The demand for new materials that provide excellent structural performance while reducing weight and being cost-effectively manufactured is increasing. For applications with high strength requirements, ultra-high strength steels (UHSS) have been widely used. However, with such a high strength level, UHSS are very sensitive to the hydrogen that could be ease by the tempering process. In this research, the correlation of hydrogen and tempering process on commercial UHSS 15B30 has been studied. Results show that the tensile strength (TS) of as-quenched 15B30 is about 1900MPa. After tempering treatment of the quenched 15B30, the TS decreases from 1600MPa to 1200MPa with tempering temperature increased from 200°C to 400°C. The 15B30 specimens, being subjected to hydrogen charge, exhibit the dramatic reduction of mechanical strengths.

The West Runway site (15BE391) is located in an upland setting in Boone County, Kentucky. Excavations at this locality revealed intact Early Woodland deposits including pit features containing Fayette Thick ceramics and straight stemmed Kramer projectile points. Until these investigations, Fayette Thick ceramics had not previously been recovered from non-mound locations or in isolation within undisturbed feature contexts. Several radiocarbon determinations bracket the primary occupation of the West Runway site between 770 B.C. and 390 B.C. These dates indicate that the site predates the well-known Adena culture and suggest a need for continuing refinement of the Early Woodland cultural sequence.

ABSTRACT The atypical hemolytic Listeria innocua strains PRL/NW 15B95 and J1-023 were previously shown to contain gene clusters analogous to the pathogenicity island (LIPI-1) present in the related foodborne gram-positive facultative intracellular pathogen Listeria monocytogenes, which causes listeriosis. LIPI-1 includes the hemolysin gene, thus explaining the hemolytic activity of the atypical L. innocua strains. No other L. monocytogenes-specific virulence genes were found to be present. In order to investigate whether any other specific L. monocytogenes genes could be identified, a global approach using a Listeria biodiversity DNA array was applied. According to the hybridization results, the isolates were defined as L. innocua strains containing LIPI-1. Surprisingly, evidence for the presence of the L. monocytogenes-specific inlA gene, previously thought to be absent, was obtained. The inlA gene codes for the InlA protein which enables bacterial entry into some nonprofessional phagocytic cells. PCR and sequence analysis of this region revealed that the flanking genes of the inlA gene at the upstream, 5′-end region were similar to genes found in L. monocytogenes serotype 4b isolates, whereas the organization of the downstream, 3′-end region was similar to that typical of L. innocua. Sequencing of the inlA region identified a small stretch reminiscent of the inlB gene of L. monocytogenes. The presence of two clusters of L. monocytogenes-specific genes makes it unlikely that PRL/NW 15B95 and J1-023 are L. innocua strains altered by horizontal transfer. It is more likely that they are distinct relics of the evolution of L. innocua from an ancestral L. monocytogenes, as postulated by others.

<strong data-sider-select-id="23310f00-5dfa-480a-a4ff-ada7c69c0c78">Background and aims: Studies have indicated a considerable change in the trend of families’ adaptation to the hearing impairment of their children following the application of hearing screening programs in the past few years. This study was conducted in line with increased rehabilitation service quality and family-based intervention. It was performed to identify factors affecting the adaptation of mothers to hearing-impaired (HI) children with cochlear implants. <strong data-sider-select-id="15b3b513-cd15-4b53-8df8-f7a27ec30b17">Methods: This qualitative-descriptive research was performed with a phenomenological approach and involved 16 mothers of two to seven-year-old HI children with cochlear implants. The research setting was the centers for speech therapy of HI children in Shiraz, Iran. Data were collected through in-depth interviews with the subjects, and data analysis was performed in MAXQDA (2018) using Colaizzi’s seven-step method. <strong data-sider-select-id="f166576e-379c-4e0c-a498-612f4d8a6ac7">Results: The mean age of the subjects was 34 years, and the children of these parents were two to seven years old. The mean age of the earliest known diagnosis was 12 months. Moreover, the mean age at cochlear implantation was 2.5 years. Based on the subjects’ experiences, the researchers extracted factors affecting mothers’ adaptation, leading to four main themes. These factors facilitated mothers’ adaptation and included the quality of medical services, condition normalization, individual capabilities of mothers, and effective use of the environment. <strong data-sider-select-id="c47ad3f8-8f76-4323-9e3f-13eec76bdee1">Conclusion: Improving the quality of medical services, condition normalization, individual capabilities of mothers, and effective use of the environment led to an improvement in mothers’ adaptation to HI children with cochlear implants.

