In this study, performance characteristics such as power take off (PTO) power consumption, fuel consumption, fuel consumption for the unit field-unit product were determined at different working speeds with two different PTO applications (540 and 540E) in a single row disc type silage machine. In particular, the 540E PTO application greatly reduces fuel consumption for unit work. The best results in terms of hourly fuel consumption were achieved in 540E PTO application and V1 working speed. When the field - product fuel consumption is evaluated, the best results were obtained with the 540E PTO application at the V3 working speed. When an evaluation is made considering all the parameters, it is concluded that the 540E PTO application will provide certain advantages in terms of fuel consumption compared to the 540 PTO application for the silage machine operating by taking the motion from PTO. 540E PTO application can be used as an important alternative to 540 PTO application for machines of similar capacity and features.

In this work, ZnO (Zinc Oxide) rods, which is a sensitive material for gas detection, are grown on a microcantilever surface by hydrothermal technique. Synthesis of ZnO rods consists of two processes, i.e., a formation of seed layer using Zinc Acetate Dihydrate by a dip-coating and a growth of the ZnO rods using Zinc-Nitrate-Tetrahydrate at a temperature 95°C for time variations of 2 hours, 4 hours and 6 hours. Drying condition for both seed layer and growth of ZnO rods was done at a temperature of 120 oC for 2 hours. The ZnO rods were then characterized by Fourier Transform Infrared (FTIR) in order to investigate the formation of Zn-O bond. The characterization results indicate that the metal oxide correlation is shown in the wavenumber spectrum of 540 cm-1 for a sample with 2-hour growth time, while the wavenumber spectrum of 548 cm-1 and 547 cm-1 for the growth time of ZnO rods for 4 hours and 6 hours. Wavenumber spectrum approximately at 540-548 cm-1 is predicted to be the absorption of the formation of a Zn-O bond. Experimental infection of a high-passage turkey coronavirus passaged serially in embryonated turkey eggs for 344 times (P344 TCoV 540) showed no enteritis-related clinical signs, decreased body weight gains, gross, and microscopic lesions. TCoV spike (S) protein specific antibodies appeared from 14 days post infection (dpi) and increased gradually. Virus neutralization (VN) titers of the serum from P344 TCoV 540-inoculated turkeys were 1:13 at 14 dpi, 1:16 at 28 dpi, and 1:36 at 56 dpi against P344 TCoV 540. P344 TCoV 540-inoculated turkeys were protected against the challenge by homologous P344 TCoV 540 completely or low passage P3 TCoV 540 partially as revealed by lack of histopathological alterations, absence of TCoV by immunofluorescent antibody assay in the intestines, and reduction in TCoV viral RNA loads in the intestines and feces. The serum from P344 TCoV 540-vaccinated turkeys had higher VN titers against P344 TCoV 540 than those against P3 TCoV 540. P344 TCoV 540 had 52 amino acid substitutions as compared to those of P3 TCoV in the S protein. The results indicated that a high passage TCoV can induce protective humoral and cellular immune response and have potentials to become an attenuated vaccine.

ABSTRACT Photodynamic inactivation of Staphylococcus aureus planktonic and biofilm cells by a phtotosensitizer, merocyanine 540 (MC 540), was investigated. For the planktonic experiments, MC 540 binding efficiency to bacterial cells was found to increase with both increasing MC 540 concentration and increasing incubation time, but the binding became saturated following 10 min of incubation. The antimicrobial activity was enhanced with an increasing light dose, but an increase in the light dose could not further improve the antimicrobial activity if the maximum excitation level attainable was less than the necessary minimum threshold level. Complete inactivation was achieved when the excitation level of MC 540 was somewhere above the threshold level. The relationship between antimicrobial activity and the excitation level of MC 540 revealed that the more MC 540 was excited, the more S. aureus cells were killed. For the biofilm experiments, the antimicrobial activity was enhanced with an increase in the light dose. No viable cells were detected when organisms were exposed to 15 μg of MC 540 per ml and a light dose of 600 J/cm2 or to 20 μg of MC 540 per ml and a light dose of 450 J/cm2. A quantitative analysis of MC 540 bound to biofilms was also performed, and the images from confocal laser scanning microscopy provided direct evidence that revealed the difference between the MC 540 remaining in the biofilms prior to irradiation and the MC 540 remaining in the biofilms after irradiation. The results of both the planktonic and biofilm experiments suggest that the antimicrobial activity of photodynamic inactivation of S. aureus is closely related to the excitation level of MC 540.

