Texto completoOptical properties of thin films depend on thickness of each layer. All coatings technologies result in a non uniform thickness distribution. This distribution must be controlled to ensure the uniformity or, on the other side, to spatially modulate optical properties. This requires the development of two tools. The first one is a model which describes the coated thickness distribution, as the second one is an optical set-up for spectrally and spatially resolved measurement of samples. A fit of this model on the measurement of a single layer of each material, allow us to optimise the configuration of the coating chambers for any kind of applications. In this way, we demonstrate how the tilt of the substrate allows us to improve the uniformity. On the contrary, we propose a mechanism to perform a straight iso-thickness lines variable filter by the mean of a mask. In this last case, the model allows us to define the mask translation law

Texto completoHumanity has come a long way from a time where common infections were seen as a death sentence. However, as the organisms driving these infections evolve resistance to treatment, we are forced to face the fast approaching reality of a post-antibiotic era. Antibiotic resistance is evolving at a pace that is significantly ahead of the present-day systems and innovation is urgently required. The aim of my PhD is to do just that by generating an open-science synthetic biology platform to accelerate antibiotic discovery and to further study resistance landscapes of pathogen targets. The platform is based on engineering E.coli as a host to express foreign pathogen targets in order to identify their corresponding antimicrobial drug candidates. This project is also founded on principles of open science and accessibility, which are evident in the development of a frugal drug screening kit and spectrophotometer to engage citizen scientists of all levels

Texto completoThe objective of this thesis was to develop a spectrophotometric sensor to quantify benzene, toluene and xylenes (BTX) in real time at the level of the occupational exposure limit values, ie 20 ppmv for toluene, 50 Ppmv for xylenes and 1 ppmv for benzene. The study was carried out with several sensitive layers, a solid silicic material synthesized by the sol-gel process or thin films (less than 5 μm) deposited on quartz by dip-coating a suspension of nanoparticles or a precursor solution leading to mesoporous silica thin films. We were able to demonstrate the effectiveness of BTX detection in real time. The use of continuous mesoporous films has enabled a correct repeatability of the sensor (standard deviation <10%). Nevertheless, neither these silica films nor their functionalization by methyl groups have solved the problem posed by moisture. Only the use of a dryer that we implemented in the measuring device allowed the detection of toluene in the presence of ambient humidity. In this configuration, interference of 40 ppmv of butanone, acetone and ethanol was also eliminated. Finally, the design of a multi-plate cell allowed to reach detection limits of 1 ppmv at 267 nm for toluene, 1 ppmv at 274 nm for p-xylene and 5 ppmv at 252 nm for benzene. Finally, this limit of detection can be lowered to 1 ppmv at 190 nm for benzene
