Texto completoBackground Androgen Deprivation Therapy (ADT) has side effects which increase the risk of accidental falls and fractures in men with prostate cancer (PCa). Although physical exercise (PE) and healthy lifestyle are recommended in cancer survivors to counteract the side effects of treatment, few individuals comply with the recommended level of PE. Objectives The aim of this Ph.D. research thesis is to ascertain the effectiveness of PE on bone health (bone mineral density (BMD), accidental falls and fractures), and to verify its feasibility and safety in patients with PCa receiving ADT. Methods Two systematic reviews were conducted. We searched in MEDLINE, EMBASE, CINAHL and the Cochrane Library for randomized controlled trials (RCTs) investigating the effectiveness of PE on bone health and, also its feasibility and safety in men with PCa receiving ADT. A cross-sectional study was conducted in an Italian hospital setting to describe the lifestyle of individuals with PCa. Men newly diagnosed with PCa were consecutively invited to participate in a structured interview that was conducted either in person or by telephone. We collected data on their PE habits and motivation to change towards healthier behaviors. Furthermore, we designed an ongoing pilot study that examines the feasibility and safety of a multicomponent experimental PE intervention in patients with PCa that are currently receiving ADT associated with radiotherapy. This PE intervention is aligned with individual preferences, it addresses psychophysical and cognitive functions, and it is specifically targeted at preventing accidental falls and fractures with an appropriate volume of exercise. Results None of the RCTs included in the two systematic reviews investigated the risk of accidental falls and fractures. Nevertheless, preliminary data suggest that multicomponent PE interventions are likely to be effective in reducing BMD loss, especially when involving resistance and impact-loading exercise or football training. Although PE seems feasible in patients with PCa receiving ADT, football training should be prescribed with caution for safety reasons. More than half of the men interviewed in the cross-sectional study (21 out of 40) did not reach the recommended PE level for cancer survivors, and were not willing to change their lifestyle. However, 40% of the sample reported their interest in participating in an exercise program. To date, five participants have been included in the ongoing pilot study: in addition to data on feasibility and safety of the experimental intervention, we investigate its impact on muscle strength, balance, fatigue, mood disturbances, cognitive function, quality of life, and participants’ satisfaction with the intervention. We are also going to record the number of accidental falls and fractures occurring during the intervention, up to one year of follow-up. Conclusions PE is recommended to counteract the side effects of ADT in individuals with PCa. Nevertheless, the evidence of the effectiveness of PE to prevent the risk of accidental falls and fractures and the loss of BMD is lacking. Multicomponent PE targeting bone health seems feasible and safe in this population, but adverse events should be systematically documented, according to current guidelines. Our data suggest that a relevant proportion of the men newly diagnosed with PCa are insufficiently active and, even when exposed to behavioral risk factors, they are not willing to change their lifestyle. Health-care professionals who deal with this population should take advantage of the teachable moment and apply strategies that support patients' motivation to exercise and adherence to healthier lifestyles.

Texto completoQuesto percorso di ricerca, che si colloca nella prospettiva teorica degli studi sociali sulla scienza e la tecnologia, ha l’obiettivo di analizzare se e in quale misura la scienza contemporanea possa essere assimilata ad un fenomeno religioso. Lo sforzo teorico è quello di rielaborare le prospettive analitiche messe a punto della sociologia per lo studio delle religioni, in particolare il paradigma della “religione implicita” e il concetto di “spiritualità”, al fine di verificare se le dimensioni della credenza e dell’esperienza religiosa costituiscano strumenti interpretativi in grado di rendere conto anche dell’impresa tecnoscientifica. Il progetto non riguarda quindi il rapporto tra scienza e religione, ma l’analisi della scienza così come viene narrata dai suoi protagonisti, con particolare attenzione ai riferimenti simbolici e religiosi utilizzati dagli scienziati per esprimere la propria esperienza scientifica. Nella consapevolezza che la comprensione del fenomeno indagato può derivare soltanto da una “teoria dell’esperienza personale”, l’uso e la scelta di strumenti metodologici propri dell’indagine qualitativa, come l’analisi del contenuto e l’intervista in profondità, derivano dalla necessità di esplorare il senso del coinvolgimento degli scienziati nella pratica del lavoro scientifico. La ricerca empirica, a partire dall’analisi della letteratura a disposizione, si articola secondo vari livelli di analisi. Sotto la convinzione che la comunicazione scientifica nello spazio pubblico risulti essere un elemento centrale nel processo di costruzione della conoscenza scientifica, una fase di ricerca è stata dedicata alla ricostruzione dell’annuncio del sequenziamento del genoma umano avvenuto nel 2000; analizzando gli articoli di tre testate nazionali, si intende dimostrare da un lato che le immagini precodificate e le metafore riferite all’universo religioso contribuiscono a promuovere il valore della scienza e, dall’altro lato, che il ruolo dei media, in questo caso delle testate giornalistiche, è rilevante non tanto perché utilizzano questo particolare tipo di linguaggio, religiosamente connotato, quanto invece perché enfatizzano il modo in cui gli scienziati stessi comunicano il proprio lavoro e le proprie scoperte. Per allargare il campo di indagine, una seconda fase di ricerca è stata dedicata all’analisi delle interviste in profondità condotte a scienziati appartenenti a due discipline con diverso grado di maturazione: la fisica e la biologia. L’obiettivo delle interviste non è stato quello di chiedere agli scienziati se la scienza possa essere concepita o meno come una forma di religione, quanto invece raccogliere le loro storie, le immagini rispetto a loro stessi e ai loro oggetti di studio, nonché il senso e i significati attribuiti alla propria esperienza scientifica al fine di verificarlo empiricamente. Una ulteriore conferma si è avuta analizzando i temi emersi nelle interviste di controllo condotte a preti-scienziati, avendo così la possibilità di analizzare, disponendo di un termine di paragone, i racconti delle vocazioni religiose confrontate con quelle scientifiche che in essi coesistono, al fine di rilevarne o meno le affinità. In sostanza, la pervasività di metafore, analogie e termini di riferimento che rimandano all’universo religioso non rappresenta soltanto una particolare forma di comunicazione della scienza, ma si pone in continuità e in sintonia con quanto sentono di vivere gli scienziati attraverso l’esperienza scientifica, dimostrando così che la scienza presenta caratteristiche tali da renderla simile ad un fenomeno religioso sia sul piano della sua rappresentazione sociale, sia su quello dell’identità degli attori che vi prendono parte.

