In a supposedly enlightened and disenchanted age, why has lyric poetry continued to make claims and perform gestures that are now otherwise inadmissible or even unimaginable? Animation, invocation, and unmotivated praise, apparently artificially imposed (dis)order, and spurious gnomic and vatic sayings that pretend to universal or transcendent knowledge are marks of the lyric as a genre. Sketching a theory of poetic license, this article addresses the lyrical entanglement of enchantment and embarrassment. The author argues for a concept of the lyric as a medium for regulating the balance between enchantment and disenchantment in an always imbalanced environment. Engaging other scholars and using examples from modern French and German poetry, the article also ventures a new understanding of lyric modernity. Rather than naming a historical event to be lamented, disenchantment unveils a risk inherent to the lyric whose regulatory function it makes explicit.

Resumen
Ancient Greek praise poems Arguing from both the surviving texts themselves and from ancient theorists, the present article deals with early Greek lyric poems in praise of human beings. This type of lyric falls under the more “secular types” of ancient Greek lyric, in the sense that they were addressed, not to a divine being, but to a human being. The context or space of such “secular” lyric performance includes, not only the public gathering of officials and the populace, but also the private and intimate circle of individuals with shared interests. Both choral odes and solo-lyrics are therefore involved. The lyric types discussed are the praise poem, the war poem, the political poem and the dirge.

The concept of lyric is a central component of Paul de Man’s theory of poetry. In several papers but first of all in his „Anthropomorphism and Trope in the Lyric” he developed a coherent understanding of the lyric as an act of modernism. Its epistemological character was given through Nietzsche’s often quoted statement on the metaphorical, anthropological and performative nature of subjective language and language use. The development of these ideas, however, took place through a detailed text-interpretation of two Baudelaire poems that served as metapoetical definitions of different poetical stances. It is possible that the idea of the lyric and reading lyric defined through anthropomorphism and phenomenality refers to, can be read like an elaborated epistemology of modern, subjective textuality too.

This thesis is an examination of the subsection of T. S. Eliot's The Waste Land that is aptly named "The Fire Sermon." The hybrid nature of this famous poem makes it open to a variety of readings, and these readings are often conflicting. Thus, the work, in spite of being a seminal text in literature, can be difficult to teach due to the complexity of the piece itself. This fact makes choosing a pedagogical approach to teaching The Waste Land a challenge. With the goal of making Eliot's poem more explorable, this thesis will undertake the task of an examination of "The Fire Sermon" using two distinctive theories. The theories in question are the theory of the lyric, exemplified by Jonathan Culler's writing, and the theory of heteroglossia established by Mikhail Bakhtin. However, that analysis will be merely a stepping stone for a more strictly pedagogical question that this project seeks to answer. That question is, namely, the query of which branch of contemporary theory, narrative or lyric, is more apt to present the issues inherent in "The Fire Sermon" in an effective and teachable manner. Both positions have a number of positive attributes and elements that make them uniquely suited to the examination of Eliot's writing.
Master of Arts
Teaching poetry can be a difficult task. The basic question of “Why should I study poetry?” is one that many a professor has had to answer. While the scholarly community has done a decent job of articulating the value of the liberal arts, the specifics of how to teach difficult poetry is more of a gray area in scholarship. Certainly, a number of articles, opinions, and theories on how to best teach poetry exist, but creating a clear blueprint with examples of how to apply complex theories to a poem is essential to guiding new instructors into the field of teaching poetic works before an audience. This thesis is a work that shows several of the methods of studying poetry via an examination of several important poetic and narrative theories and the theorists that created said methods, and then the thesis undertakes a practical examination of a poem, a section of T. S. Eliot’s The Waste Land. The purpose of this thesis is to make critical theories and abstract ideas more applicable and valuable as usable tools in the classroom, rather than having them exist as ideas without a practical application. Knowledge is, after all, something made to be shared.

The Middle English lyric, “Sir John Doth Play,” narrated in the female voice clearly depicts rape by a male authority figure and the narrator’s distress over her unplanned pregnancy, yet has been repeatedly interpreted and introduced by critics as a love song portraying the interaction of a seductive village priest and a gullible maiden. As such, the lyric provides a unique perspective on the patriarchal nature of the twentieth century and the value of critically re-examining literature rather than canonizing accepted interpretations of literary work.

