Resumen
Rainbow holography is the medium I have chosen to specialize in. Holography itself uses light as a sculptural element. In regards to my work, rainbow holography stresses the field of experimentation with the light spectrum until a certain point that I define as travelling the superhighway from reality to virtual worlds. My work appears then as the documentation, in the form of rainbow holograms, of this travel. It depicts narrative imagery while capturing the moments I existed in this virtual world set behind the rainbow. This project aims to present through still, 3D and filmic imagery the co ??? existence of the physical body and its psychological realm. The psychological reality is articulated as a fictional landscape and the rainbow is used as a metaphor for travel between real and virtual worlds. More importantly, I see holography or rainbow holography as a means of crystallising the vision of the unreachable world behind the rainbow. I see my practice as a new way of using this medium by using this rainbow world as subject matter within the rainbow hologram. By experimenting with combined image processing techniques within rainbow holography, such as analogue white light transmission holograms, full colour digital stereograms, and dot matrix holograms, it becomes possible to generate a synthetic new world. Here each pixel can have the potential to be every color of the rainbow spectrum simultaneously, depending on the angle of the eye of the perceiver. It is here that my investigation through holographic representation has led me to explore and create other worldly landscapes and to extend reality. Our longing to travel over the rainbow into our imagination is with us from a very young age. For me this desire has lasted well into adulthood and has somehow found itself at the centre of my creations over the last few years. The childlike and na??ve appearance of my imagery has the ability to evoke the feeling in the viewers of the nostalgia they may have felt as a child, when confronted with the intense experience and wonder of the imaginings of the rainbow. My work trades on a misunderstanding that the medium of holography is taken as a direct representation of an existing reality. My first hologram I???m a rainbow depicts an alter-egotistical projection of myself as a rainbow princess living in a far away fairytale rainbow galaxy, and communicating with earth beings via the technology of the message contained within the hologram. The hologram has often been associated in science fiction with a message to save the planet. This body of work invites viewers to delve into the depths of their imagination, to save this place where I have travelled by believing in it. As in the story of peter pan where the children are asked to clap their hands if they believe in fairies, by others believing in my imagination they are able to save it. The world within the imagination holds no fixed place; it is a shifting and dynamic space. This quality is shared with the rainbow, which is similarly ephemeral, vanishing and appearing within the eye of the beholder according to weather patterns. The rainbow hologram is a fixed rainbow. When replayed through the eyes of the viewer, the interaction with the real rainbow is recalled, and the viewer enters into the imagination to perceive the work. Throughout this paper I have referred to concepts and techniques in other fields such as physics, anthropology, art history and theory. My research is by no means intended as primarily a technical examination of the medium of rainbow holography. The holographic environments I have made rearrange elements from the real world with fictitious realities. They make people feel as if they are viewing a world that is real, but which imitates unreal ideas. These holographic environments enable viewers to experience ideas as a real place. As Rainbow holography is a relatively new medium, and as my own work uses the rainbow as a multi-layered tool I feel it necessary to investigate the appearance of the rainbow in nature and the reaction of humans to the rainbow as a mythical component in ancient cultures. I am interested in investigating how the rainbow has been used a metaphor for travelling from a material world to ???other worlds??? through its presence in various imaging processes through specific art works. The different ways the rainbow has been used have enabled me to more accurately understand my own work as being a nexus between depicting and generating rainbows. Furthermore, in the discussion of the application of rainbow holography I can show that my own work is necessarily different because of the way I am depicting a rainbow to explore undiscovered territory in which I am the author. Finally I look at how holography is perceived by the public, which helps me to explain the way in which my own work is perceived. Deliberately using the idea of an image in its surrounding context has helped to achieve my desired outcome: to make people believe that the world behind the rainbow really does exist and that I have travelled there, and that they too can do so via viewing my work.

Sommaren 2008 inträffade en extraordinär händelse på nöjesparken Liseberg i Göteborg. Åkattraktionen Rainbow gick sönder och flera passagerare skadades och fick föras till sjukhus. Media har följt denna händelse och det har spekulerats kring Lisebergs agerande och säkerhet.

Ämnena krishantering och medierapportering är ständigt aktuella i dagens samhälle och är kopplade till varandra. Det är viktigt för ett företag att ha bra krisberedskap och en god kontakt med media. Media rapporterar krisen till omvärlden, men en medvetenhet bör finnas om att olika källor kan spegla en händelse på olika sätt.

Syftet med denna uppsats är att se hur Lisebergs krishantering såg ut vid tiden för den extraordinära händelsen sommaren 2008. Vidare avser vi att ta reda på om krishanteringen fungerade som den skulle vid händelsen. I vårt syfte ingår även att undersöka hur tre olika tidningar i Sverige; Aftonbladet, Göteborgs Posten (GP) och Dagens Nyheter (DN) lyfte fram Lisebergs agerande vid händelsen, se hur de lyfte fram händelsen i övrigt och se om det skiljde något mellan deras rapportering.

