Cell senescence is one of the most important processes determining cell fate and is involved in many pathophysiological conditions, including cancer, neurodegenerative diseases, and other aging-associated diseases. It has recently been discovered that the E3 ubiquitin ligase STIP1 homology and U-box–containing protein 1 (STUB1 or CHIP) is up-regulated during the senescence of human fibroblasts and modulates cell senescence. However, the molecular mechanism underlying STUB1-controlled senescence is not clear. Here, using affinity purification and MS-based analysis, we discovered that STUB1 binds to brain and muscle ARNT-like 1 (BMAL1, also called aryl hydrocarbon receptor nuclear translocator–like protein 1 (ARNTL)). Through biochemical experiments, we confirmed the STUB1-BMAL1 interaction, identified their interaction domains, and revealed that STUB1 overexpression down-regulates BMAL1 protein levels through STUB1's enzymatic activity and that STUB1 knockdown increases BMAL1 levels. Further experiments disclosed that STUB1 enhances BMAL1 degradation, which is abolished upon proteasome inhibition. Moreover, we found that STUB1 promotes the formation of Lys-48–linked polyubiquitin chains on BMAL1, facilitating its proteasomal degradation. Interestingly, we also discovered that oxidative stress promotes STUB1 nuclear translocation and enhances its co-localization with BMAL1. STUB1 expression attenuates hydrogen peroxide–induced cell senescence, indicated by a reduced signal in senescence-associated β-gal staining and decreased protein levels of two cell senescence markers, p53 and p21. BMAL1 knockdown diminishes this effect, and BMAL1 overexpression abolishes STUB1's effect on cell senescence. In summary, the results of our work reveal that the E3 ubiquitin ligase STUB1 ubiquitinates and degrades its substrate BMAL1 and thereby alleviates hydrogen peroxide–induced cell senescence.

ObjectiveTo identify the genetic cause of autosomal dominant ataxia complicated by behavioral abnormalities, cognitive decline, and autism in 2 families and to characterize brain neuropathologic signatures of dominant STUB1-related ataxia and investigate the effects of pathogenic variants on STUB1 localization.MethodsClinical and research-based exome sequencing was used to identify the causative variants for autosomal dominant ataxia in 2 families. Gross and microscopic neuropathologic evaluations were performed on the brains of 4 affected individuals in these families.ResultsMutations in STUB1 have been primarily associated with childhood-onset autosomal recessive ataxia, but here we report heterozygous missense variants in STUB1 (p.Ile53Thr and p.The37Leu) confirming the recent reports of autosomal dominant inheritance. Cerebellar atrophy on imaging and cognitive deficits often preceded ataxia. Unique neuropathologic examination of the 4 brains showed the marked loss of Purkinje cells (PCs) without microscopic evidence of significant pathology outside the cerebellum. The normal pattern of polarized somatodendritic STUB1 protein expression in PCs was lost, resulting in aberrant STUB1 localization in the distal PC dendritic arbors.ConclusionsThis study confirms a dominant inheritance pattern in STUB1-ataxia in addition to a recessive one and documents its association with cognitive and behavioral disability, including autism. In the most extensive analysis of cerebellar pathology in this disease, we demonstrate disruption of STUB1 protein in PCs as part of the underlying pathogenesis.

