Abstract Sirolimus (SIR), an inhibitor of mammalian target of rapamycin (mTOR), is an immunosuppressive agent that is synergistic with CNI to prevent or treat GVHD in patients (pts) undergoing allo-HST (Cutler, Biol Blood Marrow Transplant. 2004;10:328). Transplant associated microangiopathy (TAM) is a multi-factorial complication of allo- HST, associated with CNI. Recently, the addition of SIR to CNIs was reported to result in a higher than expected (10.8%) incidence of TAM (Cutler et al. Biol Blood Marrow Transplant. 2005; 11:551). We analyzed the incidence and clinical characteristics of TAM in 2 groups of pediatric patients (pts): the prevention group (GP): pts who received SIR and tacrolimus (TAC) for prevention; and the treatment group (TG): pts treated with SIR and a CNI (TAC or cyclosporine) for exacerbation of GVHD. Methods: A retrospective chart review of pts with allo-HST who received SIR/TAC for either prevention or treatment of GVHD. Demographic data, clinical course, occurrence of GVHD and TAC and SIR levels were recorded. TAM was defined as serum creatinine (SCr) of ≥ 50% above baseline, elevated LDH levels (>X2 upper limit of normal), presence of schistocytes or persistence of nucleated red blood cells, and prolonged or progressive thrombocytopenia. For PG the risk period was defined between days −1 to +60 of HSC. The risk period for the TG was defined from the first day of SIR/TAC until day +30 after the last dose was given. Results: Records of 45 pts who were treated with SIR/TAC were reviewed. The median age of 21 patients in PG was 9.1 yr (2.9–22); male gender, n=13. The median age in 24 TG pts was14.7 yr (3.3–20.6); male gender, n= 11. Conditioning regimens consisted of TBI based regimens (64%), or chemotherapy only (36%). Diagnoses included ALL (40%), AML (27 %), NHL (9%), Fanconi’s (4 %), and other (9%). The median follow up for the surviving patients is 28.6 months. TAM criteria were met in 15 ( 30%) patients who received SIR/TAC, 5 PG pts (23.8%) and 10 TG pts ( 41.6%). Two PG patients died from multi-organ failure (MOF) related to other reasons (VOD, n=1 and multi-organism infection, n=1). Both pts had hemolytic uremic syndrome (HUS), possibly contributing to MOF. One patient developed seizures secondary to thrombotic thrombocytopenic purpura (TTP) as a complication of TAM. TAC and SIR levels were within the desired range (WDR, (≤10 ng/ml, for each) in this patient and signs and symptoms of TAM and TTP resolved after discontinuation of both TAC and SIR. In 2 patients TAM findings were asymptomatic, without progression to HUS or TTP. SIR/TAC levels in these patients were all WDR. Among 10 TG pts that met TAM criteria, 2 pts progressed to HUS and one developed severe TTP. All 3 patients with complicated TAM died (HUS, n=1, MOF, n=1, and idiopathic pneumonitis, n=1). Two of the pts with laboratory, but not clinically apparent TAM died (invasive fungal infection, n=1, IP n=1). In all TG pts with TAM either TAC (n=7) or SIR (n=6) levels exceeded WDR. To identify possible risk factors for TAM, the following parameters were analyzed: age, conditioning regimen, disease type, degree of HLA match. In the PG, only organ failure or high-grade GVHD could be identified as risk factors. In the TG, TAC or SIR levels exceeding WDR, in addition to ac GVHD or chronic GVHD present in all patients, were associated with an increased risk for clinical significant TAM. Conclusion: Organ damage, either by acute GVHD or from other reasons may contribute to development of clinically significant TAM in pts undergoing allo-HST treated with SIR plus CNI. Monitoring TAC or SIR levels and early diagnosis of TAM may prove to be critical in patients with progressive organ damage of GVHD.

Synthetic aperture radar (SAR) ship detection and classification has gained unprecedented attention due to its important role in maritime transportation. Many deep learning-based detectors and classifiers have been successfully applied and achieved great progress. However, ships in SAR images present discrete and multi-centric features, and their scattering characteristics and edge information are sensitive to variations in target attitude angles (TAAs). These factors pose challenges for existing methods to obtain satisfactory results. To address these challenges, a novel target attitude angle-guided network (TAG-Net) is proposed in this article. The core idea of TAG-Net is to leverage TAA information as guidance and use an adaptive feature-level fusion strategy to dynamically learn more representative features that can handle the target imaging diversity caused by TAA. This is achieved through a TAA-aware feature modulation (TAFM) module. It uses the TAA information and foreground information as prior knowledge and establishes the relationship between the ship scattering characteristics and TAA information. This enables a reduction in the intra-class variability and highlights ship targets. Additionally, considering the different requirements of the detection and classification tasks for the scattering information, we propose a layer-wise attention-based task decoupling detection head (LATD). Unlike general deep learning methods that use shared features for both detection and classification tasks, LATD extracts multi-level features and uses layer attention to achieve feature decoupling and select the most suitable features for each task. Finally, we introduce a novel salient-enhanced feature balance module (SFB) to provide richer semantic information and capture the global context to highlight ships in complex scenes, effectively reducing the impact of background noise. A large-scale ship detection dataset (LSSDD+) is used to verify the effectiveness of TAG-Net, and our method achieves state-of-the-art performance.

Dengan memfokuskan kajian studi pengembangan model desain pembelajaran PAI multi orientasi. Adapun hasil penelitian perasaan dan pandangan siswa terhadap proses belajar menggunakan MDP-PAI Berbasis TAM sangat positif. Siswa merasakan dan memandang bahwa proses belajar mengajar menggunakan MDP-PAI Berbasis TAM sangat menyenangkan dan bermanfaat dalam meningkatkan kemampuan tulis baca huruf al-Quran. Penggunaan MDP PAI Berbasis TAM dapat menumbuhkan motivasi dan keaktifan belajar siswa yang cukup tinggi sehingga proses belajar mengajar berlangsung kondusif dalam mencapai tujuan yang telah ditetapkan.This study discusses the instructional design of Islamic Religious Education (PAI) based Malay Arabic script (TAM). By focusing the study findings of the development model of PAI instructional design multi orientation. The result of this study is the feelings and views of the students to the learning process using the MDP-PAI-based TAM are very positive. Students feel and perceive that the learning process by using MDP-PAI-based TAM is very pleasant and helpful in improving their ability to write and read the al-Quran letters. The use of MDP PAI-based TAM can motivate and activate the student's learning so teaching and learning process can be conducive to achieving the stated goals.

e15038 Introduction: The pharmacogenetics of irinotecan indicate that a common polymorphism in the uridine diphosphate glucuronosyltransferase 1As (UGT1As) gene predict severe toxicity. However the tumor response to irinotecan is variable and unpredictable. Methods: Two multi-center phase II studies of FOLFIRI (FLIGHT-1 and FLIGHT-2 study) were conducted for patients with metastatic colorectal cancer (CRC) in Japan. FLIGHT-1 study was first-line chemotherapy, and FLIGHT-2 study was FOLFOX-refractory second-line chemotherapy. 103 patients have been enrolled in these studies (53 patients in FLIGHT-1 and 50 patients in FLIGHT-2) from 20 institutions by April 2007 from November 2005. Seventy one patients were analyzed UGT1As polymorphisms, UGT1A1*28 (TA6>TA7), UGT1A1*6 (G>A), UGT1A1*60 (T>C), UGT1A7 (N129K; T>G), UGT1A7 (-57 T>G), UGT1A9*22 (T10>T9). Results: Out of 71 patients, 34 had G3/4 neutropenia or leukopenia, and 23 had tumor responses (CR +PR). G3/4 neutropenia was more frequent in patients with *6, N129K(G), -57(G), *22 allele than patients without these allele (p<0.05). Other polymorphism was not the predictive factor for toxicity and tumor response, independently. On the other hand, genotype subset selection of multi-UGT1As polymorphisms was useful to predict severe toxicities and tumor responses. Only 7 patients of 22 patients with TA6/TA6 and T10/T10, or TA6/TA7 and -57(T/T), or *6(G/G) and *60(T/T) had grade3/4 toxicity. Similarly, high risk group of toxicity or high and low tumor responses groups was also predicted by genotype subset selections. Conclusions: Genotype subset selections of multi-UGT1As polymorphisms were the excellent predictor for severe toxicities and tumor responses of metastatic CRC patients received FOLFIRI regimen. In this study, a new technology acceptance model (TAM) based on the characteristics that are adapted from Innovation Diffusion Theory (IDT) with integrated multi-criteria decision-making (MCDM) method is developed in order to explain students’ acceptance of information system. New TAM is developed based on 650 students of a public university of Turkey. In the study, first, five innovation characteristics are prioritized with the Analytical Hierarchy Method (AHP) and independent variables of TAM are formed with the Technique for Order Preference by Similarity to Ideal Solution (TOPSIS) technique with using AHP results as the criteria weights. With the obtained results, explaining the intention to use by more than 50% shows that integrated TAM is an applicable and flexible model.