Vi har använt oss av en kvalitativ metod som har sin grund i hermeneutiken. Vi genomförde en intervju som sedan transkriberades och analyserades. Vidare samlades artiklar in från de olika tidningarna och analyserades med en systematisk textanalys och en iscensättningsmetod.

Vårt resultat som vi fick fram genom ovanstående metoder visade följande: Liseberg hade en väl genomarbetad krishantering vid tiden för olyckan. Denna krishantering ansåg de själva fungera bra och som de hade tänkt från början. Det skiljer sig i medierapporteringen mellan de olika tidningarna. De framställer händelsen på olika sätt, fokuserar på olika saker och framhåller Lisebergs agerande olika mycket.

Arisen from the Pure Parsimony Haplotyping problem in the bioinformatics, we developed the Minimum Rainbow Subgraph problem (MRS problem): Given a graph $G$, whose edges are coloured with $p$ colours. Find a subgraph $F\\\\subseteq G$ of $G$ of minimum order and with $p$ edges such that each colour occurs exactly once. We proved that this problem is NP-hard, and even APX-hard. Furthermore, we stated upper and lower bounds on the order of such minimum rainbow subgraphs. Several polynomial-time approximation algorithms concerning their approximation ratio and complexity were discussed. Therefore, we used Greedy approaches, or introduced the local colour density $\\\\lcd(T,S)$, giving a ratio on the number of colours and the number of vertices between two subgraphs $S,T\\\\subseteq G$ of $G$. Also, we took a closer look at graphs corresponding to the original haplotyping problem and discussed their special structure.

The purpose of this study was to develop a developmentally appropriate new nutrition education tool, the Rainbow Diet for Children (RDFC), to encourage and aid parents in feeding their children according to current national recommendations. In phase I of the study, the RDFC was developed. Foods were grouped based on color. This grouping provided 11 food groups and foods that provide adequate nutrition for children ages 3-6 years. Using a focus group theoretical diets/foods selections in the RDFC were tested for nutrition adequacy. Phase II of the study consisted of actual testing of the RDFC with children. Nutrition intervention was given to children at two Montessori Schools in Miami, FL. The RDFC and the Food Guide Pyramid (FGP) were used as nutrition education tools with different groups of children. Children and their parents were encouraged to follow one of the food guides for two weeks. Fifteen healthy children followed the food guides (9 children followed the RDFC and 6 the FGP) while 7 children served as control subjects. Pre and post nutrition analyses were conducted for all three groups. A pre and post intervention comparison revealed three significant differences. For the FGP group cholesterol intake was significantly (p<0.006) increased and thiamin intake was significantly (p<0.022) decreased. For the control group there was a significant increase (p<0.005) in the vitamin A intake. For the inter group mean change scores (posttest-pretest) two significant differences were found. First, cholesterol intake in the RDFC was significantly (p<0.045) decreased while for the other two groups it increased significantly. Furthermore, the mean monounsaturated fat intake for the RDFC group significantly decreased (p<0.047) from pre to post, whereas in the other two groups it was increased. These findings support our hypothesis that it is possible to create an alternative meal planning system for 3 to 6 year old children. The RDFC group had adequate nutritional intake while following the rainbow diet meal plan.

A comprehensive survey of representative rainbow trout (Oncorhynchus mykiss) hatcheries revealed that the Gram negative, yellow-pigmented, filamentous bacterium Cytophaga psychrophila was implicated in a single disease in the U.K. and other European states. The involvement of C. psychrophila as the aetiological agent of the syndrome was substantiated by the fulfilment of Koch's postulates. Infectivity studies with isolates of C. psychrophila, carried out under natural and laboratory conditions successfully reproduced clinical signs and gross pathological changes analogous to those observed during field outbreaks of the condition. Histopathological examination of artificially and naturally infected fish tissues demonstrated several features that were considered to be pathognomonic for RTFS. Preliminary electron microscopic studies described the ultra-structure of C. psychrophila and partially elucidated the cellular response to the pathogen. Environmental sampling across selected hatchery sites demonstrated that members of the family Cytophagaceae formed a substantial element of the bacterial flora from natural waters, although recovery of C. psychrophila was restricted to areas where the substantial mortalities in fry attributable to RTFS had occurred. C. psychrophila was isolated from the sexual fluids of broodstock and additionally the bacterium was demonstrated associated with surfaces of eyed ova following various disinfection regimes. The minimum inhibitory concentrations of a range of antimicrobial agents both existing in, and novel to, aquaculture were examined, revealing compounds which would potentially mitigate losses attributable to RTFS during field outbreaks. The emergence of bacterial resistance to chemotherapeutants was discussed. The minimum inhibitory and bactericidal concentrations and required exposure times to a number of disinfecting agents were demonstrated. The efficacy of these agents as disinfectants of egg surfaces and equipment associated with fish production was assessed. The potential of a number of serodiagnostic techniques were evaluated as a means of rapid detection of C. psychrophila in diseased fish.

