Основываясь на проведенных ранее исследованиях, были использованы данные, полученные в процессе сбора и анализа статистических данных. Определены внешние и внутренние факторы, влияющие на систему. Рассмотрены методы прогнозирования, оптимально подходящие для решения задач, стоящих перед предприятием лакокрасочной отрасли. С помощью метода анализа иерархий был проведен анализ альтернативных методов прогнозирования. На основании выводов, полученных в результате анализа, были выбраны нейросетевые модели. В статье рассмотрены результаты проведенной серии экспериментов по прогнозированию выпуска продукта НЦ-132 (Выбор мастера) с помощью нейросетевых моделей. По результатам экспериментов предложена схема выбора нейросетевой модели и алгоритм ее предварительного обучения. Приведены результаты проведенных экспериментов, на основании которых в дальнейшем планируется описание функционала модуля прогнозирования, разработки технического задания и внедрения разработанной системы.

RESUMEN:El objetivo del artículo es aproximarnos a la controvertida biografía del comisario Manuel Brabo Portillo. El trabajo está basado en fuentes primarias y secundarias. El método utilizado es empírico. En el imaginario del mundo sindicalista revolucionario, Brabo Portillo era el policía más odiado, la reencarnación de la cara más turbia del Estado. Fue, así mismo, un espía alemán relacionado con el hundimiento de barcos españoles, el asesinato del empresario e ingeniero Barret y el primer jefe de los terroristas vinculados a la patronal barcelonesa. La conflictividad que afectó a España en el período de la Primera Guerra Mundial es fundamental para entender los orígenes del terrorismo vinculado al pistolerismo, que marcó la historia político social española del primer tercio del siglo XX.PALABRAS CLAVE: Brabo Portillo, pistolerismo, espionaje, sindicalismo, Primera Guerra Mundial.ABSTRACT:The objective of the article is an approach to the controversial biography of Police Chief Manuel Brabo Portillo. The work is based on primary and secondary sources. The method used is empirical. In the imagery of the revolutionary syndicalist world, Brabo Portillo was the most hated policeman, the reincarnation of the murkiest face of the state. Two excavations within the medieval burgh provided the opportunity to investigate an early industrial zone on the western side of Meal Vennel and a relatively rich undeveloped area in the backlands of Scott Street. The artefact rich Meal Vennel excavation revealed evidence of a possible early western boundary of the town, timber and stone buildings and industrial activities including grain drying and iron smithing. Backland activities including sand quarrying, the digging of rubbish pits and dumping of midden, were investigated at Scott Street. A number of questions relating to the medieval development of Perth were addressed. `Excavations at Meal Vennel' by Richard Sermon & Adrian Cox (736--51), including: `The pottery' by Peter Cheer (753--9); a gold and sapphire ring and the copper alloy objects including dress fasteners/fittings and keys, a mount and fittings, needles, padlock components, pendants, pins, rivets, seal matrix, sheets and strips, thimbles and tweezers; lead alloy objects; iron objects including arrowheads, buckles, horse equipment, household ironwork, and knives are detailed along with a `Metallurgy of the knives' by Paul Harrison (776--7), locks and keys, structural ironwork, and tools; stone objects; bone and antler objects including perforated bones and horn cores, horn-working evidence, pins, antler offcuts; the glass including a bead, linen smoother(?) and window glass; `Coins and a jetton' by Nicholas Holmes (789--90); `The flint' by Jill Harden (790 & 791); ceramic roof tiles; industrial residues; `The leather' by Clare Thomas (792); `The animal bone' by Catherine Smith (792--4); and `Botanical remains' by Alan Fairweather (795). `Excavations at Scott Street' by Jonathan Burrows (795--801) is followed by the specialist reports from that site including: `The pottery' by Peter Cheer (801--4); copper alloy objects; lead alloy objects; iron objects; stone objects; the wooden cask; `Textiles' by Thea Gabra-Sanders (810--11); ceramic floor tiles; glass; clay pipes; `Coins' by Nicholas Holmes (812); `Mammal and bird bone' by Catherine Smith (812--13); `Botanical remains' by Alan Fairweather (813--14).

Background: Two billion U.S. healthcare dollars are spent on youth sports injuries each year. This staggering figure is projected to increase as athletes are sustaining more sport-related injuries and are specializing in a single sport at younger ages than ever before. Sport specialization is a modifiable injury risk factor unique to youth athlete populations. Parents are a group of key stakeholders whose knowledge and beliefs likely impact youth sport participation, however a paucity of research exists in this area. Purpose: To assess the differences in perceptions of parents on youth sport participation and specialization based on geographical region. Methods: Parents of children, ages 8-18 years, who participate in organized sports were surveyed for this study. Electronic surveys with a total of 40 questions were circulated via team/organization mailing lists and social media. Geographic regions were described as either South or North based on divisions from the U.S. Census Bureau. Sport specialization was defined as an athlete meeting >2 of the following 3 criteria: participates in one sport to the exclusion of other sports, participates in sport-specific lessons, participates on >2 teams in a single sport. Descriptive statistics and chi-square analyses were used to compare perceptions of parents with specialized and non-specialized athletes across different geographical regions. Results: Three-hundred and seventy-one responses were collected (South=204, North=167). Parents from the South reported a higher number of specialized athletes as compared to parents from the North (54% vs 38%; P=0.003). Regardless of geographical region, most parents did not think specialization had a positive impact on a youth athlete’s development ( P=0.307) or their future in that sport ( P=0.086) (Figures 1 & 2). Parents from the South did select ‘Potential to earn a college scholarship’ as a motivator for youth sport specialization more often when compared to parents from the North (30% vs 17%; P=0.002). Conclusion: Regional differences exist in the percentage of specialized youth athletes participating in organized sport. Parental knowledge of the risks and benefits of youth sport specialization does not appear to drive these observed differences. Earning a college scholarship was selected more frequently as a reason for specialization in the South, indicating that societal or socioeconomic factors may be present. Fifteen 7,4’- O-modified genistein amino acid derivatives are synthesized through nucleophilic substitution and hydrolysis, followed by condensation with diverse amino acid esters. The antiproliferative activity of all the synthesized compounds is evaluated against three cancer cell lines (MGC-803, HeLa, HCT-116) and one normal cell line (HUVEC) using 5-fluorouracil (5-Fu) as the positive control. The results show that methyl [2-({5-hydroxy-3-[4-(2-{[3-(4-hydroxyphenyl)-1-methoxy-1-oxopropan-2-yl]amino}-2-oxoethoxy)phenyl]-4-oxo-4 H-chromen-7-yl}oxy)acetyl] tyrosinate exhibits significant antiproliferative activity against the MGC-803 cell line with an IC50 value of 8.52 µM, and its inhibitory effects on HeLa and HCT-116 cancer cells are stronger than that of the positive control drug 5-Fu.

