Abstract Patients with multiple sclerosis (MS), an inflammatory, demyelinating disease of the Central Nervous System (CNS), derive the most benefit from disease modifying pharmacotherapies the earlier they are treated. Patients who have had a single episode of demyelination are at greater risk to develop clinically definite MS, but are not treated for MS, even though diagnosis of clinically definite MS could be significantly delayed if currently approved treatments are initiated immediately after the first attack. Defining biomarkers that identify patients with one demyelinating event who will convert to MS may provide a tool to allow earlier treatment of this "at risk" patient group. We have discovered a unique pattern of somatic mutations in the antibody genes of B cells isolated from the cerebrospinal fluid of patients with MS that can accurately and independently predict patients that subsequently develop definite MS after a single episode of demyelination. This antibody signature represents the first genetic biomarker candidate for MS outside of the MHC locus. If independently confirmed, our observations will have a major impact as a new diagnostic tool in patients with very early demyelinating disease of the CNS. Abstract Among 8455 people engaged in HIV care in 4 US cities, 4925 (58%) had treponemal testing at care entry. Of the 4925 tested, 3795 (77%) had a nonreactive result and might benefit from the reverse algorithm for a future incident syphilis diagnosis. Furthermore, low-barrier treponemal testing as a first step in the reverse algorithm may increase syphilis screening and decrease time to treatment.

Background In most countries, men are more likely to die by suicide than women. Adherence to dominant masculine norms, such as being self-reliant, is linked to suicide in men in Western cultures. We created a suicide prevention media campaign, “Boys Do Cry,” designed to challenge the “self-reliance” norm and encourage help-seeking in men. A music video was at the core of the campaign, which was an adapted version of the “Boys Don’t Cry” song from “The Cure.” There is evidence that suicide prevention media campaigns can encourage help-seeking for mental health difficulties. Objective We aimed to explore the reach, engagement, and themes of discussion prompted by the Boys Do Cry campaign on Twitter. Methods We used Twitter analytics data to investigate the reach and engagement of the Boys Do Cry campaign, including analyzing the characteristics of tweets posted by the campaign’s hosts. Throughout the campaign and immediately after, we also used Twitter data derived from the Twitter Application Programming Interface to analyze the tweeting patterns of users related to the campaign. In addition, we qualitatively analyzed the content of Boys Do Cry–related tweets during the campaign period. Results During the campaign, Twitter users saw the tweets posted by the hosts of the campaign a total of 140,650 times and engaged with its content a total of 4477 times. The 10 highest-performing tweets by the campaign hosts involved either a video or an image. Among the 10 highest-performing tweets, the first was one that included the campaign’s core video; the second was a screenshot of the tweet posted by Robert Smith, the lead singer of The Cure, sharing the Boys Do Cry campaign’s video and tagging the campaign’s hosts. In addition, the pattern of Twitter activity for the campaign-related tweets was considerably higher during the campaign than in the immediate postcampaign period, with half of the activity occurring during the first week of the campaign when Robert Smith promoted the campaign. Some of the key topics of discussions prompted by the Boys Do Cry campaign on Twitter involved users supporting the campaign; referencing the original song, band, or lead singer; reiterating the campaign’s messages; and having emotional responses to the campaign. Conclusions This study demonstrates that a brief media campaign such as Boys Do Cry can achieve good reach and engagement and can prompt discussions on Twitter about masculinity and suicide. Such discussions may lead to greater awareness about the importance of seeking help and providing support to those with mental health difficulties. However, this study suggests that longer, more intensive campaigns may be needed in order to amplify and sustain these results.

Abstract Natural Tregs from both WAS patients and WASp-deficient (WKO) mice were previously reported as unable to suppress the proliferation of conventional T-cells in vitro. Because WAS T-cells have a known defect of IL-2 production and this cytokine is important for the generation of Treg cells induced from naïve T-cells, we assessed the efficiency with which WKO naïve T-cells can differentiate in Tregs and observed a significant defect compared to WT. Such defect was not corrected by increasing concentration of anti-CD3 and TGF-β. Preliminary experiments indicated a role for endogenous IL-2 production in the generation of iTregs and pointed again to the IL-2 pathway as a major determinant of the Treg cells defects in WAS. Furthermore, we previously observed that the impaired suppression of T-cell proliferation could be partially rescued by pre-activation of nTregs. Because activated nTregs are known to be able to induce B-cell apoptosis, we set out to evaluate the function of WKO nTregs on B-cells. We found that pre-activated WKO Treg cells fail to effectively suppress B-cell proliferation. The reduced granzyme-B secretion by WKO nTregs compared to WT, suggest that the defective suppressive activity on B-cells may be due to a degranulation defect of WKO cells. In summary, our data suggest that both natural and induced arms of T regulatory cells are defective in WAS and that defective peripheral Treg induction may be involved in the pathogenesis of autoimmunity in this disease.