This thesis thrives to promote community health and wellbeing through smart design, celebrating culture(s), and offering efficacious and real-world solutions to mitigate certain challenges arising from the imminent threat of climate change and the gradual depletion of our planet's natural resources. The projected building harnesses naturel forces, minimizes energy consumption, and uses natural/passive strategies like thermal mass and natural ventilation. Interior spaces enjoy an abundance of Natural lighting, biophilic attributes, and thera-serlized or uninterrupted views. It generates electrical energy due to adequate solar power and clear skies, especially in hot and arid climates like the proposed location of the project in Tuba City, AZ. In my proposal of a sustainable, community-focused, wellness center, this project will attempt to embrace diversity, celebrate the Navajos heritage through incorporating their belief system and culture into the genius Loci of the place, which will have a long-term healing effect on patients during their journey of recovery. The Navajo nation is a native American reservation and a self-governing community located in the southwest of the US between four states (UT, AZ, NM, CO). Since it's an Underserved, marginalized, and medically under-resourced community, the Navajo Nation was prone to COVID-19 onslaught in 2020, which resulted in substantial number of cases compared to other US states.
Master of Architecture
In Dec 2020, the World witnessed the first case of Coronavirus disease or COVID-19 in Wuhan, China. The disease has since spread rapidly worldwide, leading to an ongoing pandemic. Like many countries across the globe, the health system in the United States of America has to grabble with this deadly virus by inducing measures such as mask mandates and lockdowns in many US states. Unfortunately, and due to economic and social disparities, COVID-19 pandemic has brought injustice and inequity to the forefront of public health. Some communities were hit hard due to lack of emergency response, the availability of health professionals, and healthcare infrastructure. Tuba city, which is the Diné or the Navajo nation second-largest community in Coconino County, AZ, was majorly hit with COVID-19 resulting in a significant number of cases compared to other US cities. This project is a critical component of an emergency preparedness matrix that can firstly; help absorb the shock of such outbreaks by providing primary and outpatient services. Secondly; it offers community-focused and wellness service that can empower underserved, under-resourced and valuable communities like the Navajo Nation. This project is unique due to its inherited and embedded characteristics of bringing the Navajo tradition into the spirit of the building, by celebrating their culture making it a key component in a patent's healing process.

The present study investigated antibacterial peptides in rainbow trout. Oncorhynchus mykiss. The growth inhibition zone assay was optimised for use with fish bacterial pathogens. Magainin 1 was inactive against all bacteria tested while cecropins P1 and B were very efficient against all Gram negative fish pathogens. The A-layer of Aeromonas salmonicida had a protective effect against cecropins. Cecropin B was more potent and had a wider spectrum of activity than cecropin P1. Defensin HNP-2 was active against three Gram positive bacteria and one Gram negative bacterium. The most sensitive strain to cecropins, A. salmonicida MT004, was chosen as the test microorganism. Purification of antibacterial compounds from rainbow trout was optimised using solid-phase extraction followed by reverse-phase C8 HPLC. Five HPLC peaks purified from gut leucocyte culture supernatants contained inducible antibacterial molecules including a 2131Da-peptide. Five antibacterial HPLC peaks obtained from blood leucocyte culture supernatants contained inducible antibacterial compounds. The most potent weighed 2149g/mol. Its small size, high potency and rapid induction post-challenge would potentate its development as a therapeutic agent. Eight very potent antibacterial peptides weighing between 2500 and 8800g/mol were isolated from serum. The N-terminal sequences of three of them, weighing 2595, 7942 and 7931Da respectively were determined and they were named salmocidins 1, 2 and 3 respectively. The effect of different stimuli on their production was assessed by HPLC and indirect ELISA using a specific polyclonal antibody raised against salmocidin 1. Salmocidin 1 was particularly interesting (large peak on reverse-phase HPLC facilitates its isolation; potency against the test microorganism, small size therefore potentially cheap to synthesise chemically). Salmocidin 1 is a very interesting, novel antibacterial peptide that may provide a satisfactory alternative to antibiotics in fish farming and may find clinical.

