Resumen
Vegetable markets in Mumbai sell “methi” at two stages of growth, the full grown vegetative to flowering stage and another at two cotyledonary-leaves stage. The latter are commonly grown in the sandy beaches of Mumbai whereas the former are the regular soil-grown ones. The beach grown Trigonella foenum-graecum L. plants are watered with saline beach- well waters whereas the soil-grown ones are watered with regular fresh water. The research work focused on studying the impact of this difference in growing conditions on many growth parameters, viz., morphology, anatomy and biochemical analyses of the sand and soil-grown plants in addition to sand, soil and water analyses. Current paper deals with the impact of these different growing conditions on some foliar parameters viz., anatomy, stomatal index, palisade ratio and venation patterns, in the two types of Trigonella foenum-graecum L. plants.

Resumen
The health benefits and medicinal properties of herbal food products are known since antiquity. Fenugreek, Trigonella foenum-graecum Linn (T. foenum-graecum, Fabaceae), a seed spice used to enhance flavor, color and texture of food is employed for medicinal purposes in many traditional systems. Ethno botanical survey of T. foenum-graecum revealed the seeds of the plant to be useful in anemia. The objective of this study was to study the ant-anemic effect of hydroalcoholic extract of seeds of T. foenum-graecum against phenylhydrazine induced anemic rat model. The hydroalcoholic extracts of seeds were prepared by soxhlation. Phytochemical analysis of the extracts was performed using standard testing procedures. Hemolytic anemia was induced in male Wistar rats by intraperitoneal administration of phenylhydrazine HCl (PHZ) at doses of 40 mg/kg of body weight during two successive days then one day after the animals were treated orally by the hydroalcoholic extracts with the amounts of 200 mg/kg and 400 mg/kg of body weight and Dexorange (reference drug) up to 13 days. The rats were analyzed for hematological parameters such as hemoglobin (Hb), red blood cell count (RBC) and white blood cell count (WBC) on day 2 and 13. Phytochemical screening of the extracts indicated the presence of carbohydrates, saponins, sterols, polyphenols, tannins and flavonoids. Anemia was induced successfully in Groups II, III, IV and V which was indicated by a mean reduction of 51.6% in RBC count; 52.85% in Hb content and 54.9% in WBC. Analysis of hematological parameters on day 13 showed that extract significantly (p<0.05) improved Hb, RBC and WBC count at a dose of 400 mg/kg body weight. This study, not only substantiates the folklore use of the seed of T. foenum-graecum, but also suggests its inclusion in the treatment of anemia as it exhibited significant anti-anemic activity Keywords: Trigonella foenum-graecum, Anemia, Phytochemical analysis, Phenyl hydrazine, Dexorange

Resumen
Trigonella foenum-graecum (TFG) is one of medicinal plants containing several steroidal sapogenins, such as diosgenin, yamogenin, gitogenin, tigogenin and trigoneoside, and also alkaloid trigonellin and some flavonoids such as vitexin, isovitexin, orientin, isoorientin, which has many activities, such as antidiabetic, estrogenic, and also anticancer. As phytoestrogen, TFG was predicted to have potency as Selective Estrogen Receptor Modulators (SERMs) which is used for hormonal-dependent breast cancer treatment. This experiment was carried out to investigate interaction of some sapogenin steroids and flavonoids in TFG to estrogen receptor alpha (ERα) and its activity to breast cancer cell line as confirmation. In silico prediction was carried out to investigate their estrogenic activity by analyzing their binding affinity to ERα using AutoDock Vina program. In vitro activity confirmation of TFG extract and its fractions were carried out using MTT assay on Erα-positive human breast cancer cell line, MCF-7. Results showed that free binding energies of diosgenin and yamogenin were -6.4 kcal/mol, estradiol was -6.0 kcal/mol, and tamoxifen was -5.1 kcal/mol. While cytotoxicity assay showed that ethyl acetate fraction gave the lowest IC50 of 41.81 ppm, with total steroid content of 20.03 ppm. From these results, we can conclude that diosgenin and yamogenin have greater binding affinity to ERα comparing to estradiol and tamoxifen. In vitro assay confirmation showed that ethyl acetate fraction has cytotoxic effect on MCF-7 cells. Keywords: Trigonella foenum-graecum, sapogenin steroids, MCF-7, estrogen receptor alpha, binding affinity

