T cell infiltration into tumours is essential for tumour antigen recognition and tumour cell elimination. The aim of this study was to develop a better understanding of T cell infiltration into tumours, focusing on two opposing arms of an immune response, anti-tumour CD8 Tcells and Regulatory T cells (Tregs). Activated CD4 T helper cells are also of importance but could not be studied due to the time constraints of the project. The effect of T cell signalling at the immunological synapse following interactions between T cells and APCs presenting cognate antigen have been well studied [1]. The endothelium is neither a stereotypical APC nor simply a passive filter barrier for non-cognate infiltrating T cells. The endothelium can activel influence the development of an inflammatory response depending on the functional state of both the endothelium and interacting T cells (resting versus recently activated T cells)and the type of interactions (cognate versus non-cognate). The hypothesis was that recognition of antigens presented in the context of major histocompatibility complex (MHC)molecules by endothelium aids T cell transmigration and hence infiltration into tissues, including into tumours. In this study, the data highlights that high avidity TRP-2 specific CD8 T cell transmigration across murine lung endothelium requires recognition of TRP-2 peptide presented by the endothelium, aiding recruitment of antigen-specific T cells into tissues in the absence of endothelial cell killing. In order for antigen specific T cells to migrate into the tumour, the tumour endothelium therefore needs to present tumour antigens. In addition to CD8 T cells, high numbers of Tregs have been found within tumours but the key mediators for this recruitment remain uncertain. The data shows a novel mechanism for Treg transmigration where cognate antigen-specific recognition of self-peptides was required for transmigration with preferential transmigration of Tregs across syngeneic rather than allogeneic endothelium. Upregulation of major histocompatibility complex (MHC) class II and adhesion molecules, by IFN-γ and TNF-α, together with a gradient of the tumour associated chemokine CXCL12 were also pre-requisites for efficient Treg transmigration. Previous studies have shown that high CXCL12 expression can induce fugetaxis of tumour cells leading to efficient metastatic spread and a poor prognosis. These results would suggest that low levels of CXCL12 and recognition of self peptides presented by self MHC on endothelial cells allows efficient migration of Tregs whereas higher levels of CXCL12 may promote tumour metastases and lead to fugetaxis of the Tregs leading to an even worse prognosis. In conclusion recognition of cognate antigen presented by endothelium enhances antigenspecific transmigration of CD8 and Regulatory T cells. This study therefore reports a novel mechanism for T cell subset infiltration into tumours where high avidity CD8 T cells require recognition of cognate tumour antigen presented on tumour endothelium in the context of MHC class I and conversely regulatory T cell infiltration into tumours depends on the repertoire of self-peptides presented on tumour endothelium in the context of MHC class II. Alteration of antigen presentation or MHC expression on tumour endothelium therefore represents a mechanism whereby T cell infiltration can be altered to re-direct anti-tumour immune responses.

This thesis discusses the agencies, transport systems, and infrastructure that enabled more than 3.15 million Europeans to emigrate to the United States, Canada, and South Africa through Britain between 1836 and 1914. Rather than travelling directly from the European mainland, these transmigrants broke their journeys by travelling to Britain where they boarded another vessel that conveyed them across the Atlantic. The control that Britain exerted over both the short-sea and long-haul passenger routes thus involved was as important to British maritime commerce as similar controls over freight or direct long-haul passenger routes to the far-flung corners of the British Empire. However the crucial significance of the transmigrant business to the British merchant marine has been largely overlooked in recent historiography, and it is this lacuna that the present dissertation seeks to redress. The study is split into three sections. The first part quantifies the patterns of the transmigrant business, answering questions such as: what were the origins of the migrants and what routes did they use to reach Britain? When did they come? Where in Britain did they land, where were they bound, and where did they re-embark? Having charted these issues, the thesis turns in the second section to investigate how the transmigrant business developed and evolved, paying particular attention to the factors that conditioned the market throughout the 78 year period. Finally, the thesis examines the significance of the transmigrant business to British ports serving as conduits for the passenger movement, to the companies involved in transporting the aliens, and to the migrants themselves. By exploring these issues this thesis has made a significant contribution to migrant and maritime historiography in the following ways. First, it has broadened the chronological and geographical focus of migrancy back from the 1880s to the 1830s and stressed Scandinavian as well as Central/East European movements. Second, it has demonstrated how European transmigrants were as important to British shipping companies as were British emigrants seeking to settle in Britain's overseas dominions. Third, immigration to Britain has been incorrectly conceptualised because historians and social commentators fail to take account of the onward movement of aliens arriving in Britain and assume instead that most were permanent settlers. Fourth, the primacy of Britain's maritime links to the United States was more important for the passenger business than has been previously acknowledged. Finally, this study disproves theories by immigrant historians that centres of alien settlement across Northern Britain arose because they were situated along the transmigrant corridor between the Humber and Mersey. In reality many of the trains carrying transmigrants never passed through the towns and cities where large-scale immigration took place. By combining a mixture of global, national and local studies, and a longer chronology, this thesis offers an important intersection of transport and maritime studies that shows how transmigration has been under appreciated by both maritime and migrants historians alike.

The passage of leukocytes across blood vessel walls plays a key role in the immune response to infection in inflammatory conditions. ADAM10 is a ubiquitously expressed molecular scissor that proteolytically cleaves key cell surface proteins including vascular endothelial (VE)-cadherin and transmembrane chemokines. Their shedding by ADAM10 promotes leukocyte transmigration in cell line models, however the precise mechanism behind ADAM I O's involvement is unknown. ADAM10 associates with six different membrane organising tetraspanins (Tspan5/10/14/15/17/33) termed the TspanC8s. These tetraspanins regulate ADAM10 enzymatic maturation and trafficking to the cell surface and emerging evidence indicates that different TspanC8s can promote ADAM10 cleavage of specific substrates. It was hypothesised that ADAM10 promotes leukocyte transmigration by cleaving one of its endothelial substrates and one or more of the TspanC8s could facilitate this process. The aim of this thesis was to test this hypothesis using in vitro leukocyte adhesion assays with primary human leukocytes and human umbilical vein endothelial cells (HUVECs). siRNA knockdown or pharmacological inhibition of ADAM10 on HUVECs impaired the transmigration of lymphocytes, but not neutrophils or monocytes. ADAM10 knockdown/inhibition caused a reduction in VE-cadherin shedding and an increase in VE-cadherin surface expression. Partial knockdown of VE-cadherin, in the presence of ADAM10 knockdown/inhibition, reduced VE-cadherin levels to normal and restored basal lymphocyte transmigration. Systematic knockdown of TspanC8s in HUVECs revealed that the presence of either Tspan5 or Tspan17 was sufficient to maintain basal lymphocyte transmigration and reduced VE-cadherin surface levels. Tspan5 and Tspan17 are functionally uncharacterised, but they are the most highly related TspanC8s by sequence (78% amino acid identity) and may share a common role in lymphocyte transmigration by regulation of ADAM10 and VE-cadherin.

