Ce memoire presente l'etude experimentale de reactions neutre-neutre (molecule-radical et radical-radical) sur un grand intervalle de temperature allant de la temperature ambiante jusqu'a des temperatures extremement basses de l'ordre de 10 k. Ce type de reaction est tres important d'un point de vue astrochimique, dans l'etude d'atmospheres planetaires et dans la confrontation avec la theorie. La technique c. R. E. S. U. (cinetique de reaction en ecoulement supersonique uniforme) permettant d'avoir un ecoulement uniforme de temperature tres basse et la technique plp-lif (pulse de photolyse laser-fluorescence induite par laser) ont permis les premieres etudes de reactions avec les radicaux cn, oh et ch a des temperatures aussi basses que 13 k. Les coefficients de vitesse des reactions de cn avec o#2 et de cn avec nh#3 croissent de facon monotone quand la temperature diminue. Ces mesures confrontees a la theorie montrent que l'on doit inclure, en plus des interactions a longue portee, celles a courte portee. Les premieres etudes de la reaction du radical oh avec trois butenes sont utiles pour une meilleure comprehension des pollutions de l'atmosphere terrestre. La forte dependance negative en temperature du coefficient de vitesse de la reaction du radical oh avec hbr a fait apparaitre une sous estimation du coefficient de vitesse aux temperatures regnant dans la stratosphere terrestre. Ceci a une incidence dans le cycle de l'ozone. Les premieres mesures du coefficient de vitesse de la reaction de oh avec no en fonction de la temperature et de la pression sont utiles dans l'etude d'atmospheres planetaires, notamment celle de la terre. Les premieres etudes de reactions entre le radical ch et plusieurs molecules et radicaux neutres indiquent que ch reagit rapidement a tres basse temperature. L'etude spectroscopique des radicaux cn et oh a montre que l'ecoulement supersonique presente un reel equilibre thermodynamique

Title: Adherences to Motoric Activities of People with a Total Knee Arthroplasty Objective: Based on availabe information analyze possible motoric activities of people with a total knee arthroplasty. To determine their physical possibilities and use. Based on the indicated data prepare and aply excercising program of algorithmic series. Method: We employed the form of "Case - Graduation Thesis" with application of a model motoric program. The data were gained by examination of the locomotive apparatus, motoric stereotypes, and a public inquiry. Results: During the realisation of the exercise program of algorithmic series, stimulation and activation of a locomotive apparatus were reached. Activities connected with a physical effort positively influenced psyche in connection with managing more everyday activities. Adherence to motoric activities of the monitored people was diverse. Many researches deal with a wide range of cause of anxiety disorders, which may include various untreated traumas, physiological causes, cognitive distortion or learned reactions. Anxiety disorders tend to create so-called the vicious circle of stress and anxiety that closes the patients, leads them to social isolation and gradual loss of life certainty. Anxiety disorders are among the most common mental illness. Major types are introduced in the thesis and they are connected with the concepts of self- compassion, shame-proneness and guilt-proneness, which have been studied abroad in recent years. The research part then brings the results of the role of self-compassion, shame- proneness and guilt-proneness in experiencing clinical anxiety, confirming the assumption that patients suffering from anxiety show a significantly lower level of self- compassion, a higher shame-proneness and internalized shame. Guilt-proneness does not show a relation with self-compassion. Details are discoussed. Key words: anxiety self-compassion shame internalized shame guilt

THE ABSTRACT OF THE DIPLOMA THESIS The Name of the Diploma Thesis: The motivation to lose the habit of smoking in patients terated for some other kind of addiction Abstract: Starting point: The prevalence of smoking in people addicted to other narcotics is usually 2 - 3-times higher than in common population. The same percentage of people addicted to tobacco is among patients treated in some in-patient treatment for people addicted to narcotics or gamblers. Simultaneous treatment of addiction to tobacco and other kinds of addiction is possible and is recommended. It seems to be also effective and favourable for treatment of other patient's addictions - it supports the improvement of physical and mental condition. The interest in treatment of addiction to tobacco often appears in patients. But the question is how much they are motivated and informed by the staff. There is no integrated schedule focused on breaking the habit of smoking in in-patient clinics for addiction treatment. The successful osteoporosis therapy is based on adequate supply of calcium and vitamin D. A lot of studies are focused on the adherence to antiresorptive treatment but the adherence to supplementation therapy is rarely evaluated. Aims: The aim of the study was to evaluate the adherence to calcium and vitamin D supplementation in women with postmenopausal osteoporosis. Methods: The adherence was evaluated using two different methods: self-reported questionnaire and the electronic monitoring by bottles type Medication Event Monitoring System (MEMS). This partial analysis of a multicentre study includes and assesses data from Osteocentre in Pardubice Regional Hospital. Results: The analysis was performed with 15 women (the average age of 73). The dissertation entitled ?Health Related Quality of Life (HRQoL) in Dialysed Patients with Respect to Age? contains a theoretical and practical part. The theoretical part describes chronic kidney disorder and the treatment of this disease. It mentions the causes, repercussions and complications of this disease. It also suggests the form of treatment as the method of replacing the function of the kidneys