Wild-type and mutant chicken integrin beta 1 subunit (beta 1c) cDNAs were expressed in NIH 3T3 cells and assayed for localization in focal adhesions of cells plated on fibronectin substrates. Focal adhesion localization in stable transfected cells was assayed by indirect immunofluorescent staining with chicken-specific anti-beta 1c antibodies. Mutant beta 1c integrins containing internal deletions of 13 amino acids adjacent to the membrane, delta 759-771, and 20 centrally located amino acids, delta 771-790, localized in focal adhesions demonstrating that sequences required for direction to focal adhesion structures were not limited to one region of the cytoplasmic domain. Point mutations revealed three clusters of amino acids which contribute to localization in focal adhesions. These three clusters or signals are: cyto-1 (764-774), cyto-2 (785-788), and cyto-3 (797-800). The 11-residue cyto-1 signal is only found on integrin beta subunit sequences, except beta 4. Four residues within this region, D764, F768, F771, and E774, could not be altered without reducing focal adhesion staining intensities, and likely form a signal that occupies one side of an alpha helix. Mutations involving two cyto-1 residues, K770 and F771, also appeared to affect heterodimer affinity and specificity. Cyto-2 (785-788,), NPIY, is an NPXY signal that forms a tight turn motif. Cyto-2 provides a structural conformation, which when perturbed by proline removal or addition, inhibits integrin localization in focal adhesions. Cyto-3 (797-800), NPKY, resembles cyto-2, however, the nonconserved proline residue can be replaced without alteration of the localization phenotype. Cyto-3, therefore, constitutes a unique integrin signal, NXXY. Both serine and tyrosine residues at positions 790 and 788, respectively, which have been implicated in integrin phosphorylation/regulation, were conservatively replaced without detectable effect on focal adhesion localization. However, acidic replacements for these amino acids reduced focal adhesion staining intensities, suggesting that phosphorylation at these sites may negatively regulate integrin function.

Children as young as five have some ability to produce contrastive focus [Wells et al. (2004) J. Child Lang. 31, 749–778]. However, adult listeners' ability to identify the location of contrastive focus depends on whether the speech came from a 4-, 7-, or 11-year-old [Patel and Brayton (2009) J. Speech. Lang. Hear. Res. 52, 790–801]. Recent acoustic studies have also reported the use of F0 vs duration in contrastive focus productions by American English–speaking 2-year-olds [Thorson and Morgan (2021) J. Child Lang. 48, 541–568] and 4-year-olds [Wonnacott and Watson (2008) Cognition 107, 1093–1101], respectively. This study, therefore, evaluated the extent to which older 6-year-olds, with more language experience, used F0 and/or duration when producing contrastive focus, and compared this to adult speech. Monosyllabic and disyllabic adjective + noun targets (e.g., green ball) in utterance medial and final position were elicited from 20 Australian English–speaking 6-year-olds and 14 adults in adjective focus and noun focus conditions. Although both adults and children used high F0, only adults consistently used word and stressed syllable duration as well. This suggests that children may master the different acoustic cues to contrastive focus at different stages of development, with late cue integration.

Solid solutions with different compositions, mNH4Cl·KCl and NH4Cl·nKCl (m，n≥1), were prepared by the constant speed cooling method. The thermal stability of the solid solutions was determined by TG-DTA. The results show that mNH4Cl·KCl crystals change its crystal form at 180°C, decompose into ammonia and hydrogen chloride gas at 330°C, melt at 790°C, and finally evaporate at around 820°C. Its evaporation temperature increases with the increasing of potassium chloride content in the samples. The Solid solution of NH4Cl·nKCl decomposes to ammonia and hydrogen chloride gas at 230°C, melts at 790°C, and evaporates at 800°C. The continuous specific heat curve of the solid solution (NH4, K)Cl were also determined. These data provide a theoretical basis for further research on solid solution properties.

Metode yang digunakan adalah overlay dan pembobotan menggunakan 4 parameter kerentanan longsor. Dari hasil penggabungan data-data yang diperoleh maka di Kecamatan Cikajang menunjukan rawan longsor ada tiga titik longsor, di Kecamatan Banjarwangi menunjukan kurang rawan longsor tidak ada kejadian titik longsor, di Kecamatan Cihurip menunjukan kurang rawan longsor ada satu titik longsor, di Kecamatan Cisompet menunjukan sangat rawan ada tujuh titik longsor dan di Kecamatan Pameungpeuk menunjukan kurang rawan tidak ada titik longsor.