The chronology of the campaigns of the years 542–545 has been the subject of debate, with Michael Whitby defending the traditional interpretation that Procopius’ long account of the bubonic plague concealed the end of the year 542, whereas Geoffrey Greatrex has championed the chronology of Kislinger and Stathakopoulos, which locates Khusro’s march to Adarbiganon and the Roman defeat at Anglon in late 542 and the siege of Edessa in 543, with Procopius failing to note the end of a year during peace negotiations in 544–545. Considerations of the progress of Khusro I’s invasion in 542 in light of his probable speed of march, and the distances he had to cover, coupled with the relatively slow advance of bubonic plague over large land masses and Procopius’ practice in arranging his material, point to the missing year-end, being that of 542/543. While the new chronology cannot absolutely be ruled out, the assumptions on which it is based are shaky.

Abstract Merocyanine 540 (MC 540) is a photoactive dye used to purge bone marrow of tumor cells in autologous bone marrow transplantation. The effects of MC 540 on the lymphoid components in the marrow are unknown. This study evaluates the treatment of lymphocytes by MC 540 (15 micrograms/mL) and light (70 W/m2) on: (1) phytohemagglutinin and Con A- induced proliferation; (2) allogeneic mixed lymphocyte cultures (MLC); (3) the regulation of Ig synthesis by T cells; and (4) the ability of B cells to produce polyclonal Igs as measured by an enzyme-linked immunosorbent assay-plaque assay. The results show that MC 540 and light treatment reduced Con A-stimulated T-cell proliferation greater than 50% after 30 minutes and greater than 80% after 60 minutes of MC 540-sensitized photoirradiation. Ninety minutes of MC 540 and light exposure (designated treatment) inhibited MLC greater than 90%. In polyclonal Ig synthesis, T-cell helper activity could be abrogated by 90 minutes of treatment in cocultures containing untreated B cells. Purified B cells treated for 90 minutes cocultured with normal T cells did not produce Ig. Treatment of B cells completely inhibited Epstein- Barr virus-stimulated Ig synthesis. These data show that T- and B-cell immunity is suppressed by the MC 540-sensitized photoirradiation. Treatment of bone marrow with MC 540 and light may have profound effects on immune reconstitution in autologous marrow graft recipients. More provocative is the fact that the same immunomodulatory effects may be applicable to partially mismatched marrow transplant situations as a means of reducing graft-versus-host reactions.

A newunsymmetrical photochromic diarylethene 1-(2-cyanophenyl)-2-[2-methy-5-(3-fluorophenyl)-3-thienyl]perfluorocyclopentene (1a) has been synthesized. Its photochromic properties in solution and PMMA film were investigated. Diarylethene 1a changed the color from colorless to purple upon irradiation with 297 nm UV light, in which absorption maxima were observed at 540 nm in hexane and at 545 nm in PMMA film, respectively. This new photochromic system also exhibited remarkable fluorescence switching in the solid state. The results demonstrated that it can be potentially used as polarization holographic optical recording medium.

Abstract The Friend virus complex was used as a model to study the effects of merocyanine 540 (MC 540)-mediated photosensitization on enveloped viruses. Simultaneous exposure to the lipophilic dye MC 540 and white light inactivated cell-free virus, cell-associated virus, and virus- transformed cells. When used under experimental conditions that are known to preserve most mature blood cells, at least some coagulation factors, and a significant portion of the pluripotent hematopoietic stem cell compartment, MC 540-mediated photosensitization reduced virus titers by greater than or equal to 4 log and the concentration of in vitro clonogenic erythroleukemia cells by greater than or equal to 5 log. Animals that received a single intravenous injection of photosensitized virus were resistant to a subsequent challenge with live virus. High sensitivity to MC 540-mediated photosensitization appears to be a property that is shared by other enveloped viruses. Thus, photosensitization mediated by MC 540 may be of benefit in the sterilization of blood products (in particular, cellular products), the production of vaccines, and selected areas of antiviral therapy.

Abstract A simple, novel, sensitive, and specific spectrophotometric method was developed and validated for the determination of azithromycin (AZ), clarithromycin (CLA), and roxithromycin (ROX) in bulk powders and their dosage forms. The proposed method was based on the interaction of any of the cited drugs with 2,4-dinitrophenylhydrazine in the presence of an acid catalyst, followed by treatment with a methanolic solution of potassium hydroxide; an intensely colored chromogen was formed that was measured in dimethylformamide, as the diluting solvent, at 542−545, 523−526, and 539−542 nm for AZ, CLA, and ROX, respectively. All variables affecting the development of the measured chromogens were studied and optimized. Beer's law was obeyed in the concentration ranges of 540, 535, and 535 μg/mL for AZ, CLA, and ROX, respectively, with good correlation coefficients (0.99910−9999). The limits of detection for this method ranged from 0.77 to 1.47 μg/mL, and the relative standard deviations were 1.24−1.8%. The proposed method was applied successfully to the determination of the 3 drugs in pure bulk form, tablets, and suspensions without interference from commonly encountered additives. The results compared favorably with those of a previously reported method. The mechanism of the reaction was also studied.