Texto completoThe diet, nutritional status and health are tightly related issues both in industrialized and developing countries. In this doctoral thesis, the main objective was the assessment of nutritional status as a prodromic step to improve it and consenquently health. In industrialized countries, a case study was the nutritional status of a population of central Italy; the intake of some groups of foods, of animal and plant origin, was estimated by food frequency questionnaires and some blood biomarkers. The results demostrated that in dietary controls, aiming to ascertain the long-term effects on health, consumption measurements by questionnaires cannot be exclusive, but some blood and anthropometric indexes could be also useful. In developing countries, two case studies in rural India and D R Congo confirmed that malnutrition is a serious issue; particularly in 3-5 years old children with 26% of severe chronic malnutrition in India and more than 60% in D R Congo. Possible pathways to improve livelihood of rural poor comunities including appropriate food production for a better diet, new income generating activities, infant nutrition improvement, food and waterborn diseases reduction, household food preservation, etc., have been analyzed and the risults of different related trials have been discussed.

Texto completoThe present Doctoral Thesis belongs to the Scientific-Disciplinary Sector of Mechanics of Solids and Structures (ICAR/08 - Scienza delle Costruzioni), and falls within the broader research field of Structural Health Monitoring (SHM), with specific reference to the civil engineering context. Nowadays, SHM-based approaches and the attached development of consistent numerical modeling, with related model updating, may constitute fundamental tools to pursue the goal of engineering structural safety, preventing possible causes of damage that may even lead to structural failure. In particular, this research work proposes complementary post-processing approaches to address the issue of noise cleaning on dynamic structural response signals typically encountered in structural engineering applications (specifically, acceleration and displacement signals). Two approaches are mainly presented, especially aiming at enhancing displacements response signals, since they are commonly affected by higher levels of noise, also due to the low-cost monitoring instrumentation that may possibly be employed. Heterogeneous Data Fusion (HDF) procedures, which involve a Kalman Filter (KF)-based implementation, are primarily investigated, by integrating data acquired from different types of sensors, so that the resulting information turns out to be characterized by a lower degree of uncertainty. A denoising approach is also inspected, as the process through which a source signal may be reconstructed, starting from a recorded, noise-affected one, by removing its noisy part, without losing the useful information incorporated within it. A HDF procedure and a denoising approach are then combined within an integrated strategy, in an effort to enhance the reliability of the monitoring process, for assessing the health conditions of bridges. Aspiring at providing a comprehensive research framework on these topics, both synthetic response signals and real response signals are considered, as well as signals displaying a different nature (non-stationary vs. stationary). These processed response signals are finally employed toward modal identification purposes, for extracting the modal properties of the monitored structure.

Texto completoThis dissertation focuses on the study of the specific articulation, both historical and conceptual, that the interactions between knowledge and life assume in Georges Canguilhem's philosophy. As a matter of fact, his entire work is oriented by the attempt to establish a knowledge of the living being which doesn't betray its vital object/subject. In particular, I address this question (like Canguilhem himself) starting from the constitutive role played by errors, aberrations, and failures, both in life and in knowledge. In this perspective, starting from the examination of both Canguilhem's published works (including the recently published Œuvres complètes) and the unpublished archive material, I outline a path divided into four main sections. First of all, I present an initial problematization (Chapter I) of the notion (which is not self-evident) of "history", used by Canguilhem to approach the medical and biological knowledge of the living being. Then, I analyse (Chapter II) Canguilhem's first articulation of the interaction between knowledge and life, addressing the connections between technique and vital normativity, mainly to the detriment of scientific knowledge. Afterwards, I focus (Chapter III) on Canguilhem's philosophical elaboration of the discoveries of genetics and molecular biology during the 1960s. I argue that this entails a re-connection between scientific knowledge and life, in favour of a conception of scientific knowledge as a vital activity of conceptualisation. In this perspective, I analyse (Chapter IV) Canguilhem's concept of "concept", which is mainly identified with its ability to perpetuate a problem by transcending the specific epistemic configurations that structure its different theoretical solutions. Then, I examine this peculiar functioning within the analysis of the concept of life. In the conclusions, finally, I show how this path leads to a characterization of Canguilhem's philosophy as a practice of thought operating from "outside" the truth of science. Thanks to its peculiar position, this practice of thought can question this same truth, in order to work it differently.