Resumen
Ischemic heart disease, often known as coronary heart disease (CHD), is a term for heart disorders caused by narrowing heart arteries (coronary arteries) that provide blood to the heart's muscles. The body is prone to heart attacks when there is less oxygen supply (hypoxia) to the heart resulting a major cell death in the heart tissues. Cardiovascular disease (CVD) is the leading cause of death worldwide. Over the last few decades, the potential therapeutic effects of herbs and other types of complementary medicines for managing risk factors for CVD have gotten a lot of attention. Fenugreek (Trigonella foenum graecum Linn.) is a very important spice in most Asian dishes. In Malaysia, fenugreek, locally known as ‘halba’, is used in the preparation of popular dishes; ‘nasi dagang’ for breakfast, and ‘putu halba’ as snacks for hi-tea and sometimes applied in drinks. Although, to date, the effect of fenugreek against diabetes and heart diseases is well investigated, most studies do not focus on its the effects at molecular levels. This review gives an insight on the ischemic heart disease and the nutritional values of fenugreek as functional food in protecting the heart.

Resumen
Background and Objective. Trigonella foenum-graecum Linn., also called fenugreek, is a popular medicinal plant cultivated all over the globe. Fenugreek seeds are known for their many medicinal properties. We present our findings on the effect of a 70% aqueous methanolic fenugreek seed extract (Tfg.Cr) on isolated GI smooth muscles (rabbit jejunum and rat ileum) and the effect of extract and its constituent diosgenin on acetylcholinesterase (AChE) enzyme. Results. When tested on the baseline of isolated tissues, Tfg.Cr was devoid of any activity (stimulant or relaxant) till 10 mg/ml. This is an interesting finding, keeping in mind that the fenugreek seeds are used to alleviate constipation and diarrhoea. When Tfg.Cr was tried for any potential AChE inhibitory activity, it did show an inhibitory effect in increasing concentrations (47-380 μg/ml). This inhibitory effect was comparable to the effect produced by a standard AChE inhibitor physostigmine. One of the known fenugreek constituents, diosgenin, was also tested, and it also showed an AChE inhibitory effect in a concentration-dependent manner (11-190 μg/ml). Interaction between diosgenin and AChE was further investigated by molecular docking and molecular dynamics simulations for 100 ns, which showed that diosgenin interacted with the active-site gorge of AChE through hydrophobic, pi-pi stacking, and hydrogen bonds with various amino acids of the AChE enzyme. Conclusion. The results show that the fenugreek extract does not possess any GI stimulant or relaxant activity even though it is used traditionally in GI motility disorders. The extract and diosgenin could inhibit the AChE enzyme pointing towards their benefit to enhance the memory.

Resumen
Background: Activation of hepatic stellate cells (HSC) plays central role in the development of liver fibrosis. In HSC activation, the transforming growth factor-β1 (TGF-β1) is considered to be the main stimuli factor. Diosgenin are the steroidal saponin and found in Trigonella foenum graecum Linn (Fenugreek) and some other species of Dioscorea. Diosgenin attenuates HSC activation by inhibiting transforming growth factor-β. Aim: In present study an attempt was made to explore the effect of diosgenin on liver fibrosis. Methods: Liver fibrosis was induced in rats by carbon tetrachloride (CCl4) 1 ml/kg intraperitoneally twice a week for 28 days and cisplatin 3mg/kg intraperitoneally at 0, 1, 3 week for 4 weeks. The extent of liver fibrosis was assessed by measuring the weight of liver and levels of total bilirubin (TBL), hydroxyproline (HP) and serum enzymes due to deposition of extracellular matrix (ECM). Results: The administration of diosgenin reduced the liver weight of CCl4 and cisplatin treated animals and reduced the TBL, HP level and serum enzymes significantly and inhibited liver fibrosis induced by CCl4 and cisplatin. Conclusion: The result obtained in the present investigation, Diosgenin treatment exerted significant hepatoprotective effect in animals by inhibiting ECM deposition and HSCs activation.