This study aims to develop and implement a new Technology Acceptance Model (TAM) to verify and explain users&rsquo; acceptance of SAP software. The structural equation model (SEM) technique is used with the AMOS program for the adoption process. General structural model, which includes Experience, Voluntariness, Perceived Enjoyment, Perceptions of External Control, Computer Self Efficacy, Output Quality and Job Relevance, is developed with Analytical Hierarchy Method (AHP) and Pareto Analysis by deciding variables from previously original developed models which are called TAM1, TAM2 and TAM3. New TAM is tested to a sample of 384 SAP users in a company that serves in metal production. Results illustrates that new TAM is an appropriate tool to explain and understand users&rsquo; acceptance of SAP besides of being flexible and easily implemented. SAP users&rsquo; job relevance and their perceptions of external control are the most important variables that affect perceived usefulness and perceived ease of use in the model.

Tubular aggregate myopathy (TAM) and Stormorken syndrome (STRMK) form a clinical continuum associating progressive muscle weakness with additional multi-systemic anomalies of the bones, skin, spleen, and platelets. TAM/STRMK arises from excessive extracellular Ca2+ entry due to gain-of-function mutations in the Ca2+ sensor STIM1 or the Ca2+ channel ORAI1. Currently, no treatment is available. Here we assessed the therapeutic potential of ORAI1 downregulation to anticipate and reverse disease development in a faithful mouse model carrying the most common TAM/STRMK mutation and recapitulating the main signs of the human disorder. To this aim, we crossed Stim1R304W/+ mice with Orai1+/− mice expressing 50% of ORAI1. Systematic phenotyping of the offspring revealed that the Stim1R304W/+Orai1+/− mice were born with a normalized ratio and showed improved postnatal growth, bone architecture, and partly ameliorated muscle function and structure compared with their Stim1R304W/+ littermates. We also produced AAV particles containing Orai1-specific shRNAs, and intramuscular injections of Stim1R304W/+ mice improved the skeletal muscle contraction and relaxation properties, while muscle histology remained unchanged. Altogether, we provide the proof-of-concept that Orai1 silencing partially prevents the development of the multi-systemic TAM/STRMK phenotype in mice, and we also established an approach to target Orai1 expression in postnatal tissues.

Abstract The major diseases responsible for the decline of profitable pepper (Capsicum annuum L.) production in Texas and other areas throughout the United States and the world have been the potato Y-type viruses (1). ‘TAM Mild Chile-2’ (TMC-2) is the first mildly pungent long green/red chile with multiple virus resistance (MVR) to tobacco etch virus (TEV), potato virus Y (PVY), pepper mottle virus (PeMV), and tobacco mosaic virus (TMV) developed by the Texas Agricultural Experiment Station (TAES). It is recommended as a multi-purpose chile for fresh market, for processing in the green stage, or dehydration into red chile powder. The fruit exhibit a very low pungency and a high concentration of extractable red color.

Abstract Down syndrome (DS) is characterized by the trisomy of chromosome 21 and complicated with multi-organ dysfunctions including the hematopoietic system. Among them, myeloproliferative disorder is known as a particular feature of the abnormality in hematopoiesis. At birth, about 10% of DS newborns show an extreme increase in blast cell number of peripheral blood and bone marrow, which is called transient abnormal myelopoiesis (TAM), because the blasts spontaneously disappear within 3 months. Morphologically, blast cells in TAM are similar to those typically found in acute megakaryoblastic leukemia (AMKL). Genetic analysis of blasts in TAM usually shows mutation in GATA1 gene. After spontaneous remission of TAM, 20 to 30% of TAM patients develop AMKL within several years. This type of AMKL is especially called DS related-AMKL (DS-AMKL). This leukemogenic transition from TAM to AMKL is considered to be a typical model of multi-step tumorigenesis. In this model, we deal with the initial part of TAM development in relationship with trisomy 21 and GATA1 mutation, especially focusing on how GATA1 mutation promotes TAM development and why hematopoietic progenitors with GATA1 mutation prevail during embryonic hematopoiesis only in the cells with trisomy 21. In order to address these unsolved issues, we have established a strictly controlled human induced pluripotent stem cell (iPSC) lines derived from DS patients with or without TAM. In this study, to recapitulate the phenotype of TAM and to specify the differentiation stage affected in hematopoietic cells in TAM patients, we differentiated established isogenic iPSC lines into megakaryocytes and erythrocytes in a step-wise manner. For this purpose, we employed two-dimensional differentiation system and compared the frequency of hematopoietic progenitor cells at various stages. For megakaryocytic lineage, we traced their differentiation as follows; hematopoietic progenitor cells committed to megakaryocytic lineage (day 9, CD34+CD41a+CD42b-CD235a-), megakaryoblasts (day 16, CD34-CD41a+CD42b-CD235a-) and promegakaryocytes (day 16, CD34-CD41a+CD42b+CD235a-). For erythrocytic differentiation, CD71+CD235a+ cells were defined as erythroid-committed hematopoietic cells. On nine days after the initiation of hematopoietic differentiation (day 9), the frequency of CD41a+CD235a- cells showed no significant differences irrespective of the status of chromosome 21 and GATA1 genotype. However, on the day 16, while the frequency of promegakaryocytes significantly decreased in GATA1-mutated iPSCs, megakaryoblasts, an earlier stage cells than promegakaryocytes, were increased in GATA1-mutated iPSCs. These data suggest that megakaryocytic maturation is arrested in GATA1-mutated iPSCs at the stage of megakaryoblasts. In GATA1-non-mutated clones, iPSCs with trisomy 21 yielded erythroid-committed CD71+CD235a+ cells more frequently than those with disomy 21. However, in GATA1-mutated clones, either trisomy 21 or disomy 21 iPSC clones never yielded the erythroid-committed cells. Taken these results together, we suspected that these in vitro phenotypes observed in both erythroid and megakaryocytic lineages were caused by the impairment of fate decision in their progenitor cells. In conclusion, we successfully recapitulated the phenotypes of TAM in vitro in regard to the abnormal differentiation into megakaryocytic and erythroid lineages. We noticed that the in vitro phenotype were associated with the GATA1 genotype and the ploidy of chromosome 21. Considering these results, analyses of the megakaryocytic and erythroid progenitor cells, such as CMP, MEP and Mk-p, are important to determine which stage of progenitors is responsible for the impairment of hematopoietic cell maturation and subsequent TAM development. Moreover, we believe that the recapitulated TAM model using iPSCs is helpful for the comprehensive understanding of pathogensis in TAM. Disclosures No relevant conflicts of interest to declare.

