Resumen
We have identified a new binding partner of the TGFβ (transforming growth factor-β)-activated protein kinase (TAK1), termed TAB3 (TAK1-binding protein-3), which shares 48% amino acid sequence identity with TAB2. Our results indicate that two distinct TAK1 complexes are present in cells. One comprises TAK1 complexed with TAB1 and TAB2, and the other TAK1 complexed with TAB1 and TAB3. Both complexes are activated in response to tumour necrosis factor-α or interleukin-1 in human epithelial KB cells or bacterial lipopolysaccharide in RAW264.7 macrophages, and are subject to feedback control by stress-activated protein kinase 2a (SAPK2a; also called p38α). The electrophoretic mobility of TAB2 and TAB3 decreases in response to these agonists or osmotic shock, and is reversed by treatment with protein phosphatase-1. The decrease in mobility of TAB3 is prevented if the cells are incubated with SB 203580 before stimulation, but treatment with SB 203580 produces forms of TAB2 with a mobility intermediate between that observed for TAB2 in unstimulated and stimulated cells. Similar results were obtained in embryonic fibroblasts from mice deficient in SAPK2a/p38α. Our results indicate that TAB3 is phosphorylated via the SAPK2a/p38α pathway, whereas TAB2 is phosphorylated at two or more sites by both an SAPK2a/p38α-dependent and an SB 203580-independent kinase. The SAPK2a/p38α-mediated phosphorylation of TAB2 and TAB3 may contribute to the SAPK2a/p38α-mediated feedback control of TAK1 activity that also involves the phosphorylation of TAB1. We also show that the agonist-induced activation of TAK1 complexes requires the phosphorylation of the TAK1 catalytic subunit at a serine/threonine residue(s).

Resumen
Interleukin-1 (IL-1) signaling induces the formation of Lys63-linked ubiquitin (K63-Ub) chains, which are thought to activate the ‘master’ protein kinase TGFβ-activated kinase 1 (TAK1) by interacting with its TAK1-binding 2 (TAB2) and TAB3 subunits. Here, we report that IL-1β can also activate the TAB1–TAK1 heterodimer present in TAB2/TAB3 double knockout (DKO) IL-1 receptor-expressing cells. The IL-1β-dependent activation of the TAB1–TAK1 heterodimer in TAB2/3 DKO cells is required for the expression and E3 ligase activity of tumor necrosis factor receptor-associated factor 6 (TRAF6) and is reduced by the small interfering RNA (siRNA) knockdown of ubiquitin conjugating 13 (Ubc13), an E2-conjugating enzyme that directs the formation of K63-Ub chains. IL-1β signaling was restored to TAB1/2/3 triple KO cells by the re-expression of either TAB1 or TAB2, but not by an ubiquitin binding-defective mutant of TAB2. We conclude that IL-1β can induce the activation of TAK1 in two ways, only one of which requires the binding of K63-Ub chains to TAB2/3. The early IL-1β-stimulated, TAK1-dependent activation of p38α mitogen-activated protein (MAP) kinase and the canonical IκB kinase (IKK) complex, as well as the NF-κB-dependent transcription of immediate early genes, was similar in TAB2/3 DKO cells and TAB2/3-expressing cells. However, in contrast with TAB2/3-expressing cells, IL-1β signaling was transient in TAB2/3 DKO cells, and the activation of c-Jun N-terminal kinase 1 (JNK1), JNK2 and p38γ was greatly reduced at all times. These observations indicate a role for TAB2/3 in directing the TAK1-dependent activation of MAP kinase kinases that switch on JNK1/2 and p38γ MAP kinases. These observations and the transient activation of the TAB1–TAK1 heterodimer may explain why IL-1β-dependent IL-8 mRNA formation was abolished in TAB2/3 DKO cells.