Introduo: O climatrio caracterizado pelo estado fisiolgico do hipoestrogenismo, com a interrupo definitiva dos ciclos menstruais. Objetivo: Analisar a qualidade do sono em mulheres climatricas ativas e sedentrias. Mtodos: A pesquisa foi realizada com 60 mulheres climatricas, onde foram dividas em trs grupos, 20 praticantes de exerccio aerbico, 20 que realizam treinamento resistido e 20 sedentrias, a coleta de dados foi realizada por meio de dois questionrios, para analise dos dados foi usado o teste Pearson Qui-quadrado com intervalo de confiana em 95% e significncia estabelecida em p 0,05. Resultados: A mdia de idade dos grupos de 49,25�0,97 a 53,4�1,08. Os resultados apontam uma dife 49,25�0,97 a 53,4�1,08. Os resultados apontam uma diferena significativa quando comparados os grupos a sintomatologia, porm com o questionrio de PSQI no houve diferenas significativas. Concluso: O estudo mostrou que comparadas quanto a sintomatologia, as mulheres praticantes de exerccios anaerbicos obtiveram um melhor resultado, entretanto em relao a qualidade do sono precisa-se de mais estudo.

Aim/Purpose: The purpose of this study is to use narrative inquiry to discover and understand how Chinese students leverage their strengths and multiple identities in socializing to American higher education and their profession. Chinese students engage with American academic culture while embracing their multiple identities. I will explore the cultural strengths they use to socialize and develop their personal, social, cultural, and professional identities in their doctoral educational experience. Background: Chinese international doctoral students encounter a unique socialization experience during their doctoral studies because they lack meaningful cross-cultural support. Likewise, it is problematic that Chinese students are often viewed as a homogeneous group and much prior research has emphasized the traditional deficit perspective in explaining how Chinese students must adjust and assimilate to the university environment. Methodology: This qualitative research uses narrative inquiry to study Chinese international doctoral students’ socialization experiences while retaining their authentic voices. Narrative inquiry allows for a more nuanced understanding of the experiences of Chinese students compared to the perceptions imposed by other stakeholders. The narrative methodology provides diverse ways to understand Chinese student interactions within American culture, place, and context. This study applies the three-dimensional approach to retell participants’ stories. The three-dimensional approach is more holistic and provides a broad lens to learn about the interactions, past, present, and future experiences of individuals through time and space. Contribution: This research shifts the narrative from the deficit view to a strength-based perspective as to how Chinese international doctoral students can rely on their cultural values and multiple identities as strengths to succeed academically, socially, and emotionally. Findings: Findings related to the literature in two important ways. First, findings support how the six cultural strengths of Yosso’s community cultural wealth apply to Chinese international doctoral students. Chinese students’ stories align with these strengths and through these strengths, they explore and develop their personal, social, cultural, and professional identity. Second, Chinese students’ stories as a counternarrative challenged and contradicted the essentialist view and misconception that Chinese students are a homogenous group personally, socially, culturally, or academically. Recommendations for Practitioners: The findings from this study offer insight for practitioners into what institutions and departments might do to support Chinese international doctoral students in their socialization journey. It is vital to support the whole student through understanding their multiple identities. Recommendation for Researchers: Chinese students and other diverse learners may benefit from peer and faculty mentors in different ways. Therefore, understanding the unique cross-cultural socialization needs and strength-based perspective will help tailor social activities and inclusive learning environments. Impact on Society: The current political, economic, and social relationships between the U.S. and China make it vital for American institutions to consider Chinese international doctoral students’ cross-cultural socialization journey. Future Research: Though it is hoped that this study is transferable, specific issues of how it can be generalized to other Chinese international doctoral students in other areas of the U.S. are beyond the scope of this study. Future research might explore how Chinese International doctoral students’ socialization experiences differ depending on where they study in the U.S.