In dengue virus (DEN) particles, the core protein is a structural protein of the nucleocapsid. The core protein is known to be present in the nucleus of DEN-infected cells but there have been conflicting reports as to whether it is also present in the nucleolus. To clarify this, the intracellular location of the core protein was examined using a monoclonal antibody, 15B11, which was produced in this study. Immunofluorescence staining with this antibody demonstrated that the core protein first appeared in the cytoplasm and then in the nuclei and nucleoli of infected cells. Nuclear localization of the core protein was determined to be independent of other DEN proteins, since recombinant core proteins still entered the nuclei and nucleoli of cells transfected with only the core protein gene. Three putative nuclear localization signal motifs have been predicted to be present on the core protein. Deletion of the first one (KKAR), located at aa 6–9, and mutation of the second one (KKSK), located at aa 73–76, did not eliminate the nuclear localization property of the core protein. The third motif with a bipartite structure, RKeigrmlnilnRRRR, located at aa 85–100, was determined to be responsible for the nuclear localization of the core protein, since the core protein without this motif was located exclusively in the cytoplasm of DEN-infected cells and that this motif mediated nuclear localization of a normally cytoplasmic protein.

This study investigated strain-rate sensitivity (SRS) in an as-extruded AZ31 magnesium (Mg) alloy with grain size of about 10 mm. Although the alloy shows negligible SRS at strain rates of >10-5 s-1 at room temperature, the exponent increased by one order from 0.008 to 0.06 with decrease of the strain rate down to 10-8 s-1. The activation volume (V) was evaluated as approximately 100b3 at high strain rates and as about 15b3 at low strain rates (where b is the Burgers vector). In addition, deformation twin was observed only at high strain rates. Because the twin nucleates at the grain boundary, stress concentration is necessary to be accommodated by dislocation absorption into the grain boundary at low strain rates. Extrinsic grain boundary dislocations move and engender grain boundary sliding (GBS) with low thermal assistance. Therefore, GBS enhances and engenders SRS in AZ31 Mg alloy at room temperature.

If the customer accesses any portal of organization website, the log is generated in web server, based on sequence of user transaction. The weblog stored in the web server as unstructured format, it contains both positive and negative responses i.e. successful and unsuccessful responses, identifying the positive and negative response is not useful for identifying user behavior of individual user. Initially the successful response is taken, from that conversion of unstructured log format to structured log format through data preprocessing technique. The process of data preprocessor contains three step process data cleaning, user identification and session identification. The pattern is discovered by preprocessing technique from that user navigation pattern is generated. From that navigation pattern classifier technique is applied, the conversion of sequence pattern to sub sequence pattern by clustering technique. This research is to identify the user navigation pattern from weblog. The Improved Spanning classification algorithm classifies the frequent, infrequent and semi frequent pattern. To identify the optimal webpage using classificatopn algorithm from thet user behavior is identified.</span></span></p>

ABSTRACT Reverse transcriptase (RT) activity has been detected recently in all chicken cell-derived measles and mumps vaccines. A study of a vaccine manufactured in Europe indicated that the RT is associated with particles containing endogenous avian retrovirus (EAV-0) RNA and originates from the chicken embryonic fibroblasts (CEF) used as a substrate for propagation of the vaccine. We investigated the origin of RT in measles and mumps vaccines from a U.S. manufacturer and confirm the presence of RT and EAV RNA. Additionally, we provide new evidence for the presence of avian leukosis virus (ALV) in both CEF supernatants and vaccines. ALV pol sequences were first identified in particle-associated RNA by amplification with degenerate retroviralpol primers. ALV RNA sequences from both thegag and env regions were also detected. Analysis of hypervariable region 2 of env revealed a subgroup E sequence, an endogenous-type ALV. Both CEF- and vaccine-derived RT activity could be blocked by antibodies to ALV RT. Release of ALV-like virus particles from uninoculated CEF was also documented by electron microscopy. Nonetheless, infectivity studies on susceptible 15B1 chicken cells gave no evidence of infectious ALV, which is consistent with the phenotypes of theev loci identified in the CEF. PCR analysis of ALV and EAV proviral sequences in peripheral blood mononuclear cells from 33 children after measles and mumps vaccination yielded negative results. Our data indicate that the sources of RT activity in all RT-positive measles and mumps vaccines may not be similar and depend on the particular endogenous retroviral loci present in the chicken cell substrate used. The present data do not support transmission of either ALV or EAV to recipients of the U.S.-made vaccine and provide reassurance for current immunization policies.