Democratic South Africa, just over two decades old, is unavoidably complex, intrinsically hybrid and, as this thesis will argue, anxiously performative. This thesis undertakes to examine bodily practices at key times in contemporary South Africa where national engagement—however divisive or complex—has been centred. It offers a nuanced reading of national performance practices: a reading that considers bodies as both historical constructs and sets of cultural possibilities. Indeed, it is to bodies and embodiment that I look throughout this thesis: the heightened moments or events which reveal tangible, grounded practices behind what I will style as the Geertzian sensibility of the ‘new’ South Africa itself. I argue that this sensibility and the material practices that make up what Geertz termed a “matrix of sensibility” are characterised by both anxiety and performativity – a curious feeling that privileges an extraordinary literacy to reading the body. With the inherent communication difficulties of eleven official languages, South African bodies are the pre-eminent site of national struggle, not only as the bearer of representation—the symbolic gesture of a single raised fist or toyi-toyi protest dance—but as the means of resistance and the enactment of struggle in confrontation or incarceration. I will argue that South Africans are, for reasons both political and practical, particularly attuned towards the nuances of bodies—their skins, their gender, their class—but also their movements, to the processes of embodiment. That even the whitest sporting events resound with the call-and-response strains of an onomatopoeic mining workers’ song speaks to the calling to, and feeling for, an embodied sense of nation. From metaphor to movement, creative expression—often embodied, always imaginative—has been the marker of a national mood. In the following pages, then, I will examine a diverse set of case studies that reveal this sensibility, using them as tethering points for a national cross-narrative constantly created by and through bodies themselves. Rather than demanding strict thematic homologies, I focus on analogues of engagement, discerning a way of seeing and being nation that showcases spectacles of power struggle playing out at the level of the critically anxious South African body. Consequently, this work considers figures such as controversial sprinters Oscar Pistorius and Caster Semenya, carefully connecting their cultural impact to, amongst others, the parliamentary political disruptions by the Economic Freedom Fighters party, the music of rap-rave music group Die Antwoord, Tweede Nuwe Jaar street carnivals and, finally, waves of recent student protest action under the #RhodesMustFall and #FeesMustFall banners.

Mechanically deboned rainbow trout (Oncorhynchus mykiss) was analyzed and then used in texture studies. The meat contained a moderately low amount of fat (10% ± 2%, x ± SD). Pre-deboned meat had more calcium than deboned meat (PIF/=0.37). A combined response model predicted formulations for fish sausage that would closely duplicate the texture of commercially available processed meats. Aquaculture represents a key sector to achieve future food security because of its ability to produce animal proteins, that are needed for human diet. Today, aquaculture accounts for 53% of global fish production, excluding non-food uses. The role of aquaculture will even be enhanced in the future, considering that human population will reach 9 billion of people before 2050 and by taking into consideration the change of diet habits of developing countries, leading to a larger consumption of animal proteins, especially those from fish. Italian fish production is dominated by rainbow trout, which is farmed in raceways, mainly, located in the mountain region of the northeast part of Italy, such as: Friuli Venezia Giulia, Veneto and Trentino Alto Adige. In this work, a bioenergetic model was developed to simulate the individual growth of rainbow trout during a grow-out phase, in relation to water temperature, quality and quantity of feed. The growth was estimated by using an energy budget, which will include anabolism and catabolism and several physiological parameters, that have been selected based on the available literature or that have been calibrated. The calibration of these parameters has been made by comparing the results of the model simulation with experimental data, provided by a fish farmer, located in the province of Trento. Also, an uncertainty analysis was conducted to estimate the confidence level of these parameters.

The Rainbow Family of Living Light is an intentional society devoted to achieving world peace through spiritual healing. A loose association of spiritual seekers that explicitly rejects all forms of leadership and imposed authority, it represents an interesting example of an anarchist and communal society. Rainbow Family events regularly draw thousands of people. These take place all over the world. While some participants may question the label, it can be described as one of the biggest and most geographically diverse New Age groups on the planet. As such, it is a very important factor in shaping the entire present day New Age movement. I conducted fieldwork with the Rainbow Family between the autumns of 1998 and 2002, traveling with the nomadic group throughout the United States. The Rainbow Family rejects any sort of official membership, accepting anyone who attends its events as an equal participant. Spending extended periods of time in the field, I became immersed in this alternative society. The distinction between ethnographic researcher and informants was highly problematic under such circumstances. This made me acutely aware of the issues surrounding fieldwork and anthropological authority. My own work began to seem quite similar to the spiritual seeking of other participants. As such, I began to consider the commonalities between anthropology and the spirituality encountered within the Rainbow Family. The spiritual discourses produced by Rainbow Family participants are uniquely eclectic and ludic in tone. In a setting explicitly championing individual freedom rather than coercion, there is no sense of spiritual orthodoxy. The ways in which spiritual discourses are treated by the Rainbow Family display interesting attitudes towards truth, authority, and reality. These attitudes are reminiscent of epistemological orientations within postmodernist anthropology. Rainbow Family participants find noteworthy solutions to the apparent ontological dilemmas postmodernism presents. It is my hope that looking at the Rainbow Family of Living Light will suggest a viable way for anthropology to productively deal with its current crisis of identity.

