. The richness and variety of the selected articles, coming from 10 countries, could not be accommodated in a single issue. The editors of the journal have therefore decided to divide the articles into two special issues, to be published one after the other. The first of these issues includes articles in three thematic categories: (a) Evidence-based research on effects of out-of-home care, (b) Historical and sociological perspectives on institutionalized child and youth care, and (c) Normalization — a challenge for foster care programs. Four further categories will provide the framework for the forthcoming second special issue: (d) Emotional and pedagogical considerations, (e) Refugee children and youth, (f) Residential staff, and (g) Care leavers.

Salmonella enterica serotype Typhimurium (S. Typhimurium) definitive phage type (DT) 104 has become a widespread cause of human and other animal infections worldwide. The severity of clinical illness in S. Typhimurium DT104 outbreaks suggests that this strain possesses enhanced virulence. ArtA and ArtB – encoded by a prophage in S. Typhimurium DT104 – are homologues of components of pertussis toxin (PTX), including its ADP-ribosyltransferase subunit. Here, we show that exposing DT104 to mitomycin C, a DNA-damaging agent, induced production of prophage-encoded ArtA/ArtB. Pertussis-sensitive G proteins were labelled in the presence of [32P]NAD and ArtA, and the label was released by HgCl2, which is known to cleave cysteine-ADP-ribose bonds. ADP-dependent modification of G proteins was markedly reduced in in vitro-synthesized ArtA6Arg-Ala and ArtA115Glu-Ala, in which alanine was substituted for the conserved arginine at position 6 (necessary for NAD binding) and the predicted catalytic glutamate at position 115, respectively. A cellular ADP-ribosylation assay and two-dimensional electrophoresis showed that ArtA- and PTX-induced ADP-ribosylation in Chinese hamster ovary (CHO) cells occur with the same type of G proteins. Furthermore, exposing CHO cells to the ArtA/ArtB-containing culture supernatant of DT104 resulted in a clustered growth pattern, as is observed in PTX-exposed CHO cells. Hydrogen peroxide, an oxidative stressor, also induced ArtA/ArtB production, suggesting that these agents induce in vivo synthesis of ArtA/ArtB. These results, taken together, suggest that ArtA/ArtB is an active toxin similar to PTX.

Artemisinin (ART) is a sesquiterpene lactone and a popular malaria drug used in many parts of the world. Artesunate (ARTS) is a semi-synthetic derivative of ART with improved pharmacokinetic properties. However, the half-life of ARTS is less than an hour in vivo. The analysis of this drug in vitro in different solvent systems using LC-MS/TOF showed a solvent-driven breakdown. ARTS breakdown formed several derivatives, including dihydroartemisinin (DHA), artemether (ARTM) and DHA-dimer among others, at different rates in different solvent composition systems. The change in temperature from room temperature to physiological temperature (37 °C) was found to enhance the rate of the ARTS breakdown. In methanol, ARTS mainly formed ARTM with a chromatographic peak decrease of about 3.13%, while methanol and water (90:10) v/v mainly gave rise to DHA and ARTM with about an 80% chromatographic peak decrease. On the other hand, ARTS in methanol and ammonium acetate (85:15) v/v formed DHA, ARTM, DHA-dimer and other reaction peaks with about a 97% peak decrease and the formation of an orange solution pointing to a molecular re-arrangement reaction. These results have an important bearing on research on the analysis of artemisinin drugs conducted on these common solvents.