Trade and growth theories generally predict a positive relationship between openness to international trade and economic growth. There are a number of channels through which openness is thought to influence economic growth. First, a liberal trade regime enhances efficiency through greater competition and improved resource allocation. Second, greater access to world markets allows economies to overcome size limitations and benefit from economies of scale. Third, imports of capital and intermediate goods can contribute to the growth process by enlarging the productive capacity of the economy. Fourth, trade can lead to productivity gains through international diffusion and adoption of new technologies. Empirical studies on the relationship between openness and economic growth have largely supported the view that openness has a favourable impact on economic growth. It is not surprising, then, that the proposition that more open economies tend to grow faster has gained wide acceptance in academic as well as policy circles. The objective of this paper is to examine the relationship between openness and economic growth in the context of Pakistan’s economy. Section 2 reviews the literature on openness and economic growth. Section 3 provides an overview of trade liberalisation in Pakistan. Data and methodology are described in Section 4, while Section 5 presents the empirical results. Section 6 concludes the discussion.

Wheat plays a leading role among cultivated crops. However, some anatomical features of the perennial wheat leaf blade structure, cell development, and metameric stem segmentation remain poorly understood. The object of the present study was six varieties of perennial wheat cultivated in the Almaty region of Kazakhstan. During the study, metameric features of the growth and development of the perennial wheat stem internodes were analyzed. The stems consisted of four internodes and very rarely of five. The variety No. 701 had the longest stem (119 ± 4 cm), while the variety Sova had the shortest one (106 ± 4 cm). The variety No. 701 also had the largest leaf blade with a length of 42 cm ± 2 cm and a width of 1.6 ± 0.09 cm, while the variety No. 704 had the smallest leaf blade with a length of 27 ± 1 cm and a width of 1.2 ± 0.06 cm. The average biological productivity of the Sova variety was 10.49 centners per hectare. The varieties No. 703, No. 704, and No. 801 demonstrated high productivity ranging from 26.08 to 28.8 centners per hectare.

Tanah longsor merupakan salah satu bencana yang sering terjadi di Indonesia. Longsor dapat terjadi karena adanya beberapa faktor utama. Salah satu faktor tersbut adalah curah hujan yang tinggi dan kelembaban tanah. Selain itu ada beberapa faktor yang dapat diperhitungkan yang dapat menyebabkan bencana longsor. Dibutuhkan suatu sistem yang dapat mengakuisisi data data dilapangan agar dapat memprediksi terjadinya bencana longsor. Pada penelitian ini dibuatlah perancangan 3 buah prorotipe akusisi data dengan 4 parameter penyebab terjadinya tanah longsor. 4 parameter itu berupa kemiringan lereng yang dapat diukur dengan sensor MPU 6050, curah hujan yang diukur dengan sensor tipping bucket dengan reed switch, kadar air dengan 2 sensor kelembaban FC-28, dan getaran tanah yang diukur dengan sensor 801s. Sistem ini pun dilengkapi dengan proses pengambilan data ataupun log data secara realtime dengan SD Card dalam proses secara langsung dan pengiriman via server mealalui SIM900. Untuk indikasi alarm digunakan SX1278. Perancangan sistem ini berdasarkan hasil pembacaan 4 parameter ynag diukur. Dari hasil pengujian sensor didapatkan hasil pembacaan nilai setiap masing masing sensor dengan error 0.165 untuk sensor MPU6050, 0 untuk sensor tipping bucket, 0 pada sensor 801S, dan 0.39 dan 0.710 untuk 2 sensor kelembaban FC-28.

IntroductionAccelerate aging is a worldwide problem. In underdeveloped countries, it is occurring without adequate planning and is attributed due to falling mortality and fertility, which contributed to increase life expectancy. However, aging was not accompanied by improvement in living conditions and health assistance. Poor sanitation and diet, low incomes and education levels contributed to increasing the prevalence of chronic diseases in elderly.ObjectivesData obtained in other countries could not represent the national population; objective was to describe a Brazilian frailty elderly sample.AimsDetermine cognitive impairment prevalence in Brazilian frailty elderly.Methods1952 frailty elderly were analyzed. Social data, tobacco and alcohol consumption were verified and mini-mental state examination was applied from February to August in 2010.ResultsFemale elderly was predominant (68.8%). They were almost married (47.5%), followed by widow (37%), single (8.1%), divorced (7.4%). White people were predominant (69.2%); black (14.6%) and brown (16.2%) were presented at the same proportion. 823 frailty elderly (64.9%) studied until 4 years; 7.7% studied between 4–8 years; 1.9% went to college and 25.6% were illiterate. Only 10.6% were smokers and 2.8% drank alcohol. The majority (79.4%) were younger than 80 years-old. Cognitive impairment prevalence were 17.2% in illiterate; 15.2% in elderly who studied until 4 years and lower, 5.3%, in the group who studied between 4–8 years.ConclusionsThe prevalence of cognitive impairment was dependent on years of study and we presented data obtained in a Brazilian sample. These data can be used to compare cognitive impairment in different countries.