Resumen
The protein kinase TAK1 (transforming growth factor-β-activated kinase 1), which has been implicated in the activation of MAPK (mitogen-activated protein kinase) cascades and the production of inflammatory mediators by LPS (lipopolysaccharide), IL-1 (interleukin 1) and TNF (tumour necrosis factor), comprises the catalytic subunit complexed to the regulatory subunits, termed TAB (TAK1-binding subunit) 1 and either TAB2 or TAB3. We have previously identified a feedback-control mechanism by which p38α MAPK down-regulates TAK1 and showed that p38α MAPK phosphorylates TAB1 at Ser423 and Thr431. In the present study, we identified two IL-1-stimulated phosphorylation sites on TAB2 (Ser372 and Ser524) and three on TAB3 (Ser60, Thr404 and Ser506) in human IL-1R cells [HEK-293 (human embryonic kidney) cells that stably express the IL-1 receptor] and MEFs (mouse embryonic fibroblasts). Ser372 and Ser524 of TAB2 are not phosphorylated by pathways dependent on p38α/β MAPKs, ERK1/2 (extracellular-signal-regulated kinase 1/2) and JNK1/2 (c-Jun N-terminal kinase 1/2). In contrast, Ser60 and Thr404 of TAB3 appear to be phosphorylated directly by p38α MAPK, whereas Ser506 is phosphorylated by MAPKAP-K2/MAPKAP-K3 (MAPK-activated protein kinase 2 and 3), which are protein kinases activated by p38α MAPK. Studies using TAB1−/− MEFs indicate important roles for TAB1 in recruiting p38α MAPK to the TAK1 complex for the phosphorylation of TAB3 at Ser60 and Thr404 and in inhibiting the dephosphorylation of TAB3 at Ser506. TAB1 is also required to induce TAK1 catalytic activity, since neither IL-1 nor TNFα was able to stimulate detectable TAK1 activity in TAB1−/− MEFs. Surprisingly, the IL-1 and TNFα-stimulated activation of MAPK cascades and IκB (inhibitor of nuclear factor κB) kinases were similar in TAB1−/−, MEKK3−/− [MAPK/ERK (extracellular-signal-regulated kinase) kinase kinase 3] and wild-type MEFs, suggesting that another MAP3K (MAPK kinase kinase) may mediate the IL-1/TNFα-induced activation of these signalling pathways in TAB1−/− and MEKK3−/− MEFs.

Resumen
Protease-activated receptor 1 (PAR1) is a G protein–coupled receptor (GPCR) for thrombin and promotes inflammatory responses through multiple pathways including p38 mitogen-activated protein kinase signaling. The mechanisms that govern PAR1-induced p38 activation remain unclear. Here, we define an atypical ubiquitin-dependent pathway for p38 activation used by PAR1 that regulates endothelial barrier permeability. Activated PAR1 K63-linked ubiquitination is mediated by the NEDD4-2 E3 ubiquitin ligase and initiated recruitment of transforming growth factor-β–activated protein kinase-1 binding protein-2 (TAB2). The ubiquitin-binding domain of TAB2 was essential for recruitment to PAR1-containing endosomes. TAB2 associated with TAB1, which induced p38 activation independent of MKK3 and MKK6. The P2Y1 purinergic GPCR also stimulated p38 activation via NEDD4-2–mediated ubiquitination and TAB1–TAB2. TAB1–TAB2-dependent p38 activation was critical for PAR1-promoted endothelial barrier permeability in vitro, and p38 signaling was required for PAR1-induced vascular leakage in vivo. These studies define an atypical ubiquitin-mediated signaling pathway used by a subset of GPCRs that regulates endosomal p38 signaling and endothelial barrier disruption.

Resumen
Abstract TGF-β activated kinase 1 (TAK1) is a mediator of various cytokine signaling pathways. Germline deficiency of Tak1 causes multiple abnormalities, including dilated blood vessels at midgestation. However, the mechanisms by which TAK1 regulates vessel formation have not been elucidated. TAK1 binding proteins 1 and 2 (TAB1 and TAB2) are activators of TAK1, but their roles in embryonic TAK1 signaling have not been determined. In the present study, we characterized mouse embryos harboring endothelial-specific deletions of Tak1, Tab1, or Tab2 and found that endothelial TAK1 and TAB2, but not TAB1, were critically involved in vascular formation. TAK1 deficiency in endothelial cells caused increased cell death and vessel regression at embryonic day 10.5 (E10.5). Deletion of TNF signaling largely rescued endothelial cell death in TAK1-deficient embryos at E10.5. However, embryos deficient in both TAK1 and TNF signaling still exhibited dilated capillary networks at E12.5. TAB2 deficiency caused reduced TAK1 activity, resulting in abnormal capillary blood vessels, similar to the compound deficiency of TAK1 and TNF signaling. Ablation of either TAK1 or TAB2 impaired cell migration and tube formation. Our results show that endothelial TAK1 signaling is important for 2 biologic processes in angiogenesis: inhibiting TNF-dependent endothelial cell death and promoting TNF-independent angiogenic cell migration.