Since the 1970s there has been concern expressed about the consequences for Third World women of economic development programs. This thesis considers the implications for women of policy and planning in the transmigration program, a resettlement scheme which has been an important element of the Indonesian government's development strategy. The discussion centres on the conventional procedures and practices of transmigration planning; the nature of projects designed specifically for transmigrant women; and the significance of the recent stress on 'the role of women' in transmigration policy. Although planners involved with the mainstream facets of transmigration planning claim to treat men and women equally, analysis of their routine procedures shows that women are often discriminated against and their special interests neglected. On the other hand, planners in women's sections within the planning bureaucracy are engaged in designing and implementing a number of projects aimed specifically at assisting transmigrant women. However, these projects are typically small-scale with low budgets, offering participants meagre and uncertain benefits. Their common strategy has been to provide skills training in areas which are overwhelmingly domestic-centred and subsistence-oriented in contrast to the stress in mainstream planning on more highly valued, male-dominated export crop production. In the mid 1980s, disquiet with low productivity in the settlements and the failure of many transmigrant families to sustain themselves, has led to an upsurge of interest among policy-makers in the potential productive contribution of transmigrant women who are viewed as an 'untapped human resource'. An outcome of the belief that women's labour could be better utilised is the greater involvement of mainstream planners in designing projects for women and collecting data about their 'roles'. The thesis argues that as transmigrant women have become a target of current development planning in Indonesia, they are being drawn into a new, essentially one-directional relationship with state and development agency bureaucracies which seek to harness their productive capabilities, although the underlying causes of the inequitable social relationships in which they are situated are not addressed. The thesis maintains that, in fact, the bureaucratic practices and conceptual frameworks which are applied in transmigration planning work to disempower women and compound their subordination both to men and the state.

Leukocyte transmigration is a critical step of the inflammatory process. In this project I have examined leukocyte responses to transmigration and endothelial responses to both chemical and mechanical stimuli which are known to be involved in leukocyte transmigration. My work has identified ~2500 differentially expressed genes following endothelial exposure to interleukin-1 beta (IL1β). Interestingly, IL1β induces up-regulation of claudin-1 and pre-b-cell colony enhancing factor and down-regulation of claudin-5 and occludin, which are all involved in maintaining endothelial cell-cell junctions. Analysis of endothelial cell (EC) transcriptional changes following neutrophil transmigration found few differentially expressed genes in comparison to IL1β treated ECs; indicating that the effects of transmigration on ECs are minimal in comparison to the global transcriptional changes induced by IL1β. Atherosclerosis, characterized by monocyte accumulation within the vessel lumen, is found in regions of flow reversal and low time averaged oscillatory shear stress. I have examined the effects of this type of shear stress on endothelial cell gene expression. My data indicates that most genes differentially expressed under these conditions are controlled by low average shear stress rather than flow reversal. These differentially expressed genes are involved in regulating the cell cycle and the immune response. My work shows that cell proliferation is increased following exposure to low steady shear stress or exposure to reversing oscillatory flow in comparison to high steady shear stress. Additionally monocyte adhesion is increased following exposure of ECs to reversing oscillatory flow. My work has also examined the impact of transmigration on monocyte gene expression. I have identified genes which are differentially expressed in monocytes by exposure to EC secretions, monocyte/EC contact, and diapedesis. I have also shown that freshly isolated human monocytes have reduced apoptosis following transmigration. Surprisingly, I also found that monocytes had reduced expression of anti-microbial peptides following transmigration. Overall my work identifies important endothelial and leukocyte transcriptional responses to the process of transmigration which extends from cytokine stimulation through diapedesis.

Leukocyte migration from the blood vessel, across the vascular wall and into the tissue underneath occurs in both an inflammatory response as well as immunosurveillance. During this process multiple adhesive interactions occur between leukocytes and vascular endothelial cells (ECs). The EC itself is rendered compliant to transmigration following inside-out signalling in response to leukocyte adhesion altering the activity of a number of different cellular components including the actin cytoskeleton, Rho GTPases and various protein kinases. For this, adhesion to endothelial intercellular adhesion molecule-1 (ICAM-1/CD54) is particularly important and the focus of this study. Indeed, previous work in our lab has shown that endothelial ICAM-1 signalling controls lymphocyte diapedesis by modulating interendothelial VE-cadherin (VEC) junction phosphorylation via a pathway involving calcium, AMP kinase and nitric oxide synthase (eNOS). In this study, I have investigated if ICAM-1-mediated endothelial mitogen-activated protein (MAP) kinases activation played a role during lymphocyte transmigration across brain microvascular ECs. All three MAP kinases, namely ERK, JNK and p38, were found to be activated in response to ICAM-1 engagement, however only JNK was important for lymphocyte transmigration. Significantly, specific neutralisation experiments using small-molecule inhibitors or dominant-negative plasmids inhibiting JNK resulted in inhibition of transmigration. Activation of JNK required Src, Rho GTPase, MKK7 and protein kinase C (PKC), with all these components also found to be important for lymphocyte transmigration. I further demonstrate that this novel pathway led to the phosphorylation of the actin-associated protein paxillin and its association with VEC. Ultimately this triggered VEC internalisation, suggesting that adherens junction modulation is an important element during transendothelial leukocyte migration. Furthermore, this also suggests that at least two endothelial signalling pathways (JNK-paxillin and eNOS-VEC) converge and cooperate to regulate ICAM-1-mediated lymphocyte transmigration.

This study analyses the interplay between women's subsistence, domestic, childcare, community and income earning work in an Indonesian transmigration settlement. The context of the analysis is, on one hand, the complex ways in which gender relationships are constituted and reconstituted within the household and in wider society; and, on the other hand, the ways in which women negotiate, accommodate and resist the organising power of these gender relations in their everyday lives. The research was undertaken in a transmigration settlement in an upland region of Riau province. While the settlement is the site of concentrated development planning by the state, inappropriate strategies, unsuitable agricultural conditions, inadequate infrastructure, lack of initial capital, and so on, mean that the settlers remain subsistence-oriented and many struggle to survive. In these economically marginal agricultural conditions, women's work is critical to their households and also to the local and regional social and economic processes. Their work as subsistence producers, for the market, and for wages in agriculture and elsewhere, contributes directly to their households’ survival. Their skills are also vital in raising children, in the management, organisation and conservation of domestic resources and in cultivating and maintaining the social networks which are an important safety net in times of crisis. While women perform these duties, men are freed from the everyday burden of domestic concerns. Their privileges and authority over women and children are sanctioned through their formally recognised positions as heads of household and in their domination of the public sphere. The study integrates narratives and qualitative data with quantitative surveys to explore the multi-dimensionality, overlapping, fragmented and unrelenting nature of women's work, and the ways in which women must juggle their tasks and obligations. It highlights the implications for women of the changing composition and structure of households throughout the family life cycle and in the frequent temporary outmigration of married men in search of a better livelihood. The analysis points to the complexity of women's work and responsibilities within diverse household structures which are not recognised or accommodated in the conventional ways in which state-directed development is implemented.