The article is devoted to the study of the opposition «Own - Alien», which is significant in intercultural communication, and the peculiarities of its implementation. «Own - Alien» is presented as a universal basic concept of culture and linguistic culture. The article presents the results of the analysis of the process of conceptualization of the opposition «Own - Alien» based on the material of proverbs and phraseological units of the Chinese language containing the terms of kinship «father / mother». In the course of the study, chengyu were selected and further divided into semantic modules and micromodules. The relevance of the article is determined by the need to achieve mutual understanding between representatives of Russian and Chinese cultures, as well as the need to analyze both general and national-specific patterns of the implementation of the opposition «Own - Alien» in Chinese chengyu, which convey information about the mentality of the nation. The object of the study was the mechanisms of language actualization of the opposition «Own - Alien». The subject of the proposed study is a system of semantic modules that actualize the opposition «Own - Alien» through the terms of kinship «father / mother». The purpose of the work is to develop a classification of the conceptual fields «one's own» and «alien». The study was carried out on base of the material of Chinese chengyu dictionaries.

The research is devoted to the analysis of the current state of chemical crop protection market in Russia. Significant industry growth has occurred in last decade due to the attractiveness of agriculture as an export-oriented industry. Russian agricultural products are in stable demand all over the world. Countries in the Middle East, South-Eastern Asia and Africa with constantly growing population rely on foodstuff import, including from the Russian Federation. Thus, country makes a significant contribution in a global food security. However, in order to develop and increase competitiveness, Russian farmers need to intensify crop production. One of technological improvement elements, which let growers to get sufficient yield and quality, is application of innovative chemical crop protection products. Pesticides help farmers struggle with crop diseases, pests and weeds, as well as minimize adverse climate factors in some cases. Agrochemicals market in Russia is represented not only by foreign players, but by domestic manufacturers as well. Market share of these two groups in value terms of market players is approximately equal. However, it’s worth mentioning, that like in many other industries, this sector is heavily dependent on import inputs. Major part of components for local pesticide production are imported and key technological process is constrained as a type of mixing necessary inputs in proper way. Domestic business, together with Russian Government and, probably, in cooperation with some international players, needs to solve the problem of deepening the production cycle, look for stimulus for development of the entire production chain in the long-term perspective, including development of fundamental science in chemical crop protection industry. Learning and adapting foreign experience can be the first step in the process of structural change of the crop protection industry. Simultaneously, it is critical to maintain openness and desire to use best practices at pesticide market, without which the competitiveness of Russian agricultural production will be constrained and development of the entire industry will have a pace inferior to other countries.

The largest video hosting service in the world is currently the YouTube platform. Having originated in the USA in 2005 the video hosting service very quickly expanded the scope of its geographical presence due to the ability to use the service in different countries. YouTube's monetization was based on an advertising model from the very beginning. The YouTube Premium service created inside the platform (previously called YouTube Red) allows you to watch content without ads by subscription. But the content model is not developing so rapidly and main financial flows continue to come from advertising. That is why it is worth understanding the possibilities of advertising on YouTube. The research identified integrated advertising on YouTube blogs which were selected according to various criteria: total number of subscribers, broadcast language, regular content publications (at least 1 publication per quarter), genre, ranking position and number of views. Advertisements were found present in all analyzed blogs while the types of advertising varied depending on the genre of the blog. The time a blogger spent on advertising also largely depended on the genre characteristics of the blog. Currently, one of the most popular types of advertising is self-promotion, since it is directly related to third-party bloggers' earnings. In the current research we analyzed advertising in the blogosphere based on monitoring several diverse thematic blogs and identified most common forms of advertising. The attempt to classify advertising opportunities in the video hosting user content seems important to us because it allows us to streamline the terminology and in the future develop a system of interaction between channels and advertisers.

Language, which is a constantly changing and developing system, is part of society. In recent time, processes such as feminization and neutralization are taking place, which are reflected in the names of professions. The authors of the article give the results of the analysis of processes, based on the theoretical basis formed by domestic and foreign researchers. These processes can be traced in the advertisements for hiring personnel. In the article, the authors distinguish four types of job advertisements in which the indicated processes are observed, presented in full or in part: advertisements using gender-neutral words (partial neutralization); announcements using gender-neutral words and evading gender agreement (complete neutralization); announcements using gender-specific job titles (partial feminization); ads using gender-specific job titles, and inclusive spelling (complete feminization). In conclusion, it is concluded that these processes fully reflect gender relations in society, they are of an evolutionary nature, and further prospects for future study of these processes are outlined.

The article offers an analysis of the creation of the image of the PRC in the context of the One Belt — One Road initiative in the Western mass media (on the example of the British news channel BBC News). The author of the study proceeds from the thesis about their important role in shaping and promoting the image of the country. In the context of information globalization, the role that the media play in international relations is multiplied many times over. A special place in such processes is occupied by global news channels, which are often of decisive importance in constructing world public opinion, setting the agenda and assessing the importance of ongoing or planned events. The Belt and Road Initiative is not only an example of global cooperation, but also a reflection of a new approach to development in the modern world as a whole, which has a serious impact on China's international prestige and image. In the course of the study, we find out that the Western mass media more often perceive the One Belt, One Road initiative in a negative aspect, ignoring the real achievements and successes of this global project. On the example of the analysis of news releases of the British television channel BBC News, it was revealed that, despite external neutrality, its editorial staff uses different methods and technologies to form a negative image of China. The preference of the editors of the TV channel to economic and political topics, specific events and positions of states as an information occasion was revealed. With the external prevailing neutral position of the TV channel towards One Belt, One Road, its real achievements and successes are ignored, the negative assessment of the Western expert community is covered, and lexemes with a negative meaning are often used.

Необходимым требованием при выполнении столь важной функции была тщательность рассмотрения министерских проектов вплоть до прогнозирования возможных последствий их применения. В ряде случаев выполнение этой задачи было невозможно без привлечения «сведущих людей» – экспертов. В статье на основании анализа широкого круга источников проанализировано содержание понятия «эксперты» во второй половине XIX в., отношение к ним в Государственном совете, значение их участия в его заседаниях. Автор приходит к выводу, что, несмотря на усложнение стоявших перед Советом задач, отношение к институту экспертов не менялось и их потенциал не был использован в полной мере.