Resumen
ABSTRACT The proinflammatory cytokine interleukin-1 (IL-1) transmits a signal via several critical cytoplasmic proteins such as MyD88, IRAKs and TRAF6. Recently, serine/threonine kinase TAK1 and TAK1 binding protein 1 and 2 (TAB1/2) have been identified as molecules involved in IL-1-induced TRAF6-mediated activation of AP-1 and NF-κB via mitogen-activated protein (MAP) kinases and IκB kinases, respectively. However, their physiological functions remain to be clarified. To elucidate their roles in vivo, we generated TAB2-deficient mice. The TAB2 deficiency was embryonic lethal due to liver degeneration and apoptosis. This phenotype was similar to that of NF-κB p65-, IKKβ-, and NEMO/IKKγ-deficient mice. However, the IL-1-induced activation of NF-κB and MAP kinases was not impaired in TAB2-deficient embryonic fibroblasts. These findings demonstrate that TAB2 is essential for embryonic development through prevention of liver apoptosis but not for the IL-1 receptor-mediated signaling pathway.

The phase composition of cathodic deposits synthesized from FLINAK-K2TaF7-KBF4 melt has been studied by the X-ray diffraction method. It is shown that boron content in the electrodeposit grows as the cathodic potential is increased from peak R1 to peak R4 in the voltammogram. The scheme of changes in the phase composition depending on the increasing current density is given as follows: {?-Ta + Ta2B}R1 ? {?-Ta + (Ta3B4 TaB)}R2 ? {TaB2}R3 ? {TaB2 + B}R4.

Resumen
ABSTRACT Interleukin-1 (IL-1) receptor-associated kinase (IRAK) plays an important role in the sequential formation and activation of IL-1-induced signaling complexes. Previous studies showed that IRAK is recruited to the IL-1-receptor complex, where it is hyperphosphorylated. We now find that the phosphorylated IRAK in turn recruits TRAF6 to the receptor complex (complex I), which differs from the previous concept that IRAK interacts with TRAF6 after it leaves the receptor. IRAK then brings TRAF6 to TAK1, TAB1, and TAB2, which are preassociated on the membrane before stimulation to form the membrane-associated complex II. The formation of complex II leads to the phosphorylation of TAK1 and TAB2 on the membrane by an unknown kinase, followed by the dissociation of TRAF6-TAK1-TAB1-TAB2 (complex III) from IRAK and consequent translocation of complex III to the cytosol. The formation of complex III and its interaction with additional cytosolic factors lead to the activation of TAK1, resulting in NF-κB and JNK activation. Phosphorylated IRAK remains on the membrane and eventually is ubiquitinated and degraded. Taken together, the new data reveal that IRAK plays a critical role in mediating the association and dissociation of IL-1-induced signaling complexes, functioning as an organizer and transporter in IL-1-dependent signaling.

Abstract: Tabu Search (TS) is a general heuristic procedure for guiding search to obtain good solutions in complex solution spaces. Fundamentally, it is characterized by mechanisms that allow the exploration of the solution space beyond local optimality. These mechanisms are generally implemented by means of parameters which impose restrictions to move selection. Since the calibration of such restrictive elements has a major impact on the algorithm's performance, some implementations use strategies for altering the values of these parameters whenever non-improving search phases are verified. Essentially, these strategies seek to intensify the exploration of promising regions of the solution space, and to abandon the search in regions where improvement possibilities seem to be minimal. The main purpose ofthis work is to propose a new adaptive Tabu metaheuristic approach. The HTA approach, considered here, assumes that the alteration of the parameters values should be defined first by identifying specific pattems in the search trajectory recently described. These pattems provide indications of the solution space topology. For each pattem, perturbation on the values of selected tabu parameters is applied, as means to adapt the search to the different conditions found. The HTA approach was developed from extensive experiments with the Symmetric and Euclidean Traveling Salesman Problem (TSP). Tests involved 12 benchmark instances and verified improvements with respect to a non-adaptive implementation, even under stressing operational conditions provided by free-Junning parameters. HTA was also applied to the Vehicle Routing Problem (VRP). We present the results obtained for 14 benchmark instances characterized by different restrictions. Our adaptive implementation is compared to three highly competitive algorithms. The results indicate that HTA is able to provide solution quality levels comparable to the other algorithms. We also present the results provided by an HTA implementation for the Capacitated Clustering Problem (CCP). They also suggest that introducing adaptive mechanisms based on the pattems of search trajectories is a robust and promising strategy