Variants in STUB1 cause both autosomal recessive (SCAR16) and dominant (SCA48) spinocerebellar ataxia. Reports from 18 STUB1 variants causing SCA48 show that the clinical picture includes later-onset ataxia with a cerebellar cognitive affective syndrome and varying clinical overlap with SCAR16. However, little is known about the molecular properties of dominant STUB1 variants. Here, we describe three SCA48 families with novel, dominantly inherited STUB1 variants (p.Arg51_Ile53delinsProAla, p.Lys143_Trp147del, and p.Gly249Val). All the patients developed symptoms from 30 years of age or later, all had cerebellar atrophy, and 4 had cognitive/psychiatric phenotypes. Investigation of the structural and functional consequences of the recombinant C-terminus of HSC70-interacting protein (CHIP) variants was performed in vitro using ubiquitin ligase activity assay, circular dichroism assay and native polyacrylamide gel electrophoresis. These studies revealed that dominantly and recessively inherited STUB1 variants showed similar biochemical defects, including impaired ubiquitin ligase activity and altered oligomerization properties of the CHIP. Our findings expand the molecular understanding of SCA48 but also mean that assumptions concerning unaffected carriers of recessive STUB1 variants in SCAR16 families must be re-evaluated. More investigations are needed to verify the disease status of SCAR16 heterozygotes and elucidate the molecular relationship between SCA48 and SCAR16 diseases.

ObjectiveTo describe a new spinocerebellar ataxia (SCA48) characterized by early cerebellar cognitive-affective syndrome (CCAS) and late-onset SCA.MethodsThis is a descriptive study of a family that has been followed for more than a decade with periodic neurologic and neuropsychological examinations, MRI, brain SPECT perfusion, and genetic analysis. Whole exome sequencing was performed in 3 affected and 1 unaffected family member and subsequently validated by linkage analysis of chromosome 16p13.3.ResultsSix patients fully developed cognitive-affective and complete motor cerebellar syndrome associated with vermian and hemispheric cerebellar atrophy, suggesting a continuum from a dysexecutive syndrome slowly evolving to a complete and severe CCAS with late truncal ataxia. Three presymptomatic patients showed focal cerebellar atrophy in the vermian, paravermian, and the medial part of cerebellar lobes VI and VII, suggesting that cerebellar atrophy preceded the ataxia, and that the neurodegeneration begins in cerebellar areas related to cognition and emotion, spreading later to the whole cerebellum. Among the candidate variants, only the frameshift heterozygous c.823_824delCT STUB1 (p.L275Dfs*16) pathogenic variant cosegregated with the disease. The p.L275Dfs*16 heterozygous STUB1 pathogenic variant leads to neurodegeneration and atrophy in cognition- and emotion-related cerebellar areas and reinforces the importance of STUB1 in maintaining cognitive cerebellar function.ConclusionsWe report a heterozygous STUB1 pathogenic genetic variant causing dominant cerebellar ataxia. Since recessive mutations in STUB1 gene have been previously associated with SCAR16, these findings suggest a previously undescribed SCA locus (SCA48; MIM# 618093).

The hepatitis B virus (HBV) core protein (HBc) functions in multiple steps of the viral life cycle. Heteroaryldihydropyrimidine compounds (HAPs) such as Bay41-4109 are capsid protein allosteric modulators that accelerate HBc degradation and inhibit the virion secretion of HBV, specifically by misleading HBc assembly into aberrant non-capsid polymers. However, the subsequent cellular fates of these HAP-induced aberrant non-capsid polymers are not well understood. Here, we discovered that that the chaperone-binding E3 ubiquitin ligase protein STUB1 is required for the removal of Bay41-4109-induced aberrant non-capsid polymers from HepAD38 cells. Specifically, STUB1 recruits BAG3 to transport Bay41-4109-induced aberrant non-capsid polymers to the perinuclear region of cells, thereby initiating p62-mediated macroautophagy and lysosomal degradation. We also demonstrate that elevating the STUB1 level enhances the inhibitory effect of Bay41-4109 on the production of HBeAg and HBV virions in HepAD38 cells, in HBV-infected HepG2-NTCP cells, and in HBV transgenic mice. STUB1 overexpression also facilitates the inhibition of Bay41-4109 on the cccDNA formation in de novo infection of HBV. Understanding these molecular details paves the way for applying HAPs as a potentially curative regimen (or a component of a combination treatment) for eradicating HBV from hepatocytes of chronic infection patients.