Gegenstand der Publikation ist eine Art der Transmigration, die nicht, wie in der bisherigen Forschungsliteratur aufgezeigt, durch die Überbrückung des Wohlstandsgefälles entsteht, sondern durch die Verwirklichung eines bestimmten Lebensstils. In mehreren Interviews mit drei Seglern, die – neben der Teilnahme an internationalen Regatten – aus Sets verschiedener Motive zwischen Neuseeland und Europa pendeln, wird deren Lebenswelt als "Long distance-Transmigranten" dargestellt und miteinander verglichen. Im theoretischen Teil werden die Befunde der Fallstudie in die wissenschaftliche Debatte eingeordnet. Ein Abriss zur Kritik sowie die Konsequenzen dieser für die Arbeit werden gezogen. Der Nachweis, dass die untersuchten Probanden "Sonderfälle" darstellen, wird erbracht und die Einführung eines eigenen wissenschaftlichen Begriffes für dieses neue Migrationsphänomen erfolgt. Obgleich diese Studie auf einer relativ schmalen empirischen Basis erwachsen ist, zeichnet sie sich durch das innovative Vorgehen der Autorin aus.

Peptide inhibitor of trans-endothelial migration (PEPITEM) is a B cell-secreted peptide which inhibits T cell trafficking across cytokine-stimulated endothelium. This homeostatic mechanism is lost in autoimmune and chronic inflammatory diseases, leading to inappropriate T cell trafficking with pathological consequences, e.g. in type-1 diabetes and rheumatoid arthritis. We aimed to investigate the structure and function of PEPITEM in vitro and in vivo to establish its pharmacokinetics, therapeutic potential, and underlying molecular mechanisms. The efficacy of PEPITEM was verified, showing the same regulatory control of T cell trafficking as previously reported. Nuclear magnetic resonance (NMR) studies revealed PEPITEM to be a linear peptide lacking secondary structure. Intravenous administration of radiolabelled native PEPTEIM in wild-type mice showed rapid clearance by the renal circulation, conferring a circulatory half-life of < 2 minutes. Common conjugation strategies employed to modify PEPITEM, e.g. PEGylation, did not affect peptide function, demonstrating its potential for therapeutic development. The evolution of PEPITEM from its parent protein, 14-3-3ζ was investigated. PEPITEM is probably cleaved from 14-3-3ζ by matrix metalloproteinase 9 extracellularly after exocytosis from B cells. Interestingly, this generates a 17aa peptide which requires additional proteolytic processing to evolve a smaller biologically active pharmacophore. These observations add to our current knowledge of the PEPITEM paradigm, which appears central to regulated trafficking of T cells. Future work will formulate new PEPITEM versions with suitable pharmacological profiles, and screen them for function in vitro and in vivo to develop suitable therapeutic agents.

Surrealism made its appearance in Canadian art in the early 1940s, twenty years after its inception in France. The distance, both in terms of space and of time, has had major implications for the manner in which surrealist ideas have been conveyed and transformed pictorially, even as they were assimilated, in this culturally-distinct society. This process may be observed in English Canada, notably in the work of Jock Macdonald and Jack Shadbolt. But it is in French-speaking Quebec that the surrealist philosophy -in particular, its appeal to the unconscious and the non-rational powers of the human mind in apprehending and imaging the real differently- has impacted most profoundly, as its effect there extended, beyond aesthetics, to the social sphere. The climate of repression maintained by the government in collusion with the clergy in the 1940s and 50s provoked dissidence -variously overt or covert-among the avant-garde: in fostering a desire for change, Surrealism played a key role in the struggle, not least at the level of semantics. Two separate groups developed around the prominent figures of Alfred Pellan and Paultmile Borduas, each with a different but ultimately complementary conception, and attendant artistic concerns, of the surrealist ideology. Bridging the two tendencies is the painter L6on Bellefleur (born in 1910), whose oeuvre over a long career epitomises the phenomenon of transmigration that characterises the Canadian experience of Surrealism. Defining himself as a surrealist-inspired artist rather than a Surrealist, Bellefleur has nonetheless consistently drawn on the movement's guiding principles (poetry, love, freedom, automatism), techniques (many based on the concept of 'objective chance'), and ancillary interests (such as the occult), to produce a body of non-figurative works which, defying conventional notions of surrealist art, offer an indigenous vision of Breton's 'convulsive beauty'.

Dans notre étude, nous nous intéressons aux personnes âgées originaires du Maghreb qui vivent en France, pendant leur retraite, après avoir immigrés et continuent à vivre en foyer SONACOTRA (devenue ADOMA). Plus spécifiquement, nous avons sélectionné des hommes considérés comme isolés car bien que mariés dans leur pays d’origine, avec à leur charge femme et enfants, ils ont une vie de célibataire, ici, en France.Nous avons utilisé l’entretien semi-directif et l’Echelle Dépressive Gériatrique comme outils d’évaluation. Nous avons sélectionné six foyers dans la région Rhône-Alpes pour leur pourcentage élevé de maghrébins parmi leurs résidents. Les entretiens se sont déroulés de Février 2007 à Mars 2009.La passation de l’échelle de dépression gériatrique a eu lieu une semaine avant leur départ pour le pays d’origine et une semaine après leur retour. Elle a permis de confirmer l’importance de la dépression dans notre échantillon. Et, une diminution des scores (donc une amélioration de la thymie) entre les deux passations est notée pour la majorité des sujets. A partir de cette simple observation, nous pouvons déduire que les voyages au pays d’origine ont un effet revalorisant sur ces hommes comme nous l’avions émis dans nos hypothèses.Il apparait, à ce propos, que c’est la réussite d’un projet, au pays d’origine qui a un effet positif sur leur moral. Ce faisant ils ont recréé un ancrage professionnel dans leur pays au moment où celui-ci est perdu, en France. Cette réalisation est, de plus, l’aboutissement du projet d’origine puisque leur but premier était de travailler, ici, pour construire là-bas
In our study, we are interested in the elderly people originating in the Maghreb who live in France, during their retirement, after having immigrant and continue to live in hearth SONACOTRA (become ADOMA). More specifically, we selected men considered as isolated because although married in their country of origin, with the responsability a woman and children, they have a single life, here, in France.We used semi-directing maintenance and the Geriatric Depressive Scale as evaluation tools. We selected six hearths in the Rhône-Alpes area for their high percentage of Maghrebians among their residents. The talks proceeded from February 2007 to March 2009.The signing of the scale of geriatric depression took place one week before their departure for the country of origin and one week after their return. It allowed to confirm the importance of the depression in our sample. And, a decrease of the scores (thus an improvement of the thymie) between both signings is noted for the majority of the subjects. From this simple observation, we can deduce that the journeys in the country of origin have an effect's revalorizing on these men as we had emitted it in our hypothese. It appears, on this subject, that it is the success of a project, in the country of origin which has a positive effect on their moral. By doing this they recreated an anchoring professional's in their country at the time when this one is lost, in France. This realization is, moreover, the result of the project of origin since their primary goal was to work, here, to build over there