Abstract: The classical resource constrained project scheduling problem consists of activities with fixed duration and subject to precedence relations, which compete for limited resources. This problem involves the determination of the starting time of the activities in order to minimize the completion time of the project. Several extensions of the classical problem have been addressed in the literature to model real problems. One of these extensions is the problem tackled in this work, named generalized resource constrained project scheduling problem. The generalized variant considers two extensions in relation to the classical problem. The first one involves the representation of precedence relations among activities, by means of a minimum time interval between two activities, which is important in the make-to-order production setting, when a machine setup time is incurred between the processing of two activities. The second extension considers that the resource availability is time varying, which represents the situation where only a subset of the total manpower and machines is available due to vacation or maintenance. Due to the high combinatorial complexity of this problem, heuristic methods are used to solve real size problems. Algorithms that combine concepts from tabu search and reactive tabu search are proposed and computationally implemented. Their performance is tested against a reactive tabu search algorithm from the literature

Resumen
Aircraft encounter increased aerodynamic loads when exposed to gusts, turbulence andmanoeuvres. Currently, these loads are mitigated through the use of ailerons and spoilers to reduce lift, in turn reducing the loads passed to the aircraft structure. However, these actuators are limited in their frequency response and cannot control loads produced by higher frequency events. Therefore, an actuator which can mitigate high frequency oscillatory loads is required, with a deployment reduced frequency, k of up to 1. One such promising load control actuator is the minitab, consisting of a small span-wise strips, similar to the Gurney flap, deployed normal to the airfoil upper surface. Key to the actuator’s high frequency response is its low inertia, meaning that a small energy input can achieve a significant effect. To investigate the efficacy of the minitab on load alleviation a series of steady state, periodic and transient measurements were conducted at a Reynolds number of 6.6 x 105. These experiments aimed to fully evaluate the effect of chordwise location, mini-tab height and angle of attack on steady state load control. The dynamic response was categorised, in terms of magnitude, phase and time delay by the periodic and transient measurements. Mini-tabs of height h/c = 0.02 and 0.04 were employed in a steady state configurationacross a range of chordwise locations to investigate the effects of mini-tab height and chordwise position. Overall, the mini-tab was found to have a lift reducing effect which increased with height. It was found that the effect of the chordwise location was highly dependent on the angle of attack. Placement close to the trailing edge induced a large effect at α = 0°, creating an effective change in camber comparable to conventional Gurney flap use. Peak suction over the lower surface increased resulting in a reduction of ΔCL = -0.48. Approaching stall, effectiveness decreased as the mini-tab became immersed in the separated flow. Placement at xf/c = 0.60 produced an almost constant lift reduction between α = 0° and 5° of ΔCL ≈ -0.60, with a gradual reduction to stall. A mini-tab positioned close to the leading edge (xf/c = 0.08) was found to separate the flow effectively at low incidences but with no noticeable change in lift observed. It was found that the flow separation produced by the mini-tab effectively eliminated the suction peak on the upper surface. However, placement close to the leading edge has increasing effectiveness towards stall, as the shear layer induced by the separation was displaced further from airfoil surface. Peak lift reduction at stall was found to be ΔCL ≈ -0.67. The optimum chordwise location for peak lift reduction is dependent on the airfoil angle of attack: the position of the mini-tab for maximum lift reduction moves towards the leading edge as the angle of attack increases. The second stage utilised a deployable mini-tab up to reduced frequencies, k = 0.79, placedat xf/c = 0.85, to assess the mini-tab’s frequency response. The force measurements indicate that the mini-tab has a decreasing effect on lift reduction with increasing actuation frequency. This trend is comparable to Theodorsen’s function, based on the change in circulation. For α = 0°, the normalised peak-to-peak lift reduction decreased from 1 for steady state deployment to around 0.6 at k = 0.79. In addition, a phase lag exists between the mini-tab deployment and the aerodynamic response which increased with actuation reduced frequency, k. However, the measured phase lag is substantially larger than Theodorsen’s prediction. Increasing the angle of attack, α reduced the mini-tab’s effect on lift while increasing the phase angle when comparing equal k values. Particle Image Velocimetry measurements indicate that the delay and reduction in effectiveness of periodic deployment is due to the presence and growth of the separated region behind the mini-tab. Overall, the mini-tab was found to be an effective, dynamic lift reduction device with the separated region behind the mini-tab key to the amplitude and phase delay of lift response. Finally, the aerodynamic response of the mini-tab was investigated during a transientdeployment. The delay in aerodynamic response to mini-tab actuation was consistent with literature. The normalised deployment period, τdeploy did not provide a significant alteration in the aerodynamic response for deployment periods below τdeploy = 3, with the aerodynamic response reaching the steady state value around τ = 6-8. The aerodynamic response of the mini-tab was approximated using a simple, 1st order system response to a ramp-step input of gradient 1/τdeploy, indicating that the aerodynamic response of the mini-tab is further delayed for higher angles of attack, due to the presence of separated flow in the vicinity of the mini-tab. PIV measurements were utilised to analyse the effect of transient mini-tab deployment, indicating a delay in the development of the separation region created by the mini-tab, producing a corresponding delay in aerodynamic response. In addition, outward deployment was found to have a slower aerodynamic response than inward deployment, as the flow was found to take to detach slower than to reattach.