The article analyzes a construction technology project as a safety standard document for a construction site and describes the application of mathematical methods at the stages of construction and design. Due to the fact that the construction project consists of (safety point of view) the site plan and technological cards (notably these documents indicate safety design solutions), the indicators making up a comparable set of indicators to evaluate the site plan, technological cards and the entire project may vary and be diverse. Practice shows that the construction technology project or its parts thereof are to be assessed using 5 to 7 parameters, as in case of a larger quantity of indicators, the significance of each indicator becomes relatively lower and has effect on the priorities of line formation. To ensure the safety of workers on construction sites, when preparing construction technology projects, the application of experimental design is proposed, thus starting the evaluation of the quality of technological solutions to construction projects in terms of safety employing multicriteria mathematical methods. Santrauka Šiame straipsnyje analizuojamos statybos darbų technologijos projekto, kaip darbuotojų saugos norminio dokumento, rengimo sąlygos ir aplinkybės bei matematinių metodų taikymo galimybės rengiant saugos darbe sprendinius statybos paruošimo ir projektavimo etape. Kadangi statybos darbų technologijos projektas susideda (saugos darbe požiūriu) iš statybvietės plano ir technologinių kortelių (būtent šiuose dokumentuose užfiksuojami saugos darbe projektiniai sprendiniai), tai ir rodikliai, sudarantys lyginamų rodiklių aibę ir vertinantys statybvietes planą, technologines korteles ir visą projektą, gali būti labai įvairūs ir skirtingi. Praktika rodo, kad statybos darbų technologijos projektas ar jo dalys vertintini pagal 5–7 rodiklius, nes, esant didesniam rodiklių skaičiui, kiekvieno rodiklio reikšmingumas santykinai mažėja ir tai turi įtakos prioritetų eilutės formavimui. Siekiant užtikrinti darbuotojų saugą statybvietėse siūloma rengiant statybos darbų technologijos projektus taikyti variantinį projektavimą, pradėti vertinti statybos darbų technologinių projektų sprendinių kokybę saugos darbe požiūriu ir tam taikyti daugiatikslius matematinius metodus.

Bioactive molecules derived from seaweed extracts are revolutionary bio-stimulants used to enhance plant growth and increase yield production. This study evaluated the effectiveness of a commercially available seaweed liquid extract, namely, True-Algae-Max (TAM®), as a plant growth stimulant on nutritional, and antioxidant activity of Capsicum annuum. Three concentrations of TAM® (0.25, 0.5, and 1%) of various NPK: TAM® ratios were investigated via foliar spray, over 2017 and 2018 cultivation seasons, under greenhouse conditions. TAM® is rich in phytochemical compounds, such as ascorbic acid (1.66 mg g−1), phenolics (101.67 mg g−1), and flavonoids (2.60 mg g−1) that showed good antioxidant activity (54.52 mg g−1) and DPPH inhibition of 70.33%. Promoting measured parameter results stated the extensive potentiality of TAM® application, in comparison with conventional NPK treatment. Yield and composition of C. annuum were significantly improved in all TAM® treated groups, especially the TAM0.5% concentration, which resulted in maximum yield (4.23 Kg m−2) and significant amounts of profuse biological molecules like chlorophyll, ascorbic, phenolic compounds, flavonoids, and total nutrients. Compared to the NPK control treatments, C. annuum treated with TAM0.5% improved the total antioxidant activity of hot Pepper from 162.16 to 190.95 mg g−1. These findings indicate that the extract of seaweed can be used as an environmentally friendly, multi-functional biostimulant in the agricultural field for more sustainable production, in addition to reducing the use of hazardous synthetic fertilizers.

Evidence supports a role of 17&-estradiol (E2) in carcinogenesis and the large majority of breast carcinomas are dependent on estrogen. The anti-estrogen tamoxifen (TAM) is widely used for both treatment and prevention of breast cancer; however, it is also carcinogenic in human uterus and rat liver, highlighting the profound complexity of its actions. The nature of E2- or TAM-induced chromosomal damage has been explored using relatively high concentrations of these agents, and only some numerical aberrations and chromosomal breaks have been analyzed. This study aimed to determine the effects of low doses of E2and TAM (10&8 mol L&1and 10&6 mol L&1respectively) on karyotypes of MCF7, T47D, BT474, and SKBR3 breast cancer cells by comparing the results of conventional karyotyping and multi-FISH painting with cell proliferation. Estrogen receptor (ER)-positive (+) cells showed an increase in cell proliferation after E2treatment (MCF7, T47D, and BT474) and a decrease after TAM treatment (MCF7 and T47D), whereas in ER& cells (SKBR3), no alterations in cell proliferation were observed, except for a small increase at 96 h. Karyotypes of both ER+ and ER& breast cancer cells increased in complexity after treatments with E2and TAM leading to specific chromosomal abnormalities, some of which were consistent throughout the treatment duration. This genotoxic effect was higher in HER2+ cells. The ER&/HER2+ SKBR3 cells were found to be sensitive to TAM, exhibiting an increase in chromosomal aberrations. Thesein vitroresults provide insights into the potential role of low doses of E2and TAM in inducing chromosomal rearrangements in breast cancer cells.

536 Background: The optimal duration of endocrine therapy ranges between 5 and 7 years. Recent data suggest that a subset of patients could benefit from extended adjuvant TAM. The goal of this biomarker study was to identify potential candidates for extended TAM. Methods: After 2–3 yrs of adjuvant TAM, 3793 patients (pts) were randomized to continue TAM up to 10 yrs, or to stop treatment (Delozier, J Clin Oncol. 2000). The tumor blocks of 587/988 pts included at the Institut Gustave-Roussy, were used to construct a tissue micro-array (TMA). Immunohistochemical stainings were performed for ER, PR, bcl2, p53 (+: > 10% of stained cells), HER-2 (+: score 3+), EGFR1 (+: any positivity), PAK1, IGFR1 (+: intensity = 2 or 3). Median follow-up was 12 years. The prognostic values of biomarkers were assessed by a Cox model adjusted for clinical prognostic parameters and predictive values by interaction tests. Only statistical tests with P<0.01 were considered significant because of multi-hypothesis testing. Time-to-relapse (TTR), excluding contralateral cancer, was the main study endpoint. Results: Median age of the cohort included in the TMA was 63. Tumor characteristics : 54% node-positive, 85% ER+, 64% PR+, 7% HER-2+, 68% bcl2+, 14% p53+, 8% EGFR1+, 46% PAK1, 45% IGFR1. None of the biomarkers were prognostically significant. However, PR expression was strongly predictive for the efficacy of extended TAM on TTR (interaction test, p = 0.003). Long-term TAM was associated with a hazard ratio of 0.56, (95% CI: 0.30–1.03) and 1.94 (95%CI = 0.80–4.70) for PR+ and PR- pts respectively. A trend towards interaction between ER (p = 0.04), HER-2 (p = 0.05) and Bcl2 (p = 0.03) expression and a benefit from TAM was also observed, with a trend towards greater efficacy of prolonged TAM in ER+/HER-2-/ bcl2+ tumors. P53, EGFR1, PAK1, IGFR1 expression had no predictive value. Conclusions: PR expression, probably reflective of ER activation and functionality, strongly predicts benefit from continuation of TAM after 2–3 yrs. These data could provide a rationale for evaluating the efficacy of TAM after 5 years of anti-aromatase therapy in post-menopausal women with ER+/PR+/bcl2+/Her2-tumors. Thermally assisted machining (TAM) is an effective method for difficult-to-cut materials, and works by locally preheating the workpiece using various heat sources, such as laser, induction, and plasma. Recently, many researchers have studied TAM because of its low manufacturing costs, high productivity, and quality of materials. Laser assisted machining (LAM) has been studied by many researchers, but studies on TAM using induction or plasma heat sources, which are much cheaper than lasers, have been carried out by only a few researchers. Lasers have an excellent preheating effect, but are expensive, and the temperature of the heated workpiece drops quickly. Here, multi heat sources were used to solve the shortage in supplied heat source with a single heat source. Induction was applied as an additional heat source. The purpose of this study is to analyze the thermal effect and temperature distribution of single heat source and multi heat sources, and compare the machining characteristics according to heat source types. In order to analyze the preheating effect according to the feed rate of the heat sources, a temperature measurement experiment using thermocouples was carried out, and the efficiency of the thermal effect using multi heat sources was verified. In addition, the effectiveness of the thermal analysis results was verified by comparison with the measured temperature distribution. The machining characteristics of Inconel 718 and Ti-6Al-4V with laser, induction, and laser-induction assisted milling (LIAMill) were analyzed, by cutting force and surface roughness.