Many new photosensitizers and laser wavelengths are being tested to improve photodynamic therapy by enhancing specific tumor uptake and/or retention, lowering systemic toxicity, and increasing laser tissue penetration. In this study the potential synergistic effects of rhodamine-123 (Rh-123) and merocyanine-540 (MC-540) sensitization of human tumor cell lines after laser exposure were explored. In a first series of experiments, the kinetics of uptake of Rh-123 and M-540 were tested on three human leukemia cell lines (K562, RAJI, 729HF2), P3 squamous carcinoma, and M26 melanoma. Our results demonstrate a clear difference in the rate and amount of uptake of MC-540 (K562 > P3 > RAJI > 729HF2 > M26) and Rh-123 (P3 > RAJI > 729HF2 > K562 > M26) by these cell lines. In a second series of experiments, M26 tumor cells were sensitized with either Rh-123 (1 μg/ml) or with MC-540 (20 μg/ml) alone or with a combination of the two dyes for 60 minutes, then exposed to the argon (514.5 nm) laser at nonthermal energy levels. Our results demonstrate a significant enhancement of the tumoricidal effects of the laser on M26 carcinoma cells after sensitization with both dyes together (MC-540 and Rh-123) when compared to each dye alone. As with combination antibiotherapy, the synergistic effects of two laser dyes that have different intracellular targeting sites appear to enhance tumoricidal effects significantly after exposure to a matching laser wavelength. The data provide evidence for effective laser phtototherapy by dye synergy.

Jumbo phages, which have a genome size of more than 200 kb, have recently been reported for the first time. However, limited information is available regarding their characteristics because few jumbo phages have been isolated. Therefore, in this study, we aimed to isolate and characterize other jumbo phages. We performed comparative genomic analysis of three Erwinia phages (pEa_SNUABM_12, pEa_SNUABM_47, and pEa_SNUABM_50), each of which had a genome size of approximately 360 kb (32.5% GC content). These phages were predicted to harbor 546, 540, and 540 open reading frames with 32, 34, and 35 tRNAs, respectively. Almost all of the genes in these phages could not be functionally annotated but showed high sequence similarity with genes encoded in Serratia phage BF, a member of Eneladusvirus. The detailed comparative and phylogenetic analyses presented in this study contribute to our understanding of the diversity and evolution of Erwinia phage and the genus Eneladusvirus.

This article seeks to support the earlier dating of campaigns on the Roman eastern frontier in the 540s. It addresses points made in a recent contribution by Michael Whitby, who argued that the traditional chronology, which places a Roman invasion of Persarmenia in 543 and a Persian siege of Edessa in 544, should be retained. The article seeks to demonstrate that the grounds he offers are inadequate and concludes therefore that the earlier dating, according to which the Romans invaded Persarmenia in autumn 542 and the Persians besieged Edessa in 543, is to be preferred.

O uso de misturas de glyphosate, em tanque, para manejo de espécies de plantas daninhas de difícil controle tem sido prática comum entre os agricultores brasileiros. Desta forma, este trabalho teve como objetivo avaliar a eficácia e seletividade de misturas, em tanque, de herbicidas com glyphosate para o controle de trapoeraba (Commelina benghalensis L.), erva-de-touro (Tridax procumbens L.) e capim-carrapicho (Cenchrus echinatus L.) na cultura da soja RR®. O experimento foi conduzido em Maracaí, São Paulo, no período de novembro de 2006 a março de 2007, utilizando-se o cultivar CD-214RR® e delineamento experimental de blocos ao acaso, com 21 tratamentos e quatro repetições. Os tratamentos foram constituídos da aplicação de: glyphosate (180; 360; 540 e 720 g ha-1); glyphosate em sequencial (180/360; 360/360 e 540/360 g ha-1); glyphosate + chlorimuron-ethyl 360+10; 540+10; 360+5/ 360+5 g ha-1); glyphosate + lactofen (360+120; 540+120; 360+60/ 360+60 g ha-1); glyphosate + cloransulam-methyl (360+30; 540+30; 360+16,9/ 360+12,9 g ha-1); glyphosate + carfentrazone (360+4 g ha-1); glyphosate + imazethapyr (360+50 g ha-1); glyphosate + imazethapyr (177,8+30 g ha-1) e testemunhas capinada e sem capina. Apesar da similaridade de produtividade de grãos entre os tratamentos com glyphosate isolado e sequencial, nas doses 540, 720 e 540/ 360 g ha-1, as misturas em tanque com chlorimuron-ethyl, cloransulam-methyl, lactofen e imazethapyr favoreceram o controle de espécies de plantas daninhas tolerantes ao glyphosate como C. benghalensis e T. procumbens.