ObjectiveTo describe the clinical and pathologic features of a novel pedigree with heterozygous STUB1 mutation causing SCA48.MethodsWe report a large pedigree of Dutch decent. Clinical and pathologic data were reviewed, and genetic analyses (whole-exome sequencing, whole-genome sequencing, and linkage analysis) were performed on multiple family members.ResultsPatients presented with adult-onset gait disturbance (ataxia or parkinsonism), combined with prominent cognitive decline and behavioral changes. Whole-exome sequencing identified a novel heterozygous frameshift variant c.731_732delGC (p.C244Yfs*24) in STUB1 segregating with the disease. This variant was present in a linkage peak on chromosome 16p13.3. Neuropathologic examination of 3 cases revealed a consistent pattern of ubiquitin/p62-positive neuronal inclusions in the cerebellum, neocortex, and brainstem. In addition, tau pathology was present in 1 case.ConclusionsThis study confirms previous findings of heterozygous STUB1 mutations as the cause of SCA48 and highlights its prominent cognitive involvement, besides cerebellar ataxia and movement disorders as cardinal features. The presence of intranuclear inclusions is a pathologic hallmark of the disease. Future studies will provide more insight into its pathologic heterogeneity.

Spinocerebellar ataxia type 48 (SCA48) is an autosomal dominantly inherited disease characterized by gait and limb ataxia, cerebellar dysarthria, cognitive impairment, psychiatric abnormalities and variable types of movement disorders. To date, more than 30 STUB1 gene (NM_005861.4) mutations have been described in the genetic background of SCA48. The aim of this short report was to demonstrate the first Hungarian SCA48 patient caused by a novel STUB1 missense mutation (c.788G>C, p.Arg263Pro). The characteristics of detailed neurological phenotype, brain MRI and genetic assessment are presented and compared to previously published cases. The most important neurological findings of the patient were gait ataxia, dysarthria, cognitive decline and psychiatric problems including depression, anxiety and mild impulsivity. The brain MRI demonstrated cerebellar atrophy with posterolateral predominance and frontal lobe cortical atrophy. Clinical exome sequencing examination identified the above-mentioned missense variant located in the significant ubiquitinase domain of the CHIP protein. In this paper the first Hungarian SCA48 patient was described with characteristic neuropsychiatric signs and brain MRI abnormalities, due to a novel STUB1 gene missense mutation.

ABSTRACTC-terminus of HSC70-interacting protein (CHIP) encoded by the gene STUB1 is a co-chaperone and E3 ligase that acts as a key regulator of cellular protein homeostasis. Mutations in STUB1 cause autosomal recessive spinocerebellar ataxia type 16 (SCAR16) with widespread neurodegeneration manifesting as spastic-ataxic gait disorder, dementia and epilepsy. CHIP−/− mice display severe cerebellar atrophy, show high perinatal lethality and impaired heat stress tolerance. To decipher the pathomechanism underlying SCAR16, we investigated the heat shock response (HSR) in primary fibroblasts of three SCAR16 patients. We found impaired HSR induction and recovery compared to healthy controls. HSPA1A/B transcript levels (coding for HSP70) were reduced upon heat shock but HSP70 remained higher upon recovery in patient- compared to control-fibroblasts. As SCAR16 primarily affects the central nervous system we next investigated the HSR in cortical neurons (CNs) derived from induced pluripotent stem cells of SCAR16 patients. We found CNs of patients and controls to be surprisingly resistant to heat stress with high basal levels of HSP70 compared to fibroblasts. Although heat stress resulted in strong transcript level increases of many HSPs, this did not translate into higher HSP70 protein levels upon heat shock, independent of STUB1 mutations. Furthermore, STUB1(−/−) neurons generated by CRISPR/Cas9-mediated genome editing from an isogenic healthy control line showed a similar HSR to patients. Proteomic analysis of CNs showed dysfunctional protein (re)folding and higher basal oxidative stress levels in patients. Our results question the role of impaired HSR in SCAR16 neuropathology and highlight the need for careful selection of proper cell types for modeling human diseases.