How we approach a poem changes it. Recently, it has been suggested that one readerly approach - a bodily orientation characterised by distance, suspicion, and resistance - risks becoming reflexive, pre-conscious, and predominant. This use-oriented reading allows us to destabilise, denaturalise, dissect, defend, and define poetic texts through its manifestation in contemporary literary critique, yet it is coming to be regarded as the sole manner and mood of intelligent, intellectual engagement. In this thesis, I demonstrate the need to pluralise this attentive orientation, particularly when it comes to contemporary lyric poetry. I suggest how an overlooked mode of response might foster a more receptive mode of approach: the 'co-creative' response. Lyric poems mean to move us, and they come to mean by moving us. Recent 'simulation theories of language comprehension', from the field of cognitive neuroscience, provide empirical evidence that language processing is not a product of a-modal symbol manipulation but rather involves 'simulations' by certain classes of neurons in areas used for real-world action and perception. As habituation and abstraction increase, however, these embodied simulations 'streamline', becoming narrow schematic 'shadows' of once broad, qualitatively rich simulations. Poems, I suggest, seek to reverse this process by situationally novel variations of language, coming to mean in the broadly embodied sense in which real-world experiences 'mean'. Readers are asked to 'enact' the poem, to 'co-create' its meaning. Where critique traditionally requires that readers resist enactive participation in the aim of objective analysis, the co-creative response - a response 'in kind' by imitation, versioning, or hommage - asks readers to receive and carry forward the enactive unfolding of a poem with a composition of their own. I assert that, by thus responding with - rather than to - poems, we might foster an attentive stance of active receptivity, thereby coming to understand poems as the enactive phenomena they are.

This paper aims to study upper secondary school seniors’ relationship to poetry. The researchers seek to discover the students’ conceptualization of the subject of poetry. Fur-thermore, this paper will present a case study, in which sixteen students participate. The chosen research approach is Glaserian Grounded Theory, which in itself has a con-siderably comprehensive methodology. However, working with Grounded Theory enables an objective approach to the field of study, requested by the researchers. In course of this study, the researchers found that the lack of experi-ence in working with poetry was the students’ main con-cern. The theory, which was generated, therefore evolved from the previously mentioned main concern.

Cette étude se consacre à l’avènement du Je anti-lyrique dans la poésie italienne des années soixante. C’est à ce moment que la pensée post-métaphysique peut se dire assimilée par la culture italienne, qui y trouve une nouvelle clef de lecture de l’expérience de la guerre et du miracle économique. C’est aussi la période où le réel, cru et changeant, fait irruption dans l’expérience artistique. Elio Pagliarani, Mario Luzi et Vittorio Sereni nous ont parus des représentants emblématiques de cette nouvelle posture du sujet poétique. Nous avons étudié leurs ouvrages des années soixante par le biais de trois catégories principales : la mise en question du Je, le rapport entre le Je et le monde, le rapport entre le Je et les autres. Cette articulation nous a permis d’une part de suivre les trois étapes principales de la reformulation anti-lyrique du Je (face à lui-même, face au monde et face aux autres), et d’autre part de reconnaître une progression dans la perte de l’identité lyrique traditionnelle. Le poète, ayant perdu la consolation métaphysique, commence à rechercher la vérité dans le monde physique ; il comprend ensuite que la vérité des choses est éphémère et que toute unité est brisée ; et il donne enfin la voix à d’autres personnages, dans l’effort de comprendre ce questionnement polyphonique qui est désormais la véritable voix du monde et du poète lui-même
This research focuses on the rise of the anti-lyrical I in the Italian poetry of the Sixties. This was the time when the post-metaphysical philosophy had been fully assimilated by the Italian culture, which started using it as a new reading key to under- stand WW2 experience and the so-called economic miracle. This was also the time when the raw and ever-changing real broke into the artistic experience. Elio Paglia- rani, Mario Luzi and Vittorio Sereni seemed to us emblematic representatives of this new poetic subject's posture. We studied their poems from the Sixties under three main categories: the questioning of the I, the relationship between the I and the world, and the relationship between the I and the others. This structure allowed us firstly to follow the three main stages of the anti-lyrical reformulation of the I (confronted to himself, to the world and to the others) and secondly to recognize a progression in the loss of the traditional lyrical identity. Aware of the loss of any metaphysical consolation, the poet begins to seek the truth inside the physical world; he the realizes that even objects/things bear a fleeting truth and that any unity is broken; and he finally gives voice to other characters in the effort to understand the polyphonic questioning which has become by now the real voice of the world and of the poet himself