which includes haemodialysis, peritoneal dialysis and transplantation. I have attempted to elaborate their principle, history and current form, benefits, negative aspects and complications. The practical part describes the actual research. Health Related Quality of Life (HRQoL) was ascertained in a group of patients undergoing haemodialysis at one dialysis centre. Questionnaire SF 36 was used which allows for a quantified assessment of the quality of life in eight domains. The results were statistically assessed and clearly depicted using tables and graphs. The quality of life was compared in the monitored group with the general population and also based on age and gender. The results found show a reduced quality of life of dialysed patients compared with the standard population. A difference was also found in the assessment of the quality of life in some age groups. No difference in results was proved with regard to gender.

The dissertation thesis deals with the skill of listening comprehension and using multimedia for its support in the French language teaching process. The issue is studied within the context of Czech curricular documents. It classifies language skills from the point of view of the Common European Framework of Reference for Languages. The process of listening comprehension is described in linguo-didactics from the historical point of view and as a process of language communication. Attention is paid especially to listening comprehension in foreign language teaching. The thesis focuses also on methodical procedures when developing and reinforcing this language skill. Integration of multimedia with respect to varied learning styles and strategies is described. Styles according to the perception preferences and listening learning strategies are focused on. The aim of the thesis is to find out what is the position of the listening comprehension skill in the French language teaching process at upper-secondary schools, what is the success rate of the students in the process of comprehension of the spoken language, whether multimedia can contribute to successful comprehension of the spoken language in a foreign language. Title: Changing attitudes of primary school pupils to integration after completing a two- month "Life with Disabilities" programme. Objective: The main aim of the thesis is to explore various possibilities of influencing primary school pupils attitudes to integration of a pupil with disabilities into physical education lessons. Method: Using the CAIPE - CZ and Adjective - Checklist questionnaires a qualitative- quantitative research has been applied to observe a change in primary school pupils' attitude to integration into the physical education lessons once a two-month "Life with Disabilities" programme was completed. A series of topical areas has been presented not only in health education lessons, but also in physical education lessons. The programme also included several sporting events taking place outside the school building and a Paralympic school day. A two-group quasi-experiment research group was formed by the 7th (headcount: 20; average age: 12) and 9th (headcount: 24; average age: 15) years of primary school. Human immunodeficiency virus (HIV) is globally spread virus without available cure. Since its life-long presence, virus is carefully monitored as well as patient's immunological status. Replicative fitness of the virus is one of important aspects which can be taken into account, when monitoring HIV. Here, we are measuring HIV replicative fitness of gag recombinant viruses and comparing the results with replicative fitness of primary isolates. Further, we are comparing our findings of replicative fitness change over time with disease progression in the patient. We found that gag can be major contributor to overall fitness, although not in all cases. Additionally, we observed a correlation of replicative fitness development and slope of patient's CD4+ T cells. Moreover, this relation was even more noticeable in patients with slow disease progression or in carriers of protective alleles. In summary, our results extend the understanding of replicative fitness and its role in disease progression; and pave the way to use the recombinant HIV for replicative fitness measurement in clinical practice. This type of cancer is divided into two subtypes, the small cell and non-small cell lung cancer. Platinum- based chemotherapy is the mainstay of treatment of advanced stages non-small cell lung cancer, to that is this work focused on. The aim of this study was to investigate whether it is possible to use the comet assay method for monitoring the progress of treatment in these patients, and if it is possible to predict a patient's response to treatment or patient's survival. This study proposes a measurement and assessment model for the construction productivity of freeway reconstruction project, according to on the previous studies of Thomas and the characteristics of the freeway reconstruction project. The model is used on the cases of the freeway No. 1 reconstruction projects and No. 2 new construction project in Taiwan to realize and compare their respective influence factors of the construction productivity. When the model evaluates the threshold construction productivity of the embankment in the cases, the estimate values for the reconstruction projects and the new construction project are 0.048~0.052 MH/ M 3 and 0.025 MH/ M 3 , respectively. In addition, the influence factors in the reconstruction projects and between the reconstruction projects and the new construction project are introduced, leading to the arrangement of improvement methods. The results based on this study can support the use of the model to various constructions to completely establish the threshold construction productivity in construction engineering for the further use of the cost analysis and construction management. The working content factor in the model, however, should be further studied in a future to obtain a better result.