The search of better layouts is a combinatorial optimisation problem that arises in a wide variety of spacial planning contexts, in looking for a Layout of departments industry. Since no polinomial-time algorithm for attainning a global optimal solution is known some heuristics techniques are used. We study this subject, for a given problem, performing the following steps: 1) We generate an initial solution by using a clustering procedure; 2) We generate another initial solution, rearranging them randomly; 3) We apply the tabu search to the two initial solutions generated above; 4) We compare the results obtained in the step 3. The procedure is applied to four cases, namely, those with 12, 15, 20 and 30 facilities. In the space assigned to facilities there were occupied areas and the facilities cannot be used there, so we increased the difficulty of the problem. When we compared the initial layouts obtained in step 1 with the obtained in step 2, we conclued that step 1 obtained better layouts. From step 4, we conclued that the tabu search obtained sligthly better final layouts when applied to initial solutions generated in step 1 than those obtained starting from solutions generated in step 2. So we conclude that tabu search is an efficient search procedure independent of the quality of the initial solution.

Resumen
Passive augmentation devices are typically added to diffusers to increase pressure recovery; however, applications such as infrared suppression exist where more uniform outlet velocity profiles are also desirable. Square tabs were added to a short annular diffuser system with diffuser effectiveness of 80% in strong swirling subsonic turbulent flow. The number, axial position, width, and height were varied for the tabs with a 135° orientation angle. Results showed that back pressure increased linearly with increasing tab projected area blockage but outlet velocity uniformity was maximum with 7% tab blockage. Tabs placed on the centre body base recovered more dynamic pressure than tabs placed downstream on the centre body with similar tab area blockage.

Resumen
The results of surface pressure measurements are presented in this paper to gain further insight into the lift changing influence of finite width micro-tabs, especially in adjacent airfoil sections. Micro-tabs are a promising concept for load control on wind turbines. Local pressure distributions were measured in several rows of pressure taps in the vicinity of the finite width micro-tab attached to a FX 63-137 profile at low Reynolds numbers. The investigation focuses on length dependency, chordwise position, and interaction between two micro-tabs. Additionally, stereo Particle-Image-Velocimetry measurements were conducted to study the structure, sense of rotation, and influence of tab-induced tip vortices, as well as the impact of a finite width micro-tab on the model’s near wake. Experiments reveal relative changes of more than 30 % in the pressure coefficient distribution upstream of several micro-tab configurations. Furthermore, increments of 20 % are recorded in neighbouring sections not directly controlled by micro-tabs. Even higher changes are obtained in the region between two tabs. These improvements are attained due to local and global changes in the effective camber.

