A novel complex [Ni(C10H8O5N3)2]and#183;H2Oand#183;(CH3OH) (1) (C10H9O5N3= 2-[(4-nitro- benzoyl)-hydrazone]-propionic acid) has been prepared by reaction of NiCl2 with C10H9O5N3 in mixed solvent of methanol and water. The Ni(II) complex in octahedral geometry was characterized by elemental analysis and X-ray single crystal diffraction. Isothermal titration calorimetry (ITC) was performed to observe the reactions between Ni(II) and C10H9O5N3 in methanol solvents. The results showed that the binding constant (Ka), enthalpy change (ΔH), entropy change (ΔS), and Gibbs free energy change (ΔG) of the system are 5.2and#215;105, -11.82 kJand#183;mol-1, 69.78 Jand#183;mol-1and#183;K-1 and -32.62 kJand#183;mol-1, respectively. In addition, the structure was optimized and frontier molecular orbital (FMO) of (1) were analyzed by B3LYP and HF methods with 6-31g level.

Методами вибрационной магнитометрии проведены исследования изменений электрофизических характеристик нескольких типов современных коммерческих многожильных композиционных проводов на основе сверхпроводящего соединения Nb3Sn при температурах 4.2, 7 и 12 K в магнитных полях до 8 T до и после облучения на циклотроне НИЦ КИ быстрыми протонами с энергией 32 MeV и флюенсами облучения phi t = 3 · 1016, 1· 1017, 3· 1017 и 1· 1018 cm-2. Для всех образцов при флюенсах облучения до phi t = 1 · 1017 cm-2 отмечен рост критической плотности тока, наиболее резко выраженный в сильных магнитных полях и при низких температурах; при флюенсе облучения phi t = 1 · 1018 cm-2 наблюдается падение критической плотности тока до уровня меньше исходного. Стабильность нового однодоменного терапевтического антитела к рецептору ErbB3 исследована методом спектроскопии флуоресценции при различных концентрациях денатурирующего вещества и температуре. Анализ экспериментальных кривых денатурации позволил построить полную термодинамическую модель процесса денатурации и определить все параметры перехода: Delta G = 8.5 kcal · mol-1, Tm = 76oC, Delta Hm= = 107 kcal· mol-1, Delta Cp = 1.8 kcal· K-1· mol-1. Полученные данные свидетельствуют о высокой стабильности антитела в широком диапазоне условий, что чрезвычайно важно для дальнейших структурно-функциональных исследований и возможного терапевтического применения. Abstract Glycogen synthase kinase 3 beta (GSK3β) is a critical protein kinase that phosphorylates numerous proteins in cells and thereby impacts multiple pathways including the β-Catenin/TCF/LEF-1 pathway. MicroRNAs (miRs) are a class of noncoding small RNAs of ∼22 nucleotides in length. Both GSK3β and miR play myriad roles in cell functions including stem cell development, apoptosis, embryogenesis and tumorigenesis. Here we show that GSK3β inhibits the expression of miR-96, miR-182 and miR-183 through the β-Catenin/TCF/LEF-1 pathway. Knockout of GSK3β in mouse embryonic fibroblast cells increases expression of miR-96, miR-182 and miR-183, coinciding with increases in the protein level and nuclear translocation of β-Catenin. In addition, overexpression of β-Catenin enhances the expression of miR-96, miR-182 and miR-183 in human gastric cancer AGS cells. GSK3β protein levels are decreased in human gastric cancer tissue compared with surrounding normal gastric tissue, coinciding with increases of β-Catenin protein, miR-96, miR-182, miR-183 and primary miR-183-96-182 cluster (pri-miR-183). Furthermore, suppression of miR-183-96-182 cluster with miRCURY LNA miR inhibitors decreases the proliferation and migration of AGS cells. Knockdown of GSK3β with siRNA increases the proliferation of AGS cells. Mechanistically, we show that β-Catenin/TCF/LEF-1 binds to the promoter of miR-183-96-182 cluster gene and thereby activates the transcription of the cluster. In summary, our findings identify a novel role for GSK3β in the regulation of miR-183-96-182 biogenesis through β-Catenin/TCF/LEF-1 pathway in gastric cancer cells.

Excessive saturated fatty acids (SFA) uptake is known to be a primary cause of obesity, a widely acknowledged risk factor of insulin resistance and type 2 diabetes. Although specific microRNAs (miRNAs) targeting insulin signaling intermediates are dysregulated by SFA, their effects on insulin signaling and sensitivity are largely unknown. Here, we investigated the role of SFA-induced miR-183-5p in the regulation of proximal insulin signaling molecules and the development of hepatic insulin resistance. HepG2 hepatocytes treated with palmitate and the livers of high-fat diet (HFD)-fed mice exhibited impaired insulin signaling resulting from dramatic reductions in the protein expressions of insulin receptor (INSR) and insulin receptor substrate-1 (IRS-1). Differential expression analysis showed the level of miR-183-5p, which tentatively targets the 3′UTR of IRS-1, was significantly elevated in palmitate-treated HepG2 hepatocytes and the livers of HFD-fed mice. Dual-luciferase analysis showed miR-183-5p bound directly to the 3′UTR of IRS-1 and reduced IRS-1 expression at the post-transcriptional stage. Moreover, transfection of HepG2 hepatocytes with miR-183-5p mimic significantly inhibited IRS-1 expression and hindered insulin signaling, consequently inhibiting insulin-stimulated glycogen synthesis. Collectively, this study reveals a novel mechanism whereby miR-183-5p induction by SFA impairs insulin signaling and suggests miR-183-5p plays a crucial role in the pathogenesis of hepatic insulin resistance in the background of obesity.