The thesis tries to describe, evaluate and analyse some negative aspects of social support provided to the Romanies who live in a socially excluded locations (considering the concrete situation in Písek) and suggest suitable measures how to improve it. Although this thesis is conceived as theoretical, for completion it contains views of social workers from Písek who provide social support to the Romanies, and in case reports, there are mapped social nets and experiences of small sample of the Roma residents from Písek´s socially excluded locations.

Abstrakt anglicky Abstract in english Abstract Chronic tromboembolic pulmonary hypertension (CTEPH) is believed to result from acute pulmonary embole. It manifests itself in the growth of blood pressure in pulmonary circulation. This condition, if not treated, can lead to the failure of right heart. The treatment of CTEPH is the surgical pulmonary endarterectomy, which consists in enucleation of organized fibrotic thrombus with the part of the vascular arterial wall. The prerequisite for the successful surgery is the selection of suitable candidates, mostly the patients with the central location of disability and the absence of the peripheral vascular remodelation. The aims of this study were to examine the relationship between symptoms of ADHD (Attention deficit/Hyperactivity disorder) and motivation for change, to explore their role in the prediction of treatment completion and to identify domains in psycho-social characteristics which can affect the motivation. The sample was composed of 250 illicit-subsatnce users, treated in therapeutic communities for drug addicts. Motivation for change was assessed with the Change Questionnaire, the presence of ADHD symptoms was assessed with DIVA2.0 (Diagnostic interview for Adult ADHD). Respondents indicated strong motivation to change the problematic behavior, but the study struggled with high rate of premature dropping out of the treatment, so the hypothesis about the effect of measured motivation for successful completing of the tretment could not be confirmed or disconfirmed. The relationship between ADHD and motivation could not be confirmed as well. The most important finding is probably the positive correlation between rate of psychopathology and ADHD, and it's negative influence on motivation. Work title: Use ofPsychological Instruments for Evaluation ofthe Parameters Influencing the Adherence to Physical Activity in the Case of Overweighted and Obese Persons. Work objectives: 1. To review a possibility to use three psychological questionaries and other parameters (BMI, age, sex) to identify the respondents with lowered adherence to physical activities. 2. To evaluate physical activity influence on some psychological aspects and eating behaviour ofoverweighted and obese persons. Methodology: With help of these three types of standardized questionaries (BDI-II, WHOQOL-BREF a TFEQ) it was evaluated a scale of depresivity, quality of life and eating behaviour in the case of persons with overweight and obesity at the begining of three-month physical activity programme. Input values from the questionaries from respondents, who had not finished the programme were compared with the values of the respondents, who had finished, and statisticaly evaluated. Consequently, there were compared and statisticaly evaluated input and output values from the quetionaries and BMI of the respondents, who had finished the programme. This diploma work studies quality of life of patients suffering from Bechtěrev disease (Ankylosing Spondylitis), who had global four-weeks cure in Berta Health Spa in Třeboň from 1st January to 31st July 2006. Hypothesis says that six months after the end of balneotherapy the quality of life of these patients will be higher than before. For confirmation or confutation of this hypothesis there was used quantitative method of research, in form of adjusted questionnaire for detection of quality of Life by SEIQoL concept (Schedule for the Evaluation of Individual Quality of Life). By this measure respondents are allowed to nominate five areas of life (aims of life), which are most important, together with percentage rate of their satisfaction with each and the relative importance of each. Average life-quality index of the respondents before balneotharapy was 60,4 %, six months after the end of balneotharapy it was 62,7 %. Thus six months after the end of the cure the respondents have higher quality of life. Then there were mentioned and discussed differences in perception of quality of life of various age groups and of gender. There was also accomplished order of the most often mentioned aims of life, as well an analysis of ``health{\crqq} as alife aim was done. This diploma work could be used in cultural work and for improvement of informedness in problematic of life quality. But mainly, it could be perhaps taken as a little controversial report to the discussion about using of assorted exploratory concepts for evaluation of quality of life.