Ingevolge artikel 15B(3)(a)(i)(aa) van die Wet op Deeltitels mag die registrateur nie oordrag van ’n deeltiteleenheid registreer nie tensy ’n transportbesorger bevestig dat die regspersoon gesertifiseer het dat alle gelde verskuldig ten opsigte van die bedoelde eenheid betaal is of dat sekerheidstelling vir betaling tot bevrediging van die regspersoon verskaf is. By die eksekusieverkoping in die Marsh Rose-saak het die koper, Steinmuller, onderneem om sekerheidstelling te verskaf dat alle gelde verskuldig aan die regspersoon deur die vorige eienaar deur hom betaal sal word. Toe Steinmuller en die regspersoon nie kon ooreenkom oor die bedrag van die sekerheidstelling nie, nader Steinmuller die hof a quo vir ’n toepaslike bevel en die hof beslis dat Steinmuller R250 000 in die trustrekening van sy prokureur moet betaal as onherroeplike sekerheidstelling vir betaling van die geld verskuldig aan die regspersoon. Op appèl na die volbank van die hooggeregshof in Johannesburg, bevestig die meerderheidsuitspraak dat die hof moet besluit of die regter a quo bevoeg was om te bepaal dat skuld wat aan Steinmuller gedelegeer is, slegs agterstallige heffings insluit. Volgens die minderheiduitspraak moet die bedrag wat as sekerheidstelling aangebied word nie slegs agterstallige heffings nie, maar álle gelde verskuldig deur die vorige eienaar insluit. Volgens die minderheidsuitspraak berus hierdie standpunt op ’n algemene aanspraak op sekerheidstelling en die feit dat die regspersoon geregtig is op alle gelde vir sy finansiële voortbestaan. Daar word aangetoon dat hierdie aanspraak slegs betrekking het op gevalle waar die eienaar van die eenheid insolvent is en alle gelde ingevolge artikel 89(1) van die Wet op Insolvensie as deel van die koste van tegeldemaking aan die regspersoon uitbetaal moet word, voordat oordrag kan geskied. Die meerderheidsuitspraak is korrek dat slegs die verskuldigde skuld van die vorige eienaar aan Steinmuller gedelegeer is. Wat regskoste betref beslis die meerderheidsuitspraak dat slegs regskoste wat getakseer is deur die regspersoon of waarop die partye ingevolge reël 25(4) van Aanhangsel 1 van die Regulasies op die Wet op die Bestuur van Deeltitelskemas ooreengekom het, afgedwing kan word. Die meerderheidsuitspraak beslis verder dat rente op agterstallige heffings en nie-betaalde regskoste slegs verskuldig is indien dit bevestig word deur ’n skriftelike trusteebesluit ingevolge reël 21(3)(c) van Aanhangsel 1 van die Regulasies op die Wet op die Bestuur van Deeltitelskemas. Ten slotte word aangevoer dat die regspersoon in plaas van die hofaansoek, die goedkoper en spoediger proses ingevolge artikel 39(1)(c) van die Wet op die Ombuddiens vir Gemeenskapskemas kon gevolg het. Volgens daardie proses sou die beregter moes bepaal of die skuld gedelegeer aan Steinmuller verkeerdelik bepaal is of onredelik was en indien wel, dit tot die korrekte bedrag aanpas.

<p><strong>Background: </strong>Clavicle fracture is a common injury involving upper limb. It accounts for 2.6-5% of injuries. Most common mechanism causing this injury is a fall on an outstretched hand. Clavicle fractures have a bimodal distribution with 1<sup>st</sup> peak in young active adult males less than 30 years of age. Second peak in elderly females with osteoporotic bones. The annual incidence is highest in male under 20 age group, decreasing with subsequent age groups. Road traffic accidents is another mode of injury which is becoming increasingly common for mid shaft clavicle fractures.</p><p><strong>Methods</strong>: A prospective study was carried out over 12 months. A total of 126 patients met the inclusion criteria. The epidemiological data was collected at the time of presentation using standard case sheet proforma. Radiographic assessment was done for fracture classification.</p><p><strong>Results</strong>: Mid Shaft clavicle fracture was most common in middle aged males (31-40 years) with right side being most commonly affected. Road traffic accidents was the most common cause of injury (62%). AO/OTA type 15B1 is the most common fracture morphology (45%). Most of the high energy road traffic accidents are associated with 15B3 type fractures while type 15B1 is more common in low energy mechanisms like ground level fall.<strong></strong></p><p><strong>Conclusions: </strong>Mid shaft clavicle fracture is a common injury in young population. It is increasingly being associated with road traffic accidents. The number of comminuted fractures, which represent high energy injury mechanism has been on the rise due to increase in high velocity accidents.<strong></strong></p>