Considerando que la Educación por el Arte posee una doctrina flexible que reúne los fines, principios y postulados que pueden contribuir a la validez y confiabilidad de la operativización de las variables, este artículo presenta siete postulados de la Educación por el Arte, definidos como elementos teórico-prácticos sistematizados con la finalidad de puntualizar algunas estructuras teóricas que sustentan esta orientación educativa. AbstractThe application of the qualitative research approach in Education through the Arts is a priority alternative, due to the demand for the most exact interpretation of the propositions and the unequivocal meaning of the variables and indicators of study, which are characteristic of this type of research. Considering that Education through the Arts has a flexible doctrine that gathers the purposes, principles and postulates that can contribute to the validity and reliability to the implementation of the variables, this paper presents seven postulates of Education through the Arts defined as systematized theoretical-practical elements, with the purpose of pointing out some theoretical structures that are the base of Education through the Arts. To meet the coming digital transformation in swine production, the need for innovative human capital strategies has never been greater. Boessen, Artz, and Schutlz 2 found labor is a critical issue for the industry. High performing swine farms achieve it because of their people. Swan 3 noted that “pigs do not achieve excellence; people achieve excellence through their pigs.” Agriculture has been slow to adapt soft skill strategies due to the ambiguity in the value proposition. Cost metrics of turnover, poor performance, and safety are traditionally buried within the P&L under labor with labor impacting 100% of the value chain in pig and pork production. The need to analyze the human impact has never been more crucial, and this led to the 2019 Labor intel Study by sponsors and Summit SmartFarms. Schmidt and Hunter‍ 4 found that the use of general mental ability testing improves the predictability and utility of hiring the right person. Mono(ADP-ribosyl)transferases regulate the function of target proteins by attaching ADP-ribose to specific amino acid residues in the proteins. We have characterized the gene for mouse arginine-specific ADP-ribosyltransferase, Art1. Southern blot analyses indicate that Art1 is a single-copy gene. Northern blot and reverse transcription–PCR analyses demonstrate prominent expression of Art1 in cardiac and skeletal muscle, and lower levels in spleen, lung, liver and fetal tissues. While human ART1 is not represented in the public expressed sequence tag (EST) database, the database contains 14 mouse Art1 ESTs. The Art1 gene encompasses four exons spanning 20 kb of genomic DNA. The deduced amino acid sequence of Art1 exhibits the characteristic features of a glycosylphosphatidylinositol-anchored membrane protein. It shows 75–77% sequence identity with its orthologues from the human and rabbit, and 33–34% identity with its paralogues from the mouse, Art2-1 and Art2-2. Separate exons encode the N- and C-terminal signal peptides, and a single long exon encodes the entire predicted native polypeptide chain. We expressed Art1 in 293T cells as a recombinant fusion protein with the Fc portion of human IgG1. This soluble protein exhibits enzyme activities characteristic of arginine-specific ADP-ribosyltransferases. The availability of the Art1 gene provides the basis for applying transgene and knockout technologies to further probe the function of this gene product.

A ideia de arquitetura como arte e gênero artístico autônomo é relativamente nova, começando a tomar forma a partir do Renascimento. Anteriormente, na Idade Média, não existiam grêmios exclusivos de arquitetos — construtores —, mas sim de canteiros, marceneiros, pintores, todos agrupados sob a mesma irmandade. Mais tarde, com o nascimento da crítica de arte moderna, especialmente com a ideia de autonomia das diferentes expressões artísticas de Lessing, no século XVIII, e a estética de gêneros autônomos de Hegel, no século seguinte, formalizou-se a ideia da produção artística como gêneros autônomos. Contudo, movimentos artísticos de final do século XIX — como o Arts-& Crafts, o Pré-Rafaelismo e as manifestações artístico-decorativas como o Art Nouveau Franco-Belga, a Sessezion Austríaca, o Jungendstill Alemão e o Modernismo Catalão — representam reações a essa esquematização da arte. Este artigo objetiva compreender os processos de autonomização da arte em relação à realidade e de autonomização entre gêneros artísticos advindos com a modernidade para logo após estudar reações a esses fenômenos que tiveram lugar nas ultimas décadas do século XIX, notadamente o ideário Arts & Crafts e o Art Nouveau.PALAVRAS-CHAVE: Arquitetura. Osnovan kao prvi dokumentacijski centar i arhiv za nacionalnu povijest umjetnosti, Arhiv za likovne umjetnosti pri Hrvatskoj akademiji znanosti i umjetnosti kontinuirano razvija svoj informacijski sustav, pružajući podršku stručnjacima i znanstvenoj zajednici. U ovom ćemo radu prikazati povijest procesa automatizacije podataka, koji je započeo 1995. godine—od jednostavnih tablica u MS Excelu i baza podataka u programu MS Access, stvorenih za specifične potrebe Arhiva, do složene repozitorijske platforme otvorenog pristupa, razvijene kao izvor za rastuće područje digitalne povijesti umjetnosti. Digitalna tehnologija kontinuirano stvara nove i brže načine dohvaćanja arhivskih podataka—od jednostavnih pretraživanja pojmova kroz nekoliko izvanmrežnih baza podataka do naprednijih mogućnosti internetskog pretraživanja utemeljenog na semantičkoj mrežnoj tehnologiji. Arhiv za likovne umjetnosti prisutan je na internetu od 2009. preko Digitalne zbirke (DiZbi), institucionalne repozitorijske platforme koja je prvi put uspostavila vezu između digitaliziranih tekstualnih i vizualnih izvora različitih Akademijinih jedinica. Kako bi se digitalni objekti prikazali na što živopisniji i interaktivniji način, stvoren je nov digitalni sadržaj—virtualna izložba, kao jedna od vizualno atraktivnijih odlika portala. Kontinuirani proces digitalizacije arhivske građe otvorio je nove mogućnosti za suradnju s domaćim i međunarodnim ustanovama kroz nekoliko projekata. Budući planovi uključuju dugoročan projekt s potencijalom razvoja u digitalnu infrastrukturu za istraživanje provenijencije. Na temelju svojih iskustava s ovakvim digitalnim projektima možemo reći da stručnjaci za digitalnu povijesti umjetnosti imaju sve važniju ulogu u snažnijoj vidljivosti hrvatske povijesti umjetnosti, kako na nacionalnoj tako i na međunarodnoj razini, kao i u omogućavanju novih oblika digitalne analize u povijesti umjetnosti.