Abstract Mucosa of the anterior human nares can harbor Staphylococcus aureus (SA), an etiologic agent that causes severe infections. Nasal carriage of SA is a complex host-pathogen interaction involving bacterial and host mediators that influence innate host defenses. We have revealed that the exoproteome of SA nasal carrier strains contains unique determinants, which enable SA to colonize the nares. In the current study, we assess how one of these bacterial mediators, Staphylococcal protein A (Spa), affects host innate immune response to SA nasal carriage. Functional disruption of spa was performed in an SA nasal carrier strain using Targetron insertion of a group II intron. Subsequent in vitro growth kinetics analysis and ex vivo culture on human nasal epithelial cells (HNECs) revealed no differences between wild type and spa-disrupted (Δspa) SA strains. However, the Δspa mutant induced lower expression of pro-inflammatory cytokines including IL6, IL8 and TNFα from HNECs compared to the levels elicited by the wild type strain. Additionally, IL17, a crucial regulator of mucosal immunity, was induced at significantly lower levels in HNECs inoculated with Δspa SA compared to those inoculated with the wild type SA strain. Furthermore, expression of the chemokines MIP1β and MCP1, as well as HL60 neutrophil migration, were significantly reduced in conditioned media from HNECs inoculated with Δspa mutants. Spa is likely involved in mediating the mucosal response to SA colonizing the nose.

To enhance enzymatic hydrolysis efficiency of the corn straw, a process microwave pretreatment of corn straw was optimized using response surface methodology (RSM). Estimated optimum parameters were as follows: water to corn straw ratio is 58:1, microwave power is 640w and processing time is 5 minutes. Under these conditions, a maximum enzyme hydrolysis ratio of corn straw (49.25%) after microwave pretreatment was reached.

The environmental problem of climate change is an issue that needs to be addressed worldwide. As the electricity-generating power sector is the largest contributor of CO2 in the country, low-carbon technologies or sustainable energy systems are being considered as viable alternatives to reduce the CO2 emissions from this sector. These are fossil-based power plants with carbon capture and storage (F-CCS) technology, nuclear energy (NE) and renewable energy (RE) technologies, particularly solar energy (SE), wind energy (WE), hydroelectricity (HE), geothermal energy (GE) and biomass (BE). However, successful implementation of any of these CCMOs depends not only on the technical and economic aspect but also the socio-political aspect of the project. This study therefore proposes an analytical decision modeling framework to evaluate these options by incorporating the subjective judgment of stakeholders. The Analytic Hierarchy Process (AHP) was used to structure the problem and quantify the relative preference of each option with respect to four criteria namely environmental effectiveness (EE), economic viability (EV), technical implementability (TI), and social acceptability (SA).Results from the decision model indicate that the most important criterion is environmental effectiveness, and the least important is social acceptability. With respect to environmental effectiveness, their most preferred CCMO was solar energy whereas their least preferred is nuclear energy mainly because of the risk posed by the generated nuclear wastes. With respect to economic viability, their most preferred CCMO was geothermal energy, and the least preferred was nuclear energy. With respect to technical implementability, the respondents gave the highest preference weight on geothermal energy and the least preferred is nuclear energy. With respect to social acceptability, the most preferred was wind energy and again, the least preferred was nuclear energy.

Thymic rosettes (ROS), structures consisting of thymic lymphoid cells attached to a central stromal cell, were isolated from mouse thymus by collagenase digestion and unit-gravity elutriation. The ROS were then separated into those where the stromal cells were either macrophage-like (M-ROS) or dendritic cell-like (D-ROS), on the basis of the differences in adherence properties or in the level of MAC-1 surface antigen. The ROS were then dissociated and the thymocyte content analyzed by immunofluorescent staining and flow cytometry. M-ROS and D-ROS differed in thymocyte composition, although the major component of both was the CD4+CD8+cortical thymocyte. D-ROS were enriched in thymocytes expressing high levels of surface T-cell antigen receptor (TcR) and the associated CD3 complex, and these included both CD4+CD8+CD3++and CD4-CD8+CD3++mature thymocytes. M-ROS were enriched in CD4-CD8-thymocytes and had a reduced content of thymocytes expressing high TcR-CD3 levels; they nevertheless contained some mature thymocytes, but only of the CD4+CD8-CD3++category. Several lines of evidence indicated that the mature thymocytes in ROS were cells recently formed in the cortex, and were not from the medullary pool. ROSassociated mature thymocytes expressed lower levels of H-2K than free, mature thymocytes. The CD4+CD8+CD3++subpopulation, believed to be a developmental intermediate between cortical thymocytes and mature T cells, was present in both ROS populations. Further, late intermediates leading to both mature T-cell categories were evident in D-ROS, but only those leading to CD4+CD8-CD3++T cells were evident in M-ROS. The results are compatible with a role for ROS in TcR-specificity selection and in the final maturation steps in the thymic cortex.