The aim of this project was the study of neutrophil recruitment and reverse transmigration using murine and human in vitro models of inflammation. Murine in vitro models of inflammation were developed using an immortalised microvascular cell line (MCEC-1) and primary murine vascular endothelial cells (mEC) isolated from heart and lung. We found that MCEC-1 could recruit murine neutrophils without the requirement of cytokine stimulation, although efficient transmigration did require such a stimulus. Primary cells required cytokine stimulation to recruit mEC. Interestingly, and in contrast to human EC, mEC were relatively insensitive to TNF-α stimulation, although IL-1β was a good stimulus for adhesion and migration. Using the IL-1β driven system we generated reverse migrated murine neutrophils and their phenotype and prolonged survival were assessed. The effect of shear stress and nitric oxide on the regulation of the process of reverse migration was examined. Using adoptive transfer strategies we investigated the fate of mRPMNs in vivo. A significant part of this work involved the study of human reverse migrated neutrophils at a proteomic level using two-Dimensional Fluorescence Gel Electrophoresis methodology to identify changes in neutrophils associated with reverse migration process. We found that murine reverse migrated neutrophils had a very similar surface phenotype to human reverse migrated cells. They also showed prolonged survival. However, our preliminary data on trafficking in vivo did not give a clear indication about their fate upon adoptive transfer into recipient mice. In vitro studies showed that flow generated shear stress and nitric oxide delayed, but did not inhibit, the process of reverse migration. Finally, the proteomics study revealed a number of metabolic, cytoskeletal and regulatory proteins that were differentially expressed in human reverse migrated neutrophils although the functional significance of these changes is yet to be explored.

Engagee en 1905 par le colonisateur neerlandais, la transmigration constitue le plus grand programme de colonisation agricole dirigee jamais entrepris par un etat. En voulant corriger l'important desequilibre demographique au sein de l'archipel, la transmigration poursuit un double objectif de politique sociale dans les iles interieures (java, madura, bali) et de politique de developpement dans les iles exterieures (sumatra, kalimantan, sulawesi, irian jaya). Le programme connait au moins autant de reussites que d'echecs. Ces derniers sont habituellement imputes au manque de moyens, a l'insuffisante selection des sites et des migrants, a la mauvaise preparation des projets et aux multiples dysfonctionnements au sein des ministeres impliques. La premiere partie montre que les difficultes auxquelles se heurte la transmigration proviennent moins de problemes de mise en uvre que d'une conception inadaptee du programme des le depart. Cette derniere est le fruit d'une perception erronee de l'evolution du paysannat javanais face a la croissance demographique, de prejuges tenaces envers les populations et le milieu des iles peripheriques et, surtout, d'une vision exclusivement agraire du developpement. Au travers de nombreux exemples, la deuxieme partie montre comment ces problemes de perception et de representation se traduisent, tant dans les choix des amenageurs que dans les reponses apportees par les paysans, au niveau des centres de transmigration. La derniere partie, enfin, apres avoir resitue la transmigration dans le cadre plus vaste des colonisations agricoles dirigees, montre qu'elle n'est en fait que le dernier avatar de l'expansion des royaumes agraires javanais, que l'origine profonde des difficultes qu'elle connait est a rechercher dans une vision et une conception particulieres du monde et du pouvoir. Mais l'indonesie n'est plus un royaume agraire. Aujourd'hui, la priorite va a l'industrialisation du pays et a la modernisation de son agriculture. Face a cette nouvelle donne, la transmigration va devoir s'adapter ou disparaitre

Cette these presente une observation des paysages dans les zones de transmigration de la province de jambi - sumatra, a partir de la teledetection aerospatiale. Des methodologies d'analyse numerique des images satellitales constituent le coeur de la these. La premiere partie est consacree a la presentation generale des elements structurants du paysage de la province de jambi, ainsi qu'a leurs dynamiques. La regionalisation du territoire jambinais montre trois zones agricoles. Les roles de ces zones sont analyses, soit au niveau provincial soit au niveau regional de sumatra. Puis, deux regions representatives de l'ensemble des situations provinciales sont selectionnees pour elaborer des methodes d'analyse des donnees de teledetection. Le travail se poursuit par des experimentations au niveau du traitement d'images. Cette etude repose sur des tests systematiques pour le choix des canaux a analyser et la comparaison de plusieurs algorithmes, ainsi que les evaluations des resultats. A la fin de ces experimentations, une methode generale et des regles locales sont definies. Les resultats du traitement d'images sont ensuite transformes en images thematiques. Une etude du paysage rural est effectuee a partir d'elles, ainsi qu'a partir des donnees exogenes et les donnees socio-economiques, obtenues par l'observation de terrain. En utilisant les informations satellitales, l'analyse du paysage peut etre realisee de facon plus precise et comprehensive. Cette contribution est tres utile dans l'etude de la transmigration.