Resumo: A proteína prion celular (PrPC) e uma glicoproteina extracelular, ancorada na membrana plasmática por uma molécula de glicofosfatidilinositol (GPI). A conversão de PrPC em uma isoforma patologica (PrPSc), a partir de modificações estruturais, e responsável pelo desenvolvimento das encefalopatias espongiformes transmissíveis (TSEs) ou doenças periódicas. Vários trabalhos indicam que formas citosólicas de PrP podem ser formadas durante erros na sua biossintese. Os príons citosólicos (CytPrP) são gerados por meio de duas vias: ERAD (ER-associated Degradation) e ineficiência do peptídeo sinal. Alguns estudos demonstram que estas formas de príons citosólicos exercem função neurotoxica, contribuindo para a manifestação das doenças periódicas. Trabalhos anteriores do nosso grupo, foram capazes de identificar uma interação entre PrPC e CHIP/Stub1. CHIP/Stub1 e uma proteína citoplasmática o qual funciona como um co-chaperona molecular e apresenta atividade ubiquitina E3- ligase, participando do controle de qualidade das proteínas desde o dobramento ate a fase de degradação. Ainda, CHIP/Stub1 e homologa a proteína STI1/Hop, uma cochaperona e um ligante bem estabelecido de PrPC. O objetivo desse trabalho foi avaliar em diferentes linhagens celulares se o efeito neurotoxico de PrP citosólico, ja descrito para algumas células, era capaz de ser modulado através dessas co-chaperonas. Em nossos resultados mostramos por diferentes ensaios de viabilidade celular, tais como MTT, vermelho neutro, que CytPrP reduziu de 30-50% a viabilidade celular de três linhagens neuronais testadas: N2a, SH-SY5Y e CF10. Esse mesmo efeito nao foi observado para linhagem renal HEK293T. A expressão das co-chaperonas CHIP e STI-1 foi capaz de reverter a toxicidade do CytPrP em todas linhagens neuronais. Nossos resultados ainda mostraram que mutantes de CHIP e STI-1 com domínios deletados também foram capazes de reverter o efeito neurotoxico causado pela CytPrP. Ensaios utilizando o inibidor MG132 e hidroxicloroquina sugerem que essa reversao do efeito neurotoxico de CytPrP por CHIP e STI-1 parece não ser dependente da via de degradação por proteossomo, nem por lisossoma. Assim, um mecanismo através do qual as co-chaperonas exercem seu efeito de proteção da neuroxicidade ainda esta sendo investigado. Os dados obtidos nesse trabalho poderão contribuir para elucidar os possíveis mecanismos da neurotoxicidade de prions, permitindo um auxilio futuro no desenvolvimento de alvos terapêuticos relacionadas a doenças prionicas. Abstract: The cellular prion protein (PrPC) is an extracellular glycoprotein, anchored to the plasma membrane by a glycophosphatidylinositol (GPI) molecule. The conversion of PrPC to a pathological isoform (PrPSc), through structural modifications, is responsible for the development of transmissible spongiform encephalopathies (TSEs) or prion diseases. Several data indicate that cytosolic forms of PrP can be formed during errors in its biosynthesis. Cytosolic prions (CytPrP) are generated by two pathways: ERAD (ER-associated Degradation) and signal peptide inefficiency. Some studies have shown that these forms of cytosolic prions exert neurotoxic function, contributing to the manifestation of prion diseases. Previous work by our group was able to identify an interaction between PrPC and CHIP/Stub1. CHIP/Stub1 is a cytoplasmic protein, identified as a co- chaperone with E3 ligase activity, participating in the quality control of proteins, from folding to the degradation phase. Still, CHIP/Stub1 is homologous to STI1 (stress - inducible protein 1 ), a