Resumen
The aim of the present study is to assess the effects Fenugreek (Trigonella foenum-graecum, Fabaceae), sunflower (Helianthus annuus, Asteraceae), green cardamom (Elettaria cardamomum, Zingiberaceae) and seed extracts, which they belonged to three different plant families, on common carp (Cyprinus carpio L.) spermatozoa motility parameters and oxidative stress conditions. For this reason, sperm samples treated with seed extracts at the ratios of 0.1%, %0.5, 1%, and 2%, were incubated at 4°C and sampled at 2nd and 48th hours. Among sperm motility parameters, curvilinear velocity (VCL), average path velocity (VAP), straight-line velocity (VSL), straightness STR), linearity (LIN), and wobble (WOB) were determined by computer-assisted sperm analysis (CASA). According to the results, sunflower and green cardamom seed extracts have attenuated motility parameters at all tested concentrations. However, motility parameters measured in %0.5 of fenugreek seed extract were higher than all other groups including the control at 48th hour. Also, TBARS values as an oxidative stress indicator in this group were decreased. 2% of all seed extracts had negative effects on the sperm samples. Particularly, 2% sunflower seed extract caused the higher oxidative stress. These results indicate that fenugreek seed extract is more proper for the maintenance of common carp spermatozoa at 4°C, comparing to those of sunflower and green cardamom.

Resumen
Systematic review and meta-analysis criteria were used to plan and conduct this systematic review (PRISMA). Different literature of reviews explicit that Fibers, flavonoids, polysaccharides, and saponins are the principal chemical components of Trigonella foenum-graecum. Some of the components may have mitogenic actions, which result in immune-competent cells being restored. Some of these ingredients are also antioxidants and may have immune-stimulant characteristics. In vitro, the extract from the seeds of the fenugreek seed was found to be cytotoxic to numerous cancers, including T-cell lymphoma, but not to normal cells. Breast, pancreatic, and prostate cancer cell lines showed growth inhibition after being treated with a specific amount of the extract. Diosgenin suppressed MCF-7 cell proliferation in a dose-dependent manner, according to in vitro tests. Diosgenin's effect on a human mammary carcinoma cell line, as well as its therapeutic potential for N-nitroso-N-methyl urea-induced breast cancer. Fenugreek extracts and their isolated constituents have been revealed to have anti-cancer activities in several investigations. Trigonelline, Diosgenin, Protodioscin, and Dioscin were found to be the most potent anticancer ingredients in fenugreek. Furthermore, fenugreek seeds are high in many essential chemicals, which show asignificant role in the suppression and inhibition of various malignancies. In addition, studies on the estrogenic effects of fenugreek seeds in vitro and in vivo have demonstrated its potential as an HRT alternative.

Resumen
Most antidiabetic polyherbal preparations contain Trigonella foenum-graecum, Momordica charantia and Cinnamomum zeylanicum as active components. Although all ingredients are popular as antidiabetic agents, but the analytical method is not available for simultaneous estimation of marker compounds from herbal formulations due to several challenges. Hence, the HPTLC method was developed for simultaneous estimation of three active phytoconstituents viz. diosgenin, charantin and hydroxychalcone in polyherbal formulation. The stationary phase of the optimized HPTLC method is silica gel 60 GF254 and the mobile phase is chloroform: glacial acetic acid: methanol: water (4:3:2:1v/v). The Rf value of phytoconstituents charantin, diosgenin, and hydroxychalcone was found to be 0.72, 0.61 and 0.30 at detection wavelength 342 nm. According to ICH criteria, the analytical method validation was performed. All three markers showed linear and proportional responses in the 400 to 1400 ng/band range, which confirmed the markers’ linearity. Precision measurements were made at the intraday and interday levels, and the results were satisfactory and in line with the specifications. Accuracy was determined by the recovery method and %recovery was found to be in the range of 85.20 to 97.19. The validated HPTLC method was utilized to analyze the marketed Quanto Diab Forte capsule formulation containing charantin, diosgenin, and hydroxychalcone. The proposed validated analytical method was found to be simple, precise and accurate.