In this paper, the mesoporous silica SBA-15 surface was modified by using silane coupling agent KH-540. In order to investigate the best modification conditions, effects of KH-540 dosage, reaction time and temperature were studied. The modified mesoporous silica SBA-15 was characterized by TGA, FTIR, XRD and N2 adsorption-desorption etc. The best modification conditions were obtained as follows: KH-540 dosage is 0.5g, reaction temperature is 90°C and reaction time is 4 hours. The results showed that KH-540 is successfully grafted on the surface of SBA-15, the SBA-15-KH540 possess a well ordered hexagonal mesoporous structure. Compared with pure SBA-15, the surface area, the pore size and the pore volume of the SBA-15-KH540 are decreased slightly.

Background Asthma Action Plans (AAPs) are recommended for pediatric patients to help improve asthma control. Studies have shown variable results for unscheduled doctor and emergency room visits. AAPs may have an impact on parental self-efficacy for asthma management as well as on other daily living factors that are valuable for patients and families, such as the number of missed school days and parental workdays, and on school and caregiver management. Objective The purpose of this study is to understand parent perceptions of AAPs. The goals of this analysis were threefold, including examining (1) the association between pediatric AAPs and parental self-efficacy, (2) parent perceptions of the helpfulness of an AAP for daily living factors, and (3) associations with the type of provider who gave the AAP (a primary care provider or an asthma specialist). Methods A national cross-sectional online survey was completed in October 2018 by parents of children with asthma aged 0-17 years. Survey questions included the presence or absence of a pediatric AAP, the Bursch Parental Self-efficacy for Asthma scale, parental perceptions of the AAP's helpfulness with regard to daily living factors ranked on a 5-point Likert scale, and the provider type who gave the AAP. Survey responses were summarized in terms of percentages or means and standard deviations. A 2-sample t test and analysis of covariance were used to compare self-efficacy for asthma and parental-perception-of-helpfulness scores between subjects with an AAP versus subjects without an AAP. All reported P values were 2-sided. Results A total of 704 parents with a child with asthma completed the survey. The parents had a mean age of 37.5 years (SD 10.9), and 82% (577/704) were women and 18% (127/704) were men. Most (564/704, 80%) parents had an AAP for their child; 65% (367/564) were written, 51% (286/564) were online, and 84% (474/564) were available at school. The Bursch Self-efficacy scale was significantly higher for parents with an AAP (mean 57.7, SD 8.6) versus no AAP (mean 55.1, SD 9.9; P<.001). Parents reported that they agreed/strongly agreed that an AAP was helpful for daily living factors, including managing asthma (446/544, 82%), decreased parental missed workdays (367/544, 68%), decreased child missed-school days (396/542, 73%), and for when a child is at school (422/541 78%), with other caregivers (434/543, 80%), doing normal activities (421/540 78%), and leading a normal life (437/540 81%). Parents agreed/strongly agreed that an AAP was helpful from all provider types: a pediatric provider (583/704, 82.8%), a family practice provider (556/704, 79%), and an asthma specialist (594/704, 84.4%). There was no significant difference (P=.53) between the type of provider who gave the AAP. Conclusions Parents who had pediatric AAPs for their children reported increased parental self-efficacy compared to those who did not have AAPs. Parents found AAPs helpful for decreasing missed time from work and school, and for asthma management when at home, school, and with other caregivers. Significant AAP helpfulness was seen regardless of the provider who gave the AAP, the parent's education, and income level. Findings support the usefulness of pediatric AAPs for families and the development of easily sharable electronic AAPs for children.

The cell mixtures were incubated in vitro with mafosfamide (AZ) or with the photoreactive dye merocyanine 540 (MC-540). A quantity of 10(4) cells of the treated suspensions was dispensed into microculture plates, and graded cell numbers of the line used to contaminate the normal marrow were added. Limiting-dilution analysis was used to estimate the frequency of leukemia cells persisting after treatment with the decontaminating agents. Treatment with AZ or MC-540 produced a total elimination (ie, 6 logs or 5.3 logs respectively) of B cell acute leukemia cells (CCRF-SB), whereas nearly 1.7 logs and 2 logs of K-562 acute myelogenous blasts were still present in the cell mixtures after treatment with MC-540 and AZ, respectively. Treatment of the Nalm-6- contaminated cell mixtures with AZ resulted in 100% elimination of clonogenic cells, whereas nearly 80% decontamination was obtained with MC-540. Our results suggest that treatment with AZ could be an effective method of eliminating clonogenic tumor cells from human bone marrow. MC-540, shown by previous studies to spare sufficient pluripotential stem cells to ensure hemopoietic reconstitution in the murine model and in clinical application, has comparable effects and merits trials for possible clinical use in autologous bone marrow transplantation.

