Texto completoPeroxynitrite is the product of the very fast reaction between nitrogen oxide and superoxide. This work tackles several sides of the chemistry of these compounds. Several SIN-1 derivatives with NO releasing properties were synthesised and studied to reduce peroxynitrite production and thus increase the NO availability. Iron Superoxide Dismutases (SOD) are enzymes that protect protozoans from the oxidative stress. Enzymes from parasites Plasmodium falciparum, Trypanosoma cruzi and Trypanosoma brucei were overexpressed and characterised in order to find some new compounds with antiparasitic activity. Iron SODs are partially inactivated by nitration of the residue tyrosine 34 by peroxynitrite. The structure of the nitrated enzyme was studied and modelised. Several methods of “de novo” conception of inhibitors are descried and the binding modes of these compounds were studied by docking experiments