Backgrounds/Aims: MicroRNAs (miRs) often contribute to the progression of non-small cell lung cancer (NSCLC) via regulation of mRNAs that are involved in lung homeostasis. We conducted a study aimed at exploring the roles of miR-183 in the proliferation, epithelial-mesenchymal transition (EMT), invasion and migration of human NSCLC cells via targeting MTA1. Methods: NSCLC and adjacent normal tissues were collected from 194 patients with NSCLC. Positive expression of MTA1 protein was detected by immunohistochemistry. The highest levels of expression of miR-183 were detected using RT-qPCR in SPC-A-1 cells, which were selected and assigned to the following groups: blank, negative control (NC), miR-183 mimic, miR-183 inhibitor, siRNA-MTA1, and miR-183 inhibitor + siRNA-MTA1. The expression of miR-183 and the mRNA and protein expression of MTA1, E-cadherin, Vimentin, Snail, PCNA, Bax and Bcl-2 in tissues and transfected cells were measured using RT-qPCR and western blot analysis. Cell proliferation, apoptosis, migration and invasion were evaluated by CCK-8, flow cytometry, scratch tests and Transwell assays. Tumor xenografts were conducted in nude mice to determine tumor growth. Results: SPC-A-1 cells with the highest levels of miR-183 expression were selected. Compared with adjacent normal tissues, the expression of miR-183 and the mRNA and protein expression of E-cadherin and Bax were decreased in NSCLC tissues, while mRNA and protein expression of MTA1, Vimentin, snail, PCNA and Bcl-2 were increased. MiR-183 was over-expressed in the miR-183 mimic group and under-expressed in the miR-183 inhibitor and miR-183 inhibitor + siRNA-MTA1 groups. In the miR-183 mimic and siRNA-MTA1 groups, the mRNA and protein expression of E-cadherin and Bax, as well as cell apoptosis, were enhanced, while the expression levels of MTA1, Vimentin, snail, PCNA and Bcl-2 mRNA and protein, cell proliferation, migration, invasion and tumor growth were reduced relative to the blank and NC groups. The miR-183 inhibitor group exhibited an opposite trend. Conclusion: Our study indicates that miR-183 down-regulates MTA1 to inhibit the proliferation, EMT, migration and invasion of human NSCLC cells.

Background: MiR-183-5p plays an important role in the pathophysiology of many tumors, while the role of MiR-183-5p in liver cancer is unclear. Methods: In this study, quantitative reverse transcription-polymerase chain reaction and Western blotting were used to detect the expression of miR-183-5p in liver cancer cell lines, liver cancer tissues, and normal tissues adjacent to the cancer, and to explore the mechanism of miR-183-5p regulating liver cancer progression. The in vitro effects of miR-183-5p were evaluated by CCK-8, colony formation test, and wound healing test. Various databases were used to predict the target mRNA of miR-183-5p and verified by luciferase report analysis. In addition, the effects of miR-183-5p and its target gene on the survival of patients with liver cancer were also analyzed. Results: miR-183-5p was highly expressed in hepatocellular carcinoma cells and tissues, and was related to some clinicopathological features. MiR-183-5p can promote the proliferation and migration of liver cancer cells. Using the bioinformatics database, we proved that miR-183-5p is related to the survival of liver cancer patients. Insulin receptor substrate 1 (IRS1) is a target of miR-183-5p, and luciferase analysis confirmed that miR-183-5p combines with the 3′-untranslated region (3′-UTR) of IRS1. Conclusion: The miR-183-5p/IRS1 axis may be a new target for liver cancer research.

Background Diarrhea is a serious health problem in children, with the highest mortality rate in sub-Saharan Africa. Diarrheagenic Escherichia coli (DEC) is among the major bacterial causes of diarrhea in children under age five. The present study aims to determine molecular epidemiology and antimicrobial resistance profiles of DEC and identify contributing factors for acquisition among children under age five in Central Ethiopia. Methods A health facility-centered cross-sectional study was conducted in Addis Ababa and Debre Berhan, Ethiopia, from December 2020 to August 2021. A total of 476 specimens, 391 from diarrheic and 85 from non-diarrheic children under age five were collected. Bacterial isolation and identification, antimicrobial susceptibility, and pathotype determination using polymerase chain reaction (PCR) were done. Results Of the 476 specimens analyzed, 89.9% (428/476) were positive for E. coli, of which 183 were positive for one or more genes coding DEC pathotypes. The overall prevalence of the DEC pathotype was 38.2% (183/476). The predominant DEC pathotype was enteroaggregative E. coli (EAEC) (41.5%, 76/183), followed by enterotoxigenic E. coli (21.3%, 39/183), enteropathogenic E. coli (15.3%, 28/183), enteroinvasive E. coli (12.6%, 23/183), hybrid strains (7.1%, 13/183), Shiga toxin-producing E. coli (1.6%, 3/183), and diffusely-adherent E. coli (0.6%, 1/183). DEC was detected in 40.7% (159/391) of diarrheic and 28.2% (24/85) in non-diarrheic children (p = 0.020). The majority of the DEC pathotypes were resistant to ampicillin (95.1%, 174/183) and tetracycline (91.3%, 167/183). A higher rate of resistance to trimethoprim-sulfamethoxazole (58%, 44/76), ciprofloxacin (22%, 17/76), ceftazidime and cefotaxime (20%, 15/76) was seen among EAEC pathotypes. Multidrug resistance (MDR) was detected in 43.2% (79/183) of the pathotypes, whereas extended spectrum ß-lactamase and carbapenemase producers were 16.4% (30/183) and 2.2% (4/183), respectively. Conclusion All six common DEC pathotypes that have the potential to cause severe diarrheal outbreaks were found in children in the study area; the dominant one being EAEC with a high rate of MDR.

AbstractAccumulating studies highlight the critical role of long non-coding RNAs (lncRNAs) in the development of various human cancers. Extracellular leucine rich repeat and fibronectin type III domain containing 1-antisense RNA 1 (ELFN1-AS1) was shown to be a newly found lncRNA that abnormally expressed in human tumors. However, till now the specific function of this lncRNA in esophageal cancer (ESCA) remains unknown. In this study, we discovered that higher ELFN1-AS1 expression indicated shorter patient survival in pan-cancer, including ESCA, using online The Cancer Genome Atlas (TCGA) tools. The lncRNA ELFN1-AS1 was significantly up-regulated in ESCA tissues and cell lines when compared with the counterparts. Down-regulation of ELFN1-AS1 restrained cell proliferation, migration, and invasion of ESCA in vitro. In addition, we found that the expression of microRNA-183-3p (miR-183-3p) and ELFN1-AS1 or glutamine-fructose-6-phosphate transaminase 1 (GFPT1) were inversely correlated in ESCA. Both ELFN1-AS1 and GFPT1 are direct targets of miR-183-3p in ESCA. The effects of ELFN1-AS1 knockdown on ESCA progression were partially rescued by inhibition of miR-183-3p or over-expression of GFPT1. In summary, the results of this study suggest that the lncRNA ELFN1-AS1 facilitates the progression of ESCA by acting as a competing endogenous RNA (ceRNA) to promote GFPT1 expression via sponging miR-183-3p.