Lymphocytes from patients with B-chronic lymphocytic leukemia (B-CLL) express large numbers of P2X7 receptors for extracellular adenosine triphosphate (ATP). Activation of P2X7 receptors induces multiple downstream effects, of which the best documented is the opening of an ionic channel that is selective for divalent cations. Another effect of ATP is to induce the shedding of L-selectin (CD62L), a molecule which is involved in the adhesive interactions of lymphocytes on endothelial cells. High levels of soluble L-selectin and CD23 are found in the serum of patients with B-CLL, although the mechanisms involved in their production are poorly characterized. Because extracellular ATP causes shedding of L-selectin, we studied the effect of ATP on shedding of CD23, an adhesion molecule expressed on the surface of B-CLL lymphocytes. ATP induced the shedding of CD23 at an initial rate of 12% of that for L-selectin, while the EC50 of ATP (35 uM) and BzATP (10 uM) was identical for shedding of both molecules. Inactivation of the P2X7 receptor by pre-incubation with OxATP, an irreversible inhibitor of P2X7 purinoceptor, abolished ATP-induced shedding of both molecules. Moreover, KN-62, the most potent inhibitor for the P2X7 receptor inhibited ATP-induced shedding of both CD23 and L-selectin with the same IC50 (12 nM). Ro 31-9790, a membrane permeant zinc chelator which inhibits the phorbol-ester stimulated shedding of L-selectin also inhibited shedding of CD23 from B-CLL lymphocytes, but the IC50 was different for the two shed molecules (25 versus 1 ug/ml respectively). Although L-selectin was completely shed by incubation of cells with phorbol-ester no CD23 was lost under these conditions. Also, Ca2+ inhibits ATP-induced CD23 shedding but not L-selectin shedding. Since soluble CD23 and L-selectin are found in the serum of normal subjects and B-CLL patients, the expression of these two adhesion molecules on lymphocytes before and after transendothelial migration was studied in an in vitro model of this process. In normal and B-CLL subjects, 71�b5% of L-selectin from both T and B cells and 90% of CD23 from B cells was lost following transmigration, while the expression of a range of other adhesion molecules such as VLA-4, ICAM-1, LFA-1 and CD44 was unchanged. Lymphocytes incubated with OxATP retained their capacity for transendothelial migration and showed the same loss of L-selectin as control leukaemic lymphocytes. Ro 31-9790, which can protect ATP-induced both L-selectin and CD23 shedding, had no effect on inhibiting L-selectin and CD23 lost during transmigration. These data show the presence of a second pathway for the downregulation of L-selectin and CD23 from the lymphocyte surface. Data in vivo from 'knock-out' mice show that L-selectin is essential for the emigration of lymphocytes through high endothelial venules into lymph nodes. The migration of normal and B-CLL lymphocytes across confluent human umbilical vein endothelial monolayers was studied in an in vitro model of this process. Lymphocytes treated with ATP or BzATP showed 56�b25% or 67�b16% loss of L-selectin on the surface and 36�b24% or 64�b19% decrease of transmigration, respectively, while OxATP, which does not alter the L-selectin level, had no effect on lymphocyte transmigration. Further experiments examined this correlation between L-selectin expression and lymphocyte transendothelial migration in this model system. A quantitative assay for cell surface L-selectin showed that expression of L-selectin was lower on B-CLL lymphocytes (8,880�b5,700 molecules/cell) than on normal lymphocytes (29,500�b7,500 molecules/cell, p less than 0.001). Also the rate of transmigration of B-CLL lymphocytes (1.5�b0.9 migrated cells/HUVEC) was lower than normal peripheral lymphocytes (2.4�b0.9 migrated cells/HUVEC, p=0.04). Incubation of lymphocytes in complete medium for 24 hrs increased the expression of L-selectin on B-CLL lymphocytes by 1.5 to 2 fold while the normal lymphocyte L-selectin remained at the initial level. This upregulation of B-CLL L-selectin correlated with a 2 fold increased rate of transendothelial migration. A correlation was found between L-selectin expression on lymphocytes and their ability for transendothelial migration (r^2=0.6). This study shows that the adhesion molecules L-selectin and CD23 can be lost from lymphocytes by two different physiological pathways. One is by P2X7 receptor activation by extracellular ATP while the second is activated by transendothelial migration of these cells. A second finding is that B-CLL lymphocytes have lower level of L-selectin expression and an impaired ability for transendothelial migration compared with normal peripheral blood lymphocytes. Do these results explain the high serum levels of soluble L-selectin and CD23 observed in B-CLL? Although B-CLL lymphocytes do not recirculate as rapidly as normal peripheral blood lymphocytes, the greatly increased number of leukaemic cells in B-CLL ensures that much more soluble L-selectin and CD23 is generated during the recirculation of these cells through the body.

Does learning English help undocumented Mexican transmigrants get better jobs in the United States? In this transborder ethnography, I worked with three households of undocumented people in Tucson, Arizona and traveled to their hometowns in Mexico, to better understand the context of their migration. For these migrants, speaking English did not lead to better jobs. Some employers tried to prevent them from learning English. Others were fired for using English to complain about unpaid wages. One person who was fired was replaced by a monolingual Spanish speaker. Many Americans think that all immigrants must learn English, and this discourse is common, both in the political and educational arenas. However, this study demonstrates that alongside this social discourse, there is a parallel economic discourse, urging the production of docile workers. Docility means not speaking English. Despite these findings, the discourse of "learning English in order to find better work" is a persistent one among the undocumented. I traced its origins and found that it begins shortly after a migrant arrives in the U.S. If English did not lead to better jobs, why did migrants learn it? For some people, it was because English helped them perform the identity of a U.S. citizen. They used self-consciously constructed semiotic and linguistic performances to appear Chicano/a, and these performances lessened their anxiety about deportation. For others, English was a conflicted symbol. Although it was a symbol of wealth, and therefore desirable, using it in public could easily reveal one's legal status to the wrong interlocutor. There are significant obstacles to the use of English among undocumented Mexican transmigrants, and language use is essential for language mastery. This study encourages those who teach English to Speakers of Other Languages (ESOL) to understand the social structures that impact their students' language use. With implications for education, border, and immigration policy, this study sheds light on the lived experiences of undocumented migrants and brings language and language use into conversations about globalization. Understanding transmigrants' experiences and ideologies offers a new lens to theorizing social inequality and human agency, and ultimately, to creating more humane borders.

Neutrophil (PMN) inflammatory functions, including cell adhesion, diapedesis, and phagocyto-sis, are dependent on the mobilization and release of various intracellular granules/vesicles. In this study, I found that treating PMN with damnacanthal, a Ras family GTPase inhibitor, resulted in a specific release of secondary granules, but not primary or tertiary granules, and caused dy-sregulation of PMN chemotactic transmigration and cell surface protein interactions. Analysis of the activities of Ras members identified Ral GTPase as a key regulator during PMN activation and degranulation. In particular, Ral was active in freshly isolated PMN, while chemoattractant stimulation induced a quick deactivation of Ral that correlated with PMN degranulation. Over-expression of a constitutively active Ral (Ral23V) in PMN inhibited chemoattractant-induced secondary granule release. By subcellular fractionation, I found that Ral, which was associatedwith the plasma membrane under the resting condition, was redistributed to secondary granules after chemoattractant stimulation. Blockage of cell endocytosis appeared to inhibit Ral transloca-tion intracellularly. In conclusion, these results demonstrate that Ral is a critical regulator in PMN that specifically controls secondary granule release during PMN response to chemoattrac-tant stimulation.

Severe septicaemia due to the bacterium Neisseria meningitidis is characterised by substantial recruitment and activation of neutrophils on inflamed endothelium, and is associated with vascular damage. N.meningitidis associates with endothelial cells, E-selectin, and neutrophils in patient biopsies, and E-selectin can co-localise beneath adherent bacteria in vitro. This thesis aims to investigate whether N.meningitidis affects E-selectin distribution on the endothelial surface and whether meningococcal-E-selectin interactions have functional consequences on neutrophil rolling, adhesion and transmigration across vascular endothelium. Lentiviral constructs were created containing full-length E-selectin (ES) or a cytoplasmic domain deficient mutant (ΔC). Transfection of primary HUVEC demonstrated that ES E-selectin forms spontaneous clusters, while ΔC is expressed in a diffuse membrane pattern. Capsulated N.meningitidis was unreliable at inducing E-selectin clustering and co-localisation. In contrast, an unencapsulated SiaD- strain could co-localise with endogenous IL-1β induced E-selectin, although not with transfected ES or ΔC. Thus the E-selectin cytoplasmic domain affects both the molecule’s distribution on the endothelial membrane, and also N.meningitidis mediated redistribution, which additionally requires some factor present on IL-1β stimulated, but not ES or ΔC transfected, endothelium. Functional aspects of N.meningitidis interactions with E-selectin on neutrophil behaviour were also investigated. Brief co-culture of N.meningitidis with ES transfected HUVEC induced increased neutrophil adhesion under static conditions and greater transmigration under physiological flow. Neutrophil adhesion to N.meningitidis stimulated ΔC transfected HUVEC however, was no greater than untransfected cells. ΔC HUVEC also only saw a small increase in transmigration despite having comparable adhesion and rolling velocity to ES cells under flow. Taken together these results show that the E-selectin cytoplasmic domain has an important yet unstudied role in neutrophil adhesion and transmigration. These data should lead to a better understanding of the underlying processes surrounding neutrophil mediated endothelial damage caused by N.meningitidis, and could have broader relevance for other inflammatory responses, particularly in sepsis.

