The development of the technical capabilities of the media to expand the audience, combined with an increase in the share of action-packed literature, films and TV series, raised the question of the nature of the impact of this cultural product on public consciousness. What is primary and what is secondary – the fight against crime in fiction or the same process in real life? Despite the seemingly obvious answer, there is no consensus on this issue. The attempts made by individual dissertators to achieve certainty in the above dilemma give interesting results, but such studies are isolated. Therefore, a purposeful and thorough study of action-packed works and the process of their impact on public consciousness seems relevant.

This research is undermined by the fact that the level of mathematical understanding in class 3B SDN 096 Sarijadi Selatan, Bandung for the academic year 2022/2023 is still low and, in particular, has not properly understood the concept of the materials taught. The use of the problem-based learning (PBL) model packed into the game of thematic snake staircase (ULTAMET) becomes one of the options to enhance the mathematical understanding of the learners. Problem-based learning, packed into the game of snake staircase," is learning that focuses on fun problem-solving activities. The purpose of this action research in general is to improve the quality of the learning process in the classroom, and the aim of this action research is to know how problem-based learning based on the game Snake Staircase can improve the mathematical understanding of students in class 3B SDN 096 Sarijadi Selatan. This class action study uses Kemmis and McTaggart designs that have four stages in the implementation of class action research: planning, action, observation, and reflection. The instruments used include mathematical understanding tests and guidelines for the observation of student learning activities. Based on the results of the action of three cycles, it was shown that: 1) the problem-based learning model based on the game of snake stairs can improve the understanding of students of class 3B SDN 096 Sarijadi Selatan; 2) on the basis of the N-gain test results, the improvement in the mathematical understanding of learners by 83.13% was effectively interpreted, and the learning activity of students continued to improve in each cycle and reached 86% with excellent categories. Based on the findings of this study, it was concluded that the mathematical understanding of students in class 3B SDN 096 Sarijadi Selatan increased after learning using a problem-based learning model based on a game of snake stairs.

The difficulty of obtaining a densely packed granular material as an initial condition is a very common problem in numerical simulations of granular materials. In this article, an Imaginary Coating Algorithm (ICA) is introduced. To avoid unreasonable particle deformation when using a longer time step and a lower Young’s modulus, the radii used in calculating the action forces in a binary collision are slightly larger than the real values. In other words, an imaginary coat is added to each particle or element. To validate this algorithm, simulations were carried out by using ÅDEM, and a A Discrete Element Method (DEM) software program was developed. Compared with traditional Simulated Annealing Algorithms (SAA), this technique can approach the densely packed state with less CPU/GPU time and is easy to operate.

1. Intracellular recordings were made from intramural neurons in the urinary bladder of guinea pigs. 2. The neurons were located in two types of ganglia: those where the cells were densely packed and those where the neurons were loosely packed. Staining of the cells by intracellular injections of markers showed that the cells had between one to three long processes and several short dendrites. 3. The resting potential measured in 230 neurons was -55.20 +/- 0.67 (SE) mV, and the input resistance was 58.37 +/- 1.78 M omega. 4. Injection of depolarizing currents from the recording electrode evoked two types of firing patterns. In 86.2% of the neurons, depolarizing currents evoked a prolonged firing of action potentials (tonic cells). In the rest of the neurons, a depolarization elicited one to three action potentials only (phasic cells). In all the cells tested, the action potentials were reversibly blocked by tetrodotoxin (TTX; 1 microM). In the presence of TTX. Ca2+ spikes were observed in 50% of the cases. 5. Single action potentials were followed by fast hyperpolarizations having mean duration of 92.7 +/- 6.0 ms and amplitude of 13.3 +/- 1.0 mV. In 62.5% of the cells repetitive firing of action potentials was followed by delayed, slow hyperpolarizations (duration 3.8 +/- 0.5 s), which were diminished by the K+ channel blocker 4-aminopyridine and in Ca+2-free high-Mg2+ medium. These results indicate that the prolonged after-spike hyperpolarizations were due to opening of Ca(2+)-induced K+ channels. 6. Electrical stimulation of nerve fiber tracts evoked fast excitatory synaptic potentials that were blocked by the nicotinic receptor antagonist hexamethonium (0.2 mM). Exogenous acetylcholine elicited depolarizations that were also blocked by hexamethonium. Nerve stimulation at frequencies of 0.1 Hz or higher caused strong facilitation of the synaptic potentials. Stimulation at 10-20 Hz did not evoke slow synaptic potentials.