This study investigates the translation of song lyrics and presents an analysis of translation outcomes. While the majority of previous studies regarding song translation focuses on the translation of lyrics from musicals and operas, this study focuses on the translation of popular song lyrics. The lyrics of eight German songs and their English versions were analysed using the approximation approach (Franzon 2009) which divides smaller textual units into the categories paraphrase, metaphrase and addition. The target texts (henceforth TT) were also categorised according to Peter Low’s (2013) song translation categories, translation, adaptation and replacement text. The aim of this study is to determine to what degree the meaning of the source texts (henceforth ST) is transferred into the TTs and to determine how Franzon’s approximation approach can be used to determine if the TTs are translations, adaptations, or replacement texts. This study found that all TTs were mostly made up of paraphrases and metaphrases, which means that all TT derived to most parts directly from the STs i.e. the TT was written using mostly direct and oblique translations. Due to the low frequency of additions and all significant details of the STs being transferred into the TTs, all song translations analysed in this study were categorised as translations.

This thesis represents an analysis of symbolic expressions used to convey traditional Chinese cultural values in marital relations as expressed through costumes and lyrics in Peking Opera plays and performances. Two symbols, dragon and phoenix, were selected from the costume collection. Four symbols--bird, tiger, wild goose, and dragon--were selected from compilations of lyrics. These symbols were selected because they expressed Chinese core cultural values, an imperial ideology based on Confucian thoughts, which were practiced rigidly during Qing Dynasty (1644-1911). Modeling Theory is applied to argue that dragon and phoenix as visual symbols convey ideas about characters' background, marital relationship, social status shifts, and socio-culturally desirable values. Social Drama Theory is employed to analyze the lyrics to understand how ideal images of husband and wife are constructed. The archetypes of Chinese traditional culture that have influenced Chinese thought and action for centuries are discovered and discussed.

The purpose of this study is to find and analyse conceptual metaphors in the lyrics, A Thousand Kissed Deep, Here It Is, and Boogie Street from the album Ten New Songs (2001) by Leonard Cohen using Conceptual Metaphor Theory (CMT). In order to detected the conceptual metaphors, the source and target domains were identified. Conceptual metaphors were found by mapping source domains onto target domains and viewing the lexical expressions in the lyrics. The result and analysis of the findings in this study show that linguistic expressions of LOVE, LIFE and DEATH are conceptually present in the lyrics.

Resumen
Music lyrics can convey a great part of the meaning in popular songs. Such meaning is important for humans to understand songs as related to typical narratives, such as romantic interests or life stories. This understanding is part of affective aspects that can be used to choose songs to play in particular situations. This paper analyzes the effectiveness of using text mining tools to classify lyrics according to their narrative contexts. For such, we used a vote-based dataset and several machine learning algorithms. Also, we compared the classification results to that of a typical human. Last, we compare the problems of identifying narrative contexts and of identifying lyric valence. Our results indicate that narrative contexts can be identified more consistently than valence. Also, we show that human-based classification typically do not reach a high accuracy, which suggests an upper bound for automatic classification. narrative contexts. For such, we built a dataset containing Brazilian popular music lyrics which were raters voted online according to its context and valence. We approached the problem using a machine learning pipeline in which lyrics are projected into a vector space and then classified using general-purpose algorithms. We experimented with document representations based on sparse topic models [11, 12, 13, 14], which aims to find groups of words that typically appear together in the dataset. Also, we extracted part-of-speech tags for each lyric and used their histogram as features in the classification process.