AbstractObjectiveTo evaluate the serum levels of microRNA-183 (miR-183) and thymidine kinase 1 (TK1) in colorectal cancer patients and their clinical value as biomarkers for colorectal cancer auxiliary diagnosis.MethodsForty-six pathology confirmed colorectal cancer patients and 46 healthy controls were included in this study. The serum levels of miR-183 and TK1 in colorectal cancer patients and healthy controls were examined by real-time PCR and chemiluminescence detection assay respectively. The diagnostic value of serum miR-183 and TK1 as tumor biomarkers for colorectal cancer detection was evaluated through receiver operating characteristic (ROC) curves.ResultsThe median serum relative expression of miR-183 was 1.33 (0.34-5.65) and 0.88 (0.26-4.67) in colorectal cancer patients and healthy controls respectively with significant statistical difference (p<0.05). Using serum miR-183 as the diagnostic reference, the colorectal cancer diagnosis sensitivity, specificity and AUC was 65.22%, 63.04% and 0.69 respectively. The median serum level of TK1 was 3.33 (0.78-5.78) pmol/L and 0.99 (0.34-4.46) pmol/L in colorectal cancer patients and healthy controls respectively with significant statistical difference (p<0.05). The diagnostic sensitivity, specificity and AUC was 84.78%, 78.26% and 0.88 respectively forserum TK1 as reference for colorectal diagnosis. The pearson correlation test was used to evaluate the serum miR-183 and TK1 correlation in colorectal cancer patients. However, no significant correlation between serum miR-183 and TK1 was found in colorectal patients (p>0.05).ConclusionSerum levels of miR-183 and TK1 arepotential biomarkers for colorectal cancer auxiliary diagnosis.

Изготовлены 4H-SiC p+-n0-n+-диоды с однородным лавинным пробоем при напряжении 1860 В. В лавинном режиме измерены импульсные вольт-амперные характеристики диодов до плотности тока 4000 А/см2. Показано,что напряжение лавинного пробоя растет при повышении температуры. Определены лавинное сопротивление диодов (8.6&#183; 10-2 Ом&#183;см2), скорость дрейфа электронов в n0-базе при полях выше 106 В/см (7.8&#183; 106 см/c), относительный температурный коэффициент напряжения пробоя (2.1&#183;10-4 K-1). Abstrak Artikel ini membahas tentang telaah buku teks bahasa Arab yang berfokus pada materi maharah qira’ah dilihat dalam setiap bab berdasarkan perspektif KMA 183 Tahun 2019 yang terdiri dari KI 1 (Spiritual), KI 2 (Sosial), KI 3 (Pengetahuan), dan KI 4 (Keterampilan). Artikel ini menggunakan metode kajian pustaka (library research) yaitu pengumpulan data dengan mengkaji kepustakaan agar diperoleh kesimpulan pada objek kajian, proses yang dilakukan dalam pengumpulan data yaitu dengan catatan dokumentasi yang berupa data-data tertulis terkait materi qira’ah buku ajar bahasa Arab siswa kelas XII KMA 183 tahun 2019. Penelitian ini bertujuan untuk mengetahui buku teks bahasa Arab apakah sudah sesuai dengan kompetensi inti KMA 183 tahun 2019 yang difokuskan pada materi maharah qira’ah dari setiap bab. Hasil penelitian pada artikel ini adalah terdapat rana spiritual, sosial, pengetahuan, maupun keterampilan, di dalam sub bab qira’ah buku teks KMA 183 tahun 2019. Jadi, dapat diambil kesimpulan bahwa buku teks bahasa Arab sesuai dengan kompetensi inti KMA 183 tahun 2019.

Aims MicroRNA-183 ( miR-183) is known to play important roles in osteoarthritis (OA) pain. The aims of this study were to explore the specific functions of miR-183 in OA pain and to investigate the underlying mechanisms. Methods Clinical samples were collected from patients with OA, and a mouse model of OA pain was constructed by surgically induced destabilization of the medial meniscus (DMM). Reverse transcription quantitative polymerase chain reaction was employed to measure the expression of miR-183, transforming growth factor α (TGFα), C-C motif chemokine ligand 2 ( CCL2), proinflammatory cytokines (interleukin (IL)-6, IL-1β, and tumour necrosis factor-α ( TNF-α)), and pain-related factors (transient receptor potential vanilloid subtype-1 ( TRPV1), voltage-gated sodium 1.3, 1.7, and 1.8 ( Nav1.3, Nav1.7, and Nav1.8)). Expression of miR-183 in the dorsal root ganglia (DRG) of mice was evaluated by in situ hybridization. TGFα, CCL2, and C-C chemokine receptor type 2 ( CCR2) levels were examined by immunoblot analysis and interaction between miR-183 and TGFα, determined by luciferase reporter assay. The extent of pain in mice was measured using a behavioural assay, and OA severity assessed by Safranin O and Fast Green staining. Immunofluorescent staining was conducted to examine the infiltration of macrophages in mouse DRG. Results miR-183 was downregulated in tissue samples from patients and mice with OA. In DMM mice, overexpression of miR-183 inhibited the expression of proinflammatory cytokines ( IL-6, IL-1β, TNF-α) and pain-related factors ( TRPV1, Nav1.3, Nav1.7, Nav1.8) in DRG. OA pain was relieved by miR-183-mediated inhibition of macrophage infiltration, and dual luciferase reporter assay demonstrated that miR-183 directly targeted TGFα. Conclusion Our data demonstrate that miR-183 can ameliorate OA pain by inhibiting the TGFα- CCL2/ CCR2 signalling axis, providing an excellent therapeutic target for OA treatment. Boron is one of the significant micronutrients for the plants; however, its excessive concentrations are lethal to plants. The accumulation of boron may cause damage to the liver, kidney, nervous system, respiratory system, digestive system, reproductive system and even threaten human life. Researchers consider using different methods to prepare boron chelating resins. In this work, two innovative, facile, and low‐cost methods were developed to prepare polystyrene-alcohol resins. The KDN and KLA resins had the highest adsorption capacity for boric acid in an aqueous solution at 288 K ,and the adsorption capacity was 36.17 mgand#183;g-1 and 43.47 mgand#183;g-1, respectively. The desorption percentages of boric acid on KDN and KLA resins were 90.75 % and 90.32 % respectively, by using 1 moland#183;L-1 HCl as an eluent. Therefore, KDN and KLA resins have the characteristics of a simple experimental process, large adsorption capacity, high elution rate, reusable, and low production cost. Therefore, it has a high commercial value and development prospects.