Like most kids of the McLuhan generation, I have vivid memories of images from television. Alongside the many comedies and adventure shows I watched, I remember commercials as miniature sagas conveniently repeated every half hour in case I missed them the first time. Sometimes, they involved an action-packed 30 seconds of Kool-Aid jugs crashing through brick walls or otherwise mild mannered men applying Hai Karate to become Kung Fu masters. Interspersed with these small vignettes, came the voice of Victor Kiam. In a fatherly way, he talked about using an electric shaver that his wife had given him and would close by saying “I liked it so much, I bought the company.”

This review focuses on the enzymatic breakdown of chitin, taking into account the latest scientific reports on the activity of lytic polysaccharide monooxygenase (LPMO). Chitin is a natural, abundant polysaccharide of great practical importance in the environment. However, the insolubility in water and the tightly packed crystalline structure of chitin pose a serious obstacle to enzymatic degradation. This substrate can be converted into soluble sugars by the action of glycosidic hydrolases (GH), also known as chitinases. LPMO could prove to be helpful in enzymatic processes that increase the rate of chitin depolymerisation by improving the availability of substrates for chitinases. The unique action of LPMO is based on the ability to catalyse the oxidative cleavage of glycosidic chains present in complex, resistant crystal networks of chitin, and this cleavage facilitates the subsequent action of glycolytic hydrolases.

Direct contact melting of a packed bed is studied experimentally. The effect of several parameters, including material properties, particle size, bed loading (applied external force), and thermal boundary conditions, is examined. Two types of melting are observed. If the load on the bed is small and the top of the bed is maintained at a temperature below the melting temperature, then crusting can occur. During this type of melting liquid flows away from the heated surface due to capillary action, and resolidifies forming a crust. The crust prevents downward movement of the packed bed, and melting of the solids below the crust results from convection and conduction. Eventually the crust is melted and the process repeats itself, resulting in a highly unsteady melting rate. If the bed load and the temperature at the top of the bed are sufficiently high, then direct contact melting persists. In this case the melting rate is steady and the parametric behavior can be predicted using a modification of an available theory for direct contact melting of monolithic solids. Although this theory accurately predicts the parametric behavior, the prediction of the actual melting rate is more than an order of magnitude higher than the experimental results.

A fluid is pumped through the tube to remove the contaminant. The convective velocity due to the fluid pumping is the nonlinear control action. The goal is to minimize a performance criterion which is a combination of the total contaminant at the final time and the cost of the control. Abstract. Oxytocin, vasopressin, cortisol and testosterone levels in the plasma were measured by radioimmunoassay in intact male goats as well as in prepubertally castrated goats injected daily, for 2 weeks, with oil vehicle and then, for 4 weeks, with testosterone propionate in oil to study the influence of gonadal steroids on posterior pituitary hormones. Packed cell volume, plasma osmolality and sodium concentration were also measured in all blood samples. Plasma levels of oxytocin, vasopressin and cortisol were similar in the intact and oil-injected castrated goats. Testosterone treatment significantly increased plasma levels of oxytocin (P < 0.01) in castrated goats but the increased levels were similar to those seen in the intact goats at the same time of year. Plasma levels of cortisol and vasopressin were unaffected by testosterone propionate treatment, whereas packed cell volume was significantly decreased (P < 0.01). Testosterone treatment of castrated male goats appears not to have any action on pituitary hormones and oxytocin increases in the spring in both intact and castrated male goats.

Focusing on the Australian KFC T20 Big Bash League (BBL), this article explores the innovative televisual technologies that represent T20 cricket as an action packed ‘smash and bash’ spectacle. An array of innovative technologies is deployed to aesthetically and affectively re-present the BBL. Cameras and microphones are embedded within the field of play, operate in highly mobile and fluid ways, and are framed in close proximity to the action – particularly when placed on the players themselves. The BBL provides intersecting affective layers for viewer engagement built upon tools for analysis, sites of commodification, visual renditions of pseudo-player perspectives and an emphasis on fast-paced entertainment. By constructing degrees of sensory invigoration and vicarious involvement for both casual and invested viewers, these innovative technologies mobilise ‘smash and bash’ cricket as an affective televisual spectacle.