We examined the function of a highly conserved Histidine rich sequence ofamino acids found in the carboxyl-terminal of the Na+/H+exchanger (NHE1). A fusion protein containing the sequenceHYGHHH (540–545) and the balance of the carboxyl terminalof the protein did not bind calcium but bound to an immobilizedmetal affinity column and could be used to partially purify theexchanger protein. Mutation of the sequence to either HYGAAA orHYGRRR did not affect activity of the intact protein. Mutationto HHHHHH did not affect proton activation of the Na+/H+exchanger or localization but caused a decreased maximal velocitysuggesting that this conserved sequence is important in maximalactivity of the Na+/H+ exchanger.

SummaryFragment X components (Mr 225,000 to 333,000) were distinguished on sodium dodecyl sulfate polyacrylamide gels. Western blotting with monoclonal antibodies to Aα-chain segments demonstrated that the Aα-chains of fibrinogen and the largest fragment X components (Mr 285,000-340,000) contained both Aα 259-276 and Aα 540-554. Fragment X components of Mr 270.000-285,000 contained Aα 259-276 but lacked Aα 540-554, whereas the smallest fragment X components (Mr 225.000-270,000) contained neither Aα 540-554 nor Aα 259-276. Studies of the small peptides generated during fragment X formation complemented the studies of the large molecules, by demonstrating peptides containing both Aα 259-276 and Aα 540-554 (Mr 41,600-41,800 and Mr 38,700-38,900), peptides containing Aα 540-554 but not Aα 259-276 (Mr 20,500-21,000 and Mr 17,300-17,500) and peptides containing only Aα 259-276 (Mr 23,600-24,000 and Mr 20,500-21,000). Cleavage of Bβ 1-42 from the amino terminal ends of the Bβ-chains, measured with a specific radioimmunoassay, was linear until 1.6 moles per mole of fibrinogen had been relased, and coincided with loss of the central and carboxy terminal Aα-chain regions, i. e. Aα 259-276 and Aα 540-554. Based on present and previously reported data, a model is proposed for the evolution of the heterogenous group of fragment X derivatives from fibrinogen with the simultaneous release of small peptides. Features of this model include1. asymmetric cleavage of the fibrinogen dimer and2. proteolyses of several different bonds occurring simultaneously but at distinct rates.

The Escherichia coli haemoglobin-like flavohaemoprotein (Hmp) has been purified to near homogeneity using two chromatographic steps. The prosthetic groups are identified as FAD and protohaem IX. SDS/PAGE has indicated a molecular mass of 44 kDa for the monomeric protein consistent with the amino-acid sequence deduced from the hmp+ gene. The protein, as isolated, is in the Fe(III) state, exhibiting absorbance maxima at 403.5, 540 (shoulder) and 627 nm. The ferrous and carbonmonoxyferrous states resemble those of haemoglobin, showing maxima at 431.5 and 558 nm, and 421, 542 and 566 nm respectively. Upon aerobic addition of NAD(P)H, the ferric state is reduced to the oxygenated Fe(II) state, characterized by maxima at 413, 544 and 580 nm. This oxy form is not stable and slowly decays to the ferric state. Addition of dithionite and nitrite to the ferric protein results in the formation of a nitrosyl complex, whose e.p.r. characteristics indicate that the b-type haem is attached to the protein through a nitrogenous ligand, probably originating from a histidine residue.