In this paper, a multi-tag optimization method based on image analysis and particle swarm optimization (PSO) neural network is proposed to verify the effect of radio frequency identification (RFID) multi-tag distribution on the performance of the system. A RFID tag detection system is proposed with two charge coupled device (CCD). This system can automatically focus on the tag according to its position, so it can obtain the image information more accurately by template matching and edge detection method. Therefore, the spatial structure of multi-tag and the corresponding reading distance can be obtained for training. Because of its excellent performance in multi-objective optimization, the PSO neural network is used to train and predict multi-tag distribution at the maximum reading distance. Compared with other neural networks, PSO is more accurate and its uptime is shorter for RFID multi-tag analysis.

The quality of multi-tag imaging greatly affects the effective detection of multi-tag. When multi-tag moves rapidly, the image may have serious dynamic blur, and tags can not be detected efficiently. In previous work, it is generally assumed that blur kernel and noise stationary to improve image quality. However, the dynamic deblurring of Radio Frequency Identification (RFID) multi-tag imaging is an ill-posed inverse problem. In this paper, firstly, blur-sharp multi-tag image pairs are made by superimposing and averaging the adjoin random frames. Then, we propose blind deblurring for dynamic RFID multi-tag imaging based on conditional generative adversarial nets (CGANs), which adds perceptual loss and content loss to generator to make image sharper. Finally, tags are detected by YOLOv3 in real time in end-to-end manner. Experimental results demonstrate that PSNR is at least 0.56dB higher and speed is at least 31.25 % faster than that of the current improved convolution neural networks (CNN). CGANs can remove image blur better, which has great superiority in the field of dynamic multi-tag imaging. In addition, YOLOv3 detects multi-tag quickly, thereby improving the detection accuracy.

Cash settlement accounts for 80% of entire transaction in Japan. Japanese government has announced the “Payment Reform Declaration,” by setting “the Cashless Vision” with a target of 40% cashless payment ratio by 2025 and aims to achieve the world's highest level of cashless payment ratio of 80% in the future. The purpose of this study is to analyze which media has affected the consumers who use QR code payment, PayPay. This research proposes a framework based on the Technology Acceptance Model (TAM) for an effective advertisement medium by using survey data on PayPay. Structure equation analysis is conducted to measure the relationship between different advertising media and consumers’ intentions to use, as well as actual usage of PayPay.

There has recently been an increasing interest in Learning Management Systems (LMSs). It is currently unclear, however, exactly how these systems are perceived by their users. This article analyzes data on user acceptance for two LMSs (Blackboard and Canvas). The respective data are collected using a questionnaire modeled after the Technology Acceptance Model (TAM); it relates several variables that influence system acceptability, allowing for a detailed analysis of the system acceptance. We present analyses at two levels of the questionnaire data: questions and constructs (taken from TAM) as well as on different analysis levels using targeted methods. First, we investigate the differences between the above LMSs using statistical tests (t-test). Second, we provide results at the question level using descriptive indices, such as the mean and the Gini heterogeneity index, and apply methods for ordinal data using the Cumulative Link Mixed Model (CLMM). Next, we apply the same approach at the TAM construct level plus descriptive network analysis (degree centrality and bipartite motifs) to explore the variability of users’ answers and the degree of users’ satisfaction considering the extracted patterns. In the context of TAM, the statistical model is able to analyze LMS acceptance on the question level. As we are also very much interested in identifying LMS acceptance at the construct level, in this article, we provide both statistical analysis as well as network analysis to explore the connection between questionnaire data and relational data. A network analysis approach is particularly useful when analyzing LMS acceptance on the construct level, as this can take the structure of the users’ answers across questions per construct into account. Taken together, these results suggest a higher rate of user acceptance among Canvas users compared to Blackboard both for the question and construct level. Likewise, the descriptive network modeling for Canvas indicates a slightly higher concordance between Canvas users than Blackboard at the construct level.

This paper presents the Topic-Aspect Model (TAM), a Bayesian mixture model which jointly discovers topics and aspects. We broadly define an aspect of a document as a characteristic that spans the document, such as an underlying theme or perspective. Unlike previous models which cluster words by topic or aspect, our model can generate token assignments in both of these dimensions, rather than assuming words come from only one of two orthogonal models. We present two applications of the model. First, we model a corpus of computational linguistics abstracts, and find that the scientific topics identified in the data tend to include both a computational aspect and a linguistic aspect. For example, the computational aspect of GRAMMAR emphasizes parsing, whereas the linguistic aspect focuses on formal languages. Secondly, we show that the model can capture different viewpoints on a variety of topics in a corpus of editorials about the Israeli-Palestinian conflict. We show both qualitative and quantitative improvements in TAM over two other state-of-the-art topic models.

9028 Background: Up to 50% of breast cancer patients on aromatase inhibitor therapy report significant musculoskeletal symptoms such as joint and muscle pain, which decreases treatment adherence. We conducted a secondary data analysis of a multi-site, phase III randomized, controlled, clinical trial examining the efficacy of yoga for improving musculoskeletal symptoms among breast cancer patients currently receiving hormone therapy (aromatase inhibitors [AI] or tamoxifen [TAM]) through the University of Rochester Cancer Center Community Clinical Oncology Program (CCOP). Methods: The original RCT randomized patients with any type of non-metastatic cancer without previous yoga participation into 2 arms: 1) standard care monitoring [controls] or 2) 4-week yoga intervention (2x/wk; 75 min/session) plus standard care. The yoga intervention utilized the UR Yoga for Cancer Survivors (YOCAS) program consisting of breathing exercises, 18 Hatha and Restorative yoga postures, and meditation. Only breast cancer patients currently receiving AI (N=95) or TAM (N=72) were included in this secondary analysis. Changes in musculoskeletal symptoms were assessed using ANCOVA with baseline values as covariates between the yoga and control groups. Results: Compared to TAM users at baseline, AI users reported higher levels of general pain (1-5 score: AI=2.65 vs. TAM=2.17; p=0.01), muscle aches (0-4 score: AI=2.14 vs. TAM=1.65; p=0.01), and total physical discomfort (0-24: AI=8.03 vs. TAM=5.92; p=0.01). Among AI users only, participants in the yoga group demonstrated greater reductions in general pain (CS=change score; Yoga CS= -0.37 vs. Control CS= +0.02; p=0.02), muscle aches (Yoga CS= -0.58 vs. Control CS= -0.15; p=0.03) and total physical discomfort (Yoga CS= -2.07 vs. Control CS= -0.58; p=0.04) from pre- to post-intervention than the control group. Conclusions: The severity of musculoskeletal symptoms was higher for AI users than for TAM users. Among breast cancer patients on AI therapy, the community-based YOCAS intervention significantly reduced general pain, muscle aches, and physical discomfort. In project development it is hardly possible to get exhaustive and accurate information. As a result, the situations occur, the consequences of which can be very damaging to the project. Inaccurate evaluation of the strategy related to capital investment and project implementation is one of the reasons why such estimates are not required in practice. Instead, a classification approaches may be used for this purpose. The decision‐making process, based on the established risk assessment principles expressed in linguistic terms, requires qualitative judgement and experiential knowledge of the construction experts. Presented structured and realistic methods deals systematically with different risk management situations and assist the investors in reaching the correct risk assessment of possible alternatives will be of great value. This paper presents a methods of multiattribute comparativite analysis (CLARA and SAW methods) of variants of investment classified risks in construction. A practical case to illustrate how the methods works is presented. Santrauka Šiame straipsnyje nagrinėjamos daugiatikslės analizės metodų taikymo galimybės. Svarbus investicijų efektyvumo garantas yra nuodugni investicinių projektų analizė ir vertinimas. Tai leidžia pagal tam tikrus kriterijus nustatyti investavimo variantų efektyvumą. Vertinimo kriterijai turi būti grindžiami tiek investicinį projektą įgyvendinančio subjekto, tiek investuotojo interesais. With the advent of electric vehicle multi-machine drives are attaining overwhelming responses from the researchers and industries in recent years, as compared to their counterpart of single machine drive. In this regard, the industries are looking for multi motor control with single inverter system with precise speed control. The solution of aforesaid problem lies with multi-leg inverter fed dual induction motor drives that are capable for high power ratings and other specific applications. Any faults in the system leads to the failure of the operation of the entire drive system. Hence condition monitoring of the entire drive system becomes of paramount significance. Considering the aforementioned points, this paper focuses on the fault analysis of five leg voltage source inverter feeding dual induction motors. The drive system is simulated using MATLAB/ SIMULINK for different pulse width modulation techniques like SPWM, SVPWM and Two Arm Modulation (TAM) Techniques. The effect on fault in the inverter like opening of the switch and shorting the switch, on the performance of the induction motors are analysed. Total Harmonic Distortion (THD) of the stator current for different modulation techniques are compared for the analysis purpose. From the results it is observed that the THD is less for SVPWM techniques as compared with SPWM and TAM method. But independent control of both the machine cannot be attained by SPWM and SVPWM method. For independent speed control TAM method is used. So a trade-off has to be done considering the requirement and THD. It is finally concluded that independent speed control is achieved at the cost of higher THD.