The main topic of this study is the Czech civic sector and its organizations. The study is based on the comparison of selected non-governmental non-profit organizations and its funding. The goal is to collect and compare sources from which were these organizations financed in the last few years, to notice possible tendencies for change and to outline potential trends. For the comparison were chosen several "general beneficial organizations", each from the different area of interest. Proposed thesis is that the Czech non-governmental non-profit organizations are relatively dependent on the funding from the public sources and they fight with lack of finances from other sources and therefore are endangered in their economic stability and often also in their mission. Before the comparison part there is a theoretical part which draws on available material about the current situation in the Czech civic sector, management of non-profit organizations, financial management and also on relevant available statistics. This theoretical material is supplemented with interviews with chosen professionals in this branch in order to achieve up-to-dateness of information. Malignant lymphoproliferative disorders include highly heterogeneous entities, i.e. lymphomas (Non-Hodgkin - NHL, as well Hodgkin's lymphoma), lymphoid leukemias, multiple myeloma and others. As currently many chromosomal aberrations with diagnostic and prognostic significance are known, molecular cytogenetic analyses of tumor cell genome has become a substantial examination also in lymphoproliferative disorders. This thesis focuses primarily on chronic lymphocytic leukemia (CLL), which is one of the mature B-cell neoplasms and represents the most common type of leukemia. We analyzed four most frequently found aberrations (13q14 deletion, ATM and TP53 gene deletion, and trisomy 12) by fluorescence in situ hybridization (FISH) and also IgH gene aberrations in some patients. We compared the findings with other factors and clinical characteristics. This work shows that the conventional G-banding is analysis relatively little relevant. FISH was more effective in detecting aberrations in CLL. Although none of the four aforementioned changes is specific to CLL, the prognostic impact is significant, particularly that of TP53 deletion. In this Diploma Thesis, I am trying to personally grasp the pericope of the Gospel of John describing the meeting of Jesus with a woman of Samar′ia. Within the stated methodological procedures, the intention was to point out to the infinite source of inspiration in Scriptura sacra, in other contexts as well. An individual approach towards the specific text is also important. In the introduction, the historical environment of this story's origin from the fourth chapter of the Gospel of John is mentioned, within the perspective of social, national and cultural phenomena. The authentic development of the meeting itself is gradually monitored. Jesus' answer is loving and at the same time, uncovers personal depths. There are a lot of dialogue levels that gradually go to the transformation of the addressed woman of Samar′ia. Working with this biblical text is primarily focused on the topics that are practically considered when investigating the connections of a specific person in the specific life situation and God as Jesus Christ. The aim of our study was to evaluate association between venous disorders and preclinical atherosclerosis and its risk factors. The study was particularly focused on the association between cardiovascular risk factors and preclinical atherosclerosis with venous diseases represented by venous thromboembolism and/or symptoms of chronic venous disease evaluated by standard protocol. In our first study we evaluated the association between preclinical atherosclerosis and prevalence of venous thromboembolic events in patients with thrombophilias. Preclinical atherosclerosis might increase through its procoagulative action the risk of venous thromboembolism. Presence of preclinical atherosclerosis in common carotid and femoral arteries measured by ultrasound was assessed by Belcaro score (based mainly on presence of plaques) and by measurements of intima media thickness in the same location in 109 patients (43 men, mean age 41.5 ± 13 years) with established thrombophilias. Other parameters under study were age, presence of traditional cardiovascular risk factors, anthropometric and clinical data including blood pressure measurements and medication. Cystic fibrosis (CF) is the most common autosomal recessive genetic disorder in Caucasians. Lower respiratory tract of CF patients is colonized by specific bacteria, often leading to chronic infection and lung tissue damage. In this thesis we characterized 338 isolates of S. aureus from 92 Czech CF patients isolated in 2011-2013. Using spa typing and PFGE we detected high clonal heterogenity of this collection with the exception of MRSA strains (resistant to oxacillin; 