C'est en fonction du raisonnement tres simple selon lequel la naissance de fronts pionniers suppose d'abord l'existence d'un contraste marque entre regions tres peuplees et regions peu humanisees, puis une dynamique susceptible de conduire les populations des regions densement occupees vers les espaces demeures forestiers, que le probleme de la colonisation agricole en indonesie a ete aborde. Le premier livre rend compte du desequilibre demographique qui oppose java et bali aux autres iles de l'archipel. Ce contraste remonte a la nuit des temps; il s'est simplement transforme en desequilibre au xixe siecle sous l'influence du "systeme des cultures". Le deuxieme livre aborde les problemes lies a la dynamique des differents groupes. Il permet de relativiser l'ampleur des migrations javanaises, sundanaises et balinaises, d'en souligner le caractere tres recent, et de faire ressortir, a cote des facteur d'ordre economique, l'existence de mobiles d'ordre culturel trop souvent ignores. L'etude des migrations organisees ne se limite pas a l'etude de la transmigration. Il existe, en effet, une filiation entre le programme colonial de kolonisatie et le programme de transmigration de l'indonesie independante meme si les obj ectifs assignes a ces deux entreprises ont considerablement evolue avec le temps. Enfin, le troisieme et dernier livre traite de la place des terres neuves dans l'organisation de l'espace indonesien contemporain. Il permet de constater que les migrations n'ont eu un impact veritablement important qu'a sumatra. Chemin faisant un certain nombre d'idees recues ont ete remises en question. L'idee d'un "exceptionnalisme" javanais a ete fortement nuancee, de meme que l'idee selon laquelle ce serait la pression demographique insoutenable qui serait reellement responsable de l'exode de millions de paysans. Au final, la conclusion qui s'impose est que l'espace national indonesien se structure aujourd'hui de la meme facon que se sont structures en leur temps mojopahit et mataram. C'est en grignotant les espaces forestiers, en multipliant les domaines agricoles, puis en federant les clairieres entre elles grace a un bon reseau de chemins en terre, que s'est elaboree la royaute javanaise. C'est en reliant entre eux les villages neufs par un dense lacis de routes secondaires bien entretenues, que la foret recule dans les iles exterieures et que les provinces peripheriques sont progressivement integrees a la construction nationale
In a very simple way, agricultural colonization in indonesia as anywhere else, supposes first a contrast between overcrowded and sparsely populated areas, then vigourous migratory flows from densely populated towards empty regions. Accordingly, the first part of the book tries to explain the demographic imbalance between java and the so-called "outer islands". This is a very old phenomenon which began acute during the xixth century under the pression of the "cultivati on system". The second part is devoted to the various migratory flows that swelled up all around the archipelago. This i s an occasion to show that beside javanese, sundanese and balinese migrations which finally are quite recent, numerous peoples spread over the different islands for a long time. In fact, economic difficulties and poverty are not the only key factors that explain the peasants'decision to sailed out their native island, cultural reasons are as less as important. Ring expansion is allowed by formation of new invadopodia at the ring periphery and disassembly of invadopodia at the ring center. The tyrosine phosphatase inhibitor, orthovanadate, enhances fast ring expansion indicating the invovlment of tyrosine phosphorylation processes both in invadopodia formation at the periphery and invadopodia disassembly at the center of the ring. The protein paxillin which is higly phosphorylated on tyrosine and promotes focal adhesion disassembly, is also a component of invadopodia and induces invadopodia disassembly at the ring center (main process of ring formation and expansion) via paxillin phosphorylation on tyrosine 31 and 118 driving to downstream Erk and calpain activations, which induce the clivage of invadopodia components

Resumen

This qualitative interpretive study explored how former newcomer English Language Learners (ELLs) in Boston Public Schools (BPS) made sense of their transmigration experiences through a digital storytelling project. The study fills a gap on transmigration experiences in the context of English-only learning environments, with a particular orientation toward the value of students’ home languages, and in turn, cultures within an urban school setting. The immigrant student population in BPS continues to increase and teachers must be able to understand and plan for newcomers’ specific needs. To this end, my conceptual framework drew on four areas: general educators and their urban students, with sections on teachers’ habitus and the hidden curriculum; cultural relevance in urban education; identity, including social identity theory and transnational identity; and school adjustment, with sections on student voice and social integration. Set in a community center in Boston and drawing on ethnographic methods, the study explored participants’ unique educational experiences in moving from Haiti to different academic programs in BPS.

The overarching research question was: How do former newcomer ELLs make sense of their transmigration experiences through a digital storytelling project? I used a combination of methods—interviews, participant observations, photography, and digital storytelling—to gather and analyze artifacts. Data analysis highlighted issues related to the process of conducting the digital storytelling project, as well as issues emerging from the participants’ narratives of transmigration. Analysis of the research process revealed a distinction between how teacher preparation and teacher preparedness might be understood for educators working with newcomer ELLs. Analysis of participant narratives suggests that language holds central importance to the transmigration and social integration experiences of newcomers. Furthermore, digital storytelling, as a pedagogical process, may serve as an effective tool for working with newcomer ELLs, both as a means to facilitate meaning making and to give significance to their transmigration experiences. It may also serve as an effective means to support language development. Adding to the body of immigration literature on how newcomers fare, the study’s implications include the role of language in social integration, ways of understanding teacher preparation and preparedness, and the utility of digital storytelling.