SummarySperm capacitation is crucial for fertilization. However, debate continues on exactly how, where and when capacitation is elicited in the bovine female genital tract. In this study we used merocyanine-540 and the chlortetracycline (CTC) assay to test how capacitation of bull spermatozoa is affected in vitro by exposure to oviductal fluid (ODF) collected in vivo, various glycosaminoglycans (GAGs) or bicarbonate. Following different durations of exposure, spermatozoa were stained with CTC or merocyanine-540, and evaluated with epifluorescent light microscopy or flow cytometry, respectively. Incubation time did not significantly affect capacitation. Exposure (30–120 min) to ODF capacitated (p < 0.05) bull spermatozoa as measured by either merocyanine-540 or CTC. Hyaluronan was the only GAG that induced a significant increase in B-pattern spermatozoa (capacitated; p = 0.012) compared with controls. Dermatan sulphate also induced capacitation (merocyanine-540 high fluorescence; p = 0.035). Exposure to bicarbonate-enriched media also yielded an increase in merocyanine-540 high fluorescence (p < 0.0001). When bicarbonate was added to the other treatments (ODF or GAGs) an equal increase in merocyanine-540 high fluorescence was noted (p < 0.0001), compared with before addition of bicarbonate and independent of the treatment before exposure. There was no significant difference in the number of B-pattern spermatozoa when bicarbonate was added, but an significant increase in spermatozoa with an acrosome-reacted (AR)-pattern (p < 0.0001) was observed. Exposure of spermatozoa to solubilized zonae pellucidae significantly increased the AR-pattern spermatozoa (p = 0.016). In conclusion, ODF was more potent in inducing capacitation of bull spermatozoa than the individual GAGs. Our results also indicate that bicarbonate is an effector of bull sperm capacitation.

We report a case of a previously healthy woman, Mst. Borna Khatun, 25 years old, who developed puerperal sepsis with extensive lower abdominal necrotizing fasciitis after an uneventful home delivery. She presented to us with the complaints of lower abdominal pain, fever & foul smelling per vaginal discharge for 09 days with history of vaginal delivery by an untrained dai at home 13 days back. On general examination she was toxic, her temperature was 103F, and lower left half of abdomen was tender & rigid. O presente trabalho teve como objetivo avaliar a influência da simulação de orvalho na eficiência do herbicida glyphosate sobre Brachiaria decumbens. Para isso, foram realizados dois ensaios simultâneos. No primeiro, os tratamentos foram constituídos pela combinação de duas doses de glyphosate (270 e 540 g ha-1), três intervalos de tempo entre a pulverização do herbicida e a simulação de orvalho (0,0; 4,0 e 8,0 horas) e cinco volumes de orvalho (250, 500, 750, 1.000 e 2.000 mL m-2), além de três testemunhas adicionais, dispostos em esquema fatorial (2x3x5)+3, com quatro repetições. No segundo ensaio, os tratamentos foram constituídos pela combinação da aplicação do orvalho simulado em cinco volumes (250, 500, 750, 1.000 e 2.000 mL m-2) com a pulverização do glyphosate, logo após o orvalho simulado, em duas doses (270 e 540 g ha-1), dispostos em esquema fatorial (5x2), com quatro repetições. A dose de 270 g ha-1 de glyphosate, independentemente do intervalo de tempo entre a pulverização do herbicida e a aplicação do orvalho simulado ou dos volumes de orvalho, não proporcionou controle de B. decumbens. Os volumes crescentes de orvalho sobre o glyphosate pulverizado na dose de 540 g ha-1 interferiram no controle de B. decumbens, sendo necessário intervalo de oito horas entre a aplicação do glyphosate e a ocorrência do orvalho para se obter controle satisfatório da planta daninha. Aplicações de glyphosate sobre plantas de B. decumbens na presença do orvalho resultaram em baixos níveis de controle na dose de 540g ha-1, não apresentando qualquer controle na dose menor. Embora tenha havido interferência do volume de orvalho simulado na eficácia do herbicida, o controle de B. decumbens pelo glyphosate pareceu estar mais relacionado com o tempo de ocorrência do orvalho após as pulverizações.

Avaliou-se a resposta de nove cultivares de algodoeiro, de importância econômica no Estado do Mato Grosso, quanto à intoxicação causada por subdoses de glyphosate. Os cultivares de algodoeiro utilizados foram Fabrika, Makina, ITA-90, FM 986, FM 966, Delta Opal, BRS Facual, Antares e Coodetec 407. As plantas foram cultivadas em tubetes preenchidos com substrato de solo e mantidas em casa telada, tendo recebido a aplicação do glyphosate aos 20 dias após a emergência, época em que apresentavam quatro folhas verdadeiras. As subdoses de glyphosate, simulando deriva, foram de 270 e 540 g ha-1. Também foi utilizada testemunha, sem aplicação do herbicida, para efeito de comparação. Foram realizadas avaliações semanais até 42 dias após a aplicação dos tratamentos (DAA), período em que também foi tomada a altura das plantas. Os sintomas visuais de intoxicação iniciaram-se aos 3 DAA, caracterizados pelo amarelecimento das pontas das folhas mais novas, seguido de murchamento do ápice das plantas. Na dose de 270 g ha-1 esses sintomas foram de baixa intensidade, mas a 540 g ha-1 causaram, na maioria dos casos, toxidez "preocupante" a "muito alta". Os cultivares BRS Facual e FM 986 mostraram-se os mais suscetíveis. A altura das plantas foi mais afetada quando se aplicou a menor dose de glyphosate. Houve recuperação de todos os cultivares tratados com 270 g ha-1 de glyphosate até os 42 DAA. Quando tratados com 540 g ha-1 de glyphosate, os cultivares Fabrika, Coodetec 407, BRS-Facual e ITA-90 foram mais sensíveis, apresentando redução de altura entre 84 e 90% aos 42 DAA. Os cultivares menos sensíveis na dose de 270 g ha-1 de glyphosate não foram os mesmos para a dose de 540 g ha-1.