Texto completoThe project was carried out with the aim of finding new alternative and low environmental impact methods for the characterization of commercial and minor wines. In fact, the analyzes carried out at the Centro Interdipartimentale Grandi Strumenti (CIGS) in Modena permitted to analyze the wines of the Lambrusco family, which have now become among the most consumed and exported wines in the world; the wines of the Malvasia family, which are increasingly establishing on the market; the lesser known wines of Emilia-Romagna, produced by the Centro di Ricerca delle Produzioni Vegetali (CRPV) of Cesena, both red and white. Regarding to the red wines, metabolomics proved to be an effective approach, and the use of cutting-edge tools such as the high-resolution mass spectrometer Orbitrap Q Exactive, permitted to identify over 100 anthocyanin molecules. Among them, in addition to the most known forms such as glycosylated, acetyl-, coumaroyl- and caffeoyl-glycosylated, many other derivative forms were identified, such as di-glucosides, which are typical of American vines, but are present in traces also in the varieties of V. vinifera, as well as the combinations with catechins and gallocatechins, with higher molecular weight. In fact, with common analytical methods such as UV spectrophotometry or UV liquid chromatography, it is not possible to see the molecules present in low concentrations, due to the lower sensitivity of the detectors. In addition, UV spectrophotometers allow the identification of molecules only based on the retention times, which often co-elute with each other, even though the chromatographic method has been properly developed. By analyzing the data through the Principal Component Analysis (PCA), the Ancellotta was confirmed, both in terms of quantity and production stability, as the most interesting variety to produce blended wine. Even the Lambrusco Salamino, Grasparossa and Reggiano, are easily distinguishable and have also shown an excellent yield, in terms of color. From a technological point of view, it was possible to distinguish the Ancellotta vinified in red from that vinified in white. In fact, the red vinified one, especially in the 2019 vintage, stands out not only for the high contents of glycosylated anthocyanins, which are typically the most appreciated by winemakers, but also for the large amount of derived anthocyanins mentioned above. Among the minor vines, Festasio proved to be the wine with characteristics very similar to those of the wines already on the market, both in qualitative and quantitative terms, and it can be used as blended wine. Lambrusco di Fiorano (or Lambrusco del Pellegrino), typical of the Modena province, even if with a less abundant color, also showed interesting characteristics for the vinification of Lambrusco. As for the analysis of white wines, an analysis was conducted using mono-dimensional Nuclear Magnetic Resonance (1H-NMR), a typical approach of "untargeted" analysis. This type of analysis, despite being a more approximated analysis than mass spectrometry, allowed the characterization of the minor red wines described above, as well as the minor white wines of Caveccia, Vernaccia del Viandante, Bertinora and Vernaccina, once widespread especially in the coastal provinces of the Adriatic Sea, and the Melara, originally from the province of Piacenza. Moreover, it was possible to discriminate minor wines from wines already on the market, such as Lambrusco and Pignoletto. Of the latter, it was also possible to distinguish the commercial product, and therefore already refermented in the bottle, from the product not yet refermented, and thus with a higher content of residual sugars, and still low in alcohol.

Texto completoG protein-coupled receptors form multimeric complexes comprising G protein and effectors. We want to characterize such complexes comprising delta opioid receptors (DOR) and Kir3 channels, which interest us due to their involvement in opioid analgesia. Immunopurification, BRET and GTPgS binding experiments were done in transfected HEK293 cells. Kir3 channels were co-immunopurified with DOR, implying a spontaneous interaction between the receptor and effector. BRET assays corroborated the presence of this interaction in living cells and allowed us to identify a spontaneous and specific interaction between DOR/Gg and Gg/Kir3, indicating their co-existence within the same complex. Since the activation of the receptor implies it undergoes conformational changes, we were interested in evaluating if the conformational information flows from the ligand-bound receptor until the downstream effector. Hence, we determined the effect of different ligands on the BRET signal that was generated by the following pairs: DOR/Gbg, DOR/Kir3 and Kir3/Gbg. We noticed a modulation of the DOR/Gbg and Gbg/Kir3 interactions that followed the order of efficacy of the ligands to activate the G protein, which we did not observe between DOR and Kir3. Therefore, we concluded that the conformational information flows from the receptor to the Kir3 channel via the Gbg protein. These results allowed us to develop a BRET biosensor (EYFP-Gg2/Kir3.1-Rluc), which could be used in high throughput screening to detect new molecules that activate Kir3 channels with high efficacy.