Il saggio affronta il rapporto tra opera e oggetto nella filosofia dell'arte di A. Danto. In particolare: 1) le opere d'arte sono sempre a "distanza" da ciň che rappresentano (contro le avanguardie artistiche, Danto difende la "differenza" tra arte e realtŕ); 2) sono oggetti semi-opachi, ovvero trasparenti e opachi allo stesso tempo; 3) come per Adorno, sono "paradossali", perché attraverso qualcosa di reale si manifesta qualcosa che non č "reale". A partire da queste considerazioni Danto distingue arti disturbanti e arti disturbazionali: le prime operano all'interno dei "confini" dell'arte senza metterli in discussione, le seconde, invece, cercano di superare la differenza tra arte e realtŕ.

El International Center for the Arts of the Americas (ICAA) del Museum of Fine Arts, Houston (MFAH) puso en línea en 2012 la primera fase del archivo ICAA Documents Project, un ambicioso repositorio digital de documentos primarios del arte latino y latinoamericano del siglo XX. Este artículo analiza la arquitectura documental del ICAA Documents Project tomando en cuenta las coyunturas que están experimentando en la actualidad las Humanidades Digitales en, desde y sobre América Latina. El artículo presta especial atención a la manera en la que este proyecto utiliza el Art & Architecture Thesaurus (Getty Foundation) para construir sus categorías meta-descriptivas y voces autorizadas. A partir del análisis de lo que llamaremos latinoamericanismo digital, en el artículo se abordan aspectos relacionados con la gestión documental del arte latinoamericano en entornos museográficos estadounidenses, así como el impacto de las Humanidades Digitales para la investigación global del arte latinoamericano.

A single gene cluster encoding components of a putative ATP-binding cassette (ABC) transporter for basic amino acids was identified in the incomplete genome sequence of the thermophilic Gram-positive bacterium Geobacillus stearothermophilus by blast searches. The cluster comprises three genes, and these were amplified from chromosomal DNA of G. stearothermophilus, ligated into plasmid vectors and expressed in Escherichia coli. The purified solute-binding protein (designated ArtJ) was demonstrated to bind l-arginine with high affinity (K d=0·39±0·06 μM). Competition experiments revealed only partial inhibition by excess l-lysine (38 %) and l-ornithine (46 %), while no inhibition was observed with l-histidine or other amino acids tested. The membrane-associated transport complex, composed of a permease (designated ArtM) and an ATPase component (designated ArtP), was solubilized from E. coli membranes by decanoylsucrose and purified by metal-affinity chromatography. The ArtMP complex, when incorporated into liposomes formed from a crude extract of G. stearothermophilus lipids, displayed ATPase activity in the presence of ArtJ only. Addition of l-arginine further stimulated the activity twofold. ATP hydrolysis was optimal at 60 °C and sensitive to the specific inhibitor vanadate. Analysis of kinetic parameters revealed a maximal velocity of ATP hydrolysis of 0·71 μmol Pi min−1 (mg protein)−1 and a K m (ATP) of 1·59 mM. Together, these results identify the ArtJMP complex as a high-affinity arginine ABC transporter.