Background and Purpose: The outcome of endovascular treatment in stroke patients with a large ischemic core is not always satisfactory. We evaluated whether the severity of baseline diffusion-weighted imaging abnormalities, as assessed by different apparent diffusion coefficient (ADC) thresholds, correlates with the clinical outcome in these patients after successful endovascular treatment. Methods: In 82 consecutive patients with a large vessel occlusion in the anterior circulation admitted ≤24 hours after onset, a baseline diffusion lesion volume (ADC ≤620×10 −6 mm 2 /s [ADC 620 ]) ≥50 mL and successful recanalization by endovascular treatment were retrospectively investigated. Lesion volumes of 3 ADC thresholds (ADC 620 , ADC ≤520×10 −6 mm 2 /s [ADC 520 ], and ADC ≤540×10 −6 mm 2 /s [ADC 540 ]) were measured using an automated Olea software program. The performance of the ADC 520 /ADC 620 and ADC 540 /ADC 620 ratios in predicting the functional outcome was assessed by receiver operating characteristic curve analysis. The ADC ratio with optimal threshold showing better receiver operating characteristic performance was dichotomized at its median value into low versus high subgroup and its association with the outcome subsequently evaluated in a multivariable logistic regression model. Results: The median baseline diffusion lesion volume was 80.8 mL (interquartile range, 64.4–105.4). A good functional outcome (modified Rankin Scale score, ≤2) was achieved in 35 patients (42.7%). The optimal threshold for predicting the functional outcome was identified as ADC 540 /ADC 620 (area under the curve, 0.833) and dichotomized at 0.674. After adjusting for age, baseline National Institutes of Health Stroke Scale score, intravenous tissue-type plasminogen activator, baseline diffusion lesion volume, and onset-to-recanalization time, a low ADC 540 /ADC 620 was independently associated with a good functional outcome (adjusted odds ratio, 10.72 [95% CI, 3.06–37.50]; P <0.001). Conclusions: A low ADC 540 /ADC 620 , which may reflect less severe ischemic stress inside a diffusion lesion, may help to identify patients who would benefit from endovascular treatment despite having a large ischemic core.

ABSTRACT Planets and stars ultimately form out of the collapse of the same cloud of gas. Whilst planets, and planetary bodies, readily loose volatiles, a common hypothesis is that they retain the same refractory composition as their host star. This is true within the Solar system. The refractory composition of chondritic meteorites, Earth, and other rocky planetary bodies are consistent with solar, within the observational errors. This work aims to investigate whether this hypothesis holds for exoplanetary systems. If true, the internal structure of observed rocky exoplanets can be better constrained using their host star abundances. In this paper, we analyse the abundances of the K-dwarf, G200-40, and compare them to its polluted white dwarf companion, WD 1425+540. The white dwarf has accreted planetary material, most probably a Kuiper belt-like object, from an outer planetary system surviving the star’s evolution to the white dwarf phase. Given that binary pairs are chemically homogeneous, we use the binary companion, G200-40, as a proxy for the composition of the progenitor to WD 1425+540. We show that the elemental abundances of the companion star and the planetary material accreted by WD 1425+540 are consistent with the hypothesis that planet and host-stars have the same true abundances, taking into account the observational errors.