Spiking neural networks (SNN) increasingly attract attention for their similarity to the biological neural system. Hardware implementation of spiking neural networks, however, remains a great challenge due to their excessive complexity and circuit size. This work introduces a novel optimization method for hardware friendly SNN architecture based on a modified rate coding scheme called Binary Streamed Rate Coding (BSRC). BSRC combines the features of both rate and temporal coding. In addition, by employing a built-in randomizer, the BSRC SNN model provides a higher accuracy and faster training. We also present SNN optimization methods including structure optimization and weight quantization. Extensive evaluations with MNIST SNNs demonstrate that the structure optimization of SNN (81-30-20-10) provides 183.19 times reduction in hardware compared with SNN (784-800-10), while providing an accuracy of 95.25%, a small loss compared with 98.89% and 98.93% reported in the previous works. Our weight quantization reduces 32-bit weights to 4-bit integers leading to further hardware reduction of 4 times with only 0.56% accuracy loss. Overall, the SNN model (81-30-20-10) optimized by our method shrinks the SNN’s circuit area from 3089.49 mm2 for SNN (784-800-10) to 4.04 mm2—a reduction of 765 times.

BackgroundDespite clinical successes of immune checkpoint blockade (ICB) antibodies blocking the inhibitory receptors CTLA-4, PD-1, or PD-L1, substantial challenges remain. Many patients do not respond, and ICB treatment is associated with serious immune-related adverse effects (irAEs) which are exacerbated by combination therapies. TIGIT blockade has been demonstrated to provide tumor control in pre-clinical studies, sparking ongoing clinical trials, including those targeting TIGIT in combination with anti-PD-1 or anti-PD-L1. The INTASYL™ platform is a self-delivering RNAi technology that (1) provides efficient delivery into target cells bypassing the need for specialized formulations, mechanical perturbation, or drug delivery systems; and (2) specifically and durably silence target gene expression when administered intratumorally (IT), providing in vivo tumor control. IT administration restricts pharmacokinetics to the tumor; an attractive strategy for mitigating ICB-mediated systemic irAEs. Additionally, using INTASYL, multiple targets can be silenced in combination. Here we demonstrate the in vivo efficacy of INTASYL specifically targeting TIGIT (PH-804), PD-1 (PH-762), PD-L1 (PH-790) alone or in combination in a CT26 model of murine colorectal carcinoma.MethodsTo assess silencing activity, activated human pan-T cells were incubated in vitro with INTASYL compounds either alone or in combination and mRNA silencing was determined by qRT-PCR and protein silencing by flow cytometry. To assess in vivo tumor efficacy CT-26 cells were implanted subcutaneously into BALB/c mice. INTASYL compounds were administered IT at 1 mg/dose on Days 1, 3, 7, and 10 either as single agents (mPH-804, mPH-762, mPH-790) or in combination (mPH-804 + mPH-762 or mPH-804 + mPH-790). Controls consisted of PBS (vehicle; (IT)), and anti-TIGIT, anti-PD-1, or anti-PD-L1 antibodies (0.2 mg/dose) administered via intraperitoneal injection (IP). Tumor volumes and body weight were recorded throughout the study. Tumors were taken at the end of the study for analysis.ResultsSingle and combination knockdown of target molecules was validated at the mRNA level (=90%) by qRT-PCR and at the protein level (=80%) in activated human pan-T cells. In vivo, combination treatment with mPH-804 + mPH-762 or mPH-790 improved tumor control compared to individual monotherapies providing evidence of potential synergy. All treatments were well tolerated.Conclusionsn/aAcknowledgementsWe demonstrate the potential of INTASYL-mediated combination therapy targeting TIGIT and PD-1/PD-L1. These findings indicate that combination of TIGIT + PD-1/PD-L1 silencing improves tumor control compared to monotherapy. As INTASYL IT is efficacious and may mitigate irAEs caused by antibody ICB, INTASYL combination therapies including PH-804, PH-762 and PH-790 warrant further investigation in patients.

