Résumé :
Abstract Introduction:. Breast cancer (BC) brain metastases (BM) are a growing clinical problem as survival for metastatic BC improves. Genomic analyses of BM are key to developing more targeted therapeutic interventions. In this study we explored the gene rearrangements of BM and compared these to a cohort of non-paired local breast cancers (BCs) and non-CNS metastasis (N-CNS). Methods:. We analyzed 822 BMs and compared them to 11,988 local, breast-biopsied BCs and 15,516 N-CNS (samples unpaired) profiled with comprehensive genomic profiling (CGP) (Foundation Medicine, Cambridge, MA, USA) examining all classes of alterations in at least 324 genes. The prevalence of individual gene rearrangements within the BM was compared to those within BC and N-CNS cases. Further analysis was undertaken by classifying BM, BC and N-CNS specimens with respect to their oestrogen receptor (ER) and human epidermal growth factor receptor 2 (HER2) status, available for a subset of samples. Results:. CGP identified 307 rearrangement events in BMs impacting 169 genes. Twenty genes exhibited recurrent rearrangement events with a prevalence greater than 0.5% in either the BM, BC or N-CNS. 10 of these 20 genes were significantly more prevalent within the BM specimens vs. BC and N-CNS (p<0.05). The 10 significant genes and their prevalence in BM were as follows: CDK12 (3.53%, 29/822), RB1 (1.95%, 16/822), ETV6 (1.70%, 14/822), FANCA (0.73%, 6/822), PALB2 (0.73%, 6/822), ATR (0.61%, 5/822), CTNNA1 (0.61%, 5/822), FBXW7 (0.61%, 5/822), KDM5C (0.61%, 5/822) and MSH6 (0.61%, 5/822). BMs were enriched with CDK12 rearrangements (3.53%, 29/822) as compared to the BC (0.86%, 103/11,988; p=4x10-9) and N-CNS specimens (0.68%, 105/15,516; p=2x10-11). CDK12 rearrangements had a significantly higher propensity within HER2+ BM (14.59%, 27/185) as compared to the HER2+ BC (7.80%, 86/1,102; p=0.0046) and HER2+ N-CNS (7.87%, 94/1,194; p=0.0048). Further analysis by receptor status is outlined in Table 1. Conclusion:. These data demonstrate that overall rearrangements are uncommon in BMs. However, we identified that the most common rearrangement involves CDK12 which has a relatively high prevalence in HER2+ BMs compared to HER2+ primary BCs and N-CNS. These data support more detailed investigation of the role and importance of CDK12 rearrangements in BM secondary to breast cancer. Résumé :
Wilson’s disease (WD) is a rare, treatable genetic disorder with multi-organ symptoms related mainly to copper accumulation. Most patients become aware of the disease as young adults, thus knowledge on fertility, pregnancy course and outcome is very important both for patients and physicians. The aim of this study was to perform a systematic review and meta-analysis of pregnancy outcomes in women with WD. This systematic literature review was performed according to Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. Studies were identified by searching the PubMed database (up to 12 January 2022) and by screening reference lists. We found 49 publications, including 13 retrospective studies and 36 series and case reports on pregnancy outcomes in WD patients. In total, descriptions of 449 pregnant women with 822 pregnancies were retrieved. Successful deliveries were achieved in 78.3% (644/822) of all pregnancies. Spontaneous abortions were observed in 21.7% (178/822) of pregnancies, more frequently in patients who were untreated 68.6% (96/140). Analyzing maternal outcome, 2.2% (18/822) of pregnancies were associated with the aggravation of neurological symptoms. Symptoms of hepatic deterioration were observed in 4.6% (38/822) of cases. These were usually transient and recovered after pregnancy; however, death due to liver failure was observed in 0.2% (2/822) of cases. Birth defects occurred in 4.7% (39/822) of pregnancies. The available meta-analysis showed statistically significant positive associations between anti-copper treatment and pregnancy outcome. Our results document the significance of anti-copper treatment as the main factor leading to successful pregnancy, as well as positive outcomes for women with WD.