Evaluation of the sternum is part of the routine examination of small animal thoracic radiographs. However, descriptions on frequency and type of abnormalities are lacking. This retrospective observational study aimed to describe abnormal radiographic findings of the sternum in a cross-section of client-owned dogs and cats undergoing thoracic radiography between 1 January 2019 and 1 January 2021 for reasons unrelated to diseases of the sternum. The study population consisted of 777 dogs (mean age, 7.3 ± 3.9 years) and 183 cats (mean age, 7.3 ± 5.1 years). Sternal abnormalities were observed in 189/777 (24%) dogs and 53/183 (29%) cats, mostly around the intersternebral cartilages, accounting for 32/80 (40%) dogs and 20/35 (57%) cats. This was followed by an abnormal number of sternal segments (8% dogs, range 3–9 sternebrae; 15% cats, range 7–9 sternebra). Pectus excavatum was observed in 6/777 (0.8%) dogs and 6/183 (3%) cats, and pectus carinatum in 18/777 (2%) dogs and 2/183 (1%) cats. Post-traumatic changes, such as subluxation, were observed in nine dogs (1.1%) and three cats (1.6%). Presumed prostatic carcinoma metastasis and malignant lymphoma were observed in two dogs (0.2%). Incidental radiographic sternal abnormalities are common in cats and dogs but mostly of unknown clinical relevance.

(1) Background: circRNAs are closed circular molecules with covalent bonds generated by reverse shearing, which have high stability and have different manifestations in different tissues, cells, or physiological conditions and play important roles in various disease processes and physiological processes. In addition, circ_PIAS1 has been screened out and verified, and the bioinformatics analyzed in previous studies. In this study, we investigated the function of circ_PIAS1 and studied its role in ALV-J infection to provide a basis for the role of circRNA in ALV-J infection. (2) Methods: the effect of circ_PIAS1 on apoptosis during ALV-J infection was studied by flow cytometry and detection of apoptotic gene expression, and miR-183 was screened by a biotin-labeled RNA pull-down technique. After overexpression and inhibition of miR-183, the effect of miR-183 on apoptosis in the process of ALV-J infection was studied by flow cytometry and detection of apoptotic gene expression. (3) Results: after overexpression of circ_PIAS1, flow cytometry and apoptotic gene expression showed that circ_PIAS1 promoted apoptosis. The results of RNA pull-down showed that 173 miRNAs could bind to circ_PIAS1, and circ_PIAS1 up-regulated the expression of miR-183. On the other hand, the same results were obtained whether miR-183 was overexpressed or inhibited that miR-183 affected ALV-J infection by promoting cell apoptosis. (4) Conclusions: circ_PIAS1 up-regulated the expression of miR-183 and influenced ALV-J infection by promoting cell apoptosis.

Astract This research aims to 1). analyze the level of Arabic proficiency in the KMA 183 Arabic text book from the perspective of ACTFL, 2). Knowing the urgency of the arrangement of material in the KMA 183 Arabic textbook on Arabic language proficiency. 3) describe the implications of the analysis of the KMA 183 Arabic textbook based on the ACTFL language proficiency guidelines. The research is library research by presenting data, documents and other supporting files with a qualitative descriptive approach, using content analysis techniques, namely analyzing data according to its contents so as to provide an overview of the suitability of the material contained in textbooks Arabic at the MA level of class XI by analyzing through the perspective of language proficiency guidelines. The results of research conducted by analyzing KMA 183 Arabic textbooks can be explained that in Productive skills, reading material is presented at the intermediate level and beginner level dominated by the middle intermediate level (intyermediate mid), while writing material is presented with inconsistent material weights. and irregular between novice mid to intermediate mid, while Receptive skills include reading skills and listening skills. On reading material as a whole has increased. In the first two chapters, it begins with the material level of proficiency at the beginner high level (Novice High), then increases to advanced high. This is different from material in listening skills which is consistently at the intermediate level, both intermediate low and medium high, thereby showing that the process of transforming material changes can be enjoyed by learners in each chapter. Penelitian merupakan penelitian kepustakaan (Library Research) dengan memaparkan data, dokumen dan berkas-berkas penunjang lainnya dengan pendekatan deskriptif kualitatif, dengan menggunakan teknis analisis isi (Content Analysis) yaitu melakukan Analisa data menurut isinya sehingga memberikan gambaran tentang kesesuaian materi yang terdapat dalam buku teks Bahasa arab pada jenjang MA kelas XI dengan menganalisis melalui perspektif pedoman kemahiran berbahasa. Hasil penelitian yang dilakukan dengan menganalisa buku teks Bahasa Arab KMA 183 dapat dipaparkan bahwa pada kemahiran Produktif yaitu materi membaca disajikan berada pada tingkat menengah dan tingkat pemula dengan didominasi oleh tingkatan menengah tengah (intyermediate mid), sedangkan materi menulis disajikan dengan bobot materi yang tidak konsisten dan tidak teratur antara pemula menengah (novice mid) sampai kepada menengah tengah (intermediate mid), sedangkan untuk kemahiran Reseptif mencakup kemahiran membaca dan kemahiran menyimak. Pada materi membaca yang secara keseluruhan mengalami peningkatan. Pada dua bab pertama diawali dengan tingkat materi kemahiran di level pemula tinggi (Novice High), lalu meningkat di mahir tinggi (advanced high). Background/Aims: Disseminated tumors, known as metastases, are responsible for ninety-percent of mortality due to cancer. Epithelial to mesenchymal transition, a phenomenon required for morphological conversion of non-motile discoid shaped epithelial cells to highly motile spindle-shaped mesenchymal cells, is thought to be a pre-requisite for metastatic progression. Metastasis-associated 1 (MTA1) protein is a prime inducer of EMT and metastatic progression in all solid tumors including hepatocellular carcinoma (HCC). However, the molecular mechanisms that regulate the expression and function of MTA1 in HCC have not been elucidated. Methods: In silico prediction algorithms were used to find microRNAs (miRNAs) that may target MTA1. We examined the relationship between the expression of MTA1 and miR-183 using quantitative real time PCR. We also determined the levels of the MTA1 protein using immunohistochemistry. Reporter assays, in the presence and absence of the miR-183 mimic, were used to confirm MTA1 as a bona fide target of miR183. The effect of miR-183 on HCC pathogenesis was determined using a combination of in vitro migration and invasion assay, together with in vivo xenograft experiments. The correlation between miR-183 and MTA1 expression was also studied in samples from HCC patients, and in The Cancer Genome Atlas dataset. Results: Analysis of the sequence database revealed that MTA1 is a putative target of miR-183. MTA1 protein and RNA expression showed opposite trends to miR-183 expression in breast, renal, prostate, and testicular tissue samples from cancer patients, and in the metastatic HCC cell line HepG2. An inverse correlation was also observed between MTA1 (high) and miR-183 (low) expression within samples from HHC patients and in the TCGA dataset. Reporter assays in HepG2 cells showed that miR-183 could inhibit translation of a reporter harboring the wild-type, but not the mutant miR-183 3’-untranslated region (UTR). In addition, miR-183 significantly inhibited in vitro migration and invasion in HepG2 cells, and in vivo hepatic metastasis. Conclusion: Our results reveal a novel post-transcriptional regulatory mechanism for MTA1 expression via miR-183, which is suppressed during HCC pathogenesis.