The paper focuses on the interlacement of literary and iconographic elements by displaying an innovatory philological and stylistic approach, from a comparative perspective, in thematizing multilingual translational and adaptive aspects, ranging across Bernard Berenson's diaristic and epistolary corpus, in conjunction with his works on Italian visual culture. This interweaving gives occasion to the elaboration of multilinguistic textual influences and their verbo-visual artistic representations deduced from his innovative interpretative readings in the domain of world literature in modern times. Such analysis of the discourse of theoretical and literary nature, and of the pictoricity, refers to Bernard Berenson's multilingual considerations about canonical authors in English, Italian, French, German language, belonging to the Neoclassical and Romantic period, as well as to the contemporary era, as conceptualized in his autobiographical works, in correlation with his writings on Italian figurative art. The scope of this presentation is to discern and articulate Berenson's aesthetic ideas evoking literary and artistic modernity, that are infused with crucial notions of translational theory and conveyed through the methodology of close reading and comprising at the same time, in an omnicomprehensive manner, a plurality of tendencies intrinsic to social paradigms of cultural studies. Unexplored premises reflecting Berenson's vision of Italian culture, most notably of a visual stamp, will be analyzed through author's understandings of such adaptive translations or volumes to be subsequently translated in Italian, and through their intertwined intertextual applications, significantly contributing to further critical and hermeneutic reception thereof. Particular attention is drawn to its instancing in the field of Romantic literary production (Emerson, Byron), originally underscoring the specificities of each literary genre and expressive mode, of the narrative, lyric or theatrical nature, as well as concomitantly involving parallel notions as adapted variants within visual arts, and in such a way expressing theoretical views pertainable to Italian artworks too. Other analogous elements relevant to literary expression in the most varied cultural sectors such as philosophy, music, civilisational history (Goethe, Hegel, Kant, Wagner, Chateaubriand, Rousseau, Mme de Staël, Taine) are furnished, as well as the examples of the resonances of non-western cultures, with the objective of exploring the effect among readership bringing also to the renewal of Italian tradition.

This paper proposes a temporal analysis for music metadata using a generative probabilistic model for collections the discrete datasets such as text corpora. This method is also a topic model that is used for discovering abstract topics from a collection of documents. The method is then applied to audio metadata and song lyrics extracted with Echo Nest® engine, Spotify® Lyrics Genius® API. Song data time series are generated by grouping data items by release date, genre and dominant topics (from LDA analysis). Using a technique from Network Theory we visualise how these topics, in this case, genres, are related to each other through time.

The study aims to identify metaphors of SEASON in English and Vietnamese song lyrics in the light of cognitive linguistics. To be specific, the study follows the theory of conceptual metaphors which was initiated by Lakoff and Johnson (1980). Within the scope of the paper, the entity SEASON is treated as a target domain which is conceptualized through the source domain SPACE, which is examined via the corpora built from English and Vietnamese song lyrics composed the duration of the 20th century onward. By employing descriptive and comparative methods, and adopting the procedure of conceptual metaphor identification (Steen, 2011), the results show that English and Vietnamese share 2 conceptual metaphors of SEASON, namely, location and path. Accordingly, this study functions as an attempt to contribute to the area of metaphor research in cognitive perspective in Vietnam.

n the eighty’s poetry from the left of the Prut, a kind of self-referential writing is highlighted, defined by literary exegesis by the term textual narcissism (Cristian Moraru). This own lyrical manner belongs the poets Emilian GalaicuPăun and Nicolae Leahu and It supposes the concentration on one’s own linguistic resources and their experimentation in the field of language. The developed lyrical constructions, frequently used in the work of these poets, they bring language games, quotations, allusions, parodies, rewritings, etc. Although, both poets don’t ignore the ideational plan, which an attentive reader can perceive, the finality of their writing consists, however, in the creating of the discourse, which It detaches itself from any other reality and It exists autonomously, being self-sufficient. Thus, such a poem returns the myth of Narcissus, stating the concept of self-referential and self-contemplative writing.

Abstract Most of the currently-enrolled undergraduate engineering students grew up with exposure to social media websites like Facebook and Youtube. Making sure that students are not distracted by their mobile devices in class has become more challenging, and one way to address the issue is to present engineering in a more entertaining and engaging way. A rap song about fluid mechanics was created by the author for entrainment, outreach, and education purposes. The song covers the fundamentals of fluid mechanics and mentions some theoretical basics, as well as some of the most widely used computational fluid dynamics and experimental fluid dynamics techniques. The song was written with the intention to be entertaining and educational — the goal was that someone with no prior fluid mechanics background will be able to understand it after spending 10–20 minutes reading through the lyrics explanations. A music video was produced for the song. The video production was sponsored by the American Society of Mechanical Engineers and includes visuals of experimental facilities and equipment. The paper provides the background of the project, marketing plans, some of the lessons learned, the lyrics, and the explanations of the lyrics.