Las herramientas de la agroindustria están sometidas a condiciones severas de desgaste en su uso. Actualmente se utiliza como material base para la producción de las mismas el acero 15B30 en el estado de templado y revenido. Cuando la aplicación lo requiere, se recubren estas herramientas con compuestos duros. Las aleaciones de matriz metálica base Fe son muy utilizadas como recubrimientos debido a su abundancia y a sus buenas propiedades mecánicas. Cuando se le agregan partículas duras, se las denominan compuestos de matriz metálica reforzada. El refuerzo puede ser añadido intencionalmente o formado durante la consolidación de la capa. En el presente estudio se diseñaron 4 compuestos de matriz metálica base Fe reforzada. Se realizaron las distribuciones granulométricas de los polvos de partida. Luego se caracterizaron los 4 recubrimientos depositados sobre sustratos de acero 15B30 mediante microscopía óptica, microdureza y dureza. Los resultados del trabajo permitieron ajustar composiciones químicas.

Abstract One of the steels commonly used in the industry in mechanical devices, tools, and structural steels is SAE 1045. A possible replacement for this alloy is SAE 15B30 with similar mechanical properties and has been used in the automotive and construction industry. To select an alloy, care is needed, for example, weldability. Making a comparative study of these alloys, can boron steel replace SAE 1045? Knowing that boron steels have high hardenability but may have improved weldability due to the low carbon equivalent. Therefore, it is extremely important to know the factors that can interfere with the quality of the final product after the welding processes, as the mechanical properties and microstructures in the heat-affected zone (HAZ) may change. Knowing that in the automotive and civil sector we have several welding processes, it is extremely important to know the results of a boron steel after a welding process; thus, this work intends to compare the weldability of SAE 15B30 and SAE 1045 welded by shielded metal arc welding (SMAW) and also to analyze the microstructural changes after welding by optical microscopy, scanning electron microscopy, and its relationship with hardness using the test of Vickers microhardness. To perform the welds, both in SAE 1045 steel and in SAE 15B30 steel, the butt joint with a V-shaped chamfer was chosen. The results showed that the boron steel, with high hardenability, obtained a microstructure with bainite in the coarse-grained heat-affected zone (CGHAZ), unlike SAE 1045, which presented pearlitic microstructure. In addition, the hardness of boron steel increased by approximately 90% over the base metal (BM), and the hardness of SAE 1045 increased close to 50% over the BM.

Abstract In this work, the heat transfer coefficients were determined utilizing an experimental&#xD;study for cooling a 15B35H boron steel, using aqueous solutions of polyalkylene&#xD;glycol (PAG) as a cooling fluid. A cooling tank was designed and built to allow&#xD;the fluid recirculation, where the effect of agitation and the PAG content in a&#xD;concentration range of 2-6% vol. were analyzed. The thermal histories of cylindrical&#xD;probes instrumented with K-type thermocouples were obtained, and the heat transfer&#xD;coefficients associated with the types of refrigeration were calculated, solving the IHCP.&#xD;The results show the presence of two maximum surface heat flux denominated q1max for&#xD;the low-temperature range and q2max for the high-temperature range. Getting higher&#xD;q1max and low q2max is desirable to avoid distortion and cracking. It was found that 4%&#xD;PAG exhibits a slight variation of heat flux values in both low and high-temperature&#xD;regions, regardless of the degree of agitation, maintaining values of about 3 MW·m-2&#xD;for q1max and 5.5 MW·m-2 for q2max . In this case, q2max is higher than q1max, leading&#xD;to higher cooling rates in the martensitic transformation zone, increasing the risk of&#xD;distortion and cracking. In the cases of 2 y 6% PAG, agitation affects q1max ranging&#xD;from 2.5 to 6 MW·m-2, and for q2max from 4 to 6 MW·m-2, where q1max is always&#xD;higher than q2max, providing better conditions for quenching.