This research begins to reflect on how multiracial families navigate racialised difference in everyday life in South Africa. It utilises qualitative data collected in both Mahikeng and Cape Town, to throw light on various people’s lived experience of race in South Africa, whilst concurrently drawing from the large discourse on race in South Africa and elsewhere. The findings suggest that multiracial families are interacting with the remnants of Apartheid still evident in South Africa - most notably in discourses of racially homogenous kinship and racial categorisation – whilst concurrently thinking about new ways to engage with and envision possibilities beyond the dominant discourses of race evident in South Africa at present. These possibilities take the forms of recognising kinship which crosses racial and biological boundaries, engaging with the limitations of Apartheid racial categorisation in a space where Apartheid and all legislation pertaining to interracial relationships has been dismantled, and formulating new language with which to accommodate racial diversity. This implies that whilst South Africa remains haunted by its past, possibilities for alternative ways of engaging with race are emerging. The research contributes to on-going debates about how racialized difference is accommodated within post-apartheid South Africa. It allows for critical reflection on (a) the state of the family in South Africa; (b) formations of difference and similarity and(c) the ways in which historically racialised discourse and practice remain embedded in everyday social interactions.

Monoclonal antibodies (mAbs) have been essential tools in the elucidation of the immune system of mammals, and their application to identify surface molecules on leukocytes have allowed important functions of these cell to be identified (such as receptors that bind antigens, ligands involved in cell to cell signaling or in initiating immune response activity). Not only have mAbs been used to discriminate cells during different stages of cell development, but have also assisted in understanding the dynamics of molecules expressed during functional processes. Such molecules detected on human leukocytes are called human leukocyte differentiation antigens or HLDA. In order to group the antibodies that detect similar molecules and have similar patterns of reaction, immunologists have organised the mAbs that bind to these antigens into Clusters of Differentiation (CD). So far, there are about 350 leukocyte surface molecules detected by mAbs with a CD nomenclature for human leukocytes (www.hcdm.org). In fish immunology there is a great need to produce mAbs that are able to differentiate the various components of the fish immune system to assist in the elucidation of the fish immune system. The present study was an endeavour to develop and characterise mAbs that could be accredited to such scheme. A better understanding of the fish immune system is urgently required so that effective strategies of control can be developed for significant diseases during fish farming. Monoclonal antibodies were prepared by immunizing mice with thymic leukocytes from rainbow trout. The leukocytes were activated with the lectin Concanavalin A to promote the activation and proliferation of the target T-cell population. The selection of clones producing antibodies during screening was performed on the basis of the response of the supernatant from hybridomas using three consecutive assays. First, selection was determined by the positive staining of cells from the thymus in a Dot blot assay. Secondary screening was performed by means of flow cytometry (FCM) and the criterion for selection was the preferential detection of leukocytes gated in the lymphocyte region. Finally, the positive supernatants from hybridomes were evaluated to determine their effectiveness in the detection of modifications in the labelled cells during a multiple way activation by detection of foreign histocompatibility complex enhanced with mitogens. Monoclonal antibody TcOm15 was selected from 564 hybridomas produced and then used to stain cells from various Rainbow Trout tissues. It was clear from FCM, microscopy and Western blot analysis that mAb TcOm15 not only reacted with thymic cells but also with cells from other tissues. Differential staining of cells with mAb TcOm15 was observed with 27.1 ±1.4 % of leukocytes from peripheral blood leukocytes (PBL) stained in comparison to 2.0 ±0.2 % from the thymus, 13.8 ±0.4 % from the spleen, and 5.6 ±0.6 % cells stained from head kidney. The labeled cells showed characteristics of lymphocytes and monocytes, presenting a distinctive staining in immunohistochemistry and confocal microscopy. Western blot analysis, using electrophoresed proteins under denaturing conditions with leukocytes from several different tissues, showed that mAb TcOm15 did not detect a single protein. At least three proteins appeared to be identified by the mAb at 105, 160 and 200 kDa. The proteins were identified as α Actinin-4, non-erythroid Spectrin αII chain or Ig-like protein and non-muscle Myosin (MYH10) by MALDI-TOF analysis. Three of these identities are for compositional molecules for the cytoskeleton of different types of cells, and one it is associated to immunoglobulin superfamily. The identification of these proteins by mAb TcOm15 suggests an ability of this mAb to detect a specific function, possibly related with the synchronicity of expression or interaction of cytoskeleton-membrane proteins forming a multiprotein complex. Another possibility is as a carrier role for a protein during interactions. Colocalization of the mAb with F actin from the cytoskeleton was also observed suggesting the possibility that mAb TcOm15 detects a specific site in a multi-protein complex from the cytoskeleton. The molecule detected showed down-regulation in a dose dependant way with Concanavalin A and the expression was almost lost following stimulation of cells with phorbol 12-myristate 13-acetate stimulation. Leukocytes from the PBL and thymus up-regulated the expression of the TcOm15 molecule under mitogenic conditions in vitro, and results from in vivo experiments suggested the possibility of up-regulation on thymic cells. In conclusion, the results obtained in the present study provide information on a potentially useful marker (mAb TcOm15) for a cytoskeleton-membrane antigen that is modulated during stimulation of teleost lymphocytes. Additionally, this may enable insights into the relationship between cytoskeletal proteins and membrane associated immunoglobulin. Future research is necessary in order to explain this relationship and to determine the functional participation of the TcOm15 molecule during the activation of rainbow trout cells.