Résumé :
A field experiment was conducted during winter season (1 998-99 and 1999-2000) to assess the effect of 4 ir- rigation schedules based on irrigation water : cumulative pan evaporation (IW : CPE) ratios on performance 2 oat varieties ('JHO 822' and 'JHO 851') revealed that the varieties did not differ for fodder yield and N concentration. However, based on crude protein yield (qlha) 'JHO 851' (6.08) was found significantly superior to 'JHO 822' (5.29), whereas in respect of water-use efficiency (kg dmlha-mm) 'JHO 822' (28.6) was superior to 'JHO 851' (20.3). Among the 4 irrigation schedules, fodder yield and water-use efficiency increased significantly with increasing IW: CPE ratio from 0.0 to 0.9 ratio.

Résumé :
The blooming period of the cucumber plant depended on the characteristics of the varieties, meteorological conditions, the time of planting, and the order of harvesting the fruits. Generative organs appeared in the leaf axils on the 15th-18th day after germination in the “Uzbeksky 740” variety, and on the 18th-22nd day in the “Margelansky 822” variety. After some time, it was possible to determine the gender of the flower. Male flowers bloomed first: 35-40 days after germination in “Uzbeksky 740” variety, 50-55 days in “Margelansky 822” variety. Blooming of female flowers is observed after 42-47 days after germination in “Uzbeksky 740” variety and after 57-62 days in “Margelansky 822” variety. In the Tashkent region, the blooming of female flowers in the “Uzbeksky 740” variety coincided with the 40th day from the day of emergence of sprouts at the optimal density between plants. This was 45% of the period from germination to fruit ripening. When the plant thickened to 142,800/ha, the blooming of female flowers was delayed by 2 days, fruit formation by 8 days, and fruit ripening by 22 days. This was found that the period of branching - blooming occupied 38% of the period of branching - fruit ripening.

Celem niniejszego artykułu jest dokonanie syntetycznej analizy prawnoporównawczej wybranych aspektów obowiązkowegoraportowania krajowych schematów podatkowych w Polsce i Portugalii w związku z implementacją dyrektywyRady (UE) 2018/822 z dnia 25 maja 2018 r. zmieniającej dyrektywę 2011/16/UE w zakresie obowiązkowej automatycznejwymiany informacji w dziedzinie opodatkowania w odniesieniu do podlegających zgłoszeniu uzgodnień transgranicznych1(tzw. dyrektywy DAC-6).

While the general idea is to illustrate how William Shakespeare reflected the contemporary conflicts and problems of the Elizabethan society, the particular aim of the thesis is to offer a close critical analysis of Shakespeare's Henry IV Part 1 and Part 2, Henry V and Henry VI Part 1, Part 2 and Part 3 plays in an eclectic critical approach derived from the theoretical principles of New Historicism and Cultural Materialism. In order to provide a better understanding of the plays studied in the thesis, there is a presentation of the development of drama, both religious and secular, in the Reformation period. In addition to this, main features of Cultural Materialism and New Historicism are given. The English Reformation and its effects on drama have been given in the introductory chapter. In the first chapter, contemporary religious controversies as reflected in Shakespeare's 1 and 2 Henry VI plays are discussed. The second chapter deals with the reflections of contemporary social conflicts in especially the Jack Cade episode of Shakespeare's 2 Henry VI. In the third chapter, reflections of political conflicts in Shakespeare's Henry V, Henry V, and Henry VI plays are analysed in terms of the appropriation of commoners by the ruling class for the preservation of the dominant order. The thesis concludes that the plays are polyvalent in meaning and thus open to further academic discussions for the years to come.

Song, Dongzhe. « Cardiovascular function in animal models of metabolic syndrome and type 2 diabetes : the role of inducible nitric oxide synthase (iNOS) ». Thesis, University of British Columbia, 2008. http://hdl.handle.net/2429/822.