In this study, the failure mode of flexible reinforced soil slopes under earthquake action was investigated by shaking table tests. The distribution law of a potential failure surface of a flexible no-faceplate reinforced soil slope under earthquake action was obtained based on the analysis results. A simplified trilinear failure surface suitable for flexible reinforced soil slopes without faceplate was proposed. Subsequently, based on the upper-bound theorem of limit analysis, we derived the formula for calculating the yield seismic acceleration coefficient of a flexible no-faceplate reinforced soil slope under a seismic load. The main parameters that affect its seismic performance were determined. The flexible geogrid reverse-packed reinforced earth structure can effectively limit the fracture of a slope body and improve the stability of the slope. This provides a theoretical basis for facilitating the engineering of flexible reinforced soil slopes.

Abstract The critical property of tire tread material for ice-skid resistance was analyzed. This skid resistance in the temperature range of 0 to −15°C is related to the compliance, not the hysteresis, of the tread material. Mechanistic analysis of the phenomena is presented in terms of the viscous resistance of a thin liquid-like layer on the surface of ice. The microscopic contact area and the wiping action of the tread material seem to be important factors for ice skid resistance. In a winter tire, the critical tread material property for traction on packed snow follows the same relationship.

Skin delivery of biomacromolecules holds great advantages in the systemic and local treatment of multiple diseases. However, the densely packed stratum corneum and the tight junctions between keratinocytes stand as formidable skin barriers against the penetration of most drug molecules. The large molecular weight, high hydrophilicity, and lability nature of biomacromolecules pose further challenges to their skin penetration. Recently, novel penetration enhancers, nano vesicles, and microneedles have emerged as efficient strategies to deliver biomacromolecules deep into the skin to exert their therapeutic action. This paper reviews the potential application and mechanisms of novel skin delivery strategies with emphasis on the pharmaceutical formulations.

While ostensibly an action-packed narrative about the hunt for indigenous Ainu gold, Golden Kamuy has earned a reputation among fans as a gurume , or gourmet, manga and anime. Cooking scenes of both Ainu and Wajin food feature prominently throughout the series. This paper analyzes the rhetorical role of food in the fictional narrative and its connection to official and unofficial ideologies about ethnic harmony between Ainu and Wajin communities in Japan.

The melting of a multi-component mixture that consists of melting and nonmelting components is examined. A numerical solution is obtained by solving the energy equation and continuity equations for both solid and liquid phases for a one-dimensional system. Both constant volume and constant porosity melting models are explored. Experiments were conducted to confirm the numerical results. During the experiments, heat was applied at the bottom of a rectangular packed bed containing salol (benzoic acid 2-hydroxyphenylester) and glass beads. The experiments were conducted with various sizes of glass beads ranging from 120 μm to 450 μm in diameter with salol having an average diameter of 90 μm. During the melting process, the volume fraction of the phase-change material (salol), temperature, and applied heat flux were measured. The volume fraction of the salol was measured utilizing gamma attenuation. Upon melting, the salol moves as two fronts, one downward due to gravity and one upward due to capillary action. The constant porosity model yields results which compare well with the experimental data.

Use of biofilms in food enable increase of shelf life and ensure the conservation thereof. Chitosan has been used on obtainment of biofilms due its excellent properties of antimicrobial, antifungal, permeable and resistant film formation. The aim of this study was to evaluate the application effect of chitosan biofilms on conservation of sausages vacuum-packed or bulk packaging, stored at 10 °C for 10 days. For biofilm obtaining, chitosan (85% of deacetylation) was dissolved in acetic acid (0.5 mol L-1), at concentrations of 1% (T1) and 2% (T2). Sausages produced in industrial line were coating with T1 and T2 to immersion, stored at vacuum-packed or bulk packaging, and compared to a control (T3, without coating). Microbiologic analyses to determination of total and thermotolerant coliforms, mesophilic aerobic count, coagulase-positive Staphylococcus, and molds and yeasts were carried out during the storage at 10 °C, at times of 0, 14, 21 and 28 days, in order to study the antimicrobial and antifungal behavior of biofilm on the sausage. The biofilm showed no inhibitory action for mesophilic aerobic in all samples during the storage period. In molds and yeasts analyses were no significant difference between the samples T1, T2 and T3, for the vacuum packed sausages, indicating that the biofilm chitosan showed antifungal properties in the studied conditions. Further, in yeast count was observed that T2 was more effective inhibition than T1. In all experimental conditions, the coagulase-positive Staphylococcus count, and determination of total and thermotolerant coliforms showed values lower than 2 CFU g-1 and 0.3 MPN g-1, respectively. Application of chitosan biofilms in sausages can result in antifungal effect when associated to vacuum packing.