HIV associated neurocognitive disorders (HAND) encompass a spectrum of cognitive deficits that affect 40-70% of HIV-infected individuals, despite antiretroviral therapy. Monocytes are among the first cells infected by HIV and are critical mediators of HAND as they facilitate viral seeding of the central nervous system (CNS) upon their transmigration across the blood brain barrier (BBB). Monocyte subpopulations exist with differing levels of maturation and functions. Monocytes that express CD14, the LPS receptor, and CD16, the Fcylll receptor, are a mature population of cells that are highly susceptible to HIV. While CD14+CD16+ monazites are believed to mediate the neuropathogenesis of HIV, little is known about the mechanisms that contribute to their diapedesis across the BBB. As the CD14+CD16 + monocyte represents a small percentage of monocytes in healthy individuals, we developed a tissue culture model to enrich for this population. We found that there was a selective transmigration of CD14+CD16 + monocytes across our BBB model, with little migration of other monocyte populations. HIV infection resulted in the increased transmigration of CD14 +CD16+ monocytes in response to CCL2 relative to uninfected cells, which was due to increased CCR2 and a heightened sensitivity to the chemokine. The junctional proteins JAM-A and ALCAM were also critical for this transmigration as antibody blockade reduced the number of migrating monocytes. These CD14+CD16+ monocytes were present in significantly greater numbers in HIV-infected people, despite viral suppression, in contrast to individuals without HIV. CCR2 was increased on CD14+CD16+ monocytes in HIV-infected individuals with HAND compared to those with normal cognition and was predictive of fluctuations in cognitive impairment upon longitudinal study. ALCAM and JAM-A were increased on CD14+CD16 + monocytes in those with HIV compared to HIV seronegative people. Blocking antibodies to ALCAM and JAM-A inhibited the transmigration of CD14 +CD16+ monocytes, but not of T cells, suggesting their importance in specifically facilitating monocyte transmigration across the BBB. Our findings indicate therapeutic strategies to monitor HAND, and that may decrease the entry of CD14+CD16+ monocytes into the CNS of HIV-infected individuals, contributing to the eradication of neuroinflammation, HAND, and CNS viral reservoirs.

In the 1970s, the Indonesian government undertook a massive national development program which involved the relocation of 1.5 million people throughout the islands of the archipelago. Known as transmigration, the program resettled people from Java and Bali, two islands experiencing overpopulation, urbanization and increasing poverty, to the "Outer Islands" of Sumatra, Kalimantan, Sulawesi, the Moluccas and Papua. One of the objectives of transmigration was the integration of the many ethnic and religious sub-communities throughout Indonesia to fashion Indonesian citizens which collectively would represent the national motto of Bhinneka Tunggal Ika, or "Unity in Diversity." Unfortunately, many of the transmigration settlements were established as exclusively Javanese or Balinese enclaves resulting in instances of inter-communal conflict with the indigenous groups. This thesis examines the unusual transmigration settlement of Mopugad, in Northern Sulawesi. Here the locus of integration is between two groups settled in the same town, creating an opportunity to assess whether the shared experience of migration is a condition of unification.

We can see that in Mopugad the two communities, one Javanese and the other Balinese, have remained largely distinct and are apparently becoming increasingly distinct due to the evolution of religious culture. The relationship between the two communities can partly be seen in the negotiation of cultural and administrative jurisdictions visible in the changing physical order of the town. Though diversity has been sustained at the expense of unity, it is not impervious to the changing circumstances facing the town which could allow a change in trajectory towards increased unity and a diminished diversity. Should residents of Mopugad jointly decide that diversity is a goal worth pursuit, they will have to work deliberately to sustain it by building local interdependence. The impending threat that nearby informal gold mining poses to the health and rice-farming livelihood of both sets of residents may be an opportunity upon which to base a conditional community, a precursor to shared communalism. The resulting shared communalism would be particularly applicable in other parts of the nation as Indonesia undergoes massive political and fiscal decentralization. The children of the pioneers of transmigration have the opportunity to become the new pioneers of decentralization.

This exegesis outlines the procedures, processes and methodologies used in developing a body of studio work that investigates the artistic potential of scientific photomicrography made by the Scanning Electron Microscope (SEM) with a view to its possible social and cultural impact of this practice. The outcomes of this research are of two kinds: theoretical findings and the work developed and exhibited during the candidature and at the final exhibition, H2O+. Reflecting the chemical composition of water and interpreting photomicrographic images of it, my practice seeks to provide insights and awareness about water quality, that might be later used to encourage behavioural change to water management, and demonstrate productive transdisciplinary relationships and innovation between the arts and sciences. Artistic manipulation of a scientific process through experimenting with the SEM fuses science and technology with art and proposes a new meaning for what such images can say about water to a viewer. Transforming the microworld to a macro level causes a dilemma for viewers, mainly because enlarged photomicrographs placed in the gallery space are seen differently from those viewed for their scientific information within the laboratory. An aesthetic engagement with photomicrography gives viewers the possibility to see in the particular configurations of inherent features of water opening a horizon of meaning that was previously unseen as well as referring to the creative ability to observe natural phenomena over and beyond the directly visible. Building on the fact that scientific tools have brought new ways of seeing the world, the primary purpose of this research is to show that the artistic use of SEM-made photomicrography can shift the visual outcomes of scientific photomicrography to function within the context of art.

The small intestinal permeability barrier is dependent on tight junction (TJ) complexes that separate the external lumen from the underlying mucosa. Apical TJs consist of integral transmembrane proteins including occludin, claudins and ZO-1 as well as the cytoplasmic plaque of TJ-associated adaptor, scaffolding and signalling proteins. Although the function of occludin at the TJ remains unclear, the dynamic mobility of occludin, claudins and ZO-1 to and from the TJ suggests occludin may play a key role in regulation of TJ structure and function, regulated by occludin phosphorylation status. Defects in TJ barrier function have been implicated in a range of inflammatory diseases such as inflammatory bowel disease (IBD) and pathogens such as Toxoplasma gondii target this complex as a route of infection. As oral infection is the primary cause of toxoplasmosis, the first point of contact between T. gondii and the host is the small intestinal epithelium and studies by Weight, 2012 show occludin may be involved in T. gondii paracellular transmigration of the small intestinal epithelium. The aim of this research was to investigate T. gondii paracellular transmigration using an in vitro model of the small intestinal epithelium and elucidate the role of occludin both in regulation of the TJ barrier and as a receptor for T. gondii infection. The results presented in this thesis demonstrate that T. gondii infects the small intestinal epithelium via the paracellular pathway and occludin was shown to play a key role both in regulation of the TJ paracellular barrier and as a receptor for T. gondii infection by parasite-mediated modulation of the occludin C-terminus phosphorylation status and direct binding to occludin ECL1; suggesting T. gondii interactions with occludin are a potential mechanism of paracellular transmigration of the small intestinal epithelium.