803 Background: Ripretinib is a switch-control tyrosine kinase inhibitor approved for the fourth-line therapy for GIST. In the INTRIGUE study (NCT03673501), ripretinib as second-line (2L) treatment demonstrated similar progression-free survival (PFS) and fewer Grade 3/4 treatment-emergent adverse events (TEAEs) compared to sunitinib in the overall population. It showed a numerically longer PFS and a nominally higher objective response rate (ORR) than sunitinib in patients (pts) with KIT exon 11 mutations. This study aims to evaluate the efficacy and safety of ripretinib vs sunitinib as 2L treatment in GIST pts in China. Methods: This multicenter, randomized, open-label phase 2 study (NCT04633122) enrolled adults with GIST who progressed on or had intolerance to imatinib. Pts were randomized 1:1 to ripretinib 150 mg once daily (QD) or sunitinib 50 mg QD (4 weeks on/2 weeks off) in 42-day cycles. Randomization was stratified by KIT mutational status. The primary endpoint was PFS by independent radiologic review (IRR) using modified RECIST version 1.1. Secondary endpoints included ORR by IRR, overall survival (OS) and safety. Efficacy analysis was performed in all-patients intent-to-treat (AP ITT) and KIT exon 11 mutation intent-to-treat (Ex11 ITT) populations. This study was designed to bridge the global trial (INTRIGUE) to show consistency in the efficacy of ripretinib between the two studies. No statistical testing was pre-specified and nominal p-values were presented for descriptive purpose. Results: Overall, 108 pts were randomized to ripretinib (AP ITT n= 54; Ex11 ITT n=35) or sunitinib (AP ITT n= 54; Ex11 ITT n=35). Median age was 59.0 years (range 25–82); 63.9% of pts were male and 57.4% had ECOG PS≥1. By primary data cut-off (20 Jul 2022), PFS by IRR was similar between ripretinib and sunitinib in AP ITT population (HR 0.99, 95% CI 0.57, 1.69; p=0.92; median PFS [mPFS] 10.3 vs 8.3 months). ORR by IRR was numerically higher for ripretinib than sunitinib (29.6% vs 20.4%). Median OS was not reached in either arm. In Ex11 ITT population, a longer PFS by IRR (HR 0.46, 95% CI 0.23, 0.92; p=0.03; mPFS not reached for ripretinib and 4.9 months for sunitinib) and a numerically higher ORR by IRR (37.1% vs 22.9%) were observed for ripretinib vs sunitinib. Fewer Grade 3/4 treatment-related TEAEs were reported with ripretinib than with sunitinib (16.7% vs 55.6%), as were treatment-related TEAEs leading to dose interruption (7.4% vs 42.6%), dose reduction (20.4% vs 29.6%) and treatment discontinuation (1.9% vs 7.4%). Conclusions: Similar to the INTRIGUE trial, ripretinib demonstrated comparable overall efficacy and favorable safety vs sunitinib as 2L therapy in Chinese pts with advanced GIST. 2L GIST pts harboring a KIT exon 11 mutation may benefit from ripretinib vs sunitinib. 8014 Background: Maintenance monotherapy has been associated with improved survival for NSCLC pts. AVAPERL was designed to evaluate the safety and efficacy of bevacizumab with or without pemetrexed as continuation maintenance treatment and demonstrated improved progression-free survival (PFS) (Barlesi, JCO in press). Methods: Pts with advanced nsNSCLC received first-line Bev (7.5 mg/kg), cisplatin (75 mg/m2), and Pem (500 mg/m2) every 3 weeks (q3w) for 4 cycles. Those non progressing were randomized to maintenance Bev (7.5 mg/kg) +/-Pem (500 mg/m2) q3w until progressive disease or unacceptable toxicity. The primary end point (PFS) was met. An independent update analysis has been conducted to assess OS. Results: A total of 376 pts receive induction treatment; 125 and 128 were randomized to the Bev-alone and Bev+Pem arms, respectively. Induction therapy resulted in confirmed disease control in 71.9% of pts. After a median follow-up of 14.8 months, PFS for the Bev+Pem arm was significantly improved both from induction (10.2 v 6.6 m, HR 0.58 [0.45-0.76], p<.0001) and randomization (7.4 v 3.7 m, HR 0.57 [0.44-0.75], p<.0001). With 58% of events, OS for the Bev+Pem arm was also improved both from induction (19.8 v 15.9 m, HR 0.88 [0.64-1.22], p=.32) and randomization (17.1 v 13.2 m, HR 0.87 [0.63-1.21], p=.29). The PFS and OS improvements were comparable across age, PS, smoking status, and response to induction (SD vPR/CR) subgroups. No new safety signal was observed during this updated analysis. Conclusions: Continuation maintenance with Bev+Pem in an unselected population of nsNSCLC pts who had achieved disease control after induction was associated with an increase by almost 4 months in OS benefit over standard Bev alone. 8013 Background: Outcomes for younger patients (pts) with R/R cHL are poor, particularly for those without complete metabolic response (CMR) before autologous transplant (auto-HCT). Nivolumab + BV has shown 67% CMR and a high 2-y PFS rate as first salvage in adults with R/R cHL. CheckMate 744 (NCT02927769) is an ongoing phase 2 study for CAYA with R/R cHL, evaluating a risk-stratified, response-adapted approach using nivolumab + BV and, for pts without CMR, BV + bendamustine. In the initial analysis of the standard-risk cohort (R2), the regimen was well tolerated with high CMR rates before consolidation with high-dose chemotherapy plus auto-HCT. We report data from the primary analysis. Methods: Pts were aged 5–30 y and had first-line treatment (tx) without auto-HCT. Risk stratification has been described previously (Harker-Murray, ASH 2018). Pts received 4 induction cycles of nivolumab + BV; pts without CMR by blinded independent central review (BICR) received BV + bendamustine intensification. Pts with CMR at any time could proceed to consolidation off study. Response was per Lugano 2014 criteria. Primary endpoint: CMR rate (Deauville ≤3) per BICR any time before consolidation. Results: At database lock, 44 pts were treated in R2 (median follow up: 20.9 mo); 43 received 4 induction cycles and 11 received intensification. Median age was 16 y (range 9–30); 24 (55%) pts had primary refractory cHL and 20 had relapsed cHL. CMR rates and ORR any time before consolidation and after induction are shown in Table. 1-y PFS rate by BICR was 91% (90% CI 77–96). During induction, 8 (18%) pts experienced grade (G) 3–4 tx-related adverse events (TRAEs); the most common any grade TRAEs were nausea and hypersensitivity (20% each). 1 TRAE led to discontinuation (G3 anaphylaxis). Most tx-related immune-mediated AEs were G1–2 (1 pt had 2 G3 infusion-related reactions). Conclusions: This risk-stratified, response-adapted approach offers a well-tolerated salvage strategy with high CMR rates and no new safety signals for CAYA with R/R cHL. Most pts avoided alkylator exposure prior to consolidation. Further follow up may confirm durability of disease control. Clinical trial information: NCT02927769 . [Table: see text]

Nuevos artículos consideran que aproximadamente 30 000 muertes anuales se deben a trastornos hipertensivos como preeclampsia, eclampsia y síndrome HELLP(1,2). En los Estados Unidos, los trastornos relacionados con la hipertensión representan aproximadamente 7,4% de las casi 800 muertes asociadas al embarazo que ocurren cada año, con importante disminución de esta tasa de 17,6% en 1990(3). En el Perú, esta probabilidad es aproximadamente 32%(4). Las mujeres con preeclampsia o eclampsia tienen 3 a 25 veces el riesgo de complicaciones graves del embarazo incluyendo desprendimiento prematuro de placenta, coagulación intravascular diseminada, insuficiencia renal, edema pulmonar y neumonía por aspiración(5,6).