Targeted endocytosis of plasma membrane (PM) proteins allows cells to adjust their complement of membrane proteins to changing extracellular conditions. For a wide variety of PM proteins, initiation of endocytosis is triggered by ubiquitination. In yeast, arrestin-related trafficking adaptors (ARTs) enable a single ubiquitin ligase, Rsp5, to specifically and selectively target a wide range of PM proteins for ubiquitination and endocytosis. However, the mechanisms that allow ARTs to specifically recognize their appropriate substrates are unknown. We present the molecular features in the methionine permease Mup1 that are required for Art1-Rsp5–mediated ubiquitination and endocytosis. A combination of genetics, fluorescence microscopy, and biochemistry reveals three critical features that comprise an ART sorting signal in the Mup1 N-terminal cytosolic tail: 1) an extended acidic patch, 2) in close proximity to the first Mup1 transmembrane domain, and 3) close to the ubiquitinated lysines. We show that a functionally similar ART sorting signal is also required for the endocytosis of a second Art1-dependent cargo, Can1, suggesting a common mechanism for recognition of Art1 substrates. We isolate two separate suppressor mutations in the Art1 C-terminal domain that allele-specifically restore endocytosis of two Mup1 acidic patch mutants, consistent with an interaction between the Art1 C-terminus and the Mup1 acidic patch. We propose that this interaction is required for recruitment of the Art1-Rsp5 ubiquitination complex.

Many bacterial pathogens encode ADP-ribosyltransferase toxins. The authors identified an ADP-ribosyltransferase toxin homologue (ArtA, ArtB) in Salmonella enterica serovar Typhimurium (S. typhimurium) DT104. ArtA is most homologous to a putative pertussis-like toxin subunit present in Salmonella typhi (STY1890) and Salmonella paratyphi A (SPA1609), while ArtB shows homology to a hypothetical periplasmic protein of S. typhi (STY1364) and S. paratyphi A (SPA1188), and a putative pertussis-like toxin subunit in S. typhi (STY1891) and S. paratyphi A (SPA1610). The artA gene was detected from the phage particle fraction upon mitomycin C induction, and the flanking region of artAB contains a prophage-like sequence, suggesting that these putative toxin genes reside within a prophage. Southern blotting analysis revealed that artA is conserved in 12 confirmed DT104 strains and in four related strains which are not phage-typed but are classified into the same group as DT104 by both amplified-fragment length polymorphism and pulsed-field gel electrophoresis. Except for one strain, NCTC 73, all 13 S. typhimurium strains which were classified into different groups from that of DT104 lacked the artA locus. The results suggest that phage-mediated recombination has resulted in the acquisition of art genes in S. typhimurium DT104 strains.