The tyrosine kinase receptors of the TAM family—Tyro3, Axl and Mer—and their main ligand Gas6 (growth arrest-specific 6) have been implicated in several human diseases, having a particularly important role in the regulation of innate immunity and inflammatory response. The Gas6/TAM system is involved in the recognition of apoptotic debris by immune cells and this mechanism has been exploited by viruses for cell entry and infection. Coronavirus disease 2019 (COVID-19) is a multi-systemic disease, but the lungs are particularly affected during the acute phase and some patients may suffer persistent lung damage. Among the manifestations of the disease, fibrotic abnormalities have been observed among the survivors of COVID-19. The mechanisms of COVID-related fibrosis remain elusive, even though some parallels may be drawn with other fibrotic diseases, such as idiopathic pulmonary fibrosis. Due to the still limited number of scientific studies addressing this question, in this review we aimed to integrate the current knowledge of the Gas6/TAM axis with the pathophysiological mechanisms underlying COVID-19, with emphasis on the development of a fibrotic phenotype.

M-Attendance merupakan sistem absensi online yang dapat menyederhanakan metode pencatatan kehadiran mahasiswa sehingga dapat mengurangi kesalahan serta mempercepat verifikasi. Penelitian ini bertujuan untuk mengetahui tingkat kesuksesan penerapan sistem M-Attendance Student sebagai absensi mahasiswa/i di Universitas Sari Mulia (UNISM) dengan menggunakan metode TAM (Technology Acceptance Model) dan UTAUT (Unified Theory of Acceptance and Use of Technology). Metode TAM dapat digunakan untuk menjelaskan faktor-faktor dari perilaku pengguna terhadap penerimaan teknologi perilaku penerimaan pengguna terhadap teknologi, sedangkan melalui UTAUT dapat dipahami bahwa reaksi dan persepsi pengguna terhadap teknologi dapat mempengaruhi sikapnya dalam penerimaan penggunaan teknologi. Penelitian ini menggunakan semua variabel dari kedua metode tersebut. Pengukuran data yang dianalisis pada penelitian ini menggunakan kuesioner yang disebarkan kepada 100 responden yaitu mahasiswa/i di Universitas Sari Mulia. Berdasarkan hasil analisis metode TAM diperoleh bahwa niat penggunaan berpengaruh terhadap penggunaan sistem dengan sesungguhnya sebesar 77%, niat penggunaan dipengaruhi oleh sikap terhadap penggunaan sebesar 41,7% dan persepsi terhadap kebermanfaatan sebesar 43%, sikap terhadap penggunaan dipengaruhi oleh persepsi terhadap kebermanfaatan sebesar 41,5% dan persepsi terhadap kemudahan penggunaan sebesar 49,7%, persepsi terhadap kemudahan penggunaan berpengaruh terhadap persepsi terhadap kebermanfaatan sebesar 87,9%. Sedangkan berdasarkan hasil analisis metode UTAUT diperoleh bahwa niat penggunaan dan kondisi yang memfasilitasi berpengaruh terhadap perilaku pengguna sebesar 68% dan 31%, perilaku penggunaan hanya dipengaruhi oleh ekspektasi usaha sebesar 56% dan pengaruh sosial sebesar 44,6%. Ekspektasi terhadap kinerja tidak berpengaruh karena hanya mendapat nilai sebesar 0,13%.

The world of underground engineering and construction has acquired a wide‐ranging and high‐level experience on tunnel construction with Tunnel Boring Machines (TBM) and nowadays remarkable progresses are traceable in the number of tunnels that are becoming longer, going deeper, and growing larger in diameter and in other words becoming more difficult to realize. Tabriz‐one of the big cities in northern west of Iran has four subway lines which are under construction or investigation. The phase1 design of Tabriz urban railway line 2 (TURL2) has completely been done. Method statement of this line in the length of about 20 km and much interference due to tunneling in urban area dictates the application of TBM. Two kinds of TBM such as EPB (earth pressure balance) and SS (slurry shield) are usually used for urban areas. In this paper, the process of choosing TBM for TURL2 using MCA method (Multi Criteria Analysis) is expressed. Generally in this method some technical, economical and environmental parameters affected the TBM type are identified and taken into account by assigned weights related to the case study. Finally the results show that EPB‐TBM will be more appropriate choice for TURL2 excavation. e13019 Background: TAM is mainly metabolized by CYP2D6 to form its most active metabolites, 4hydroxy-tamoxifen (4OH-T) and endoxifen (END). Because of its long half-life, steady state is reached after around 4 months of continuous intake. The wide variable inter-patient activity of CYP2D6 might influence drug efficacy. A multi-institutional study in north Italy is evaluating the relationship between END levels and outcome. As a part of it, we investigated the role of dextromethorphan (DM), a probe drug for CYP2D6 enzymatic activity, as a potential phenotyping test for TAM activation. Methods: Twenty-nine breast cancer patients (75% postmenopausal) on adjuvant TAM therapy (20 mg/die) were investigated. They received a single dose (15 mg) of oral DM before starting TAM and their urines were collected over the10 following hours. Simultaneous quantitative determination of DM and its metabolite dextrorphan (DO) was performed in urines to estimate their log transformed metabolic ratio (LMR=logDM/DO). After 4 months a blood sample was collected to characterize TAM exposure at steady state; plasma levels of TAM, END, 4OH-T and the non active END precursor N-desmethyltamoxifen (NDT) were quantified by HPLC. Linear regression analysis and t test were performed for correlating LMR and drug plasma levels. Results: LMR varied between -2.15 and 0.90 (median: -1.37) while steady state plasma levels of END varied between 1.9 and 15.0 ng/ml (median: 4.36) and 4OH-T between 0.9 to 3.1 ng/ml (median:1.72). A significant correlation (r = 0.56; p= 0.0013) was found between LMR and END plasma concentrations. The patients with high LMR (> median value), compared to patients with low LMR, had lower END (3.7 vs 7.5 ng/ml, p=0.0003), lower 4OH-T (1.6 vs 2.1 ng/ml, p=0.04) and, accordingly, higher NDT (291.2 vs 198.2 ng/ml, p=0.025). Conclusions: DM/DO urine ratio obtained before starting therapy correlates with TAM biotransformation activity and can predict steady state active metabolites exposure in individual patients. This phenotyping test is fast, simple and unexpensive and could contribute to the personalization of adjuvant breast cancer treatment. Funded by Regione Veneto.