Resumen
Fenugreek (Trigonella foenum-graecum L.) has long been used as a medicinal plant for the treatment of nutritional and metabolic dysfunctions. It has been shown that the daily intake of fenugreek seeds improves feeding behavior in animals. A number of studies have also suggested a link between Fenugreek seeds extracts and fertility but have yielded contradictory results. In this paper, particular attention has been paid to the impact of consumption of the aqueous extract of fenugreek seeds, prepared according to the traditional Moroccan method, on the spermatic parameters and the pathophysiology of the reproductive system of Wistar rats receiving 450 and 900 mg / kg /day, respectively. Our study results show the benefits of fenugreekseeds aqueous extract, used in the traditional Moroccan pharmacopoeia on appetite stimulation and weight gain without any significant impact on the fertility and the reproductive performances. Both quantitative and qualitative analysis of sperm parameters (morphology, mobility and vitality) showed no significant variation compared to the controls. In addition, histological examination of seminiferous tubules and reproductive accessory glands (prostate and seminal vesicles) in fenugreek treated rats showeda normal tissue architecture without visible lesion. These data exclude all potential risks from the consumption of aqueous extract of fenugreek seeds on the fertility and reproductive function in male rats. However, taking into account other data from the literature, this regular consumption should be closely monitored in order to allow the organism to fully benefit from a multiple phytotherapeutic virtues of this plant.

Resumen
Diabetes mellitus (DM) is a chronic metabolic disorder. Streptozotocin is a naturally occurring cytotoxic chemical, particularly toxic to the pancreas and insulin producing beta cells in mammals and induces diabetes. Glimepiride is a second generation sulfonylurea, used as second-line or add-on treatment options for type 2 diabetes. Fenugreek (Trigonellafoenum graecum) seeds have been documented as a traditional plant treatment for diabetes. Soluble dietary fiber of Fenugreek significantly improved oral glucose tolerance in diabetic rats. It also exerts anti-diabetic effects mediated through the inhibition of carbohydrate digestion and absorption and the enhancement of peripheral insulin action. Most herbal remedies can interact with allopathic drugs resulting in altered activity and toxicity. At the same time, herbal remedies might produce the same kind of effects as the drug produce. Current published research information on herb-drug interactions is scanty. So, the aim of this study was to investigate the possible interaction between conventional drug used for the management of diabetes; (Glimepiride) and a traditional herbal remedy; Fenugreek aqueous extract in Streptozotocin induced diabetic male albino rats. In conclusion, combination therapy induces better hematological, biochemical effects and improves the oxidative stress biomarkers and antioxidant enzymes. Histological studies showed better results on some organ functions. The results emphasize the benefit of using the combination of Fenugreek seeds aqueous extracts as supportive complementary anti-diabetic therapy.