Abstract Abstract 67 Dose optimized imatinib (IM) at doses of 400– 800mg has been shown to induce faster and deeper cytogenetic and molecular – responses than standard IM (400mg/day). Since complete molecular remission (CMR 4.5) identifies a subgroup of patients who may stay in remission even after discontinuation of treatment, it was of interest to analyse whether CMR 4.5 is reached faster with dose optimized IM and whether CMR 4.5 correlates with survival. CMR 4 and CMR 4.5 are defined as ≤ 0.01% BCR-ABL IS or ≥ 4. log reduction and ≤ 0.0032% BCR-ABL IS or ≥ 4.5 log reduction, respectively, from IRIS baseline as determined by real-time PCR. CML-Study IV is a five arm randomized study of IM 400 mg vs IM 400 mg + IFN vs. IM 400 mg + Ara C vs. IM after IFN failure vs. IM 800 mg. In the IM 800 arm, a 6 weeks run in period at IM 400 mg was followed by a dose increase to 800 mg and then by a dose reduction according to tolerability. Grade 3 or 4 adverse effects (AE) were to be avoided. From July 2002 to March 2012 a total of 1551 patients with newly diagnosed chronic phase CML were randomized of whom 1525 were evaluable. Median age was 52 years, 88% were EUTOS low risk, 12% high risk, 36% were Euro score low risk, 52% intermediate and 12% high risk, 38% were Sokal low risk, 38% intermediate and 24% high risk. 113 patients were transplanted, 246 received 2nd generation TKI. 152 patients have died, 90 of CML or unknown reasons, 62 of not directly CML-related causes. After a median observation time of 67,5 months 6 years OS was 88.2% and PFS 85.6%. CCR, MMR, CMR 4 and CMR 4,5 were achieved significantly faster with dose optimized IM (400 – 800 mg). No significant differences in remission rates were observed between IM 400 mg and the combination arms IM 400 mg + IFN and IM 400 mg + Ara C, whereas IM after IFN failure thus far yielded significantly slower response rates. After 4 years CCR rates were for IM 400, IM 400 + IFN, IM 400 + Ara C, IM 400 after IFN, and IM 800, 80%, 75%, 73%, 59% and 80%, respectively, MMR rates 84%, 77%, 82%, 61% and 88%, CMR 4 rates 57%, 55%, 55%, 40% and 65%, and CMR 4.5 rates 40%,42%, 42%, 28% and 52%, respectively. CMR 4 was reached after a median of 27 months with IM 800 and 41.5 months with IM 400. CMR 4.5 was reached after a median of 41.5 months with IM 800 and 63 months with IM 400. EUTOS low risk patients reached all remissions faster than EUTOS high risk patients. The differences of CMR 4 rates between IM 800 and IM 400 at 3 years were 13% and at 4 years 8%, and of CMR 4.5 rates at 3 years 10% and at 4 years 13%. Grade 3 and 4 AE were not different between IM 400 and dose optimized IM 800. Independent of treatment approach, CMR 4 and more clearly CMR 4.5 at 3 years predicted better OS and PFS, if compared with patients without CMR 4 or CMR 4.5, respectively. CMR 4 and 4.5 were stable. After a median duration of CMR 4 of 3.7 years only 4 of 792 patients with CMR 4 have progressed. Life expectancy with CMR 4 and 4.5 was identical to that of the age matched population. We conclude that dose optimized IM induces CMR 4.5 faster than IM 400 and that CMR 4 and CMR 4.5 at 3 years are associated with a survival advantage. The present study investigated the occurrence of recurring errors in the learning process. Our goal was to determine the reasons for recurring errors. In considering these reasons we were trying to find the characteristic features of incorrect responses at the beginning of a learning process in order to predict the occurrence of recurring errors in the learning process. Response times and confidence measures were used as the predictors of error repetition. The results of two experiments have shown that response times and confidence levels for the recurring and singular errors at the beginning of learning are different. Response times were shorter for recurring errors than for singular errors at the beginning of a learning process. This information can be used to predict the repetition of such errors further along during learning. In addition, in both experiments, the correct responses were slower for stimuli that would cause recurring errors in the future. This allowed us to predict the repetition of such errors. Participants’ confidence estimates also varied with different types of erroneous responses and allowed the repetition of errors during the learning process to be predicted. Thus, our results allow the prediction of error repetition and also can be used to make suggestions about the phenomenon of recurring errors and the causes of their occurrence.