Artificial intelligence (AI) chatbots are set to be the defining technology of the next decade due to their ability to increase human capability at a low cost. However, more research is required to assess individuals’ behavioural intentions to use this technology when it becomes publicly available. This study applied an extended Technology Acceptance Model (TAM), with additional predictors of trust and privacy concerns, to assess individuals’ behavioural intentions to use AI chatbots across three industries: mental health care, online shopping, and online banking. These services were selected due to the current popularity of regular chatbots in these fields. Participants ( N = 360 , 202 females) aged between 17 and 85 years ( M = 38.17 , SD = 17.66 ) completed a 71-item online, cross-sectional survey. As hypothesised, perceived usefulness and trust were significant positive predictors of behavioural intentions across all three behaviours. However, the influence of the perceived ease of use and privacy concerns on behavioural intentions differed across the three behaviours. These findings highlight that the combination of predictors within the extended TAM have different influences on behavioural intentions to use AI chatbots for mental health care, online shopping, and online banking. This research contributes to the literature by demonstrating that the influence of the variables in one field cannot be generalised across all uses of AI chatbots.

Abstract Extensive infiltration of tumor-associated macrophages (TAMs) into the solid tumor microenvironment (TME), where they promote angiogenesis, initiate metastasis, and tune immunosuppression, has long been proven to correlate with worse patient prognosis. Thus, much significance has been attached on either impeding TAM infiltration or reversing pro-tumoral TAMs back into phagocytic M1 phenotype. Nevertheless, while revealing underlying mechanisms of blocking TAM recruitment, widely adopted in vitro migration assays like Transwell, µ-slides fail to distinguish cell’s random migration from chemotaxis (biased migration) and introduce 3D extracellular matrix environment. Here we present data that monocytes, as major replenishment source of TAMs, massively boost their random migration via paracrine pathway to initiate early-stage infiltration using an advanced 3D multi-compartment organoid model. In addition, before re-educated into TAMs, monocytes in the TME are capable of inducing triple negative breast cancer cells’ hyper-proliferation by activating their MEK/ERK signaling pathway. As a non-contact automatic identification technology, Radio Frequency Identification (RFID) is of great significance to improve the simultaneous identification of multi-target. This paper designs a more efficient and accurate multi-tag reading performance measurement system based on the fusion of YOLOv3 and Elman neural network. In the machine vision subsystem, multi-tag images are collected by dual CCD and detected by neural network algorithm. The reading distance of 3D distributed multi-tag is measured by laser ranging to evaluate the reading performance of RFID system. Firstly, the multi-tag are detected by YOLOv3, which realizes the measurement of 3D coordinates, improves the prediction accuracy, enhances the recognition ability of small targets, and improves the accuracy of 3D coordinate detection. Secondly, the relationship between the 3D coordinates and the corresponding reading distance of RFID multi-tag are modelled by Elman recurrent neural network. Finally, the reading performance of RFID multi-tag is optimized. Compared with the state-of-the-arts, the multi-tag detection rate of YOLOv3 is 17.4% higher and the time is 3.27 times higher than that of the previous template matching algorithm. In terms of reading performance, the MAPE of Elman neural network is 1.46 %, which is at least 21.43 % higher than other methods. In running time, Elman only needs 1.69s, which is at least 28.40% higher than others. Thus, the system not only improves the accuracy, but also improves the speed, which provides a new insight for the measurement and optimization of RFID performance.

Fimbriae are long, adhesive structures widespread throughout members of the family Enterobacteriaceae. They are multimeric extrusions, which are moved out of the bacterial cell through an integral outer membrane protein called usher. The complex folding mechanics of the usher protein were recently revealed to be catalysed by the membrane-embedded translocation and assembly module (TAM). Here, we examine the diversity of usher proteins across a wide range of extraintestinal (ExPEC) and enteropathogenic (EPEC) Escherichia coli , and further focus on a so far undescribed chaperone–usher system, with this usher referred to as UshC. The fimbrial system containing UshC is distributed across a discrete set of EPEC types, including model strains like E2348/67, as well as ExPEC ST131, currently the most prominent multi-drug-resistant uropathogenic E. coli strain worldwide. Deletion of the TAM from a naive strain of E. coli results in a drastic time delay in folding of UshC, which can be observed for a protein from EPEC as well as for two introduced proteins from related organisms, Yersinia and Enterobacter . We suggest that this models why the TAM machinery is essential for efficient folding of proteins acquired via lateral gene transfer.

The abundance of Web 2.0 social media in various media formats calls for integration that takes into account tags associated with these resources. The authors present a new approach to multi-modal media search, based on novel related-tag graphs, in which a query is a resource in one modality, such as an image, and the results are semantically similar resources in various modalities, for instance text and video. Thus the use of resource tagging enables the use of multi-modal results and multi-modal queries, a marked departure from the traditional text-based search paradigm. Tag relation graphs are built based on multi-partite networks of existing Web 2.0 social media such as Flickr and Wikipedia. These multi-partite linkage networks (contributor-tag, tag-category, and tag-tag) are extracted from Wikipedia to construct relational tag graphs. In fusing these networks, the authors propose incorporating contributor-category networks to model contributor’s specialization; it is shown that this step significantly enhances the accuracy of the inferred relatedness of the term-semantic graphs. Experiments based on 200 TREC-5 ad-hoc topics show that the algorithms outperform existing approaches. In addition, user studies demonstrate the superiority of this visualization system and its usefulness in the real world.