Abstract To assess the potential of photoradiation therapy for the in vitro purging of residual tumor cells from autologous bone marrow (BM) transplants, we studied normal marrow and tumor cell clonogenicity in response to different light-activated compounds by using the fluorescent dyes dihematoporphyrin ether (DHE) and merocyanine-540 (MC- 540). After photoradiation of cells with white light, both DHE and MC- 540 showed high cytocidal activity toward lymphoid and myeloid neoplastic cells but had a significantly lesser effect on normal granulocyte-macrophage (CFU-GM), erythroid (BFU-E), and mixed colony- forming (CFU-GEMM) progenitor cells. Acute promyelocytic leukemia (HL- 60), non-B, non-T, CALLA-positive acute lymphoblastic leukemia (Reh), and diffuse histocytic B cell lymphoma (SK-DHL-2) cell lines were exposed to different drug concentrations in combination with white light at a constant illumination rate of 50,000 lux. With DHE doses varying from 2.0 to 2.5 micrograms/mL and MC-540 concentrations of 15 to 20 micrograms/mL, clonogenic tumor cells could be reduced by more than 4 logs when treated alone or in mixtures with normal irradiated human marrow cells. However, preferential cytotoxicity towards neoplastic cells was highly dependent on the mode of light activation. MC-540 had no substantial effect on malignant lymphoid (SK-DHL-2) and myeloid (HL-60) cells and on normal marrow myeloid (CFU-GM) precursors when drug incubation was performed in the dark and followed by light exposure of washed cells. Equal doses of MC-540 (15 to 20 micrograms/mL) could preferentially eliminate tumor cells under conditions of simultaneous light and drug treatment (30 minutes at 37 degrees C). When using DHE (2.5 micrograms/mL), 29.3%, 46.8%, and 27.5% of normal marrow CFU-GM, BFU-E, and CFU-GEMM, respectively, were spared after sequential drug and light exposure of cells, whereas simultaneous treatment reduced both normal (CFU-GM) and neoplastic cells below the limits of detection. In summary, our results indicate the usefulness of various photoradiation models for the ex vivo treatment of leukemic and lymphomatous bone marrow autografts.

To investigate the genetic variation of trembling aspen ( Populus tremuloides Michx.) in ecophysiological responses to [CO2] elevation, 1-year-old seedlings of four provenances (three families per provenance) from northwestern Ontario were exposed to three [CO2] levels in the greenhouse: ambient (360 ppm), 1.5 × ambient (540 ppm), and 2 × ambient (720 ppm). Biomass and foliage gas exchange were examined after 60 d of treatment. [CO2] elevation significantly increased the rate of net CO2 assimilation and photosynthetic water use efficiency. The stimulation was generally greater in the 540 ppm [CO2] than in 720 ppm [CO2]. The 720 ppm [CO2] resulted in a 10% photosynthetic down-regulation, but no down-regulation was detected in the 540 ppm CO2 treatment. The 540 ppm [CO2] (but not the 720 ppm) treatment significantly decreased stomatal conductance and transpiration rate in the provenances from the Great Lakes – St. Lawrence Region but not in those from the Boreal Region. The intercellular to ambient CO2 concentration ratio (Ci/Ca) was significantly higher under 720 ppm [CO2] than under the other two [CO2]. The CO2 elevations generally increased the total and root biomass, and the stimulation was greater in the 540 ppm [CO2] than in the 720 ppm [CO2] treatment. The two provenances from the Great Lakes – St. Lawrence region generally had greater biomasses than those from the boreal region, while there were no significant differences between them in the physiological variables. However, we did not find any significant differences between provenances in the responses of biomass to [CO2] treatments.

Objetivos: Determinar los niveles de hormona de crecimiento (GH) en sujetos varones adultos, a nivel del mar (Lima, 150 msnm) y en la altura (Cajamarca: 2 750 msnm; Huancayo: 3 280 msnm; Morococha: 4 540 msnm). Diseño: Estudio descriptivo, observacional, transversal y prospectivo. Lugar: Ciudades de Lima, Cajamarca, Huancayo y Morococha, Perú. Participantes: Adultos varones, nativos de localidades de diferentes altitudes. Intervenciones: Previo consentimiento informado, a 41 adultos varones nativos de su localidad de origen [12 sujetos alnivel del mar (Lima 150 m), 10 sujetos en Cajamarca (2 750 m), 12 sujetos en Huancayo (3 280 m) y 7 sujetos en Morococha (4 540 m)] se tomó muestras de sangre en ayunas y en condiciones de reposo. Principales medidas de resultados: Niveles sanguíneos de hormona de crecimiento (GH) en sangre. Resultados: La edad promedio de los participantes fue 28 años (rango de 20 a 35 años). Los valores promedios de GH encontrados fueron: a 4 540 m, 3,87 ± 0,16 ng/mL; a 3 280 m, 1,31 ± 0,09 ng/mL; a 2 750 m, 0,63 ± 0,08 ng/mL; y a 150 m, 0,54 ± 0,04 ng/mL. Hubo diferencia significativa (p &lt; 0,05) entre los valores de GH encontrados en la altura a 4 540 m y 3 280 m con respecto al nivel del mar 150 m. No se encontró diferencia significativa entre los valores encontrados a 2 750 m con respecto a 150 m. Conclusiones: Los niveles de GH son mayores en los sujetos nativos a 3 280 m (Huancayo, n=12) y 4 540 m (Morococha, n=7) respecto a los sujetos que viven a 150 m (Lima, n=12).