Purpose: Objective: to evaluate the effect of treatment of bacterial corneal ulcers of varying severity using a combination of antibacterial and corticosteroid drugs.Material and methods. 63 patients with bacterial corneal ulcers of varying severity were treated. The study used a quantitative scale to assess the severity of corneal ulcers. Group 1 — patients with mild corneal ulcers (18 patients); Group 2 — patients with moderate corneal ulcer (17 patients); group 3 — severe corneal ulcer (11 patients); control group — patients with mild corneal ulcer (17 patients). Patients of groups 1, 2 and 3, in addition to the conventional treatment, received Dexamethasone 0.1 % — 0.3 ml in parabulbar injections from the first day of treatment for the entire treatment period. Kr). In addition to the standard ophthalmological examination, all patients were assessed for the ulcer defect using measurements on OCT-POG accessing the parameters of the diameter of the corneal ulcer (d) and the depth coefficient (Kr).Results. The patients in the 1st group received antibacterial treatment and the addition of parabulbar injections of a corticosteroid drug (Dexamethasone). It allowed to decrease the treatment time and the period of hospitalization, promoted faster epithelialization and a reduction in the depth of the ulcer, less coarse scarring of the cornea, allowed to increase the functional results of treatment. The proposed treatment for severe corneal ulcers in 55 % of cases allows to use only conservative treatment. It is possible to obtain moderate opacity with vascularization in 60 % cases, which contributes to the preservation and/or improvement of visual acuity, despite the severity of the inflammatory process without the use of surgical intervention.Conclusion. The proposed treatment, involving the use of the corticosteroid drug Dexamethasone 0.1 % in parabulbar injections of 0.3 ml once daily for a bacterial corneal ulcer of mild severity for, average, 15.0 ± 1.4 days, a bacterial corneal ulcer of moderate severity — 18.0 ± 1.3 days, severe bacterial corneal ulcer — 25.0 ± 4.4 days, seems to be effective if the proposed parameters of the diameter and depth of the corneal ulcer are controlled.

Idiopathic intracranial hypertension (ICH) is a neuroophophthalmologic syndrome, the main ophthalmologic symptoms of which are vision loss and bilateral swelling of optic nerve disks (OD). The article provides a review covering various aspects of ICH. Epidemiology: ICH occurs mainly in women aged 20–45 years with body overweight. In this group of patients world incidence is 12–20 per 100 000 of population per year. In the total population it represents 0.5–2 cases per 100 000 of population per year. The eightfold gender predominance of women with ICH is observed. Etiology is not fully known until present time. The reliable connection between obesity in childbearing age women and menstrual cycle disorders as evidences of hormonal changes has been established. The probable mechanisms of increase of ICP are supposed: hyperproduction of and disturbances of its absorption, CSF mechanical pressure on the optic nerve sheath; restriction of venous outflow from the brain due to the pathology of venous sinuses; stimulating effect of abundant vitamin A in diet resulting in the fat tissue formation with the development of obesity; expression of the protein aquaporin, involved in the regulation of body mass and water metabolism in the subarachnoid space. Clinical manifestations. Ophthalmologic symptoms of ICH: transient visual impairment, sustained loss of visual acuity, photopsy, retrobulbar pain, diplopia. Non-ophthalmologic symptoms: headache, throbbing tinnitus, hearing loss, dizziness. Diagnostics. The valuable diagnostic data can be obtained by visual field investigation, ophthalmoscopic examination with revealing of bilateral OD-edema, OD and retinal optical coherent tomography, orbital ultrasound examination, brain MRI with venography. Treatment. Non-drug therapy: weight loss, lowcalorie diet with limited water and salt intake. Drug therapy: long-term oral use of carbonic anhydrase inhibitors (acetazolamide, topiramate). High doses of steroids (methylprednisone) are used for short-term treatment of patients with fulminant disease type before surgery. Surgery: bypass surgery, cerebral venous sinus stenting and fenestrations of the optic nerve sheath: bariatric surgery with reducing of stomach volume.

Introduction. Sprouted grain can cause food poisoning, since inappropriate conditions can promote the growth of pathogenic microorganisms on the grain surface. As a result, products of long-term storage use thermally-treated sprouted grain, the parameters of which depend on the initial bacteria content. There are different ways to reduce bacterial contamination of sprouted grain, each of which has its own advantages and disadvantages. Natural substances with antimicrobial properties, such as medicinal herbs, can serve as decontaminators. However, no scientific research has been performed so far to determine the exact temperature of grain sprouting to minimize its microbiological contamination. The research objective was to investigate the effect of antimicrobial agents and sprouting conditions on the microflora of wheat and buckwheat grain.
Study objects and methods. The study featured wheat grain and green buckwheat grain. A set of experiments was performed to define the effect of antimicrobial agents and sprouting conditions on the quantity of mesophilic aerobic and facultative anaerobic microorganisms (QMAFAnM), molds, and yeasts. During sprouting at 10–30°C for 90 h, the grain was irrigated with distilled water, potassium permanganate solution (KMnO4), calendula infusion, and celandine i nfusion. QMAFAnM and the count of molds and yeasts were determined by standard methods; the qualitative analysis of the microflora was based on their morphological and cultural characteristics.
Results and discussion. Microflora development during sprouting of wheat and buckwheat grains was controlled by selecting appropriate conditions and grain treatment methods. The herbal infusions for sprouting reduced the total microbial insemination of grain during sprouting by 52–68%; the calendula infusion reduced the contamination with molds by 47–51%, yeasts – by 100%.
Conclusion. The research revealed the total microbial count and the count of mold and yeast colonies in dry sprouted grain. The optimal temperature of sprouting wheat and buckwheat was 20 ± 2°C in the infusion of medicinal herbs: it minimized the microflora of sprouted grain and reduced the sprouting time to 46 h. Calendula infusion could be recommended for commercial use in order reduce the microbiological contamination of sprouted grain. The initial microbial population of the product was found to affect the mode of heat treatment in long-term storage products.