The substantial increase in production by the aquaculture industry in the last few decades has been accompanied by a greater demand for prophylactic measures to control fish health, since fish diseases are a major impediment for the profitability of fish farms. To date vaccination has played a major role in the success of fish farming, offering resistance to a limited number of infectious diseases. In order to expand the availability of effective vaccines, a better knowledge of the piscine immune system is crucial. Although a tremendous effort has been put into studies of the fish immune system over recent years, our understanding is still far behind when compared to that of mammals, with many gaps in knowledge still requiring to be addressed. To gain a better understanding of fish immune responses, this thesis has focused on characterising a number of immune molecules considered crucial for disease resistance against extracellular pathogens, in a commercially important species, the rainbow trout (Oncorhynchus mykiss). Adaptive immunity is regulated by a group of specialised lymphocytes, the T Helper (T H) cells. They exert their helper functions through the release of a range of cytokines, which can be used to further categorise them into subsets. One of the latest subsets to be characterised was the T H 17 lineage, which secretes various cytokines, including interleukin (IL )-17 A and IL-22, pivotal in the eradication of extracellular pathogens. To gain a better insight into TH17-type cytokines and their responses in fish, Chapters II and III focused on the characterisation of such molecules, through expression studies, and by analysing their bioactivity as recombinant proteins for the rust time in piscine species. Homology studies confirmed that both molecules are likely to belong to the IL- 22 and IL-17 cytokine families, noting that the trout IL-17 AlP protein shared a close relationship with other piscine IL-17 AlP2 molecules. Interestingly, analysis of their biological activities in splenocyte primary cultures indicated that the two proteins had an effect on the expression of antimicrobial peptides, with IL-22 displaying a potentially more important role than IL-17 AlF. Cytokines are only able to conduct their effects through binding to specific cell surface receptors, acting as ligands. The presence of these particular receptors dictates which cells can be targeted by such cytokines and highlights the importance of the ligand-receptor interaction. To further understand the already identified cytokines, or their family members, a range of receptors was characterised in Chapter IV, with two of them (CRFB4 and IL-17RA) being potentially involved in the signalling of IL-22 and IL-17 AlF ligands. Although homology studies confirmed they belong to the IL-17 and class II cytokine receptor families, orthology of all nine receptors to mammalian homologues could not be inferred. Therefore, further work is required to achieve a better understanding of the homology of these molecules with the mammalian receptors, and their potential involvement with IL-22 and IL-17 AlF intracellular signalling. TH17 cells are characterised not only by the unique range of effector cytokines they secrete, but also through the presence of a master transcription factor, the retinoid-related orphan receptor (ROR)-yt. In Chapter V, two members of the ROR family were characterised, sharing 90% identity between each other. Trout ROR-y1 and -y2 were found to have a close relationship with known ROR-y family members, sharing a particularly high homology with the mammalian splice variant ROR-yt. This suggests that the piscine molecules are likely to be homologues of ROR-yt. However, it still remains to be elucidated whether they can drive piscine TH17 cell differentiation and indeed trigger a TH17 response in fish. T Helper (TH) cells are vital in promoting immune responses. Thus, Chapter VI aimed at developing a potentially effective antibody which could recognise the CD4 marker, typically expressed by this cell subset. The polyclonal antibody, produced using a recombinant protein approach, revealed encouraging results. It was found by Western blot analysis that this antibody could recognise proteins with a molecular weight approximating to that expected for the trout CD4 molecule and its potential splice variant. However, to confirm these observations, further work is still required t? better characterise this antibody and the cells it targets. The results of this thesis have revealed that despite the similarities between the mammalian and piscine immune systems, clear differences exist and also that fish have developed unique molecules and mechanisms not found in mammals. It is therefore of great interest to understand in more detail the role of these novel components so that future progress can be made towards gaining a better comprehension of fish immunity and help advance new vaccination strategies.