Texte intégral
Résumé :
Activation of inducible nitric oxide synthase (iNOS) and oxidative stress have been shown to be associated with compromised cardiovascular function in streptozotocin (STZ)-induced type 1 diabetes. The aim of the project is to investigate cardiovascular abnormalities in a rat model of type 2 diabetes (Zucker diabetes fatty or ZDF rats) and two models of metabolic syndrome (fructose-fed rats and Zucker obese rats), and to provide direct evidence linking iNOS and oxidative stress to abnormal cardiovascular function in these disorders. Blood pressure, cardiac contractility, cardiac index, regional flow, vascular resistance and venous tone were measured in diseased as well as normal rats. Biochemical analyses such as activities of iNOS, immunostaining of iNOS and western-blot analysis of iNOS in the heart tissue were carried out. The results showed that cardiac contractile response to dobutamine was compromised in the ZDF rats, and this was associated with increased myocardial protein expression as well as activity of iNOS. The formation of peroxynitrite was increased in the heart tissue of the ZDF rats. Selective inhibition of iNOS by 1400W (N-3-aminomethyl-benzyl-acetamidine) did not alter responses to dobutamine in the control rats, but augmented the contractile effects of dobutamine in the diabetic rats. The regional blood flow was altered in the ZDF rats, and iNOS played a negligible role in regulating regional flow in the ZDF rats. Although venous response to noradrenaline was also altered in the Zucker obese rats, NOS may not be involved in venous tone regulation. Anti-oxidative treatment with N-acetylcysteine inhibited the development of insulin resistance, blood pressure elevation and the increase of 8-isoprostane formation in the fructose-fed rats. We conclude that heart function is compromised and regional blood flow is altered in the ZDF rats. Activation of iNOS plays an important role in suppressing heart dysfunction but does not affect regional blood flow. In Zucker obese rats with metabolic syndrome, iNOS may not be involved in changes of venous function. Oxidative stress is associated with both abnormality of heart dysfunction in type 2 diabetes (by formation of peroxynitrite due to iNOS activation) and development of hypertension and insulin resistance in metabolic syndrome.

Many infectious agents have been associated with reproductive failure in swine, representing significantly economic losses for production. Recently, Porcine Circovirus Type 2 (PCV2), etiologic agent of PCVAD or PCV2 associated diseases, was associated with reproductive failure in swine around the world. To confirm the pathogenic potential of PCV2 inducing reproductive failure in sows, it s necessary the viral isolation and antigen and nucleic acid demonstration in fetuses. Other viral agent, Torque Teno Vírus (TTV), also have been recently associated with infections caused by PCV2. TTV alone has not showed pathogenic signals in swine, but, its role in co-infections with other pathogens has been investigated. The present study aimed the diagnostic of PCV2 in natural infections where there was reproductive failure, as well as to establish and apply the Polimerase Chain Reaction (PCR) technique for TTV from organs. Samples from field cases, as aborted fetuses, mummified, stillborn, fragile piglets and material from abattoir sows were collected and processed to diagnostic infection in order to detect PCV2 by PCR and immunohistochemistry (IHC). Samples were collected from 21 farms; and a total of 169 fetuses were necropsied. Moreover, reproductive samples from 83 abattoir sows were collected in 4 slaughterhouses of Santa Catarina State. In the present study was possible detect viral DNA by PCR in 29 (17,1%) of 169 analyzed fetuses, where heart and lymphoid tissues showed virus DNA more frequently, 41,4% and 37,8%, respectively. Viral presence was confirmed by IHC in tissues, which detected viral antigens in 17 PCV2 positives fetuses by PCR. Samples of reproductive tissues from sows also were tested by PCR and PCV2 was identified in 4 sows (4,8%). PCR technique aimed to detect TTV was established for viral DNA from organs. Samples of reproductive tissues from sows were tested, and were found both genogroups of TTV (TTV1 and TTV2), in 25 (30,1%) and 41 (49,3%) sows, respectively. Fetuses samples that resulted positive to PCV2 by PCR were also tested to TTV, and it was observed the occurrence of co-infection between these agents. The results obtained here suggest the involvement of PCV2 in reproductive failure in sows, besides show that TTV was present in analyzed samples, corroboring the association with PCV2