co-chaperone and a well-established PrPC bind protein. The objective of this work was to evaluate in different cell lines whether the neurotoxic effect of cytosolic PrP, already described for some cells, was able to be modulated through these co-chaperones. In our results we showed by different cell viability assays, such as MTT, neutral red, that CytPrP reduced from 30% to 50% the cell viability of three tested neuronal lines: N2a, SH-SY5Y and CF10. This same effect was not observed for HEK293T renal lineage. Our results also show that CHIP and STI-1 mutants with deleted domains were also capable of reversing the neurotoxic effect caused by CytPrP in all neuronal lines. Assays using the proteasome inhibitor MG132 and Hydroxychloroquine suggest that this reversal of the neurotoxic effect does not appear to be dependent of the proteosome or lysosomal degradation pathways. Thus, a mechanism through which co-chaperones exert their protective effect of CytPrP neuroxicity is still being investigated. The data obtained in this work may contribute to elucidate possible mechanisms of neurotoxicity of prions, allowing future aid in the development of therapeutic targets related to prion diseases. Esta dissertação apresenta o projeto e implementação de um sistema gerador de sfubs, para utilização no ambiente computacional do Laboratório de Sistemas Digitais do ICMSC/USP, em plataformas DOS. O gerador de sfubs é uma ferramenta de auxílio ao desenvolvimento de aplicações distribuídas e tem como função criar automaticamente os procedimentos sfubs, a partir da definição dos serviços que serão executados remotamente. Esse software de apoio libera os programadores da implementação de rotinas que envolvam os protocolos básicos de comunicação. Os procedimentos sfubs são responsáveis pela comunicação entre os processos clientes e os serviços oferecidos. Com esses procedimentos, os processos cliente e servidor podem ser compilados e executados separadamente, em máquinas diferentes. Dentre as vantagens do sistema gerador de sfubs, a mais importante, e que está bem caracterizada neste trabalho, é o ganho de produtividade nos projetos de aplicações distribuídas, tornando extremamente atrativa sua adoção nesses projetos. O sistema proposto está implementado na linguagem \"C\" e pode ser utilizado facilmente para a geração de aplicações distribuídas envolvendo equipamentos compatíveis com a linha IBM-PC, executando DOS versão 3 ou superior.
This dissertation presents the project and implementation of a stub generator system, to be used in the Digital Systems Laboratory at ICMSC/USP, running on DOS platforms. The stub generator is an auxiliary tool for the development of distributed applications and has the responsability of creating the stub procedures automatically, based on the definitíon of the services that will be executed remotely. The auxiliary software releases the programmers from the routine implementation tasks that involve the basic communication protocol. The stub procedures are responsible for the communication between the clients processes and the available services. With these procedures, the client and the server can be both compiled and executed separately, on diferent machines. The most attractive advantage in adopting the stub generator described in this work is the gain of productivity observed in distributed applications designs. The proposed system is implemented in the \"C\" language and can be easily used for the generation of distributed applications involving IBM-PC compatible platforms, executing the DOS version 3 or higher.