ENGLISH ABSTRACT: A breeding programme aimed at the genetic improvement of growth rate of rainbow trout was initiated in 1988 by the Department of Genetics, University of Stellenbosch, in collaboration with the local trout producer's organisations. The first phase of the breeding programme included the collection, evaluation and selection of the best available genetic material from 13 different genetic groups (nine local and four overseas) to make up two separate base populations as odd and even year-groups. This was done to establishment a base population with high genetic merit and variation at the onset of the breeding programme. Statistically significant and commercially valuable genetic differences in terms of weight and length gain were detected between the various hatchery groups. The next two generations of the breeding program included a series of single and double crosses in order to increase the levels of genetic variation in the base populations, and to investigate possible heterosis and specific and general combining ability among the crosses. Significant levels of heterosis (6.7% to 9.6%) and general combining ability was found for weight and length gain during consecutive growth stages. No evidence was found for specific combining ability among the crosses. The crossing of selected offspring from the original genetic groups followed by the application of intensive multi-stage selection for growth rate within progeny groups has led to the establishment of second and third generation parental populations with higher levels of genetic variation and improved individual genetic merit with regard to growth rate. The exploitation of non-additive genetic variation within the base populations through crossbreeding and heterosis during the early stages of the selection programme was delayed in favour of the utilization of additive genetic variance through a procedure of multi-stage selection that incorporated high intensities of selection within and between family groups. The estimation of genetic parameters during the fourth generation on the basis of a hierarchical half-sib family structure confirmed the presence of high levels of additive genetic variation within the respective populations/year-groups. High heritability values in the range of 0.40 to 0.53 were recorded for body weight and length at 150 days. Genetic correlations between the traits were also high, in the range of 0.74 to 0.82. The cumulative realized response of 50% in body length for the EVEN year-group after six generations of selection (8.3% per generation), and the 33% for the ODD year-group after five generations of selection (6.6% per generation) confirms the efficiency of the multi-stage selection procedure to exploit the available additive genetic variation for growth rate within the respective populations. The programme is still ongoing, entering its 7th generation in 2004 and is supplying about 50-60% of commercial material through direct supplies of broodstock, ova and fingerlings and indirect supplies via multiplier stations (commercial hatcheries). The programme was the first of its kind in relation to aquaculture species in the Southern African region, and has since initiated the introduction of programmes of genetic improvement in three other indigenous species, namely tilapia (Oreochromis mossambicus), African catfish (Clarias gariepinus) and abalone (Haliotis midae).
AFRIKAANSE OPSOMMING: ‘n Teelprogram gerig op die verbetering van groeitempo in reënboogforel is in 1988 ingestel onder toesig van die Departement Genetika aan die Universiteit van Stellenbosch, in sameweking met die plaaslike forelprodusenteverenigings. Die eerste fase van die teelprogram behels die versameling, evalasie en seleksie van die beste beskikbare genetiese materiaal vanuit, 13 verskillende genetiese groepe (nege plaaslike en vier van oorsee) om twee basispopulasies te ontwikkel in elk van die gelyke en ongelyke jaargange. Die doel daarvan was om ’n basispopulasie met hoë genetiese meriete en variasie te ontwikkel met die aanvang van die teelprogram gerig op genetiese verbetering, deur middel van seleksie. Statisties betekenisvolle en ekonomies belangrike genetiese verskille in massa- en lengtetoename is aangetref, tussen die onderskeie genetiese groepe. Daar kon geen aanduiding van betekenisvolle, spesifieke kombineringsvermoë gevind word nie. Die kruising van geselekteerde nageslag vanuit die oorspronklike genetiese groepe, gevolg deur ‘n multi-fase seleksiemetode vir groeitempo binne nageslaggroepe, het bygedra tot die ontwikkeling van ‘n tweede en derde generasie broeipopulasie wat beskik oor hoër vlakke van genetiese variasie en verbeterde individuele meriete ten opsigte van groeitempo. The rainbow parrotfish Scarus guacamaia is a prominent herbivore in the coastal waters of southeastern Florida whose life history is strongly linked to a dependence on both mangrove and coral reef habitats. Rainbow parrotfish in turn serve in maintaining the health of coral reefs by keeping algal populations in check. This study used NOAA Fisheries data from the Mangrove Visual Census and the Reef Visual Census in Biscayne Bay and Upper Florida Bay. Observations of abiotic factors at individual sites were used to correlate and predict presence and absence of this species. This was done to visualize habitat presence and ontogenetic shifts present in this species between juvenile and adult stages through ArcGIS mapping. Logistic regression analysis was used to predict presence or absence using the environmental variables of temperature, dissolved oxygen, salinity, average depth, distance from channel openings, mangrove presence, temperature Δ, and salinity Δ. Average depth, distance from channel openings, temperature Δ and salinity Δ were significant in predicting the presence of this species, while salinity, temperature, dissolved oxygen, and mangrove presence were not. Conservation efforts for this species, listed as vulnerable under the IUCN, need to be given greater consideration. The health of this and other parrotfish may have a greater impact on coral reef ecosystems across the Caribbean Sea than currently acknowledged and management breadth and priorities should be adjusted to reflect this role.