Resumen
Household processing of fenugreek seeds and leaves, including soaking, germination, and boiling of the seeds, and air-drying of the leaves, has improved the levels of human consumption of the bitter seeds and increased the shelf life of fresh leaves, respectively. The potential anticancer activity of either unprocessed or processed fenugreek seeds or leaves and the relative expression of pro-apoptotic and anti-apoptotic genes of the studied cancerous cell lines exposed to IC50 crude extracts was investigated to observe the apoptotic-inducing property of this plant as an anticancer agent. The protein expression of IKK-α and IKK-β, as inhibitors of NF-KB which exhibit a critical function in the regulation of genes involved in chronic inflammatory disorders, were studied in the tested cancerous cell lines. In this study, the anticancer activity of household-processed fenugreek leaves and seeds against HepG2, HCT-116, MCF-7, and VERO cell lines was measured using an MTT assay. DNA fragmentation of both HepG2 and MCF-7 was investigated by using gel electrophoresis. RT-PCR was used to evaluate the relative expression of each p53, caspase-3, Bax, and Bcl-2 genes, whereas ELISA assay determined the expression of caspase-3, TNF-α, and 8-OHDG genes. Western blotting analyzed the protein-expressing levels of IKK-α and IKK-β proteins in each studied cell line. Data showed that at 500 µg mL−1, ADFL had the highest cytotoxicity against the HepG2 and HCT-116 cell lines. Although, each UFS and GFS sample had a more inhibitory effect on MCF-7 cells than ADFL. Gel electrophoresis demonstrated that the IC50 of each ADFL, UFS, and GFS sample induced DNA fragmentation in HepG2 and MCF-7, contrary to untreated cell lines. Gene expression using RT-PCR showed that IC50 doses of each sample induced apoptosis through the up-regulation of the p53, caspase-3, and Bax genes and the down-regulation of the Bcl-2 gene in each studied cell line. The relative expression of TNF-α, 8-OHDG, and caspase-3 genes of each HepG2 and MCF-7 cell line using ELISA assays demonstrated that ADFL, UFS, and GFS samples reduced the expression of TNF-α and 8-OHDG genes but increased the expression of the caspase-3 gene. Protein-expressing levels of IKK-α and IKK-β proteins in each studied cell line, determined using Western blotting, indicated that household treatments decreased IKK-α expression compared to the UFS sample. Moreover, the ADFL and SFS samples had the most activity in the IKK-β expression levels. Among all studied samples, air-dried fenugreek leaves and unprocessed and germinated fenugreek seeds had the most anti-proliferative and apoptotic-inducing properties against human HepG2, MCF-7, and HCT-116 cell lines, as compared to the VERO cell line. So, these crude extracts can be used in the future for developing new effective natural drugs for the treatment of hepatocellular, breast, and colon carcinomas.

Resumen
High levels of free radicals in diabetic wounds often cause chronic inflammation. Nigella sativa and Trigonella foenum-graecum seed extracts are rich in antioxidants and have anti-inflammatory effects. Trigonella foenum-graecum seed extracts can also accelerate the reepithelialization process in wound healing. This study aimed to determine the combination of ethanol extract of Nigella sativa and Trigonella foenum-graecum seeds on wound length and the number of neutrophils, macrophages, fibroblasts, and collagen density in the incision wound of diabetic mice induced by streptozotocin. T. This study used three types of controls, namely Normal (Normal mice, HPMC 3%), DM (Diabetic mice, HPMC 3%), and Iodine (Diabetic mice, Povidone Iodine 10%). Topical treatment with 70% ethanol extract ointment combined with Nigella sativa and Trigonella foenum-graecum in various variations, namely N (20%:0%), T (0%:10%), NT (10%:5%), Observations of the wound healing process were carried out on 3rd, 7th and 14th day. Data were analyzed using the One Way ANOVA and Duncan Multiple Range Test. The study's results by observing the morphology of the wound length on day 7th day showed that the combination treatment of extracts of Nigella sativa and Trigonella foenum-graecum resulted in the shortest wound, which was significantly different from the wound in untreated diabetic mice. In addition, diabetic wounds without treatment showed infection, and on the contrary, the infection did not occur in wounds treated with a combination of extracts of Nigella sativa and Trigonella foenum-graecum. The results of wound histology observations showed that the combination treatment of extracts of Nigella sativa and Trigonella foenum-graecum showed a decrease in inflammation which was indicated by a significant decrease in the number of neutrophil cells, macrophages, and accelerated reepithelialization of wound healing as indicated by a significant increase in the number of fibroblasts and collagen density since the third day of treatment.