Abstract Patients with hematological malignancies can be successfully treated with allogeneic stem cell transplantation (alloSCT). The therapeutic potential of alloSCT in the HLA-matched setting is primarily mediated by donor T cells directed against minor histocompatibility antigens (MiHA) presented in self-HLA. In addition, tumor associated antigens (TAA) represent a diversity of proteins overexpressed by various malignancies (self-peptide recognized in self-HLA) that may be recognized by donor T cells. Early application of donor T cells after alloSCT often results in graft versus host disease (GVHD) besides graft versus leukemia (GVL) responses. After T cell depleted transplantation, patients are vulnerable to opportunistic viral infections and disease relapses. Adoptive transfer of a multi-antigen specific T cell product containing T cells directed against viruses, TAA and MiHA may restore anti-viral immunity and support the GVL effect early after alloSCT. Detectable frequencies of high avidity memory virus specific T cells can be easily enriched from blood of seropositive healthy donors. However, due to the extremely low precursor frequencies of TAA and MiHA specific T cells, the isolation of these cells is more complicated. Moreover, high avidity, self-HLA restricted TAA specific T cells may have been deleted during thymic selection. In this study (T-control, EU FP7), we aim to treat patients with a prophylactic infusion of a multi-antigen specific T cell product consisting of T cells directed against MHC class I restricted peptides of CMV, EBV and adenovirus, HLA-A2 restricted peptides from the TAA WT1, RHAMM, NY-eso and the peptide PR1 derived from proteinase 3, and the MiHA HA-1h. The reversible streptamer technology allows purification of minimally manipulated T cells from donor peripheral blood mononuclear cells (PBMC) and is based on strep-tag labeled peptide/MHC complexes coupled to Strep-Tactin coated magnetic nanobeads, followed by purification using the CliniMACS under Good Manufacturing Practice (GMP) conditions. In large scale validation runs (n=3) we purified up to 12 viral, TAA and MiHA specificities out of ≥500*10^6 donor PBMC in one procedure. This resulted in an immediately clinical applicable multi-antigen specific T cell product with a purity of ≥98% within the T cell compartment. As expected the the different viral specificities comprised the main portion of the product (98%). To confirm that the remaining part of the product contains T cells directed against TAA and MiHA, we performed subsequent individual streptamer enrichments. After each enrichment the isolated cells were non-specifically expanded with allogeneic feeders, IL-2 and PHA. Starting with 100*10^6 PBMC (n=4), we could detect in every donor ≥ 3 different TAA and/or MiHA specific T cell populations after 2 or 3 enrichments. Moreover, when starting with 500*10^6 PBMC (n=3) all TAA and MiHA specific T cells could be enriched to detectable frequencies (1-78%) after 2 or 3 enrichments. This indicates that TAA and MiHA specific T cells are commonly present in healthy donors, although at very low frequencies. The TAA and MiHA specific T cell populations were clonally isolated and expanded, evaluated for CD8 and tetramer positivity and screened for antigen specific reactivity measured by cytokine release (IFN-g and GM-CSF) after 24 hours of stimulation with TAP deficient T2 cells loaded with 10^-5M of the relevant peptide. From the tetramer positive T cell clones 8/132 NY-eso clones, 8/17 RHAMM clones, 31/47 WT1 clones and 13/16 PR1 clones showed peptide recognition. Among these were TAA clones of intermediate avidity recognizing up to 10^-7M peptide, despite the anticipated self-tolerance. As expected, the HA-1h clones recognizing a foreign MiHA in self HLA displayed high functional avidity reflected by recognition of peptide-loaded T2 cells in the pM range. This illustrates that intermediate avidity TAA-specific and high avidity HA-1h specific T cell clones can be enriched from healthy donors and that these cells may have therapeutic potential. In conclusion, we are able to generate from donor PBMC a highly purified multi-antigen specific T cell product for clinical application. Besides the main component of MHC class I restricted viral specific T cells, we have demonstrated that this product also contains highly proliferative high avidity MiHA specific T cells and TAA specific T cells of intermediate avidity. People who live in urban areas can be explained as social groups that are sociologically in the middle class, have a productive life, and are part of social change. Shopee as one of the marketplaces in Indonesia always needs to consider the factor of technology usage that allows users to make purchases with a variety of products (multi-product) available. This study aims to analyze the behavior of using Urban Consumer Behavior with the Technology Acceptance Model (TAM) approach. Data analysis using SMART PLS 3.0 involving 163 samples in Jakarta with survey-questionnaire as a method of data collection. The results of this study explain that two hypotheses were accepted and two more hypotheses were rejected. In this case, Shopee's website/app usage behavior is influenced by the Ease of Use Factor. Furthermore the Benefit factor is influenced by the Ease of Use factor. On the other hand, usage is not influenced by usage behavior. Then the use is not influenced by the expediency factor. TAM as a classic model in explaining technology adoption in aspects of daily life is still needed to gain more understanding, especially on the characteristics of various or different samples.

The main objective of this paper is to understand user interaction behaviour on social networking sites (SNS), and to investigate cultural influence on acceptance and use behaviour. SNS are growing in importance, and the many advantages they offer to companies are on the increase. The model described in this paper has been defined on the basis of a technology acceptance model (TAM) to examine the adoption and use of SNS, by adding trust and perceived risk constructs. To analyse the culture as a moderating effect of the causal relationships proposed, we focus on two European countries, using a multi-group structural equation model (SEM). This study shows that extended TAM (ETAM) is appropriate for use in predicting the acceptance of voluntary-use technologies, and it focuses on social relationships. The cultural effects may moderate some theoretical relationships in the adoption process.

Synopsis The present case study discusses the multi-branded strategy of hotels by the Indian Hotels Company Limited (IHCL) in 2012. The brand architecture of the group in 2012 consisted of four brands, namely Taj, Vivanta, Gateway and Ginger. However such brand architecture posed quite a few challenges for the group such as positioning, sustain the different brands and avoiding brand dilution since both The Gateway and Vivanta had a tag “by Taj” which could erode the premium associations of the parent brand. Research methodology The case is based on secondary research and has been developed using published information collected from online and offline sources. Store-operated Ca2+ entry (SOCE) is a ubiquitous mechanism regulating extracellular Ca2+ entry to control a multitude of Ca2+-dependent signaling pathways and cellular processes. SOCE relies on the concerted activity of the reticular Ca2+ sensor STIM1 and the plasma membrane Ca2+ channel ORAI1, and dysfunctions of these key factors result in human pathologies. STIM1 and ORAI1 gain-of-function (GoF) mutations induce excessive Ca2+ influx through SOCE over-activation, and cause tubular aggregate myopathy (TAM) and Stormorken syndrome (STRMK), two overlapping disorders characterized by muscle weakness and additional multi-systemic signs affecting growth, platelets, spleen, skin, and intellectual abilities. In order to investigate the pathophysiological effect of overactive SOCE on muscle function and structure, we combined transcriptomics with morphological and functional studies on a TAM/STRMK mouse model. Muscles from Stim1R304W/+ mice displayed aberrant expression profiles of genes implicated in Ca2+ handling and excitation-contraction coupling (ECC), and in vivo investigations evidenced delayed muscle contraction and relaxation kinetics. We also identified signs of reticular stress and abnormal mitochondrial activity, and histological and respirometric analyses on muscle samples revealed enhanced myofiber degeneration associated with reduced mitochondrial respiration. Taken together, we uncovered a molecular disease signature and deciphered the pathomechanism underlying the functional and structural muscle anomalies characterizing TAM/STRMK.

Abstract <Introduction> Transient abnormal myelopoiesis (TAM) occurs in about 10% of neonates with Down syndrome. While TAM is generally considered as self-limiting disease, substantial number of patients suffer from serious complications including liver fibrosis or pericardial effusion and eventually leads to fatal clinical outcomes. Low-dose cytosine arabinoside (CA) therapy has been reported to be effective for clinical symptoms of TAM patients, but it is still controversial as to which patient should be treated. We have retrospectively reviewed the clinical outcomes of TAM patients in our institution to confirm the efficacy as well as the safety of low-dose CA therapy by comparing the clinical outcomes before and after introduction of CA. <Patients> Before introduction of CA, we have experienced 20 TAM patients of 13 male and 7 female between September 1992 and November 2008. The median gestational age of those was 37w4d (range, 31w5d - 39w2d) , median peak value of white blood cell (WBC) count was 48×109/L (range, 19.8 - 399×109/L), and median peak direct bilirubin(DB) level was 0.9 mg/dL (range, 0.1 - 18.4 mg/dL), respectively. Sixteen of twenty patients (80%) had various complications including dyspnea, hepatosplenomegaly, pericardial effusion, and ascites. Since 2009, we have introduced low dose CA (0.4 - 1.0mg/kg/day) for those with high WBC count (>100×109/L) until WBC count decrease to 10×109/L . Three patients received chemotherapy, but one patient with high WBC count did not, because consent was not obtained. The median gestational age of those was 36w4d (range, 33w6d - 39w2d) , median peak value of WBC count was 106×109/L (range, 101 - 267×109/L), and median peak DB level was 1.95mg/dL (range, 1.0 - 11.0 mg/dL), respectively. All of them had various complications including dyspnea, hepatosplenomegaly, pericardial effusion, liver fibrosis and ascites. <Results> Before introduction of low dose CA therapy, five of six patients (83%) with high WBC count (>100×109/L) died of liver failure(4 of 5 patients) or AML, and one of 14 patients (7%) with low WBC count (<100×109/L) died of pneumonia at the age of eight without any relation to TAM. After introduction of CA, all three patients who received chemotherapy are alive without serious complications related to TAM or chemotherapy. Their blast cells in the peripheral blood disappeared promptly and liver dysfunction as well as biomarker of liver fibrosis is getting normalized. One patient who did not receive chemotherapy died of liver failure in spite of supportive care. <Conclusion> It is suggested that low dose CA therapy is safely conducted and is effective to improve the clinical outcomes of TAM patients with high WBC count. Prospective analysis is mandatory to confirm our findings by multi-institutional basis. Disclosures No relevant conflicts of interest to declare.