Методом Бриджмена выращены монокристаллы твердых растворов (FeIn2S4)x&#183;-0.8pt(AgIn5S8)1-x во всем диапазоне концентраций. Установлено, что выращенные монокристаллы кристаллизуются в кубической структуре шпинели. Методом ядерной гамма-резонансной спектроскопии изучены локальные конфигурации ионов железа в структуре твердых растворов (FeIn2S4)x&#183;-0.8pt(AgIn5S8)1-x. По спектрам пропускания в области края собственного поглощения определена ширина запрещенной зоны (Eg) указанных твердых растворов и построена ее концентрационная зависимость. Textile industry is a continuous source of colored wastewater. This wastewater frequently used for irrigation purpose in many underdeveloped countries including Pakistan. In this study, we isolated the bacterial strains capable of decolorizing dyes and promote the plant growth. Hence to decolorize the reactive red 120 (RR120), the strain WS-D/183 was optimized following response surface methodology (RSM) based modeling approach. Moreover, strain WS-D/183 was also assessed for plant growth promoting characteristics. Results revealed that the strain WS-D/183 showed a good potential for decolorization of structurally diverse types of azo dyes on reaction with a mixture of heavy metal ions (Cr6+, Cd2+, Zn2+, Pb2+). This strain concurrently removed reactive dyes (100 mg L-1 ) and reduced Cr(VI). Results showed that each dye was decolorized up to 90% except reactive yellow-2 which was decolorized up to 57.4%. Furthermore, the bacterium reduced Cr(VI) by 41 to 95% along with concurrent decolorization of RR120. This bacterium was also found to carry plant growth promoting traits including inorganic phosphate solubilization (497.6 ± 14.8 µg mL-1 ) and indole-3-acetic acid production (21.07 ± 0.9 µg mL-1 ). A phytotoxicity evaluation study indicated that irrigation of mung bean [Vigna radiata (L.) Wilczek] with RR120, Cr(VI) and RR120+Cr(VI) contaminated waters treated with the strain WS-D/183 enhanced germination along with plumule and radical length of seedlings. Results suggested that Pseudomonas sp. WS-D/183 is a valuable addition to the bioresources, which can be used to devise textile wastewater treatment strategies as well as for integrated bioremediation and plant growth promotion in agricultural soils contaminated with textile wastewaters. © 2020 Friends Science Publishers

A large‐scale immunoaffinity (IA) purification process was developed for the isolation of recombinant soluble antigen CD4 (sCD4) from Escherichia coli fermentations. The monoclonal antibody used for IA purification of sCD4 recognized a conformation‐dependent epitope on the surface of domain 1 of CD4. IA chromatography was used to purify both sCD4‐183, consisting of the N‐terminal 183 amino acids of human CD4, and sCD4‐PE40, a fusion protein consisting of the N‐terminal 178 amino acids of CD4 and amino acids 1–3 and 253–613 of Pseudomonas exotoxin A (PE40). sCD4‐183 was purified from E. coli cell pellets using cell disruption, protein solubilization, oxidation, Q‐Sepharose anion‐exchange and IA chromatography steps. sCD4‐PE40 was purified from cell pellets using cell disruption, protein solubilization, oxidation, Cu(2+)‐immobilized metal‐affinity chromatography, anion‐exchange and IA chromatography steps. The IA‐purified sCD4 analogues demonstrated the correct apparent molecular masses on SDS/PAGE. The immobilized monoclonal antibody appeared to select for correctly folded CD4 protein, since sCD4‐183 and sCD4‐PE40 purified by the IA method bound human‐immunodeficiency‐virus glycoprotein gp120 (HIV gp120) in vitro. sCD4‐PE40 purified by IA chromatography also inhibited protein synthesis in CV‐1 cells expressing HIV gp120/160 at the cell surface. Relatively high recoveries of sCD4‐183 and sCD4‐PE40 were observed in the IA step of the purification process (71 and 79% recovery respectively). The results demonstrate that immobilized monoclonal antibodies directed against conformational epitopes may be used for rapid purification of gram amounts of correctly folded protein from mixtures of oxidized E. coli proteins.