But the conception of the arts that emerged in the 18th century, and remained the orthodoxy in philosophy for about two centuries, either excludes architecture from the fine arts or relegates it to the intermediate or decorative arts. This essay addresses this puzzle, assesses the truth in certain formalist doctrines about architecture, and advances the view that works of art are organic unities, i.e. integrated sets of solutions to various problems, some aesthetic and others technical, mathematical, theological, political, etc.Key words: art, architecture, aesthetics, formalism.Resumen: Los edificios y monumentos se encuentran entre las obras de arte más importantes. Madero de la Ciudad de México, en 2013 y 2014. Posterior a ello, impulsamos talleres de verano de artes escénicas dirigidos a niñas y niños en el barrio de Tepito (2016, 2017, 2018, 2019). ABSTRACT This is the first study reporting the inactivation of a member of the mouse gene family of toxin-related ecto-ADP-ribosyltransferases (ARTs). Transfer of the ADP-ribose moiety from NAD onto extracellular arginine residues on T-cell membrane proteins is mediated by glycosylphosphatidylinositol-linked cell surface ARTs. Exposure of T cells to ecto-NAD blocks T-cell activation and induces T-cell apoptosis. To determine a possible role of ecto-ART2.1 and ART2.2 in these processes, we generated ART2.1/ART2.2 double-knockout mice. ART2-deficient mice were healthy and fertile and showed normal development of lymphoid organs. ART2-deficient T cells showed a dramatically reduced capacity to ADP-ribosylate cell surface proteins, indicating that most if not all ART activity on the T-cell surface can be attributed to the ART2s. Moreover, ART2-deficient T cells were completely resistant to NAD-induced apoptosis and partially resistant to NAD-mediated suppression of proliferation. These results demonstrate that the ART2 ectoenzymes are an essential component in the regulation of T-cell functions by extracellular NAD, e.g., following release of NAD upon lysis of cells in tissue injury and inflammation.

Background: This study is to evaluate the predisposing factors and cause of resistance which are associated with ART1 failure and shifted to ART 2 regime.Methods: This study was conducted in ART plus centre K.P.S. Post Graduate Institute of Medicine (G.S.V.M. Medical College, Kanpur, India) tertiary care teaching hospital (G.S.V.M. Medical College, Kanpur, India). It will be a clinical (assessment with investigation), continuous, longitudinal, prospective and retrospective, observational, single centre hospital-based study at ART Centre, Kanpur, India. All the subjects who were on 1st line ART regime, attended in our centre were screened for treatment failure of based on clinical, immunological and virological criteria as decided by SACEP. Duration of this study was DEC 2016 TO DEC 2018.This study was taking as regime ART2 as TLATV/R, ZLATV/R, TLLP/R and ZLLP/R.Results: In this study there is PL HIV subjects that are considered for ART2 are mostly living in rural area and more are female having less adherence to ART1. Smoking, alcohol and tobacco chewing were also having less adherence to ART and cause resistance to ART1.Conclusions: In this study subjects were having associated with predisposing factor as Alcohalic 53 (45%), Tobacco Chewing 8 (07%), SMOKING 13 (11%). Alcohalic, Tobacco Chewer and Smoker have significant association of predisposing factor for low adherence to ART1 and resistance to ART1 drugs. There is also concluded that females and rural areas subject are having low adherence and cause ART1 failure.

ABSTRACT. Much attention is paid to the problem of secondary osteoarthritis caused by various microorganisms and fungi of the genus S. albicans, associated with their significant growth and prevalence throughout the world among the population of various age groups, high temporary and permanent disability. At the same time, the problem of immunopathology in the pathogenesis of secondary osteoarthritis is being actively studied. The developed immunopathological process induced by pathogenic and opportunistic microflora in articular cartilage and synovial membrane is the main factor in the degenerative destruction of articular cartilage. Goal. to determine the role of pathogenic and opportunistic microflora localized in the oropharynx in the etiopathogenesis of secondary osteoarthritis. Research materials and methods. 65 patients aged from 19 to 45 years, who were undergoing inpatient treatment at Artz Klinic, Kharkiv, were examined for secondary osteoarthritis of the knee joint. The diagnosis was established according to the International Classification of Diseases of the 10th revision (ICD-10) - M18.1. Patients with a history of a high frequency of accompanying pathologies of the oropharynx were selected for the study: chronic tonsillitis - 48 (73.8%), tonsillitis - 32 (49.2%), pharyngitis - 34 (52.3%), sinusitis - 13 (20) %), otitis – 9 (13.8%) the etiology of which is determined by gram-positive and gram-negative microorganisms. All patients underwent microbiological examination of throat swabs, and 24 patients underwent microbiological examination of synovial fluid, which was taken for examination before intra-articular therapy. The obtained data were statistically processed by calculating the arithmetic mean and its error. The results. Presented modern ideas about secondary osteoarthritis caused by etiologically significant macroflora persisting in the oropharynx and as a result of organotropy due to the inclusion in its structure of mimicking antigens of articular cartilage play a leading role in the degradation of articular cartilage and articular bag, transferring the pathological process to an autoimmune basis. This approach makes it possible to develop early immunodiagnostics, prognosis of the disease, and immunocorrective therapy. Conclusions. Pathogenic and conditionally pathogenic microflora persisting in the oropharynx is characterized by organotropy (community of lipopolysaccharides and glycoproids), is identical to the cell-tissue structures of articular cartilage and synovial membrane in the causative agent and modulates the immune response through mimicking antigens.