Abstract Background: Pediatric sarcoma represents the second most common solid tumor in children. Despite advances in multimodal therapies, one-third of sarcoma patients do not survive the disease. Over the past few decades, the development of novel therapeutic strategies for sarcoma has been impeded by an insufficient understanding of the tumor microenvironment (TME). In particular, the potential of immunotherapy remains constrained by the gap of knowledge about the molecular and functional phenotypes of the immune cells in the sarcoma TME. Methods: To address this knowledge gap, we collected treatment-naïve tumor samples from 32 patients with five pediatric sarcoma subtypes. We employed a single-nuclei RNA+ATAC-Seq multi-omics assay to comprehensively characterize the transcriptomic and epigenomic heterogeneity of tumor and immune cells within the sarcoma TME. Results: Our analysis revealed that tumor-associated macrophages (TAMs) were the most abundant immune cell type in the sarcoma TME. We identified seven distinct TAM subpopulations, each exhibiting unique gene expression and chromatin accessibility patterns. Among these, TAM subsets with higher expression of SPP1 and C1QC genes displayed the strongest immunosuppressive potential. Transcriptional regulatory network (TRN) analysis identified transcription factors that were most likely to drive the phenotypes of SPP1+ and C1QC+ TAMs, including ELF2, JUND and RUNX1. Through cell-cell communication analysis, we discovered that SPP1+ TAMs interact with T cells via SPP1-CD44 signaling, while C1QC+ TAMs interact with NK cells through HLA-E-KLRC1 signaling, suggesting the potential involvement of these signaling pathways in TAM-mediated suppression of these cytotoxic lymphocytes. We prioritized ligand-receptor gene pairs, such as SEMA3D-NRP1 and LTBP3-ITGB5, which likely mediate the interaction between malignant sarcoma cells and SPP1+/C1QC+ TAMs based on their dominance across sarcoma subtypes and association with poorer clinical outcomes. On the other hand, our analysis revealed that sarcoma subtype-specific interactions between TAMs and malignant cells are common, highlighting the heterogeneity of signaling events across sarcoma subtypes. Conclusion and Significance: Our study presents a comprehensive single-cell multi-omics characterization of pediatric sarcomas and sheds light on its tumor microenvironment. By identifying unique markers for immunosuppressive TAM subpopulations and elucidating intercellular interactions promoting tumor development, this study proposes potential therapeutic targets for sarcoma treatment based on the role of TAMs in sarcoma TME. In RFID-assisted applications, the customers often request to provide the tag searching service to determine which specific tags are present in the system. In practice, tags are usually divided into different groups to represent different categories or brands. We found that the traditional tag searching protocols are not very appropriate for multi-group RFID scenarios because they cannot ensure that the searching results of each group satisfy the predefined reliability requirements. Therefore, we develop a series of effective multi-group tag searching schemes. B-Search is a basic method that leverages the filter vector and the indicator vector to perform tag searching for each group sequentially. G-Search and A-Search are two parallel multi-group tag searching schemes. G-Search selects the longest frame length as the frame length of all groups, while A-Search can adaptively adjust the frame length of each group to improve the searching efficiency. We evaluate the performance of the proposed protocols through theoretical analysis and discuss the optimal parameter settings to minimize the execution time. Extensive simulations illustrate that both G-Search and A-Search can achieve fast and effective multi-group tag searching.

Interpreting how multicellular interactions in the tumor affect resistance pathways to BRAF and MEK1/2 MAPK inhibitors (MAPKi) remains a challenge. To investigate this, we profiled global ligand-receptor interactions among tumor and stromal/immune cells from biopsies of MAPK-driven disease. MAPKi increased tumor-associated macrophages (TAMs) in some patients, which correlated with poor clinical response, and MAPKi coamplified bidirectional tumor-TAM signaling via receptor tyrosine kinases (RTKs) including AXL, MERTK, and their ligand GAS6. In xenograft tumors, intravital microscopy simultaneously monitored in situ single-cell activities of multiple kinases downstream of RTKs, revealing MAPKi increased TAMs and enhanced bypass signaling in TAM-proximal tumor cells. As a proof-of-principle strategy to block this signaling, we developed a multi-RTK kinase inhibitor nanoformulation that accumulated in TAMs and delayed disease progression. Thus, bypass signaling can reciprocally amplify across nearby cell types, offering new opportunities for therapeutic design.

We propose an improved image matching algorithm that combines the minimum feature value algorithm to extract feature points and the direction gradient histogram to calculate the description vector. This algorithm is oriented to RFID multi-tag identification and distribution optimization in the actual scenario, and the traditional SURF algorithm has the problems of low matching accuracy and high complexity in multi-tag matching. This algorithm effectively improves the positioning accuracy of the RFID multi-tag positioning system. The experimental results show that the matching success rate of the improved algorithm in this paper is 87.4%, which is 50% higher than the SURF algorithm. Not only the matching accuracy is greatly improved, but the running speed is also increased by 48%. The algorithm in this paper has high matching accuracy and real-time performance.It provides an effective way for RFID multi-tag real-time fast matching and precise positioning.

The selection of texture information and block ringing effect in multi-focus fusion process, a new multi-focus image fusion algorithm based on Three Activity Measures (TAM) excitation Spiking Cortical Model, (SCM) in shearlet (SHT) domain is proposed. Firstly, in SHT domain, using local spatial frequency (SF), local energy of gradient (EOG) and different measurements (SF, EOG, and local laplace energy sum (SML)) motivated SCM selected the texture information and construct the initial fusion image (P). Then, the focal region was extracted from the significant feature of the difference between the image P and the original image. Finally, the joint focus area produces fusion images. To verify the superiority of the proposed algorithm, compare the results of this paper with seven competing methods. Experimental results show that the algorithm can produce clear edges, good visual perception and less distortion.

Transportation asset management (TAM) practices continue to grow and to develop as transportation agencies seek to make more objective and defensible decisions, as well as responding to recent legislation. One primary goal of TAM is to provide a structure in which decisions on how to distribute resources across many disparate assets can be made using a systematic process. Resource allocation is analogous to multi-objective optimization, and thus presents the complication that many potential optimal solutions (i.e., a Pareto set) can be found. To solve the multi-objective resource allocation problem, many approaches have been recommended, such as the use of utility theory and weighting functions to express preferences that result in a single solution being selected. This paper discusses those recommendations, as well as describing the current state of the resource allocation process in an effort to identify gaps in practice and, in turn, to provide recommendations for addressing those gaps. First, the process of cross-asset resource allocation is deconstructed to highlight the key steps. Then, current practices identified from the literature, as well as from interviews with five State agencies are discussed. Finally, a set of recommendations for improving resource allocation and the supporting processes within resource allocation are presented.

The increasing rate of population growth in urban areas to find work or study and the high Basic Credit Interest Rates for Home Ownership Loans, the choice of renting a boarding house can be an alternative for those who do not want to stay in the long term. Implementation of Decision Support System provide choices for information system in order to assist the community in choosing a place to live such as a boarding house. One method of Decision Support System is Multi Attribute Utility Theory (MAUT). MAUT can be interpreted as method of systematic comparison by finding the total weight of a set of values in the criteria to obtain results. MAUT method on a web-based information system, it can help people determine the choice of the desired boarding house. The results from the MAUT method will be used an objective consideration for users. The results of this study were tested by Technology Acceptance Model to measure the acceptance of systems. The calculation of TAM uses questionnaire distributed to 88 respondents and based on t-statistics on the TAM test, Perceived Ease of Use (PEOU) against Attitude Toward Using (ATU) is 2.660, Perceived Usefulness (PU) against ATU is 4.218. Then Behavior Intention to Use (BITU) for Actual System Use (ASU) is 16,122 and PU for BITU is 4,218. Where the indicator to have a positive influence when the value is above 1.9894. Meanwhile, ATU against BITU is only 1.179 which means that it does not have a positive influence.
