Texto completo”Kommunikation med synligt ljus” (VLC) som ett design material är ett komplement till ”radio frekvens kommunikation” och har många applikationer inom trådlös kommunikation, Inomhuspositionering, och situationer där RFC inte är applicerbart. Problematiken med teknologi är att den ofta inte är synlig, och därför är svår att inkludera i dagens design processer. För att designers ska kunna använda sig av VLC i deras kreativa process, utgår denna studie från ”Inspirational Bits” förhållningsätt till design, och ämnar utforska följande frågeställning: Hur kan vi designa artefakter som tillåter designers att engagera sig i en konversation med ”kommunikation med synligt ljus” (VLC) som ett design material? Genom ”konstruktiv designforskning”, studerades prototyper av teknologin följt av analys. Processen upprepades med ett flertal modeller. Prototyperna utvärderades i två design-workshops, genom observation av och respons från 11 deltagare. Som resultat, byggdes sju stycken artefakter för att belysa sju stycken potentiella materiella egenskaper av VLC. Observationerna och den samlade responsen visade också att artefakterna hjälpte designers att få taktil kunskap om VLC. Artefakterna baserades på ”the Arduino Platform” och elektriska standard komponenter. En biprodukt av denna studie är ”the lumoino toolkit for tinkering and prototyping with VLC”. Eventuella styrkor och svagheter diskuteras och stycket framtida arbete visar potentiella utvecklingar och expantioner av ”the lumoino toolkit”.

Texto completoA lipofuscina é um pigmento autofluorescente acumulado progressivamente durante o envelhecimento celular em diversos tecidos, como o músculo cardíaco e retina, principalmente no período pós-mitótico. Esse fenômeno pode ocorrer em decorrência do estresse oxidativo, quando biomoléculas e organelas (principalmente mitocôndrias) sofrem danos, gerando produtos não degradáveis no interior dos lisossomos. A lipofuscina pode ser fotossensibilizada promovendo processos fotoxidativos nos componentes celulares. Muitos estudos de lipofuscina foram feitos em células do epitélio pigmentar da retina de olho humano, mas conhece-se muito pouco sobre a lipofuscina de pele humana. Neste trabalho nós investigamos a formação fotoinduzida (UVA e luz visível) de lipofuscina e as consequências da sua fotossensibilização pela luz visível. Nós também estabelecemos protocolos eficazes na indução de lipofuscinogênese, por meio de dano específico em mitocôndrias e lisossomos. Células que acumularam lipofuscina, após exposição à UVA ou luz azul, tornaram-se sensíveis à luz visível (400-750 nm). Caracterizamos as propriedades de absorção e de emissão da lipofuscina e seu tempo de vida de fluorescência, utilizando a microscopia de fluorescência resolvida no tempo (FLIM). Observamos que lipofuscina em queratinócitos tem máximo de absorção na região do azul (420-450 nm), com emissão máxima de fluorescência no vermelho. As células HaCaT carregadas com lipofuscina efotossensibilizadas no visível, tiveram redução da viabilidade celular, que foi relacionada com a geração de oxigênio singlete, bem como acumularam lesões pré-mutagênicas 8-oxo-dG e quebras na fita de DNA. Também, investigamos a eficiência de diferentes comprimentos de onda da luz visível (408, 466, 522 e 650 nm) em promover a formação de lipofuscina em consequência de lesões em mitocôndrias e lisossomos. Tanto a luz azul (408 e 466 nm) quanto a luz verde (522 nm), mas não vermelha (650 nm) promoveram dano em mitocôndrias (integridade de membrana e DNA) e lisossomos (integridade de membrana e atividade autofágica), induzindo eficientemente lipofuscinogênese. Logo, além de UVA, o próprio espectro do visível aumenta a sensibilidade de queratinócitos à luz visível, através da geração de lipofuscina. Por fim, testamos o potencial carcinogênico da luz azul de alta energia (408 nm), irradiando células HaCaT cronicamente. Identificamos quatro mudanças principais envolvidas com o processo de transformação maligna: instabilidade genômica, redução da expressão de proteína supressora de tumor p16INK4a, aumento da taxa de proliferação, e resistência à apoptose. Além disso, a formação de dímeros de pirimidina ciclobutano (CPD) no DNA nuclear de células HaCaT logo após ou depois de vários ciclos de irradiação com 408 nm foi observada pela primeira vez na literatura.