To observe internalization of the yeast pheromone receptor Ste2 by fluorescence microscopy in live cells in real time, we visualized only those molecules present at the cell surface at the time of agonist engagement (rather than the total cellular pool) by tagging this receptor at its N-terminus with an exocellular fluorogen-activating protein (FAP). A FAP is a single-chain antibody engineered to bind tightly a nonfluorescent, cell-impermeable dye (fluorogen), thereby generating a fluorescent complex. The utility of FAP tagging to study trafficking of integral membrane proteins in yeast, which possesses a cell wall, had not been examined previously. A diverse set of signal peptides and propeptide sequences were explored to maximize expression. Maintenance of the optimal FAP-Ste2 chimera intact required deletion of two, paralogous, glycosylphosphatidylinositol (GPI)-anchored extracellular aspartyl proteases (Yps1 and Mkc7). FAP-Ste2 exhibited a much brighter and distinct plasma membrane signal than Ste2-GFP or Ste2-mCherry yet behaved quite similarly. Using FAP-Ste2, new information was obtained about the mechanism of its internalization, including novel insights about the roles of the cargo-selective endocytic adaptors Ldb19/Art1, Rod1/Art4, and Rog3/Art7.

Este artigo apresenta a discussão sobre o ensino de Arte e de Literatura nos anos iniciais do Ensino Fundamental, bem como a estratégia metodológica desenvolvida no Projeto Leiturarte. Destaca nos documentos oficiais o encaminhamento para o ensino de Arte e de leitura/ literatura, apontando as contribuições das estratégias metodológicas, que visam à interdisciplinaridade e justifica o desenvolvimento do Projeto Leiturarte como possibilidade metodológica qualitativa no ensino de Arte e Literatura no Ensino Fundamental. Para alcançar tais objetivos, a metodologia utilizada foi pesquisa bibliográfica e a pesquisa descritiva e de campo por meio da pesquisa-ação. Dessa forma, aponta uma contribuição para prática docente destacando que a interdisciplinaridade deve ser reconhecida como estratégia metodológica ao estabelecer conexões entre os conhecimentos a serem ensinados aos alunos. O Projeto Leiturarte demonstrou, qualitativamente, resultados para o processo ensino e aprendizagem nos anos iniciais no Ensino Fundamental, redimensionamento a compreensão do ensino de Arte e de Literatura.Palavras-chave: Ensino de Arte. AbstractThis article presents a discussion about the teaching of Art and Literature in the initial Primary School years, as well as the methodological strategy developed in the Leiturarte Project. To achieve the goals, the bibliographical, descriptive and field researches, throughthe action-research, were used as the methodology. Thus, it brings a contribution for the teaching practice emphasizing that the interdisciplinaritymust be recognized as a methodological strategy establishing connections withinthe knowledge to be taught to the students. The Leiturarte Project showed, qualitatively, results for the teaching and learning process in the initial Primary School years resizing the Arts and Literature’s teaching comprehension. Adaptive Resonance Theory (ART) model is a special neural network based on unsupervised learning which simulates the cognitive process of human. However, ART1 can be only used for binary input, and ART2 can be used for binary and analog vectors which have complex structures and complicated calculations. In order to improve the real-time performance of the network, a minimal structural ART is proposed which combines the merits of the two models by subsuming the bottom-up and top-down weight. The vector similarity test is used instead of vigilance test. Therefore, this algorithm has a simple structure like ART1 and good performance as ART2 which can be used for both binary and analog vector classification, and it has a high efficiency. Finally, a gas turbine fault diagnosis experiment exhibits the validity of the new network.