Natural Language Processing is in growing demand with recent developments. This Generator model is one such example of a music generation system conditioned on lyrics. The model proposed has been tested on songs having lyrics written only in English, but the idea can be generalized to various languages. This paper’s objective is to mainly explain how one can create a music generator using statistical machine learning methods. This paper also explains how effectively outputs can be formulated, which are the music signals as they are million sized over a short period frame. The parameters mentioned in the paper only serve an explanatory purpose. This paper discusses the effective statistical formulation of output thereby decreasing the vast amount of estimation of output parameters, and how to reconstruct the audio signals from predicted parameters by using ‘phase-shift algorithm’.

Enhancing the activity of biocontrol agents could be the most important factor in their success in controlling fruit disease and their ultimate acceptance in commercial disease management. Direct manipulation of a biocontrol agent resulting in enhancement of diseases control could be achieved by using recent advances in molecular biology techniques. The objectives of this project were to isolate genes from yeast species that were used as postharvest biocontrol agents against postharvest diseases and to determine their role in biocontrol efficacy. The emphasis was to be placed on the yeast, Candida oleophila, which was jointly discovered and developed in our laboratories, and commercialized as the product, Aspire. The general plan was to develop a transformation system for C . oleophila and either knockout or overexpress particular genes of interest. Additionally, biochemical characterization of the lytic peptides was conducted in the wild-type and transgenic isolates. In addition to developing a better understanding of the mode of action of the yeast biocontrol agents, it was also our intent to demonstrate the feasibility of enhancing biocontrol activity via genetic enhancement of yeast with genes known to code for proteins with antimicrobial activity. Major achievements are: 1) Characterization of extracellular lytic enzymes produced by the yeast biocontrol agent Candida oleophila; 2) Development of a transformation system for Candida oleophila; 3) Cloning and analysis of C.oleophila glucanase gene; 4) Overexpression of and knockout of C. oleophila glucanase gene and evaluating its role in the biocontrol activity of C. oleophila; 5) Characterization of defensin gene and its expression in the yeast Pichiapastoris; 6) Cloning and Analysis of Chitinase and Adhesin Genes; 7) Characterization of the rnase secreted by C . oleophila and its inhibitory activity against P. digitatum. This project has resulted in information that enhanced our understanding of the mode of action of the yeast C . oleophila. This was important step towards enhancing the biocontrol activity of the yeast. Fungal cell wall enzymes produced by the yeast antagonist were characterized. Different substrates were identified to enhance there production in vitro. Exo-b-1, 3 glucanase, chitinase and protease production was stimulated by the presence of cell-wall fragments of Penicillium digitatum in the growing medium, in addition to glucose. A transformation system developed was used to study the role of lytic enzymes in the biocontrol activity of the yeast antagonist and was essential for genetic manipulation of C . oleqphila. After cloning and characterization of the exo-glucanase gene from the yeast, the transformation system was efficiently used to study the role of the enzyme in the biocontrol activity by over-expressing or knocking out the activity of the enzyme. At the last phase of the research (still ongoing) the transformation system is being used to study the role of chitinase gene in the mode of action. Knockout and over expression experiments are underway.