Resumen
This thesis proposes a new methodology to extend the software performance engineering process. Common performance measurement and tuning principles mainly target to improve the software function itself. Hereby, the application source code is studied and improved independently of the overall system performance behavior. Moreover, the optimization of the software function has to be done without an estimation of the expected optimization gain. This often leads to an under- or overoptimization, and hence, does not utilize the system sufficiently. The proposed performance improvement methodology and framework, called dynamic performance stubs, improves the before mentioned insufficiencies by evaluating the overall system performance improvement. This is achieved by simulating the performance behavior of the original software functionality depending on an adjustable optimization level prior to the real optimization. So, it enables the software performance analyst to determine the systems’ overall performance behavior considering possible outcomes of different improvement approaches. Moreover, by using the dynamic performance stubs methodology, a cost-benefit analysis of different optimizations regarding the performance behavior can be done. The approach of the dynamic performance stubs is to replace the software bottleneck by a stub. This stub combines the simulation of the software functionality with the possibility to adjust the performance behavior depending on one or more different performance aspects of the replaced software function. A general methodology for using dynamic performance stubs as well as several methodologies for simulating different performance aspects is discussed. Finally, several case studies to show the application and usability of the dynamic performance stubs approach are presented.

Resumen
Dans les systèmes actuels de communication, qu’ils soient terrestre ou spatial, qu’ils soient mobile ou fixe, il y a un réel intérêt à développer des front-ends radiofréquences et hyperfréquences miniatures et performants. Ceci s’applique en particulier aux dispositifs de filtrage où l’encombrement et les facteurs de qualité sont clairement antagonistes. Pour les bandes de fréquences basses aux alentours du GHz, les longueurs d’onde restent encore importantes, rendant difficiles les efforts de miniaturisation. D’autre part il faut aussi s’assurer que ces filtres viendront s’interconnecter aisément avec les autres composants du système, notamment les actifs.Pour toutes ces raisons, le développement de structures de filtres multicouches utilisant des substrats à haute permittivité (εr = 68) selon une approche LTCC apparait comme une alternative intéressante. Elle peut en effet conduire à une réduction significative de l'empreinte (footprint) sans pour autant trop nuire aux performances électriques.Dans le cadre de ce travail, deux structures de filtres multicouches ont été développées pour répondre à des spécifications proposées en bandes L et S, par un équipementier du spatial. Ces filtres ont pour caractéristiques principales un haut niveau de rejection et des faibles pertes dans la bande passante. Pour atteindre les spécifications, un filtre SIW empilé verticalement et un filtre à stubs en court-circuit en configuration triplaque ont été étudiés. Le filtre SIW se distingue par un facteur de qualité élevé, ce qui entraîne des faibles pertes d’insertion et une bonne platitude. La solution à stub permet quant à elle de réduire l’encombrement mais au prix d’un impact sur les performances électriques. Dans les deux cas on tire parti de la souplesse offerte par la technologie LTCC, puisqu’elle offre finalement un degré de liberté supplémentaire, par rapport à une approche planaire classique. Si dans le cas SIW, c’est surtout l’architecture topologique qui a été étudiée finement pour pouvoir agencer et coupler douze cavités, dans le cas du filtre à stub une synthèse mettant à profit tous les degrés de liberté offerts a été spécifiquement développée.Compte tenu de la complexité des filtres, notamment à cause de l’ordre élevé et de la mise en oeuvre de murs « électriques » à partir d’arrangements de via spécifiques, une attention particulière doit être apportée lors des phases de simulation et d’optimisation. De plus la très forte permittivité du substrat ne permet pas d’utiliser de ligne 50 Ohms. Enfin les transitions constituent un point dur de l’exercice surtout dans le cas SIW.Cette thèse co-financée par le CNES (Centre National d'Etudes Spatiales) et Thales Alenia Space, était accompagnée par un projet R&T financé par le CNES. In current communication systems, whether terrestrial or spatial, whether fixed or mobile, there is a real interest in developing high performance miniature RF front-ends. This is applied in particular to filter devices, in which the size and the quality factors are clearly in conflict. For low frequency bands around the GHz, the wavelengths remain significant, making it difficult the miniaturization efforts. On the other hand, we must also ensure that these filters will be easily interconnected with other other system components, including active devices.For all these reasons, the development of multilayer filter structures using high permittivity substrates (Er = 68) in an LTCC approach is consolidated as an interesting alternative. It may lead to a significant footprint reduction without decreasing the electrical performances.As