Neuroblastoma, a malignant tumor of the sympathetic nervous system, is an aggressive extracranial tumor in childhood. Long noncoding RNAs (lncRNAs) have been discovered to play a key role in the eukaryotic regulatory gene network and be involved in a wide variety of biological processes. We observed that the expression of lncRNA nuclear-enriched abundant transcript-1 (NEAT1) was significantly decreased in human neuroblastoma tissues and cell lines, compared with the normal. We observed cell proliferation, migration, and invasion with Cell Counting Kit-8 assay, colony formation assay, and Transwell assay to investigate the effects of NEAT1, miR-183-5p, or FOXP1 on neuroblastoma cells. And we also used StarBase and luciferase reporter gene assay to predict and confirm the interaction of NEAT1, miR-183-5p, and FOXP1 in neuroblastoma cells. First, overexpression of NEAT1 suppressed cell proliferation and played a key role in cell migration and invasion. In addition, NEAT1 was demonstrated to directly interact with miR-183-5p and exerted its antioncogenic role in neuroblastoma by negatively regulating miR-183-5p expression. miR-183-5p suppressed the expression of FOXP1 and regulated cell proliferation and migration by directly targeting FOXP1 mRNA 3′-untranslated region. Moreover, FOXP1 antagonized the effect of miR-183-5p on the phosphorylation of extracellular-regulated kinase/protein kinase B (ERK/AKT), while FOXP1 siRNA increased the reduced phosphorylation of ERK/AKT caused by miR-183-5p inhibitor in neuroblastoma cells. Taken together, these data showed that NEAT1 negatively regulated cell proliferation and migration of neuroblastoma by the miR-183-5p/FOXP1 axis via suppression of the ERK/AKT pathway. Our findings may provide a new target for the study of pathogenesis and treatment of neuroblastoma.

Objek yang diamati adalah MIN 1 Wonosobo. Data berupa dokumen. Strategi penelitian menggunakan analisis isi. Hasil penelitian menunjukkan KMA Nomor 183 tahun 2019 mengenai perancangan kurikulum di MIN 1 Wonosobo dirancang dengan berbagai pola, diantaranya adalah penggunaan struktur kurikulum yang berlaku pada semua mata pelajaran umum dan Mapel PAI- Bahasa Arab untuk semua kelas. Kurikulum pendukung seperti intrakurikuler, kurikuler dan ekstrakurikuler. Kemudian KMA Nomor 184 tahun 2019 mengenai pedoman implementasi berupa program prioritas, pengembangan diri dan pendidikan kecakapan hidup. Sedangkan untuk implementasi merdeka belajar, MIN 1 Wonosobo belum sepenuhnya menerapkan. Hal ini dibuktikan dari tabel yang menunjukkan banyaknya beban belajar dan mengajar.

Among other microRNA clusters, we previously showed that the miR-183~96~182 cluster (miR-183, miR-96, and miR-182) is abundantly expressed in bovine granulosa cells (bGC) of preovulatory dominant follicles obtained at the follicular phase of the bovine oestrous cycle. Moreover, this miRNA cluster are validated to coordinately target the Fork head O1 (FOXO1), a subfamily of transcription factors that regulate genes involved in cell proliferation, apoptosis, cell cycle arrest, and metabolism. However, the functional involvement of miR-183~96~182 cluster in bGC function by regulation of FOXO1 is not yet determined. Here, we aimed to investigate the function of miR-183~96~182 cluster in bGC using in vitro cell culture model. For this, bGC were aspirated from ovarian follicles (Ø 3–5 mm) obtained from local abattoir. Cells were plated in 24-well plate (2.5 × 105 cells well–1) in DMEM/F-12 (Sigma, Germany) supplemented with 10% FBS (GIBCO, Grand Island, NY) and 1% penicillin/streptomycin (GIBCO) and incubated at 37°C in 5% CO2. Transfection of bGC with miRNA mimics, inhibitors, FOXO1-siRNA, and appropriate controls (Exiqon, Vedbæk, Denmark) was performed using Lipofectamine 2000 (Invitrogen, Carlsbad, CA, USA). Cell proliferation was determined using Cell Counting Kit-8 (CCK-8; Dojindo Molecular Technology, Kumamoto, Japan). Cell cycle distribution was determined with flow cytometric analysis. Total RNA was isolated using miRNeasy mini kit (Qiagen, Hilden, Germany), quantification of target gene was performed using qPCR, and data were analysed using ΔΔCT method. Differences in the mean expression values between treatments were analysed with two-tailed Student’s t-test and statistical significance was defined at P ≤ 0.05. Results showed that a sponge effect was observed upon inhibition in individual miRNA of the cluster, which could be attributed to the partial sequence similarity among cluster members. Both FOXO1 mRNA and protein expression were significantly reduced upon transfection of bGC with miR-183~96~182 cluster mimics, while miR-183~96~182 cluster inhibition increased both FOXO1 mRNA and protein expression. Transfection of bGC with miR-183~96~182 mimics promoted cell proliferation, while inhibition tends to slow down proliferation. Furthermore, the proportion of bGC under G0/G1 arrest markedly declined (P < 0.05), while the S and G2/M phases increased in response to miR-183~96~182 mimicking. Selective knockdown of FOXO1 with FOXO1-siRNA significantly reduced FOXO1 mRNA and protein expression. Interestingly, knockdown of FOXO1 showed similar phenotypic effects such as that of miR-183~96~182 mimics transfection, which resulted in elevated bGC proliferation and reduction in the proportion of cells under G0/G1 arrest. In conclusion, overexpression of miR-183~96~182 cluster promote bGC proliferation and G0/G1 to S and G2/M cell cycle transition through coordinated regulation of genes in the FOXO1 signaling axis.

The objectives of this research were to determine if 1-methylcyclopropene (1-MCP) improves fruit quality and extends shelf life of Ontario greenhouse tomatoes. `Beefsteak' tomatoes between breaker and turning maturity stages were harvested from commercial growers in Leamington, Ontario, and randomly sorted into uniform lots for 1-MCP treatment. Application of 1-MCP concentrations from 0 to 1200 nL/L was done at 22 °C for 12 hours in sealed bags. After treatment, fruit were held at 22 °C. Color change, fruit firmness, and production rates of CO2 and ethylene were followed for a period of 2 weeks. Significant differences (P ≤ 0.05) were found in color, ethylene, and CO2 production rates between treatments. However, there were few significant differences among cultivars and growers. This suggests that different tomato cultivars respond similarly to 1-MCP, and that commercial growing conditions and practices may not affect its efficacy. Over the 2-week ripening period, fruit from treatments of less than 300 nL/L 1-MCP exhibited similar color changes while treatments of more than 600 nL/L resulted in blotchy ripening, causing fruit to be unmarketable. 1-MCP treatment led to an increase in the rates of ethylene and CO2 production, two processes correlated with the onset of fruit ripening. This increase was unexpected and other studies showed that 1-MCP delayed the onset of these processes in tomatoes, and inhibited them in other fruits. Tomatoes treated at a maturity between breaker and turning did not respond well to 1-MCP, perhaps due to the ripening process having already begun. This implies that maturity stages earlier than breaker to turning may respond better to 1-MCP, and it may be more beneficial to target greenhouse tomatoes at an earlier maturity.