Biogenic gas is an important end-product of methanogenesis in an anoxic environment. Based on the coal, coal-gangue, mine water of Songzao Coal Mine and in the case of adding inoculums, the quantities of gas and the contents of methane are measured by the microorganism action, and also respectively measured fermentation broth and residues by NO3--N, NH4+-N, S2- , SO42-, the values of pH, metals and the organic matter. The results show that coal and coal-gangue are involved in the microbial role; the quantity of inoculums is directly related to the quantities of gas and the contents of methane, at the same time it has a certain degree of impact on the contents of NO3--N, NH4+ -N, S2- , SO42- and metals; besides, because of the microbial anaerobic fermentation, the contents of NH3+ are seriously substandard; overall, the group B which has a smaller proportion of coal-gangue has a less impact to the environment compared to the group A which has a larger proportion of coal-gangue. In the further research into the direction of organic matter and obtaining and using the biogas, we must prevent secondary pollution of groundwater at the same time.

Cellular polymers because of their inherent light weight are particularly attractive for use as insulation materials but their fire characteristics have to be closely monitored so that they do not present an unacceptable hazard.

We solve a challenging yet practically useful variant of 3D Bin Packing Problem (3D-BPP). In our problem, the agent has limited information about the items to be packed into a single bin, and an item must be packed immediately after its arrival without buffering or readjusting. The item's placement also subjects to the constraints of order dependence and physical stability. We formulate this online 3D-BPP as a constrained Markov decision process (CMDP). To solve the problem, we propose an effective and easy-to-implement constrained deep reinforcement learning (DRL) method under the actor-critic framework. In particular, we introduce a prediction-and-projection scheme: The agent first predicts a feasibility mask for the placement actions as an auxiliary task and then uses the mask to modulate the action probabilities output by the actor during training. Such supervision and projection facilitate the agent to learn feasible policies very efficiently. Our method can be easily extended to handle lookahead items, multi-bin packing, and item re-orienting. We have conducted extensive evaluation showing that the learned policy significantly outperforms the state-of-the-art methods. A preliminary user study even suggests that our method might attain a human-level performance.

Essential plant oils added to products, packaging or animal feed are used as a method of preserving food quality because they extend the shelf-life of meat due their antioxidant and/or antimicrobial capacity. This action can be achieved with the correct packaging that preserves the meat’s quality and safety. This study investigates the effects of plant-derived extracts (PDE) on the meat quality and shelf-life of pork packaged in vacuum or modified atmosphere packaging (MAP). Thirty-six barrows and thirty-six gilts were allocated into three experimental groups: the control, garlic extract (1 kg/ton of feed) and oregano–rosemary oil (2 kg/ton of feed) with the same base-diet. Two packaging were used: vacuum and a commercial MAP (70% O2, 30% CO2). The meat fat content, pH, colour, TBARS values and Warner–Bratzler shear stress were investigated. The sex of the animals did not affect any of the studied variables, whereas PDE affected some of the colour variables and the shear stress; both the packaging type and the storage time affected the colour variables, lipid oxidation and shear stress. Vacuum-packed meat was more stable in terms of colour, lipid oxidation and shear stress than MAP-packed meat.

The study was planned to investigate the anti-urolithiatic activity of fresh juice of Citrus limon L. fruit. The phytochemical screening was also undertaken to reveal the alkaloids, flavonoids, reducing sugar, vitamin C, amino acid and sterols present in the sample. Macromolecule supersaturation of higher molecular weight components of plant extract exerts its action similar to natural urinary inhibitors and inhibits crystal aggregation and growth. Pharmacological evaluation of Citrus limon L. was performed by the percentage dissolution and turbidity method. Calcium oxalate crystals were synthetically prepared and packed in a semi-permeable membrane in both methods. Result was compared statistically and it showed significant P<0.001 anti-urolithiasis activity as compared to control and standard (Cystone).

ABSTRACT To study the antimalarial action of chloroquine, normal mouse erythrocytes were used as surrogates for erythrocytoid bodies. These bodies form in the endosomes of intraerythrocytic malaria parasites as they feed on their host and consist of erythrocyte cytoplasm enclosed in a vestige of the erythrocyte membrane. In suspensions of normal erythrocytes or lysates (equivalent to 5 μl of erythrocytes per ml in each case), hemoglobin underwent denaturation when it was incubated at 38°C in 150 mM sodium acetate (pH 5). It is reasonable to assume that the same phenomenon occurs in acidic endosomes. Addition of 100 μM chloroquine to the incubation mixture caused the rate of hemoglobin denaturation to double to 40 nanomoles per hour per ml of packed erythrocytes. This effect required the presence of erythrocyte stroma and was inhibited by reducing the temperature to 24°C or increasing the pH to 6. We propose that the primary antimalarial action of chloroquine is to bind to ferriprotoporphyrin IX (FP) and remove it from oxidized hemoglobin, thus producing toxic FP-chloroquine complexes and an excess of denatured globin. Furthermore, we suggest that these substances inhibit endosomal maturation and thereby cause hemoglobin accumulation in immature endosomes and masking of the lipids needed for FP dimerization. The term “masking” is used to signify that unsaturated lipids are present in parasitized erythrocytes but are specifically unavailable to promote FP dimerization.