There is a need for renewable antimicrobial surface treatments that are semi- permanent, can eradicate both biofilms and planktonic pathogens over long periods of time and that do not select for resistant strains. This proposal describes a dopamine binding technology that is inexpensive, bio-friendly, non-toxic, and uses straight-forward commercially available products. The antimicrobial agents are peptidoglycanhydrolase enzymes that are non-toxic and highly refractory to resistance development. The goal of this project is to create a treatment that will be applicable to a wide variety of surfaces and will convey long-lasting antimicrobial activity. Although the immediate goal is to create staphylolytic surfaces, the technology should be applicable to any pathogen and will thus contribute to no less than 3 BARD priorities: 1) increased animal production by protecting animals from invasive and emerging diseases, 2) Antimicrobial food packaging will improve food safety and security and 3) sustainable bio- energy systems will be supported by coating fermentation vats with antimicrobials that could protect ethanolic fermentations from Lactobacillus contamination that reduces ethanol yields. The dopamine-based modification of surfaces is inspired by the strong adhesion of mussel adhesion proteins to virtually all types of surfaces, including metals, polymers, and inorganic materials. Peptidoglycanhydrolases (PGHs) meet the criteria of a surface bound antimicrobial with their site of action being extracellular peptidoglycan (the structural basis of the bacterial cell wall) that when breached causes osmotic lysis. As a proof of principle, we will develop technology using peptidoglycanhydrolase enzymes that target Staphylococcus aureus, a notoriously contagious and antimicrobial-resistant pathogen. We will test for susceptibility of the coating to a variety of environmental stresses including UV light, abrasive cleaning and dessication. In order to avoid resistance development, we intend to use three unique, synergistic, simultaneous staphylococcal enzyme activities. The hydrolases are modular such that we have created fusion proteins with three lytic activities that are highly refractory to resistance development. It is essential to use multiple simultaneous activities to avoid selecting for antimicrobial resistant strains. This strategy is applicable to both Gram positive and negative pathogens. We anticipate that upon completion of this award the technology will be available for commercialization within the time required to achieve a suitable high volume production scheme for the required enzymes (~1-2 years). We expect the modified surface will remain antimicrobial for several days, and when necessary, the protocol for renewal of the surface will be easily applied in a diverse array of environments, from food processing plants to barnyards.

Steroidal glycoalkaloids (SGAs) are secondary metabolites being part of the plant defense response. The two major SGAs in cultivated potato (Solanum tuberosum) are α-chaconine and α-solanine, which exhibit strong cellular lytic properties and inhibit acetylcholinesterase activity, and are poisonous at high concentrations for humans. As SGAs are not destroyed during cooking and frying commercial cultivars have been bred to contain low levels, and their content in tubers should not exceed 20 mg/100 g fresh weight. However, environmental factors can increase tuber SGA content above the safe level. The focus of the proposed research was to apply genomic approaches to identify candidate genes that control potato SGA content in order to develop tools for potato improvement by marker-assisted selection and/or transgenic approaches. To this end, the objectives of the proposal included identification of genes, metabolic intermediates and allelic variations in the potato SGAbiosynthetic pathway. The SGAs are biosynthesized by the sterol branch of the mevalonic acid/isoprenoid pathway. Transgenic potato plants that overexpress 3-hydroxy-3-methylglutaryl-CoA reductase 1 (HMG1) or squalene synthase 1 (SQS1), key enzymes of the mevalonic acid/isoprenoid pathway, exhibited elevated levels of solanine and chaconine as well as induced expression of genes downstream the pathway. These results suggest of coordinated regulation of isoprenoid (primary) metabolism and SGA secondary metabolism. The transgenic plants were further used to identify new SGA-related candidate genes by cDNA-AFLP approach and a novel glycosyltransferase was isolated. In addition, genes involved in phytosterol biosynthesis may have dual role and synthesize defense-related steroidal metabolites, such as SGAs, via lanosterol pathway. Potato lanosterol synthase sequence (LAS) was isolated and used to prepare transgenic plants with overexpressing and silencing constructs. Plants are currently being analyzed for SGA content. The dynamics of SGA accumulation in the various organs of a potato species with high SGA content gave insights into the general regulation of SGA abundance. Leaf SGA levels in S. chacoense were 10 to 20-fold greater than those of S. tuberosum. The leptines, SGAs with strong antifeedant properties against Colorado potato beetles, were present in all aerial tissues except for early and mid-developmental stages of above ground stolons, and accounted for the high SGA content of S. chacoense. These results indicate the presence of regulatory mechanisms in most tissues except in stolons that limit the levels of α-solanine and α-chaconine and confine leptine accumulation to the aerial tissues. The genomes of cultivated and wild potato contain a 4-member gene family coding for SQS. Three orthologs were cloned as cDNAs from S. chacoense and heterologously expressed in E. coli. Squalene accumulated in all E. coli lines transformed with each of the three gene constructs. Differential transcript abundance in various organs and amino acid sequence differences in the conserved domains of three isoenzymes indicate subfunctionalization of SQS activity and triterpene/sterol metabolism. Because S. chacoense and S. phureja differ so greatly for presence and accumulation of SGAs, we selected four candidate genes from different points along the biosynthetic pathway to determine if chcor phuspecific alleles were associated with SGA expression in a segregating interspecific diploid population. For two of the four genes (HMG2 and SGT2) F2 plants with chcalleles expressed significantly greater total SGAs compared with heterozygotes and those with phualleles. Although there are other determinants of SGA biosynthesis and composition in potato, the ability of allelic states at two genes to affect SGA levels confirms some of the above transgenic work where chcalleles at two other loci altered SGA expression in Desiree. Present results reveal new opportunities to manipulate triterpene/sterol biosynthesis in more targeted ways with the objective of altering SGA content for both human health concerns and natural pesticide content without disrupting the essential metabolism and function of the phytosterol component of the membranes and the growth regulating brassinosteroids.