Abstract Abstract 2558 Juvenile myelomonocytic leukemia (JMML) is a rare disease of early childhood with a predilection for the monocyte/macrophage lineage. The pathogenesis of JMML is linked to dysregulated signal transduction through the NF1/RAS signaling pathway that is partially caused by genetic mutation of Ras, PTPN11, and c-CBL, or loss-of heterozygosity of Nf1. The hallmark of JMML is that JMML cells are selectively hypersensitive to GM-CSF in vitro. We previously reported that protein deficiencies of PTEN, CREB, and Egr-1 were frequently observed in JMML (67–87%). Recent research indicated that CREB was regulated by miR-34b, and Egr-1 was targeted by miR-183. We hypothesized that microRNAs may play an important role in contributing to the deficiency of these proteins. Using relative-quantitative real-time PCR, we evaluated the expression levels of miR-34b and miR-183 in mononuclear cells from 47 JMML patients. We found that the median level of miR-183 was significantly higher in JMML in comparison to normal controls (median=13.8 vs 4.2, p<0.001); but the median level of miR-34b was only slightly higher in JMML subjects, and not significantly so, compared to normal individuals (median=1.4 vs 1.0, p>0.05). This suggests that miR-34b does not play a significant role in JMML. Since extreme monocyte accumulation is one of the critical characteristics of JMML, we analyzed the correlation between the expression level of miR-183 and the monocyte percentage in the peripheral blood. Strikingly, there was a significant correlation between the expression level of miR-183 and the monocyte percentage in the peripheral blood from 34 patients who had available data (p<0.05). Based on a robust regression analysis, for every unit increase in the square root of RQ miR-183, the monocyte percentage significantly increased by 0.73% (SE=0.32%, p=0.023). This is the first evidence suggesting that microRNAs may contribute to the pathogenesis of JMML. miR-183 may also serve as an important biomarker that can be directly and quantitatively linked to significant clinical parameters in JMML. Using a gemini surfactant ([C18H37(CH3)2-N+-(CH2)3-N+-(CH3)2C18H37]Cl2) as the directing agent, K2SiO3 and KAlO2 as the source of silicon and aluminum, a Nano-crystal has been successfully synthesized under a strong alkali hydrothermal condition. The material shows hexahedron morphology and its crystal size is between around 250-500 nm. Its liquid specific surface area is 1313.2 m2and#183;g-1. The adsorption process of Pb on the Nano-crystal follows quasi-second-order kinetic model and Langmuir adsorption isotherm equation. The maximum Pb adsorption capacity of Nano-crystal is around 1105 mgand#183;g-1.

BackgroundOsteoarthritis(OA) is a common comorbidity in the elderly, characterized by articular cartilage degeneration, hyperosteogeny and synovitis. Long non-coding RNAs (LncRNAs) have been shown to be involved in several human diseases, including OA. However, the effect of NUTM2A-AS1 on chondrocytes remains unknown.ObjectivesThe purpose of this study was to evaluate the role of LncRNA NUTM2A-AS1 in OA pathological changes in vitro.MethodsChondrocyte cells were treated for 24 hours to mimic OA pathological conditions. The experimental center used chondrocytes with interleukin-1 beta (IL-1β) Intervention to simulate the pathological state of OA.The expression levels of LncRNA NUTM2A-AS1, miR-183-5p and TGFA mRNA were detected by quantitative real-time PCR (qRT-PCR), along with CCK-8 to determine cell viability. Inflammatory response was assessed by determining the release of pro-inflammatory factors such as TNF-α and IL-6 using ELISA kits. Cell apoptosis was examined by flow cytometry assay. The binding relationship between miR-183-5p and LncRNA NUTM2A-AS1 or TGFA was confirmed by dual-luciferase reporter assay.ResultsIL-1β induced chondrocytes to express LncRNA NUTM2A-AS1 and TGFA, and the miR-183-5p expression was decreased in IL-1β-treated cells. Low expression of LncRNA NUTM2A-AS1 or TGFA mitigated IL-1β-caused chondrocyte viability reduction and apoptosis promotion. miR-183-5p overexpression alleviated IL-1β-mediated chondrocyte apoptosis and inflammatory injury via decreasing TGFA expression. In addition, our work revealed that miR-183-5p is a target of LncRNA NUTM2A-AS1. Rescue experiments showed that TGFA overexpression could reverse the effects of low expression of LncRNA NUTM2A-AS1 on the pathological changes in IL-1β-induced chondrocytes.ConclusionLow expression of LncRNA NUTM2A-AS1 significantly mitigated the chondrocytes damage induced by IL-1β through regulating miR-183-5p/TGFA axis, which might be an important target to regulate the promotion of OA.References[1]Kong H, Sun ML, Zhang XA, Wang XQ. Сообщается о результатах исследования структурных и электрических свойств гомоэпитаксиального наномасштабного слоя (001) n-GaP, толщиной 70 nm, выращенного методом газофазной эпитаксии из металлоорганических соединений на проводящей сильнолегированной подложке кристалла n-GaP, ориентированного по оси (001). В спектре рамановского рассеяния света такого нанослоя n-GaP в образце n-GaP/n-GaP (001) в сравнении со спектром высокоомного кристаллического образца (001) si-GaP удалось обнаружить две достаточно узкие линии, приписанные колебаниям поперечных TO()- фононов и высокочастотным продольным связанным плазмон-фононным LO()+ колебаниям. При этом установлено, что спектральные параметры LO()+ колебаний как в нанослое n-GaP, так и в подложке (001) n-GaP существенно отличаются между собой, а также и от спектральных параметров линии продольных оптических LO()- фононов. Анализ выявленных строгих количественных особенностей спектральных параметров позволил получить ценную информацию о совершенстве кристаллической структуры наномасштабного гомоэпитаксиального слоя (001) n-GaP. Помимо этого, показано что, численные расчеты на основе микроскопической модели рассеяния света LO()+ колебаниями, обусловленного механизмами деформационного потенциала и электрооптическим рассеянием, позволили бесконтактным и неразрушающим образом определить онцентрацию n и подвижность &mu; свободных носителей заряда гомоэпитаксиального слоя наномасштабной толщины и проводящей сильнолегированной подложки. Полученные значения оказались nhepi=(3.25&#177;0.1)&#183;1017 cm-3 и подвижности &mu;hepi=(40.0&#177;0.1) cm2&#183;V-1&#183;s-1 для гомоэпитаксиального слоя (001) n-GaP в сравнении с nsubs=(2.52&#177;0.1)&#183;1017 cm-3 и &mu;subs=(51.0&#177;0.1) cm2&#183;V-1&#183;s-1 для подложки (001) n-GaP образца n-GaP/n-GaP (001). Nghiên cứu hồi cứu nhằm nhận xét kết quả điều trị phình động mạch não vỡ bằng can thiệp nội mạch hoặc phẫu thuật ở bệnh nhân chảy máu dưới nhện tại Bệnh viện Bạch Mai từ 7/2021 đến 7/2022. Tổng số 183 bệnh nhân, 42,1% (77/183) là nam và tuổi trung bình là 57,27 ± 11,48 năm. Ngoài ra, 45,9% bệnh nhân (84/184) được can thiệp nội mạch và 54,1% (99/183) được phẫu thuật. Có sự khác biệt giữa bệnh nhân được can thiệp nội mạch và phẫu thuật theo phân loại của Liên hiệp Phẫu thuật Thần kinh thế giới (1 [Q1 - Q3: 1 - 1] và 1 [Q1 - Q3: 1 - 2], p = 0,046) khi vào viện. Tỷ lệ máu tụ nhu mô não (1,2% [1/82] so với 20,4% [20/98], p < 0,001) và phình động mạch não giữa (15,5% [13/84] so với 28,3% [28/99], p = 0,038) thấp hơn ở bệnh nhân được can thiệp nội mạch so với phẫu thuật. Tuy nhiên, không có sự khác biệt về kết quả chức năng thần kinh xấu (8,3% [7/84] và 10,1% [10/99], p = 0,681) và tỷ lệ tử vong (2,4% [2/84] và 0,0% [0/99], p = 0,209) ở thời điểm 90 ngày giữa bệnh nhân được can thiệp nội mạch và phẫu thuật.