ABSTRACT A key challenge in managing transmissible spongiform encephalopathies (TSEs) or prion diseases in medicine, agriculture, and wildlife biology is the development of practical tests for prions that are at or below infectious levels. Of particular interest are tests capable of detecting prions in blood components such as plasma, but blood typically has extremely low prion concentrations and contains inhibitors of the most sensitive prion tests. One of the latter tests is quaking-induced conversion (QuIC), which can be as sensitive as in vivo bioassays, but much more rapid, higher throughput, and less expensive. Now we have integrated antibody 15B3-based immunoprecipitation with QuIC reactions to increase sensitivity and isolate prions from inhibitors such as those in plasma samples. Coupling of immunoprecipitation and an improved real-time QuIC reaction dramatically enhanced detection of variant Creutzfeldt-Jakob disease (vCJD) brain tissue diluted into human plasma. Dilutions of 1014-fold, containing ~2 attogram (ag) per ml of proteinase K-resistant prion protein, were readily detected, indicating ~10,000-fold greater sensitivity for vCJD brain than has previously been reported. We also discriminated between plasma and serum samples from scrapie-infected and uninfected hamsters, even in early preclinical stages. This combined assay, which we call “enhanced QuIC” (eQuIC), markedly improves prospects for routine detection of low levels of prions in tissues, fluids, or environmental samples. IMPORTANCE Transmissible spongiform encephalopathies (TSEs) are largely untreatable and are difficult to diagnose definitively prior to irreversible clinical decline or death. The transmissibility of TSEs within and between species highlights the need for practical tests for even the smallest amounts of infectivity. A few sufficiently sensitive in vitro methods have been reported, but most have major limitations that would preclude their use in routine diagnostic or screening applications. Our new assay improves the outlook for such critical applications. We focused initially on blood plasma because a practical blood test for prions would be especially valuable for TSE diagnostics and risk reduction. Variant Creutzfeldt-Jakob disease (vCJD) in particular has been transmitted between humans via blood transfusions. Enhanced real-time quaking-induced conversion (eRTQ) provides by far the most sensitive detection of vCJD to date. The 15B3 antibody binds prions of multiple species, suggesting that our assay may be useful for clinical and fundamental studies of a variety of TSEs of humans and animals.

Introduction: Acute postoperative heart failure (PoHF) is encountered in 20-35% of elderly patients after cardiac surgery but predictors are not well defined. Circulating microRNAs (miRNAs) predict HF or postoperative atrial fibrillation, yet, their role in identifying those at risk for PoHF with preserved LV function and gender-related differences is not known. Hypothesis: miRNAs involved in CVD can identify patients who develop PoHF in gender-specific manner. Methods: Preoperative blood samples from patients with preserved LV function undergoing cardiac surgery with no prior history of HF, supraventricular, or ventricular tachycardia was used for RNA isolation. Differences in relative plasma levels of miRNA between PoHF vs. No-PoHF were assessed using qPCR. Univariate and multiple logistic regressions were performed to assess risk factors for PoHF. Results: Out of 68 patients (mean age 68.3±12.4), 13 (19%) developed PoHF (54% males; mean age 64.1±15.1; p=0.23), while 55 (51% males; mean age 69.3y ± 11.6) remained free of PoHF. Although all patients had preserved LV function, those who developed PoHF had lower LVEF, (52 vs 58; P<0.05). No significant differences were observed in the prevalence of hypertension, diabetes, myocardial infarction, heart failure, stroke, COPD, or the use of cardiac medications. Out of 13 miRNAs analyzed, stepwise discriminant analysis showed miR-423 (11-fold), -187 (6-fold), -26a2 (4-fold), and -15b (-1.9-fold) were significantly (p<0.05) altered in patients who developed PoHF. When miRNAs were assessed for gender differences, levels of only four miRNAs [miR-423(P<0.01), -23b2(p<0.05), -15b1(p<0.01), and -29b (p<0.05)] principally involved in TGFβ1-SMAD 2/3 regulated extracellular matrix (ECM) turnover pathways were significantly altered in male but not in female patients who developed PoHF compared to No-PoHF group. Conclusions: The differences in preoperative miRNA expression in pathways involved in ECM turnover in male patients who develop PoHF is suggestive of pathophysiological substrate differences between males and females and identifies a group at risk of PoHF with preserved LV function.