Terpenes are a very promising class of skin penetration enhancers especially due to their low potential of irritation in the skin. In this work, we have used the electron paramagnetic resonance (EPR) spectroscopy of the small spin label di-tert-butyl nitroxide (DTBN), which partitions the aqueous and hydrocarbon phases, to study the interaction of the terpenes ®-terpineol, 1,8-cineole, L(-)-carvone and (+)-limonene with the uppermost skin layer, the stratum corneum, and the membrane models of 1,2-dipalmitoyl-sn-glycero- 3-phosphatidylcholine (DPPC) and 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC). The EPR spectra indicated that the terpenes increase both the partition coe±cient and the rotational di®usion rate of the spin labels in the stratum corneum membranes whereas for DMPC and DPPC bilayers were observed similar e®ects only at temperatures below the liquid-crystalline phase. The EPR parameter associated to probe polarity inside the membranes showed thermotropic induced changes, suggesting relocations of spin probe, which were dependent of the membrane phases. While DMPC and DPPC bilayers showed abrupt changes in the partitioning and rotational correlation time parameters at the phase transitions, the SC membranes were characterized by slights changes in whole interval of measured temperatures, presenting the greatest changes or membranes reorganizations in the temperature range of v50 to v74±C. The results suggest that the terpenes act as spacers that weaken the hydrogen-bonded network at the polar interface thus °uidizing the stratum corneum lipids and, in consequence, increase the permeation of small polar molecules across the membranes. 2

Abstract This project drilled in Sin Phu Horm field. The main challenge in this field is the formation. The 8.5-in section is designed to drill through the hard and abrasive sandstone formation (known as Nam Phong formation) with unconfined compressive strength (UCS) between 6,000 and 24,000 psi and peak up to 55,000 psi. Multiple bit runs and heavy set of Polycrystalline Diamond Compact (PDC) bits were observed in the offset wells with slow rate of penetration (ROP) and short intervals, which resulted in a high drilling cost. In the offset runs, the average interval was observed between 200 and 300 meters and average on-bottom ROP ranged from 2 to 8 m/hr. Worn cutters were the main dull characteristic in the offset PDC bits and the bits were pulled out of hole due to slow ROP. Due to the challenging formation, the goal was to increase the interval per bit run and ROP which resulted to reduce the number of bit trips and drilling cost. Looking at the dull grading of the offset PDC bits, it was obvious that the slow ROP was caused by the cutters worn by the abrasive and hard Nam Phong formation. The fixed-cutter PDC bits were run in the offset wells and worn cutters were observed in the shoulder area. The worn portion of the cutter occurred only in the exposed side, while the portion in the cutter pocket remained intact. Utilizing the portion in the cutter pocket helps to prolong cutter life, increase the ROP, and bit life longevity. Thus, it can help to reduce undesired bit trips. Based on the worn cutter observation, the new design of the 8.5-in PDC bit equipped with innovative 360 rolling cutter (RC) bit was proposed. A comprehensive vibration simulation drilling parameters roadmap were provided to minimize shock and vibration. Two bits were run with rotary steerable BHA to drill Nam Phong formation in the field. The first bit drilled 431 meters at an average ROP of 6.8 m/hr and the second bit drilled 391 meters at an average ROP of 5.5 m/hr. Two runs using the 360 RC bits drilled 822 meters in total of 1,236 meters entire interval of Nam Phong formation, which was equivalent to 66%, achieving the operator's goal while saving 2.2 days solely from two runs of RC bit. This success increased the operator's confidence to run 360 RC bits in the subsequent wells to reduce the number of bit trips and increase the ROP. This paper will discuss the application and evolution of 360 RC bit, along with the result achieved by the bit fitted equipped with this cutter in Thailand onshore.