5% prevalence) which were clonally related. The prevalence of S. aureus MLSB resistance in our collection was high (69 %), which is a serious problem due to common usage of these antimicrobials in clinical practice. A half of the MLSB resistant strains lacked any known determinant of this resistance (ermA, ermC, ermT, msrA). Sequencing of the ribosomal genes revealed a high number of S. aureus strains carrying target site mutations resulting in MLSB resistance (37 %). This is new important information about the staphylococcal strains associated with chronic infections in Czech Republic. Introduction: Adenoids are common disease in childhood, especially at preschool age. The main symptoms of adenoids are nose obstruction, chronic rhinitis, cough, relapsing otitis media, sinusitis, sleeping disorders and otitis media with effusion. The nasal patency influences the nasal nitric oxid (nNO) concentration. However, there are only a few published data about concentration of nNO in patients with adenoids. Aims: The prospective study explores the influence of adenoids on nNO in children. The investigated parameters in probands are the age, atopic diseases, allergic rhinitis and bronchial asthma. The results of nNO are compared with epipharyngoscopy, rhinomanometry and effect of adenoidectomy. Methods: The study was carried out on children with adenoids (n=105) at the age from 5 to 18 years old. The control group consists of 38 healthy children. All children underwent epipharyngoscopy, allergology examination including prick test and measurement of nNO and fractional exhaled nitric oxide (FENO) by the chemoluminescence analyzer. 7 ABSTRACT The occurrence of enthesopathies is associated with muscle insertion sites and physical activities. The aim of this work is focused on two basic target areas. In the methodical section, I tested two methods of enthesopathy evaluation (Crubézy, 1988; Villotte, 2006) and I monitored the influence of age and physical stress on the incidence of enthesopathies in a documented skeletal collection from Greece. The main aim was then the evaluation of enthesopathies in the Great Moravian population from the settlement agglomeration Mikulčice-Valy (9th -10th century) using the selected method and the subsequent interpretation of the results attained, taking into consideration the archaeological facts relating to the given locality. Based on the observed degrees of concordance between the repeated evaluations, I used Villotte's method (2006) for final evaluation. I proved the correlation between the prevalence of enthesopathies and age in individuals from a recently documented skeletal collection. Background: Despite a slight decline in the prevalence of substance use among adolescents in last years, there is a constant need for effective, available and well-targeted preventive interventions. Children of parents with substance use experiences are considered to be more endangered. Although the quality and availability of universal prevention programs has improved, their effectiveness is not usually systematically verified, moreover, the impact on children of parents with substance use experiences is unknown. The Unplugged preventive intervention was supplemented by nPrevention booster sessions in order to support its research-proven preventive impact. Introduction: Obstructive sleep apnea (OSA) is a condition significantly increasing cardiovascular mortality. In the population of Type 2 diabetes (T2D) patients, OSA is highly prevalent, but mostly undiagnosed. Screening for OSA in T2D patients is still not routine practice and it is also uncertain what the most effective screening method is. The aim of this study was 1) to establish the prevalence of OSA in T2D patients 2) to compare the performance of screening questionnaires and to determine a suitable screening method 3) to find out the adherence of T2D patients to the diagnostic process and OSA treatment 4) to compare adherence to treatment between T2D and regular sleep clinic patients. Methods: 494 consecutive patients with T2D were recruited and asked to fill in screening questionnaires, to undergo screening and, if indicated, diagnostic home sleep monitoring. Those with clinically significant OSA (apnea hypopnea index ≥ 15) were recommended CPAP (continuous positive airway pressure) treatment. Changes in cell immune parameters with focus on NK cells in patients with pancreatic cancer in relation to therapy and depression Pancreatic cancer (further PC) is one of the diseases with the greatest malignant potential. Most of patients die within 1 year from diagnosis, and only 2-5 % of patients survive more than 5 years. A major cause of late diagnosis of PC and so his insufficient therapeutic management is due to the late clinical signs of PC. Despite of progress in understanding of the molecular and biological basis of tumor genesis, the prognosis of patients with PC stays during the years unchanged. One of the often clinical