The changes on microstructure, including cortical cell structure, density and degree of crystalline, of yak hair fiber have been analyzed after the stretching slenderization in this paper. It was found that the cortical cell has been elongated and became thinner, which average length-diameter ratio could be raised from 18 to about 40. By means of ultrasonic separation technology, the effect of stretching on packing action of fiber cortical cells has been investigated. It was proved that the stretching slenderization would improve the structure of yak hair cortex, and cortical cells were packed tighter along the fiber. The density and degree of crystalline also increased as raise of stretching rate. At the same time, the results form tensile test indicated that the fiber physical property would be improved through stretching treatment.

The geometric nonlinear dimensionless dynamical equations of suspension spring cushioning packaging system were established and a new concept of damage boundary curves of the system was developed under the action of rectangular pulse. Considering the angle of the suspension spring ,a three-dimension damage boundary curve is obtained.The mathematic results of the equation are calculated by using the four steps Runge-Kutta method. Studies have shown that the dimensionless pulse peak, the angle of suspension spring and the damping ratio of the system can affect the safe region, and that increasing damping can enlarge the safe region of the system. The shock of the packed product can reduce by adjusting the parameters of the system properly. These results have important application value in design of the suspension cushioning system.

In this film, Harrison Ford returns to his iconic role as Indiana Jones, showcasing a new perspective on action-packed adventures with seasoned heroes, which brings forth exciting new possibilities. Examining Indiana Jones and the Dial of Destiny, this study delves into the development of masculinity in older action heroes, highlighting how these seasoned protagonists can challenge traditional notions of male hegemony. The study employs discourse analysis to examine the portrayal of Harrison Ford's Indiana Jones and draws comparisons to previous iterations and other characters within the genre. This analysis highlights the film's departure from conventional portrayals of masculinity, indicating a movement towards dismantling gender norms and challenging male hegemony. The analysis highlights the importance of older heroes in portraying changing perspectives on masculinity and male roles in today's society. This study presents a fresh outlook on gender studies, highlighting the importance of portraying masculinity in film in a more nuanced way. It advocates for breaking away from traditional norms and promoting diversity in storytelling and character development.

Pseudoalteromonas carrageenovora κ-carrageenase is a glycoside hydrolase involved in the bioconversion of carrageenans. Carrageenans are sulfated galactans that are densely packed in red algal cell walls. Previous crystallographic investigations revealed that the active site of κ-carrageenase has a tunnel-shaped topology, suggesting a processive mode of action for this enzyme. To biochemically characterize the enzymatic depolymerization of κ-carrageenan, soluble and solid substrates (in both gel and powder forms) were incubated with P. carrageenovora κ-carrageenase. The average molecular mass of soluble carrageenan decreased rapidly, and all possible degradation products were observed, suggesting random degradation of κ-carrageenan. In contrast, as expected for a processive-type carrageenase, the average molecular mass of solid carrageenan decreased very slowly, and tetrasaccharide production was high. Interestingly, experimentally determined processivity was similar for gel and powder, suggesting that, in addition to an adapted catalytic site, the substrate must be in the solid state for κ-carrageenase processivity to operate, whatever the level of carrageenan ordering.

DNA-encoded libraries (DELs) have been broadly applied to identify chemical probes for target validation and lead discovery. To date, the main application of the DEL platform has been the identification of reversible ligands using multiple rounds of affinity selection. Irreversible (covalent) inhibition offers a unique mechanism of action for drug discovery research. In this study, we report a developing method of identifying irreversible (covalent) ligands from DELs. The new method was validated by using 3C protease (3CP) and on-DNA irreversible tool compounds (rupintrivir derivatives) spiked into a library at the same concentration as individual members of that library. After affinity selections against 3CP, the irreversible tool compounds were specifically enriched compared with the library members. In addition, we compared two immobilization methods and concluded that microscale columns packed with the appropriate affinity resin gave higher tool compound recovery than magnetic beads.