Resumen
Abstract The effect of discrete delta-shaped tabs on the film cooling performance from a row of cylindrical holes is investigated. The holes are inclined at 35° along the streamwise direction. Three tab locations are investigated: (1) tabs placed along the upstream edge of the hole; (b) tab placed along the downstream edge of the hole; and (3) tabs placed along the lateral edges of the hole. Measurements were carried out in a low-speed wind tunnel using the transient liquid crystal technique. The mainstream velocity and free-stream turbulence intensity in the low speed wind tunnel are 8.5 m/s and 6% respectively and the Reynolds number based on hole diameter is 6375. Three blowing ratios of 0.56, 1.13, and 1.7 are tested. Results show that the tab placed upstream of the hole covering the 33% of the hole exit provides the highest film effectiveness compared to other tab locations studied. This is because the jet lift-off in the presence of the tabs is reduced and there is greater lateral spreading of the jet.

Resumen
This paper describes an experimental investigation into the effects of geometrical variations on ejector system performance when the driving nozzle includes delta mixing tabs. Mixing tabs have been shown to provide good mixing performance with comparable back-pressure penalties to other types of enhanced mixing nozzles. The performance parameters of most interest are pumping, mixing, and back-pressure. Geometric parameters studied include standoff distance, mixing-tube diameter, and tab angle. Experimental testing showed significant performance improvements in mixing and pumping with a decrease in tab angle. Maximum mixing was found to occur with tab angles positioned at 120°. Exceptional mixing was also observed with increased standoff. Back-pressure was shown to increase with increasing standoff and decreasing tab angle, but was not affected by mixing-tube diameter. In addition, a zone of recirculation was identified at the entrance to the mixing-tube. This zone is thought to have an influence on ejector performance.

Resumen
Micro-electro-mechanical (MEM) translational tabs are introduced for active lift control on aircraft. These tabs are mounted near the trailing edge of lifting surfaces such as aircraft wings and tails, deploy approximately normal to the surface, and have a maximum deployment height on the order of one percent of the section chord. Deployment of the tab effectively changes the sectional camber, thereby changing the aerodynamic characteristics of a lifting surface. Tabs with said deployment height generate a change in the section lift coefficient of approximately ±0.3. The microtab design and the techniques used to fabricate and test the tabs are presented.

Resumen
A uniaxial tensile test is a useful method for determination of material properties, especially longitudinal tensile strength. To accurately derive the longitudinal tensile strength, it is desired that the specimen fails in in the gauge section defined here as ‘working zone’. Unidirectional (UD) composites require use of end tabs during this tensile testing to avoid damage to the specimen due to grip serrations. The grip pressure, along with sudden geometry change at the edge of end tabs leads to longitudinal stress concentrations. The conventionally used rectangular and tapered end tabs suffer from these longitudinal stress concentrations under the edge of end tabs, causing premature failure of specimen outside of the working zone. In the present paper, a simulation study is performed for comparison of conventional end tabs with hybrid specimen geometry [1] and a novel arrow-shaped end tab geometry to determine the effect of end tab geometry on longitudinal stress concentrations. The study is focused on high modulus carbon fibre HS40/epoxy UD (0°) composite. The numerical model replicates the actual set-up for uniaxial tensile testing, including contact interactions between testing machine components. The simulation results are used to further optimise the geometry and provide recommendations to eliminate or minimise longitudinal stress concentrations.

Resumen
Wind turbines generally suffer from unsteady inflow caused by yaw misalignment, gusts, and turbulence which induce fatigue loads. Spanwise distributed active micro-tabs at the mid and outer blade regions are able to countervail these unsteady loads. However, during the actuation process of these devices, transient effects play an important role. This work aims to give a deeper insight in the process of the tab deployment and retraction to evaluate the effectiveness of active micro-tabs for load control on wind turbines. Wind tunnel experiments on a two-dimensional NACA 0018 airfoil with an active micro-tab were conducted. The tab deployment- and retraction time was varied for an application on the suction or the pressure side of the airfoil. Time resolved surface pressure measurements were performed at Reynolds numbers of Re = 7 · 105 and 1 · 106. Transient responses showed a significant delay and post deployment behavior of the lift which strongly depend on the actuation time.