Vers la fin des années 1980, les gardes-chasse de l'Île Manitoulin ont organisé et effectué une infiltration technique pour faire cesser une pratique de braconnage et de vente de gibier par des personnes de l'île à des acheteurs de l'extérieur. Cette opération a impliqué 60 gardes-chasse et a abouti à la mise en accusation de 21 membres des Premières Nations. Dans le rocès qui suivit, tous les «Blancs» furent condamnés à des amendes après avoir déposé un plaidoyer de culpabilité tandis que les Amérindiens subirent un long procès de sept ans qui s'est terminé par une décision sans sentence. Ce procès a vu plusieurs rebondissements. Les avocats de la défense ont fait appel à tous les arguments ossibles en se référant au droit constitutionnel. L'argument final fut que les membres des Premières Nations ont des droits ancestraux et des droits reconnus par les traités, des droits de chasse et de pêche sur toute la surface de l'île et des terres nvironnantes. Selon ces traités, ils possèdent parallèlement un droit d'échange de archandise. La question fondamentale est donc de savoir si les droits ancestraux et les traités sont encore compatibles avec l'utilisation de moyens modernes de récolte. Peut-il y avoir pluralisme juridique anachronique? Les politiques modernes sont-elles compatibles avec les coutumes ancestrales?

Resumen
The completion of the color palette has yielded a family of electrochromic polymers (ECPs) each able to absorb in unique regions across the visible spectrum. Synthetically, by varying the electronic content of phenylene type moieties coupled with the donor 3,4-propylenedioxythiophene (ProDOT), high band gaps can be achieved absorbing short wavelength light, yielding a family of yellow-to-transmissive electrochromic polymers. Using the synthetic approach to tune specific absorptions in a discrete region of the visible spectrum, a family of electrochromic polymers that possess sharpened or broadened absorption spectra relative to electrochromic materials previously produced has been developed. By varying the steric hindrance of dioxythiophenes along a conjugated backbone, new hues of magenta and blue have been achieved. Through progressively adding more steric hindrance and twisting the polymer backbone, the absorbance of a polymer can be pushed towards shorter wavelengths, allowing more red light and less blue light to pass through a film. This unequal passing of long and short wavelengths reduces the overall purple color that is normally exhibited by a previous magenta ECP, thereby giving brighter, truer magenta colored materials. By reducing steric hindrance and relaxing the polymer backbone, the opposite can be achieved: pushing the absorbance of a polymer to longer wavelengths allows more blue and less red light to transmit. These polymers also exhibit highly transmissive oxidized states that are attainable at low potentials. In the quest to achieve black (or dark as defined by low L*) to transmissive ECPs with suitable contrast for window or eyewear applications, a relaxed donor-acceptor architecture has been explored. These materials give broad neutral state absorptions with a %Tint (380-780 nm) > 50 %, bringing these materials closer to realization.

This thesis aims to assess what perceptions a sample population of Dutch students in Amsterdam have of South Africa from a broad range of social, political and cultural indicators. Until now, research into the existent perceptions regarding South Africa in the international community has been limited, which has implications for the formulation of its branding strategies and the possibility of their successful implementation at a crucial stage in the development of the country’s international reputation. Based on a theoretical framework which assumes the potential of nation branding for developing states, this thesis aims to provide an assessment of several historical and contemporary challenges faced by Brand South Africa, the most salient of which are linked to the fundamental need for consistency in the promotion of the nation’s identity. This analysis introduces the empirical research upon which the study is based and thereby explains the ambiguous nature of South Africa’s post-Apartheid brand identity.

Le spectacle d'une telle profusion reconduit la "bifurcation" majeure du roman au plan thématique : entre, d'une part, l'intégration tautologique de toute variété et de tout écart, et d'autre part la désintégration du monde et des significations dans la dissipation des formes. La dissipation affecte l'origine et l'autorité des discours, étudiés dans le premier chapître : la circulation des voix effectue un partage, et une dépossession des locuteurs ; la simulation et l'imitation sont une stratégie majeure du pouvoir comme de la résistance au pouvoir. Le deuxième chapître est une étude de la phrase. La multiplication des rapports à la faveur de l'ellipse parataxique, met en crise les continuités et produit des configurations instables. Ce métamorphisme est volontiers ironique et met en doute tout détournement : la révolte romantique contre la synthèse hypotaxique est à la fois ironisée et relayée. Th. Pynchon's style, in his major work Gravity's rainbow, is a sum of expressive ressources. This spectacular profusion exhibits the main "bifurcation" in the novel : between the tautological integration of variety and deviations on the one hand, and on the other hand the disintegration of the world and of meanings in a dissipative formlessness. The origins and authorities of discourses are affected, as shown in the first chapter : the circulation of voices points to a redistribution, but also a dispossession of the speakers ; impersonation and simulation are a major strategy of power, and of the resistance to power as well. The second chapter is a study of the sentence. The multiplication of relationships, thanks to the predominant paratactic ellipsis, produces a crisis of continuity and an instability of the configurations. Such metamorphism is often potentially ironic, and casts a doubt on the forms of deviation : the romantic rebellion against synthesis is replayed, between irony and the search for another subversion. Denomination and metaphor are examined in the third chapter. In the crisis of analogical relationships, ceaseless exchanges appear between symbolical exhaustion, poetic invention, and simulation : the figuration of the other, and of the singular, are everywhere pursued and threatened.