In this study new chalcone derivative (2E)-3-(1-benzothiophen-2-yl)-1-(4-hydroxyphenyl)prop-2-en-1-one (I), which has important pharmacological applications, was designed and synthesized. IR, NMR and LC-MS/MS empirical methods were used in order to confirm the molecular structure of the synthesized compound I. Both two tautomeric forms of the molecule were optimized by using density functional theory (DFT) method. The structural parameters formed after optimization and structural parameters obtained from similar compounds by X-ray diffraction show a good correlation. The theoretical and experimental results of IR and NMR are generally compatible, but difference arises between some values. This difference shows the existence of O–Hand#183;and#183;and#183;O type intermolecular hydrogen bond. By using its UV-Vis data, the visible absorption maximums of the molecule were analyzed. Compound I was tested against kind of Leishmania major from the point of antileishmanial activity, against kind of Candida albicans from the point of antifungal activity and against fourteen kinds of bacteria from the point of antibacterial activity in this study. The test results showed, that compound I was effective on eight kinds of bacteria and the activity strength on six bacteria among the eight was MIC: 5000 μg/mL and on the rest two kinds MIC: 1250 μg/mL. Besides, it was also found out, that compound I has MIC: 5000 μg/mL antileishmanial activity. Since bacterial and parasitic infections form serious health problems globally, compound I may be a medication candidate in future; if its antibacterial and antiparasitic effect are taken into consideration.

10683 Background: Many chemotherapeutic options exist for metastatic breast cancer (MBC) but there is no standard regimen. We tested a sequential combination regimen using Cisplatinum and Vinorelbine (PVn) followed by Docetaxel as first-line chemotherapy in a phase II clinical trial. This is the first study using a non-anthracycline sequential combination chemotherapy of PVn with Docetaxelin MBC. Methods: Thirty-five patients were enrolled. Cisplatin 80 mg/m2, which was given on day 1 and Vinorelbine 30 mg/m2 on days 1 and 8 q3 weeks for 4 cycles. After the 4th cycle, responding patients received Docetaxel 75 mg/m2 every 21 days for a maximum of 4 cycles. Patients were evaluated every two cycles and those who had no response were offered other therapy. Patients who showed progression after 2 cycles of PVn were offered Docetaxel but were considered off protocol. Three patients were excluded from analysis because of death unrelated to treatment. Results: After a median follow up of 14 months (range 1–36), 32 patients (91.4%) completed the study. Overall response rate was 53.1%. Complete remission was seen in 5 pts (15.6%), partial response in 12 patients (37.5%), stable disease in 6 patients (18.75%), and progressive disease in 9 patients (28.1%). 20 patients (62.5%) were alive at 12 months. Median time to disease progression was 8 months (range 1–24). At 24 months 37.5% (12 patients) were alive. Response rates to PVn were not significantly improved by sequential addition of Docetaxel. A total of 183 cycles were administered with a median of 6 cycles per patient. Delay of chemotherapy because of Grade II neutropenia occurred in 41/183 cycles (22.4%). Febrile neutropenia was observed in 4 patients (2.2%). Grade II anemia was observed in 40/183 cycles (21.8%) and Grade III in 24/183 cycles (13.1%). Grade II nephrotoxicity occurred in 12 cycles (6.5%) and Grade III vomiting was observed in 31/183 cycles (16.9%). No treatment-related mortality has been reported. Conclusions: Sequential PVn-Docetaxel is a feasible and effective non-anthracycline option as first-line chemotherapy in patients with MBC. More effective Docetaxel may be better used upfront and followed by PVn. This regimen has acceptable toxicity as well. No significant financial relationships to disclose.