Texto completoENGLISH ABSTRACT: South Africa is experiencing a number of challenges, which have led to developmental backlogs, housing shortages and growing informal areas. At the same time, the country is also experiencing sustainability problems, such as climate change, biodiversity loss and resource shortages. Strategies to address these challenges do not always provide adequate space for participatory structures as suggested by Local Agenda 21. Social sustainability (the ability of communities to collaborate in order to promote sustainability) is a fundamental component of sustainable development. An essential ingredient of social sustainability is to maintain and develop the stock of social capital (social bonds, relationships of trust, and connectedness in groups and networks). The existence of conflict and distrust in communities therefore makes cooperation for development and sustainability very difficult. The purpose of this study was to examine and evaluate a specific case study, namely the Witsand Integrated Energy Environment Empowerment Cost Optimisation (iEEECO) Human Settlement Project, situated near Atlantis within the City of Cape Town, with regard to lessons about the process towards sustainable human settlements and sustainability. This case study was chosen because it is a so-called People’s Housing Project that has also been developed as an ecological sustainable project. The study specifically aimed to establish whether and how participation was implemented as part of this project and how effective this had been in promoting social sustainability, since it had also been a conflict-ridden project. The case study methodology was used where interviews, direct observation and focus groups sessions were conducted. Finally, the study reports on the findings and formulates recommendations based on the case study on some of the ways to improve sustainability. The process of learning about this community was like unpeeling an onion, that started with the impression of a successful sustainable human settlement, but ended up with the knowledge that it is a project, which despite more than usual efforts made to implement sustainability principles and participation, remains conflict ridden and fragmented. The community is presently represented by two community organisations (of which one does not support sustainability principles). It also seems as if few community members felt that they had really been involved in the participation process, with many relegated to make up numbers in meetings, with no real involvement in the process. The level of illiteracy within the community is high and rumours abound, which at one stage led to houses of community leaders, as well as field workers employed by the municipality to monitor extension of shacks, being stoned and burnt. Municipal policies to use local people as gate-keepers to keep informal areas from growing are contributing to these problems. Changes in housing policies regarding contributory payments are also adding to conflict within the community, which adds fuel to rumours of corruption. In the context of a shortage of space where people can legally build their own shacks, a shortage of public facilities like schools and clinics, as well as employment opportunities, this project cannot yet be regarded as an integrated sustainable human settlement.The lessons that this case study teaches us is about the path towards sustainability, is that it is a complex process, which requires a bigger focus on social sustainability and on conflict transformation. Social sustainability requires more and better participation and transparency in policy-making, as well as changes in policies to promote more fairness, justice, and the building of social capital. Co-production, where state and citizens work together to provide basic services, has much to offer as a method of participation, especially if initiated and run by grassroots organisations.
AFRIKAANSE OPSOMMING: Suid-Afrika beleef 'n aantal uitdagings, wat gelei het tot ontwikkelingsagterstande, behuisingstekorte en groeiende informele gebiede. Terselfdertyd ondervind die land ook volhoubaarheidsprobleme, soos klimaatverandering, die verlies aan biodiversiteit en hulpbron-tekorte. Strategieë om hierdie uitdagings aan te spreek, laat nie altyd voldoende ruimte vir deelnemende strukture soos voorgestel deur Plaaslike Agenda 21 nie. A review of project portfolio management (PPM) literature has shown that human resources allocation is rarely revisited beyond the initial planning cycle, and that it is often treated as a static problem. Therefore, this thesis sought to understand modern PPM practices further and to underscore variables that correlate with proficient portfolio planning, management, and execution. A survey of current practices has yielded several unexpected results. For example, the extent of employee involvement in resource allocation decisions, via active participation in the PMO, is positively correlated with highly effective PPM practices. Organizations experience schedule delays on the order of 10-20%, even though they classify their PPM practices as highly effective. Furthermore, 54% of survey participants indicated their firms do not evaluate nor model resource uncertainties, risks or interdependencies, of which 85% conceded these variables should be addressed. Given the survey results and given that PPM methods were borne of Markowitz's Modern Portfolio Theory, this thesis sought to frame the human resource allocation problem as a sociotechnical system instead. As such, nine critical system design decisions were identified and combined to yield distinct process architectures. Next, these architectures were scored and evaluated against performance metrics levied by the system stakeholders. An architectural tradespace of 11,664 feasible human resource allocation systems was generated; of which 42 architectures are nondominated. The systematic analysis in this thesis revealed that 100% of the architectures on the Pareto Front are analogous to a transparent, market-like resource allocation system as opposed to an anonymous, centralized system. Furthermore, 83% of these architectures appointed the employee as the sole decision-maker of its allocation to tasks. Roughly 70% of these architectures required agents to frequently updated task start and end times, hence reducing uncertainty and risk in planning. Future work shall re-assess the architecture scores and stakeholder requirements prior to application on a pilot portfolio.

Texto completoThe global marketplace is rapidly intensifying. Longer product sales lives, greater profit margins or simply survival, is dependent on management¿s ability to create and lead change. Project Management has become an important competency, combined with other business practices to adapt to the trend of changing conditions. Critical Chain is a relatively new project methodology, elaborated by Eliyahu Goldratt in order to complete projects faster, make more efficient use of resources and securing the project deliverables. The methodology is based on the assumption that traditional project techniques such as CPM and PERT, do not recognize critical human behavior. The methodology claims that many project failures are a direct result of how safety is built into the task delivery times, and then wasted by human behavior such as Student Syndrome, Parkinson Law and Multitasking. However, there has been little or no previous research regarding this topic in the Argentine marketplace. This study intended to investigate to what extent the human behavior concepts of critical chain project management are present, by performing in-depth interviews with Argentine project stakeholders. It appears that the four human behavior concepts are present in Argentina and that the majority of Argentine companies are yet to apply project management techniques.