signs related with PC is the presence of depressive symptoms or already developed clinically depression. The association of depression and cancer constitutes an important factor affecting the quality of life and can lead to increased morbidity. Even when the relationship between the function of subcortical centers of the brain, immune and endocrine system is known, the role of long-term stress and depression in the homeostasis is often overlooked. Tuto práci bylo určitě možno napsat na mnoha místech jinak, zcela jistě lépe a často také více do hloubky. Nepopírám, že řady vytčených cílů bylo dosaženo pouze částečně a některé otázky zůstaly dokonce nezodpovězeny, přesto si ale myslím, že práce jako celek svůj základní účel splnila. Výsledky archeologického výzkumu často svojí nekonkrétností svádí ke spekulaci. V takovém případě jsem se vždy snažil o co možná nejserióznější přístup, který by bral ohled na všechny možné varianty řešení. Jestliže jsem byl nucen formulovat konkrétní závěry, pokoušel jsem se je podat vždy tak, aby bylo zřejmé, jaké důvody mne ke konečnému rozhodnutí vedly. Netvrdím, že výsledky, k nimž jsem při práci dospěl, odrážejí nějakou historickou pravdu a jsou tak jediné možné. Jsem si vědom, že jiný badatel může mít na věc odlišný názor, a budu proto rád za každou věcnou připomínku ke způsobu provedení i výsledkům mého dosavadního výzkumu. Čtenáři se pak dodatečně omlouvám, za všechny případné chyby, které v práci najde, a které padají pouze a jen na mou hlavu.

In recent years, the trend of Taiwan’s educational reforms in elementary and secondary education is heading for deregulation; however, U.S. federal government is trying to strengthen its influences on states’ educational policies. With theories and policies combined, the thesis tries to analyze the changes of K-12 educational policies in U.S. federal government to understand the shift of the role of state in education, and then to provide suggestions for our government. This thesis uses laws and theories as the bases of discussion. In terms of laws, although U.S. Constitution did not mention any word on education, the doctrine of implied power, General Welfare Clause and the First and Fourteen Amendments provide federal government with power to intervene in educational affairs. From theoretical dimension, the externality effect, public good, equality of educational opportunity, and social harmony are reasons for state to intervene in educational affairs. In addition, the Conservatism and the Liberalism in the U.S. also exercise their respective influences on the roles of federal government in education. The former believes a lack of competition in education will result in low efficiency; moreover, they are concerned about the possibility of using education as a means of state control. On the contrary, the Liberals are asking for more governmental intervention, hoping that positive freedom and equality being realized. Based on the changes of educational policies, the purposes of federal laws can be divided into three periods. Before 1958, educational laws are made for the establishment of public education system. Between 1958 and 1982, the spotlight was on the improvement of student achievement and equality of education opportunity. After 1983, student achievement and accountability becomes the main concern of educational reform. According to the analysis, the thesis divides the role of U.S. federal government into three periods. Before 1958, it played an insignificant role. Between 1958 and 1982, it turned to a role which provided financial support but without intense control. After 1983, it not only provided financial support but also attempted to lead and control educational affairs. Three implications are mentioned. First, the state should play a positive and active role in education. Second, the state should use accountability to improve student achievement. Third, educational reforms should proceed with more flexibility.

Driving license can be taken from addictological patients by diagnosing substance abuse syndrome or driving a motor vehicle under the influence of an addictive substance. After that, drivers are often sent by specialists to the outpatient addictolocial care for examination by an addictologist. However, there is currently no uniform workflow and the role of addictologist is unclear in this process. The main aim of this thesis is to describe the role of addictologist in outpatient addictolgical care in the process of assessment of the return of driving license after its previous removal in patients diagnosed with substance abuse syndrome. Data obtained by semi-structured interview with staff of 9 outpatient addictological cares, which have practical experience with the given issue, was fixed by audio recording and then transcribed into text form. Analysis of this data was done by the method of creating clusters, linking them to larger units, and organizing data into groups based on their similarity.