1. Two experiments were carried out. The first with five normal male subjects was placebo controlled and single blind, each subject being studied on two occasions. Lower body subatmospheric pressure (LBSP) was used to assess the cardiovascular effects of graded hypovolaemia before and during either a hyperinsulinaemic, euglycaemic clamp or a placebo clamp using 0.9% (w/v) NaCl only. 2. During hyperinsulinaemia, resting systolic blood pressure rose and was accompanied by forearm vasodilatation. Forearm blood flow (FABF) and heart rate (HR) were higher at each level of LBSP during than before hyperinsulinaemia. In addition, hyperinsulinaemia was accompanied by a small increase in noradrenaline, but packed cell volume did not change. 3. In the second experiment, the effects of a hyperinsulinaemic euglycaemic clamp on the cardiovascular responses to LBSP were assessed in seven diabetic subjects with peripheral and autonomic neuropathy. 4. In contrast to the normal subjects, there was a slight fall in systolic blood pressure during the clamp but no effect was noted on HR or FABF. Mean arterial blood pressure was lower at each level of LBSP during hyperinsulinaemia compared with the pre-elamp period. Packed cell volume fell during the clamp and plasma noradrenaline rose. In one of the diabetic subjects, a precipitous fall in blood pressure occurred during hyperinsulinaemia when LBSP of 10 mmHg (1.3 kPa) was applied, this manoeuvre having been well tolerated before the clamp. 5. The mode of action of hyperinsulinaemia is not clear, but there was, however, no evidence that a fall in plasma volume had occurred.

Contact surface of nanoscale sliding friction represent some new features that are different from the macro scale sliding friction, which need to seek new analysis methods. Molecular dynamics simulation is an effective method to describe microscopic phenomena. Therefore, Molecular dynamics method was used to study mechanical behavior of contact surface of nanoscale sliding friction. A molecular dynamics model of hemisphere sphere sliding on rectangular solid plane was built. State change of the micro contact area and friction force variation in the process of sliding friction were observed and analyzed after solution and simulation. The results show that, at the beginning position of the sliding, with different contact depth, contact action region of hemisphere and plane generated the atoms displacement, re-arranged and close-packed accumulation is also different. The deeper the contact depth is, the greater the atoms close-packed accumulation is, and the greater the contact deformation is. In the process of sliding friction, the contact surface of the basal body has produced lattice destruction, surface upheaval and silicon atoms close-packed accumulation, and then formed furrow scratches. At the same time the silicon atoms of the hemisphere generated atomic migration obviously and adhered on the basal body surface. The top of the hemisphere was torn and peeled, which resulted in wear. The deeper contact depth is, the more loss of the material of the hemisphere is, and wear become heavier. The curve of friction force and sliding displacement in different contact depths shows that the deeper contact depth is, the greater friction force is. The friction force increases quickly at the beginning of the sliding. Then the friction force remains steady relatively at stable sliding phase. In subsequent sliding process, due to hemisphere was worn and the original contact surface changed in size, shape and configuration state, friction force decreases obviously. Besides, in process of sliding friction, due to stick-slip effect, friction force appears obviously fluctuations. Moreover, if the sliding speed is large the changes of sliding speed have less effect on friction force when the nanoscale sphere sliding on the plane at the different speeds.

Abstract Previous work has shown that mutation of the gene that encodes the microtubule motor subunit kinesin heavy chain (Khc) in Drosophila inhibits neuronal sodium channel activity, action potentials and neurotransmitter secretion. These physiological defects cause progressive distal paralysis in larvae. To identify the cellular defects that cause these phenotypes, larval nerves were studied by light and electron microscopy. The axons of Khc mutants develop dramatic focal swellings along their lengths. The swellings are packed with fast axonal transport cargoes including vesicles, synaptic membrane proteins, mitochondria and prelysosomal organelles, but not with slow axonal transport cargoes such as cytoskeletal elements. Khc mutations also impair the development of larval motor axon terminals, causing dystrophic morphology and marked reductions in synaptic bouton numbers. These observations suggest that as the concentration of maternally provided wild-type KHC decreases, axonal organelles transported by kinesin periodically stall. This causes organelle jams that disrupt retrograde as well as anterograde fast axonal transport, leading to defective action potentials, dystrophic terminals, reduced transmitter secretion and progressive distal paralysis. These phenotypes parallel the pathologies of some vertebrate motor neuron diseases, including some forms of amyotrophic lateral sclerosis (ALS), and suggest that impaired fast axonal transport is a key element in those diseases.