Auch unter physiologischen Bedingungen finden sich T-Zellen und andere Leukozyten nicht nur in den Sinusoiden, sondern auch im Parenchym der Leber. Da die Leber u. a. verschiedene Aufgaben für das Immunsystem übernimmt (z. B. Deletion aktivierter T Zellen, Induktion peripherer Toleranz), könnte die Akkumulation der T-Zellen in der Leber - neben der immunologischen Überwachung der Leber - Voraussetzung für ihre Modulation sein. In der vorliegenden Arbeit wurde der Einfluss von Leber-sinusoidalen Endothelzellen (LSEC), der Barriere zwischen Blut und Leber-Parenchym, auf CD4+ T-Zellen untersucht. Zum einen zeigte sich, dass die LSEC sowohl die spontane Transmigration der T-Zellen, als auch ihre Chemotaxis zu CXCL9 und CXCL12 effizienter unterstützen als andere Endothelien. Eine endotheliale Aktivierung durch die Chemokine wurde als Mechanismus ausgeschlossen. Dagegen schien eine effiziente Präsentation der Chemokine auf der luminalen LSEC-Oberfläche nach Aufnahme von abluminal für die gesteigerte Transmigration der T Zellen verantwortlich zu sein. Die LSEC könnten somit in vivo an der Rekrutierung von T-Zellen in die Leber beteiligt sein, indem sie eine rasche Wanderung der T-Zellen aus dem Blut ins Parenchym und möglicherweise auch zurück in die Zirkulation zulassen. Des Weiteren konnte gezeigt werden, dass die LSEC fähig sind, naive CD4+ T-Zellen in vitro Antigen-spezifisch zu aktivieren. Im Vergleich zu professionellen APZ war hierfür eine höhere Antigen-Dosis notwendig, die Expansion schwächer und es waren kaum Effektorzytokin-Produzenten detektierbar. Diese konnten jedoch durch Restimulierung mit professionellen APZ induziert werden (reversibler Phänotyp), was auf einen unreifen Differenzierungsstatus der T-Zellen schließen ließ. Es bleibt zu prüfen, in welchem Maße die Aktivierung naiver CD4+ T-Zellen durch LSEC in vivo stattfindet und diese durch LSEC aktivierten CD4+ T-Zellen funktionelle Bedeutung, z. B. regulatorische Kapazität, für das Immunsystem besitzen.
The liver plays a major role for the metabolism, but it is also of general importance for the immune system, e.g. for the deletion of activated T cells or the induction of peripheral tolerance. Under physiological conditions T cells and other leukocytes can be found in the liver, in the sinusoids as well as in the parenchyma. This hepatic accumulation of T cells might be due to immunosurveillance, but it would also be a prerequisite for modulation of T cells by hepatic cells. The present study investigated two different aspects of the interaction of liver sinusoidal endothelial cells (LSEC), the barrier between the sinusoidal lumen and the hepatic parenchyma, and CD4+ T cells. In the first part of the study it could be demonstrated that LSEC support the spontaneous transmigration of CD4+ T cells as well as their chemotaxis to CXCL12 and CXCL9 more efficiently than other endothelial cells. Whereas a direct endothelial activation by chemokines could be excluded the efficient chemokine presentation at the luminal LSEC surface (after abluminal uptake) might be responsible for the enhanced T cell transmigration. The findings suggest that LSEC might be involved in the recruitment of T cells by supporting a rapid transendothelial migration. The second part of the study focused on the characteristics of LSEC in the context of antigen presentation. LSEC were able to prime and expand naïve CD4+ T cells in vitro but less effective than professional APC as proven by weaker expansion of cells, a requirement for higher antigen concentration and the lack of cytokine producing T cells. The “immature effector” phenotype of the CD4+ T cells primed on LSEC was reversible since it could be overcome by restimulation on professional APC. In conclusion these data suggest that antigen presentation by LSEC results in activation but incomplete differentiation of CD4+ T cells.

Les cellules dendritiques (CDs) patrouillent les tissus et transportent les antigènes vers les ganglions lymphatiques pour initier des réponses immunitaires adaptatives. Au sein des tissus, les CDs constituent une population cellulaire complexe composée de sous-types distincts pouvant présenter différents états d'activation et fonctions. La façon dont les signaux tissulaires orchestrent la diversification des CDs reste insaisissable. La Lamina Propria (LP) de l'intestin grêle (IG) est enrichie par une population particulière de cCD2 exprimant les intégrines CD103 et CD11b. Une fraction de ces cellules peut transmigrer dans l'épithélium à l'état d'équilibre et en proportion plus élevée lors d'une infection. Cependant, les conséquences d'un tel événement sur l'identité et le destin de ces cellules sont inconnues. En utilisant l'analyse de séquençage d'ARN unicellulaire, nous avons constaté que la transmigration des CDs CD103+CD11b+ dans l'épithélium modifie profondément leur profil transcriptomique caractérisé par une baisse d'expression des gènes inflammatoires et une augmentation d'expression des gènes associés à l'activité antimicrobienne. Nous avons ensuite décrit que l'intestin grêle comprend deux pools de cCD2s provenant de précurseurs de préCDs communs : (1) cCD2s CD103+CD11b+ de la LP qui sont des cellules pro-inflammatoires de type mature et (2) des cCD2s intraépithéliales qui présentent un phénotype similaire à celui des CDs immatures induisant des propriétés tolérogènes des lymphocytes T. Ce phénotype résulte de l'action de l'acide rétinoïque d'origine alimentaire (ATRA), qui améliore la contractilité de l'actomyosine et favorise la transmigration des cCD2s de la LP vers l'épithélium. Les cCD2s sont alors influencées par des indices environnementaux spécifiques à l'épithélium comprenant l'ATRA lui-même ainsi que le composant muqueux Muc2. Par conséquent, en atteignant des niches sous-tissulaires distinctes, les CDs peuvent exister sous forme de cellules immatures et matures dans le même tissu, révélant un nouveau mécanisme de diversification fonctionnelle des CDs
Dendritic cells (DCs) patrol tissues and transport antigens to lymph nodes to initiate adaptive immune responses. Within tissues, DCs constitute a complex cell population made of distinct subsets that can exhibit different activation states and functions. How tissue-specific cues orchestrate DC diversification remains elusive. Particularly, the small intestine (SI) Lamina Propria (LP) is enriched in a peculiar population of cDC2s expressing the integrins CD103 and CD11b. Interestingly, a fraction of these cells can transmigrate into the epithelial layer both at steady state and in higher proportion upon infection. However, the consequences of such event on the identity and fate of these cells is unknown. By using single cell RNAseq analysis, we found that their epithelial colonization deeply modifies their transcriptomic profile, downregulating inflammatory genes expression and stimulating the transcription of antimicrobial genes. We then further described that the small intestine includes two pools of cDC2s originating from common preDC precursors: (1) lamina propria CD103+CD11b+ cDC2s that are mature-like pro-inflammatory cells and (2) intraepithelial cDC2s that exhibit an immature-like phenotype and induce tolerogenic T lymphocyte properties. Intraepithelial cDC2 phenotype results from the action of food-derived retinoic acid (ATRA), which enhances actomyosin contractility and promotes LP cDC2 transmigration into the epithelium. There, cDC2s are imprinted by environmental cues including ATRA itself and the mucus component Muc2. Hence, by reaching distinct sub-tissular niches, DCs can exist as immature and mature cells within the same tissue, revealing a novel mechanism of DC functional diversification