Résumé :
Cathodic protection (CP) is one of the primary methods to protect buried piping and pressure components from corrosion and is a critical element in asset management of buried piping at nuclear power plants. Implementation of cathodic protection requires non-structural attachments to the buried piping for electrical leads and connections. The method of attaching copper-copper alloy CP leads to carbon steel piping and components using traditional arc welding processes can be difficult and time consuming. A two-step process is frequently used where a carbon steel weld tab is first welded to the pipe or component by a traditional arc welding process. The copper-copper alloy CP lead is then joined to the carbon steel weld tab by the exothermic welding process. An alternative to this cumbersome two-step process is pin brazing which is an automatic brazing process that uses electric current resistance to heat the interface between a pin capsule and the component. An arc between the pin capsule and the outside surface of the electrical connector is then used to melt the capsule or pin that contains the brazing filler metal. The process is similar to stud welding in that the brazing pin is loaded into an automatic pistol and the brazement is made when the trigger is pulled. ASME Section XI Code Case N-882 delineates rules and requirements for application of pin brazing on Class 2 and 3 pressure boundary components. This paper provides the background and description of the pin brazing process with a summary of the technical basis for Case N-882.

Résumé :
In recent years, Europe has been moving towards a concept of inclusivity as highlighted by the sixteenth goal of the 2030 Sustainable Development Goals Agenda that promotes peaceful and inclusive societies. (UN Dept. of Global Communications, 2015). The increasing awareness of social diversity has attracted the attention of designers who started to adopt an inclusive design approach and design products or services to be usable by as many people as reasonably possible, without the need for specialised adaptions. The inclusive design approach has been largely applied in adaptive sports to improve levels of functioning and independence in daily living activities and increase physical capability, physiological capacity, social status, and sense of belonging. Adaptive sports can become a way to promote involvement as an active part of the rehabilitation exercise to stimulate neuromotor recovery, particularly in children with disabilities (Canina et al., 2020). Recent research has demonstrated that climbing could be an excellent rehabilitation tool that involves the child with disabilities in a natural way. This sport exploits the propensity to play, to sport, to compete, to stimulate the execution of specific exercises, can transform this effort into a game and multiply the effectiveness of the rehabilitation process (Reljin, V., 2019). An intensive rehabilitation from an early age guarantees the recovery of part of their neuromotor abilities. In order to achieve better results in rehabilitation, adaptive sports must adopt a holistic approach to the user considering both the physical and the psycho-perceptual aspects, i.e. the ability to do it but also the feeling of fulfilment in doing it. However, current climbing walls do not include these aspects of the adaptive sport. An adaptive climbing wall design requires identifying a methodology that could lead to a coherent and effective solution, using explicit attention for inclusiveness. The paper describes the Design for Sustainable Behaviour (DfSB) approach adopted to design an adaptive climbing wall as a tool for the rehabilitation of children with Cerebral Palsy (CP) by identifying the sustainable, inclusive requirements that consider children’s diversity. The DfSb approach, as user- and use-centred design that create preconditions for a sustainable everyday life, considers the sustainability aspects from two essential points of view. The user's sustainable behaviour, in which inclusiveness is a fundamental part of these attitudes, and the product's sustainability that uses new recycled materials create a more natural environment (similar to climbing in natural environments). Indeed, the project considers first the sustainable behavioural aspects, spreading climbing as a tool to improve the health conditions of CP children, introducing them to climbing by making it accessible and inclusive, intending to help children with different abilities to build trust and awareness of their potentialities, and a sense of accomplishment while training problem-solving and decision-making skills. As a second point of DfSB, the climbing wall and holds are designed with sustainable materials (waste material content) that provide the feeling of natural stone considering the entire product lifecycle. This paper shows how the DfSB approach can support the definition of design requirements of a training tool introducing children with CP to climbing as a natural approach to rehabilitation, making it accessible and inclusive. The project brings children with disabilities closer to the adapted sport through an indoor and democratic recreational activity. Bibliography 1. AA.VV. (2020). What is inclusive design?