Original objectives . 1. Test anti-fungal gene products for activity against Uncinula necator, Aspergillus niger, Rhizopus stolonifer and Botrytis cinerea. 2. For Agrobacterium transformation, design appropriate vectors with gene combinations. 3. Use biolistic bombardment and Agrobacterium for transformation of important cultivars. 4. Characterize gene expression in transformants, as well as level of powdery mildew and Botrytis resistance in foliage of transformed plants. Background The production of new grape cultivars by conventional breeding is a complex and time-consuming process. Transferring individual traits via single genes into elite cultivars was proposed as a viable strategy, especially for vegetatively propagated crops such as grapevines. The availability of effective genetic transformation procedures, the existence of genes able to reduce pathogen stress, and improved in vitro culture methods for grapes, were combined to serve the objective of this proposal. Effective deployment of resistance genes would reduce production costs and increase crop quality, and several such genes and combinations were used in this project. Progress The efficacy of two-way combinations of Trichoderma endochitinase (CHIT42), synthetic peptide ESF12 and resveratrol upon the control of growth of Botrytis cinerea and Penicillium digitatum were evaluated in vitro. All pairwise interactions were additive but not synergistic. Per objective 2, suitable vectors with important gene combinations for Agrobacterium transformation were designed. In addition, multiple gene co-transformation by particle bombardment was also tested successfully. In New York, transformation work focused on cultivars Chardonnay and Merlot, while the technology in Israel was extended to 41B, R. 110, Prime, Italia, Gamay, Chardonnay and Velika. Transgenic plant production is summarized in the appendix. Among plants developed in Israel, endochitinase expression was assayed via the MuchT assay using material just 1-5 days after co-cultivation. Plants of cv. Sugraone carrying the gene coding for ESF12, a short anti-fungal lytic peptide under the control of the double 358 promoter, were produced. Leaf extracts of two plants showed inhibition zones that developed within 48 h indicating the inhibitory effect of the leaf extracts on the six species of bacteria. X fastidiosa, the causal organism of Pierce's disease, was very sensitive to leaf extracts from ESF12 transformed plants. Further work is needed to verify the agricultural utility of ESF12 transformants. In New York, some transformants were resistant to powdery mildew and Botrytis fruit rot. Major conclusions, solutions, achievements and implications The following scientific achievements resulted from this cooperative BARD project: 1. Development and improvement of embryogenesis and tissue culture manipulation in grape, while extending these procedures to several agriculturally important cultivars both in Israel and USA. 2. Development and improvement of novel transformation procedures while developing transformation techniques for grape and other recalcitrant species. 3. Production of transgenic grapevines, characterization of transformed vines while studying the expression patterns of a marker gene under the control of different promoter as the 35S CaMV in different part of the plants including flowers and fruits. 4. Expression of anti-fungal genes in grape: establishment of transgenic plants and evaluation of gene expression. Development of techniques to insert multiple genes. 5. Isolation of novel grape specific promoter to control the expression of future antimicrobial genes. It is of great importance to report that significant progress was made in not only the development of transgenic grapevines, but also in the evaluation of their potential for increased resistance to disease as compared with the non engineered cultivar. In several cases, increased disease resistance was observed. More research and development is still needed before a product can be commercialized, yet our project lays a framework for further investigations.