part of this work, two multilayer filter structures have been developed to meet the given specifications in L and S bands, given by a space manufacturer. These filters have as main features a high rejection level and low losses in the passband. To meet the specifications, a vertically stacked SIW filter and a short-circuited stubs filter in a stripline configuration were studied. The SIW filter is characterized by a high quality factor, which results in low insertion loss and good flatness. The stubs filter allows in contrast to reduce the footprint but at the price of impacting the electrical performance. In both cases we take advantage of the flexibility offered by the LTCC technology as it finally provides an additional freedom degree compared to a conventional planar approach. For the SIW filter, the topological architecture was studied and designed in detail, to be able to arrange and synthetize couplings between twelve cavities. In a similar way, for the stub filter a synthesis that takes profit of all the offered freedom degrees was developed.Given the filters complexity, especially due to the high order and the implementation of “electrical walls" based on specific vias patterns, a close attention must be paid during the simulation and optimization phase. In addition, the high permittivity substrate does not allow to conceive 50-Ohms lines. Finally, access transitions constitute a challenging task, especially for the SIW case.This thesis was co-funded by CNES (Centre National d'Etudes Spatiales) and Thales Alenia Space, and was accompanied by an R&T project funded by CNES. Este estudo tem por objetivo avaliar o comportamento estrutural de um sistema de vigamento de piso tipo stub-girder, modificado para melhor aproveitamento dos perfis em sua fabricação. O estudo é baseado em formulações não lineares a partir do método dos elementos finitos (MEF), com uso do software ANSYS. Através do software FTOOL foi criado um modelo simplificado para a análise estrutural e o cálculo das resistências nominais foi feito com base na norma canadense CAN-S16-10 da Canadian Standards Association. Além disso, foram avaliadas as cargas de máximas e de serviço do sistema, bem como os deslocamentos associados a estes carregamentos. Com base nos resultados obtidos, foram feitas recomendações de projeto do sistema e foram avaliadas as condições de economia de material e possibilidade de utilização na prática de construção de edificações de andares múltiplos. Com vistas à fase de construção, é feita a discussão sobre a possibilidade de obtenção de um método construtivo industrializado, já que o sistema de fabricação é caracterizado pela rapidez de produção e montagem, que são facilitadas pelo fato deste sistema permitir a reutilização de pequenos comprimentos de perfis que sobram no processo normal de fabricação.
This study s purpose was to evaluate the structural behavior of a stub-girder floor-system, which was modified to better utilize the steel profiles in its manufacturing process. The study is based on nonlinear analysis through the use of the finit element method (FEM), employing the software ANSYS. For the structural analysis, a simplified model was created through the use of the software FTOOL and the nominal resistances was obtained based on the canadian CAN-S16-10 Standard, by the Canadian Standards Association. Furthermore, the working and tensile loads were evaluated, as were the displacements associated to them. Based on the results, designs recommendations were made and the conditions of economy of material, as well the possibility of the system s practical use in multi-storey buildings were discussed. With regards to the construction phase, a discussion was made on the possibility of obtaining an industrialized construction method, since the manufacturing process is characterized by the fast production and assembly, which are facilitade by the fact that this system allows the reuse of small lenghts of remaining profiles from the normal manufacturing process.

Resumen

Testdriven utveckling samt användning av enhetstestning är ett växande område. Vid användning av dessa tekniker är det önskvärt att kunna använda stubb- och mockobjekt för att ersätta en DOC(Dependent on Component).

I dagsläget finns det ett verktyg hos Enea som genererar stubbar för C++-källkod. Examensarbetet består av att vidareutveckla detta verktyg i flera avseenden. Stöd för C-kod adderas till verktyget. För C-kod adderas även möjligheten att generera stubb- och mockobjekt. Testning av verktyget görs mot ett befintligt kundprojekt hos Enea med goda resultat.

The current deliverable describes the initial API available for the solvers. The API is intended to be based on the Kratos Multiphysics fraemework and will evolve during the project. The current deliverable describes the essential features of the interface and provides an initial working implementation to be used as a basis for the future developements. The initial implementation described here is currently operative on the master branch of Kratos. As of the end of July 2018 (moment of handing in of current deliverable) the interface is operative and is being used “in production”. Nevertheless, it still does not fully support the model serialization capabilities that are needed for pyCompSS and HyperLoom. The interface is documented in the project wiki page [wiki]. The same documentation is also presented in the current deliverable.
